Abstract:Stover and manure are the main solid waste in agricultural industry. The generation of stover and manure could lead to serious environmental pollution if not handled properly. Composting is the potential greener solution to remediate and reduce agricultural solid waste, through which stover and manure could be remediated and converted into organic fertilizer, but the long composting period and low efficiency of humic substance production are the key constraints in such remediation approach. In this study, we explore the effect of lignocellulose selective removal on composting by performing chemical pretreatment on agricultural waste followed by utilization of biochar to assist in the remediation by co-composting treatment and reveal the impacts of different lignocellulose component on organic fertilizer production. Aiming to discover the key factors that influence humification during composting process and improve the composting quality as well as comprehensive utilization of agricultural solid waste. The results demonstrated that the removal of selective lignin or hemicellulose led to the shift of abundances lignocellulose-degrading bacteria, which in turn accelerated the degradation of lignocellulose by almost 51.2%. The process also facilitated the remediation of organic waste via humification and increased the humic acid level and HA/FA ratio in just 22 days. The richness of media relies on their lignocellulose content, which is negatively correlated with total nitrogen content, humic acid (HA) content, germination index (GI), and pH, but positively correlated with fulvic acid (FA) and total organic carbon (TOC). The work provides a potential cost effective and efficient framework for agricultural solid waste remediation and reduction.

Abstract:The base analog, 2-aminopurine (2AP), was used as a fluorescent reporter of the biochemical steps in the proofreading pathway catalyzed by bacteriophage T4 DNA polymerase. “Mutator” DNA polymerases that are defective in different steps in the exonucleolytic proofreading pathway were studied so that transient changes in fluorescence intensity could be equated with specific reaction steps. The G255S- and D131N-DNA polymerases can hydrolyze DNA, the final step in the proofreading pathway, but the mutator phenotype indicates a defect in one or more steps that prepare the primer-terminus for the cleavage reaction. The hydrolysis-defective D112A/E114A-DNA polymerase was also examined. Fluorescent enzyme-DNA complexes werepreformed in the absence of Mg2+, and then rapid mixing, stopped-flow techniques were used to determine the fate of the fluorescent complexes upon the addition of Mg2+. Comparisons of fluorescence intensity changes between the wild type and mutant DNA polymerases were used to model the exonucleolytic proofreading pathway. These studies are consistent with a proofreading pathway in which the protein loop structure that contains residue Gly255 functions in strand separation and transfer of the primer strand from the polymerase active center to form a preexonuclease complex. Residue Asp131 acts at a later step in formation of the preexonuclease complex.
Screening of lignin-degrading fungi and bioaugmentation on the directional humification of garden waste composting

Abstract:The effective lignin-degrading strains mesophilic Sordaria tomentoalba (MSDA1) and thermophilic Thermomyces lanuginosus (HDGA2) were screened and used for the directional humification of garden waste (GW) composting. The strains MSDA1 and HDGA2 demonstrated significant Laccase (Lac), Lignin peroxidase (LiP), and Manganese peroxidase (MnP) activities, of which MnP activity was the highest at 746.31 U/L and 901.48 U/L, respectively. The composting BA-1 group was inoculated with the fungi MSDA1, HDGA2, M1 (mesophilic cellulose-degrading strain Aspergillus oryzae ), and ZJ3 (thermophilic cellulose-degrading strain Thermoascus aurantiacus ) on day 0, while BA-2 group was inoculated with the fungi MSDA1 and M1 on day 0 and the fungi HDGA2 and ZJ3 on day 3, respectively. The attained humic acid (HA) was 122.2 g/kg and 131.81 g/kg in BA-1 and BA-2 groups, respectively, significantly higher than the control group (72.97 g/kg). The organic matter contents of BA-2 (51.28%) were higher than that of CK (30.55%) and BA-1 (22.04%), and HA/fulvic acid (FA) were 1.26 (CK), 2.04 (BA-1) and 3.10 (BA-2), respectively. The results revealed that bioaugmentation could promote the directional humification by providing more FA precursors with the addition of lignocellulosic degradation fungi. Ammonium nitrogen ( NH4+−N ) could be fixed by FA during the high-temperature period, promoting NH4+−N being assimilated into amino acids and then converted to HA. Bioaugmentation effectively increased the HA and N content of the compost. The sequential addition of mesophilic and thermophilic fungi improved the quality of the directional humification compost.

Abstract:Regulating nitrogen source composition is efficient approach to accelerate the spent mushroom substrate (SMS) composting process. However, currently, most traditional composting study only focuses on total C/N ratio of initial composting material. Rarely research concerns the effect of carbon or nitrogen components at different degradable level and their corresponding decomposed-substances on humification process. This study deciphers and compares the mechanism of mixed manure-N sources on SMS humification from bioavailability and molecular perspective. Two different biodegradable manure-N sources, cattle manure (CM) and Hainan chicken manure (CH), were added into the SMS composting with the different CM:CH ratio of 1:0, 3:1, 1:1, 1:3, and 0:1, respectively. The physicochemical properties and humic substances were determined to evaluate the compost quality. Coupling analysis of spectroscopy, fluorescence, and humic intermediate precursors were conducted to characterizing molecular formation process of humic acid (HA). The results indicated that regulating the carbon-nitrogen nutrient biodegradability of composting material by adding mixed nitrogen sources is an effective strategy to accelerate the SMS humification process. The C1H3 (CM:CH ratio of 1:3) and CH treatments obtained great physicochemical properties and the highest growth rate of HA (31.96% and 27.02%, respectively). The rapid reaction of polysaccharide, ketone, quinone, and amide in DOM (LCP1) might be the key for the fast humification in C1H3 and CH. The polyphenol, reducing sugar and amino acid originated from the labile-carbon-proportion I (LCP1) and recalcitrant-carbon-proportion (RCP), labile-carbon-proportion II (LCP2) and RCP, and labile-nitrogen-proportion I (LNP1), respectively, were the main driving intermediate precursors for the formation of HA. This study deciphers the SMS humification mechanism at molecular level and provides a strategy in accelerating-regulating the composting process. which will be beneficial for enhancing the disposing efficiency of SMS, producing high-quality organic fertilizer, and even popularizing to the similar types of organic waste in practical field.

Abstract:The inoculation of compound microorganisms effectively processes lignocellulosic biomass and promotes the formation of humic acid (HA). However, the basic mechanism of HA formation in compost remains unclear. In this study, we comprehensively characterized the transformation of HA molecules during co-composting. The addition of compound microorganisms enhanced the degradation of proteinoids and lignocellulose in the dissolved organic matter (OM), thereby accelerating the humification of bagasse cow dung co-compost. Elemental and UV–Vis spectral analyses showed that inoculation with the compound microorganisms enhanced the oxidation level and aromatization of the HA structure. Furthermore, two-dimensional Fourier-transform infrared spectroscopy correlation analysis suggested that the addition of compound microorganisms accelerated the decomposition and oxidation of OM. Accordingly, premature aggregation of HA was avoided, and the side chains bonded to HA were more abundant, promoting the evolution of HA to higher-order structures. By influencing the degree of aromaticity, molecular weight, and the C2 fluorescence peak of HA, the inoculation of compound microorganisms promoted HA synthesis during bagasse composting. These results provide an important basis for revealing the mechanism by which compound microorganisms are inoculated to promote HA formation at the molecular level and serve as a reference for the production of high-quality and high-maturity compost.

Effects of humic acid modified oyster shell addition on lignocellulose degradation and nitrogen transformation during digestate composting

Abstract:The aim of this work was to investigate the performance of a novel humic acid modified oyster shell (MOS) bulking agent on the digestate composting. MOS was prepared by immobilizing humic acid onto oyster shell using solid phase grafting method, and then applied to the composting process. Results showed more obvious degradation of lignocellulose was observed in the MOS treatment, which was probably due to the high relative abundance of Actinobacteria. Moreover, the addition of MOS could significantly preserve NH4+ and reduce the NO3- generation with the decreasing abundance of ammonia-oxidizing bacteria and archaea. Besides, adding MOS reduced the N2O emission by 59.63% compared with the control. After composting, excitation-emission matrix fluorescence spectra demonstrated that the humification degree as well as compost maturity was enhanced with MOS added.

Abstract:Chinese medicine residues (CMHRs) resource is attracting widespread attention, as it is expected to be produced into Humus-rich fertilizer for soil application. This study aimed to promote effective humus (HS) production through lignocellulose-degrading bacteria (LDB) addition and explore the biological regulation mechanism of LDB affecting lignocellulose-to-humus conversion. The results showed higher HS production was achieved, with 109.73 and 111.44 g·kg −1 , and HA/FA was raised by 12.70–16.02 % in compost products by LDB addition stimulation. Significant upregulation of β-glucanase and xylanase activities catalyzed higher decomposition of lignocellulose toward more HS potential precursors supply. Furthermore, exogenous LDB intervention induced microbial community restructure and microbial network establishment via enriching synergism functional bacteria, i.e., Thermobifida , Paenibacillus , Nonomuraea , etc. Mantel test results showed that it was variation of cellulose, hemicellulose and HS that affected microbial community succession ( p < 0.01, r > 0.6), which represented the positive action of LDB addition stimulation on HS synthesis upregulation. Further exploration suggested LDB had an indirect effect on HS formation by enhanced lignin and hemicellulose conversion based on the Random Forest model and Partial least-squares path modeling results. This research provides new insights into the trigger effects of LDB introduction on upregulating HS synthesis and is expected to propose new perspectives for HS efficient production in CMHRs composting.

Abstract:Respiratory muscle weakness may induce dyspnoea, secretion retention and respiratory failure. Assessing respiratory muscle strength is mandatory in neuromuscular diseases and in case of unexplained dyspnoea. A step by step approach is recommended, starting with simple volitional tests. Using spirometry, respiratory muscle weakness may be suspected on the basis of an abnormal flow-volume loop or a fall of supine vital capacity. When normal, maximal inspiratory and expiratory pressures against a near complete occlusion exclude significant muscle weakness, but low values are more difficult to interpret. Sniff nasal inspiratory pressure is a useful alternative because it is easy and it eliminates the problem of air leaks around the mouthpiece in patients with neuromuscular disorders. The strength available for coughing is easily assessed by measuring peak cough flow. In most cases, these simple non invasive tests are sufficient to confirm or to eliminate significant respiratory muscle weakness and help the timely introduction of ventilatory support or assisted cough techniques. In a minority of patients, a more complete evaluation is necessary using non volitional tests like cervical magnetic stimulation of phrenic nerves.
The microbial mechanisms of enhanced humification by inoculation with Phanerochaete chrysosporium and Trichoderma longibrachiatum during biogas residues composting

Abstract:This study investigated effects of composite microbes (CMs) (Phanerochaete chrysosporium and Trichoderma longibrachiatum) on humification during co-composting of biogas residue, spent mushroom substrate and rice straw. Results showed that CMs inoculation elevated degradation ratios of cellulose, hemicellulose and lignin by 7.86%, 8.87% and 6.45%, and contents of humus and humic acid were correspondingly promoted by 15.5% and 23.6%, respectively. Relative abundances of bacteria associated with refractory macromolecules degradation (Flavobacterium, Anseongella and Actinomadura) and cellulolytic fungi (Hypocreales_Incertae_sedis, Hypocreaceae and Psathyrellaceae) were raised by CMs addition. Redundancy analysis demonstrated a positive correlation between microbial communities and temperature, fulvic acid and lignocellulose contents. Moreover, CMs inoculation promoted pathways of xenobiotics biodegradation and metabolism, and biosynthesis of other secondary metabolites, which was closely associated with lignocellulose degradation and humus formation. These results suggested that biological inoculation could enhance composting efficiency and improve compost quality, benefiting biogas residues composting.

Abstract:The study examined the impact of adding cattle manure to the composting process of Agaricus bisporus mushroom substrate on compost humification. A control group CK comprised entirely of Agaricus bisporus mushroom substrate, while the experimental group CD (70 percent Agaricus bisporus mushroom substrate and 30 percent cattle manure) comprised the two composting treatments that were established. The study determined that the addition of cow dung has promoted the formation of humus components. Particularly, humic substance (HS-C) and humic acid (HA) increased by 41.3 and 74.7%, respectively, and the ratio of humic acid to fulvic acid (HA/FA) also increased by 2.78. It showed that the addition of cow dung accelerated the synthesis and decomposition of precursors, such as polysaccharides, polyphenols, and reducing sugars. Thereby promoting the formation of humic acid. Network analysis revealed that adding cow dung promoted microbial interactions increased the complexity and stability of the bacterial and fungal symbiotic network, enhanced cooperation and reciprocity among microbes, and assisted in transforming fulvic acid (FA) components. Structural equation modeling (SEM) is a multivariate data analysis method for analyzing complex relationships among constructs and core indicators. SEM illustrated that introducing cattle manure into the composting process resulted in alterations to the correlation between physicochemical parameters and the microbial community, in addition to humus formation. Polysaccharides are the primary precursors for polymerization to form HA, which is an essential prerequisite for the conversion of fulvic acid to humic acid. Additionally, microbes affected the formation of humus, with bacteria substantially more influential than fungi. These findings provide new ideas for regulating the degree of humification in the composting process and have important practical implications for optimizing mushroom cultivation and composting techniques today.

Abstract:Composting can convert organic materials into stable humus (HS), resulting in high-fertility organic fertilizers that were widely used in the treatment of agricultural wastes. Therefore, it would be beneficial to strengthen the degree of composting humification by exploring the formation law and the main influencing factors of composting humification for different agricultural wastes. In this study, straw, cabbage, chicken manure, and pig manure were used as research objects, and the industrial mixed-material strip-stack composting and the single-material laboratory reactor composting were carried out, respectively. The results showed that the industrial mixed-material composting mode was beneficial to the continuation of the high-temperature period for composting (the heat lasts for 20 days), and the high lignin treatment T2 (straw + chicken manure composting) helped to promote the degradation of lignin and formation of humic acid (HA). At the end of composting, the absolute degradation of lignin in T2 treatment increased by 13.18% and 120.02% ( p < 0.01) compared with T1 (straw + cabbage + chicken manure) and T3 (straw + pig manure) treatments, respectively ( p < 0.01), and the humic acid (HA) content increased by 11.78% and 26.59% ( p < 0.05). In the single-material laboratory reactor composting system, the continuous high-temperature period (31 days) contributed to the degradation of the composting materials and the formation of HA in the straw (TS) composting treatment; at the end of composting, the absolute degradation amount of TS lignin increased by 137.98%, 170.99%, and 196.82% compared with TV (cabbage), TC (chicken manure), and TP (pig manure) treatments ( p < 0.01), and the HA content increased by 281.76%, 377.57%, and 460.71% ( p < 0.01), respectively. Redundancy analysis showed that temperature was the main environmental factor affecting the decomposition and humification process of composting organic materials; the increase of lignin absolute degradation was helpful to promote the formation of HA. This study found that temperature was the main environmental factor that drove lignin degradation and promoted the humification process of agricultural waste composting; the lignin content in the composting material was the basis for the formation of HA, and the absolute degradation amount of lignin was the main material factor that promoted the humification process.

Abstract:This study aimed to understand the effects of using apple branch biochar (ABC) and manganese (Mn (II) ) to enhance the humification process when sawdust and hog manure are being composted aerobically. Four treatments of sawdust and hog manure were established, namely, CK (no additives), ABC (5 % ABC), Mn (0.5 % MnSO 4 ), and ABC-Mn (5 % ABC + 0.5 % MnSO 4 ). The results from the 42-day composting showed that ABC-Mn treatment was the most effective. It increased the total organic matter reduction by 19.24 %, decreased CH 4 emissions, produced a high level of humic acid (HA) (60.45 g/kg), and significantly elevated the degree of polymerization (HD) to 3.15 ( P < 0.05) upon the end of the composting process. In addition, the presence of ABC abiotically reduced Mn (II) salt stress and promoted compost maturation. Spectroscopic examination revealed that ABC-Mn treatment facilitated the breakdown of polysaccharides and the production of stable, aromatized humus-like materials throughout the composting process. The addition of Mn (II) and ABC also changed the microbial community composition of the compost substrate, stimulated the activities of laccase and dehydrogenase, and raised the relative abundances of Ascomycetes , Firmicutes , Proteus , Actinomycetes , and Basidiomycetes . ABC addition accelerated the transformation of Mn (II) to reducible Mn (II) and Mn (IV) , potentially enhancing the abiotic degradation of organic matter and the formation of humus-like substances through redox reactions. In conclusion, this study highlighted the biotic and abiotic effects of adding ABC and Mn (II) to accelerate humification during the co-composting of hog manure and sawdust.

Abstract:Present study investigated the effects of hydrothermal pretreatment (HTP) and addition of compound microbial agent (CMA) on humification, odour generation and metabolism functions of bacterial communities during composting of kitchen waste (KW). Surprisingly, HTP and CMA addition treatment could promote the humification of compost and the control of odour units in contrast to the control (without HTP and CMA addition). The humic acid to fulvic acid ratio of end compost increase by 187.30 %, while humification index (HIX) increased by 18.87 %. 3D-EEM fluorescence spectroscopy of dissolved organic matter (DOM) demonstrated that it facilitated the synthesis of humified compounds and the decomposition of biodegradable compounds. Moreover, the SUVA 254 , SUVA 280 and E 253 /E 203 increased by 118.6 %, 115.25 % and 42.11 % after HTP and CMA addition indicating an increase in aromatic carbon abundance. VFAs had the higher degradation rate (84.91 %) than other treatments (57.46–77.72 %). Meanwhile, the main contributor to the malodorous odour was isovaleric acid, followed by butyric acid and acetic acid during composting. Mantel test indicated that the humification degree was significantly influenced by environmental parameters (temperature, pH, etc.) and metabolic products (HA, DOC and VFAs). Metagenomic analysis indicated that the biodegradation processes at the thermophilic stage were controlled mainly through genes involved in microbial metabolism. HTP and CMA addition was an eco-friendly and efficient strategy to reduce odour emission and improve the compost quality.

Abstract:The aim is to enhance lignin humification by promoting laccase activities which can promote lignin depolymerization and reaggregation during composting. 1-Hydroxybenzotriazole (HBT) is employed to conduct laccase mediator system (LMS), application of oxidized graphene (GO) in combination to strengthen LMS. Compared with control, the addition of GO, HBT, and GH (GO coupled with HBT) significantly improved laccase expression and activities ( P < 0.05), with lignin humification efficiency also increased by 68.6 %, 36.7 %, and 107.8 %. GH treatment induces microbial expression of laccase by increasing the abundance and synergy of core microbes. The unsupervised learning model, vector autoregressive model and Mantel test function were combined to elucidate the mechanism of action of exogenous materials. The results showed that GO stabilized the composting environment on the one hand, and acted as a support vector to stabilize the LMS and promote the function of laccase on the other. In GH treatment, degradation of macromolecules and humification of small molecules were promoted simultaneously by activating the dual function of laccase. Additionally, it also reveals the GH enhances the humification of lignocellulosic compost by converting phenolic pollutants into aggregates. These findings provide a new way to enhance the dual function of laccase and promote lignin humification during composting. It could effectively achieve the resource utilization of organic solid waste and reduce composting pollution.

Bacillus licheniformis inoculation promoted humification process for kitchen waste composting: Organic components transformation and bacterial metabolic mechanism

Abstract:Kitchen waste (KW) composting always has trouble with slow humification process and low humification degree. The objective of this study was to develop potentially efficient solutions to improve the humification of KW composting, accelerate the humus synthesis and produce HS with a high polymerization degree. The impact of Bacillus licheniformis inoculation on the transformation of organic components, humus synthesis, and bacterial metabolic pathways in kitchen waste composting, was investigated. Results revealed that microbial inoculation promoted the degradation of organic constituents, especially readily degradable carbohydrates during the heating phase and lignocellulose fractions during the cooling phase. Inoculation facilitated the production and conversion of polyphenol, reducing sugar, and amino acids, leading to an increase of 20% in the content of humic acid compared to the control. High-throughput sequencing and network analysis indicated inoculation enriched the presence of Bacillus, Lactobacillus, and Streptomyces during the heating phase, while suppressing the abundance of Pseudomonas and Oceanobacillus, enhancing positive microbial interactions. PICRUSt2 analysis suggested inoculation enhanced the metabolism of carbohydrates and amino acids, promoting the polyphenol humification pathway and facilitating the formation of humus. These findings provide insights for optimizing the humification process of kitchen waste composting by microbial inoculation.

Abstract:The innovative biodrying-enhanced composting (BEC) process produces highly matured fertilizer within 10 d. To clarify the biodrying-accelerated humification mechanism, structural and molecular variations in humic acid (HA) during BEC were compared to those during 16-d bioaugmented mechanical composting without biodrying. Results showed that BEC produced HA with significantly higher aromaticity and molecular size (p < 0.01). More aromatic skeletons, mainly from biodrying-enhanced lignin decomposition (p < 0.05), contributed to HA aromatization. Reactive phenolic hydroxyls on these skeletons facilitated the binding of other humic precursors, promoting HA elongation. Microbial analysis indicated that Bacillus, Sinibacillus, and Issatchenkia, enriched by drastic heating and dehydration during days 0–3, participated in lignin decomposition. Saccharomonospora, Georgenia, Oceanobacillus, Nigrospora, Kluyveromyces, and Aspergillus contributed to HA elongation during the maturation phase (days 3–9). This study’s findings that biodrying enhanced lignin-related humification pathways by enriching functional microorganisms provides a theoretical foundation for further improving compost humification efficiency.
Deciphering the carbon and nitrogen component conversion in humification process mediated by distinct microbial mechanisms in composting from different domestic organic wastes

Abstract:This study aimed to compare the process of maturity and humus fraction evolution as well as bacterial community dynamics in composting from different domestic organic wastes (food waste (FW), and vegetable waste (VW)) and decipher the key biotic influencing factors of humic acid formation through correlation analysis and ecological network. The results showed that organic carbon components in FW with high ratio of soluble organic carbon and hemicellulose were more easily to be degraded in composting compared to VW. After 30 days of composting, the content of HA-C generated by VW was 35.41%, higher than 29.01% of FW, and the growth rate of HA-C generated was 38.42% and 28.34%, respectively. PARAFAC analysis showed that the structure of HA generated in VW was more complex, and the proportion of humic acid-like components (C3 + C4) was 60.32%, while FW only accounted for 43.86%. However, the evolution growth rate of aromatic components in HA in FW was 26.88% in 30 days of compost, which was higher than 15.17% in VW. High-throughput sequencing indicated that Lactobacillus was the initial dominated genera in composting from different domestic wastes. Thermobifida, Thermovum, and Pusillimas as well as Aeribacillus were core bacterial genera that promoted the humification process in FW and VW, respectively. Network analysis showed that there was higher bacterial interacted connection degree and complexity in FW compared to VW. This study was of great significance for optimizing organics conversion and humification efficiency of household waste composting.

Abstract:Abstract This prospective study compares the results of pre-operative clinical examination and simple ultrasound examination of the neck in 18 patients with proven head and neck primary tumours and palpable cervical lymphadenopathy, who then underwent 21 radical neck dissections. Neck palpation and ultrasound examination were compared with histological examination. Ultrasound did not add significantly to the information obtainable by simple neck palpation in this group of patients.
Effect of thermophilic bacterial complex agents on synergistic humification of carbon and nitrogen during lignocellulose-rich kitchen waste composting

Abstract:This work reported the effects of thermophilic bacterial agents on degrading persistent lignocellulose and reducing the loss of valuable nitrogen in kitchen waste (KW) composting. The results showed that thermophilic bacterial compound agents improved the high temperature period by 8 days, and increased the ligninase activity by 0.5-3 times during the composting process. The activity of cellulase increased up to 1 time in agent A (Geobacillus, Clostridium caenicola, Haloplasma) adding group by improving the microbial activity of lignocellulosic degradation metabolic pathways. Nitrogen storage increased to 70% in group added with agent B (Clostridium caenicola, Geobacillus, Clostridium sp. TG60-81) by increasing the population abundance of nitrogen-fixing microorganisms such as Bacillus, Hungateiclostridium and Herbaspirillum, and changed amino acid metabolic pathways. In general, agents A and B could increase the thermophilic phase, optimize the microbial community structure, realize the synergistic humification of carbon and nitrogen, and convert KW into mature and high quality fertilizers.
