Abstract:Discovery of a remnant habitable environment by the Mars Science Laboratory in the sedimentary record of Gale Crater has reinvigorated the search for evidence of martian life. In this study, we used a simulated martian mudstone material, based on data from Gale Crater, that was inoculated and cultured over several months and then dried and pressed. The simulated mudstone was analysed with a range of techniques to investigate the detectability of biosignatures. Cell counting and DNA extraction showed a diverse but low biomass microbial community that was highly dispersed. Pellets were analysed with bulk Elemental Analysis - Isotope Ratio Mass Spectrometry (EA-IRMS), high-resolution Laser-ablation Ionisation Mass Spectrometry (LIMS), Raman spectroscopy and Fourier Transform InfraRed (FTIR) spectroscopy, which are all techniques of relevance to current and future space missions. Bulk analytical techniques were unable to differentiate between inoculated samples and abiotic controls, despite total levels of organic carbon comparable with that of the martian surface. Raman spectroscopy, FTIR spectroscopy and LIMS, which are high sensitivity techniques that provide chemical information at high spatial resolution, retrieved presumptive biosignatures but these remained ambiguous and the sedimentary matrix presented challenges for all techniques. This suggests challenges for detecting definitive evidence for life, both in the simulated lacustrine environment via standard microbiological techniques and in the simulated mudstone via analytical techniques with relevance to robotic missions. Our study suggests that multiple co-incident high-sensitivity techniques that can scan the same micrometre-scale spots are required to unambiguously detect biosignatures, but the spatial coverage of these techniques needs to be high enough not to miss individual cellular-scale structures in the matrix.
Remote and in-Situ Characterization of Mars Analogs: Coupling Scales to Improve the Search for Microbial Signatures on Mars

Abstract:Past environments on Mars contained abundant water, suggesting certain regions may have been conducive to life as we know it and implying the potential for microbial inhabitants. Gale and Jezero craters, home of the Perseverance and Curiosity rovers, hosted ancient lakes that experienced periods of active hydrologic cycling and prolonged drying intervals. Exploration of these basins (and future operations on Mars) will benefit from detailed characterizations of analogous environments on Earth, where life detection strategies at various spatial scales (i.e., rover to orbiter) can be tested and validated. Investigations of terrestrial analogs are critical for understanding (1) how microorganisms generate chemical biosignatures in environments characterized by multiple extreme conditions; (2) the impact of environmental conditions and mineralogy on biosignature preservation; and (3) what technologies and techniques are needed to detect biosignatures remotely or in situ . Here, we survey five terrestrial sites analogous to climate conditions proposed for Late Noachian to Early Hesperian Mars, when craters are thought to have hosted active lakes. We review the geologic setting, environmental conditions, microbial habitability, extant microbial communities, and preserved biomarkers at each analog and discuss their relevance to the search for signs of life in Martian craters with in situ and remote instrumentation. The analogs range from active to desiccated lake systems, temperate to hyper-arid climates, and have acidic to neutral-pH and hypo- to hyper-saline waters. Each analog hosts microorganisms adapted to multiple extremes (polyextremophiles), including aspects of water availability (i.e., surface waters versus shallow subsurface water versus groundwater) and physiochemistry (e.g., water activity, salinity, temperature, alkalinity, pH, and redox potential) that can form macrobiological features such as microbial mats. Comparing the expected achievable spatial resolution of several key Mars instruments to the spatial extent of macrobiological features at each analog reveals that most features are unlikely to be resolved from orbit and require rover-scale instruments for detection. We recommend that future studies at these analogs use multi-scale remote sensing surveys to determine thresholds for detecting macrobiological features and map how patterns in mineralogy or physical characteristics of environments correlate to modern-day microbial communities or preserved biomarkers. It will also be critical to determine how the characteristics of macrobiological features, such as areal extent, percent cover, thickness, pigments, etc., impact detectability thresholds. These findings can provide vital information on potential topographic or spectroscopic signatures of life, and at what scales they are detectable. This research is critical to guide sample collection locations within craters like Jezero, and for selecting landing sites for future missions in evaporative Martian basins and other rocky bodies.

Abstract:This paper empirically analyzes the impact of the Sarbanes-Oxley Act on the relation between measures of corporate governance and a firm’s dividend policy in the U.S. equity market. Using the IRRC database, we find that there is a statistically significant relation between governance measures and a firm’s dividend policy in the years prior to the introduction of the Sarbanes-Oxley Act. However, following Sarbanes-Oxley, the relation between a firm’s governance structure and dividend policy changes. In particular, shareholders’ rights and the proportion of outside directors are no longer significant in explaining a firm’s dividend policy.

Abstract:The NASA Mars 2020 Perseverance rover is actively exploring Jezero crater to conduct analyses on igneous and sedimentary rock targets from outcrops located on the crater floor (Máaz and Séítah formations) and from the delta deposits, respectively. The rock samples collected during this mission will be recovered during the Mars Sample Return mission, which plans to bring samples back to Earth in the 2030s to conduct in-depth studies using sophisticated laboratory instrumentation. Some of these samples may contain traces of ancient martian life that may be particularly difficult to detect and characterize because of their morphological simplicity and subtle biogeochemical expressions. Using the volcanic sediments of the 3.45 Ga Kitty's Gap Chert (Pilbara, Australia), containing putative early life forms (chemolithotrophs) and considered as astrobiological analogues for potential early Mars organisms, we document the steps required to demonstrate the syngenicity and biogenicity of such biosignatures using multiple complementary analytical techniques to provide information at different scales of observation. These include sedimentological, petrological, mineralogical, and geochemical analyses to demonstrate macro- to microscale habitability. New approaches, some unavailable at the time of the original description of these features, are used to verify the syngenicity and biogenicity of the purported fossil chemolithotrophs. The combination of elemental (proton-induced X-ray emission spectrometry) and molecular (deep-ultraviolet and Fourier transform infrared) analyses of rock slabs, thin sections, and focused ion beam sections reveals that the carbonaceous matter present in the samples is enriched in trace metals ( e.g. , V, Cr, Fe, Co) and is associated with aromatic and aliphatic molecules, which strongly support its biological origin. Transmission electron microscopy observations of the carbonaceous matter documented an amorphous nanostructure interpreted to correspond to the degraded remains of microorganisms and their by-products (extracellular polymeric substances, filaments...). Nevertheless, a small fraction of carbonaceous particles has signatures that are more metamorphosed. They probably represent either reworked detrital biological or abiotic fragments of mantle origin. This study serves as an example of the analytical protocol that would be needed to optimize the detection of fossil traces of life in martian rocks.

Abstract:To assess whether life existed on Mars, it is crucial to identify geochemical biosignatures that are relevant to specific Martian environments. In this paper, thermochemical modeling was used to investigate fluid chemistries and secondary minerals that would have evolved biotically over geological time scales in Martian fluvio‐lacustrine and evaporitic settings, and that could be used as potential inorganic biosignatures for life detection on Mars. Modeling was performed using fluid and rock chemistries relevant to Gale crater aqueous environments. Potential inorganic biosignatures were identified investigating alteration deposits found at the surface of a simulant exposed to short‐term bio‐mediated weathering and comparing experimental and modeling results. In a fluvio‐lacustrine setting (water/rock of 2000–278), models suggest that less complex mineral assemblages form during biotic basalt dissolution and subsequent brine evaporation compared to what would happen in an abiotic system. Mainly nontronite, kaolinite, and quartz form under biotic conditions, whereas celadonite, talc, and goethite would also precipitate abiotically. Quartz, sepiolite, and gypsum would precipitate from the evaporation of fluids evolved biotically, whereas nontronite, talc, zeolite, and gypsum would form in an abiotic evaporitic environment. These results could be used to distinguish products of abiotic and biotic processes, aiding the interpretation of data from Mars exploration missions.

Abstract:A high priority goal for past, present and future missions to Mars is the search for evidence of past or present life. Some of the most information-rich signals are those represented by organic biomarkers. Thermal extraction has historically been the most popular in situ analysis technique employed on Mars owing to its elegance and ability to liberate both small compounds and large macromolecular networks. The geological record of Mars contains a variety of minerals, some of which can interact with organic matter when subjected to thermal extraction. Here we discuss the organic records that may be associated with these mineral hosts and the problems encountered when mineral-organic mixtures are analysed by thermal-based methods. We also suggest potential mitigations for future experiments of a similar nature and note that these mitigating steps can be applied not only in situ on Mars but also after samples are returned to Earth as part of Mars Sample Return where more resources and time for sample preparation are available. Abstract:Siliceous hot spring deposits, or sinters, deposit from hot spring discharge at Earth's surface and are sites of exceptional preservation of biosignatures. Their macro‐ and micro‐textures are regarded as important evidence of past microbial activities in hydrothermal environments. However, biology mimics do occur, and bona fide microbial textures could be destroyed by subsequent diagenesis or other post‐depositional processes. Thus, it is paramount to narrow the search for prospective Martian silica‐rich deposits that may contain biosignatures from both orbital and rover‐based perspectives. This study investigates hydrothermal deposits in Chile, which are analogs of high‐silica deposits discovered in the Gusev crater on Mars, through remote sensing and laboratory analysis. Results indicate that compositional remote sensing based on multispectral data with a high spatial resolution of <4 m/pixel reflects various concentrations of silica, which assisted in identifying the direction of discharged hydrothermal flows from the vent to the apron. Micro‐infrared mapping of sinters from similar hydrothermal fields linked spectral features to specific textures revealed by scanning electron microscope and chemical compositions confirmed by electron microprobe analysis, indicating that sinters with no shift in their emissivity minimum in the thermal infrared range were more likely to preserve cellular structures. An instrument for collecting multispectral data with higher spatial resolution could aid in characterizing the geologic settings of potential hot springs on Mars. Locating emissivity minima in the infrared regions of silica that do not shift to a lower position would suggest the potential for well‐preserved microbial structures in Martian sinters, if life ever did exist there.

Abstract:Few traces of Earth’s geologic record are preserved from the time of life’s emergence, over 3,800 million years ago. Consequently, what little we understand about abiogenesis — the origin of life on Earth — is based primarily on laboratory experiments and theory. The best geological lens for understanding early Earth might actually come from Mars, a planet with a crust that’s overall far more ancient than our own. On Earth, surface sedimentary environments are thought to best preserve evidence of ancient life, but this is mostly because our planet has been dominated by high photosynthetic biomass production at the surface for the last ~2,500 million years or more. By the time oxygenic photosynthesis evolved on Earth, Mars had been a hyperarid, frozen desert with a surface bombarded by high-energy solar and cosmic radiation for more than a billion years, and as a result, photosynthetic surface life may never have occurred on Mars. Therefore, one must question whether searching for evidence of life in Martian surface sediments is the best strategy. This Perspective explores the possibility that the abundant hydrothermal environments on Mars might provide more valuable insights into life’s origins.

Abstract:Spectroscopic technologies are prominent tools for the detection of potential extraterrestrial chemical biomarkers, such as those on Mars. However, the biomass in Martian samples, if existed, is expected to be extremely low, so it is crucial to understand the low boundary of spectroscopic detections. In this study, a Martian basalts-like igneous rock and four representative bacterial strains were used to examine the biomarker detection potentials of the laser-induced breakdown (LIBS), Raman, infrared (IR), and X-ray spectroscopies. A series of concentration gradients of these strains were prepared and dropped onto the basaltic sample, the areas of which were then analyzed by spectrometers. The multivariate analysis of LIBS data can differentiate areas with several to tens of cells per cm2 from basaltic components. In addition, Raman and Fourier-transform IR (FTIR) spectrometers are robust to detecting organic chemical bonds or biological materials when cells are directly targeted. If more water-rich conditions are present, LIBS, Raman, and FTIR spectrometers were all able to acquire this more habitable information. Moreover, transmission electron microscopy (TEM) and affiliated energy dispersive X-ray spectroscopy (EDS) can examine the microscale shapes and chemical distribution within suspicious compartment structures. These findings imply that LIBS-associated statistics and suites of spectrometers can be promising tools for in situ life detection on Mars or returned sample analyses in laboratories if biotic materials were present in a small measured region even at low biomass levels.

Abstract:Abstract The Mars 2020 Perseverance rover has examined and sampled sulfate‐rich clastic rocks from the Hogwallow Flats member at Hawksbill Gap and the Yori Pass member at Cape Nukshak. Both strata are located on the Jezero crater western fan front, are lithologically and stratigraphically similar, and have been assigned to the Shenandoah formation. In situ analyses demonstrate that these are fine‐grained sandstones composed of phyllosilicates, hematite, Ca‐sulfates, Fe‐Mg‐sulfates, ferric sulfates, and possibly chloride salts. Sulfate minerals are found both as depositional grains and diagenetic features, including intergranular cement and vein‐ and vug‐cements. Here, we describe the possibility of various sulfate phases to preserve potential biosignatures and the record of paleoenvironmental conditions in fluid and solid inclusions, based on findings from analog sulfate‐rich rocks on Earth. The samples collected from these outcrops, Hazeltop and Bearwallow from Hogwallow Flats, and Kukaklek from Yori Pass, should be examined for such potential biosignatures and environmental indicators upon return to Earth.

Abstract:The search for organic matter on Mars is one of the major objectives of Mars exploration. However, limited detection of organic signals by Mars rovers to date demands further investigation on this topic. The Curiosity rover recently discovered numerous nodules in Gale Crater on Mars. These nodules have been considered to precipitate in the neutral-to-alkaline and saline diagenetic fluids and could be beneficial for organic preservation. Here, we examine this possibility by studying the carbonate nodules in the western Qaidam Basin, NW China, one of the terrestrial analog sites for Mars. Fourier transform infrared spectra of the carbonate nodules reveal that the aliphatic and aromatic molecules can be readily preserved inside nodules in Mars-like environments. The chain-branching index of the Qaidam nodules suggests that the diagenetic fluids where nodules precipitated were able to support diverse microbial communities that could vary with the water salinity. Findings of this study provide new perspectives on the astrobiological significance of nodules in Gale Crater and the further detection of organic matter on Mars.

Abstract:Twelve hair dye formulations were tested for systemic toxicity by topical application twice weekly for 13 wk to groups of 12 New Zealand white rabbits and for teratologic effects following applications to groups of 20 pregnant Charles River CD rats on days 1, 4, 7, 10, 13, 16, and 19 of gestation. The three semipermanent formulations were applied as is, and the nine oxidation dyes were mixed 1:1 with 6% hydrogen peroxide just prior to application, as in normal use. The formulations induced a broad spectrum of dyes and dye intermediates used or considered useful in oxidative and semipermanent hair color products. In the teratology study no biologically significant soft tissue or skeletal changes were noted. Similarly, the mean numbers of corpora lutea, implantation sites, live fetuses, and resorptions per pregnancy, as well as numbers of litters with resporptions, were not significantly affected by the dye treatment. In the percutaneous toxicity study there was no evidence of compound-induced systemic effects. Microscopic examination of 25 tissues from each animal gave no indication of histomorphologic evidence of toxicity. No dye discoloration of urine was seen at any time during the test or at necropsy. Some of the dye groups showed epidermal hyperplasia, which was probably a reflection of slight irritation due to the frequency of application of the oxidation formulations.

Abstract:We shortly discuss the observable consequences of the two hypotheses about the origin of life on Earth and Mars: the Lithopanspermia (Mars to Earth or viceversa) and the origin from a unique progenitor, that for Earth is called LUCA (the LUCA hypothesis). To test the possibility that some lifeforms similar to the terrestrial ones may survive on Mars, we designed and built two simulators of Martian environments where to perform experiments with different bacterial strains: LISA and mini-LISA. Our LISA environmental chambers can reproduce the conditions of many Martian locations near the surface trough changes of temperature, pressure, UV fluence and atmospheric composition. Both simulators are open to collaboration with other laboratories interested in performing experiments on many kind of samples (biological, minerals, electronic) in situations similar to that of the red planet. Inside LISA we have studied the survival of several bacterial strains and endospores. We verified that the UV light is the major responsible of cell death. Neither the low temperature, nor the pressure, nor the desiccation or the atmospheric changes were effective in this sense. We found that some Bacillus strains have a particular capability to survive for some hours in Martian conditions without being screened by dust or other shields. We also simulated the coverage happening on a planet by dust transported by the winds, blowing on the samples a very small quantity of volcanic ash grains or red iron oxide particles. Samples covered by these dust grains have shown a high percentage of survival, indicating that under the surface dust, if life were to be present on Mars in the past, some bacteria colonies or cells could still be present.

Abstract:To search more efficiently for a record of past life on Mars, it is critical to know where to look and thus maximize the likelihood of success. Large-scale site selection for the Mars 2020 mission has been completed, but small (meter to 10 cm)-scale relationships of microenvironments will not be known until the rover reaches the surface. Over a 2 m transect at a modern volcanic deposit on the flank of Askja volcano in the barren highlands of Iceland, we compared two biological indicators (ATP activity and 16SrRNA amplicon sequence composition) to physical characteristics including bulk chemical composition, spectral signatures of mineralogy, and grain size. Using analytical instrumentation analogous to those available on Mars rovers, we were able to characterize the geological setting of the deposits and link physical parameters to microbial abundance and diversity. In general, methanogenesis, methanotrophy/methylotrophy, and nitrate reduction were the functional traits most associated with microbial community shift along the transect. Core microbiome members tended to be associated with nitrate reduction, and relative abundance of core microbes was strongly related to free water in the deposit. Community compositional shift of the rare microbiome was related to microenvironmental changes such as change in grain size, geochemistry, and age of deposit. These correlations lead us to suggest a sampling strategy that accounts for Martian geology, looking for undisturbed (not remobilized) explosive volcanic ash below pumice that could maximize diversity and abundance of different bioindicators. Our study also illustrates the importance of studying the variability across microenvironments in low biomass settings on earth.

Abstract:Here we review published studies on the abundance and diversity of terrestrial rock-hosted life, the environments it inhabits, the evolution of its metabolisms, and its fossil biomarkers to provide guidance in the search for life on Mars. Key findings are (1) much terrestrial deep subsurface metabolic activity relies on abiotic energy-yielding fluxes and in situ abiotic and biotic recycling of metabolic waste products rather than on buried organic products of photosynthesis; (2) subsurface microbial cell concentrations are highest at interfaces with pronounced chemical redox gradients or permeability variations and do not correlate with bulk host rock organic carbon; (3) metabolic pathways for chemolithoautotrophic microorganisms evolved earlier in Earth's history than those of surface-dwelling phototrophic microorganisms; (4) the emergence of the former occurred at a time when Mars was habitable, whereas the emergence of the latter occurred at a time when the martian surface was not continually habitable; (5) the terrestrial rock record has biomarkers of subsurface life at least back hundreds of millions of years and likely to 3.45 Ga with several examples of excellent preservation in rock types that are quite different from those preserving the photosphere-supported biosphere. These findings suggest that rock-hosted life would have been more likely to emerge and be preserved in a martian context. Consequently, we outline a Mars exploration strategy that targets subsurface life and scales spatially, focusing initially on identifying rocks with evidence for groundwater flow and low-temperature mineralization, then identifying redox and permeability interfaces preserved within rock outcrops, and finally focusing on finding minerals associated with redox reactions and associated traces of carbon and diagnostic chemical and isotopic biosignatures. Using this strategy on Earth yields ancient rock-hosted life, preserved in the fossil record and confirmable via a suite of morphologic, organic, mineralogical, and isotopic fingerprints at micrometer scale. We expect an emphasis on rock-hosted life and this scale-dependent strategy to be crucial in the search for life on Mars.

Abstract:The second ExoMars mission will be launched in 2020 to target an ancient location interpreted to have strong potential for past habitability and for preserving physical and chemical biosignatures (as well as abiotic/prebiotic organics). The mission will deliver a lander with instruments for atmospheric and geophysical investigations and a rover tasked with searching for signs of extinct life. The ExoMars rover will be equipped with a drill to collect material from outcrops and at depth down to 2 m. This subsurface sampling capability will provide the best chance yet to gain access to chemical biosignatures. Using the powerful Pasteur payload instruments, the ExoMars science team will conduct a holistic search for traces of life and seek corroborating geological context information. Abstract:There is growing evidence suggesting the presence of aqueous environment on ancient Mars, raising the question of the possibility of life in such an environment. Subsequently, with the erosion of the Martian atmosphere resulting in drastic changes in its climate, surface water disappeared, shrinking habitable spaces on the planet, with only a limited amount of water remaining near the surface in form of brines and water-ice deposits. Life, if it ever existed, would have had to adapt to harsh modern conditions, which includes low temperatures and surface pressure, and high radiation dose. Presently, there is no evidence of any biological activity on the planet's surface, however, the subsurface environment, which is yet to be explored, is less harsh, has traces of water in form of water-ice and brines, and undergoes radiation-driven redox chemistry. I hypothesize that Galactic Cosmic Ray (GCR)-induced radiation-driven chemical disequilibrium can be used for metabolic energy by extant life, and host organisms using mechanisms seen in similar chemical and radiation environments on Earth. I propose a GCR-induced radiolytic zone, and discuss the prospects of finding such life with Rosalind Franklin rover of the ExoMars mission.

Abstract:Over 20% of organic carbon in sediments on Earth are bound to reactive Fe mineral phases [1]. These reactive Fe phases are generally Fe (oxyhydr)oxides, often associated with clay minerals. It is important to note that they occur as nanoparticulate and X-ray amorphous phases that are challenging to identify. On Earth, proxy methods such as chemical sequential extractions are often used but they can produce misleading results when used for mineral identification [2,3]. We develop and use M&#246;ssbauer spectroscopy applications to identify these phase [2-4] and compare these to Raman spectroscopy because the Mars 2020 Perseverance rover and the ExoMars 2022 Rosalind Franklin rover use Raman spectrometers for mineralogical identification. Reactive Fe phases are abundant on Mars. It is important to note that they are not the well-crystalline expression of Fe (oxyhydr)oxides such as hematite and goethite that have been observed from orbit and with a variety of rover-based instruments. Instead, reactive Fe phases are represented by as yet unidentified Fe phases: Aqueously altered rocks and soils in Gusev crater and at Meridiani Planum (including the Burns formation) contain large amounts of nanophase iron oxides (npOx and Fe3D3) [5]; and 20-60 wt% of minerals in fluvio-lacustrine deposits in Gale crater are X-ray amorphous and this amorphous phase is rich in iron [6]. Mineralogical interpretation of CRISM data of Rosalind Franklin's landing site at Oxia Planum also suggest the presence of these phase. These reactive Fe phases can be any combination of a number of minerals including ferrihydrite, lepidocrocite, akagan&#232;ite, hissingerite, schwertmannite, and superparamagnetic (i.e. nanoparticulate) hematite and goethite [5]. The preservation of organic compounds by reactive Fe species is effective over hundreds of thousands of years in Earth sediments [1]. In return, organic compounds slow down the transformation of reactive Fe species such as ferrihydrite into the more crystalline and thermodynamically stable Fe (oxyhydr)oxides hematite or goethite during diagenetic processes. With temperature and pressure rising further during diagenesis, however, organic compounds are oxidized and destroyed through the reduction of Fe (resulting in the diagenetic formation of the Fe carbonate siderite, for example), and the non-reduced Fe species are transformed into thermodynamically stable minerals. Thus, the presence of reactive Fe species in Martian sediments/sedimentary rocks indicates only little diagenetic overprinting and therefore a high preservation potential of organic compounds. Such samples will be of high priority for analysis with MOMA. However, the presence of Fe species during pyrolysis can reduce the detectability of certain organic compounds. This effect depends on the specific Fe species present and is mitigated in the presence of clay minerals [7,8].
