Abstract:A palladium-catalyzed one-pot two-step radical mediated carbonylative cyclization of 1,7-enynes with perfluoroalkyl iodides and Mo(CO)(6) has been developed for the construction of polycyclic 3,4-dihydroquinolin-2(1H)-one scaffolds. This method realizes a facile synthesis of various polycyclic 3,4-dihydroquinolin-2(1H)-one derivatives containing perfluoroalkyl and carbonyl units in high yields. Moreover, modifications of several bioactive molecules were demonstrated using this protocol.

Abstract:An efficient synthesis of novel fused heterocycles has been established via microwave-assisted palladium-catalyzed intramolecular direct arylation. The acyclic aryl bromides are readily available from microwave-assisted one-pot annulation of N-(2-bromobenzyl)-2-aminophenois and ethyl 2-bromoalkanoates. The intramolecular direct arylation is then performed in the presence of palladium(II) acetate and dppf (10 mol% each) in toluene using potassium carbonate (2 equiv) as the base under microwave heating (150 degrees C, 1 h) to afford the products in 43-99% yields. Steric effect is observed for Ar-Ar bond formation, giving a substantial amount of the debromination byproduct.

Abstract:A direct, efficient, and practical protocol to install a chiral oxazoline unit onto aryl/heteroaryl rings via palladium-catalyzed C-H functionalization of 2-positions of oxazolines with a variety of halides using dppe as the ligand has been developed. Various chiral oxazoline ligands could be synthesized, even in a 10-g scale process. This protocol is a good supplement to traditional methods and for diverse synthesis of chiral oxazoline ligands.

Abstract:Aromatization-driven deconstruction and functionalization of spiro dihydroquinazolinones via dual photoredox/nickel catalysis is developed. The aromatization effect was introduced to synergistically drive unstrained cyclic C-C bond cleavage, with the aim of overcoming the ring-size limitation of nitrogen-centered radical induced deconstruction of carbocycles. Herein, we demonstrate the synergistic photoredox/nickel catalyzed deconstructive cross-coupling of spiro dihydroquinazolinones with organic halides. Remarkably, structurally diverse organic halides including aryl, alkenyl, alkynyl, and alkyl bromides were compatible for the coupling. In addition, this protocol is also characterized by its mild and redox-neutral conditions, excellent functional group compatibility, high atom economy, and easy scalability. A telescoped procedure involving condensation and ring-opening/coupling was found to be accessible. This work provides a complementary strategy to the existing radical-mediated C-C bond cleavage of unstrained carbocycles.

Abstract:A convenient method for the synthesis of aryl-functionalized spirocyclohexadienone scaffolds from alkyne-containing phenol-based biaryls with aryl halides via palladium-catalyzed cyclization/dearomatization/arylation is developed. The approach provides a series of spirocyclohexadienone molecules in moderate to high yields. This tandem process involves the formation of three new chemical bonds, providing access to novel quinoline-substituted hydroxyl isoindolones skeleton under simple reaction conditions. The deuterium-labeled competition reaction has revealed that C-H bond cleavage is the turnover limiting step.

The transformation involves cascade syn-carbopalladation, C−H activation, and a decarboxylation sequence, enabling simultaneously formation of three C-C bonds. Furthermore, 8-bromo-1-naphthoic acid can act as a viable substrate to forge a seven-membered ring fused isoquinolinone.

Abstract:Summary of main observation and conclusionAn efficient strategy for the formation of alkenyl‐functionalized spirocarbocyclic scaffolds from alkyne‐containing phenol‐based biaryls via sequential iodine‐induced cyclization/dearomatization and Pd‐catalyzed coupling of N‐tosylhydrazones is developed. The approach provides various spirocarbocyclic compounds in moderate to excellent yields with good functional tolerance. The results also demonstrate the feasibility for the direct cross‐couplings of N‐tosylhydrazones with sterically congested tetrasubstituted alkenyl halides.

Abstract:A novel method for the construction of a cyclopenta[c]quinoline ring via cyclization of 3-bromoindoles with internal alkynes in the presence of palladium is described. The formation of the cyclopenta[c]quinoline ring is proposed from a double [1,5] carbon sigmatropic rearrangement of the spirocyclic cyclopentadiene intermediate, which is generated in situ from the cyclization of 3-bromoindoles with internal alkynes involving a sequential double alkyne insertion into the carbon-palladium bond and dearomatization of indole. The present studies have developed a novel ring-expansion reaction of the pyrrole ring to pyridine via one carbon insertion into the C2-C3 bond of indoles and provided a simple and distinct route for the construction of tricyclic fused-quinoline derivatives that are not easy to access with conventional methods.

Abstract:Aryl-aryl bond formation constitutes one of the most important subjects in organic synthesis. The recent developments in direct arylation reactions forming aryl-aryl bond have emerged as very attractive alternatives to traditional cross-coupling reactions. Here, we describe a general palladium-catalyzed direct arylation and cyclization of o-iodobiaryls to build a library of tetraphenylenes. This transformation represents one of the very few examples of C-H activation process that involves simultaneous formation of two aryl-aryl bonds. Oxygen plays a vital role by ensuring high reactivity, with air as the promoter furnished the best results. We anticipate this ligand-free and aerobic catalytic system will simplify the synthesis of tetraphenylenes as many of the reported methods involve use of preformed organometallic reagents and will lead to the discovery of highly efficient new direct arylation process.

Abstract:A novel asymmetric Pd-catalyzed decarboxylative [3 + 2] cycloaddition of vinylethylene carbonates and alpha,beta-unsaturated pyrazolones has been developed to afford highly functionalized tetrahydrofuran-fused spiro-heterocyclic skeletons with excellent stereoselectivity. A diverse range of enantioenriched spiro-tetrahydrofuran-pyrazolones, possessing three contiguous stereogenic centers including one all-carbon quaternary, were obtained in high yields and excellent enantioselectivities (up to 90% yield and 98% ee). In addition, the practical application of this protocol was also demonstrated by gram-scale synthesis and further transformations into various compounds. Preliminary mechanistic experiments indicated that the allylic 1, 3-strain was crucial to the stereoselective control. We developed an asymmetric Pd-catalyzed [3 + 2] cycloaddition of alpha,beta-unsaturated pyrazolones with vinylethylene carbonates. An array of spiro-tetrahydrofuran-pyrazolones were obtained in high yields with excellent enantioselectivities.

Abstract:C-aryl glycosides are widely found in nature and play important roles in drug design. Despite the significant progress made over the past few decades, efficient and stereoselective synthesis of complex C-aryl glycosides remains challenging, lagging far behind the state of the art of the synthesis of O- or N-glycosides. Here, we report a simple and powerful bioinspired strategy for the stereoselective synthesis of C-aryl glycosides via palladium-catalysed ortho-directed C(sp2)−H functionalization of arenes and heteroarenes with easily accessible glycosyl chloride donors. The catalytic palladacycle intermediate generated via C−H palladation provides a soft aryl nucleophile that can react with glycosyl oxocarbenium ion partners with high efficiency and excellent stereocontrol. The method can be applied to a wide range of arene and heteroarene substrates, glycosyl chloride donors and auxiliary groups. It can simplify the synthesis of a variety of complex C-aryl glycosides and offers a tool for late-stage modification of drug molecules.

Abstract:A highly diastereo- and enantioselective phosphine-catalyzed sequential [3 + 2]/[3 + 2] annulation of allenoates with arylidenemalononitriles has been developed. This reaction allows for the facile construction of multifunctionalized <i>cis</i>-fused bicyclic[3,3,0]octene scaffolds, encompassing three consecutive stereogenic centers with one quaternary carbon center, in a one-step operation from readily available materials. The reported protocol is scalable, operates under mild reaction conditions, and creates the core structural motif of a number of natural products.
