Abstract:Background L-(+)-tartaric acid (L-TA) is an important organic acid, which is produced from the cream of tartar or stereospecific hydrolysis of the cis -epoxysuccinate. The former method is limited by the availability of raw material and the latter is dependent on the petrochemical material. Thus, new processes for the economical preparation of L-TA from carbohydrate or renewable resource would be much more attractive. Production of 5-keto-D-gluconate (5-KGA) from glucose by Gluconobacter oxydans is the first step to produce L-TA. The aim of this work is to enhance 5-KGA accumulation using combinatorial metabolic engineering strategies in G. oxydans . The sldAB gene, encoding sorbitol dehydrogenase, was overexpressed in an industrial strain G. oxydans ZJU2 under a carefully selected promoter, P 0169 . To enhance the efficiency of the oxidation by sldAB , the coenzyme pyrroloquinoline quinone (PQQ) and respiratory chain were engineered. Besides, the role in sldAB overexpression, coenzyme and respiratory chain engineering and their subsequent effects on 5-KGA production were investigated. Results An efficient, stable recombinant strain was constructed, whereas the 5-KGA production could be enhanced. By self-overexpressing the sldAB gene in G. oxydans ZJU2 under the constitutive promoter P 0169 , the resulting strain, G. oxydans ZJU3, produced 122.48 ± 0.41 g/L of 5-KGA. Furthermore, through the coenzyme and respiratory chain engineering, the titer and productivity of 5-KGA reached 144.52 ± 2.94 g/L and 2.26 g/(L · h), respectively, in a 15 L fermenter. It could be further improved the 5-KGA titer by 12.10 % through the fed-batch fermentation under the pH shift and dissolved oxygen tension (DOT) control condition, obtained 162 ± 2.12 g/L with the productivity of 2.53 g/(L · h) within 64 h. Conclusions The 5-KGA production could be significantly enhanced with the combinatorial metabolic engineering strategy in Gluconobacter strain, including sldAB overexpression, coenzyme and respiratory chain engineering. Fed-batch fermentation could further enlarge the positive effect and increase the 5-KGA production. All of these demonstrated that the robust recombinant strain can efficiently produce 5-KGA in larger scale to fulfill the industrial production of L-TA from 5-KGA.
Engineering Escherichia coli for l-Threonine Hyperproduction Based on Multidimensional Optimization Strategies

Abstract:Exploring effective remodeling strategies to further improve the productivity of high-yield strains is the goal of biomanufacturing. However, the lack of insight into host-specific metabolic networks prevents timely identification of useful engineering targets. Here, multidimensional engineering strategies were implemented to optimize the global metabolic network for improving l-threonine production. First, the metabolic bottleneck for l-threonine synthesis was eliminated by synergistic utilization of NADH and an enhanced ATP supply. Carbon fluxes were redistributed into the TCA cycle by rationally regulating the GltA activity. Subsequently, the stress global response regulator UspA was identified to enhance l-threonine production by a transcriptomic analysis. Then, l-threonine productivity was improved by enhancing the host's stress resistance and releasing the inhibitory reaction of glucose utilization. Eventually, the l-threonine yield of THRH16 reached 170.3 g/L and 3.78 g/L/h in a 5 L bioreactor, which is the highest production index reported. This study provides rational guidance for increasing the productivity of other chemicals.

Abstract:The market of L-phenylalanine has been stimulated by the great demand for the low-calorie sweetener aspartame. In this paper, the effects of pivotal genes on L-phenylalanine production were evaluated by metabolic engineering of wild type Escherichia coli. The bifunctional PheA protein contains two catalytic domains (chorismate mutase and prephenate dehydratase activities) as well as one R-domain (for feedback inhibition by L-phenylalanine). The catalytic domain of PheA was overexpressed to increase L-phenylalanine production. It was firstly indicated that this domain could enhance the metabolic influx to overproduce L-phenylalanine and improve the survival ability under m-Fluoro-DL-phenylalanine stress. Furthermore, the fermentation performance of aroG feedback inhibition resistant mutants was firstly compared, aroG29 and aroG15 increased the L-phenylalanine concentration by 5-fold. After that the expression of aroK and ydiB was also elevated, and the L-phenylalanine yield on cell (0.79 g/g) and maximum L-phenylalanine productivity (0.073 g/L/h) were subsequently doubled. Meanwhile, the L-phenylalanine yield on glucose increased from 0.124 g/g to 0.153 g/g. It was found that genes ydiB and aroK could elevate the L-phenylalanine yield and productivity and shorten the lag phase. (C) 2013 Elsevier Ltd. All rights reserved.
Expression regulation of multiple key genes to improve l-threonine in Escherichia coli

Abstract:Abstract Background Escherichia coli is an important strain for l -threonine production. Genetic switch is a ubiquitous regulatory tool for gene expression in prokaryotic cells. To sense and regulate intracellular or extracellular chemicals, bacteria evolve a variety of transcription factors. The key enzymes required for l -threonine biosynthesis in E. coli are encoded by the thr operon. The thr operon could coordinate expression of these genes when l -threonine is in short supply in the cell. Results The thrL leader regulatory elements were applied to regulate the expression of genes iclR , arcA , cpxR , gadE , fadR and pykF , while the threonine-activating promoters P cysH , P cysJ and P cysD were applied to regulate the expression of gene aspC , resulting in the increase of l -threonine production in an l -threonine producing E. coli strain TWF001. Firstly, different parts of the regulator thrL were inserted in the iclR regulator region in TWF001, and the best resulting strain TWF063 produced 16.34 g l -threonine from 40 g glucose after 30 h cultivation. Secondly, the gene aspC following different threonine-activating promoters was inserted into the chromosome of TWF063, and the best resulting strain TWF066 produced 17.56 g l -threonine from 40 g glucose after 30 h cultivation. Thirdly, the effect of expression regulation of arcA , cpxR , gadE , pykF and fadR was individually investigated on l -threonine production in TWF001. Finally, using TWF066 as the starting strain, the expression of genes arcA , cpxR , gadE , pykF and fadR was regulated individually or in combination to obtain the best strain for l -threonine production. The resulting strain TWF083, in which the expression of seven genes ( iclR , aspC , arcA , cpxR , gadE , pykF , fadR and aspC ) was regulated, produced 18.76 g l -threonine from 30 g glucose, 26.50 g l -threonine from 40 g glucose, or 26.93 g l -threonine from 50 g glucose after 30 h cultivation. In 48 h fed-batch fermentation, TWF083 could produce 116.62 g/L l ‐threonine with a yield of 0.486 g/g glucose and productivity of 2.43 g/L/h. Conclusion The genetic engineering through the expression regulation of key genes is a better strategy than simple deletion of these genes to improve l -threonine production in E. coli . This strategy has little effect on the intracellular metabolism in the early stage of the growth but could increase l -threonine biosynthesis in the late stage.

Abstract:The production of L-tryptophan was investigated in a recombinant strain Escherichia coli W3110-ZDrr. It was observed that phosphate and feeding strategies are key factors to ensure the good cell growth and high production of L-tryptophan. The simple exponential feeding strategy could only produce 10.6 g/L L-tryptophan due to the improper feeding rate; while the manual glucose-feedback feeding approach could effectively control the substrate and inhibit the formation of acetate, and thus improvemed the L-tryptophan production to 25.5 g/L. The modified exponential feeding approach avoided overfeeding or underfeeding and achieved high production of L-tryptophan. Moreover, the addition of Tween 60 or PL61 could enhance the cell growth and the production of L-tryptophan in the fed-batch mode. Subsequent metabolic flux analysis showed that more carbon flux was distributed into the biosynthesis of L-tryptophan when Tween 60 or PL61 was supplied. The present work presents one base for further large-scale production of this important amino acid.

Abstract:Development of microbial cell factory for L-tryptophan (L-trp) production has received widespread attention but still requires extensive efforts due to weak metabolic flux distribution and low yield. Here, the riboswitch-based high-throughput screening (HTS) platform was established to construct a powerful L-trp-producing chassis cell. To facilitate L-trp biosynthesis, gene expression was regulated by promoter and N-terminal coding sequences (NCS) engineering. Modules of degradation, transport and by-product synthesis related to L-trp production were also fine-tuned. Next, a novel transcription factor YihL was excavated to negatively regulate L-trp biosynthesis. Self-regulated promoter-mediated dynamic regulation of branch pathways was performed and cofactor supply was improved for further L-trp biosynthesis. Finally, without extra addition, the yield of strain Trp30 reached 42.5 g/L and 0.178 g/g glucose after 48 h of cultivation in 5-L bioreactor. Overall, strategies described here worked up a promising method combining HTS and multidimensional regulation for developing cell factories for products in interest.

Abstract:In this study, we constructed an l-methionine-producing recombinant strain from wild-type Escherichia coli W3110 by metabolic engineering. To enhance the carbon flux to methionine and derepression met regulon, thrBC, lysA, and metJ were deleted in turn. Methionine biosynthesis obstacles were overcome by overexpression of metA (Fbr) (Fbr, Feedback resistance), metB, and malY under control of promoter pN25. Recombinant strain growth and methionine production were further improved by attenuation of metK gene expression through replacing native promoter by metK84p. Blocking the threonine pathway by deletion of thrBC or thrC was compared. Deletion of thrC showed faster growth rate and higher methionine production. Finally, metE, metF, and metH were overexpressed to enhance methylation efficiency. Compared with the original strain E. coli W3110, the finally obtained Me05 (pETMA(Fbr)-B-Y/pKKmetH) improved methionine production from 0 to 0.65 and 5.62 g/L in a flask and a 15-L fermenter, respectively.

Abstract:l -alanine is a critical natural amino acid that is widely used in the food, pharmaceutical, and personal care industries. However, low titer and long fermentation period limits the industrial viability of l -alanine production. In this study, we engineered a microbial cell factory for efficient l -alanine synthesis using a variety of metabolic engineering strategies. Initially, l -alanine was biosynthesized through the heterologous introduction of alanine dehydrogenase from Geobacillus stearothermophilus . Thereafter, the l -alanine biosynthetic pathway and transport module were modified to improve its titers. Furthermore, a dynamic response system was constructed based on the l -alanine-driven P Ala-7 promoter to regulate the tolerance target BetT expression and enhance the tolerance of Escherichia coli to high l -alanine concentrations. Finally, the resultant strain E. coli W-P Ala-7 -BetT could produce 134.5 g/L of l -alanine in 24 h in a 5-L bioreactor, with 5.6 g/L/h of productivity, which is the highest productivity reported to date. Our findings may help in constructing high-performance microbial cell factories for the production of l -alanine and other valuable chemicals.

Abstract:2,5-Dimethylpyrazine (2,5-DMP) is a high-value-added alkylpyrazine compound with important applications in both the food and pharmaceutical fields. In response to the increasing consumer preference for natural products over chemically synthesized ones, efforts have been made to develop efficient microbial cell factories for the production of 2,5-DMP. However, the previously reported recombinant strains have exhibited low yields and relied on expensive antibiotics and inducers. In this study, we employed metabolic engineering strategies to develop an Escherichia coli strain capable of producing 2,5-DMP at high levels without the need for inducers or antibiotics. Initially, the biosynthesis pathway of 2,5-DMP was constructed that realized 2,5-DMP production from glucose. Subsequently, efforts focused on enhancing 2,5-DMP production by improving the availability of the cofactor NAD+ and precursor l-threonine. Additionally, the supply and conversion of l-threonine were balanced by optimizing the copy number of the key gene tdh on the chromosome and by modifying the l-threonine transport system. The final engineering strain D19 produced 3.1 g/L of 2,5-DMP, which is the highest titer for fermentative production of 2,5-DMP using glucose as the carbon source up to date. The strategies used in this study lay a good foundation for the production of 2,5-DMP on a large scale.

Systems metabolic engineering of Corynebacterium glutamicum for efficient L-tryptophan production

Abstract:Corynebacterium glutamicum is a versatile industrial microorganism for producing various amino acids. However, there have been no reports of a well-defined C. glutamicum strain capable of hyperproducing L-tryptophan. This study presents a comprehensive metabolic engineering approach to establish a robust C. glutamicum cell factory for L-tryptophan biosynthesis, including: (1) identification of potential targets using an enzyme-constrained genome-scale model; (2) targeted enhancement of the L-tryptophan biosynthetic pathway; (3) reconfiguration of central metabolic pathways; (4) identification of metabolic bottlenecks through comparative metabolome analysis; (5) engineering of the transport system, shikimate pathway, and precursor supply; and (6) repression of competing pathways and iterative optimization of key targets. The resulting engineered C. glutamicum strain achieved a remarkable L-tryptophan titer of 16.2 g/L with a yield of 0.16 g/g glucose in shake-flask fermentation. This study highlights the efficacy of integrating computational modeling with systems metabolic engineering for significantly enhancing the production capabilities of industrial microorganisms.

Abstract:l -Threonine is an important feed additive with the third largest market size among the amino acids produced by microbial fermentation. The GRAS (generally regarded as safe) industrial workhorse Corynebacterium glutamicum is an attractive chassis for l -threonine production. However, the present l -threonine production in C. glutamicum cannot meet the requirement of industrialization due to the relatively low production level of l -threonine and the accumulation of large amounts of by-products (such as l -lysine, l -isoleucine, and glycine). Herein, to enhance the l -threonine biosynthesis in C. glutamicum , releasing the aspartate kinase (LysC) and homoserine dehydrogenase (Hom) from feedback inhibition by l -lysine and l -threonine, respectively, and overexpressing four flux-control genes were performed. Next, to reduce the formation of by-products l -lysine and l -isoleucine without the cause of an auxotrophic phenotype, the feedback regulation of dihydrodipicolinate synthase (DapA) and threonine dehydratase (IlvA) was strengthened by replacing the native enzymes with heterologous analogues with more sensitive feedback inhibition by l -lysine and l -isoleucine, respectively. The resulting strain maintained the capability of synthesizing enough amounts of l -lysine and l -isoleucine for cell biomass formation but exhibited almost no extracellular accumulation of these two amino acids. To further enhance l -threonine production and reduce the by-product glycine, l -threonine exporter and homoserine kinase were overexpressed. Finally, the rationally engineered non-auxotrophic strain ZcglT9 produced 67.63 g/L (17.2% higher) l -threonine with a productivity of 1.20 g/L/h (108.0% higher) in fed-batch fermentation, along with significantly reduced by-product accumulation, representing the record for l -threonine production in C. glutamicum . In this study, we developed a strategy of reconstructing the feedback regulation of amino acid metabolism and successfully applied this strategy to de novo construct a non-auxotrophic l -threonine producing C. glutamicum . The main end by-products including l -lysine, l -isoleucine, and glycine were almost eliminated in fed-batch fermentation of the engineered C. glutamicum strain. This strategy can also be used for engineering producing strains for other amino acids and derivatives.

Integrated laboratory evolution and rational engineering of GalP/Glk-dependent Escherichia coli for higher yield and productivity of L-tryptophan biosynthesis

Abstract:L-Tryptophan (Trp) is a high-value aromatic amino acid with diverse applications in food and pharmaceutical industries. Although production of Trp by engineered Escherichia coli has been extensively studied, the need of multiple precursors for its synthesis and the complex regulations of the biosynthetic pathways make the achievement of a high product yield still very challenging. Metabolic flux analysis suggests that the use of a phosphoenolpyruvate:sugar phosphotransferase system (PTS) independent glucose uptake system, i.e. the galactose permease/glucokinase (GalP/Glk) system, can theoretically double the Trp yield from glucose. To explore this possibility, a PTS- and GalP/Glk-dependent E. coli strain was constructed from a previously rationally developed Trp producer strain S028. However, the growth rate of the S028 mutant was severely impaired. To overcome this problem, promoter screening for modulated gene expression of GalP/Glk was carried out, following by a batch mode of adaptive laboratory evolution (ALE) which resulted in a strain K3 with a similar Trp yield and concentration as S028. In order to obtain a more efficient Trp producer, a novel continuous ALE system was developed by combining CRISPR/Cas9-facilitated in vivo mutagenesis with real-time measurement of cell growth and online monitoring of Trp-mediated fluorescence intensity. With the aid of this automatic system (auto-CGSS), a promising strain T5 was obtained and fed-batch fermentations showed an increase of Trp yield by 19.71% with this strain compared with that obtained by the strain K3 (0.164 vs. 0.137 g/g). At the same time, the specific production rate was increased by 52.93% (25.28 vs. 16.53 mg/g DCW/h). Two previously engineered enzyme variants AroGD6G-D7A and AnTrpCR378F were integrated into the strain T5, resulting in a highly productive strain T5AA with a Trp yield of 0.195 g/g and a specific production rate of 28.83 mg/g DCW/h.

Abstract:Stocks are short-sale constrained when there is a strong demand to sell short and a limited supply of shares to borrow. Using data on both short interest (a proxy for demand) and institutional ownership (a proxy for supply) we find that constrained stocks underperform during the period 1988–2002 by a significant 215 basis points per month on an equally weighted basis, although by only an insignificant 39 basis points per month on a valueweighted basis. For the overwhelming majority of stocks, short interest and institutional ownershiplevels make short selling constraints unlikely.

Abstract:Background: L-phenylalanine (L-Phe) is an essential amino acid for mammals and applications expand into human health and nutritional products. In this study, a system level engineering was conducted to enhance L-Phe biosynthesis in Escherichia coli. Results: We inactivated the PTS system and recruited glucose uptake via combinatorial modulation of galP and glk to increase PEP supply in the Xllp01 strain. In addition, the HTH domain of the transcription factor TyrR was engineered to decrease the repression on the transcriptional levels of L-Phe pathway enzymes. Finally, proteomics analysis demonstrated the third step of the SHIK pathway (catalyzed via AroD) as the rate-limiting step for L-Phe production. After optimization of the aroD promoter strength, the titer of L-Phe increased by 13.3%. Analysis of the transcriptional level of genes involved in the central metabolic pathways and L-Phe biosynthesis via RT-PCR showed that the recombinant L-Phe producer exhibited a great capability in the glucose utilization and precursor (PEP and E4P) generation. Via systems level engineering, the L-Phe titer of Xllp21 strain reached 72.9 g/L in a 5 L fermenter under the non-optimized fermentation conditions, which was 1.62-times that of the original strain Xllp01. Conclusion: The metabolic engineering strategy reported here can be broadly employed for developing genetically defined organisms for the efficient production of other aromatic amino acids and derived compounds.
Flux redistribution of central carbon metabolism for efficient production of l ‐tryptophan in Escherichia coli

Abstract:Microbial production of L‐tryptophan has received considerable attention because of its diverse applications in food additives and pharmaceuticals Overexpression of rate‐limiting enzymes and blockage of competing pathways can effectively promote microbial production of L‐tryptophan. However, the biosynthetic process remains suboptimal due to imbalanced flux distribution between central carbon and tryptophan metabolism, presenting a major challenge to further improvement of L‐tryptophan yield. In this study, we redistributed central carbon metabolism to improve phosphoenolpyruvate (PEP) and erythrose‐4‐phosphate (E4P) pools in an L‐tryptophan producing strain of E. coli for efficient L‐tryptophan synthesis. To do this, a phosphoketolase from Bifidobacterium adolescentis was introduced to strengthen E4P formation, and the L‐tryptophan titer and yield increased to 10.8 g/L and 0.148 g/g glucose, respectively. Next, the phosphotransferase system was substituted with PEP‐independent glucose transport, meditated by a glucose facilitator from Zymomonas mobilis and native glucokinase. This modification improved L‐tryptophan yield to 0.164 g/g glucose, concomitant with 58% and 40% decreases of acetate and lactate accumulation, respectively. Then, to channel more central carbon flux to tryptophan biosynthetic pathway, several metabolic engineering strategies were applied to rewire the PEP‐pyruvate‐oxaloacetate node. Finally, the constructed strain SX11 produced 41.7 g/L L‐tryptophan with an overall yield of 0.227 g/g glucose after 40 h fed‐batch fermentation in 5‐L bioreactor. This is the highest overall yield of L‐tryptophan ever reported from a rational engineered strain. Our results suggest the flux redistribution of central carbon metabolism to maintain sufficient supply of PEP and E4P is a promising strategy for efficient L‐tryptophan biosynthesis, and this strategy would likely also increase production of other aromatic amino acids and derivatives.This article is protected by copyright. Biomechanical and histomorphometric characterizations of osseointegration during mini-screw healing in rabbit tibiae.

Abstract:OBJECTIVE To evaluate effects of different healing times on integration of titanium mini-screws and bones under unloaded conditions. MATERIALS AND METHODS Biomechanical stability measurements and histomorphometric observations were performed in a rabbit tibia model after different healing times: 0 (immediate) and 1, 2, 4, and 8 weeks. RESULTS Biomechanical stability and both maximum torque and maximum pullout load increased with healing time but increased significantly only after 4 weeks. Maximum torque and maximum pullout load both significantly correlated with healing time and with each other. Similarly, histomorphometric analyses showed that new bone formation increased with healing time but increased dramatically only after 4 weeks. The data may provide guidance for determining optimal implant plans in clinical practice. CONCLUSION Mini-screw healing is a continuous process, and week 4 is a critical time point in this process.
Spatiotemporal Gene Expression by a Genetic Circuit for Chemical Production in Escherichia coli

Abstract:Gene expression in spatiotemporal distribution improves the ability of cells to respond to changing environments. For microbial cell factories in artificial environments, reconstruction of the target compound's biosynthetic pathway in a new spatiotemporal dimension/scale promotes the production of chemicals. Here, a genetic circuit based on the Esa quorum sensing and lac operon was designed to achieve the dynamic temporal gene expression. Meanwhile, the pathway was regulated by an l-cysteine-specific sensor and relocalized to the plasma membrane for further flux enhancement to l-cysteine and toxicity reduction on a spatial scale. Finally, the integrated spatiotemporal regulation circuit for l-cysteine biosynthesis enabled a 14.16 g/L l-cysteine yield in Escherichia coli. Furthermore, this spatiotemporal regulation circuit was also applied in our previously constructed engineered strain for pantothenic acid, methionine, homoserine, and 2-aminobutyric acid production, and the titer increased by 29, 33, 28, and 41%, respectively. These results highlighted the applicability of our spatiotemporal regulation circuit to enhance the performance of microbial cell factories.
