Abstract:The aim of this study was to identify the influence of the anesthetic eugenol on crucian carp. The muscle quality and freshness were little influenced by anesthesia from the perspective of cooking loss (CL) and total volatile basic nitrogen (TVB-N). In contrast, lower redness, liquid loss (LL), and 2-thiobarbituric acid (TBA) values were observed in anesthetized fish. It was found that anesthesia increased serum cortisol content and reduced serum glucose concentration. There was little influence of eugenol on serum glutamic-oxaloacetic transaminase (GOT), glutamic-pyruvic transaminase (GPT), and alkaline phosphatase (AKP) activities, while serum and muscle acid phosphatase (ACP) activities significantly increased after the use of the anesthetic. Results suggested that the stress of eugenol to crucian carp was nontoxic and did not lead to hyperglycemia. On the basis of these findings, eugenol as an anesthetic is effective and safe for the transportation of crucian carp.

Abstract:Eugenol is a widely used fishery anesthetic. This study investigated the effects of various concentrations of eugenol on blood physiological and biochemical indexes, and muscle flavor, in crucian carp (Carassius auratus). To determine the appropriate concentration of eugenol anesthetic for use in crucian carp transportation and production operations, we evaluated seven anesthesia groups of 20, 30, 40, 50, 60, 70, and 80 mg/L and one control group (without eugenol) to determine the effects on blood physiological and biochemical indexes, and muscle flavor. The red blood cells and platelets of crucian carp decreased significantly (p < 0.05) with eugenol treatment. With increasing eugenol concentration, the white blood cells and hemoglobin did not change significantly, whereas lactate dehydrogenase, alkaline phosphatase, alanine aminotransferase, and aspartate aminotransferase increased significantly (p < 0.05). The content of phosphorus, magnesium, and sodium increased after anesthesia, whereas the content of potassium decreased with increasing eugenol concentration. After anesthesia, the content of albumin and total protein in the serum decreased with increasing eugenol concentration (p < 0.05); triglyceride first increased and subsequently decreased (p < 0.05); blood glucose content first increased and then decreased (p < 0.05); and no significant difference was observed in total cholesterol content (p > 0.05). No significant difference was observed in muscle glycogen and liver glycogen content after eugenol anesthesia (p > 0.05). The eugenol-based anesthesia test did not indicate major liver histomorphological effects, but the very small number of gill sheet edema cases observed requires further study. Analysis of electronic nose data indicated that eugenol treatment affected the flavor of the fish. The anesthesia concentration of 20-80 mg/L had some effect on the physiology and biochemistry of crucian carp, thus providing a reference for the application of eugenol in crucian carp transportation and experimental research.

Abstract:The aim of this study was to evaluate the effects of eugenol, benzocaine, and ice water during the sedative, anesthetic or euthanasia processes on the welfare of adult grass carp (Ctenopharyngodon idella). The experimental design was randomized and the animals were divided into eight groups. Sixty-two animals underwent an acclimation period. The neutral group used to obtain basal data of grass carp were not subjected to treatments, but anesthetized to collect blood samples and euthanized by medullary section. The others seven groups were submitted to seven treatments with eight repetitions (control group; ethanol; eugenol 50 mgL-1, eugenol 250 mgL-1, benzocaine 100 mgL-1, benzocaine 300 mgL-1, and ice water 2:1), their behavior was observed. Blood samples was collected and then euthanized by medullary sectioning. Biometric data were measured and a part of the liver was collected for hepatic glycogen analysis. There was a statistically significant difference in the time required to reach the anesthetic stage between the groups (p < 0.01). Benzocaine and eugenol at the higher concentration provided the fastest responses to sedatives and anesthetics, respectively. The animals subjected to higher anesthetic concentrations reached stage five and did not return from anesthesia, therefore, benzocaine and eugenol were effective euthanizing agents. Benzocaine at the lowest concentration showed the highest concentrations of glucose and cortisol (p < 0.05). Although benzocaine at 100 mgL-1 concentrations is widely used as an anesthetic in fish, this study demonstrated its use as a stressor agent. Basal data of grass carp for stress parameters are presented for the first time.

Abstract:The anaesthetic effect of vanillin on crucian carp was investigated using different concentrations of vanillin, with a nonvanillin control. The effective concentration range of vanillin anaesthesia was determined from the behavioural characteristics of crucian carp during the anaesthesia onset and recovery phases. Physiological and biochemical indices, and the electronic nose response to the fish muscle, were measured over the range of effectiveanaestheticc concentrations. An increased concentration of vanillin shortened the time taken to achieve deep anaesthesia but increased the recovery time. The levels of white blood cells, red blood cells, haemoglobinn, platelets, alanine aminotransferase, alkaline phosphatase, lactate dehydrogenase, phosphorus, potassium, magnesium, total protein, and serum albumin were lower than the control in the vanillin treatment group. Triglycerides and total cholesterol were not significantly affected. Histology showed no effect of vanillin on the liver, except at 1.00 g/L vanillin. Vanillin resulted in a nondose-responsive effect on the gill tissue, increasing the width and spacing of the gill lamellae. E-Nose analysis of the carp-muscle flavour volatiles was able to distinguish between different vanillin treatment concentrations. GC-IMS identified 40 flavour compounds, including 8 aldehydes, 11 alcohols, 10 ketones, 2 esters, and 1 furan. Vanillin had aanaestheticic effect on crucian carp and these findings provide a theoretical basis for improving the transport and experimental manipulation of crucian carp.

Abstract:hile hereditary (familial) amyloidosis has probably been around for as long as recorded hisW tory, it has only been recognized as a distinct entity in the latter half of the 20h Century. The first documentation of transthyretin amyloidosis as an autosomal dominant inherited disease was in 1952 by Corino Andrade' . This disease which was named familial amyloidotic polyneuropathy (FAP) because of the predominance of peripheral neuropathy in affected individuals of Portuguese origin has grown in importance as the number of mutations in transthyretin which are associated with pathology (presently over 70) and the sytemic nature of the disease have become appreciated*. It is truly the prototype of the hereditary systemic amyloidoses and displays all of the characteristics of this group of fibril protein deposition diseases. 1) Amyloid affects many organ systems with variation fiom kindred to kindred and individual to individual. 2) The disease is adultonset; usually starting after the childbearing years, as would be expected for a fatal hereditary condition. 3) The disease is dominantly inherited, a common feature of diseases due to mutations in genes for structural proteins. 4)Anumber of mutations in the same protein may govern expression of the genetic disease. 5 ) The occurrence of amyloid deposition in old age in the absence of any genetic mutation. Transthyretin amyloidosis is only one of several forms of systemic amyloidosis that have been characterized in the last 20 years. Mutations in the gene for apolipoprotein AI cause a number of syndromes of systemic amyloidosis with restrictive cardiomyopathy the most prominent clinical feature, but with cutaneous and renal amyloidosis in some kindreds. As with transthyretin, a number of disease causing mutations of apoA 1 have been described; at least eight within the last ten years. Four mutations in the gene for fibrinogen Aa-chain are associated with renal amyloidosis. Two mutations in lysozyme have been associated with either renal or hepatic amyloidosis. Two mutations in gelsolin and one in cystatin C can cause systemic amyloid deposition. In the case of transthyretin and apolipoprotein AI, it is now known that normal protein can be associated with amyloid deposits; transthyretin in the heart, apoAI in the aorta. Whether amyloid deposits can be derived fiom normal fibrinogen Aa-chain, lysozyme, gelsolin or cystatin C has yet to be explored. Besides the hereditary systemic amyloid syndromes, several inherited forms of localized amyloidosis have been described. The most common is Alzheimer disease due to mutations in APPP, presenilin1 or presenilin-2. GertstmannStraussler-Scheinker disease and familial Cruetzfeld-Jacob diseases are due to mutations in the pnon protein. As with the systemic amyloidoses, several mutations have been found to cause the localized forms and, at least with Alzheimer disease, senile amyloid deposits fiom normal ApPP occur without obvious genetic basis. Despite the amazing amount of knowledge gained in the last two decades about these diseases, we still have little in the way of specific therapy to offer individuals and families affected with hereditary amyloidoses. We can offer DNA testing, genetic counseling and the reassurance that science is progressing at an incredible rate.

In the first experiment, fish were exposed to 30, 80, 130, 180, 200, and 230 μL EOLA L−1. The second experiment evaluated smaller and larger fish with 180 μL EOLA L-1. In the third experiment, ventilatory rates (VR) for up to 120 min in animals during exposure to 5 and 10 μL EOLA L−1 were evaluated. In the fourth experiment, fish anesthetized with 30 and 180 μL EOLA L-1 were assessed at 0, 30, and 60 min after anesthesia recovery to evaluate biochemical and antioxidant parameters. The best mild and deep anesthesia times were obtained with 30 and 180 μL EOLA L-1, and larger fish had the highest times. The VR increased in fish exposed to EOLA. Blood glucose and whole-body cortisol levels were higher in fish anesthetized with 180 μL EOLA L-1. Fish exposed to EOLA had higher liver glutathione S-transferase and superoxide dismutase activities without affecting catalase and lipid peroxidation levels. The 180 μL EOLA L-1 is recommended for fat snook anesthesia because it increases VR, blood glucose, and whole-body cortisol levels and prevents oxidative stress.

A meta‐analytic review of monoterpene for fish anaesthesia

Abstract:The use of anaesthetic agents has been increasing to address the welfare needs of different fish species in scientific and aquaculture practices. MS‐222 is the most used synthetic anaesthetic but some limitations and side effects have been reported. Natural substances have been used as potential substitutes with clove oil playing an important role due to its eugenol content. Yet, other monoterpenes have shown anaesthetic properties in different fish species. As such, a quantitative assessment has been conducted to explicitly relate and summarise the use of monoterpenes with described anaesthetic properties in fish. A combined PubMed, Web of Science, Scopus, PubMed Central and Europe PMC electronic database search was performed, from January 2002 to August 2022, following the PRISMA guidelines. Out of 1555 articles retrieved from the literature search, 30 English reports met the criteria for inclusion in the meta‐analysis through a generic inverse‐variance method (random‐effects model) and according to the fish age. The findings show a total of 10 different monoterpenes with high homogeneity and effectiveness in inducing anaesthesia and recovery according to the fish anaesthetic criteria. Further subgroup analysis showed these effects were independent of fish age. Meta‐regression of the included studies revealed an inverse linear association between anaesthesia induction and recovery time and water bath temperature while no further correlation was found with other confounding factors (body weight, water pH and dissolved oxygen levels). Although further studies will be required, this meta‐analysis provides robust evidence that different monoterpenes serve as an attractive and effective alternative for fish anaesthesia although water bath temperature also played an important role in anaesthesia outcome.

Effects of Nisin, Cecropin, and Penthorum chinense Pursh on the Intestinal Microbiome of Common Carp (Cyprinus carpio)

Abstract:Following a ban on antibiotic use in the feed industry, trials on the effects of various immunostimulants (prebiotics, probiotics, antimicrobial peptides [AMPs], and herbs) on the survival, growth, immunity, and disease control of farmed fish in aquaculture are being rapidly conducted. The wide variety of microbes with roles in nutrition, metabolism, and immunity in the fish intestine is the primary factor affecting the fermentability and functionality of dietary immunostimulants. For this reason, the dynamic interactions between immunostimulants and the intestinal microbiome may influence fish health. In this study, the effects of two agriculturally important AMPs (nisin and cecropin) and one herb (Penthorum chinense) on the gut microbiome of common carp were investigated, using 16S rDNA high-throughput sequencing. The results suggest that all three substances can alter the richness, diversity, and composition of the intestinal microbiota of common carp. P. chinense had a similar effect on the gut microbiota of common carp to that of nisin, and both promoted more striking changes in the gut microbiota community than did cecropin. The relative abundance of Proteobacteria was lower in the nisin and P. chinense groups than in the control and cecropin groups. The relative abundance of Bacteroidetes in the nisin, cecropin, and P. chinense groups was markedly increased, compared with that of the control group. Additionally, nisin, cecropin, and P. chinense showed obvious anti-inflammatory effects on the fish intestine, which was reflected by significantly increasing the expression levels of two anti-inflammatory cytokines IL-10 and TGF-β. Some digestive enzyme activities in the fish intestine were also significantly enhanced by supplementing these three substances in feeds.
Combined Transcriptomics and Metabolomics Analyses in Grass Carp Under Anesthetic Stress

Abstract:Ctenopharyngodon Idella , as a common freshwater bony fish, is more susceptible to various diseases than other carp species, so it has been proposed as a test organism for toxicological analysis In this study, C. idella were anesthetized with MS-222 and 2-PE, and the related anesthetic mechanism and toxic effects were revealed by transcriptomics and metabolomics analyses. When the concentration of MS-222 was 80 mg/L and 200 mg/L, 179 and 887 differentially expressed genes (DEGs), respectively, were identified in the brain tissue of C. idella . When the concentration of 2-PE was 0.6 mL/L and 1.2 mL/L, 498 and 514 DEGs were identified. The DEGs associated with MS-222 treatment were enriched in immune pathways, lipid metabolism, amino acid metabolism, and various signaling pathways; DEGs associated with 2-PE treatment were enriched in immunity and amino acid metabolism. In total, 304 metabolites were identified using a combination of positive and negative ion modes in mass spectrometry. The common differential metabolites identified in the MS-222 high and low concentration groups were 20-HETE and 12(R)-HETE; the common significant differential metabolite identified in the 2-PE high and low concentration groups was salidroside. In combination with the transcriptomics analysis and metabolomics analysis, the results showed that with the MS-222 and 2-PE concentrations used in this experiment, the metabolism of arachidonic acid in C. idella was inhibited by MS-222, and 2-PE affected the upstream and downstream metabolic pathways of arachidonic acid metabolism, thereby affecting the metabolism of arachidonic acid. Both anesthetics induce sedation by affecting related metabolites that affect stress response and autoimmunity. Metabolomics results showed that neither anesthetic had a significant effect on cortisol expression.

This work evaluated the safety and sedative and anesthetic efficacy of EOs from three native Brazilian plants, Acmella oleracea (jambu), Aloysia hatschbachii and Cordia verbenacea (whale herb) in juvenile Rhamdia quelen (silver catfish). Anesthetic induction and recovery protocols (20 to 400 mg L-1) and long exposure (48 h) from 10 to 100 mg L-1 were tested. The EOs performed sedative and/or anesthetic activities: AOOi at a concentration of 20 mg L-1, AOOl at 50 and 100 mg L-1, AHOl, and CVOL (only sedation) 50 mg L−1, as there were no important adverse effects and/or mortality. The results obtained indicate that Cordia verbenacea EO is the most promising as a sedative for juvenile silver catfish at a concentration of 50 mg L−1.

Abstract:Traumatic brain or spinal cord injury (SCI) has devastating consequences for patients, and can lead to life-long disability. However, even in the most serious cases, there is normally some recovery of function. For example, 73% of patients who initially present with complete paralysis but sacral sensory sparing recover some motor function within a year (Marino et al ., 1999). The mechanisms of this recovery are still poorly understood and are likely to include the following: reduction of local ischaemia, oedema and inflammation; recovery from spinal shock of traumatized but undamaged pathways; remyelination of demyelinated axons; functional synaptic plasticity allowing spared pathways to take over the lost functions; and anatomical plasticity leading to new, and functionally effective, circuitry. In the past few years it has become apparent that the last of these possibilities, anatomical plasticity, may play a significant role. Although the CNS is not able to regenerate a major pathway such as the corticospinal tract, there is accumulating evidence that some pathways are able to sprout locally to establish novel circuits that restore a limited degree of function. The key question is, can such plasticity be promoted to enable patients to recover complex functions such as fine hand control? This exciting possibility is addressed by two papers in this issue of Brain from James Fawcett and colleagues at the Cambridge Centre for Brain Repair (Galtrey et al ., 2007; Smith et al ., 2007). After injury to a pathway such as the corticospinal tract, axons normally die back a short distance but then initiate a regenerative response which is unsuccessful and confined to local sprouts at the injury site (Kerschensteiner et al ., 2005). Abstract:Fucoidan with its excellent biological activities such as growth promotion, antioxidant and strong immunity, is widely used in animal production. The present study was conducted to investigate the influences of feeding fucoidan on growth performance, biochemical indices, immunity, the antibacterial ability of plasma, the digestive enzyme activity of the intestine, antioxidant capacity, and the histological structure of liver in juvenile common carp. Five experimental diets added with 0 (Diet 1), 500 (Diet 2), 1,000 (Diet 3), 1,500 (Diet 4), and 2,000 (Diet 5) mg/kg fucoidan were fed to triplicate groups of 30 fish (35.83 ± 0.24 g) respectively for 8 weeks. The results showed that fish fed diets with a fucoidan supplementation of 1,666.67–1,757 mg/kg might have the best growth performance ( p < 0.05). The levels of plasma total protein (TP) and albumin (ALB) in Diet 3, Diet 4, and Diet 5 were higher than those in Diet 1 and Diet 2 ( p < 0.05). Moreover, the contents of plasma C3, LYZ, and IgM; the antibacterial ability of serum; and the activity of SOD, CAT, POD, and GPX in the liver, and ACP, AKP, LPS, AMS, and TRY in the intestine significantly improved; the contents of LPO and MDA in the liver were notably decreased in diets with fucoidan supplement ( p < 0.05). Furthermore, the activity of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) and the contents of total bilirubin (TB) and glucose (Glu) in Diet 5 were the highest among the groups. Meanwhile, proinflammatory factors (plasma IL-6 and IL-1β) had a higher expression, but anti-inflammatory factors (plasma IL-1) had a lower expression in Diet 5 ( p > 0.05). It indicated that a higher dose (2,000 mg/kg) of fucoidan may induce inflammation and metabolic disorders. Interestingly, histological results of liver also indicated that dietary fucoidan intake in certain amounts (500–1,500 mg/kg) could ameliorate hepatic morphology, but the high dosage (2,000 mg/kg) probably damaged the liver. To the best of our knowledge, this is the first study on the application of fucoidan as a functional additive to juvenile common carp. The results of the present study can be used to guide the application of fucoidan in healthy aquaculture and can further reveal the effect and mechanism of fucoidan on the nutritional physiology of aquatic animals.

Modulatory effects of laurel-leaf cistus (Cistus laurifolius) ethanolic extract on innate immune responses and disease resistance in common carp (Cyprinus carpio)

Abstract:Medicinal herbs are used for growth promotion, disease control and other health benefits in aquaculture industry. Here, we examined the effect of dietary laurel-leaf cistus (Cistus laurifolius) ethanolic extract on growth performance, digestive enzyme activity, haematological profile and nonspecific immune responses in common carp (Cyprinus carpio). In addition, resistance against Aeromonas hydrophila infection was examined. Common carp was fed diets containing 0 (Control), 0.1 (CL0.1), 0.5 (CL0.5) and 1 (CL1) g kg-1 laurel-leaf cistus extract for 45 days. After 30 days, superoxide anion production (SAP) increased in CL0.1 and CL0.5 fish groups and at the end of the study all experimental fish groups had higher SAP compared to that of the control (P ˂ 0.05). Lysozyme activity (LA) was elevated in CL0.5 and CL1 treated groups on 30th day (P < 0.05), and this increase was only observed in C0.1 fish group at the end of study compared to control (P ˂ 0.05). Myeloperoxidase activity was significantly increased in CL0.5 and CL1 fish groups at the end of study. IL-1βgene expression was significantly increased in treated fish in a dose-depended manner. Similar results were observed for transcription of IL-6 and IL-8 (P < 0.05). Anti-inflammatory cytokines, IL-10 and TGF-β were highly up-regulated in the intestine and head kidney of CL treated fish groups compared to control (P < 0.05). At the end of experiment, significantly higher final body weight, weight gain, and specific growth rate were obtained in CL0.1 treated fish group compared to control. However, growth was negatively affected in CL1 fish group (P < 0.05). CL1 fish group had also a significantly higher FCR. Amylase activity was significantly increased in all experimental fish groups compared to control (P ˂ 0.05). Trypsin activity was decreased in CL0.1 and CL1 fish groups (P ˂ 0.05). WBC and RBC were significantly increased (P ˂ 0.05) in CL0.5 and CL1 fish groups, whereas haemoglobin, haematocrit, mean cell, mean cell haemoglobin contents were no significantly changed among control and treatment groups. Result of challenge test with A. hydrophila exhibited that survival rate in all treatment groups was significantly higher than that of control. These findings demonstrated that laurel-leaf cistus at 0.1 g kg-1 can be a suitable candidate for growth promotion, immune system induction and infection control in fish.
Toxicological effects of meloxicam on physiological and antioxidant status of common carp (Cyprinus carpio)

Abstract:Background: Fish in aquatic environments are end consumers of the food chain and are widely used for the evolution effects of environmental pollution and their interactions in aquatic ecosystem. Objective: In the present study, common carp (Cyprinus carpio) fingerlings were selected to assess the potential risk and aquatic toxicity of meloxicam as a non-steroidal anti-inflammatory and a commonly used pharmaceutical drug. Methods: In order to evaluate meloxicam toxicological effect on haematological, antioxidant status, enzymological and histological parameters, based on its LC50 24 h acute toxicity (10.05 mg L-1 ), fish fingerlings were exposed to four doses of meloxicam including; 0 (control), 0.1 (low), 1 (medium) and 2 mg L-1 (high) under static bioassay method for 28 days. Results: The results showed that sublethal doses of meloxicam significantly decreased alanine aminotransferase, alkaline phosphatase, superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX) levels in comparison with the control group after 28 days (p < 0.05). However, red blood cell, haematocrit, haemoglobin and malondialdehyde values in fish exposed to meloxicam significantly increased alongside its concentration (p < 0.05) more than the control group after 28 days. SOD, CAT and GPX mRNA expression levels in gill, liver, kidney and brain organ of fish under meloxicam treatment were significantly down-regulated compared to the control group (p < 0.05). Histopathological assessment showed the increased vacuolation in hepatocytes in liver of fish exposure to medium and high doses of meloxicam. Conclusion: In conclusion, meloxicam induces oxidative stress in common carp which results a disruption of physiological and health status of this species based on our current findings.

Abstract:The aim of the study was to evaluate the effects of emerging environmental contaminants, the non-steroidal anti-inflammatory drugs (NSAIDs) diclofenac (DCF) and ibuprofen (IBP), on physiological functions in juvenile common carp ( Cyprinus carpio ). Fish were exposed for 6 weeks, and for the first time, NSAIDs were administered through diet. Either substance was tested at two concentrations, 20 or 2000 μg/kg, resulting in four different treatments (DCF 20, DCF 2000, IBP 20, IBP 2000). The effects on haematological and biochemical profiles, the biomarkers of oxidative stress, and endocrine disruption were studied, and changes in RNA transcription were also monitored to obtain a comprehensive picture of toxicity. Fish exposure to high concentrations of NSAIDs (DCF 2000, IBP 2000) elicited numerous statistically significant changes ( p < 0.05) in the endpoints investigated, with DCF being almost always more efficient than IBP. Compared to control fish, a decrease in total leukocyte count attributed to relative lymphopenia was observed. Plasma concentrations of total proteins, ammonia, and thyroxine, and enzyme activities of alanine aminotransferase (ALT), aspartate aminotransferase, and alkaline phosphatase (ALP) were significantly elevated in either group, as were the activities of certain hepatic antioxidant enzymes (superoxide dismutase, glutathione-S-transferase) in the DCF 2000 group. The transcriptomic profile of selected genes in the tissues of exposed fish was affected as well. Significant changes in plasma total proteins, ammonia, ALT, and ALP, as well as in the transcription of genes related to thyroid function and the antioxidant defense of the organism, were found even in fish exposed to the lower DCF concentration (DCF 20). As it was chosen to match DCF concentrations commonly detected in aquatic invertebrates (i.e., the potential feed source of fish), it can be considered "environmentally relevant". Future research is necessary to shed more light on the dietary NSAID toxicity to fish.

Abstract:The study explored the effects of Angelica sinensis extract (AsE) on oxidative lesions and apoptosis in branchiae and red blood corpuscles in hypoxia–reoxygenation (HR) and Cu-treated carp (Cyprinus carpio var. Jian). After feeding trial for 30 days, the carp were exposed to HR and CuSO4. The results indicated that dietary AsE increased the durative time, decreased the oxygen consumption rate, suppressed ROS generation and cellular component oxidation, decreased enzymatic antioxidant activity and reduced glutathione (GSH) levels in red blood corpuscles and branchiae in carp under hypoxia. Moreover, dietary AsE avoided the loss of Na+,K+-ATPase, metabolic and antioxidant enzyme activities, ROS generation and cellular component oxidation, as well as the increase in caspase-8, 9, and 3 activities in the branchiae of the carp and inhibited ROS generation. It furthermore avoided the loss of Na+,K+-ATPase and metabolic enzyme activities, the decrease in GSH levels and hemoglobin content, the increase in the activities of caspase-8, 9, and 3 and the increase in the levels of cytochrome c and phosphatidylserine exposure in the red blood corpuscles of Cu-exposed carp. The present results suggested that dietary AsE improved hypoxia tolerance and inhibited HR or Cu-triggered oxidative lesions and apoptosis. Therefore, AsE can be utilized as a natural inhibitor of Cu and HR stress in fish.

Abstract:Species feeding lower at the food web like cyprinids (carp) are often advocated for sustainable aquaculture. However, their gastrointestinal biology (lack of stomach) has limited their acid‐peptic digestion, acid lysis of plants and invertebrate tissues, and phosphorus uptake. This review presents data from 118 articles on the use of plant‐based feed additives (PFAs) in Cyprinus carpio (common carp). Our review shows that most PFA research in carp is focused on improving feed and nutrient utilization efficiency along various levels (in vivo digestibility, metabolizability to in situ nutrient loading), while other half is focused on ensuring animal welfare standards (biomass, immunity, welfare). Several reviewed plant‐based additives however tend to do both. The PFAs include essential oils (EOs), plant‐part extracts (PPEs), medicinal plants (herbs) and spices. The effective inclusion rate of EOs is lower (0.5%–0.75%). Whereas PPEs, herbs and spices work at a generic inclusion rate of 2%–5% in carp feeds. Even at an optimum inclusion rate, there are factors controlling PFAs efficacy such as methods of preparation (reviewed). PFAs like lemon beebrush, geranium, dill, Rosemary pepper and common mallow with/without certain pre‐conditions are better to avoid in cyprinid diets. Although certain PFAs may potentially alter organoleptic qualities of carp flesh or might assist in training carp immunity, information on these aspects is limited. The review also provides a quick guide to researchers and feed companies interested in exploring the potential of PFAs in C. carpio aquaculture. Potential PFAs that could be valuable for cyprinids, but not yet explored are also listed.

Abstract:Avermectin (AVM) has been utilized extensively in agricultural production since it is a low-toxicity pesticide. However, the pollution caused by its residues to fisheries aquaculture has been neglected. As an abundant polyphenolic substance in plants, ferulic acid (FA) possesses anti-inflammatory and antioxidant effects. The goal of the study is to assess the FA's ability to reduce liver damage in carp brought on by AVM exposure. Four groups of carp were created at random: the control group; the AVM group; the FA group; and the FA + AVM group. On day 30, and the liver tissues of carp were collected and examined for the detection of four items of blood lipid as well as the activity of the antioxidant enzymes catalase (CAT), glutathione (GSH) and malondialdehyde (MDA) in carp liver tissues by biochemical kits, and the transcript levels of indicators of oxidative stress, inflammation and apoptosis by qPCR. The results showed that liver injury, inflammation, oxidative stress, and apoptosis were attenuated in the FA+AVM group compared to the AVM group. In summary, dietary addition of FA could ameliorate the hepatotoxicity caused by AVM in carp by alleviating oxidative stress, inflammation, apoptosis in liver tissues.

Abstract:Use of pesticides or insecticides can be highly toxic to aquatic life forms due to leaching and agricultural runoff, rains or flood. Fipronil (FP) is a GABA receptor inhibitor, while buprofezin (BPFN) is an insect growth regulator. Presently, we exposed groups of aquaria acclimated carp fish (Cyprinus carpio) for 96h to sub-lethal concentrations of fipronil (400μgL(-1); 9.15×10(-7)molL(-1)) and buprofezin (BPFN, 100mgL(-1); 1.072×10(-6)molL(-1)) singly or in combination. The extent of damage was assessed at biochemical, hematological, molecular biological and histopathological level. Results obtained in treated fish were compared statistically with those of control non-treated fish and also among treatment groups. Significance level was p<0.05. Compared to control, serum total protein and globulin concentrations decreased significantly (p<0.0001) in fish treated with FP; while albumin concentration remained unaltered with all treatments. Glucose concentration decreased significantly (p<0.002) in fish treated with FP. In contrast, combined FP+BPFN treatment and BPFN treatment caused insignificant elevation of glucose concentration. Hematological assessment demonstrated significant decrease in red blood cell and thrombocyte counts, hemoglobin concentration and hematocrit percent; while white blood cell count showed an increase in all treatment groups (p<0.0001). Blood smears from pesticide treated fish revealed aberrant erythrocyte morphologies which included necrosis, micronuclear formation and hyperchromatosis. DNA laddering assay carried out on whole blood demonstrated excessive smear formation in combined FP+BPFN and BPFN treatment groups but no smear formation was noticeable in FP treated fish. Compared to control, whole blood DNA content increased significantly in the combined FP+BPFN and BPFN treatment groups (p<0.001 and p<0.009). With all treatments histopathological changes observed in the gills were: epithelial uplifting and necrosis of lamellae, lamellar atrophy, disruption of cartilaginous core, fusion and disorganization of lamellae and telangiectasia. In liver these were: karyorrhexis, hepatocellular hypertrophy, nuclear hypertrophy, melanomacrophage aggregates and central vein contraction, while in the kidney: deterioration of glomerulus and dilatation of Bowman's space, dilatation of renal tubules, thyroidisation, altered tubular lumen, nuclear hypertrophy, cellular atrophy, and cellular necrosis were the outcome. Our study revealed that FP and BPFN produce highly toxic effects on fish when given in combination or singly. To our knowledge, this is the first report on toxicity caused by FP and BPFN in single and combined state.

Abstract:Common carp (Cyprinus carpio), a valuable aquaculture species susceptible to various infections, requires effective immune enhancement strategies. This study investigates the immunomodulatory effects of orally administered terpenoids and phenol fraction (TPF) from Padina gymnospora in C. carpio, focusing on stimulation of nonspecific immune response, immune gene expression, and protection against experimental infection. P. gymnospora is a brown seaweed species known for its bioactive compounds and medicinal properties. TPF was extracted using the Harborne fractionation method, and the presence of terpenoids and phenol compounds was confirmed by qualitative analysis and high-performance thin layer chromatography (HPTLC). TPF was administered orally in different doses to carp. Nonspecific immune responses were evaluated by measuring cellular ROS, RNI, and peroxidase production. The expression of immune genes (lysozyme and interleukin-1β) was assessed by reverse transcriptase PCR. Furthermore, the protective efficacy of TPF was determined by infecting carp with a virulent pathogen, Aeromonas hydrophila, and monitoring mortality rates and disease symptoms. The results demonstrate that oral TPF administration significantly enhances nonspecific immune responses, with increased ROS, RNI, and peroxidase production, indicating improved immune function. Expression levels of lysozyme and interleukin-1β were upregulated, suggesting immune system activation. Moreover, TPF exhibited significant protection against experimental infection, with lower mortality rates compared to the control group. These findings highlight TPF's potential as an effective immunostimulatory agent, enhancing immune responses and providing infection protection in carp. In conclusion, oral TPF administration stimulates nonspecific immune responses, modulates immune gene expression, and confers protection against experimental infection in carp, displaying its potential for enhancing immune responses and disease resistance in aquaculture species, and contributing to sustainable fish health management.
