Abstract:Heat stress affects the physiology and production performance of Chinese Holstein dairy cows. As such, the selection of heat tolerance in cows and elucidating its underlying mechanisms are vital to the dairy industry. This study aimed to investigate the heat tolerance associated genes and molecular mechanisms in Chinese Holstein dairy cows using a high-throughput sequencing approach and bioinformatics analysis. Heat-induced physiological indicators and milk yield changes were assessed to determine heat tolerance levels in Chinese Holstein dairy cows by Principal Component Analysis method following Membership Function Value Analysis. Results indicated that rectal temperature (RT), respiratory rate (RR), and decline in milk production were significantly lower (p < 0.05) in heat tolerant (HT) cows while plasma levels of heat shock protein (HSP: HSP70, HSP90), and cortisol were significantly higher (p < 0.05) when compared to non-heat tolerant (NHT) Chinese Holstein dairy cows. By applying RNA-Seq analysis, we identified 200 (81 down-regulated and 119 up-regulated) significantly (|log2fold change| >= 1.4 and p <= 0.05) differentially expressed genes (DEGs) in HT versus NHT Chinese Holstein dairy cows. In addition, 14 of which were involved in protein-protein interaction (PPI) network. Importantly, several hub genes (OAS2, MX2, IFIT5 and TGFB2) were significantly enriched in immune effector process. These findings might be helpful to expedite the understanding for the mechanism of heat tolerance in Chinese Holstein dairy cows.

Abstract:Heat stress imposes a challenge to the dairy industry, even in northern latitudes. In this study, publicly available weather station data were combined with test-day records for milk, fat, and protein yields to identify the temperature–humidity index (THI) thresholds at which heat load starts affecting milk production traits in Canadian Holstein cows. Production loss per THI unit above the threshold for each trait was estimated. Test-day records from 2010 to 2019 from 166,749 cows raised in Ontario and from 221,214 cows raised in Quebec were analyzed. Annual economic losses (EL) due to heat stress were estimated from the average losses of fat and protein yields based on the annual average of 156 d with THI exceeding the calculated thresholds. Average thresholds for the daily maximum (THI max ) and daily average (THI avg ) THI estimated across lactations in both provinces were THI max (THI avg ) 68 (64), 57 (50), and 60 (58) for milk, fat, and protein yield, respectively, indicating that milk components are more sensitive to heat stress. An EL of about $34.5 million per year was estimated. Our findings contribute to an initial investigation into the impact of heat stress on the Canadian dairy industry and provide a basis for genetic studies on heat tolerance.

Abstract:Heat stress has been recognized as a serious problem in dairy farms around the world due to the increasing heat waves and higher genetic potential of dairy cows. In Chile, milk production is concentrated in the southern regions of the country, where animals graze all year around, consequently being exposed directly to environmental conditions. Nevertheless, there are few studies conducted in Chile that have evaluated at the commercial level the impact of heat stress on milk production. The aim of this study was to assess the effects of summer conditions, across periods, on the milk production of cows at different stages of lactation in a dairy farm located in Southern Chile. Daily meteorological and milk yield records of three summers from a dairy farm were collected to characterize the relationship between two thermal stress indices and milk yield. The thermal comfort indices used were the comprehensive climate index (CCI), and the adjusted temperature humidity index (THIadj). The average values of CCI and THIadj were dependent on the period (P < 0.0001) with maximum CCI of 40.2 °C, 31.7 °C, and 27.5 °C for the 2012-2013, 2015-2016, and 2016-2017 periods, respectively. A similar response was recorded when THIadj was used (85.5, 78.0, and 73.9, respectively). In the 2012-2013 summer, 44.4% of the days presented conditions of heat stress (CCI ≥23), a value that fell to 26.7% in the summer of 2015-2016 and only 5.6% in the 2016-2017. On the opposite, when the THIadj was used, these values were 50%, 48.9%, and 5.6%, respectively. In conclusion, both comfort thermal indices are good tools to determine the risk of thermal stress in dairy cows, with a large variation between the three summer periods but also between indices. Likewise, cows in the early and mid-lactation periods are more affected in terms of milk yield.

Abstract:A versatile meteorological index for predicting heat stress in dairy cattle remains elusive. Despite numerous attempts at developing such indices and widespread use of some, there is growing skepticism about the accuracy and adequacy of the existing indices as well as the general statistical approach used to develop them. At the same time, precision farming of high-yielding animals in a drastically changing climate calls for more effective prediction and alleviation of heat stress. The present paper revisits classical work on human biometeorology, particularly the apparent temperature scale, to draw inspiration for advancing research on heat stress in dairy cattle. The importance of a detailed, mechanistic understanding of heat transfer and thermoregulation is demonstrated and reiterated. A model from the literature is used to construct a framework for identifying and characterizing conditions of potential heat stress. New parameters are proposed to translate the heat flux calculations based on heat-balance models into more tangible and more useful meteorological indices, including an apparent temperature for cattle and a thermoregulatory exhaustion index. A validation gap in the literature is identified as the main hindrance to the further development and deployment of heat-balance models. Recommendations are presented for systematically addressing this gap in particular and continuing research within the proposed framework in general.

Abstract:Lipocalin 2 (Lcn2), a member of the lipocalin family that carries small lipophilic ligands, has gained recent attention as both a potential biomarker and a modulator of human cancers. Here we describe recent findings of the functions of Lcn2 in breast cancer and the potential mechanisms that underlie its actions. Lcn2 has been shown to induce the epithelial to mesenchymal transition (EMT) in breast cancer cells and to promote breast tumor invasion. Estrogen receptor α may participate in the pathway that leads to Lcn2-induced EMT. Preliminary evidence also suggests that Lcn2 may be used as a potential non-invasive urinary biomarker of breast cancer. Elevated levels of Lcn2 have also been reported in other human cancers. The potential roles of Lcn2 in some other epithelial tumors as well as leukemia are also reviewed and discussed here.

Abstract:Heat stress exerts a substantial effect on dairy production. The temperature and humidity index (THI) is widely used to assess heat stress in dairy operations. Herein, we review the effects of high temperature and humidity on body temperature, feed intake, milk production, follicle development, estrous behavior, and pregnancy in dairy cows. Analyses of the effects of THI on dairy production have shown that body temperature is an important physiological parameter in the evaluation of the health state of dairy cows. Although THI is an important environmental index and can help to infer the degree of heat stress, it does not reflect the physiological changes experienced by dairy cows undergoing heat stress. However, the simultaneous measurement of THI and physiological indexes (e.g., body temperature) would be very useful for improving dairy production. The successful development of automatic detection techniques makes it possible to combine THI with other physiological indexes (i.e., body temperature and activity), which could help us to comprehensively evaluate heat stress in dairy cows and provide important technical support to effectively prevent heat stress.

Abstract:Heat stress is a major challenge in dairy cattle farming and leads to economic losses. This study aims to determine the impact of heat stress on dairy cattle under a hot and humid climate. Milk yield test records between 2015 to 2019 were collected from 532 first lactating Friesian crossbred cows. Hourly values of temperature (dry bulb), relative humidity, surface wind speed, and solar radiation over 24 h collected from the national weather station in Malaysia were averaged and computed into four temperature indexes (TI). The heat stress threshold was identified using a two-slope broken line regression and the magnitude of milk loss was determined by the degree of decline fitted to a Legendre polynomial regression. Environmental parameters and TI values were observed to be influenced by the dry and monsoon season. Thresholds reported ranged between 73 (TI 4 ) to 79 (TI 3 ) according to the respective TI measured. The index values were higher than the reported studies in temperate, semi-arid and mediterranean climates. The largest milk decline was determined as 0.181 kg in mild, 0.267 kg in moderate, and 0.523 kg in extreme heat stress conditions, respectively, when using the conventional TI 1 . Thresholds determined in the current study can be used for annual forecasting and targeted heat stress mitigation efforts.

Abstract:The present study was undertaken to determine the effect of heat stress on milk production (test day milk yield) and compositional traits (fat%, protein%, fat yield, protein yield) as well as to observe the pattern of response to increasing heat load on these traits in Jersey crossbred cows, maintained at ICAR-National Dairy Research Institute, Eastern Regional Station, Kalyani, West Bengal, India. The weather information, obtained from the NASA POWER database based on the location of the farm latitude and longitude, was used to calculate the Temperature Humidity Index (THI). To analyze the data, a linear model was fitted to the milk production and compositional records, which were adjusted for additive genetic effect of animal, permanent environmental effect of animals and known environmental sources of variations. Subsequently, a segmented linear regression model was fitted, and the least squares estimates of production and composition traits in different classes of THI were used as the dependent variable. Two THI break-points (BP) for milk yield and one THI BP for fat yield, protein %, and protein yield were found. The first and second BP for milk yield was at THI 59 and 77, respectively, with a significant decline in milk yield of -0.04 kg/unit of THI at second BP. The BP for fat and protein yield was at THI 76, with a decline rate of -1.18 and - 0.61 g/unit of THI increase, respectively. The findings revealed the significant adverse effects of THI on milk production and composition traits in Jersey crossbred cattle.

Abstract:Heat stress with measurable effects in dairy cattle is a growing concern in temperate regions. Heat stress in temperate regions differs between environments with different geophysical characteristics. Microclimates specific to each environment were found to greatly impact at what level heat stress occurs and will occur in the future. The landlocked state of Baden-Württemberg, Germany, provides several different environments, hence, a good case-study. Temperature-Humidity Index (THI) from 17 weather stations for the years 2003 to 2022 was calculated and milking yields from 22 farms for the years 2017 to 2022 were collected. The occurrences and evolving patterns of heat stress were analyzed with the use of a THI, and the effect of heat stress on milk yield was analyzed based on milking records from Automated Milking Systems. Daily average THI was calculated using hourly readings of relative humidity and ambient temperature, disregarding solar radiation and wind, as all animals were permanently stabled. Based on studies conducted in Baden-Württemberg and neighboring regions, cited ahead in the section of THI, THI = 60 was the threshold for heat stress occurrence. Findings show that the heat stress period varied between stations from 64 to 120 d with THI ≥ 60 in a year. This aligns with yearly and summer averages, also steadily increasing from May to September. The length of the heat stress period was found to increase 1 extra day every year. Extreme weather events such as heat waves did not increase the heat stress period of that year in length but increased the average THI. Milk yield was found to be significantly (α = 0.05) different between counties grouped into different zones according to heat stress severity and rate of increase in daily average THI. Future attempts at managing heat stress on dairy cattle farms in the temperate regions should account for microclimate, as geographical proximity does not mean that the increase in heat stress severity will be the same in the 2 neighboring areas.

Abstract:There are clear signs that milk production growth is leveling off, and recently even declining, in China. Heat stress is one of the main reasons for the recent reduction in milk production. In this study, we computed the change in milk production as a result of heat stress in major milk production areas in China. We constructed a temperature–humidity index (THI) spatial layer to understand the monthly distribution of heat and moisture. We documented specific areas in northern China where cattle were at high risk to heat stress in specific months. THI values exceeded the threshold above which milk production declines during months of June, July, and August. Especially during July, the THI value was higher than the production threshold in recent years (2008 to 2016) and in projected future scenarios (2050 and 2070). THI-based milk yield losses were up from 0.7 to about 4 kg per cow per day in July 2016. These losses are projected to increase from 1.5 to 6.5 kg in 2050 and 2 to 7.2 kg in 2070 (representing production losses between 15 and 50%). These results suggest that climate change will have significant consequences for the dairy sector in major milk-producing areas in China. Our results are useful in identifying areas susceptible to heat stress where adaptive livestock management practices are needed to prevent significant production decreases.

Abstract:Dairy cattle are highly susceptible to heat stress. Heat stress causes a decline in milk yield, reduced dry matter intake, reduced fertility rates, and alteration of physiological traits (e.g., respiration rate, rectal temperature, heart rates, pulse rates, panting score, sweating rates, and drooling score) and other biomarkers (oxidative heat stress biomarkers and stress response genes). Considering the significant effect of global warming on dairy cattle farming, coupled with the aim to reduce income losses of dairy cattle farmers and improve production under hot environment, there is a need to develop heat tolerant dairy cattle that can grow, reproduce and produce milk reasonably under the changing global climate and increasing temperature. The identification of heat tolerant dairy cattle is an alternative strategy for breeding thermotolerant dairy cattle for changing climatic conditions. This review synthesizes information pertaining to quantitative genetic models that have been applied to estimate genetic parameters for heat tolerance and relationship between measures of heat tolerance and production and reproductive performance traits in dairy cattle. Moreover, the review identified the genes that have been shown to influence heat tolerance in dairy cattle and evaluated the possibility of using them in genomic selection programmes. Combining genomics information with environmental, physiological, and production parameters information is a crucial strategy to understand the mechanisms of heat tolerance while breeding heat tolerant dairy cattle adapted to future climatic conditions. Thus, selection for thermotolerant dairy cattle is feasible.

Negative association between high temperature-humidity index and milk performance and quality in Korean dairy system: big data analysis

Abstract:The aim of this study was to investigate the effects of heat stress on milk traits in South Korea using comprehensive data (dairy production and climate). The dataset for this study comprised 1,498,232 test-day records for milk yield, fat- and protein-corrected milk, fat yield, protein yield, milk urea nitrogen (MUN), and somatic cell score (SCS) from 215,276 Holstein cows (primiparous: n = 122,087; multiparous: n = 93,189) in 2,419 South Korean dairy herds. Data were collected from July 2017 to April 2020 through the Dairy Cattle Improvement Program, and merged with meteorological data from 600 automatic weather stations through the Korea Meteorological Administration. The segmented regression model was used to estimate the effects of the temperature-humidity index (THI) on milk traits and elucidate the break point (BP) of the THI. To acquire the least-squares mean of milk traits, the generalized linear model was applied using fixed effects (region, calving year, calving month, parity, days in milk, and THI). For all parameters, the BP of THI was observed; in particular, milk production parameters dramatically decreased after a specific BP of THI (p < 0.05). In contrast, MUN and SCS drastically increased when THI exceeded BP in all cows (p < 0.05) and primiparous cows (p < 0.05), respectively. Dairy cows in South Korea exhibited negative effects on milk traits (decrease in milk performance, increase in MUN, and SCS) when the THI exceeded 70; therefore, detailed feeding management is required to prevent heat stress in dairy cows.

Abstract:Context: Heat stress is an increasing concern for the Australian dairy industry. Aims: This study aimed to evaluate the effect of temperature–humidity index (THI) on rumination time (RT), milk yield and quality, and milking frequency in a pasture-based voluntary-movement automatic milking system (AMS). Methods: Data were collected from the University of Melbourne Dookie College AMS farm for 3 years (June 2016 to March 2019). Daily RT was collected through the transponder collar (Qwes-HR, Lely). Climatic data (maximum ambient temperature and relative humidity) were obtained from the Dookie Meteorological station to calculate daily maximum THI (THI max ). Key results: Daily milk yield increased with a rising THI max to 65, then declined after THI max 65. Milking frequency was highest at THI max 90, followed by a steady decline afterwards. Rumination time was maximum at mid-range THI max and declined for high and low values. Conclusions: The findings of this study clearly indicated that under pasture-based voluntary-movement AMS, high THI resulted in a drop in the milk yield, milking frequency and RT. Implications: With the provision of automation of data collection from AMS, further study with mathematical modelling describing the daily patterns and thresholds in conjunction with the different heat stress levels can be useful for assessing animal welfare and to mitigate heat stress and seek alternative management strategies.

Abstract:Climate change is a problem that causes many environmental issues that impact the productivity of livestock species. One of the major issues associated with climate change is an increase of the frequency of hot days and heat waves, which increases the risk of heat stress for livestock species. Dairy cattle have been identified as being susceptible to heat stress due to their high metabolic heat load. Studies have shown heat stress impacts several biological processes that can result in large economic consequences. When heat stress occurs, dairy cattle employ several physiological and cellular mechanisms in order to dissipate heat and protect cells from damage. These mechanisms require an increase and diversion in energy toward protection and away from other biological processes. Therefore, in turn heat stress in dairy cattle can lead numerous issues including reductions in milk production and reproduction as well as increased risk for disease and mortality. This indicates a need to select dairy cattle that would be thermotolerant. Various selection strategies to confer thermotolerance have been discussed in the literature, including selecting for reduced milk production, crossbreeding with thermotolerant breeds, selecting based on physiological traits and most recently selecting for enhanced immune response. This review discusses the various issues associated with heat stress in dairy cattle and the pros and cons to the various selection strategies that have been proposed to select for thermotolerance in dairy cattle.

Abstract:<p>The objectives of this study were to investigate the effects of the cow-related factors on rectal temperature (RT) and respiration rate (RR) of lactating dairy cows under different heat stress (HS) conditions and establish the temperature-humidity index (THI) thresholds at which RT and RR begin to increase for cows in China. Cow-related factors included body posture (standing and lying), milk yield (<26 kg/d, ≥ 26–39 kg/d, and ≥39 kg/d), days in milk (≤60 d, > 60 and ≤ 150 d, and >150 d), and parity (1, 2, and ≥3). Records of RT, RR, and individual characteristics were collected from July to October 2020 on a commercial dairy farm in Northern China, where 826 Holstein lactating cows were measured. Using the broken-stick regression models and the entire dataset, the THI thresholds for RT and RR were 69.8 and 67.1, respectively. Therefore, the heat stress conditions during this study were classified according to the THI levels as thermoneutrality (TN, 60 < THI ≤ 67), mild (67 < THI ≤ 72), moderate (72 < THI ≤ 80), and severe (80 < THI ≤ 86). Results showed that lying cows exhibited the higher RT and RR but the lower THI threshold for RT (68.8 vs. 70.7) and RR (65.6 vs. 68.4) than standing cows; milk yield is positively associated with the values of RT and RR under TN or HS conditions, and the THI thresholds for RT (70.2 vs. 70.0 vs. 68.0) and RR (68.1 vs. 64.7 vs. 64.7) were progressively lower for low-yielding, middle-yielding, and high-yielding cows; there was a significant increase in RT and RR in early-lactation cows compared to late-lactation cows under TN or HS conditions (P < 0.001), and the lowest THI threshold (67.8 for RT and 64.7 for RR) was observed in early-lactation cows, followed by mid-lactation cows (68.2 for RT and 65.3 for RR) and late-lactation cows (70.0 for RT and 67.3 for RR); the effects of parity were not significant on RT (P > 0.05), but significant on RR (P < 0.001). The THI thresholds for RT (69.2) and RR (66.0) were lowest for cows in 3rd-parity and higher, followed by cows in 2nd-parity (70.0 for RT and 68.9 for RR) and 1st-parity (70.7 for RT and 66.6 for RR). This study highlighted the great significance of considering the cow-related factors in heat stress responses and THI threshold assessment. For dairy cows in China, we suggest that cooling should be initiated when THI reaches 65 to 66.</p>

Abstract:Abstract Modeling the individual animal response to heat stress (HS) conditions is challenging because of the complex interactions that characterize the system behavior. In explaining the resulting animal behavior, the dynamicity, nonlinearity, and delays in the HS response are often unaccounted for or misinterpreted. The system dynamics (SD) methodology, a mathematical modeling approach based on feedback loop structures, allows for modeling and understanding the behavior of complex systems over time. By applying SD methodology, this study developed a preliminary conceptual model to capture the cow response and observed milk yield (MY) under HS. The data on the temperature-humidity index (THI) and MY used for model development were collected from a dairy cattle farm in August 2021. The parameters related to the HS response of 20 selected cows were used for calibration and parameterization of the model. To minimize the effect of the lactation stage on milk production and model results, 20 cows were selected for days in milk (DIM) to be between 70 and 220 d. After the parameter calibration using MY data, it was found that the historical data pattern of 13 out of 20 cows followed the expected behavioral pattern generated by the model. In contrast, the behavior of the remaining seven cows did not align with that generated by the model. Therefore, based on their patterns, the cows were identified as fitting or non-fitting the model’s structure. The structure of the model captured the effect of HS on fitting cows with high accuracy (mean absolute percentage error, MAPE < 5%; R2 > 0.6; concordance correlation coefficient, CCC > 0.6). At the same time, the behavior of the non-fitting cows could not be explained by the defined parameter space. We believe they either had heat-resistant behavior or experienced different biological delays than average. Based on the obtained results, the evaluation of parameter values should be done only for the fitting cows, as the work aimed to develop a model to understand the HS response. The behaviors generated by the model can help farmers and decision-makers distinguish heat-sensitive from heat-tolerant cows and quantify the animal response in terms of MY so that mitigation strategies can be implemented.

Abstract:Numerous climatic indices have been utilized to predict the effect of hot, and cold, climatic conditions on animal production and welfare. To date, the dairy industry has relied extensively on the Temperature Humidity Index (THI) to predict adverse climatic conditions; however, neither solar radiation nor air movement is accounted for in the THI equation. The Dairy Heat Load Index (DHLI) was initially developed as an alternative climate index. In its current format, the DHLI does not account for the effects of heat load mitigation strategies, such as shade, which decreases the negative effects of hot climatic conditions on lactating cows. Therefore, this experiment aimed to determine the effectiveness of the DHLI as a predictor of heat load responses in both shaded and unshaded cows, as compared with the THI. Forty lactating Holstein Friesian (n = 40) cows were selected and paired based on live weight, milk yield, and days in milk. One cow from each pair was randomly allocated to one of two treatments: shaded (n = 20) or unshaded (n = 20). Cows were given 7 days to acclimate prior to the commencement of data collection. After 28 days, cows were transitioned into the alternate treatment in a crossover design and given 7 days to acclimate prior to data collection. Behavioral observations (0800, 1200, 1400, and 1800 h daily), daily milk yield (kg), milk composition (various days), and vaginal temperature (TVAG, °C; 5 pairs/week, over a 4-week rotation) were recorded. Overall, data from this experiment indicated that the DHLI was a better predictor of standing and feeding behaviors in unshaded cows and drinking behaviors in shaded cows. Conversely, the THI was a better predictor of standing behavior and shade usage in shaded cows. Furthermore, the THI was a better predictor of mean panting score (MPS) in shaded cows, whereas the DHLI performed better in unshaded cows. Additionally the DHLI was a better predictor of TVAG in these cows. Finally, when evaluating the 7-day average of each climatic index, the DHLI was a better predictor of change in milk yield. Incorporation of additional animal and management factors is required if the DHLI is to become an effective heat load management tool.

Abstract:Introduction: Increasing global temperatures present a significant challenge to livestock production. The dry period is an important stage in the production cycle of cow, and environmental heat stress (HS) during this period can have adverse effects on the subsequent lactation performance. In this study, we aimed to investigate the effects of HS on endocrine, thermoregulatory, and lactation parameters in heat-tolerant dry cows (HTDC) and heat-sensitive dry cows (HSDC). Methods: We measured the respiratory rate (RR), body temperature (BT), and temperature-humidity index (THI) in 66 dry cows during HS. The slopes of RR and BT to THI were determined through analysis of measurements and dry cows background information using a mixed-effects model. Subsequently, the heat tolerance or sensitivity of dry cows was assessed using clustering method (HTDC = 19 and HSDC = 47). Results: Compared with that of HSDC, the RR of HTDC significantly increased after exposure to HS ( p < 0.05). The average reduction in milk yield from new lactation to the previous lactation was significantly lower in HTDC compared to HSDC ( p < 0.05). Plasma cortisol and non-esterified fatty acid levels were significantly lower in HTDC compared to HSDC ( p < 0.05), while plasma triiodothyronine ( p = 0.07) and growth hormone ( p = 0.08) levels tended to be higher in HTDC relative to HSDC. Discussion: HTDC can more effectively alleviate the impacts of HS through their superior thermoregulation and metabolism, thereby ensuring optimal postpartum lactation performance.
