Abstract:In recent years, multi-component therapies are increasingly utilized to treat complex diseases such as cancer, diabetes, and other chronic complex diseases. Here, we proposed the protocol for rational design of drug combination with poly-pharmacological effects by integration of design of experiments (DOE), computational modeling, and multiple-objective optimization algorithm. Here, we introduce a common workflow for modeling quantitative relationship of chemical composition and multiple activities of drug combinations. As an example, anti-oxidation, neuroprotective, and anti-platelet activities of three different salvia polyphenols were measured, which were mathematically represented by multivariant regression models to evaluate the effect of combination. In validation, the optimized combination which obtained by weighed-sum method showed good activities in all three models. Our results demonstrate that the multiple-objective optimization strategy is suitable to optimize the ratio of the compounds so to induce the best therapeutic action.
Prescription Function Prediction Using Topic Model and Multilabel Classifiers.

Abstract:Determining a prescription's function is one of the challenging problems in Traditional Chinese Medicine (TCM). In past decades, TCM has been widely researched through various methods in computer science, but none concentrates on the prediction method for a new prescription's function. In this study, two methods are presented concerning this issue. The first method is based on a novel supervised topic model named Label-Prescription-Herb (LPH), which incorporates herb-herb compatibility rules into learning process. The second method is based on multilabel classifiers built by TFIDF features and herbal attribute features. Experiments undertaken reveal that both methods perform well, but the multilabel classifiers slightly outperform LPH-based method. The prediction results can provide valuable information for new prescription discovery before clinical test.

Abstract:Multicomponent therapeutic has become an increasingly favored strategy for treating complex diseases in recent years. In this study, a multiple objective optimization approach was proposed to design the optimal combination of three components for antiplatelet activity. The platelet aggregation assays induced by three different ways, adenosine diphosphate, arachidonic acid, and collagen, were applied to evaluate the in vitro antiplatelet activities of three active components derived from a traditional Chinese medicine. After analyzing this dataset by quantitative composition-activity relationship modeling, a weighted-sum optimization method was adopted to calculate the optimal ratio between three components for antiplatelet effects. Further experiments validated our method and showed that better antiplatelet activity was exerted by the optimized combination than the individual component or other combinations. Our findings suggested that the proposed multiobjective optimization approach is a novel method for multicomponent drug design.
Recovery Prediction of Danshensu, Salvianolic Acid B, and Saivianolic Acid D in Danshen Ethanol Precipitation

Abstract:Objective To develop recovery prediction models for danshensu,salvianolic acid B,and salvianolic acid D in danshen ethanol precipitation.Methods Recorded the operation parameters of 15 normal batches of danshen ethanol precipitation in one Chinese medicine plant and measured five active component contents in danshen concentrated liquid and danshen ethanol precipitated liquid,the Stepwise-MLR recovery prediction models of dashensu,salvianolic acid B,and salvianolic acid D were developed based on an approach which combined the process parameters and the active component contents in danshen concentrated liquid.The importance of variables in the prediction models was analyzed later.Results The coefficient correlations of dashensu,salvianolic acid B,and salvianolic acid D recovery prediction models were all above 0.95.Conclusion The models developed for active component recovery prediction show a good predictability.Models can be used to help manufacturers to understand the ethanol precipitation process and improve the quality control ability of danshen ethanol precipitation.
[Preparation of personalized traditional Chinese medicine condensed water pill without excipients based on correlation between concentrate viscosity and molding quality]

Abstract:To explore the correlation between concentrate viscosity and molding quality of personalized traditional Chinese medicine(TCM) condensed water pill, this study established a concentrate viscosity characterization method with rotational rheometry. Seven model prescriptions were respectively concentrated to different degrees and the viscosity of each concentrate was determined. The pre-sence of 'viscosity jump' in the middle stage of 'flag hanging' of all the model prescriptions implied that there might be an ideal viscosity range in the preparation of condensed water pill. The further study of 22 model prescriptions demonstrated that the optimum viscosity range of concentrate was 5-15 Pa·s(25 ℃) for approximately 82% of the prescriptions. About 18% of the prescriptions had a wide range, which might be caused by the high proportions of mineral and crustacean drugs in the crushing part and sugar and fibrous drugs in the decocting part. This study clarified the optimum viscosity range for concentrates of personalized TCM condensed water pills and achieved a preparation technology without any excipient, laying a foundation for the on-line control of the preparation.

Abstract:The purpose of this study was to optimize the formulation of tea catechin-loaded nano-liposomes using response surface methodology. Response surface methodology based on central composite rotatable design has been successfully used to model and optimize biochemical and biotechnological processes. The mass ratio of phosphatidylcholine and cholesterol (1-3), catechin concentration (3-5 mg/mL), pH values of phosphate buffer solution (6-7) and the volume ratio of organic phase and aqueous phase (2-4) were selected as independent variables with encapsulation efficiency and particle size as dependent variables. For each response, a second-order polynomial model was developed using multiple linear regression analysis. Applying a desirability function method the optimum parameters were: phosphatidylcholine to cholesterol mass ratio of 2.17, catechin concentration of 5 mg/mL, pH values of phosphate buffer solution of 6.62 and organic phase to aqueous phase volume ratio of 3.05. At this optimum point, particle size and encapsulation efficiency were found to be 220 nm and 60.18%, respectively. Furthermore, leakage ratio of nano-liposomes was used to determine the influence of storage period.

Abstract:To control the risks of powder caking and capsule shell embrittlement of Guizhi Fuling Capsules, a predictive model for hygroscopicity of contents in Guizhi Fuling Capsules was built. A total of 90 batches of samples, including raw materials, intermediate powders and capsules, were collected during the manufacturing of Guizhi Fuling Capsules. According to the production sequence, 47 batches were used as the calibration set, and the properties of raw materials and the four intermediate powders were comprehensively characterized by the physical fingerprint. Then, the partial least squares(PLS) model was developed with the content hygroscopicity as the response variable. The variable importance in projection(VIP), variance inflation factor(VIF) and regression coefficients were used to screen out potential critical material attributes(pCMAs). As a result, five pCMAs from 54 physical parameters were screened out. Furthermore, different models were built by different combinations of pCMAs, and their predictive robustness of 43 batches was evaluated on the basis of the validation set. Finally, the tap density(D_c) of wet granules obtained from wet granulation and the angle of repose(α) of raw materials were identified as the critical material attributes(CMAs) affecting the hygroscopicity of the contents of Guizhi Fuling Capsules. The prediction model established with the two CMAs as independent variables had an average relative prediction error of 2.68% for samples in the validation set, indicating a good accuracy of prediction. This paper proved the feasibility of predictive modeling toward the control of critical quality attributes of Chinese medicine oral solid dosage(OSD). The combination of the continuous quality improvement, the industrial big data and the process modeling technique paved the way for the intelligent manufacturing of Chinese medicine oral solid preparations.

Abstract:This paper deals with the influence of network topological changes on the resistive losses for a significant period. It is based on a previous work [1] where the goal was to reduce power losses on a peak load situation. The second part of the work, presented here, attempts to reduce the energy losses of transmission network by implementing a cyclic search of topology changes and phase shifting adjustments over a week time period. As the whole problem for such a long period is not tractable in a Mixed Integer Non-Linear Programming approach, a piecewise-linear formulation is presented in order to find a convenient relation between active losses and real flows for each branch. Subsequently, a mixed integer linear formulation is developed to find an optimal topology that tends to minimize the active losses. Considered topological configurations concern the splitting or merging of electric buses and the line switching. Once the new topology has been identified, the interactions between real and reactive power are handled to adjust the taps of phase shifting transformers through a Mathematical Program with Equilibrium Constraints (MPEC). A practical example based on several hundreds of observations (snapshots) of the French national grid is presented. Obtained results confirm the interest of the method.

Abstract:This paper proposed an approach to predict the breakthrough curves for herbal medicine adsorption. The saponins adsorption process was taken as an example. The Yoon-Nelson model was used to fit the breakthrough curves of saponins and the model parameters in different batches were obtained. The model parameters were correlated with saponin concentrations in loading solution and flow rate using polynomial regression models. According to the polynomial regression models, the breakthrough curves could be predicted and the optimal adsorption end-point could be calculated early-on. This research provided a new method for predicting the end-points of the adsorption process of herbal extracts.

Abstract:Natural medicines are now gaining increasing acceptance around the global scope. Due to the complexity of composition, it is imperative to adopt a unique evaluation system, which is not confined to the content of finite marker compounds. This research presents the successful application of pharmacodynamics-based optimization to the ultrasound-assisted extraction of Chansu, a widely used natural medicine with complex bioactive ingredients. The effect of solvent concentration, solvent to solid ratio, extraction time and particle size, on two response indexes (extraction yield of total content of bufalin, cinobufagin and resibufogenin and anticancer activity) were evaluated by response surface methodology (RSM). Two models were built based on two response indexes, and both of them provided accurate predictions. Compared with extraction yield-based optimization, pharmacodynamics-based optimization aligns well with the integrity and complexity of natural medicines. Moreover, the pharmacodynamics-based optimization gives a more economical and efficient optimal condition, which is specifically embodied in the coarser requirement for particle size and the saving of extraction time and solvent. The recommended extraction conditions were methanol concentration of 83.54% (v/v), solvent to solid ratio of 26.23 mL/g, extraction time of 40.43 min and particle size of 61 mesh (0.212-0.25 mm). The present study elaborated the feasibility and implication of pharmacodynamics-based optimization, which provides useful information for further research on the extraction process of natural medicines.
Refining the rheological characteristics of high drug loading ointment via SDS and machine learning

Abstract:This paper presents an optimized preparation process for external ointment using the Definitive Screening Design (DSD) method. The ointment is a Traditional Chinese Medicine (TCM) formula developed by Professor WYH, a renowned TCM practitioner in Jiangsu Province, China, known for its proven clinical efficacy. In this study, a stepwise regression model was employed to analyze the relationship between key process factors (such as mixing speed and time) and rheological parameters. Machine learning techniques, including Monte Carlo simulation, decision tree analysis, and Gaussian process, were used for parameter optimization. Through rigorous experimentation and verification, we have successfully identified the optimal preparation process for WYH ointment. The optimized parameters included drug ratio of 24.5%, mixing time of 8 min, mixing speed of 1175 rpm, petroleum dosage of 79 g, liquid paraffin dosage of 6.7 g. The final ointment formulation was prepared using method B. This research not only contributes to the optimization of the WYH ointment preparation process but also provides valuable insights and practical guidance for designing the preparation processes of other TCM ointments. This advanced DSD method enhances the screening approach for identifying the best preparation process, thereby improving the scientific rigor and quality of TCM ointment preparation processes.

Comparison of several variable selection methods for quantitative analysis and monitoring of the Yangxinshi tablet process using near-infrared spectroscopy

Abstract:Variable selection methods can simplify the modelling process and improve the accuracy of models constructed for rapid monitoring using near-infrared (NIR) spectroscopy. In this study, NIR spectroscopy was applied in combination with chemometrics to determine the icariin, salvianolic acid B, and puerarin content, as well as the relative density through the concentration process of a Yangxinshi tablet (YXST) water extraction solution. Partial least squares with several variable selection algorithms were used for the modelling. Variable selection methods, including synergy interval partial least squares (Si-PLS), a genetic algorithm, competitive adaptive reweighted sampling (CARS), and their combination, were comparatively applied to calibrate the regression model. The performances of the models obtained were systematically evaluated according to the correlation coefficients of prediction (Rp), the relative standard error of prediction (RSEP), and the residual predictive deviation (RPD). For icariin, salvianolic acid B, and puerarin, the Si-PLS model showed optimal results. The Rp, RSEP, and RPD of Si-PLS were 0.9905, 7.84%, and 7.38 for icariin; 0.9781, 9.48%, and 4.80 for salvianolic acid B; and 0.9668, 12.81%, and 3.92 for puerarin, respectively. For the relative density, the CARS-Si-PLS acquired the best prediction results. The Rp, RSEP, and RPD of CARS-Si-PLS were 0.9725, 0.47%, and 4.31 for the relative density, respectively. This study demonstrated that variable selection methods can simplify the modelling process and improve the accuracy of the models. In addition, the methods developed in this study are suitable for a quality monitoring of the concentration process of YXST water extraction solution, and they provide reference for real-time monitoring of the concentration processes used in the preparation of other types of traditional Chinese medicine.
Analytical Quality by Design Oriented Development of the UPLC Method for Analysing Multiple Pharmaceutical Process Intermediates: A Case Study of Compound Danshen Dripping Pills

Abstract:This work presented the integration of the Analytical Quality by Design concept in developing a UPLC method for the quantification of saponins and phenolic acids in two Compound Danshen Dripping Pills intermediates of a concentrated extract and a supernatant. Prior knowledge and systematic risk analysis were conducted to identify potential critical method parameters (CMPs) that affected critical quality attributes (CQAs). A definitive screening design was employed to investigate the quantitative relationship among the intermediate type, CPPs, and CQAs. Intermediate type was added in the experimental design table as a categorical variable. After the models were built, the method operable design region (MODR) was calculated with an exhaustive search-Monte Carlo method. The CPP combinations with a 95 % or higher probability of reaching the preset standard were collected in MODR. The MODR robustness test was conducted with a Plackett-Burman design. The separation of analytes was achieved with a Waters Acquity HSS T3 (100 x 2.1 mm, 1.8 mu m) column at 30 degrees C, using 0.05 % phosphoric acid and acetonitrile as the mobile phases in gradient elution at a flow rate of 0.40 mL/min. The elution was monitored at UV detection wavelengths of 280 nm and 203 nm. The optimized method was also validated. Good robustness, accuracy, precision, stability and repeatability, and linear regression (R > 0.9997) were obtained. All recoveries of the 9 compounds ranged from 97.8 % to 101.7 %, with an RSD of less than 3.49 %. Nine batches of Danshensu, protocatechualdehyde, rosmarinic acid, salvianolic acid B, salvianolic acid A, notoginsenoside R1, ginsenoside Rg1, ginsenoside Re, and ginsenoside Rb1 were analysed. Danshensu was found to be the compound with the highest content.
Optimization of membrane dispersion ethanol precipitation process with a set of temperature control improved equipment.

Abstract:Ethanol precipitation is an important separation and purification process in the traditional Chinese medicines (TCMs) industry. In the present study, a membrane dispersion micromixer was applied to achieve good mixing for the ethanol precipitation process of Astragali radix concentrate. New experimental apparatus was set up to rapidly lower the temperature of ethanol solution before mixing with the concentrate. Ethanol precipitation process was optimized according to Quality by design concept. To identify critical material attributes (CMAs), ten batches of Astragali radix were used to prepare concentrates. Calycosin-7-O-β-D-glucoside content, the sucrose content, and the electrical conductivity were found to be CMAs after the correlation analysis and stepwise regression modelling. Definitive screening design was used to investigate the relationships among critical process parameters, CMAs, and process critical quality attributes (CQAs). Quadratic models were developed and design space was calculated according to the probability of attaining process CQA standards. A material quality control strategy was proposed. High quality and low quality Astragali radix concentrates can be discriminated by the inequalities. Low quality Astragali radix concentrates should not be released for ethanol precipitation process directly. Verification experiment results indicated accurate models and reliable design space. The temperature control method and control strategy are promising for ethanol precipitation process of other TCMs or foods.
Uniform Design Method for Optimizing Proportion of Prescription of Xiaozheng

Abstract:Objective To optimize the proportion of the prescription of Xiaozheng by uniform design method.Methods The mice were divided into twelve groups,including seven groups treated with the prescriptions which contained rhizoma paridis total saponins,evodiamine and rhymchophylla total alkaloids with different proportion,three groups treated with single traditional Chinese medicine mentioned before,cyclophosphamide group and distilled water group.On the second day,each group was treated with relevant medicinal for fourteen days.The proportion was fixed using uniform design method.The inhibition rates of different formulations of prescription of Xiaozheng on murine xenograft was compared.Results The regression equation was Y=0.851 78-0.014 92×X2-0.000 390 07×X1X2-0.000 878 39×X1X3.The best proportion of the prescription for rhizoma paridis total saponins was 37.5mg and rhymchophylla total alkaloids was 12.5mg,and tumor inhibitory ration was 44%.The body weight of murine were increased significantly in the rhymchophylla total alkaloids group,from(25.12±2.36)g to(33.06±2.47)g,which was higher than that of distilled water group(P0.05).Conclusion Compatibility of medicines in prescription is the soul of traditional Chinese medicine theory.It's a courageous method to explore the law of prescription by the uniform design method.The optimized prescription could achieve the best effect.The optimized proportion of prescription using modern mathematical statistics method can promote the modernization of traditional Chinese medical research.

Abstract:Objective Using solid dispersion technique to prepare Silymarin Dropping Pill to accelerate dissolution and to improve bioavailability. A central composite design-response surface method was employed to select the optimum formulations. Methods Independent variables were Poloxamer 188 content and silymarin content, while dependent variables were disintegrating time and percent of silymarin dissoluted at a definite time. Linear, two and three order quadratic models were used to estimate the relationship between independent and dependent variables. Response surfaces were delineated according to best-fit mathematic models and optimum formulations were selected there from. Prediction was carried out through comparing the observed and predicted values. Results Three order quadratic equation was the best-fitted mathematic models to describe the relationship between dependent and independent variables, with a regression coefficient of 0.998. Bias between observed and predicted values of disintegrating time and dissolution percentage of optimum formulation dropping pill were negligible, indicating the high predictability of the fit models. Percent dissolution of Silymarin Dropping Pill at 60 min was 19 times as that of conventional tablets. Conclusion Dissolution speed of silymarin can be effectively improved through incorporating into dropping pills. It shows that the optimum mathematic model is highly predictive. The central composite design-response surface method can be fairly used in formulation screening.

Abstract:The aim of this study was to verity the feasibility of in-vitro dissolution behavior prediction using multi-source data fusion strategy, taken the prepared sinomenine hydrochloride (SH) sustained-release tablets as examples. In this work, 180 batches of mixed powders of SH sustained-release tablets were scanned using near-infrared (NIR) spectroscopy technology and Raman spectroscopy technology respectively. Tables were then prepared with experiment design, which consisted a 3-level full factorial design and a D-optimal design. After obtaining in-vitro dissolution profiles of SH sustained release tablets, the prediction models were developed using the partial least squares (PLS) regression algorithm based on NIR spectra or Raman spectra individually. Subsequently, the data fusion models were built based on optimal principal components (PCs) combined formulation variables and process parameters using PLS regression algorithm and back-propagation artificial neural network (BP-ANN) algorithm respectively. The results of PLS regression model established by the data fusion strategy showed better performance than prediction models built using spectra individually. The value of variable importance for projection (VIP) indicated that the content of hydroxypropyl methylcellulose (HPMC) was the main factor influencing the dissolution behavior of SH sustained release tablets. The results of the difference factor (f1) and similarity factor (f2), as well as the paired t-test for the parameters of the fitted equations showed there was no difference between the measured and predicted values of the dissolution profiles of the validation set samples. In summary, the data fusion strategy established in this paper can provide a solution for the quality evaluation of sustained release tablets in a rapid way.

Abstract:The work aims to explore the feasibility of Raman mapping in predicting the dissolution profiles of solid oral dosage form. In this study, N=36 batches of representative sinomenine hydrochloride sustained-release tablets were prepared, using a D-optimal design, to introduce adequate variability, and the Raman mapping data of each tablet were acquired. The partial least squares regression models were established using three kinds of different modes, named single point mode, average mode and multi-point mode, to predict the dissolution profiles based on Raman mapping data. The percent dissolutions at specific time points and the parameters of an exponential function, which was employed to fit the dissolution profiles, were predicted, and the accuracy and precision of prediction were tested. The results showed that the multi-point mode displayed the best accuracy and precision in the prediction of both the dissolutions at the specific time points and the function parameters. In summary, the established method based on Raman mapping avoids the shortcomings of traditional dissolution testing protocols, such as complex operation, time-consuming and high analysis cost, thus has great potential of application and popularization.
