Abstract:Understanding the evolutionary processes that shape the landscape of genetic variation and influence the response of species to future climate change is critical for biodiversity conservation. Here, we sampled 27 populations across the distribution range of a dominant forest tree, Quercus acutissima, in East Asia, and applied genome‐wide analyses to track the evolutionary history and predict the fate of populations under future climate. We found two genetic groups (East and West) in Q. acutissima that diverged during Pliocene. We also found a heterogeneous landscape of genomic variation in this species, which may have been shaped by population demography and linked selections. Using genotype‐environment association analyses, we identified climate‐associated SNPs in a diverse set of genes and functional categories, indicating a model of polygenic adaptation in Q. acutissima. We further estimated three genetic offset metrics to quantify genomic vulnerability of this species to climate change due to the complex interplay between local adaptation and migration. We found that marginal populations are under higher risk of local extinction because of future climate change, and may not be able to track suitable habitats to maintain the gene‐environment relationships observed under the current climate. We also detected higher reverse genetic offsets in northern China, indicating that genetic variation currently present in the whole range of Q. acutissima may not adapt to future climate conditions in this area. Overall, this study illustrates how evolutionary processes have shaped the landscape of genomic variation, and provides a comprehensive genome‐wide view of climate maladaptation in Q. acutissima.

Abstract:Rapid global climate change is posing a substantial threat to biodiversity. The assessment of population vulnerability and adaptive capacity under climate change is crucial for informing conservation and mitigation strategies. Here we generate a chromosome-scale genome assembly and re-sequence genomes of 230 individuals collected from 24 populations for Populus koreana , a pioneer and keystone tree species in temperate forests of East Asia. We integrate population genomics and environmental variables to reveal a set of climate-associated single-nucleotide polymorphisms, insertion/deletions and structural variations, especially numerous adaptive non-coding variants distributed across the genome. We incorporate these variants into an environmental modeling scheme to predict a highly spatiotemporal shift of this species in response to future climate change. We further identify the most vulnerable populations that need conservation priority and many candidate genes and variants that may be useful for forest tree breeding with special aims. Our findings highlight the importance of integrating genomic and environmental data to predict adaptive capacity of a key forest to rapid climate change in the future.

Abstract:A rapidly changing climate and frequent human activity influences the distribution and community structure of forests. Increasing our knowledge about the genetic diversity and distribution patterns of trees is helpful for forest conservation and management. In this study, nSSRs (nuclear simple sequence repeats) were integrated with a species distribution model (SDM) to investigate the spatial genetic patterns and distribution dynamics of Quercus chungii F.P.Metcalf, a rare oak in the subtropics of southeast China. A total of 188 individuals from 11 populations distributed across the natural range of Q. chungii were genotyped using nine nSSRs. The STRUCTURE analysis indicated that genetic admixture was present in all populations, but the population genetic variation and genetic differentiation were related to their geographical distributions. The SDM result indicated that Q. chungii retreated to the Nanling Mountains and adjacent areas during the Last Glacial Maximum (LGM) period, which corresponds to higher genetic diversity for populations in this region. Landscape genetic analysis showed that the Nanling Mountains served as a corridor for organism dispersal at the glacial and interglacial periods within the Quaternary. Based on these results, we propose that establishing nature reserves to protect the ecological corridor across the Nanling Mountains is necessary for the conservation of regional species genetic diversity, as well as the ecosystem of evergreen broadleaved forests in southern China. The study combines species distribution models and genetic diversity to provide new insight into biodiversity conservation and forest management under future climate change.

Abstract:The unprecedented habitat fragmentation or loss has threatened the existence of many species. Therefore, it is essential to understand whether and how these species can pace with the environmental changes. Recent advantages in landscape genomics enabled us to identify molecular signatures of adaptation and predict how populations will respond to changing environments, providing new insights into the conservation of species. Here, we investigated the pattern of neutral and putative adaptive genetic variation and its response to changing environments in a tertiary relict tree species, Taxus cuspidata Sieb. et Zucc, which is distributed in northeast China and adjacent regions. We investigated the pattern of genetic diversity and differentiation using restriction site-associated DNA sequencing (RAD-seq) and seven nuclear microsatellites (nSSRs) datasets. We further explored the endangered mechanism, predicted its vulnerability in the future, and provided guidelines for the conservation and management of this species. RAD-seq identified 16,087 single nucleotide polymorphisms (SNPs) in natural populations. Both the SNPs and nSSRs datasets showed high levels of genetic diversity and low genetic differentiation in T. cuspidata. Outlier detection by F ST outlier analysis and genotype-environment associations (GEAs) revealed 598 outlier SNPs as putative adaptive SNPs. Linear redundancy analysis (RDA) and nonlinear gradient forest (GF) showed that the contribution of climate to genetic variation was greater than that of geography, and precipitation played an important role in putative adaptive genetic variation. Furthermore, the genetic offset and risk of non-adaptedness (RONA) suggested that the species at the northeast edge may be more vulnerable in the future. These results suggest that although the species has maintained high current genetic diversity in the face of recent habitat loss and fragmentation, future climate change is likely to threaten the survival of the species. Temperature (Bio03) and precipitation (Prec05) variables can be potentially used as predictors of response of T. cuspidata under future climate. Together, this study provides a theoretical framework for conservation and management strategies for wildlife species in the context of future climate change.

Abstract:Abstract Background and Aims Understanding adaptive genetic variation and whether it can keep pace with predicted future climate change is critical in assessing the genetic vulnerability of species and developing conservation management strategies. The lack of information on adaptive genetic variation in relict species carrying abundant genetic resources hinders the assessment of genetic vulnerability. Using landscape genomics approach, this study aimed to determine how adaptive genetic variation shapes population divergence and predict the adaptive potential of Pterocarya macroptera (a vulnerable relict species in China) under future climate scenarios. Methods We applied restriction site-associated DNA sequencing (RAD-seq) to obtain 8,244 single nucleotide polymorphisms (SNPs) from 160 individuals across 28 populations. We examined the pattern of genetic diversity and divergence, and then identified outliers by genetic differentiation (FST) and genotype–environment association (GEA) methods. We further dissected the effect of geographic/environmental gradients on genetic variation. Finally, we predicted genetic vulnerability and adaptive risk under future climate scenarios. Key Results We identified three genetic lineages within P. macroptera: the Qinling-Daba-Tianmu Mountains (QDT), Western Sichuan (WS), and Northwest Yunnan (NWY) lineages, which showed significant signals of isolation by distance (IBD) and isolation by environment (IBE). IBD and IBE explained 3.7–5.7% and 8.6–12.8% of the genetic structure, respectively. The identified GEA SNP-related genes were involved in chemical defence and gene regulation and may exhibit higher genetic variations to adapt to the environment. Gradient forest analysis revealed that the genetic variation was mainly shaped by temperature-related variables, indicating its adaptation to local thermal environments. A limited adaptive potential was suggested by the high levels of genetic vulnerability in marginal populations. Conclusions Environmental gradient mainly shaped the population differentiation of P. macroptera. Marginal populations may be at high risk of extinction, and thus, proactive management measures, such as assisted gene flow, are required to ensure the survival of these populations.

Abstract:Tree species have good tolerance to a range of environmental conditions, though their ability to respond and persist to environmental changes is dramatically reduced at the rear-edge distribution limits. At those edges, gene flow conferring adaptation is impaired due to lack of populations at lower latitudes. Thus, trees mainly rely on phenotypic changes to buffer against long-term environmental changes. Interspecific hybridization may offer an alternative mechanism in the generation of novel genetic recombinants that could be particularly valuable to ensure persistence in geographically isolated forests. In this paper, we take advantage of the longevity of a temperate-submediterranean mixed-oak forest to explore the long-term impact of environmental changes on two different oak species and their hybrid. Individual trees were genetically characterized and classified into three groups: pure Quercus petraea (Matt.), Liebl, pure Q. pyrenaica Willd, and hybrids. We calculated basal area increment and intrinsic water-use efficiency (iWUE) from tree-ring width and δ13C per genetic group, respectively. Tree-growth drivers were assessed using correlation analyses and generalized linear mixed models for two contrasting climatic periods: (1880–1915, colder with [CO2] < 303 ppm; and 1980–2015, warmer with [CO2] > 338 ppm). The three genetic groups have increased radial growth and iWUE during the last decades, being the least drought-tolerant QuPe the most sensitive species to water stress. However, no significant differences were found among genetic groups neither in mean growth rate nor in mean iWUE. Furthermore, little differences were found in the response to climate among groups. Genetic groups only differed in the relationship between δ13C and temperature and precipitation during the earlier period, but such a difference disappeared during the recent decades. Climate change may have promoted species-level convergence as a response to environment-induced growth limitations, which translated in synchronized growth and response to climate as well as a tighter stomatal control and increased iWUE across coexisting oak species.

Abstract:Despite widespread biodiversity loss, our understanding of how species and populations will respond to accelerated climate change remains limited. In this study, we predict the evolutionary responses of Populus lasiocarpa, a key alpine forest tree species primarily found in the mountainous regions of a global biodiversity hotspot, to climate change. We accomplish this by generating and integrating a new reference genome for P. lasiocarpa, re-sequencing data for 200 samples, and gene expression profiles for leaf and root tissue following exposure to heat and waterlogging. Analyses of the re-sequencing data indicate that demographic dynamics, divergent selection, and long-term balancing selection have shaped and maintained genetic variation within and between populations over historical timescales. In examining genomic signatures of contemporary climate adaptation, we found that haplotype blocks, characterized by inversion polymorphisms that suppress recombination, play a crucial role in clustering environmentally adaptive variations. Comparison of evolved and plastic gene expression show that genes with expression plasticity generally align with evolved responses, highlighting the adaptive role of plasticity. Lastly, we incorporated local adaptation, migration, genetic load, and plasticity responses into our predictions of population-level climate change risks. Our findings reveal that western populations, primarily distributed in the Hengduan Mountains-a region known for its environmental heterogeneity and significant biodiversity-are the most vulnerable to climate change and should be prioritized for conservation and management. Overall, our study advances understanding of the relative roles of long-term natural selection, local environmental adaptation, and immediate plasticity responses in driving evolutionary adaptation to climate change in keystone species.

Abstract:Understanding the genetic basis underpinning local adaptation is one of the fundamental issues in ecological and evolutionary biology. In this study, we investigated the genomic basis underlying local adaptation of the Chinese wingnut (Pterocarya stenoptera C. DC). Our population genomic analyses revealed nine spatial genetic clusters across the current distribution range of this species. Based on the assessment of genetic–environment association, we found that adaptive divergence of the P. stenoptera populations were mainly shaped by solar radiation during fruit development, temperature seasonality, annual temperature, precipitation, and air humidity. In particular, our genome‐wide scanning identified a total of 801 candidate single nucleotide polymorphisms (SNPs) that are highly correlated with diverse environmental factors. Further functional annotation of the SNPs identified some candidate genes that are involved into temperature, water, and light adaptation. Taken together, our results suggest that natural selection during local adaptation has contributed to the success of survival to diverse heterogenous environmental conditions. Our study provides important insights into the fundamental knowledge of the genetic basis underlying the local adaptation of non‐model species.
Genetically based variation in fitness and carbon assimilation among bur oak populations

Abstract:Ongoing climate change will negatively impact tree populations unless they are able to acclimate to the changes in their local environment. Effective planning for climate adaptation management requires an understanding of the current state of local adaptation and physiological performance to assess whether populations are at risk of local extinction, determine if assisted migration is appropriate, and to select appropriate seed sources if intervention is needed. We established a new reciprocal transplant experiment (ACE, Adaptation to Climate and Environment) across a latitudinal gradient in North America to examine the impacts of warming on three bur oak (Quercus macrocarpa) populations across much of the species range. We established common gardens in Minnesota, Illinois, and Oklahoma with seedlings grown from seeds collected within 50 km of each of those locations from a total of sixty maternal families . We aimed to 1) assess local adaptation in each of the populations using survival and size as fitness metrics, and 2) evaluate physiological responses to different environments along the latitudinal gradient. We found that northern populations are maladapted to hotter climates as evidenced by their low survival, growth, and photosynthetic rates in the warmest common garden. The southernmost population had the highest survival rate, growth rate, and fitness of the three populations in the southernmost garden, providing evidence for local adaptation to the warmest site. However, conditions in the middle garden resulted in the highest fitness and best physiological performance for all populations. Growth and survival were correlated in the middle garden but were decoupled in the northern and southern gardens. This decoupling is likely due to stress associated with the relatively extreme climates in the gardens at the ends of the gradient that led to greater resource allocation to survival than growth. Our results suggest that southern seed sources may perform well in warmer conditions in the north brought on by climate change, which has important implications for managers assisting broadly ranged tree species in adapting to climate change.

New insights into adaptation and population structure of cork oak using genotyping by sequencing

Abstract:Species respond to global climatic changes in a local context. Understanding this process, including its speed and intensity, is paramount due to the pace at which such changes are currently occurring. Tree species are particularly interesting to study in this regard due to their long generation times, sedentarism, and ecological and economic importance. Quercus suber L. is an evergreen forest tree species of the Fagaceae family with an essentially Western Mediterranean distribution. Despite frequent assessments of the species' evolutionary history, large-scale genetic studies have mostly relied on plastidial markers, whereas nuclear markers have been used on studies with locally focused sampling strategies. In this work, "Genotyping by sequencing" is used to derive 1,996 single nucleotide polymorphism markers to assess the species' evolutionary history from a nuclear DNA perspective, gain insights into how local adaptation is shaping the species' genetic background, and to forecast how Q. suber may respond to global climatic changes from a genetic perspective. Results reveal (a) an essentially unstructured species, where (b) a balance between gene flow and local adaptation keeps the species' gene pool somewhat homogeneous across its distribution, but still allowing (c) variation clines for the individuals to cope with local conditions. "Risk of Non-Adaptedness" (RONA) analyses suggest that for the considered variables and most sampled locations, (d) the cork oak should not require large shifts in allele frequencies to survive the predicted climatic changes. Future directions include integrating these results with ecological niche modeling perspectives, improving the RONA methodology, and expanding its use to other species. With the implementation presented in this work, the RONA can now also be easily assessed for other organisms.

Abstract:The investigation of the genetic basis of local adaptation in non-model species is an interesting focus of evolutionary biologists and molecular ecologists. Identifying these adaptive genetic variabilities on the genome responsible can provide insight into the genetic mechanism of local adaptation. We investigated the spatial distribution of genetic variation in 22 natural populations of Pterocarya stenoptera across its distribution area in China to provide insights into the complex interplay between multiple environmental variables and adaptive genetic differentiation. The Bayesian analysis of population structure showed that the 22 populations of P. stenoptera were subdivided into two groups. Redundancy analysis demonstrated that this genetic differentiation was caused by the divergent selection of environmental difference. A total of 44 outlier loci were mutually identified by Arlequin and BayeScan, 43 of which were environment-associated loci (EAL). The results of latent factor mixed model analysis showed that solar radiation in June (Sr6), minimum temperature of the coldest month (Bio6), temperature seasonality (Bio4), and water vapor pressure in January (Wvp1) were associated with the highest numbers of EAL. Sr6 was associated with the ecological habitat of “prefered light”, and Bio6 and Wvp1 were associated with the ecological habitat of “warm and humid environment”. Our results provided empirical evidence that environmental variables related to the ecological habitats of species play key roles in driving adaptive differentiation of species genome.

Abstract:Background Global climate change poses severe threats to biodiversity and ecosystem stability. Rapid climate oscillations potentially lead to species geographic range shifts, population declines, and even extinctions. The rare and endangered species, being critical components of regional biodiversity, hold the key to understanding local adaptation and evolutionary processes shaping species distributions. Therefore, assessing the evolutionary mechanisms of local adaptation and population vulnerability under climate change is crucial for developing conservation strategies of endangered species. Results In this study, we assembled a high-quality, chromosome-level genome of the rare and endangered herb Bergenia scopulosa in the Qinling Mountains in East Asia and resequenced 37 individual genomes spanning its entire geographic distributional ranges. By integrating population genetics, landscape genomics, and climate datasets, a substantial number of adaptive single-nucleotide polymorphism loci associated with climate variables were identified. The genotype–environment association analysis showed that some cold-tolerant genes have played pivotal roles in cold environmental adaptation of B. scopulosa. These findings are further corroborated through evolutionary analysis of gene family and quantitative PCR validation. Population genomic analysis revealed 2 distinct genetic lineages in B. scopulosa. The western lineage showed higher genomic vulnerability and more rare cold-tolerance alleles, suggesting its heightened sensitivity to impending climate shifts, and should be given priority conservation in the management practices. Conclusions These findings provide novel insights into local adaptation and genomic vulnerability of B. scopulosa under climate change in the Qinling Mountains in East Asia. Additionally, the study also offers valuable guidance for formulating conservation strategies for the rare and endangered plants.

Abstract:Understanding the impact of climate change on the geographical distribution of species is a fundamental requirement for biodiversity conservation and resource management. Quercus oxyphylla, an evergreen oak endemic to China, plays a crucial role in maintaining the ecological stability in subtropical regions and high economic value attributed to its dark and high-density heartwood, but the existing resources are close to endangered. Currently, limited knowledge exists regarding its distribution and potential influences of climate change on suitable areas. This study utilized 63 occurrence records and Biomod2 platform, to predict changes in suitable areas for Q. oxyphylla under future climate change. The results revealed that (1) Q. oxyphylla showed a pattern of three disjunctive geographical centers in the eastern subregion of subtropical evergreen broad-leaved forest region (IVA): Qinling-Daba Mountains, Nanling Mountains and Wuyi Mountains center. Currently, the highly suitable areas concentrated in two zones divided by the Yangtze River, that is, the northern subtropical evergreen and deciduous broad-leaved forest zone (IVAii) and the mid-subtropical evergreen broad-leaved forest zone (IVAi). (2) The temperature-related variables, such as annual temperature range (Bio7), the mean diurnal range (Bio2), and annual mean temperature (Bio1), were identified as the key determinants of the distribution pattern. Because of its considerable climatic variations in temperature and water conditions, Q. oxyphylla's habitat displayed a wider climate niche and strong physiological tolerance to climate change. (3) Under future climate scenarios, the suitable area of the species was expected to overall expand with significant regional differences. The suitable area in IVAi was expected to expand significantly northward while that in IVAii was expected to gradually shrink. To address the impact of climate change, it is necessary to develop conservation plans focused around the three distribution centers, implement localized and regional conservation policies, and conduct educational outreach among local people.
Predicting the Potential Distribution of Quercus oxyphylla in China under Climate Change Scenarios

Abstract:Global climate changes are expected to profoundly shape species distribution. Quercus oxyphylla, a valuable evergreen broad-leaved tree species, is rigorously conserved and managed in China owing to its substantial scientific, economic, and ecological value. However, the impact of projected climate change on its future distribution and potential climatic drivers remains unclear. Here, a maximum entropy model (MaxEnt) was used to explore the distribution of Q. oxyphylla in China under current conditions and three future scenarios (SSP1-2.6, SSP2-4.5, and SSP5-8.5) for the 2050s and 2070s. We optimized the model using the 'ENMeval' package to obtain the best parameter combination (RM = 1, FC = LQHPT), and multiple evaluation metrics (AUC ≥ 0.9; TSS ≥ 0.6; Kappa ≥ 0.75) verified the high accuracy of the model and the reliability of the prediction results. We found the following: (1) The potential distribution of Q. oxyphylla spans across 28 provinces in China under current climatic conditions, predominantly in southern regions, with Sichuan exhibiting the largest suitable area for survival. The total suitable habitat covers 244.98 × 104 km2, comprising highly, moderately, and poorly suitable habitats of 51.66 × 104 km2, 65.98 × 104 km2, and 127.34 × 104 km2, respectively. (2) Under future climate conditions, the overall geographical boundaries of Q. oxyphylla are predicted to remain similar to the present one, with an increase of 10.29% in the 2050s and 11.31% in the 2070s. In the 2050s, the total suitable habitats for Q. oxyphylla under the three scenarios (SSP1-2.6, SSP2-4.5, and SSP5-8.5) might increase by 8.83%, 9.62%, and 12.42%, while in the 2070s they might increase by 10.39%, 17.21%, and 6.33%, respectively. (3) Moreover, the centroid of the suitable area is expected to migrate southwestward under the three scenarios in the future. (4) Annual precipitation, isothermality, and temperature annual range emerged as the main factors influencing the distribution of Q. oxyphylla, with contributions of 55.9%, 25.7%, and 13.5%, respectively. Our findings refined the spatial arrangement of Q. oxyphylla growth and revealed its climate resilience. This suggested that under climate change, Sichuan and Shaanxi are the optimal regions for cultivation and management, while appropriate conservation strategies should be formulated in Tibet and Hubei.

Abstract:There is substantial interest in uncovering the genetic basis of the traits underlying adaptive responses in tree species, as this information will ultimately aid conservation and industrial endeavors across populations, generations, and environments. Fundamentally, the characterization of such genetic bases is within the context of a genetic architecture, which describes the mutlidimensional relationship between genotype and phenotype through the identification of causative variants, their relative location within a genome, expression, pleiotropic effect, environmental influence, and degree of dominance, epistasis, and additivity. Here, we review theory related to polygenic local adaptation and contextualize these expectations with methods often used to uncover the genetic basis of traits important to tree conservation and industry. A broad literature survey suggests that most tree traits generally exhibit considerable heritability, that underlying quantitative genetic variation (QST) is structured more so across populations than neutral expectations (FST) in 69% of comparisons across the literature, and that single-locus associations often exhibit small estimated per-locus effects. Together, these results suggest differential selection across populations often acts on tree phenotypes underlain by polygenic architectures consisting of numerous small to moderate effect loci. Using this synthesis, we highlight the limits of using solely single-locus approaches to describe underlying genetic architectures and close by addressing hurdles and promising alternatives towards such goals, remark upon the current state of tree genomics, and identify future directions for this field. Importantly, we argue, the success of future endeavors should not be predicated on the shortcomings of past studies and will instead be dependent upon the application of theory to empiricism, standardized reporting, centralized open-access databases, and continual input and review of the community’s research.

Abstract:The subtropical forests of East Asia are renowned for their high plant diversity, particularly the abundance of ancient relict species. However, both the evolutionary history of these relict species and their capacity for resilience in the face of impending climatic changes remain unclear. Using whole‐genome resequencing data, we investigated the lineage differentiation and demographic history of the relict and endangered tree, Bretschneidera sinensis (Akaniaceae). We employed a combination of population genomic and landscape genomic approaches to evaluate variation in mutation load and genomic offset, aiming to predict how different populations may respond to climate change. Our analysis revealed a profound genomic divergence between the East and West lineages, likely as the result of recurrent bottlenecks due to climatic fluctuations during the glacial period. Furthermore, we identified several genes potentially linked to growth characteristics and hypoxia response that had been subjected to positive selection during the lineage differentiation. Our assessment of genomic vulnerability uncovered a significantly higher mutation load and genomic offset in the edge populations of B. sinensis compared to their core counterparts. This implies that the edge populations are likely to experience the most significant impact from the predicted climate conditions. Overall, our research sheds light on the historical lineage differentiation and contemporary genomic vulnerability of B. sinensis. Broadening our understanding of the speciation history and future resilience of relict and endangered species such as B. sinensis, is crucial in developing effective conservation strategies in anticipation of future climatic changes.

Abstract:The biogeographic history affects adaptive evolution by altering the pattern of genetic variation and provides the background upon which other evolutionary processes are operating. Assessment of the consequence of such eco‐evolutionary interactions is crucial to population's genetic diversity maintenance. Yet, considering how biogeographic and multiple evolutionary processes interact in complex environments to alter genetic diversity in conservation planning remains unresolved. The Pacific Northwest. Herein I document the impact of geographic landscapes and past climatic fluctuations on the evolutionary processes that drive population divergence of Populus trichocarpa by assembling genomic, climatic and species occurrence data. Based on inferences of demographic history of the species and tests for niche divergence, results show that the British Columbia (BC)‐North and BC‐South populations are ancestral and subject to more recent bottlenecks than the Oregon population. Consistently, ecological niche modelling illustrates that compared to the Last Glacial Maximum (~21 Ka), the present niche suitability of the BC‐North and ‐South populations is overall marginally inferior to that of the Oregon population. However, genomic analysis demonstrates that the Oregon harbours the lowest genetic diversity, possibly due to an extended bottleneck experienced by the population. Model prediction indicates that in the 2050s, the impact of climate change is minor compared to the niche suitability of the species as a whole, but climate change will likely result in considerable genetic shifts over two generations especially in the Oregon and BC‐South populations, which will attenuate along a northward gradient. This study underpins the long‐lasting impact of demographic histories and natural selection on genetic diversity, rather than highlighting the consequence of recent demographic events (e.g. bottleneck in the Holocene epoch; ~12 Ka) potentially cascading to eroding genetic diversity and yielding conservation urgency.

Abstract:Information about the resistance and adaptive potential of tree species and provenances is needed to select suitable planting material in times of rapidly changing climate conditions. In this study, we evaluate growth responses to climatic fluctuations and extreme events for 12 provenances of northern red oak ( Quercus rubra L.) that were tested across three trial sites with distinct environmental conditions in Germany. Six provenances each were sourced from the natural distribution in North America and from introduced stands in Germany. We collected increment cores of 16 trees per provenance and site. Dendroecological methods were used to compare provenance performance and establish climate-growth relationships to identify the main growth limiting factors. To evaluate the provenance response to extreme drought and frost events, three site-specific drought years were selected according to the Standardized Precipitation Evapotranspiration Index (SPEI) and 2010 as a year with an extreme late frost event. Resistance indices for these years were calculated and assessed in relation to overall growth performance. We observed a high variation in growth and in the climate sensitivity between sites depending on the prevailing climatic conditions, as well as a high intra-specific variation. Overall, summer drought and low temperatures in the early growing season appear to constrain the growth of red oak. The resistance of provenances within sites and extreme years showed considerable rank changes and interaction effects. We did not find a trade-off between growth and resistance to late frost, namely, fast growing provenances had a high frost hardiness. Further, there was no evidence for a trade-off between growth and drought hardiness. Still, responses to drought or late frost differ between provenances, pointing to dissimilar adaptive strategies. Provenances from introduced (i.e. German) stands represent suitable seed sources, as they combine a higher growth and frost hardiness compared to their North American counterparts. Drought hardiness was slightly higher in the slow-growing provenances. The results provide a better understanding of the variable adaptive strategies between provenances and help to select suitable planting material for adaptive forest management.

Abstract:In situ digestion kinetics of neutral detergent insoluble nitrogen (NDIN) from alfalfa (Medicago sativa L.) harvested at one-tenth bloom and eastern gamagrass (Tripsacum dactyloides L.) harvested at the boot (GGB), anthesis (GGA), and physiological maturity (GGM) stages of growth were determined with nonlinear regression techniques. Whole-plant tissue and associated leaf and stem fractions were incubated in the ventral rumen simultaneously. On a wholeplant basis, potential extents of degradation were particularly high (> or =904 g/kg NDIN) for GGB and GGA, relative to those of GGM and alfalfa (772 and 658 g/kg NDIN, respectively). For all plant parts, degradation rates of NDIN were faster (P<.05) for alfalfa than for all gamagrass forages. Degradation rate of NDIN did not differ (P>.05) across maturities for any gamagrass tissue type. These results indicate 1) that phenological development and lignification do not limit the rate of NDIN degradation in gamagrass forages but do markedly limit the potential extent of NDIN availability and 2) that most of the NDIN in these forages is potentially available in the rumen and can contribute to the ruminal N supply. Our secondary objective was to compare estimates of N escaping ruminal degradation that were determined on the basis of NDIN degradation kinetics (NDIN method) with those determined traditionally, on the basis of total residual N. The NDIN method mathematically eliminates all neutral detergent soluble N from consideration as part of the pool of dietary N potentially escaping the rumen intact. Estimates of rumen escape nitrogen determined on the basis of degradation rates of NDIN were consistently less than corresponding estimates that were determined on the basis of total residual N. When ruminal escape N that was determined with the NDIN method was regressed on corresponding estimates with the total residual N method, the slopes of the regression lines were .53 and .66 for assumed passage rates of .02 and .06 h(-1), respectively. For the forages evaluated in this study, these results indicate that neutral detergent soluble N may make important contributions to the pool of N escaping ruminal degradation.

Abstract:Environmental factors are strong drivers of local adaptation in forest tree species. Toona ciliata var. pubescens, an endangered tree species endemic to China, is widely distributed across Eastern and Southwestern China. In this study, we used 8 genomic microsatellite markers and 17 EST-SSR markers across nine populations from the Yunnan–Kweichow Plateau and Eastern China, to explore the adaptive variation and genetic structure of T. ciliata var. pubescens. Patterns of population structure were apparent using a Bayesian clustering program, STRUCTURE, which identified four distinct clusters. We identified four outlier loci that were potentially under selection using the Dirichlet-multinomial and hierarchic simulation models. Through the Mantel test, it was found that geographic and climatic factors have jointly affected the genetic structure of T. ciliata var. pubescens in the study area. Based on redundancy analysis (RDA), it was shown that the correlation between climatic variables associated with variation is stronger than that of geographic variables. It is worth mentioning that the eight alleles from outlier loci have potentially adaptive and are associated with either precipitation or temperature variables. All analyses revealed high genetic diversity and significant genetic differentiation in the populations of T. ciliata var. pubescens. This indicated that the climatic variables including precipitation and temperature are drivers of local adaptation in the populations of T. ciliata var. pubescens.
