Abstract:Cervical cancer is the most common reproductive malignancy in the female reproductive system. The incidence rate and mortality rate of cervical cancer among women in China are high. In this study, Raman spectroscopy was used to collect tissue sample data from patients with cervicitis, cervical precancerous low-grade lesions, cervical precancerous high-grade lesions, well differentiated squamous cell carcinoma, moderately differentiated squamous cell carcinoma, poorly differentiated squamous cell carcinoma and cervical adenocarcinoma. The collected data were preprocessed using an adaptive iterative reweighted penalized least squares (airPLS) algorithm and derivatives. Convolutional neural network (CNN) and residual neural network (ResNet) classification models were constructed to classify and identify seven types of tissue samples. The attention mechanism efficient channel attention network (ECANet) module and squeeze-and-excitation network (SENet) module were combined with the established CNN and ResNet network models, respectively, to make the models have better diagnostic performance. The results showed that efficient channel attention convolutional neural network (ECACNN) had the best discrimination, and the average accuracy, recall, F1 and AUC values after five cross-validations could reach 94.04%, 94.87%, 94.43% and 96.86%, respectively.

Artificial Classification of Cervical Squamous Lesions in ThinPrep Cytologic Tests Using a Deep Convolutional Neural Network.

Abstract:The diagnosis of squamous cell carcinoma requires the accurate classification of cervical squamous lesions in the ThinPrep cytologic test (TCT). It primarily relies on a pathologist's interpretation under a microscope. Deep convolutional neural networks (DCNN) have played an increasingly important role in digital pathology. However, they have not been applied to diverse datasets and externally validated. In the present study, a DCNN model based on VGG16 and an ensemble training strategy (ETS) based on 5-fold cross-validation was employed to automatically classify normal and abnormal cervical squamous cells from a multi-center dataset. First, we collected a dataset comprising 82 TCT samples from four hospitals and fine-tuned our model twice on the dataset with and without the ETS. Then, we compared the classifications obtained from the models with those provided by two skilled pathologists to discriminate the performance of the models in terms of classification accuracy and efficiency. Finally, paired sample t-tests were used to validate the consistency between the classification provided by the proposed methods and that of the pathologists. The results showed that ETS slightly, though not significantly, improved the classification accuracy compared with that of the pathologists: P0=0.387>0.05 (DCNN without ETS vs. DCNN with ETS), P1=0.771>0.05 (DCNN with ETS vs. pathologist 1), P2=0.489>0.05 (DCNN with ETS vs. pathologist 2). The DCNN model was almost 6-fold faster than that of the pathologists. The accuracy of our automated scheme was similar to that of the pathologists, but a higher efficiency in the accurate identification of cervical squamous lesions was provided by the scheme. This result allows for wider and more efficient screening and may provide a replacement for pathologists in the future. Future research should address the viability of the practical implementation of such DCNN models in the laboratory setting.
Diagnosis of Cervical Precancerous Lesions Based on Multimodal Feature Changes.

Abstract:To realize the automatic diagnosis of cervical intraepithelial neoplasia (CIN) cases by preacetic acid test and postacetic acid test colposcopy images, this paper proposes a method of cervical precancerous lesion diagnosis based on multimodal feature changes. First, the preacetic acid test and postacetic acid test colposcopy images were registered based on cross-correlation and projection transformation, and then the cervical region was extracted by the k-means clustering algorithm. Finally, a deep learning network was used to extract features and classify the preacetic acid test and postacetic acid test cervical images after registration. Finally, the proposed method achieves a classification accuracy of 86.3%, a sensitivity of 84.1%, and a specificity of 89.8% in 60 test cases. Experimental results show that this method can make better use of the multimodal features of colposcopy images and has lower requirements for medical staff in the process of data acquisition. It has certain clinical significance in cervical cancer precancerous lesion screening systems.

Abstract:Introduction: Cervical cancer is a high incidence of cancer in women and cervical precancerous screening plays an important role in reducing the mortality rate. Methods: In this study, we proposed a multichannel feature extraction method based on the probability distribution features of the acetowhite (AW) region to identify cervical precancerous lesions, with the overarching goal to improve the accuracy of cervical precancerous screening. A k-means clustering algorithm was first used to extract the cervical region images from the original colposcopy images. We then used a deep learning model called DeepLab V3+ to segment the AW region of the cervical image after the acetic acid experiment, from which the probability distribution map of the AW region after segmentation was obtained. This probability distribution map was fed into a neural network classification model for multichannel feature extraction, which resulted in the final classification performance. Results: Results of the experimental evaluation showed that the proposed method achieved an average accuracy of 87.7%, an average sensitivity of 89.3%, and an average specificity of 85.6%. Compared with the methods that did not add segmented probability features, the proposed method increased the average accuracy rate, sensitivity, and specificity by 8.3%, 8%, and 8.4%, respectively. Conclusion: Overall, the proposed method holds great promise for enhancing the screening of cervical precancerous lesions in the clinic by providing the physician with more reliable screening results that might reduce their workload.

Abstract:<p>Cervical cancer has a long latency, and early screening greatly reduces mortality. In this study, cervical adenocarcinoma and cervical squamous cell carcinoma tissue data were collected by Raman spectroscopy, and then, the adaptive iteratively reweighted penalized least squares (airPLS) algorithm and Vancouver Raman algorithm (VRA) were used to subtract the background of the collected data. The following five feature extraction algorithms were applied: partial least squares (PLS), principal component analysis (PCA), kernel principal component analysis (KPCA), isometric feature mapping (isomap) and locally linear embedding (LLE). The k-nearest neighbour (KNN), extreme learning machine (ELM), decision tree (DT), backpropagation neural network (BP), genetic optimization backpropagation neural network (GA-BP) and linear discriminant analysis (LDA) classification models were then established through the features extracted by different feature extraction algorithms. In total, 30 types of classification models were established in this experiment. This research includes eight good models, airPLS-PLS-KNN, airPLS-PLS-ELM, airPLS-PLS-GA-BP, airPLS-PLS-BP, airPLS-PLS-LDA, airPLS-PCA-KNN, airPLS-PCA-LDA, and VRA-PLS-KNN, whose diagnostic accuracy was 96.3 %, 95.56 %, 95.06 %, 94.07 %, 92.59 %, 85.19 %, 85.19 % and 85.19 %, respectively. The experimental results showed that the model established in this article is simple to operate and highly accurate and has a good reference value for the rapid screening of cervical cancer.</p>

Abstract:Breast and cervical cancers are becoming the leading causes of death among women worldwide, but current diagnostic methods have many drawbacks, such as being time-consuming and high cost. Raman spectroscopy, as a rapid, reliable, and non-destructive spectroscopic detection technique, has achieved many breakthrough results in the screening and prognosis of various cancer tumors. Therefore, in this study, Raman spectroscopy technology was used to diagnose breast cancer and cervical cancer. A total of 225 spectra were recorded from 87 patients with cervical cancer, 60 patients with breast cancer, and 78 healthy individuals. The obvious difference in Raman spectrum between the three groups was mainly shown at 809 cm −1 (tyrosine), 958 cm −1 (carotenoid), 1004 cm −1 (phenylalanine), 1154 cm −1 (β-carotene), 1267 cm −1 (Amide III), 1445 cm −1 (phospholipids), 1515 cm −1 (β-carotene), and 1585 cm −1 (C = C olefinic stretch). We used one-way analysis of variance for these peaks and demonstrated that they were significantly different. Then, we combined the detected Raman spectra with multivariate statistical calculations using the principal component analysis-linear discrimination algorithm (PCA-LDA) to discriminate between the three groups of collected serum samples. The diagnostic results showed that the model's accuracy, precision, recall, and F1 score of the model were 92.90%, 92.62%, 92.10%, and 92.36%, respectively. These results suggest that Raman spectroscopy can achieve ultra-sensitive detection of serum, and the developed diagnostic models have great potential for the prognosis and simultaneous screening of cervical and breast cancers.

Abstract:The mortality of ovarian cancer is closely related to its poor rate of early detection. In the search of an efficient diagnosis method, Raman spectroscopy of blood features as a promising technique allowing simple, rapid, minimally-invasive and cost-effective detection of cancers, in particular ovarian cancer. Although Raman spectroscopy has been demonstrated to be effective to detect ovarian cancers with respect to normal controls, a binary classification remains idealized with respect to the real clinical practice. This work considered a population of 95 woman patients initially suspected of an ovarian cancer and finally fixed with a cancer or a cyst. Additionally, 79 normal controls completed the ensemble of samples. Such sample collection proposed us a study case where a ternary classification should be realized with Raman spectroscopy of the collected blood samples coupled with suitable spectroscopic data treatment algorithms. In the medical as well as data points of view, the appearance of the cyst case considerably reduces the distances among the different populations and makes their distinction much more difficult, since the intermediate cyst case can share the specific features of the both cancer and normal cases. After a proper spectrum pretreatment, we first demonstrated the evidence of different behaviors among the Raman spectra of the 3 types of samples. Such difference was further visualized in a high dimensional space, where the data points of the cancer and the normal cases are separately clustered, whereas the data of the cyst case were scattered into the areas respectively occupied by the cancer and normal cases. We finally developed and tested an ensemble of models for a ternary classification with 2 consequent steps of binary classifications, based on machine learning algorithms, allowing identification with sensitivity and specificity of 81.0% and 97.3% for cancer samples, 63.6% and 91.5% for cyst samples, 100% and 90.6% for normal samples.
High-Precision Intelligent Cancer Diagnosis Method: 2D Raman Figures Combined with Deep Learning

Abstract:Raman spectroscopy, as a label-free detection technology, has been widely used in tumor diagnosis. However, most tumor diagnosis procedures utilize multivariate statistical analysis methods for classification, which poses a major bottleneck toward achieving high accuracy. Here, we propose a concept called the two-dimensional (2D) Raman figure combined with convolutional neural network (CNN) to improve the accuracy. Two-dimensional Raman figures can be obtained from four transformation methods: spectral recurrence plot (SRP), spectral Gramian angular field (SGAF), spectral short-time Fourier transform (SSTFT), and spectral Markov transition field (SMTF). Two-dimensional CNN models all yield more than 95% accuracy, which is higher than the PCA-LDA method and the Raman-spectrum-CNN method, indicating that 2D Raman figure inputs combined with CNN may be one reason for gaining excellent performances. Among 2D-CNN models, the main difference is the conversion, where SRP is based on the structure of wavenumber series with the best performances (98.9% accuracy, 99.5% sensitivity, 98.3% specificity), followed by SGAF on the wavenumber series, SSTFT on wavenumber and intensity information, and SMTF on wavenumber position information. The inclusion of external information in the conversion may be another reason for improvement in the accuracy. The excellent capability shows huge potential for tumor diagnosis via 2D Raman figures and may be applied in other spectroscopy analytical fields.
Cervical cancer biomarker screening based on Raman spectroscopy and multivariate statistical analysis

Abstract:Cervical cancer (CC) stands as one of the most prevalent malignancies among females, and the examination of serum tumor markers(TMs) assumes paramount significance in both its diagnosis and treatment. This research delves into the potential of combining Surface-Enhanced Raman Spectroscopy (SERS) with Multivariate Statistical Analysis (MSA) to diagnose cervical cancer, coupled with the identification of prospective serum biomarkers. Serum samples were collected from 95 CC patients and 81 healthy subjects, with subsequent MSA employed to analyze the spectral data. The outcomes underscore the superior efficacy of Partial Least Squares Discriminant Analysis (PLS-DA) within the MSA framework, achieving predictive accuracy of 97.73 %, and exhibiting sensitivities and specificities of 100 % and 95.83 % respectively. Additionally, the PLS-DA model yields a Variable Importance in Projection (VIP) list, which, when coupled with the biochemical information of characteristic peaks, can be utilized for the screening of biomarkers. Here, the Random Forest (RF) model is introduced to aid in biomarker screening. The two findings demonstrate that the principal contributing features distinguishing cervical cancer Raman spectra from those of healthy individuals are located at 482, 623, 722, 956, 1093, and 1656 cm-1, primarily linked to serum components such as DNA, tyrosine, adenine, valine, D-mannose, and amide I. Predictive models are constructed for individual biomolecules, generating ROC curves. Remarkably, D-mannose of V (C-N) exhibited the highest performance, boasting an AUC value of 0.979. This suggests its potential as a serum biomarker for distinguishing cervical cancer from healthy subjects.
Multispectral Image under Tissue Classification Algorithm in Screening of Cervical Cancer.

Abstract:The objectives of this study were to improve the efficiency and accuracy of early clinical diagnosis of cervical cancer and to explore the application of tissue classification algorithm combined with multispectral imaging in screening of cervical cancer. 50 patients with suspected cervical cancer were selected. Firstly, the multispectral imaging technology was used to collect the multispectral images of the cervical tissues of 50 patients under the conventional white light waveband, the narrowband green light waveband, and the narrowband blue light waveband. Secondly, the collected multispectral images were fused, and then the tissue classification algorithm was used to segment the diseased area according to the difference between the cervical tissues without lesions and the cervical tissues with lesions. The difference in the contrast and other characteristics of the multiband spectrum fusion image would segment the diseased area, which was compared with the results of the disease examination. The average gradient, standard deviation (SD), and image entropy were adopted to evaluate the image quality, and the sensitivity and specificity were selected to evaluate the clinical application value of discussed method. The fused spectral image was compared with the image without lesions, it was found that there was a clear difference, and the fused multispectral image showed a contrast of 0.7549, which was also higher than that before fusion (0.4716), showing statistical difference (P < 0.05). The average gradient, SD, and image entropy of the multispectral image assisted by the tissue classification algorithm were 2.0765, 65.2579, and 4.974, respectively, showing statistical difference (P < 0.05). Compared with the three reported indicators, the values of the algorithm in this study were higher. The sensitivity and specificity of the multispectral image with the tissue classification algorithm were 85.3% and 70.8%, respectively, which were both greater than those of the image without the algorithm. It showed that the multispectral image assisted by tissue classification algorithm can effectively screen the cervical cancer and can quickly, efficiently, and safely segment the cervical tissue from the lesion area and the nonlesion area. The segmentation result was the same as that of the doctor's disease examination, indicating that it showed high clinical application value. This provided an effective reference for the clinical application of multispectral imaging technology assisted by tissue classification algorithm in the early screening and diagnosis of cervical cancer.

Abstract:In this study, a minimally invasive test method for cervical cancer in vitro was proposed by comparing Raman spectroscopy with support vector machine (SVM) model and deep belief network (DBN) model. The serum Raman spectra of cervical cancer, hysteromyoma, and healthy people were collected. After data processing, SVM classification model and DBN classification model were built respectively. The experimental results show that when the DBN network algorithm is used, the sample test set can be divided accurately and the result of cross-validation is ideal. Compared with the traditional SVM algorithm, this method firstly screened the effective feature matrix from the data, and then classified the data. With high efficiency and accuracy, based on 445 samples collected, this method improved the accuracy by 13.93%±2.47% compared with the SVM method, and provided a new direction and idea for the in vitro diagnosis of cervical diseases.

Raman spectroscopy combined with convolutional neural network for the sub-types classification of breast cancer and critical feature visualization

Abstract:Problems: Raman spectroscopy has emerged as an effective technique that can be used for noninvasive breast cancer analysis. However, the current Raman prediction models fail to cover all the molecular sub-types of breast cancer, and lack the visualization of the model. Aims: Using Raman spectroscopy combined with convolutional neural network (CNN) to construct a prediction model for the existing known molecular sub-types of breast cancer, and selected critical peaks through visualization strategies, so as to achieve the purpose of mining specific biomarker information. Methods: Optimizing network parameters with the help of sparrow search algorithm (SSA) for the multiple parameters in the CNN to improve the prediction performance of the model. To avoid the contingency of the results, multiple sets of data were generated through Monte Carlo sampling and used to train the model, thereby improving the credibility of the results. Based on the accurate prediction of the model, the spectral regions that contributed to the classification were visualized using Gradient-weighted Class Activation Mapping (Grad-CAM), achieving the goal of visualizing characteristic peaks. Results: Compared with other algorithms, optimized CNN could obtain the highest accuracy and lowest standard error. And there was no significant difference between using full spectra and fingerprint regions (within 2 %), indicating that the fingerprint region provided the most contribution in classifying sub-types. Based on the classification results from the fingerprint region, the model performances about various sub-types were as follows: CNN (95.34 %±2.18 %)>SVM(94.90 %±1.88 %)>PLS-DA(94.52 %±2.22 %)> KNN (80.00 %±5.27 %). The critical features visualized by Grad-CAM could match well with IHC information, allowing for a more distinct differentiation of sub-types in their spatial positions. Conclusion: Raman spectroscopy combined with CNN could achieve accurate and rapid identification of breast cancer molecular sub-types. Proposed visualization strategy could be proved from biochemistry information and spatial location, demonstrated that the strategy might be used for the mining of biomarkers in future.
Enhanced Cancer Classification and Critical Feature Visualization Using Raman Spectroscopy and Convolutional Neural Networks

Abstract:Cell misuse and cross-contamination pose a significant threat to the accuracy of cell research outcomes, often leading to the wasteful expenditure of time, manpower, and material resources. Consequently, the accurate identification of cell lines is paramount. However, traditional identification methods, which often involve staining and culturing procedures, are not only time-consuming but also laborious. This underscores the need for a novel approach that enables rapid and automated cell line identification, thereby enhancing research efficiency and accuracy. Raman spectroscopy, renowned for its label-free, rapid, and noninvasive nature, offers invaluable molecular insights into samples, making it a widely utilized technique in the biological field. In this study, the identification of one normal and five cancer cell lines was achieved using a sparrow search algorithm-convolutional neural networks (SSA-CNN), considering both the full spectra and fingerprint region perspectives. The SSA-CNN model demonstrated exceptional performance, not just in binary classification, but also in accurately distinguishing among six cell lines. It achieved the highest accuracy (around 95 %), and the lowest standard error (≤3%), for both the full spectra and fingerprint region. Based on the highly accurate SSA-CNN model, proposed the application of gradient-weighted class activation mapping (Grad-CAM) to visualize the Raman feature peaks. Upon comparing the visualized Raman features with reported biomarkers, found that not only were common biomolecules such as glucose, proteins, and liquids visualized, but specific feature peaks also aligned with reported biomarkers. The aforementioned results clearly demonstrated that the proposed strategy not only classifies cancer cell lines with remarkable accuracy but also served as a valuable tool for the discovery of novel biomarkers.

Abstract:The early detection of laryngeal cancer significantly increases the survival rates, permits more conservative larynx sparing treatments, and reduces healthcare costs. A non-invasive optical form of biopsy for laryngeal carcinoma can increase the early detection rate, allow for more accurate monitoring of its recurrence, and improve intraoperative margin control.In this study, we evaluated a Raman spectroscopy system for the rapid intraoperative detection of human laryngeal carcinoma. The spectral analysis methods included principal component analysis (PCA), random forest (RF), and one-dimensional (1D) convolutional neural network (CNN) methods.We measured the Raman spectra from 207 normal and 500 tumor sites collected from 10 human laryngeal cancer surgical specimens. Random Forest analysis yielded an overall accuracy of 90.5%, sensitivity of 88.2%, and specificity of 92.8% on average over 10 trials. The 1D CNN demonstrated the highest performance with an accuracy of 96.1%, sensitivity of 95.2%, and specificity of 96.9% on average over 50 trials. In predicting the first three principal components (PCs) of normal and tumor data, both RF and CNN demonstrated high performances, except for the tumor PC2.This is the first study in which CNN-assisted Raman spectroscopy was used to identify human laryngeal cancer tissue with extracted feature weights. The proposed Raman spectroscopy feature extraction approach has not been previously applied to human cancer diagnosis. Raman spectroscopy, as assisted by machine learning (ML) methods, has the potential to serve as an intraoperative, non-invasive tool for the rapid diagnosis of laryngeal cancer and margin detection.

Automatic model for cervical cancer screening based on convolutional neural network: a retrospective, multicohort, multicenter study

Abstract:Abstract Background The incidence rates of cervical cancer in developing countries have been steeply increasing while the medical resources for prevention, detection, and treatment are still quite limited. Computer-based deep learning methods can achieve high-accuracy fast cancer screening. Such methods can lead to early diagnosis, effective treatment, and hopefully successful prevention of cervical cancer. In this work, we seek to construct a robust deep convolutional neural network (DCNN) model that can assist pathologists in screening cervical cancer. Methods ThinPrep cytologic test (TCT) images diagnosed by pathologists from many collaborating hospitals in different regions were collected. The images were divided into a training dataset (13,775 images), validation dataset (2301 images), and test dataset (408,030 images from 290 scanned copies) for training and effect evaluation of a faster region convolutional neural network (Faster R-CNN) system. Results The sensitivity and specificity of the proposed cervical cancer screening system was 99.4 and 34.8%, respectively, with an area under the curve (AUC) of 0.67. The model could also distinguish between negative and positive cells. The sensitivity values of the atypical squamous cells of undetermined significance (ASCUS), the low-grade squamous intraepithelial lesion (LSIL), and the high-grade squamous intraepithelial lesions (HSIL) were 89.3, 71.5, and 73.9%, respectively. This system could quickly classify the images and generate a test report in about 3 minutes. Hence, the system can reduce the burden on the pathologists and saves them valuable time to analyze more complex cases. Conclusions In our study, a CNN-based TCT cervical-cancer screening model was established through a retrospective study of multicenter TCT images. This model shows improved speed and accuracy for cervical cancer screening, and helps overcome the shortage of medical resources required for cervical cancer screening.

Abstract:Among the most frequently diagnosed cancers in developing countries, esophageal squamous cell carcinoma (ESCC) ranks among the top six causes of death. It would be beneficial if a rapid, accurate, and automatic ESCC diagnostic method could be developed to reduce the workload of pathologists and improve the effectiveness of cancer treatments. Using micro-FTIR spectroscopy, this study classified the transformation stages of ESCC tissues. Based on 6,352 raw micro-FTIR spectra, a one-dimensional convolutional neural network (1D-CNN) model was constructed to classify-five stages. Based on the established model, more than 93% accuracy was achieved at each stage, and the accuracy of identifying proliferation, low grade neoplasia, and ESCC cancer groups was achieved 99% for the test dataset. In this proof-of-concept study, the developed method can be applied to other diseases in order to promote the use of FTIR spectroscopy in cancer pathology.
A novel attention-guided convolutional network for the detection of abnormal cervical cells in cervical cancer screening

Abstract:Early detection of abnormal cervical cells in cervical cancer screening increases the chances of timely treatment. But manual detection requires experienced pathologists and is time-consuming and error prone. Previously, some methods have been proposed for automated abnormal cervical cell detection, whose performance yet remained debatable. Here, we develop an attention feature pyramid network (AttFPN) for automatic abnormal cervical cell detection in cervical cytology images to assist pathologists to make a more accurate diagnosis. Our proposed method consists of two main components. First, an attention module mimicking the way pathologists reading a cervical cytology image. It learns what features to emphasize or suppress by refining extracted features effectively. Second, a multi-scale region-based feature fusion network guided by clinical knowledge to fuse the refined features for detecting abnormal cervical cells at different scales. The region proposals in the multi-scale network are designed according to the clinical knowledge about size and shape distribution of real abnormal cervical cells. Our method, trained and validated with 7030 annotated cervical cytology images, performs better than the state of art deep learning-based methods. The overall sensitivity, specificity, accuracy, and AUC of an independent testing dataset with 3970 cervical cytology images is 95.83%, 94.81%, 95.08% and 0.991, respectively, which is comparable to that of an experienced pathologist with 10 years of experience. Besides, we further validated our method on an external dataset with 110 cases and 35,013 images from a different organization, the case-level sensitivity, specificity, accuracy, and AUC is 91.30%, 90.62%, 90.91% and 0.934, respectively. Average diagnostic time of our method is 0.04s per image, which is much quicker than the average time of pathologists (14.83s per image). Thus, our AttFPN is effective and efficient in cervical cancer screening, and improvement of clinical workflows for the benefit of potential patients. Abstract:Background: Raman spectroscopy has been extensively utilized as a marker-free detection method in the complementary diagnosis of cancer. Multivariate statistical classification analysis is frequently employed for Raman spectral data classification. Nevertheless, traditional multivariate statistical classification analysis performs poorly when analyzing large samples and multicategory spectral data. In addition, with the advancement of computer vision, convolutional neural networks (CNNs) have demonstrated extraordinarily precise analysis of two-dimensional image processing. Result: Combining 2D Raman spectrograms with automatic weighted feature fusion network (AWFFN) for bladder cancer detection is presented in this paper. Initially, the s-transform (ST) is implemented for the first time to convert 1D Raman data into 2D spectrograms, achieving 99.2% detection accuracy. Second, four upscaling techniques, including short time fourier transform (STFT), recurrence map (RP), markov transform field (MTF), and grammy angle field (GAF), were used to transform the 1D Raman spectral data into a variety of 2D Raman spectrograms. In addition, a particle swarm optimization (PSO) algorithm is combined with VGG19, ResNet50, and ResNet101 to construct a weighted feature fusion network, and this parallel network is employed for evaluating multiple spectrograms. Class activation mapping (CAM) is additionally employed to illustrate and evaluate the process of feature extraction via the three parallel network branches. The results demonstrate that the combination of a 2D Raman spectrogram along with a CNN for the diagnosis of bladder cancer obtains a 99.2% accuracy rate，which indicates that it is an extremely promising auxiliary technology for cancer diagnosis. Significance: The proposed two-dimensional Raman spectroscopy method has an improved precision than one-dimensional spectroscopic data, which presents a potential methodology for assisted cancer detection and providing crucial technical support for assisted diagnosis.

Abstract:Cervical cancer is a prevalent disease affecting the cervix cells in women and is one of the leading causes of mortality for women globally. The Pap smear test determines the risk of cervical cancer by detecting abnormal cervix cells. Early detection and diagnosis of this cancer can effectively increase the patient's survival rate. The advent of artificial intelligence facilitates the development of automated computer-assisted cervical cancer diagnostic systems, which are widely used to enhance cancer screening. This study emphasizes the segmentation and classification of various cervical cancer cell types. An intuitive but effective segmentation technique is used to segment the nucleus and cytoplasm from histopathological cell images. Additionally, handcrafted features include different properties of the cells generated from the distinct cervical cytoplasm and nucleus area. Two feature rankings techniques are conducted to evaluate this study's significant feature set. Feature analysis identifies the critical pathological properties of cervical cells and then divides them into 30, 40, and 50 sets of diagnostic features. Furthermore, a graph dataset is constructed using the strongest correlated features, prioritizes the relationship between the features, and a robust graph convolution network (GCN) is introduced to efficiently predict the cervical cell types. The proposed model obtains a sublime accuracy of 99.11% for the 40-feature set of the SipakMed dataset. This study outperforms the existing study, performing both segmentation and classification simultaneously, conducting an in-depth feature analysis, attaining maximum accuracy efficiently, and ensuring the interpretability of the proposed model. To validate the model's outcome, we tested it on the Herlev dataset and highlighted its robustness by attaining an accuracy of 98.18%. The results of this proposed methodology demonstrate the dependability of this study effectively, detecting cervical cancer in its early stages and upholding the significance of the lives of women.

Abstract:Pancreatic cancer is the deadliest cancer type with a five-year survival rate of less than 9%. Detection of tumor margins plays an essential role in the success of surgical resection. However, histopathological assessment is time-consuming, expensive, and labor-intensive. We constructed a lab-designed, hand-held Raman spectroscopic system that could enable intraoperative tissue diagnosis using convolutional neural network (CNN) models to efficiently distinguish between cancerous and normal pancreatic tissue. To our best knowledge, this is the first reported effort to diagnose pancreatic cancer by CNN-aided spontaneous Raman scattering with a lab-developed system designed for intraoperative applications. Classification based on the original one-dimensional (1D) Raman, two-dimensional (2D) Raman images, and the first principal component (PC1) from the principal component analysis on the 2D image, could all achieve high performance: the testing sensitivity, specificity, and accuracy were over 95%, and the area under the curve approached 0.99. Although CNN models often show great success in classification, it has always been challenging to visualize the CNN features in these models, which has never been achieved in the Raman spectroscopy application in cancer diagnosis. By studying individual Raman regions and by extracting and visualizing CNN features from max-pooling layers, we identified critical Raman peaks that could aid in the classification of cancerous and noncancerous tissues. 2D Raman PC1 yielded more critical peaks for pancreatic cancer identification than that of 1D Raman, as the Raman intensity was amplified by 2D Raman PC1. To our best knowledge, the feature visualization was achieved for the first time in the field of CNN-aided spontaneous Raman spectroscopy for cancer diagnosis. Based on these CNN feature peaks and their frequency at specific wavenumbers, pancreatic cancerous tissue was found to contain more biochemical components related to the protein contents (particularly collagen), whereas normal pancreatic tissue was found to contain more lipids and nucleic acid (particularly deoxyribonucleic acid/ribonucleic acid). Overall, the CNN model in combination with Raman spectroscopy could serve as a useful tool for the extraction of key features that can help differentiate pancreatic cancer from a normal pancreas.
