Abstract:Oil sands process-affected water (OSPW), generated by surface mining in Canada’s oil sands, require treatment of environmentally persistent dissolved organic compounds before release to the watershed. Conventional chemical and mechanical treatments have not proved suitable for treating the large quantities of stored OSPW, and the biological recalcitrance of some dissolved organics may not be adequately addressed by conventional passive treatment systems. Previous work has evaluated photocatalytic treatment as a passive advanced oxidation process (P-AOP) for OSPW remediation. This work expands upon this prior research to further characterize the effects of water chemistry on the treatment rate and detoxification threshold. Under artificial sunlight, buoyant photocatalysts (BPCs) detoxified all OSPW samples within 1 week of treatment time with simultaneous treatment of polycyclic aromatic hydrocarbons, naphthenic acid fraction components (NAFCs), and un-ionized ammonia. Overall, these results further demonstrate passive photocatalysis as an effective method for treatment of OSPW contaminants of potential concern (COPCs).

Dual Recognition of Lipid A and DNA by Human Antibodies Encoded by the VH4‐21 Gene A Possible Link between Infection and Lupus a

Abstract:The VH4-21 (V4-34) gene segment, a member of the VH4 family, is expressed early in B-cell maturation and is utilized by approximately 6% of normal adult B lymphocytes. This prevalence indicates an importance of VH4-21 in the B-cell repertoire. The gene also encodes certain autoantibodies being mandatory for pathological IgM anti-red cell antibodies directed against the I/i antigen, and also capable of encoding anti-DNA antibodies. Recognition of I/i antigen or DNA appears to be via two distinct sites on VH, with I/i binding mediated by sequences in the framework region, and DNA binding correlating with the presence of positively charged amino acids in complementarity-determining region 3. However, these positively charged residues appear to suppress the ability of the framework region to interact with I/i, rendering a single sequence monospecific for I/i or DNA. The IgM anti-DNA antibodies also recognize bacterial lipid A, whereas the anti-I/i antibodies do not, indicating that CDR3 may be involved in binding the negatively charged lipid A. Structural similarities between the DNA backbone and lipid A provide a possible explanation for this cross-reactivity. This dual recognition of bacterial antigen and autoantigen provides a potential link between infection and autoimmunity.
Naphthenic acid anaerobic biodegrading consortia enriched from pristine sediments underlying oil sands tailings ponds

Abstract:Seepage from oil sands tailings ponds (OSTP), which contain toxic naphthenic acids (NAs), can infiltrate into groundwater. Clay sediment layer beneath is a critical barrier for reducing the infiltration of NAs into the sand sediment layer, where groundwater channels reside. Biodegradation has great potential as a strategy for NAs removal, but little is known about NAs biodegradability and potential functional microbes in these pristine sediments. This study investigated the potential for anaerobic biodegradation of NAs by microbial consortia enriched from clay and sand sediments underlying OSTP, amended with either acid extracted organics or Merichem NAs, under nitrate- and sulfate-reducing conditions. Degradation of NAs only be detected after DOC concentration reached to steady state after 163 days. Microbial community analysis shows that different electron acceptors, sediment types, and NAs sources associated with specific microbial taxa and can explain 14.8, 13.9 % and 5% of variation of microbial community structures, respectively. The DOC and methane were the most important geochemical properties for microbial community variations. This study approved the potential capability of indigenous microbial communities from the pristine sediments in NA degradation, demonstrating the barrier function of pristine clay sediments underlying OSTP in prohibiting organic contaminants from entering into groundwater.

Treatment of naphthenic acids in oil sands process-affected waters with a surface flow treatment wetland: mass removal, half-life, and toxicity-reduction

Abstract:This study is the first to investigate the removal of naphthenic acids in a full-scale constructed wetland within the Alberta Oil Sands region. The average mass-removal efficiency for all O2-naphthenic acids measured in three separate deployments in the wetland ranged from 7.5% to 68.9% and appeared sensitive to physicochemical properties of the naphthenic acids, environmental conditions, and water quality. Treatment efficiency of individual naphthenic acids was found to increase with increasing carbon number and decreasing number of double bond equivalents in the molecule. Treatment efficiency was also found to increase with both higher initial turbidity in OSPW entering the wetland, and warmer average OSPW temperatures during wetland operation. Half-life times of naphthenic acids in the treatment wetland ranged between 8.9 and 39 days and were substantially lower than those in tailings ponds (i.e., 12.9-13.6 years) and laboratory studies focussed on bench-scale aerobic microbial biodegradation (i.e., 44-315 days). Using published dose-response data, biomimetic extraction measurements using solid phase microextraction fibers indicate that 14 days of wetland treatment resulted in a reduction in (4 d) deformity of Danio rerio from 50 to 16%, while exhibiting less than 1% toxic response for less sensitive toxic endpoints. The study concludes that wetland treatment is a feasible and productive treatment method for naphthenic acids in oil sands process-affected water due to a combination of sorption and biodegradation.

Abstract:This study investigated for the first time the synergistic effect of coupling persulfate (PDS) and peroxymonosulfate (PMS) with UVA LED light for the treatment of real oil sands process water (OSPW). The performance of the UVA LED/PDS and UVA LED/PMS processes was studied at different oxidant concentrations to remove fluorophore organic compounds and naphthenic acids (NAs), commonly considered significant contributors to the toxicity of raw OSPW. Both systems were highly efficient in eliminating fluorophore organic contaminants and classical NAs (O 2 -NAs) from the matrix; however, concentrations of 1 and 2 mM of PDS and PMS, respectively, were the most suitable conditions to achieve one order of magnitude removal of O 2 -NAs (i.e., 90 %). In addition, oxidized NAs (O 3 - and O 4 -NAs) were moderately degraded under these conditions. Considering economic factors, the UVA LED/PDS (1 mM) system proved to be more cost-effective than the UVA LED/PMS (2 mM) process and a viable alternative to other advanced treatments for OSPW remediation. Moreover, the pre-existing toxic effect of raw OSPW was markedly mitigated after treatment with UVA LED/PDS (1 mM), reducing by 80 % the ability of the matrix to induce pro-inflammatory cytokines in human leukemic monocytic and without significantly affecting the viability of the Staphylococcus epidermidis bacteria. Specifically for the microbial viability assay, the exposure period of the microorganism was exceptionally more extended than those considered in standard bioassays, reinforcing the scientific relevance of the obtained results. Finally, these results suggest a high environmental compatibility of the treated OSPW, making its reuse in further applications safe.

Abstract:The Alberta oil sands contain one of the world's largest reserves of oil - over 169 billion barrels of bitumen are economically recoverable with current extraction technologies. Surface mining and subsequent hot water extraction of bitumen from the ore generates about nine cubic meters of raw tailings per cubic meter of oil. Oil sands facilities are required to operate under a policy of zero water discharge, resulting in ponds containing more than one billion cubic meters of tailings, a mixture of sand, fines and process-affected water. Process-affected water contains numerous organic compounds, including naphthenic acids (NAs), which have been identified as the primary source of acute toxicity of process-affected water. Developments in analytical techniques, aerobic biodegradability, and treatment via chemical oxidation (ozone) of NAs are reviewed. The field continues to be challenged by the lack of a cost-effective, accurate analytical technique for NAs or an understanding of all the organic constituents in process-affected water that may be contributing to observed toxicity and thus requiring treatment.

Abstract:Oil sands operations to recover bitumen in Alberta, Canada generate large volumes of contaminated water which are stored in tailings ponds. These ponds emit large quantities of intermediate / semi- volatile organic compounds (IVOC/SVOCs) which upon emission can react or partition to form secondary organic aerosols (SOAs), a significant issue concerning human and environmental health. These ponds are one of the largest sources of SOAs in North America, and the responsible VOCs (polycyclic aromatic hydrocarbons (PAHs), alkanes, single ring aromatics) are difficult to treat. Passive technologies that can reduce aqueous emissions and degrade volatiles in-situ are desired. A promising technology is buoyant photocatalysts (BPCs), which float on the surface of water bodies and absorb sunlight to oxidize contaminants. In this work, a glass microsphere TiO 2 -coated BPC is studied, which forms a semi-porous floating reactive barrier on the surface of water, intercepting VOC emissions and photocatalytically oxidizing them in water before they can be released to air. Based on a group of F2 – F3 hydrocarbons, 16 PAHs (C 10 – C 22 ) and 8 alkanes (C 11 -C 18 ), this BPC technology reduced 89% of these SOA precursor emissions while degrading 84% of the compounds in the system under 2 days of simulated sunlight. In a synthetic tailings pond environment, BPCs were demonstrated to block and treat VOCs of particular concern well (phenanthrene, pyrene, chrysene), while further preventing emissions from free phase hydrocarbons. In context of tailings pond SOA precursor emissions, application of this floating reactive barrier technology to all ponds could reduce the environmental impact of oil sands operations by preventing release of between 2400 to 11000 tonnes of SOAs per year. The proven ability to intercept and treat VOCs in water extends the potential of this technology past the oil sands industry, providing one of the first options for passive fugitive emission management for various industrial water bodies. Abstract:Naphthenic acids (NAs) present in oil sands process water (OSPW) have brought increasing environmental concerns due to their potential risk to aquatic and mammalian species. In this study, sustainable biochar zinc oxide (BC/ZnO) composites were synthesized using wood waste with different ZnO content and applied under simulated solar light for the photocatalytic degradation of different NAs for the first time. The best experimental conditions were determined as 0.5 g/L BC/30%ZnO and 4 h of simulated solar irradiation time, achieving 93.7 % degradation of cyclohexanecarboxylic acid (CHA) following the pseudo-first order kinetics. BC, with a porous structure and roughened surface, acted as an excellent platform for ZnO particles and an electron reservoir to inhibit the recombination of photogenerated electron-hole pairs. Hydroxyl radicals ( • OH) were identified to play the dominant role in CHA degradation, and the enhanced photocatalytic performance of the BC/30%ZnO composite was proved by more • OH species detected compared to synthesized ZnO. The composite showed good reusability and stability after 4 successive cycles of use, still achieving 92.9 % degradation of CHA. Moreover, this is the first study applying BC/30%ZnO composite for the simultaneous degradation of a complex mixture of eight NAs with significantly different chemical structures. The competition kinetics were observed among different NAs in the mixture, resulted in over 95 % degradation of total NAs after 6 h treatment. By applying wood waste-based BC/ZnO composite with excellent photocatalytic performance in combination with solar light as green energy source, this research highlights a promising sustainable approach for real OSPW remediation.

Abstract:Oil sands process-affected water (OSPW) is generated during extraction of bitumen in the surface-mining oil sands industry in Alberta, Canada, and is acutely and chronically toxic to aquatic organisms. It is known that dissolved organic compounds in OSPW are responsible for most toxic effects, but knowledge of the specific mechanism(s) of toxicity, is limited. Using bioassay-based effects-directed analysis, the dissolved organic fraction of OSPW has previously been fractionated, ultimately producing refined samples of dissolved organic chemicals in OSPW, each with distinct chemical profiles. Using the Escherichia coli K-12 strain MG1655 gene reporter live cell array, the present study investigated relationships between toxic potencies of each fraction, expression of genes and characterization of chemicals in each of five acutely toxic and one non-toxic extract of OSPW derived by use of effects-directed analysis. Effects on expressions of genes related to response to oxidative stress, protein stress and DNA damage were indicative of exposure to acutely toxic extracts of OSPW. Additionally, six genes were uniquely responsive to acutely toxic extracts of OSPW. Evidence presented supports a role for sulphur- and nitrogen-containing chemical classes in the toxicity of extracts of OSPW.
Low adsorption affinity of athabasca oil sands naphthenic acid fraction compounds to a peat-mineral mixture

Abstract:Much of the toxicity in oil sands process-affected water in Athabasca oil sands tailings has been attributed to naphthenic acids (NAs) and associated naphthenic acid fraction compounds (NAFCs). Previous work has characterized the environmental behaviour and fate of these compounds, particularly in the context of constructed treatment wetlands. There is evidence that wetlands can attenuate NAFCs in natural and engineered contexts, but relative contributions of chemical, biotic, and physical adsorption with sequestration require deconvolution. In this work, the objective was to evaluate the extent to which prospective wetland substrate material may adsorb NAFCs using a peat-mineral mix (PMM) sourced from the Athabasca Oil Sands Region (AOSR). The PMM and NAFCs were first mixed and then equilibrated across a range of NAFC concentrations (5-500 mg/L) with moderate ionic strength and hardness (∼200 ppm combined Ca2+ and Mg2+) that approximate wetland water chemistry. Under these experimental conditions, low sorption of NAFCs to PMM was observed, where sorbed concentrations of NAFCs were approximately zero mg/kg at equilibrium. When NAFCs and PMM were mixed and equilibrated together at environmentally relevant concentrations, formula diversity increased more than could be explained by combining constituent spectra. The TOC present in this PMM was largely cellulose-derived, with low levels of thermally recalcitrant carbon (e.g., lignin, black carbon). The apparent enhancement of the concentration and diversity of components in PMM/NAFCs mixtures are likely related to aqueous solubility of some PMM-derived organic materials, as post-hoc combination of dissolved components from PMM and NAFCs cannot replicate enhanced complexity observed when the two components are agitated and equilibrated together.

Abstract:Semiconductor-based photocatalysis is emerging as a potent oxidation for degrading Naphthenic acids (NAs) in oil sands process water (OSPW). Here, the impacts of some of the OSPW-relevant parameters and properties were investigated to address some questions about the previously observed varied performance of a given catalyst in different OSPW samples and the differences of some catalysts in OSPW that could not be explained by their intrinsic electrophysical properties. To examine the individual impacts of the selected factors, model NAs were used with ultra-performance liquid chromatography (UPLC) for the analyses. The photocatalytic degradation kinetics was enhanced by the presence of Cl − and SO 4 2− in the water while HCO 3 − dampened the kinetics. The impact of a significant change in water pH appeared to be dependent on the direction of the change relative to the isoelectric points (IEP) of the catalyst. The reactivity of NAs in the photocatalysis was strongly structure-dependent with the alkyl branching and substituent being influential factors. The study of the reaction mechanism indicated that both O 2 − and OH mediated in the reactions. The study provides more understanding of the potentially varied dynamics of the photocatalytic process in treating different streams and can help make better-informed process designs.

Abstract:Large volumes of oil sands process water (OSPW) containing naphthenic acids (NAs) and other contaminants are generated by oil sands operations. For the first time, pristine and zinc chloride-activated biochars from sludge from a municipal wastewater treatment plant are studied in the adsorption of NAs in real OSPW. By using activated biochar instead of pristine biochar, the adsorption capacity (qt) for total NAs (classical and oxidized NAs) increased from 2.3 to 26.6 mg/g. The increase in qt for activated biochar was related to the improvement of surface area from 7.9 to 513 m2/g, the abundance of mesopores (89% of pore volume), and the oxygen-containing surface groups. Equilibrium was reached at 2 h of contact time. Considering the thousands of structures of NAs in the OSPW, the activated biochar presented higher affinity for NAs with higher carbon and double bond equivalency numbers, therefore, NAs with more hydrophobic and higher cyclicity characteristics. Pore-filling, hydrogen bonding, and π-π interactions are the adsorption mechanisms. Thermal regeneration of spent activated biochar was studied, and in the fifth cycle of regeneration, the regenerated biochar had qt of 7.1 mg/g, which was 5 times higher than the adsorption capacity of fresh pristine biochar. Adsorption treatment of OSPW using activated biochar decreased the following parameters: metals Mg (7%), Ca (21%), Sr (27%), and Ba (60%), organic matter by means of DOC (63%) and COD (66%), acute toxicity towards V. fischeri (66%) and bioavailability of organics (91%). These findings promote circular economy as it presents an efficient adsorbent for NAs in OSPW developed from sludge, reducing the environmental impact associated with sludge disposal and allowing the efficient reclamation and reuse of OSPW.

Abstract:Naphthenic acids (NAs) are a complex mixture of organic compounds released during bitumen extraction from mined oil sands that are important contaminants of oil sands process-affected water (OSPW). NAs can be toxic to aquatic organisms and, therefore, are a main target compound for OSPW. The ability of microorganisms to degrade NAs can be exploited for bioremediation of OSPW using constructed wetland treatment systems (CWTS), which represent a possible low energy and low-cost option for scalable in situ NA removal. Recent advances in genomics and analytical chemistry have provided insights into a better understanding of the metabolic pathways and genes involved in NA degradation. Here, we discuss the ecology of microbial NA degradation with a focus on CWTS and summarize the current knowledge related to the metabolic pathways and genes used by microorganisms to degrade NAs. Evidence to date suggests that NAs are mostly degraded aerobically through ring cleavage via the beta-oxidation pathway, which can be combined with other steps such as aromatization, alpha-oxidation, omega-oxidation, or activation as coenzyme A (CoA) thioesters. Anaerobic NA degradation has also been reported via the production of benzoyl-CoA as an intermediate and/or through the involvement of methanogens or nitrate, sulfate, and iron reducers. Furthermore, we discuss how genomic, statistical, and modeling tools can assist in the development of improved bioremediation practices.
Effects of Hot Water-Based Extraction Process on the Removal of Petroleum Hydrocarbons from the Oil-Contaminated Soil

Abstract:The properties including oil content, density, high viscosity, and corresponding components were analyzed in the samples of the oil-contaminated soil from one country in the Middle East; also, chemical composition of the solids in the oil-contaminated soil was analyzed by using the X-ray deflection structure analysis method. Characterization results of obtained samples showed the similarities of the oil-contaminated soil with the Canadian oil sand, and then the hot water-based extraction technology that is successfully used in commercial oil sand operation in Canada was fine-tuned to be a novel cleaning technology to clean up oil-contaminated soils. Then, the different cleaning agents, cleaning agent dosage, liquid–solid ratio, temperature, stirring rate, and time as the primary parameters that would affect the cleaning effect were all optimized. Under the optimum conditions, the oil removal results demonstrated that oil removal by this technology was very effective; the oil content of the cleaned soils could be reduced to less than 0.3 wt %, which met the treatment and disposal requirements stipulated in GB4284-2018, and the reaction process was theoretically analyzed by a kinetic method. The oil phase obtained by water washing after further solvent purification treatment has a higher recycling value. This study provides a cost-effective way to recover and deal with the oil-contaminated soil from the perspective of economic efficiency and environmental protection.

Abstract:This paper reported the impact of UV/H2O2, UV/chlorine, and UV/persulfate advanced oxidation processes on the molecular transformations of dissolved organic matter (DOM), removal of naphthenic acids (NAs) and acute toxicity in oil sands process water (OSPW). The UV/persulfate process exhibited the highest removal (81.2% with 2 mM dose) towards classical NAs and highest reduction in acute toxicity to Vibrio fischeri among the three processes. The fraction of DOM such as CHOS class species decreased along with the increase of the oxidant doses in all processes. The increase in O/C ratio and lack of change in the H/C and double bond equivalence indicated that H-abstraction followed by the OH-addition was the main reaction pathway for all processes. This observation aligned with previous studies using model compounds and proved that OSPW DOM reacted similarly to model compounds. Sulfur containing organic matters were the most liable compounds in OSPW NOM, while UV/chlorine was the most effective process to oxidize nitrogen containing organic matters. Overall results revealed that the UV/persulfate process could be used as a promising technique for the removal of OSPW NA and reduction of acute toxicity towards Vibrio fischeri. In addition, this DOM characterization approach could be utilized to investigate the transformation of complicated OSPW DOM and to identify the byproducts generated during different water treatment processes.

Abstract:The open pit oil sands mining operations north of Fort McMurray, Alberta, Canada, are accumulating tailings waste at a rate approximately equal to 4.9 million m(3) /d. Naphthenic acids are among the most toxic components within tailings to aquatic life, but structural components have largely remained unidentified. In the present study, electrospray ionization high-resolution mass spectrometry (ESI-HRMS) and synchronous fluorescence spectroscopy (SFS) were used to characterize fractions derived from the distillation of an acid-extractable organics (AEO) mixture isolated from oil sands process-affected water (OSPW). Mean molecular weights of each fraction, and their relative proportions to the whole AEO extract, were as follows: fraction 1: 237 Da, 8.3%; fraction 2: 240 Da, 23.8%; fraction 3: 257 Da, 26.7%; fraction 4: 308 Da, 18.9%; fraction 5: 355 Da, 10.0%. With increasing mean molecular weight of the AEO fractions, a concurrent increase occurred in the relative abundance of nitrogen-, sulfur-, and oxygen-containing ions, double-bond equivalents, and degree of aromaticity. Structures present in the higher-molecular-weight fractions (fraction 4 and fraction 5) suggested the presence of heteroatoms, dicarboxyl and dihydroxy groups, and organic acid compounds with the potential to function as estrogens. Because organic acid compositions become dominated by more recalcitrant, higher-molecular-weight acids during natural degradation, these findings are important in the context of oil sands tailings pond water remediation.
Effect Of Ozonation On The Estrogenicity And Androgenicity Of Oil Sands Process-Affected Water

Abstract:There is increasing environmental concern about the volume of oil sands process-affected water (OSPW) produced by the oil sands industry in Alberta, Canada. There is limited knowledge of the toxic effects of OSPW and one of the primary organic constituents, naphthenic acids (NAs), which are thought to be one of the toxic constituents of OSPW. OSPW and NAs can have endocrine disrupting potential. The NAs in OSPW are persistent, but ozonation can significantly reduce concentrations of NA, while increasing their biodegradability, and consequently reduce OSPW toxicity. However, it is of concern that OSPW ozonation might generate hydroxylated cycloaliphatics with endocrine disrupting potential. In this study, the estrogen receptor- (ER) and androgen receptor- (AR) mediated effects of OSPW and ozone-treated OSPW were investigated in vitro by use of T47D-kbluc (estrogen responsive) and MDA-kb2 (androgen responsive) cells. Ozonation neither attenuated nor intensified the estrogenicity of OSPW. The estrogenic responses to untreated OSPW and ozone treated OSPW were 2.58(+/- 0.22)-fold and 2.48(+/- 0.13)-fold greater than those of controls, respectively. Exposure to untreated OSPW produced significant antiandrogenicity in the presence of 0.01, 0.05, or 0.1 nM testosterone (T), while ozone-treated OSPW produced significant antiandrogenicity in the presence of 0.01 or 0.05 nM T. Exposure to untreated and ozone-treated OSPW also caused potentiation of androgen receptor-mediated effects of T. OSPW could cause estrogenic and antiandrogenic effects through receptor mediated pathways, and ozonation can partially mitigate the OSPW antiandrogenicity as well as androgen potentiating effect, without increasing estrogen potency.
Application of an indigenous microorganisms-based fixed-bed GAC-biofilm reactor for passive and sustainable treatment of oil sands process water through combined adsorption and biodegradation processes

Abstract:In this study, a fixed-bed biofilm reactor (biofilter) was developed and applied for oil sands process water (OSPW) remediation by using granular activated carbon (GAC) as packing media. Using quantitative polymerase chain reaction (qPCR) detection, the total bacterial copy number (16S) in the GAC biofiltration system was found to reach a relatively stable level (1.3 ± 0.2 × 109 copies/g GAC) after 62 days of operation, and the thickness of biofilm on GAC surface was 26.7 ± 4.3 μm based on the scan of confocal laser scanning microscopy (CLSM). The established GAC-biofilter showed 95.4% naphthenic acids (NAs) removal from raw OSPW after 2 months of operation. The GAC-biofilter also showed 88.3% NAs removal after a long operation time (2 years), indicating its sustainable bioremediation capacity for OSPW. 16S and 18S rRNA gene-targeted metagenomic sequencing showed that the microbial community in the GAC biofilter had higher diversity and richness than that found in the sand biofilter which was used for OSPW treatment previously. Comamonadaceae and Saccharomycotina were found to be the dominant bacterial and fungal families in the GAC biofilter, respectively. Xenobiotic metabolism function of the microbial community may contribute significantly to the biodegradation of NAs. The GAC biofiltration process is a promising passive OSPW treatment approach that can be used in-situ.
