Abstract:Seeds represent essential stages of the plant life cycle: embryogenesis, the intermittent quiescence phase and germination. Each stage has its own physiological requirements, genetic program and environmental challenges. Consequently, the effects of developmental and environmental hypoxia can vary from detrimental to beneficial. Past and recent evidence shows how low-oxygen signalling and metabolic adaptations to hypoxia affect seed development and germination. Here, we review the recent literature on seed biology in relation to hypoxia research, and present our perspective on key challenges and opportunities for future investigations.

Abstract:Seed dormancy in rice is an important trait related to the pre-harvest sprouting resistance. In order to understand the molecular mechanisms of seed dormancy, gene expression was investigated by transcriptome analysis using seeds of the strongly dormant cultivar N22 and its less dormant mutants Q4359 and Q4646 at 24 days after heading (DAH). Microarray data revealed more differentially expressed genes in Q4359 than in Q4646 compared to N22. Most genes differing between Q4646 and N22 also differed between Q4359 and N22. GO analysis of genes differentially expressed in both Q4359 and Q4646 revealed that some genes such as those for starch biosynthesis were repressed, whereas metabolic genes such as those for carbohydrate metabolism were enhanced in Q4359 and Q4646 seeds relative to N22. Expression of some genes involved in cell redox homeostasis and chromatin remodeling differed significantly only between Q4359 and N22. The results suggested a close correlation between cell redox homeostasis, chromatin remodeling and seed dormancy. In addition, some genes involved in ABA signaling were down-regulated, and several genes involved in GA biosynthesis and signaling were up-regulated. These observations suggest that reduced seed dormancy in Q4359 was regulated by ABA-GA antagonism. A few differentially expressed genes were located in the regions containing qSdn-1 and qSdn-5 suggesting that they could be candidate genes underlying seed dormancy. Our work provides useful leads to further determine the underling mechanisms of seed dormancy and for cloning seed dormancy genes from N22.
Metabolic strategies in hypoxic plants

Abstract:Complex multicellular organisms have evolved in an oxygen-enriched atmosphere. Oxygen is therefore essential for all aerobic organisms including plants, for energy production through cellular respiration. However, plants can experience hypoxia following extreme flooding events and also under aerated conditions in proliferative organs or tissues characterized by high oxygen consumption. When oxygen availability is compromised, plants adopt different strategies to cope with hypoxia and limited aeration. A common feature among different plant species is the activation of an anaerobic fermentative metabolism to provide adenosine triphosphate (ATP) to maintain cellular homeostasis under hypoxia. Fermentation also requires many sugar substrates, which is not always feasible, and alternative metabolic strategies are thus needed. Recent findings have also shown that the hypoxic metabolism is also active in specific organs or tissues of the plant under aerated conditions. Here, we describe the regulatory mechanisms that control the metabolic strategies of plants and how they enable them to thrive despite challenging conditions. A comprehensive mechanistic understanding of the genetic and physiological components underlying hypoxic metabolism should help to provide opportunities to improve plant resilience under the current climate change scenario.

Abstract:Reduced oxygen availability (hypoxia) represents a key plant abiotic stress in natural and agricultural systems, but conversely it is also an important component of normal growth and development. We review recent advances that demonstrate how genetic adaptations associated with hypoxia impact the known plant oxygen sensing mechanism through the PLANT CYSTEINE OXIDASE (PCO) N-degron pathway. Only three protein substrates of this pathway have been identified, and all adaptations identified to date are associated with the most important of these, the group VII ETHYLENE RESPONSE FACTOR transcription factors (ERFVIIs). We discuss how geography, altitude, and agriculture have all shaped molecular responses to hypoxia, and how these responses have emerged at different taxonomic levels through the evolution of land plants. Understanding how ecological and agricultural genetic variation acts positively to enhance hypoxia tolerance will provide novel tools and concepts to improve the performance of crops in the face of increasing extreme flooding events.

Abstract:In higher plants, seed is a propagule which ensures dissemination and survival of species. Developmental phases of a seed comprise embryogenesis, maturation and germination paving a way to its final fate i.e. seedling establishment. The final stage of seed maturation is marked by dehydration, acquisition of dessication tolerance and induction of dormancy. A precise Abscisic acid (ABA) to Gibberellins (GA) ratio, accumulation of miRNA 156, low level of reactive oxygen species (ROS) and enzyme inactivity govern seed dormancy. This also prevent pre harvest sprouting of the seeds. Overtime, stored seed mRNAs and proteins are degraded through oxidation of specific nucleotides in response to ROS accumulation. This degradation alleviates seed dormancy and transforms a dormant seed into a germinating seed. At this stage, ABA catabolism and degradation accompanied by GA synthesis contribute to low ABA to GA ratio. GA as well as ROS acts downstream, to mobilize reserve food materials, rupture testa, enhance imbibition and protrude radicle. All these events mark seed germination. Further, seedling is established under the governance of auxin and light. ABA and GA are master regulators while auxin, cytokinins, ethylene, jasmonic acid, brassinosteroids act through interdependent pathways to tightly regulate seed dormancy, germination and seedling establishment. In this review, the role of phytohormones and ROS in accordance with environmental factors in governing seed dormancy, promoting seed germination and thus, establishing a seedling is discussed in detail.

Abstract:Abstract The breaking of dormancy mediated by reactive nitrogen species (RNS) is related to the accumulation of reactive oxygen species (ROS) in germinating embryos but the underlying mechanism is unclear. The objectives of this study were: (1) to explore the relationship between RNS-mediated dormancy release and ROS accumulation in germinating embryos of Sorbus pohuashanensis ; and, (2) to investigate the relationships among germination time, ROS metabolism, and endogenous hormone synthesis. We studied the effects of exogenous nitric oxide (NO) donor sodium nitroprusside (SNP), the NO scavenger (4-carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide (PTIO), abscisic acid (ABA), the exogenous ethylene donor ethrel, and the ethylene receptor inhibitor 2,5-norbornadien (NBD) on embryo germination and seedling growth. Embryos were released from dormancy by pretreatment with NO or ethylene, which was related to increased ethylene biosynthesis and decreased ABA levels. Breaking of dormancy by SNP was related to increased levels of ethylene, hydrogen peroxide, and glutathione, increased activities of superoxide dismutase and glutathione peroxidase, and decreased levels of ABA, superoxide anions, and malondialdehyde. These effects of nitric oxide were especially significant in seedling hypocotyls and radicles. These results demonstrate that NO can break S. pohuashanensis embryo dormancy by inducing ethylene biosynthesis, and that this signalling pathway is closely related to ROS accumulation and the antioxidant defence response.

Abstract:1. Many internal (inherent) and environmental (imposed) factors control seed dormancy and germination that we divide into three basic dormancy‐release pathways: Maternal structures and embryo physiology control inherent dormancy that is broken by various types of scarification and physiological changes, followed by imposed‐dormancy release when the prevailing environment is replaced by certain 'standard' conditions that stimulate germination (Pathway 1); imposed dormancy prevails even if inherent dormancy is broken or not applicable that is released when replaced by certain 'standard' environmental conditions which stimulate germination (Pathway 2); release from inherent dormancy by light/dark or cold stratification is contingent on the pre‐existence of certain environmental conditions that stimulate germination (Pathway 3). On‐plant seed storage (serotiny) and frugivorous seeds are recognized here as representing special types of physical dormancy, as their properties are consistent with those of hard diaspores. Warm stratification does not require seeds to be moist as it is just a physical response. Heat may promote germination of non‐hard, as well as hard, seeds as it may increase their permeability further. 3. Levels of germination gauge the net effect of inherent‐ and imposed‐dormancy release so that it is only possible to identify the extent of inherent‐dormancy release when conditions for germination are optimal (imposed dormancy has been annulled). While imposed dormancy may be protracted after inherent dormancy is broken by heat or chilling during the dry or cold seasons, release from both states may effectively coincide if smoke chemicals or light are received during the (wet) growing season. 4. We suggest reserving the term secondary dormancy for seeds that return to (inherent or imposed) dormancy due to changed environmental conditions. Under seasonal climates, fluctuations in environmental conditions can lead to secondary dormancy and even dormancy cycling. 5. We recognize four types of functional interactions between any two environmental factors that induce inherent‐dormancy release: binary interactions are ineffective, only one stimulus is effective, both are effective but non‐additive, or both are additive/synergistic. Two environmental stimuli that individually break dormancy but have no additive effect must be affecting the same process; this was demonstrated here for some interactions between heat and smoke. 6. The three dormancy‐release pathways, together with internal, seasonal and stochastic interactions, are coordinated by the non‐dormant seed to ensure maximum germination under optimal conditions. To ignore any aspect outlined here leads to an impoverished understanding of the disparate seed ecology of species adapted to different stressful and disturbance‐prone habitats.

Abstract:From 50 to 90% of wild plant species worldwide produce seeds that are dormant upon maturity, with specific dormancy traits driven by species' occurrence geography, growth form, and genetic factors. While dormancy is a beneficial adaptation for intact natural systems, it can limit plant recruitment in restoration scenarios because seeds may take several seasons to lose dormancy and consequently show low or erratic germination. During this time, seed predation, weed competition, soil erosion, and seed viability loss can lead to plant re‐establishment failure. Understanding and considering seed dormancy and germination traits in restoration planning are thus critical to ensuring effective seed management and seed use efficiency. There are five known dormancy classes (physiological, physical, combinational, morphological, and morphophysiological), each requiring specific cues to alleviate dormancy and enable germination. The dormancy status of a seed can be determined through a series of simple steps that account for initial seed quality and assess germination across a range of environmental conditions. In this article, we outline the steps of the dormancy classification process and the various corresponding methodologies for ex situ dormancy alleviation. We also highlight the importance of record‐keeping and reporting of seed accession information (e.g. geographic coordinates of the seed collection location, cleaning and quality information, storage conditions, and dormancy testing data) to ensure that these factors are adequately considered in restoration planning.This article is protected by copyright. Abstract:Oxygen (O 2 ) is an inhibiting factor for plant growth and development in submerged and flooding environments. Plants experience different O 2 concentrations, such as normoxia, hypoxia, and anoxia, which can change over space and time. Plants have evolved various morphological, physiological, and biochemical adaptations to withstand low O 2 stress, many of which have been well investigated. This review provides a detailed analysis of how plants respond to hypoxia, a significant stress factor primarily caused by flooding. Hypoxia affects plants at various cellular, developmental, and environmental levels. This review highlights genetic, molecular, and metabolic adaptations crops employ to cope with O 2 deficiency. The roles of various transcription factors (TFs) and gene regulation mechanisms in enabling plants to modulate their physiological responses under hypoxic conditions are notable. The review also identifies a significant gap in research on plant responses during reoxygenation, the phase of returning to normal O 2 levels, especially under natural lighting conditions. This transition poses ROS generation and photoinhibition challenges, affecting plant recovery post-hypoxia. We discuss various strategies to enhance plant hypoxia tolerance, including traditional breeding, genetic modification, and grafting techniques. It emphasizes integrating these approaches with a comprehensive understanding of hypoxia sensing and response mechanisms. We underscore the complexity of plant adaptations to hypoxia and the need for continued research in this field, especially in the face of global climate change. This is vital for developing sustainable agricultural practices and ensuring future food security.

Abstract:The seed represents a critical stage in the life cycle of flowering plants. It corresponds to a dry structure carrying the plant embryo in dormant or quiescent state. Orthodox seeds possess a very low water content, preventing biochemical reactions, especially respiration. If the desiccation of living organisms leads to a loss of homeostasis, structure, and metabolism, the seeds go through it successfully thanks to their structure, cellular organization, and growth regulation. Seeds set up a certain number of sophisticated molecules to protect valuable macromolecules or organelles from dehydration/rehydration cycles. Moreover, dormancy takes place in a coordinated process with environmental cues in order to ensure embryo development at the most appropriate conditions for the establishment of the new plant. Moreover, repair processes are programmed to be ready to operate to maximize germination success and seed longevity. This review focuses on the physiology of the seed as related to hydration forces, respiration, and biochemical reactions in the transition from thermodynamically undefined dry state to self-sustained living system. Such processes are of importance for basic knowledge of the regulation of metabolism of living organisms, but also for the control of germination in the context of climate change due to global warming.

Abstract:As sessile organisms, plants enter periods of dormancy in response to environmental stresses to ensure continued growth and reproduction in the future. During dormancy, plant growth is suppressed, adaptive/survival mechanisms are exerted, and stress tolerance increases over a prolonged period until the plants resume their development or reproduction under favorable conditions. In this review, we focus on seed dormancy and bud dormancy, which are critical for adaptation to fluctuating environmental conditions. We provide an overview of the physiological characteristics of both types of dormancy as well as the importance of the phytohormones abscisic acid and gibberellin for establishing and releasing dormancy, respectively. Additionally, recent epigenetic analyses have revealed that dormancy establishment and release are associated with the removal and deposition of histone modifications at the loci of key regulatory genes influencing phytohormone metabolism and signaling, including DELAY OF GERMINATION 1 and DORMANCY-ASSOCIATED MADS-box genes. We discuss our current understanding of the physiological and molecular mechanisms required to establish and release seed dormancy and bud dormancy, while also describing how environmental conditions control dormancy depth, with a focus on the effects of histone modifications.
New insights into the role of lipids in plant hypoxia responses

Abstract:In plants, hypoxia (low-oxygen stress) is induced by soil waterlogging or submergence and this major abiotic stress has detrimental effects on plant growth, development, distribution, and productivity. To survive low-oxygen stress, plants have evolved a set of morphological, physiological, and biochemical adaptations. These adaptations integrate metabolic acclimation and signaling networks allowing plants to endure or escape from low-oxygen environments by altering their metabolism and growth. Lipids are ubiquitously involved in regulating plant responses to hypoxia and post-hypoxic reoxygenation. In particular, the polyunsaturation of long-chain acyl-CoAs regulates hypoxia sensing in plants by modulating acyl-CoA-binding protein–Group VII ethylene response factor dynamics. Moreover, unsaturated very-long-chain ceramide species protect plants from hypoxia-induced cellular damage by regulating the kinase activity of CONSTITUTIVE TRIPLE RESPONSE1 in the ethylene signaling pathway. Finally, the oxylipin jasmonate specifically regulates plant responses to reoxygenation stress by transcriptionally modulating antioxidant biosynthesis. Here we provide an overview of the roles of lipid remodeling and signaling in plant responses to hypoxia/reoxygenation and their effects on the downstream events affecting plant survival. In addition, we highlight the key remaining challenges in this important field.

The signalling role of ROS in the regulation of seed germination and dormancy.

Abstract:Reactive oxygen species (ROS) are versatile compounds which can have toxic or signalling effects in a wide range living organisms, including seeds. They have been reported to play a pivotal role in the regulation of seed germination and dormancy but their mechanisms of action are still far from being fully understood. In this review, we sum-up the major findings that have been carried out this last decade in this field of research and which altogether shed a new light on the signalling roles of ROS in seed physiology. ROS participate in dormancy release during seed dry storage through the direct oxidation of a subset of biomolecules. During seed imbibition, the controlled generation of ROS is involved in the perception and transduction of environmental conditions that control germination. When these conditions are permissive for germination, ROS levels are maintained at a level which triggers cellular events associated with germination, such as hormone signalling. Here we propose that the spatiotemporal regulation of ROS production acts in concert with hormone signalling to regulate the cellular events involved in cell expansion associated with germination.

Production of Microcrystalline Cellulose by Reactive Extrusion Production of Microcrystalline Cellulose by Reactive Extrusion

Abstract:This process involves feeding cellulose into an extruder with an acid solution. The extruder is comprised of a screw and a barrel. The screw is rotated so as to pressurize the cellulose, and the cellulose undergoes acid hydrolysis and forms microcrystalline cellulose. The invented process can be accomplished by using pure cellulose or a lignocellulosic material as the starting material. If a lignocellulosic material is used, then a basic solution is added to it and the lignocellulosic material is fed through an extruder so as to obtain a mixture comprising lignin, hemicellulose and cellulose. The lignin and hemicellulose are extracted and the remaining cellulose is processed, as discussed previously, to form microcrystalline cellulose.

Abstract:Reactive oxygen species (ROS) play an essential role in the regulation of seed dormancy, germination, and deterioration in plants. The low level of ROS as signaling particles promotes dormancy release and triggers seed germination. Excessive ROS accumulation causes seed deterioration during seed storage. Maintaining ROS homeostasis plays a central role in the regulation of seed dormancy, germination, and deterioration in crops. This study highlights the current advances in the regulation of ROS homeostasis in dry and hydrated seeds of crops. The research progress in the crosstalk between ROS and hormones involved in the regulation of seed dormancy and germination in crops is mainly summarized. The current understandings of ROS-induced seed deterioration are reviewed. These understandings of ROS-dependent regulation on seed dormancy, germination, and deterioration contribute to the improvement of seed quality of crops in the future.

Abstract:Quiescence is an essential property of meristematic cells, which restrains the cell cycle while retaining the capacity to divide. This crucial process not only facilitates life-long tissue homeostasis and regenerative capacity but also provides protection against adverse environmental conditions, enabling cells to conserve the proliferative potency while minimising DNA damage. As a survival attribute, quiescence is inherently regulated by the products of aerobic life, in particular reactive oxygen species (ROS) and the redox (reduction/ oxidation) mechanisms that plant have evolved to channel these into pervasive signals. Adaptive responses allow quiescent cells to compensate for reduced oxygen tension (hypoxia) in a reversible manner, while the regulated production of the superoxide anion (O 2 − ) facilitates cell division and the maintenance of stem cells. Here we discuss the role of ROS and redox reactions in the control of the quiescent state in plant meristems, and how this process is integrated with cellular energy and hormone biochemistry. We consider the pathways that sense and transmit redox signals with a focus on the central significance of redox regulation in the mitochondria and nucleus, which is a major regulator of quiescence in meristems. We discuss recent studies that suggest ROS are a critical component of the feedback loops that control stem cell identity and fate and suggest that the ROS/hypoxia interface is an important "outside/ in" positional cue for plant cells, particularly in meristems.

Abstract:Seeds are unique time capsules that can switch between 2 complex and highly interlinked stages: seed dormancy and germination. Dormancy contributes to the survival of plants because it allows to delay germination to optimal conditions. The switch between dormancy and germination occurs in response to developmental and environmental cues. In this review we provide a comprehensive overview of studies that have helped to unravel the molecular mechanisms underlying dormancy and germination over the last decades. Genetic and physiological studies provided a strong foundation for this field of research and revealed the critical role of the plant hormones abscisic acid and gibberellins in the regulation of dormancy and germination, and later natural variation studies together with quantitative genetics identified previously unknown genetic components that control these processes. Omics technologies like transcriptome, proteome, and translatomics analysis allowed us to mechanistically dissect these processes and identify new components in the regulation of seed dormancy and germination.

Abstract:Two major stages of seed development are dormancy and germination which finally promotes the growth of a plant. Some internal and external factors such as hormonal, genetic, chromatin development and environmental factors which maintain the seed dormancy with passing time and other suitable factors, these dormancy promoters are gradually decreased causing release of dormancy and promoting germination by the mechanisms of ROS in plant signalling, cell elongation and reverse mobilization. But dormancy has some benefits in protecting the seed from extreme condition even after natural disaster as well as serving as food for predators in order to maintain balance of nature. So, dormancy can be inhibited by some phytochemical components like terpenoids, polyphenoliic compounds, flavonoids, alkaloids and glycosides by the mechanism of inhibiting water uptake system III & II, surface sterilization, reverse mobilization, cell elongation etc.

Oxygen dependency of germinating Brassica seeds

Abstract:Establishing plants in space, Moon or Mars requires adaptation to altered conditions, including reduced pressure and composition of atmospheres. To determine the oxygen requirements for seed germination, we imbibed Brassica rapa seeds under varying oxygen concentrations and profiled the transcription patterns of genes related to early metabolism such as starch degradation, glycolysis, and fermentation. We also analyzed the activity of lactate dehydrogenase (LDH) and alcohol dehydrogenase (ADH), and measured starch degradation. Partial oxygen pressure (pO2) greater than 10% resulted in normal germination (i.e., protrusion of radicle about 18 hours after imbibition) but lower pO2 delayed and reduced germination. Imbibition in an oxygen-free atmosphere for three days resulted in no germination but subsequent transfer to air initiated germination in 75% of the seeds and the root growth rate was transiently greater than in roots germinated under ambient pO2. In hypoxic seeds soluble sugars degraded faster but the content of starch after 24 h was higher than at ambient oxygen. Transcription of genes related to starch degradation, α-amylase (AMY) and Sucrose Synthase (SUS), was higher under ambient O2 than under hypoxia. Glycolysis and fermentation pathway-related genes, glucose phosphate isomerase (GPI), 6-phosphofructokinase (PFK), fructose 1,6-bisphosphate aldolase (ALD), glyceraldehyde-3-phosphate dehydrogenase (GAPDH), pyruvate decarboxylase (PDC), LDH, and ADH, were induced by low pO2. The activity of LDH and ADH was the highest in anoxic seeds. Germination under low O2 conditions initiated ethanolic fermentation. Therefore, sufficient oxygen availability is important for germination before photosynthesis provides necessary oxygen and the determination of an oxygen carrying capacity is important for uniform growth in space conditions.

Abstract:Dormancy is an adaptive trait that blocks seed germination until the environmental conditions become favorable for subsequent vegetative plant growth. Seed dormancy is defined as the inability to germinate in favorable conditions. Dormancy is alleviated during after-ripening, a dry storage period, during which dormant (D) seeds unable to germinate become non-dormant (ND), able to germinate in a wide range of environmental conditions. The treatment of dormant seeds with ethylene (D/ET) promotes seed germination, and abscisic acid (ABA) treatment reduces non-dormant (ND/ABA) seed germination in sunflowers (Helianthus annuus). Metabolomic and transcriptomic studies have been performed during imbibition to compare germinating seeds (ND and D/ET) and low-germinating seeds (D and ND/ABA). A PCA analysis of the metabolites content showed that imbibition did not trigger a significant change during the first hours (3 and 15 h). The metabolic changes associated with germination capacity occurred at 24 h and were related to hexoses, as their content was higher in ND and D/ET and was reduced by ABA treatment. At the transcriptional level, a large number of genes were altered oppositely in germinating, compared to the low-germinating seeds. The metabolomic and transcriptomic results were integrated in the interpretation of the processes involved in germination. Our results show that ethylene treatment triggers molecular changes comparable to that of after-ripening treatment, concerning sugar metabolism and ABA signaling inhibition.
