Abstract:The 2,2′-((1,2 phenylenebis (azaneylylidene)) bis (methaneylylidene)) bis(4-bromophenol) (5BSOP) Schiff base was synthesized, characterized using different spectroscopic techniques, and the data are compared with predictions from DFT computations. The calculated energy values are -8.65 eV (HOMO) and -6.66 eV (LUMO), and the energy gap was found to be 1.99 eV. The reactive atom sites in the chemical are identified by MEP analysis. The locations of localized and delocalized orbitals are revealed through ELF and LOL surface maps. The sites of non-covalent interactions are exposed by RDG and NCI analysis. The possible inter and intra-molecular interactions were determined by NBO analysis and the highest stabilization energy observed was 40.07 kcal/mol. Biological performance as a HMGCS2 expression enhancer was predicted by Pass online studies. Molecular docking simulation with protein 706I having HMGCS2 expression inhibition characteristics revealed a stable receptor-ligand interaction pose at a binding energy of -4.74 kcal/mol. The interaction of the Phe61 amino acid chain of 706I with 5BSOP through conventional H-bond expressed a bond distance of 3.24 Å .

Abstract:The study extensively examined N,N'-(4-methyl-1,3-phenylene)bis(1-(2,4-dichlorophenyl)methanimine) (6D). Advanced spectroscopic techniques, including IR, Raman, NMR, and UV-VIS spectroscopies, were employed to analyse the molecule. The Schiff base was ultimately confirmed using NMR spectrum analysis. The UV-VIS study revealed a notable bathochromic change in the compound, indicating their electronic transitions. By using the HOMO–LUMO bond gap the titled compound reactivity sites were identified. The compound 6D HOMO–LUMO band gap is 3.70 eV. Various wave function investigations like MESP, HOMO–LUMO, RDG, ELF, LOL, and ALIE were conducted to elucidate the distribution of electronic charge, providing valuable insights into the behaviour of molecules. The biological probability of the synthesised compound was extensively assessed using a docking study. Using MEP and HOMO–LUMO investigations, we confirm nucleophilic and electrophilic attacking sites. In the docking study, the protein Bovine cytochrome bc1 complex stigma Tellin bound (PDB ID–1PP9) was downloaded. The docking study identified the most stable minimum binding energy is –6.26 kcal/mol.

Abstract:A novel Schiff base (CDA) was synthesized from camphor and it was characterized by spectroscopic analysis. UV‐Vis and fluorescence spectra were used to quantify the electronic absorption and emission of the compound. The frontier molecular orbital energy gap (3.78 eV) indicated the studied molecule has higher polarizability and chemical reactivity with lower kinetic stability. A new class of (N‐(pyrimidin‐2‐yl)‐4‐((1,7,7‐trimethylbicyclo [2.2.1] heptan‐2‐ylidene) amino) benzenesulfonamide (CDA) was synthesized from commercially available camphor and sulfadiazine. The synthesized Schiff‐base was characterized by FT‐IR, 1H and 13C NMR spectroscopic analyses. The HOMO, LUMO, MEP, chemical reactivity parameters, and NBO were calculated using the WB97XD/cc‐pVDZ basis set. For better understanding, the electronic absorption and emission of the compound were recorded using UV‐Vis and fluorescence spectra. Molecular docking simulations were investigated between ligand (compound) and target protein. The computational results correlate well with observed values. The frontier molecular orbital energy gap (3.78 eV) revealed that the studied molecule has higher polarizability and chemical reactivity with lower kinetic stability. In addition, compound showed the highest binding score −5.72 kcal/mol, which was confirmed by a molecular docking simulation.

Abstract:The Schiff base (5FSATH) was synthesized and characterized by different experimental techniques. In the context of local energy decomposition analysis (LED), the binding energy of 5FSATH-water interaction is determined to be -15.63 kcal/mol, while the bond distance between the two objects is measured to be 1.9781 A°. The absorption spectrum was recorded using different solvents like DMSO, water, and chloroform. The experimental absorption spectrum wavelengths 206, 220, and 300 nm, were recorded from DMSO solvent. In the emission (fluorescence) spectrum we observed three different wavelengths such as 352, 510, and 609 nm. For the calculated NMR spectrum, used the standard GIAO method and DMSO as the solvent. The HOMO-LUMO analysis indicates the charge transfer within the molecule, it is calculated by different solvents. NBO confirms the synthesized compound stability, hyper conjugative interaction, and charge delocalization. To test for antibacterial and antifungal activity, an agar disc well diffusion method is utilized. Synthesized compound tested against Escherichia coli, Candida albicans, Staphylococcus aureus, and Streptococcus pneumoniae microorganisms. Amoxicillin was used as the reference drug for testing antibacterial and antifungal activity. According to the findings of the antimicrobial activity, the result of the molecular docking experiment showed that the compound 5FSATH had an extremely high binding affinity score.

Abstract:In the current study, a molecule (E)-3-(((9H-purin-6-yl)imino)methyl)phenol (A3H) was made and its structure was characterized through the application of spectroscopic techniques. The optimization of the structure of the title chemical was performed using the B3LYP/cc-pVDZ basis set, employing the most stable conformational structure according to the DFT approach. The geometrical characteristics, vibrational wavenumbers, and electrical properties have also been investigated. The electronic characteristics of HOMO-LUMO, UV–Vis, and MEP maps were analysed, including different solvation effects. When compared to the other solvents the water has the lowest energy gap value, due to solvent interaction. The NLO activity has been investigated using the DFT/B3LYP approach, employing the cc-pVDZ basis set. In the NLO study water has the highest dipole moment, polarizability, and hyper-polarizability values. In this study, we have successfully obtained a set of lone pairs of donor–acceptor contacts and a hydrogen bond donor/acceptor surface. These findings provide a basis for explaining a charge transfer process. The utilization of molecular docking techniques has been employed to get insight into the binding interaction between the ligand and targeted proteins.

Abstract:The N, N'-(1,2-phenylene) bis (1- (4- chlorophenyl) methanimine) (CS4) was synthesized and characterized by infrared (IR), absorption (UV-vis) and NMR (1H and 13C) spectral analyses. The structural parameters, vibrational frequencies, potential energy and the distribution analysis (PED) were calculated by using DFT with the basis set of B3LYP/cc-pVDZ and these spectral values were compared to the experimental values. HOMO and LUMO studied were performed in order to understand the stability and biological activity of the compound. The most reactive sites on the compound were investigated by utilizing MEP energy surface and Fukui function descriptor with the natural population analysis (NPA) of the charges. The study of the natural bond orbitals (NBO) reveals the delocalization of the intramolecular interaction of the charges in the compound. Additionally, topological investigations (ELF, LOL), determination of thermodynamic parameters and noncovalent interaction (NCI) study by using topology (RDG) analysis were also carried out. Finally, the molecular docking and molecular dynamics simulations was carried out by examining against glycosylphosphatidylinositol phospholipase D inhibitor receptor for distinct protein targets (3MZG).Communicated by Ramaswamy H. Abstract:The Schiff base (E)-2-((4-chlorobenzylidene)amino)phenol (4CL2AM) has been synthesized from 4-chlorobenzaldehyde and 2-aminophenol. Spectroscopic techniques like FTIR, Raman, UV, fluorescence, and NMR were used to characterize the synthesized compound. The experimental data well matched with the DFT method (B3LYP/cc-pVDZ basis set). The frontier molecular orbital (FMO), molecular electrostatic potential (MEP), and electronic spectra (UV–visible) analysis were conducted on three different solvents such as gas, DMSO, and water. The frontier molecular orbital (FMO) analysis calculated the HOMO-LUMO energy gap as 4.0109 eV, 4.0406 eV, and 4.0411 eV for the gas phase, DMSO, and water respectively. Molecular electrostatic potential (MEP), localized orbital locator (LOL), and electron localize function (ELF) analysis revealed positions of localized and delocalized orbitals as well as electrophilic and nucleophilic attack sites. The natural bond orbital analysis (NBO) predicted highest stabilization energy was 33.37 kcal/mol. The locations of the covalent and non-covalent interactions were investigated by density overlaps region indicator (DORI), interaction region indicator (IRI), and noncovalent interaction (NCI) analysis. Drug likeness and ADME analysis were investigated. In vitro, anticancer activity studies revealed against MCF-7 breast cancer cell lines. In the molecular docking study, the predicted lowest binding energy is −5.77 kcal/mol for the 6NLV-4CL2AM system.

Abstract:A new synthesized Schiff base, N -((anthracen-9-yl) methylene-2-methyl benzenamine) (AMMB), was prepared from o- toluidine and 9-anthracenecarboxaldehyde. The structure of the compound was confirmed by single crystal X-ray diffraction and characterized through various analytical tools such as high resolution mass, 1 H and 13 C NMR, infrared, UV-vis and elemental composition analysis. The molecular geometries of AMMB were optimized by means of DFT/B3LYP/6311-G++ (d,p) basis set. The optimized geometric parameters of the compound were concordant with the single crystal structure. The optimized structure was utilized in comparison of experimental FT-IR with calculated frequency calculation. TD-DFT analysis was performed to compute the electronic absorption of the compound. SCF-GIAO was employed to compute 1 H and 13 C NMR, which was consistent with the experimental results. Additionally, NLO, NBO, MEP and global reactivity parameters were also computed using the same level of theory. Hirshfield surface analysis, fingerprint plot and energy frameworks were presented to explore the intermolecular interactions within the crystal. Molecular docking study was carried out to predict the probable binding site of AMMB into lipid bilayers such as DMPC, DPPC, POPC, DLPE, DOPC and POPE. The molecular docking analysis of AMMB showed complex formation with binding affinity energy ranging from −9.3 to −3.4 kcal/mol.

Abstract:Optimisation of molecular geometry of the headline compound, 4-pyrrolidin-2-yl-pyridine (4P2YLP) was achieved theoretically exercising Density Functional Theory using B3LYP standard approach utilising basis set, 6-311++G(d,p). Using Gaussian 09, HOMO–LUMO analysis was achieved to comprehend the chemical activity and electronic properties of the heading compound. The least HOMO–LUMO gap was obtained for gas phase (5.6486). Bonding interlinkage of the fragments is accomplished by Natural Bonding Orbitals (NBO), as steadiness and chemical reactivity depend on the border molecular orbitals. The nucleophilic & electrophilic spots along with 3D charge transmission areas are determined using the Molecular Electrostatic Potential (MEP). Multiwfn 3.8 software with Pauli Repulsion (PR) & electron localization has been used to conduct ELF and LOL research. While LOL simply displays the most closely spaced orbitals overlapping, ELF displays the electron pair density. Non-linear response properties are analysed in a variety of solvents. The dipole moment (1.9039), polarizability (3.23017E-23 esu) & first order hyperpolarizability (1.51981E-30 esu) of water are the highest values among the selected solvents. Different solvents endured UV–Vis analysis employing TD-DFT technique and the absorption of maximum wavelength is accomplished. Fructose 5-dehydrogenase inhibitor activity by docking is investigated using molecular modelling procedures.

Abstract:A new class of Schiff base, 4-(butan-2-ylideneamino)-N-(thiazol-2-yl) benzenesulfonamide (EMKTH) was synthesized from sulfathiazole by simple condensation with methyl ethyl ketone. The molecular structure was characterized by infrared, 1 H and 13 C NMR, and UV–visible spectroscopic studies. These results were compared with theoretical values. Density functional theory (DFT) calculations were performed to determine the structure and reactivity of the molecule. Further, HOMO-LUMO, MEP, and NBO calculations were performed on this system to examine the electronic and chemical properties of this EMKTH system. The UV spectra were analyzed at ambient temperature to determine the electronic absorption maximum using TD-DFT. In the UV study, there was more solvent interaction in the solvent phase than in the gas phase, which is where the maximum wavelength was observed. Docking simulations were performed on the target protein (1W3R) in order to evaluate the binding affinity of EMKTH. It was found that the target protein had a larger energy of −3.99 kcal/mol. The findings showed that there is a significant correlation between the experimental and theoretical values. The molecule's effective charge transfer mechanism, which accounts for its high chemical reactivity, was suggested by a small orbital gap. The synthesized molecule EMKTH exhibited both intra- and intermolecular charge transfer capabilities in line with the NBO results. Analysis of the EMKTH compound's molecular electrostatic potential (MEP) revealed its reactive sites.

Abstract:The structural and spectroscopic properties of a new -ONNO- type Schiff base were analyzed. The compound 6,6'-((1E,1'E)-(hexane-1,6-diylbis(azaneylylidene))bis(phenylmethaneylylidene))bis(3-(octyloxy)phenol) was synthesized by condensing a diamine and a ketone in tetrahydrofuran, resulting in a high yield. The symmetric Schiff base, in which both amine ends converted to the imine structure, was examined using XRD, NMR, IR and UV-vis. spectroscopies. The theoretical calculations were done successfully using DFT approach at 6-31G(d, p) basis set to investigate the properties of the title compound that cannot be studied by experimental analyses. NBO analysis was used to study hyper-conjugative interactions involving the overlap of sigma (σ) or pi (π) electrons and electron density between adjacent atoms and to determine the stabilization energy. The DFT theoretical parameters and XRD experimental parameters were compared and showed good agreement. 2D fingerprint and 3D Hirshfeld surface plots were obtained to determine the intermolecular interactions. The features of the optimized structure were investigated by several analyses such: NBO, Frontier molecular orbitals, ESP, MEP and ECT which cannot be studied by theoretical methods. The UV-vis. spectroscopy was used to investigate the interaction of the synthesized benzophenone Schiff base compound with lyotropic media. The synthesized compound is significantly compatible with the prepared lyotropic medium due to its highly symmetrical and amphiphilic structure.

Abstract:This research examines the green synthesis of bis-Schiff bases with benzyl substitutions at 3,4-dimethoxy and 3,4,5-trimethoxy positions through solvent-free microwave-assisted condensation between para-phenylenediamine and 3,4-dimethoxybenzaldehyde (I) and 3,4,5-trimethoxybenzaldehyde (II), using sulfated-TiO 2 as a solid acid catalyst. The structures were confirmed through various physicochemical and spectroscopic techniques, including IR, 1H NMR, 13C NMR, and SC-XRD. The compound II crystallized in the monoclinic system (space group of P-21 /C ). The primary objective was to explore the geometry using both experimental methods and density functional theory (DFT) with the B3LYP/6-311G (d,p) level. A comparison of experimental (FT-IR, NMR, SC-XRD) and simulated data showed strong agreement. Additionally, Hirshfeld analysis, ELF, LOL, and RDG topological studies were performed. Docking simulations on the 2V54 monkeypox target protein revealed binding affinities, with compounds I and II. The target protein had a binding energy of -4.83 and -4.98 kcal/mol with I and II. ADMET predictions further demonstrated the pharmacological profile of the synthesized compounds.

Abstract:A new Schiff base compound of N′-[(2,6-dichlorophenyl)methylidene]-2-{[3-(trifluoro-methyl)phenyl] amino}benzohydrazide was synthesized and characterized through various spectroscopic techniques, including infrared, 1 H NMR, 13 C NMR spectroscopy and X-ray diffraction. Experimental results collected by XRD were compared with theoretical results obtained from Density functional theory method. Hirshfeld surface analysis was used to obtain three-dimension molecular surface and two-dimension fingerprint plots to illustrate the intermolecular bonding. Theoretical calculations provide valuable insights into both global and local chemical activity, as well as the properties of molecules and chemicals, including their nucleophilic and electrophilic nature. The DFT method at B3LYP/6-311++G( d , p ) basis set was employed to study the optimized structure and geometric parameters, as well as to explore the frontier molecular orbitals, global reactive parameters, Mullikan population analaysis, Natural bond orbital and molecular electrostatic potential characteristics which cannot be obtained by experimental methods. Additionally, electrophilicity based charge transfer study was carried out with DNA bases to determine the direction of charge transfer. Finally, an investigation was carried out using molecular docking analysis to examine the binding energies of the title compound with PDB ID: 2QDJ protein target. The analysis yielded significant insights into the possible interactions, offering valuable findings in the process.

Abstract:A Schiff base compound 4-(E)-[2-(benzylamino)phenylimino)methyl-2]ethoxy phenol (4BPM2EP) was synthesized and spectroscopic characterization was performed using experimental methods such as FT-IR, FT-Raman and UV-Vis spectroscopy. Density functional theory (DFT/B3LYP/6-311++G(d,p)) computation was used to investigate the optimized molecular geometry, harmonic vibrational wavenumber, NMR chemical shifts, natural bond orbital (NBO) analysis, non-linear optical (NLO) properties, molecular electrostatic potential (MEP) map and Mulliken atomic charges of 4BPM2EP molecule. TD-DFT calculations have been carried out on the optimized geometry at gaseous phase, DMSO and ethanol to further understand the electronic transitions and solvents effect on the UV-Vis spectra of the compound. The assignments of vibrational modes were performed on the basis of total energy distribution (TED) using VEDA 4 program and were compared with experimental data. Molecular docking study was performed using Glide program to establish the information about the interactions between the topoisomerase DNA gyrase enzymes and the novel compound in order to explore the biological behaviour of the examined compound. The compound screened against four pathogens two gram positive, two gram negative and two fungal strains had shown good anti-bacterial and antifungal behaviour. Abstract:Abstract In this work, we consider the multivariate aggregate model introduced in [11], model that takes into account the case when different types of claims affect in the same time an insurance portfolio under some specific assumptions related to the number of claims. For the probability function of the corresponding multivariate compound distribution, [11] obtained an exact recursive formula proved using the properties of the probability generating function. In this paper, we present a new shorter proof of the same formula that we also extend to a new form. Moreover, we present an alternative approximate method to evaluate the compound distribution based on the Fourier transform, and we compare both methods on a numerical example.

Abstract:The research described involves the synthesis and characterization of a new benzohydrazide based Schiff base compound namely 4-methyl-N'-(2,4,5-trimethoxybenzylidene) benzohydrazide (HL). The compound was synthesized by condensing a primary amine and an aldehyde functional group. The characterization of the compound was carried out using various spectroscopic techniques, including elemental analysis, Ultraviolet spectroscopy, nuclear magnetic resonance spectroscopy, and infrared spectroscopy. These techniques helped confirm the excellent quality of the synthesized molecules. In addition to the characterization, the Schiff base compound was subjected to docking studies and biological studies. The antibacterial activity of the compound was tested against three strains of bacteria, namely E. faecalis, B. subtills, and E.coli. The results of these tests provided information on the compound effectiveness against these bacterial strains. Furthermore, docking studies was performed to assess the interaction between the synthesized compound and three target enzymes like GlcN-6-p synthase, DNA polymerase, and EGFR tyrosine kinase. Docking studies are computational simulations that provide insights into the binding interactions between Schiff base ligand and target proteins. In this case, the docking studies helped understand the potential mechanisms of action of the synthesized compound by calculating binding constants and the number of binding modes. The biological activity studies revealed interesting fragmentation patterns, which could be further investigated to understand the compound modes of action. Moreover, the docking studies evaluated the compound's potential as a drug candidate by assessing its binding interactions with the target enzymes. The present study suggests that the synthesized benzohydrazide based Schiff base compound exhibits potential antibacterial activity and shows promising binding interactions with the target protein/enzymes. This information is valuable for designing and developing more potent compounds in the future.

Abstract:The sulfonamide Schiff base (C16H14N4O3S) was successfully synthesized and experimentally ascertained. The main purpose of this research is to investigate the geometry of the aforesaid molecule using both experimental and density functional theory (DFT) techniques and determine its drug likeness characteristics, docking ability as an insulysin inhibitor, and its NLO property. For the computational investigations the DFT approaches were utilized at the B3LYP level with the 6-311G+(d,p) basic set. The experimental results of the compound (such as FT-IR, UV-Vis, and 1H NMR) were compared with simulated data. The both results were well and consistent with previously related published data. The obtained spectral results confirm the formation of the Schiff base compound. Both π-π* and n-π* interactions were found in experimental and computational UV-Vis spectra, as well as in the natural bond orbital (NBO) study. The molecular, electronic, covalent, and non-covalent interactions were analyzed using DFT studies. Both experimental and simulation results revealed that the compound is successfully formed and relatively stable. The compound with a lower band gap showed high chemical reactivity. The medicinal characteristics of the compound were evaluated using in silico medicinal methods. The investigated compound was also followed Pfizer, Golden Triangle, GSK as well as Lipinski's rules. Therefore, the compound has more favorable absorption, distribution, metabolism, excretion, and toxicity (ADMET) profile and it can be used as non-toxic oral drug candidate. The compound was exhibited good insulysin inhibitory activity and it has almost eighteen times higher non-linear optical properties than urea and three times higher than potassium dihydrogen phosphate (KDP).

Abstract:In this study, experimental methods X-ray single crystal analysis, UV-Vis spectroscopy and FTIR spectroscopy, and theoretical method Density Functional Theory (DFT) were used to study the structure of the new Schiff base ( E )- N -(2,6-dichlorophenyl)-1-(5-nitro-2-(piperidin-1-yl)phenyl) methanamine. Theoretical calculations of the title compound and FTIR harmonic vibration frequencies were carried out using B3LYP methods with the 6-31G( d , p ) basis set. The optical properties have been studied by DFT calculation and the experimental UV-Vis spectrum presents its contribution to the HOMO‒LUMO boundary. The molecular electrostatic potential map (MEP-ESP), analysis of frontier molecular orbitals (FMO), and determination of thermodynamic properties for the title compound were conducted using identical levels of theory. Besides, the structural state and crystal packing of the title compound were analyzed using molecular structure, Hirshfeld surfaces, interactions energies, energy frameworks and electrophilicity-based charge transfer (ECT) with DNA bases.
