Abstract:A novel series of 2-styryl-5-hydroxy-4-pyrone derivatives and analogues were designed and synthesized as H3 receptor antagonism based multitarget-directed ligands (MTDLs) for AD therapy using pharmacophore-combine strategy. The 2-styryl-5-hydroxy-4-pyrone pharmacophore with metal ion chelation, antioxidation, and Aβ aggregation inhibition activities was employed as the "eastern part", and a typical phenoxyalkylamine moiety was used as "central ring + western part" of the H3 receptor antagonist. The biological evaluation revealed that the majority of the target compounds demonstrated desirable multiple functions. The two most promising compounds 8a and 8b exhibited nanomolar IC50 values on H3 receptor antagonism, excellent metal ion chelating capability, more potent ABTS+ scavenging activity than Trolox, efficient Aβ self-aggregation and Cu2+-induced aggregation inhibitory activities, as well as disaggregation activities against Aβ self/Cu2+-induced aggregation.
Rational Design of Nitrogen-Doped Carbon Dots for Inhibiting β-Amyloid Aggregation

Abstract:The fibrillization and abnormal aggregation of β-amyloid (Aβ) peptides are commonly recognized risk factors for Alzheimer's disease (AD) brain, and require an effective strategy to inhibit the Aβ deposition and treat AD. Herein, we designed and synthesized nitrogen-doped carbon dots (N-CDs) as an Aβ-targeted probe, which exhibits the capacity of inhibiting the 1–42 Aβ (Aβ1–42) self-assembly in vitro. The N-CDs exhibited orange emission with an emission wavelength of 570 nm, which demonstrates their excellent optical properties with excitation-independent behavior. Meanwhile, the N-CDs have spherical morphologies with an average size of 2.2 nm, whose surface enriches the amino, carboxyl, and hydroxyl groups. These preparties are conducive to improving their biological water solubility and provide a large number of chemical bonds for further interaction with proteins. Contrary to this, the kinetic process, size evolutions, and morphologies changes of Aβ1–42 were inhibited in the presence of N-CDs in the determination of a thioflavin T assay, dynamic light scattering, transmission electron microscope, etc. Finally, the safety application of N-CDs on Aβ1–42-induced cytotoxicity was further demonstrated via in vitro cytotoxicity experiments. This work demonstrates the effective outcome of suppressing Aβ aggregation, which provides a new view into the high-efficiency and low-cytotoxicity strategy in AD theranostics.

Abstract:The abnormal self-assembly of cerebral β-amyloid (Aβ) peptides into toxic aggregates is a hallmark of Alzheimer's disease (AD). Here, we report on multifunctional carbon dots that can chelate Cu(ii) ions, suppress Aβ aggregation, and photooxygenate Aβ peptides. Copper ions have high relevance to AD pathogenesis, causing Cu(ii)-mediated Aβ aggregation and oxidative damage to neuronal cells. For effective conjugation with Cu(ii)-bound Aβ complexes, we have designed carbon dots that possess nitrogen (N)-containing polyaromatic functionalities on their surface by employing o-phenylenediamine (OPD) as a polymerization precursor. We demonstrate that the polymerized OPD (pOPD)-derived carbon dots exhibit multiple capabilities against Cu(ii)-mediated Aβ aggregation. Furthermore, the pOPD-derived carbon dots exhibited dramatically enhanced absorption and fluorescence upon coordination with Cu(ii) ions and effectively photooxygenated Aβ peptides. The photodynamically modulated Aβ residues lost the propensity to coordinate with Cu(ii) and to assemble into toxic aggregates. This work demonstrates the potential of carbon dots as a multifunctional β-sheet breaker and provides a promising anti-amyloidogenic strategy for future Aβ-targeted AD treatments.

Abstract:A series of novel 1-phenyl-3-hydroxy-4-pyridinone derivatives were designed and synthesized as multifunctional agents for AD therapy through incorporation of 3-hydroxy-4-pyridinone moiety from deferiprone into the scaffold of H3 receptor antagonists. Most of these new compounds displayed designed quadruple functions H3 receptor antagonism, Aβ aggregation inhibition, metal ion chelation and radical scavenging. Especially, the most promising compound 5c displayed nanomolar IC50 values in H3 receptor antagonism with high selectivity, efficient capability to interrupt the formation of Aβ1-42 fibrils, good copper and iron chelating properties, and more potent ABTS•+ scavenging activity than Trolox. The further biological evaluation revealed that it did not show obvious cytotoxicity and hERG potassium channel inhibition in micromolar concentration. In addition, compound 5c demonstrated suitable pharmacokinetic properties and acceptable blood-brain barrier (BBB) permeability in vivo. All these results indicate that compound 5c is a potential multifunctional candidate for AD therapy.
Modulating surface functional groups in carbon dots for enhanced inhibition of β‐amyloid aggregation

Abstract:Compared to CDs with more carboxyl functional groups, CDs possessing more amino functional groups effectively induced alterations in the initial conformation of Aβ1–42, leading to a significant inhibition of Aβ1–42 aggregation. Gaining a profound understanding of carbon dots (CDs) structure and their potential to inhibit β‐amyloid (Aβ) aggregation holds significant importance in the quest for Alzheimer's disease (AD) prevention and treatment. Herein, CDs with varying structures were synthesized by adjusting the molar ratio of citric acid (CA) and ethylenediamine (EDA). Transmission electron microscopy (TEM) was used to characterize their size and morphology. Fourier transform infrared spectroscopy (FTIR) and X‐ray photoelectron spectroscopy (XPS) were applied to study their structure. Optical properties were analyzed through Ultraviolet (UV) spectroscopy absorption and fluorescence spectroscopy. The inhibitory effect of CDs on Aβ1–42 aggregation was explored using circular dichroism and atomic force microscopy (AFM). The results highlighted the substantial influence of the CA to EDA molar ratio on the surface structure of CDs, subsequently impacting their optical properties and efficacy in inhibiting Aβ1–42 aggregation. With increasing EDA content, CDs exhibited a rise in the number of amino functional groups while experiencing a reduction in carboxyl groups. CDs possessing more amino functional groups effectively induced alterations in the initial conformation of Aβ1–42, leading to a significant inhibition of Aβ1–42 aggregation.

Dual-targeted carbon-dot-drugs nanoassemblies for modulating Alzheimer's related amyloid- β aggregation and inhibiting fungal infection

Abstract:Amyloid aggregation and fungal infection, especially amyloid beta (Aβ) peptide and Candida albicans are considered as two of the crucial pathogenic agents in Alzheimer's disease (AD). In this work, we propose an innovative treatment strategy for AD, targeting at not only Aβ aggregation but also Candida albicans infection. Here, a high-performance nanomaterial, namely gCDs-E, have been prepared by self-assembled of glycosylated carbon dots (gCDs) and epigallocatechin-3-gallate (EGCG). Surprisingly, gCDs-E can not only suppress the fibrillation of Aβ and disaggregate Aβ fibrils, but also effectively inhibit the activity of Candida albicans. More importantly, the prepared gCDs-E can effectively cut down the cytotoxicity of amyloid aggregations, and the cell viability reached to 99.2%. In addition, the capability of the gCDs-E for blood brain barrier (BBB) penetration was also observed using a normal mice model. Above all, the gCDs-E greatly cleaned Aβ deposition and improved memory impairment in APP/PS1 transgenic AD model mice, confirming its potential as therapeutic agent for AD treatment.

Abstract:Diagnostics of Alzheimer's disease (AD) commonly employ the use of fluorescent thioflavin derivatives having affinity for the amyloid-β (Aβ) proteins associated with AD progression. However, thioflavin probes have limitations in their diagnostic capabilities arising from a number of undesireable qualities, including poor photostability, weak emission intensity, and high emission overlap with the backgound tissue autofluorescence. To overcome such limitations, we have developed nanoformulated probes consisting of a red-emitting fluorescent quantum dot (QD) core encapsulated in a PEGylated shell with benzotriazole (BTA) targeting molecules on the surface (QD-PEG-BTA). The combination of strong red fluorescence, multivalent binding, and decreased backgound signal and nonspecific binding provided the ability of the QD-PEG-BTA probes to achieve detection sensitivites 4 orders of magnitude greater than those of conventional thioflavin derivatives. This study opens the door for the use of QDs in AD detection applications.
A multi-target theranostic nano-composite against Alzheimer's disease fabricated by conjugating carbon dots and triple-functionalized human serum albumin

Abstract:The complex pathogenesis of Alzheimer's disease (AD) involves the aggregation and accumulation of amyloid β-protein (Aβ) as well as elevated levels of reactive oxygen species (ROS), which requires the development of comprehensive diagnostic and therapeutic interventions. In this work, a multifunctional theranostic nano-composite (HSA-BFP@CDs) is constructed by conjugating triple-functionalized human serum albumin (HSA-BFP) as a theranostic agent targeting Aβ and carbon dots (CDs) as an ROS scavenger. HSA-BFP@CDs exhibits a fluorescence “off-on” effect at 700 nm upon interaction with Aβ aggregates, showing the capability for detection of Aβ plaques and potential for early diagnosis of AD. Besides, HSA-BFP@CDs effectively inhibits the aggregation of Aβ, increasing the viability of Aβ-treated cells from 74% to over 95% at 100 μg/mL. Moreover, multiple ROS, including hydroxyl radicals, superoxide radicals, hydrogen peroxide, and Aβ-Cu2+-induced-ROS, can be scavenged by HSA-BFP@CDs, thus resulting in the mitigation of cellular oxidative damages. Experiments with the AD model of Caenorhabditis elegans further demonstrate the multifunctionality of HSA-BFP@CDs in imaging amyloid plaques, reducing Aβ deposition, and relieving oxidative stress in vivo, showing the prospect for Aβ- and ROS-targeted AD diagnosis and treatment. This work provided new insight into the design of protein-carbon dots conjugate and the development of multi-target therapy of AD.Statement of significanceAlzheimer's disease (AD) is the most common form of dementia, which currently affects over 55 million people worldwide. Abstract:Metal dyshomeostasis is one of the pathological features of Alzheimer's disease (AD). Cu2+ is believed to induce the formation of neurotoxic β-amyloid (Aβ) aggregates and aggravate oxidative damage. It is challenging to take effective therapeutic interventions towards the complicated pathogenesis related to Cu2+. Herein, chiral carbon dots were proposed for Cu2+-related AD therapy. The chiral CDs were prepared based on the ternary carbon sources, using L/d-cysteine as the chiral carbon source, citric acid as the carbonization source and β-cyclodextrin as the chirality inducer, reserving and enhancing the chirality of CDs. Compared to L-Cys-CDs, D-Cys-CDs exhibited enhanced chirality, stronger affinity towards Cu2+ and comparable antioxidant ability, but better inhibition on peroxidase-like activity of Cu2+ or Aβ42-Cu2+. Thus, D-Cys-CDs performed better to inhibit Cu2+-induced Aβ aggregation. Meanwhile, D-Cys-CDs can alleviate the cytotoxicity of Aβ42-Cu2+ with satisfactory anti-inflammation effect and scavenging ability towards intracellular reactive oxygen species. D-Cys-CDs succeeded in reducing Aβ deposits and releasing paralysis for the AD model CL2120 nematodes. D-Cys-CDs with an enantiomeric effect on Cu2+-induced Aβ aggregation and the peroxidase-like activity of Cu2+ was promising as the multifunctional therapeutic agent for Cu2+-induced AD therapy.

Abstract:Mitochondrial dysfunction and metal ion imbalance are recognized as pathological hallmarks of Alzheimer's Disease (AD), leading to deposition of β-amyloid (Aβ) thereby and inducing neurotoxicity, activating apoptosis, eliciting oxidative stress, and ultimately leading to cognitive impairment. In this study, the red blood cell membrane (RBC) was used as a vehicle for encapsulating carbon quantum dots (CQD) and polydopamine (PDA), creating a nanocomposite (PDA-CQD/RBC). This nanocomposite was combined with near-infrared light (NIR) for AD treatment. The RBC offers anti-immunorecognition properties to evade immune clearance, PDA exhibits enzyme-mimicking activity to mitigate oxidative stress damage, and CQD acts as a chelating agent for metal ions (Cu 2+ ), effectively preventing Cu 2+ -mediated aggregation of Aβ. Furthermore, the local heating induced by near-infrared laser irradiation can dismantle the formed Aβ fibers and enhance the blood–brain barrier's permeability. Both in vitro and animal experiments have shown that PDA-CQD/RBC, in combination with NIR, mitigates neuroinflammation, and ameliorates behavioral deficits in mice. This approach targets multiple pathological pathways, surpassing the limitations of single-target treatments and enhancing therapeutic efficacy while decelerating disease progression.

Abstract:Multi-target intervention and synergistic treatment are critical for the drug development of Alzheimer’s disease (AD) due to its complex and multifactional nature. Oxidative stress and amyloid β peptides (Aβ) accumulation have been recognized as therapeutic targets for AD. Herein, with ability to inhibit Aβ aggregation and the broad-spectrum antioxidant properties, the large amino acid mimicking selenium-doped carbon quantum dots (SeCQDs) are presented as novel nanoagents for multi-target therapy of AD. Compared with the precursor, selenocystine, SeCQDs which maintain the intrinsic properties of both selenium and carbon quantum dots (CQDs) possess good biocompatibility and a remarkable ROS-scavenging activity. Moreover, the functionalized α-carboxyl and amino groups on edge of SeCQDs can trigger multivalent interactions with Aβ, leading to the ability of SeCQDs to inhibit Aβ aggregation. In vivo study demonstrated that SeCQDs can significantly ameliorate the Aβ induced memory deficits, reduce Aβ accumulation and inhibit neuron degeneration in AD model rats. The versatility of functionalization and potential ability to cross the blood-brain barrier (BBB) make SeCQDs as prospective nanodrugs for treating AD.

Abstract:The pathological aggregation of some proteins is claimed to be highly related to several human diseases, such as β-amyloid 1-42 (Aβ<sub>42</sub> ) to Alzheimer's disease (AD), islet amyloid polypeptide, and insulin to type 2 diabetes mellitus. Therefore, it is in desperate need to develop effective methods for detection of protein aggregates and inhibition of abnormal aggregation. Herein, to construct all-in-one probe with both diagnosis and treatment potentials for protein aggregation diseases, Congo red (CR), a classical staining reagent with red fluorescence signal output for protein aggregates, is deliberately adopted to react with three different reductive carbon sources and ammonium persulfate to generate three CR-derived carbon dots (CDs). The obtained CDs exhibit the capabilities of turn-on red fluorescence imaging of protein aggregates, and/or inhibition of protein aggregation as well as scavenging of free radicals. Among them, CA-CDs, using citric acid as the reductive carbon source, demonstrate the superiority to the other two studied CDs in integrating all of these functions, and particularly exert excellent cytoprotection effect against toxic Aβ<sub>42</sub> species, possessing tremendous potential in diagnosis and treatment of AD for future study. The present study paves a new way to develop all-in-one CDs for the protein disease research.

Abstract:The abnormal aggregation of β-amyloid protein (Aβ) is a major contributor to Alzheimer's disease (AD). Cu 2+ homeostasis imbalance can lead to the aggregation of Aβ, resulting in cytotoxic oligomers and fibrous aggregates, causing neuroinflammation and nerve cell damage, ultimately leading to AD. In this study, we synthesized nitrogen-doped carbon quantum dot (CQD), and designed a macrophage membrane (RAW-M) encapsulated CQD nanosystem for the first time. The abundant nitrogen-containing groups on the surface of CQD effectively capture excess Cu 2+ and inhibit rapid Aβ aggregation. Additionally, the good photothermal properties of CQD dissolve the formed fiber precipitates under near-infrared light (NIR). In vitro and in vivo studies showed that the nanosystem significantly improved BBB permeability under laser irradiation, enhancing its ability to cross the BBB and overcome traditional anti-AD drug limitations. In vivo investigations conducted on APP/PS1 mice indicate that the nanosystem strongly reduced Aβ deposition, mitigated neuroinflammation, and ameliorates deficits in learning and memory. Overall, our nanocarrier approach adjusts metal ion homeostasis, inhibits abnormal Aβ aggregation, and uses excellent photothermal properties to depolymerize mature Aβ fibrils to protect cells from Aβ neurotoxicity, providing an effective strategy for Aβ-targeted treatment of AD.

C3N Nanodots Inhibits Aβ Peptides Aggregation Pathogenic Path in Alzheimer's Disease.

Abstract:Despite the accumulating evidence linking the development of Alzheimer's disease (AD) to the aggregation of Aβ peptides and the emergence of Aβ oligomers, the FDA has approved very few anti-aggregation-based therapies over the past several decades. Here, we report the discovery of an Aβ peptide aggregation inhibitor: an ultra-small nanodot called C3N. C3N nanodots alleviate aggregation-induced neuron cytotoxicity, rescue neuronal death, and prevent neurite damage in vitro. Importantly, they reduce the global cerebral Aβ peptides levels, particularly in fibrillar amyloid plaques, and restore synaptic loss in AD mice. Consequently, these C3N nanodots significantly ameliorate behavioral deficits of APP/PS1 double transgenic male AD mice. Moreover, analysis of critical tissues (e.g., heart, liver, spleen, lung, and kidney) display no obvious pathological damage, suggesting C3N nanodots are biologically safe. Finally, molecular dynamics simulations also reveal the inhibitory mechanisms of C3N nanodots in Aβ peptides aggregation and its potential application against AD.

Abstract:Alzheimer's disease is a progressive neurodegenerative disorder with an insidious onset of symptoms. At present, there are no treatments that can effectively stop or reverse the disease from progressing. The development of effective methods for early diagnosis and treatment of Alzheimer's disease is therefore still urgently needed. In this article, we designed a family of Q-series probes ( Q16~Q21 ) that bind to Aβ aggregates by introducing different electron donors to bind to benzothiazole salts electron acceptors with different substituents to achieve imaging and photooxidation of aggregated Aβ proteins. The structure of the six probes was composed of N, N-dimethylaminophenyl and triphenylamine (donor groups), benzothiazole salts (acceptor groups), and π-conjugated double bonds. Among them, the probe Q17 with triphenylamine as the electron donor had a good singlet oxygen production efficiency, which can effectively carry out photooxidation after combining with Aβ aggregates, effectively reducing the degree of aggregation, and the cytotoxicity of Aβ aggregates was significantly reduced after bright light oxidation through cell experiments, which has great potentials in the treatment of Aβ photooxidation. On the other hand, probes Q16 and Q18 with N, N-dimethylaminophenyl as donors had excellent imaging abilities, good affinities for Aβ (Kd -Q16 = 26.95 nM, Kd -Q18 = 21.56 nM), can effectively image Aβ plaques in AD mouse brain slices and monitor the change of cell viscosity, and have a good application prospect in fluorescence imaging.

Abstract:Abnormal accumulation amyloid-β (Aβ) is recognized as one of the crucial factors for the progression and pathology of AD, which has accelerated the development of new chemical agents or technology for early mapping and monitoring Aβ plagues. While Aβ plaques is from diffuse to highly compacted (dense-core), most of reported dyes or probes could only stain the core of Aβ, due to the lack of monitoring technology, the research on periphery of Aβ was always ignored. Herein, we reported a new near-infrared (NIR) fluorescent probe named Dicyano-3, which had the capability to stain both core and periphery of Aβ plaques. Dicyano-3 was based on dicayno-scaffold and input N,N’-dimethyl group connecting with double bond. Meanwhile, cyclopropyl group applied in drug discovery was introduced to the structure which could lead to good photophysical properties. Dicyano-3 displayed good binding affinity for Aβ aggregates and better histological staining capability compared to Thioflavin T. Furthermore, Dicyano-3 showed good blood–brain barrier (BBB) permeability and low cytotoxicity. More importantly, in vivo imaging results indicate that the fluorescent intensity in the brain in 10-month APP/PS1 mice is higher than age-matched wild type mice. We believe that Dicyano-3 is benefit for the research of Aβ mechanism and AD drug discovery.

Curcumin-derived carbon quantum dots: Dual actions in mitigating tau hyperphosphorylation and amyloid beta aggregation

Abstract:The amyloid cascade and tau hypotheses both hold significant implications for the pathogenesis of Alzheimer's disease (AD). Curcumin shows potential by inhibiting the aggregation of amyloid beta (Aβ) and reducing tau hyperphosphorylation, however, its use is limited due to issues with solubility and bioavailability. Carbon dots, recognized for their high biocompatibility and optimal water solubility, have demonstrated the capability to inhibit either Aβ or tau aggregation. Nonetheless, their effects on tau hyperphosphorylation are yet to be extensively explored. This study aims to evaluate the water-soluble curcumin-derived carbon quantum dots (Cur-CQDs) synthesized via an eco-friendly method, designed to preserve the beneficial effects of curcumin while overcoming solubility challenges. The synthesis of Cur-CQDs involves a single-step dry heating process using curcumin, resulting in dots that exhibit negligible cytotoxicity to SH-SY5Y cells at the examined concentrations. Notably, Cur-CQDs have shown the ability to simultaneously mitigate Aβ aggregation and tau hyperphosphorylation. Therefore, it is suggested that Cur-CQDs may hold potential for AD treatment, a hypothesis deserving of further research.

Abstract:Photo-oxygenation has emerged as an effective modality for scavenging Alzheimer's amyloid-β (Aβ) plaques. However, limitations of the current photo-oxidants, such as low Aβ-targeting and single functionality, hinder the scavenging of Aβ plaques via photo-oxygenation. Herein, based on an aggregation-induced emission (AIE)-active fluorogen (named TPMD), we designed AIE photo-oxidant nanoparticles (T-LD NPs) for Aβ imaging, inhibition, and disaggregation. The T-LD NPs were prepared by the assembly of hydrophobic TPMD with Aβ-targeting peptide (LPPFD, L) conjugated amphiphilic polymer (DSPE-PEG). Such T-LD NPs could specifically label Aβ plaques for image-guided therapy. Under laser irradiation, T-LD NPs generated a plethora of reactive oxygen species (ROS), including 1O2, •OH, and O2•-, to oxygenate Aβ species, leading to the potent inhibition of Aβ fibrillization, and significant alleviation of Aβ-mediated neurotoxicity (36% to 10% at 20 μg mL-1). Notably, T-LD NPs could rapidly disaggregate mature Aβ fibrils into fractured β-sheet rich aggregates via photo-oxygenation, resulting in alleviated cytotoxicity. In vivo studies revealed that the photo-activated T-LD NPs scavenged amyloid plaques in the transgenic C. elegans strain CL2006 and extended the lifespan by 4 days. Taken together, this multifunctional T-LD NPs integrated Aβ-targeting, near-infrared fluorescence imaging, and photo-oxygenation, providing a new strategy for the development of multifunctional AIE photo-oxidants for the treatment of Alzheimer's disease.

Abstract:Alzheimer's Disease (AD) is a neurodegenerative disease, of which β-amyloid (Aβ) deposition is one of the most important pathological features. It has been reported that during Aβ aggregation, the microenvironment around the Aβ protein is altered in terms of viscosity and polarity. In this work, we developed five novel hemicyanine fluorescent probes (MZs). After screening the photochemical properties, MZ-2 and 3 were found to enable the rapid detection of Aβ42 aggregates, which were also sensitive to ambient viscosity. After comparison the structure of probes, we also observed that extensions of conjugated π-systems effectively cause redshifts of excitation wavelength. In the meanwhile, hydroxyl groups with weaker ionization strengths are more responsive to Aβ42 aggregates than sulfonate groups, probably due to the small size of the hydroxyl group and the acidity. Overall, MZ-2 showed the best response to Aβ42 aggregates (15.35-fold) and viscosity (17.6-fold). MZ-2 can quickly cross the blood-brain barrier (BBB), enabling high-fidelity imaging of Aβ42 aggregates in the mice brain.
