Abstract:Zinc oxide nanoparticles (nano-ZnOs) are widely used and possess great potentials in agriculture and biomedicine. It is inevitable for human exposure to these nanoparticles. However, no study had been conducted to investigate the long term effects of nano-ZnOs. This study aimed at investigating effects of nano-ZnOs on development, zinc metabolism and biodistribution of minerals (Zn, Fe, Cu, and Mn) in mice from week 3 to 35. After the characteristics of nano-ZnOs were determined, they were added into the basal diet at 0, 50, 500 and 5000 mg/kg. Results indicated that added 50 and 500 mg/kg nano-ZnOs showed minimal toxicity. However, 5000 mg/kg nano-ZnOs significantly decreased body weight (from week 4 to 16) and increased the relative weights of the pancreas, brain and lung. Added 5000 mg/kg nano-ZnOs significantly increased the serum glutamic-pyruvic transaminase activity and zinc content, and significantly enhanced mRNA expression of zinc metabolism-related genes, including metallothionein 1(32.66 folds), metallothionein 2 (31.42 folds), ZIP8 (2.21folds), ZIP14 (2.45 folds), ZnT1 (4.76 folds), ZnT2 (6.19 folds) and ZnT4 (1.82 folds). The biodistribution determination showed that there was a significant accumulation of zinc in the liver, pancreas, kidney, and bones (tibia and fibula) after receiving 5000 mg/kg nano-ZnO diet, while no significant effects on Cu, Fe, and Mn levels, except for liver Fe content and pancreas Mn level. Our results demonstrated that long term exposure to 50 and 500 mg/kg nano-ZnO diets showed minimal toxicity. However, high dose of nano-ZnOs (5000 mg/kg) caused toxicity on development, and altered the zinc metabolism and biodistribution in mice.
Maternal oxidized soybean oil exposure in rats during lactation damages offspring kidneys via Nrf2/HO-1 and NF-κB signaling pathway

Abstract:Background: Cooking oil is an indispensable component of the human diet. However, oils usually undergo thermal oxidation. Oxidized soybean oil (OSO) has been shown to have detrimental effects on humans and has emerged as a root cause of many chronic diseases. The objective of this work was to evaluate the effects of puerpera exposure to OSO on kidney damage in the mother and offspring using lactating rats as an experimental model. Results: Pathological sections and ultrastructure showed that OSO exposure resulted in various levels of damage to lactating rats and their offspring. OSO induced oxidative stress in the kidneys of lactating rats, as evidenced by increased levels of hydrogen peroxide, interleukin (IL)-1β, and IL-8. OSO increased the activities of glutathione peroxidase and superoxide dismutase. OSO upregulated the expression of apoptosis-related genes, nuclear factor-erythroid 2-related factor 2 (Nrf2), and nuclear factor κB-related inflammatory factor genes. In the offspring of the OSO-exposed mothers, hydrogen peroxide, malondialdehyde, IL-6, and tumor necrosis factor-alpha contents were increased. Furthermore, OSO enhanced the levels of Nrf2, NAD(P)H quinone oxidoreductase 1, heme oxygenase 1, and p65 and decreased B-cell lymphoma 2. Conclusion: These findings indicated that the kidneys of two generations of rats were compromised by oxidative damage when fed OSO during lactation. This study provides evidence for increasing the genes expression of the Nrf2/heme oxygenase 1 pathway to alleviate the kidney damage caused by OSO in the mother and offspring. © 2021 Society of Chemical Industry.

Abstract:Dietary oxidized fat contains harmful materials such as hydrogen peroxide and malondialdehyde (MDA). Excessive oxidized fat intake during pregnancy and lactation not only leads to maternal body injury but also damages offspring health. Our previous study demonstrated that vitamin D (VD) had antioxidative capability in sows. This study was conducted to investigate the effect of maternal VD and inulin supplementation in oxidized oil diet on the growth performance and oxidative stress of their offspring. Sixty 5-month-old C57BL/6N female mice were randomly divided into five groups: Control group (basal diet, n = 12), OF group (oxidized-soybean-oil-replaced diet, n = 12), OFV group (oxidized-soybean-oil-replaced diet + 7000 IU/kg VD, n = 12), OFI group (oxidized-soybean-oil-replaced diet + 5% inulin, n = 12) and OFVI group (oxidized-soybean-oil-replaced diet + 7000 IU/kg VD + 5% inulin, n = 12). Mice were fed with the respective diet during pregnancy and lactation. The offspring were then slaughtered on day 21 of age at weaning. Results showed that a maternal oxidized oil diet impaired body weight and liver weight gain of offspring during lactation compared to the control group, while maternal VD, inulin or VD and inulin mixture supplementation reversed this effect. In addition, the activity of T-AOC in the liver of offspring was lower in the OF group than that in the control group, but could be restored by maternal VD and inulin mixture supplementation. Furthermore, the gene expression of both proinflammatory and anti-inflammatory cytokines, such as Il-6, Tnfα and Il-10, in offspring liver were downregulated by a maternal oxidized oil diet compared with the control group, but they were restored by maternal VD or VD and inulin mixture supplementation. The expressions of Vdr and Cyp27a1 were decreased by a maternal oxidized oil diet compared with the control group, while they could be increased by VD or VD and inulin mixture supplementation. Conclusion: maternal oxidized oil diet intake could impair the growth performance by inducing oxidative stress, but this can be relieved by maternal VD and inulin supplementation.

Abstract:The present study aimed to investigate the preventive effect of mussel oil (MO) on gestational diabetes mellitus (GDM) in mice fed by a high-fat and high-sucrose (HFHS) diet. Pregnant mice were allocated to four groups: normal diet + corn oil (CO), HFHS + CO, HFHS + fish oil (FO), and HFHS + MO. The total n-3 polyunsaturated fatty acids (PUFAs) in MO (51.30%) and FO (48.25%) were comparable (mainly C22:6n-3 and C20:5n-3). HFHS + MO and HHFS + FO had a significantly lower area under the curve (AUC) for the oral glucose tolerance test (OGTT) than the HFHS + CO group. The HFHS + MO group but not HFHS + FO group had a significantly lower AUC for the insulin tolerance test (ITT) than the HFHS + CO group. The HFHS + MO group had significantly lower homeostasis model assessment-insulin resistance (HOMA-IR) and fasting serum insulin than the HHFS + FO and HFHS + CO groups. Liver sphingosine kinase 1 (SphK1) was significantly higher, while SphK2, Akt, and P-Akt were significantly lower in the HFHS + CO group compared with the normal diet + CO group. The HFHS + MO group but not the HFHS + FO group had significantly higher SphK2, Akt, and P-Akt than the HFHS + CO group. SphK2 had a strong negative correlation with the AUC for the OGTT ( = -0.759, = 0.001) and insulin tolerance test (ITT) ( = -0.637; = 0.008), fasting serum insulin ( = -0.594, = 0.015), fasting blood glucose ( = -0.587, = 0.017) and HOMA-IR ( = -0.629, = 0.009) and a strong positive correlation with Akt ( = 0.594, = 0.015) and P-Akt ( = 0.676, = 0.004). In conclusion, mussel oil improved glucose intolerance and insulin resistance during mice pregnancy, which was superior to the effects of fish oil.

Abstract:Corn oil, sesame oil, and 10% ethanol in corn oil are commonly used as dosing vehicles in toxicology studies. Since these vegetable oils contain bioactive compounds, it is important for toxicology studies to characterize the toxicities of the dosing vehicles themselves. It has been recently proposed that the width of the genital tubercle (GT), the dorsal-ventral length (D-V length) of the GT, and urethral tube closure in mouse fetuses can be used as novel markers for monitoring sexual development in mice. However, how these parameters are influenced by the dosing vehicles themselves remains unclear. Therefore, we evaluated the effects of corn oil, sesame oil, and 10% ethanol in corn oil on GT width, D-V length, and GT morphology in ICR mice. Our results showed that all three vehicles influenced GT width and D-V length, but not GT morphology, suggesting that the effects of dosing vehicles themselves might need to be considered when GT width or D-V length is used as a parameter to evaluate the effects of chemicals on GT development.
Maternal organophosphate flame-retardant exposure alters offspring feeding, locomotor and exploratory behaviors in a sexually-dimorphic manner in mice

Abstract:Increased usage of organophosphate flame retardants (OPFRs) has led to detectable levels in pregnant women and neonates, which is associated with negative neurological outcomes. Therefore, we investigated if maternal OPFR exposure altered adult offspring feeding, locomotor, and anxiety-like behaviors on a low-fat (LFD) or high-fat diet (HFD). Wild-type C57Bl/6J dams were orally dosed with vehicle (sesame oil) or an OPFR mixture (1 mg/kg combination each of tris(1,3-dichloro-2-propyl)phosphate, triphenyl phosphate and tricresyl phosphate) from gestation day 7 to postnatal day 14. After weaning, pups were fed either a LFD or HFD until 19 weeks of age. Locomotor and anxiety-like behaviors were evaluated with the open field test, elevated plus maze, and metabolic cages. Feeding behaviors and meal patterns were analyzed by a Biological Data Acquisition System. Anogenital distance was reduced in OPFR-exposed male pups, but no effect was detected on adult body weight. We observed interactions of OPFR exposure and HFD consumption on locomotor and anxiety-like behavior in males, suggesting an anxiogenic effect while reducing overall nighttime activity. We also observed an interaction of OPFR exposure and HFD on weekly food intake and feeding behaviors. OPFR-exposed males consumed more total HFD than oil-exposed males during the 72-hour trial. However, when arcuate gene expression was analyzed, OPFR exposure induced Agrp expression in females, which would suggest greater orexigenic tone. Collectively, the implications of our study are that the behavioral effects of OPFR exposure are modulated by adult HFD consumption, which may influence the metabolic and neurological consequences of maternal OPFR exposure.
Molecular phylogenetics supports widespread cryptic species in moonworts (Botrychium s.s., Ophioglossaceae).

Abstract:PREMISE OF THE STUDY Previous phylogenetic studies of moonworts (Botrychium sensu stricto (s.s.)) included few taxa from outside of North America. This low geographical representation limited interpretations of relationships of this group rich in cryptic species. With 18 out of 30 species in the genus being polyploid, understanding their evolutionary history remains a major challenge. METHODS A new molecular phylogeny was reconstructed using Maximum Likelihood (ML) and Bayesian Inference (BI) analyses based on multiple accessions of the most wide-ranging Arctic taxa of Botrychium in North America and Europe using three noncoding plastid DNA regions (psbA-trnH(GUG), trnL(UAA)-trnF(GAA) intergenic spacer, and rpL16 intron). KEY RESULTS The new phylogeny confirms the identity of several recently described species and proposed new taxa. Nine subclades are newly identified within the two major clades in Botrychium s.s.: Lanceolatum and Lunaria. Chloroplast DNA was variable enough to separate morphologically cryptic species in the Lunaria clade. On the contrary, much less variation is seen within the morphologically variable Lanceolatum clade despite sampling over the same broad geographic range. The chloroplast region psbA-trnH(GUG) is identified as an efficient DNA barcode for the identification of cryptic taxa in Botrychium s.s. CONCLUSIONS The combined increase in species representation, samples from throughout the geographic range of each species, and sequencing of multiple plastid DNA regions supports morphologically cryptic species in Botrychium s.s.
[Suppression effect of secoisolariciresinol diglucoside against trans fatty acids-induced oxidative damage and inflammatory in brain of offspring mice]

Abstract:Objective: To probe into the protective effect of different dose of secoisolariciresinol diglucoside(SDG) on brain of offspring of mice anainst oxidative damage and inflammatory reaction induced by maternal exposure to trans fatty acids(TFA) during gestation, and observe the the changes of regulating Nrf2/Keap1 pathway in the course. Methods: 30 healthy female mice(C57BL/6) were divided into 5 groups randomly, they are respectively control group, TFA-exposed group, and three SDG-intervention groups(low-(TFA+LSDG), medium-(TFA+MSDG) and high-(TFA+HSDG)). The pregnancy mice of control group and TFA group were treated with distilled water and 60 mg/kg·d TFA by gavage, in the same time, the mice of three SDG-intervention groups were treated with 60 mg/kg·d TFA by gavage and fed with feed included SDG(10, 20 and 30 mg/kg). The treatment to pregnancy mice continued to birth of offspring. After 21 days of lactation, the offspring were killed under anesthesia and the experiment was ended. The coefficient of brain was calculated. The levels of superoxide dismutase(SOD), glutathione peroxidase(GSH-Px), malondialdehyde(MDA), tumor necrosis factor-α(TNF-α), interferon-γ(IFN-γ) and amyloid-β(Aβ)of brain were detected. RT-PCR and Western Blot was used to detected gene expression and protein levels of nuclear factor erythroid-2 related factor 2(Nrf2), kelch-like ECH-associated protein 1(Keap1), quinone oxidoreductase 1(NQO1) and hemeoxygenase-l(HO-1). Results: Compared with control group, the brain coefficient and Aβ1-40 of offspring of TFA-group had no significant changes(P>0.05), the activity of SOD and GSH-Px reduced, the content of MDA, IFN-γ, TNF-α and Aβ1-42 increased, the level of mRNA and protein expression of Nrf2, NQO1 and HO-1 decreased and the level of mRNA and protein expression of Keap1 increase because of the exposion to TFA during gestation and all the differences were statistically significant(P<0.05). Compared with TFA-group, the brain coefficient, Aβ1-40 and the level of NQO1 mRNA of offspring of three SDG-intervention groups had no significant changes(P>0.05), the activity of SOD(the middle and high dose SDG intervention groups) and GSH-Px(three SDG-intervention groups) increased, the content of MDA(the middle and high dose SDG intervention groups), IFN-γ(the middle and high dose SDG intervention groups), TNF-α(three SDG-intervention groups) and Aβ1-42(the middle and high dose SDG intervention groups) decreased, the mRNA expression of Nrf2 and HO-1(the middle and high dose SDG intervention groups) was up-regulated, the mRNA expression of Keap1(the middle and high dose SDG intervention group) decreased, proteic expression of Nrf2, NQO1 and HO-1 of three SDG-intervention groups increase and the level of protein of Keap1 decreased because of the intervention of SDG during gestation(P<0.05). Conclusion: These result suggest that maternal TFA exposure during gestation can result in oxidative stress and inflammation to brain of offspring in a way. SDG can protect brain of mice of offspring from TFA-induced oxidative injury by up-regulating the expression of mRNA and protein of Nrf2, down-regulating the expression of Keap1, accelerating expression of protein of NQO1 and HO-1 which are antioxidant protein lying downstream of pathway of Nrf2/Keap1.

Abstract:In this work, the paraffin oil was first molecularly distilled into two mineral oil saturated hydrocarbons (MOSH) subfractions with their main carbon number range of C20-30 (84.58%) and C30-50 (89.65%), which were determined using a liquid chromatography-gas chromatography-flame ionization detector (LC-GC-FID) technique. Then, both female and male Fischer 344 (F344) rats were fed with the diets containing one of these two MOSH subfractions at the low (1.5 g/kg) or high dose (15 g/kg) for 6 months. It was found that the exposure of MOSH C20-30 subfraction could result in a significant increase of spleen weight index (4.64 ± 0.18 mg/g for female and 3.79 ± 0.31 mg/g for male rats) (P < 0.05), and irregular shape of white medulla, and the macrophage infiltration in mesenteric lymph node for both female and male rats, especially for the 15 g/kg MOSH C20-30-fed rats. Meanwhile, both MOSH C20-30 and C30-50 subfraction promoted inflammation and oxidative stress and had a negative effect on the immune function in both female and male rats. The metabolomics results further supported these observations, and indicated the down-regulation of cysteine and methionine metabolism, and glutathione metabolism pathway in female and male rats, respectively.
Toxicological effects of repeated subcutaneous administration of corn oil for 4 weeks in rats

Abstract:In toxicity studies, it is important to select a vehicle that does not affect the toxicity assessment of the test substance. To obtain toxicity information on corn oil as a Vehicle for subcutaneous toxicity studies of non-aqueous test substances, Crl:CD (SD) rats (6 animals/sex/group) were repeatedly dosed with corn oil at 0 (negative control; saline), 1, 2 and 5 mL/kg/day for 4 weeks. High body weight due to retention of the administered substance were found in corn oil groups in both sexes at 2 mL/kg/day or higher. Some relative organ weights at 2 mL/kg/day or higher were decreased due to high body weight or body weight gain. Subcutaneous retention of the administered substance found in all corn oil-treated groups at necropsy was observed on histopathological analysis as accumulation of the administered substance and granulation tissue. Accumulation of the administered substance in inguinal skin, the axillary lymph node, and alveolus of the lung and bronchus was observed. Multiple white nodules in the abdominal cavity and liver observed in a female at 5 mL/kg/day at necropsy were considered to be related to lipogranuloma at the peritoneum and hepatic capsule. In hematology, RBC was lower in females at the 5 mL/kg/day than in the control group.. In conclusion, since the effects of corn oil administration were found in groups at all volumes, it is necessary to take into account that corn oil induces these changes when used as a vehicle of a test substance in toxicity studies.
Soybean oil induces neuroinflammatory response through brain-gut axis under high-fat diet

Abstract:Neuroinflammation is considered the principal pathogenic mechanism underlying neurodegenerative diseases, and the incidence of brain disorders is closely linked to dietary fat consumption and intestinal health. To investigate this relationship, 60 8-week-old C57BL/6J mice were subjected to a 20-week dietary intervention, wherein they were fed lard and soybean oil, each at 15% and 35% fat energy. At a dietary fat energy level of 35%, inflammation was observed in both the soybean oil and lard groups. Nevertheless, inflammation was more pronounced in the mice that were administered soybean oil. The process by which nerve cell structure is compromised, inflammatory factors are upregulated, brain antioxidant capacity is diminished, and the TLR4/MyD88/NF-κB p65 inflammatory pathway is activated resulting in damage to the brain-gut barrier. This, in turn, leads to a reduction in the abundance of Akkermansia and unclassified_f_Lachnospiraceae, as well as an increase in Dubosiella abundance, ultimately resulting in brain inflammation and damage. These results suggested that soybean oil induces more severe neuroinflammation compared to lard. Our study demonstrated that, at a dietary fat energy level of 35%, compared to soybean oil, lard could be the healthier option, the outcomes would help provide a reference basis for the selection of residents' daily dietary oil.
Effects of repeatedly heated cooking oil consumption in mice: a study on health implications

Abstract:Background: Cooking oils are a major part of human diets, but repeated use of heated oils can have detrimental effects on consumer health. This study aims to investigate the impact of different heating grades of vegetable oils on the hemato-biochemical parameters and vital organs like the heart, liver, kidney, and intestine in mice. Methods: Thirty mice were randomly assigned to different treatment groups, including a control group (diet only), unheated cooking oil (UHCO) group, single heated cooking oil (SHCO) group, three times repeatedly heated cooking oil (3RHCO) group, and repeatedly heated cooking oil (ReHCO) group. Blood and organ samples were collected on day 31 to investigate hemato-biochemical parameters and histo-morphological alterations in response to the oil treatments. Results: The oil-treated groups showed significant (P<0.05) decreases in the total erythrocyte, leukocyte, and hemoglobin levels. Meanwhile, serum levels of total cholesterol, triglyceride, high-density lipoprotein, glucose, and creatinine increased significantly (P<0.05), while low-density lipoprotein and protein levels dropped markedly in the treatment groups. Severe histo-morphological alterations were also found in the liver (hepatocytic degeneration with hydropic change in the 3RHCO and ReHCO groups), kidney (glomerular atrophy with increased glomerular space, tubular degeneration, and lymphocytic infiltration in the SHCO, 3RHCO, and ReHCO groups), and colon (lymphocytic infiltration in the mucosal layer of ReHCO group). Conclusions: These findings suggest that the consumption of heated oils can have severe adverse effects on consumers' health, leading to alterations in blood chemistry and damage to vital organs.
The Oil Formulation Derived from Moringa Oleifera Seeds Ameliorates Behavioral Abnormalities in Water-immersion Restraint Stress Mouse Model

In spite of the traditional uses of Moringa oleifera (MO), no experimental evidence for its use against chronic stress exists. Here, we investigated whether seed oil from MO (MOO) could improve behavior abnormalities of chronic stress mice induced by water-immersion restraint stress (WIRS) and the underlying mechanism. Methods: BALB/C male mice at 12 weeks of age were exposed to chronic WIRS for two weeks and divided in to four groups: normal group, WIRS group, WIRS+MOO1 group (treated with MOO at the dose of 1 mL/kg BW), and WIRS+MOO2 group (treated with MOO 2 mL/kg BW). The MOO treatment was given orally for 23 days. On day 24, we checked the behavior parameters, the plasma level of cortisol, acetylcholinesterase (AChE) activity in hippocampus, mRNA expression level of brain-derived neurotrophic factor ( BDNF ) and oxidative stress parameters in brain tissues. In addition, we also checked the histopathological features of the gastric mucosa wall. Results: Administration of MOO ameliorated anxiety-like, depression-like and memory impairment phenotypes in the WIRS mouse model although the plasma cortisol concentrations were comparable among the groups. Of note, MOO both in two doses could suppress the AChE activity in hippocampus tissue and ameliorated the MDA level in prefrontal cortex tissue in mice exposed to WIRS. Although only WIRS+MOO2 group could increase the mRNA expression of BDNF , the histopathological gastric mucosa wall features were improved in all MOO groups. Conclusion: Taken together, these finding suggested that MOO may have a neuroprotective effect in the mouse model of WIRS as evidenced by improving the abnormal behaviors through enhancing mRNA expression level of BDNF , inhibited AChE activity, and prevented the increase of MDA level in the brain. Stress causes structural changes in the hippocampus, prefrontal cortex, amygdala, anterior cingulate cortex, and basal ganglia. 1 Excessive external stress is a widely accepted theory in the development of mood disorders such as depression and anxiety. 2 Stress negatively affects many parts of the brain which are involved in the regulation of emotion including the cortex and hippocampus. 3,4 In addition, for decades the idea that maladaptive stress impairs cognitive function has been a cornerstone to be explored in basic and clinical research. Effects of Daily Intake of Plant and Animal Oils on the Incidence of Depression in Adult Mice

Abstract:Background: Depression is a complicated disturbance affected by a collection of biological and environmental factors. The first aim of psychiatric studies is to recognize biological markers that could be utilized to predict improvement and increase reactions to antidepressant treatments. Diet affects different aspects of health, including depression. The aim of study was to determine antidepressant-like effects of some consumable oils, the effects of oils on depression were compared. Methods: Thirty-two male and female mice (Mus musculus (BALB/c)) weighing 25- 35 g were randomly divided into 8 groups (4 mice in each group, 2 male and 2 female, A: Laden sunflower liquid oil, B: 50% vegetable oil + 50% olive oil, C: Kermanshah Rojin animal oil, D: Spring frying oil, E: Distilled water, F: BAHAR solid vegetable oil, G: Olive oil, H: 50% Kermanshah animal oil + 50% of olive oil). In different groups, 30 g of vegetable or animal oil was gavaged every day at 1:00 pm. Five types of vegetable and animal oils among the high consumption oils in the market were selected, including spring frying oil (used for several times in 305 ̊f, usually used in eastern Asia countries), Laden sunflower liquid oil, olive oil, Kermanshah Rojin animal oil, and BAHAR solid vegetable oil. After 6 weeks of using the oils diet, the forced swim test was utilized as a test of depression like behavior. Results: There was a significant difference between all groups (P < 0.0001). Based on the results, the latency time of immobility in group A significantly decreased in comparison with groups C (P ˂ 0.02), D (P ˂ 0.003), and G (P < 0.001). However, it increased in groups B and C compared to group H (P ˂ 0.02). Also, this parameter in group D increased significantly compared to groups E (P < 0.01), F (P < 0.05), and H (P < 0.002). Conclusion: The results indicated that olive oil had a preventive effect against forced swimming-induced depression-like symptoms.

Abstract:Sesame oil and sunflower oil are popular cooking oils in southern India. Deep-frying is a frequent method of food preparation. Deep-frying at high temperatures has been linked with several disorders, including cancer, diabetes, and unknown metabolic problems. There have been no long-term investigations on the influence of deep-fried oils on PUFA metabolism and pathogenesis. As a result, the current study aimed to explore the effect of deep-fried frying oil on Wistar rats by continuous treatment. Furthermore, the pathophysiology of MSG-induced neurotoxicity in Wistar rats was investigated.

Abstract:Diet adjustment will affect the health of gut microbiota, which in turn influences the development and function of the organism's brain through the gut–brain axis. Walnut oil (WO), peony seed oil (PSO) and camellia seed oil (CSO), as typical representatives of woody plant oils, have been shown to have the potential to improve cognitive impairment in mice, but the function mechanisms are not clear. In this study, we comparatively investigated the neuroprotective effects of these three oils on D -galactose ( D -gal)-induced cognitive impairment in mice, and found that the ameliorative effect of WO was more prominent. During the behavioral experiments, supplementation with all three oils would improve spatial learning and memory functions in D -gal mice, with a significant reduction in the error times ( p < 0.001) and a significant increase in step-down latency ( p < 0.001); walnut oil supplementation also significantly increased the number of hidden platform traversals, the target quadrant spent times and percentage of distance ( p < 0.05). The results of biomarker analysis showed that WO, in addition to significantly inhibiting D -gal-induced oxidative stress and neuroinflammation as did PSO, significantly increased the ACh content in the mouse brain ( p < 0.05) and modulated neurotransmitter levels. The results of further microbiota diversity sequencing experiments also confirmed that dietary supplementation with all three oils affected the diversity and composition of the gut microbiota in mice. Among them, WO significantly restored the balance of the mouse gut microbiota by increasing the abundance of beneficial bacteria ( Bacteroidetes , Actinobacteria , Firmicutes ) and decreasing the abundance of harmful bacteria ( Clostridium , Shigella , Serratia ), which was consistent with the results of behavioral experiments and biomarker analyses. Based on the analysis of the fatty acid composition of the three oils and changes in the gut microbiota, it is hypothesized that there is a correlation between the fatty acid composition of the dietary supplement oils and neuroprotective effects. The superiority of WO over PSO and CSO in improving cognitive impairment is mainly attributed to its balanced composition of omega-6 and omega-3 fatty acids.
