Abstract:Sharp eyespot is a crucial disease affecting cereal plants, such as bread wheat (Triticum aestivum) and barley (Hordeum vulgare), and is primarily caused by the pathogenic fungus Rhizoctonia cerealis. As disease severity has increased, it has become imperative to find an effective and reasonable control strategy. One such strategy is the use of the trehalose analog, validamycin, which has been shown to have a potent inhibitory effect on several trehalases found in both insects and fungi, and is widely used as a fungicide in agriculture. In this study, we demonstrated that 0.5 μg/mL validamycin on PDA plates had an inhibitory effect on R. cerealis strain R0301, but had no significant impact on Fusarium graminearum strain PH-1. Except for its inhibiting the trehalase activity of pathogenic fungi, little is known about its mechanism of action. Six trehalase genes were identified in the genome of R. cerealis, including one neutral trehalase and five acidic trehalase genes. Enzyme activity assays indicated that treatment with 5 μg/mL validamycin significantly reduces trehalase activity, providing evidence that validamycin treatment does indeed affect trehalase, even though the expression levels of most trehalase genes, except Rc17406, were not obviously affected. Transcriptome analysis revealed that treatment with validamycin downregulated genes involved in metabolic processes, ribosome biogenesis, and pathogenicity in the R. cerealis. KEGG pathway analysis further showed that validamycin affected genes related to the MAPK signaling pathway, with a significant decrease in ribosome synthesis and assembly. In conclusion, our results indicated that validamycin not only inhibits trehalose activity, but also affects the ribosome synthesis and MAPK pathways of R. cerealis, leading to the suppression of fungal growth and pesticidal effects. This study provides novel insights into the mechanism of action of validamycin.
Isothiazolinones As Novel Candidate Insecticides for the Control of Hemipteran Insects.

Abstract:Hemipteran insects, such as whiteflies, aphids and planthoppers, resemble one of the most important pest groups threating food security. While many insecticides have been used to control these pests, many issues such as insecticide resistance have been found, highlighting the urgent need to develop novel insecticides. Here, we first observed that a commercial tetramycin solution was highly effective in killing whitefly. The major bioactive constituents were identified to be isothiazolinones, a group of biocides. We then tested the toxicity of several isothiazolinones to five hemipteran insects. The results show that Kathon, a widely used biocide against microorganisms, and its two constituents, chloromethylisothiazolinone (CMIT) and methylisothiazolinone (MIT), can cause considerable levels of mortality to whiteflies and aphids when applied at concentrations close to, or lower than, the upper limit of these chemicals permitted in cosmetic products. The results also indicate that two other isothiazolinones, benzisothiazolinone (BIT) and octylisothiazolinone (OIT) can cause considerable levels of mortality to whitefly and aphids but are less toxic than Kathon. Further, we show that Kathon marginally affects whitefly endosymbionts, suggesting its insecticidal activity is independent of its biocidal activity. These results suggest that some isothiazolinones are promising candidates for the development of a new class of insecticides for the control of hemipteran pests.
The impact of three thioxothiazolidin compounds on trehalase activity and development of Spodoptera frugiperda larvae

Abstract:Trehalases (TREs), serving as crucial enzymes regulating trehalose and chitin metabolism in insects, represent prime targets for pest control strategies. We investigated the impact of three thioxothiazolidin compounds (1G, 2G, and 11G) on TRE activity and summarized their effects on the growth and development of Spodoptera frugiperda (Lepidoptera, Noctuidae). The experimental larvae of S. frugiperda were injected with the three thioxothiazolidin compounds (1G, 2G, and 11G), while the control group received an equivalent volume of 2% DMSO as a control. All three compounds had a strong effect on inhibiting TRE activity, significantly prolonging the pre-pupal development stage. However, compared with the 11G-treated group, the survival rate of larvae treated with 1G and 2G was significantly reduced by 31.11% and 27.78% respectively, while the occurrence of phenotypic abnormalities related to growth and development was higher. These results manifest that only the TRE inhibitors, 1G and 2G, modulate trehalose and chitin metabolism pathways of larvae, ultimately resulting in the failure molting and reduction of survival rates. Consequently, the thioxothiazolidin compounds, 1G and 2G, hold potential as environmentally friendly insecticides.

Abstract:Novel approaches for pest control are essential to ensure a sufficient food supply for the growing global population. The development of new insecticides must meet rigorous regulatory requirements for safety and address the resistance issues of existing insecticides. Proteolysis-targeting chimeras (PROTACs), originally developed for human diseases, show promise in agriculture. They offer innovative insecticides tailored to overcome resistance, opening avenues for agricultural applications. In this study, we developed small-molecule degraders by incorporating pomalidomide as an E3 ligand. These degraders were linked to a ligand (spirotetratmat enol) targeting the ACC protein through a flexible chain, aiming to achieve the efficient control of insects. Compounds 9a-9d were designed, synthesized, and evaluated for biological activities and mechanisms. Among them, 9b exhibited superior potency against Aphis craccivora (LC50 = 107.8 μg mL-1) compared to others and effectively degraded ACC proteins through the ubiquitin-proteasome system. These findings highlight the potential of utilizing PROTAC-based approaches in the development of insecticides for efficient pest control.
Synthesis and Pesticidal Activities of Ester Derivatives of the Labdane Diterpenoid Andrographolide at the C-3 Position Containing the Isoxazoline Fragment and Their Toxicology Study against Tetranychus cinnabarinus Boisduval

Abstract:Due to the long-term overuse of chemical pesticides, serious resistance and negative problems on human health and the ecological environment have appeared. To develop plant-product-based pesticide candidates, a series of novel andrographolide ester derivatives containing the isoxazoline skeleton were prepared at the C-3 position. Their pesticidal activities were evaluated against three typical pests such as Mythimna separata Walker, Aphis citricola Van der Goot, and Tetranychus cinnabarinus Boisduval. Against M. separata, compounds Ik, IIf, IIg, and IIk showed 1.6-1.8 times insecticidal activity compared to that of andrographolide; against A. citricola, compounds 6, Ih, and IIh possessed 3.7-3.9-fold aphicidal activity compared to that of andrographolide; against T. cinnabarinus, compounds Ib, Ig, and IIk exhibited 7.4-9.1-fold promising acaricidal activity compared to that of andrographolide. It is worth mentioning that effects of IIk on morphological changes of the treated mite cuticle layer structures were observed by the scanning electron microscope imaging method. Compound IIk can be studied as a pesticidal lead for further structural modification.
In Silico Studies of Lamiaceae Diterpenes with Bioinsecticide Potential against Aphis gossypii and Drosophila melanogaster

Abstract:Background: The growing demand for agricultural products has led to the misuse/overuse of insecticides; resulting in the use of higher concentrations and the need for ever more toxic products. Ecologically, bioinsecticides are considered better and safer than synthetic insecticides; they must be toxic to the target organism, yet with low or no toxicity to non-target organisms. Many plant extracts have seen their high insecticide potential confirmed under laboratory conditions, and in the search for plant compounds with bioinsecticidal activity, the Lamiaceae family has yielded satisfactory results. Objective: The aim of our study was to develop computer-assisted predictions for compounds with known insecticidal activity against Aphis gossypii and Drosophila melanogaster. Results and conclusion: Structure analysis revealed ent-kaurane, kaurene, and clerodane diterpenes as the most active, showing excellent results. We also found that the interactions formed by these compounds were more stable, or presented similar stability to the commercialized insecticides tested. Overall, we concluded that the compounds bistenuifolin L (1836) and bistenuifolin K (1931), were potentially active against A. gossypii enzymes; and salvisplendin C (1086) and salvixalapadiene (1195), are potentially active against D. melanogaster. We observed and highlight that the diterpenes bistenuifolin L (1836), bistenuifolin K (1931), salvisplendin C (1086), and salvixalapadiene (1195), present a high probability of activity and low toxicity against the species studied.

Abstract:The insecticidal and repellent activities of the four wild plant extracts: stinging nettle, Urtica dioica L., white henbane, Hyoscyamus albus L., camphorweeds, Pluchea dioscoridis L., and cocklebur, Xanthium strumarium L. were tested compared to the insecticide spirotetramat against the 3 rd nymphal instar of Aphis gossypii Glover and adult females of Phenacoccus solenopsis Tinsley after 24 and 48 h of treatment. Their effects on acetylcholinesterase (AChE) and glutathione-S-transferase (GST) enzyme activities were also assessed to explore their possible mechanisms of action. The phenolic compounds of the plant extracts were investigated by high-performance liquid chromatography (HPLC) analysis. The most abundant compounds in the methanol extract from P. dioscoridis were gallic acid (14.45 μg/mL), and cinnamic acid (11.44 μg/mL); from U. dioica were caffeic acid (15.32 μg/mL) and syringic acid (13.47 μg/mL); from H. albus were syringic acid (7.12 μg/mL) and catechol (5.33 μg/mL); and from X. strumarium were p -coumaric acid (24.53 μg/mL) and pyrogallol (18.35 μg/mL). According to the LC 50 values, the contact toxicity of U. dioica , H. albus , P. dioscoridis , and X. strumarium against A. gossypii was 40.3, 60.6, 56.2, and 32.3 mg/L, whereas, the contact toxicity against P. solenopsis was 56.4, 67.5, 64.3, and 36.2 mg/L, compared to the spirotetramat (17.2 and 24.5 mg/L), respectively, at 48 h of treatment. The highest repellency rates were observed with LC 50 values for X. strumarium (63.2 and 60.3%), followed by U. dioica (46.2 and 43.5%), P. dioscoridis (45.3 and 42.8%), and H. albus extract (38.8 and 35.2%) compared to the spirotetramat (78.5 and 76.2%) against A. gossypii and P. solenopsis at 24 h, but decreased with time. The AChE and GST activities in pests were significantly inhibited, compared to control and spirotetramat, with dose and time-dependent effects. These findings indicate that tested wild extracts may be suggested as viable alternatives for aphids and mealybugs control.

Abstract:The oriental fruit fly, Bactrocera dorsalis (Hendel), is a notorious invasive pest that has raised concerns worldwide. Validamycin has been demonstrated to be a very strong inhibitor against trehalase in a variety of organisms. However, whether validamycin can inhibit trehalase activity to suppress trehalose hydrolysis and affect any other relevant physiological pathways in B. dorsalis remains unknown. In this study, the effects of validamycin injection on the synthesis and metabolism of trehalose and chitin were evaluated. The results show that validamycin injection significantly affected trehalase activity and caused trehalose accumulation. In addition, the downstream pathways of trehalose hydrolysis, including the synthesis and metabolism of chitin, were also remarkably affected as the expressions of the key genes in these pathways were significantly regulated and the chitin contents were changed accordingly. Intriguingly, the upstream trehalose synthesis was also affected by validamycin injection due to the variations in the expression levels of key genes, especially BdTPPC1. Moreover, BdTPPC1 was predicted to have a binding affinity to validamycin, and the subsequent in vitro recombinant enzyme activity assay verified the inhibitory effect of validamycin on BdTPPC1 activity for the first time. These findings collectively indicate that validamycin can be considered as a promising potential insecticide for the management of B. dorsalis.

Inhibition of trehalase affects the trehalose and chitin metabolism pathways in Diaphorina citri (Hemiptera: Psyllidae)

Abstract:The Asian citrus psyllid, Diaphorina citri is the principal vector of huanglongbing, which transmits 'Candidatus Liberibacter asiaticus'. Trehalase is a key enzyme involved in trehalose hydrolysis and plays an important role in insect growth and development. The specific functions of this enzyme in D. citri have not been determined. In this study, three trehalase genes (DcTre1‐1, DcTre1‐2, and DcTre2) were identified based on the D. citri genome database. Bioinformatic analysis showed that DcTre1‐1 and DcTre1‐2 are related to soluble trehalase, whereas DcTre2 is associated with membrane‐bound trehalase. Spatiotemporal expression analysis indicated that DcTre1‐1 and DcTre1‐2 had the highest expression levels in the head and wing, respectively, and DcTre2 had high expression levels in the fat body. Furthermore, DcTre1‐1 and DcTre1‐2 expression levels were induced by 20‐hydroxyecdysone and juvenile hormone Ⅲ, but DcTre2 was unaffected. The expression levels of DcTre1‐1, DcTre1‐2, and DcTre2 were significantly upregulated, which resulted in high mortality after treatment with validamycin. Trehalase activities and glucose contents were downregulated, but the trehalose content increased after treatment with validamycin. In addition, the expression levels of chitin metabolism‐related genes significantly decreased at 24 and 48 h after treatment with validamycin. Furthermore, silencing of DcTre1‐1, DcTre1‐2, and DcTre2 reduced the expression levels of chitin metabolism‐related genes and led to a malformed phenotype of D. citri. These results indicate that D. citri trehalase plays an essential role in regulating chitin metabolism and provides a new target for controlling of D. citri.This article is protected by copyright. Design, Synthesis, and Insecticidal Activity of Novel Doramectin Derivatives Containing Acylurea and Acylthiourea Based on Hydrogen Bonding

Abstract:Our recent investigation on the insecticidal activities of several doramectin derivatives preliminarily revealed that the presence of hydrogen bonds at the C4″ position of the molecule with target protein γ-aminobutyric acid (GABA) receptor was crucial for retaining high insecticidal activity. As a continuation of our research work on the development of new insecticides, two series of novel acylurea and acylthiourea doramectin derivatives were designed and synthesized. The bioassay results indicated that the newly synthesized compounds (5o, 5t, and 6t) exhibited higher insecticidal activity against diamondback moth, oriental armyworm, and corn borer than the control compounds doramectin, commercial avermectins, chlorbenzuron, and lead compound 3g in our laboratory. Specifically, compound 5t was identified as the most promising insecticide against diamondback moth, with a final mortality rate of 80.00% at the low concentration of 12.50 mg/L, showing approximately 7.75-fold higher potency than the parent doramectin (LC50 value of 48.1547 mg/L), 6.52-fold higher potency than commercial avermectins (LC50 value of 40.5507 mg/L), and 3.98-fold higher potency than compound 3g (LC50 value of 24.7742 mg/L). Additionally, molecular docking simulations revealed that compound 5t (2.17, 2.20, 2.56, and 2.83 Å) displayed stronger hydrogen-bond action in binding with the GABA receptor, better than that of compound 5o (1.64 and 2.15 Å) and compound 6t (2.20 and 2.31 Å) at the C4″ position. This work demonstrated that these compounds containing hydrogen-bond groups might contribute to the improvement of insecticidal activity and supply certain hints toward structure optimization design for the development of new insecticides.
Regulation of three novel pepper thiothiazolidinones on the fecundity of Spodoptera frugiperda

Abstract:Spodoptera frugiperda has emerged as a major invasive pest worldwide. The utilization of chemical pesticides not only poses numerous ecological concerns but also fosters resistance in S. frugiperda. In this study, we designed and synthesized three novel thiothiazolidinone compounds (6a, 7b, and 7e) and incorporated innovative thiothiazolidinone structural elements into the piperine skeleton. Treatment with compounds 6a and 7e resulted in the blackening and agglomeration of oviduct eggs within the ovaries of certain female moths, impeding the release of normal eggs. The levels of vitellogenin and vitellogenin receptor, along with three trehalase inhibitors, exhibited a dynamic equilibrium state, leading to no discernible change in egg production but a notable increase in the generation of low-hatching-rate egg fragments. Compared with the injection of 2%DMSO, the eclosion rate of 6a injection was significantly decreased, as followed the spawning time and longevity were prolonged or significantly prolonged in the trehalase inhibitors of 6a, 7b, and 7e. We aimed to investigate the regulatory impacts of three new pepper thiothiazolidinone compounds on the reproduction of S. frugiperda, and to authenticate the efficacy of novel alginase inhibitors in inhibiting the reproduction of S. frugiperda. This research endeavors to aid in the identification of efficient and steadfast trehalase inhibitors, thereby expediting the research and development of potent biological pesticides.
Scaffold hopping approaches for the exploration of herbicidally active compounds inhibiting Acyl-ACP Thioesterase

Abstract:Background: The sustainable control of weed populations is a significant challenge facing farmers around the world. Although various methods for the control of weeds exist, the use of small molecule herbicides remains the most effective and versatile approach. Striving to find novel herbicides that combat resistant weeds via the targeting of plant specific modes of action (MoAs), we further investigated the bicyclic class of acyl-acyl carrier protein (ACP) thioesterase (FAT) inhibitors in an effort to find safe and efficacious lead candidates. Results: Utilizing scaffold hopping and bioisosteric replacements strategies, we explored new bicyclic inhibitors of FAT. Amongst the investigated compounds we identified new structural motifs that showed promising target affinity coupled with good in vivo efficacy against commercially important weed species. We further studied the structure-activity relationship (SAR) of the novel dihydropyranopyridine structural class which showed promise as a new type of FAT inhibiting herbicides. Conclusion: The current work presents how scaffold hopping approaches can be implemented to successfully find novel and efficacious herbicidal structures that can be further optimized for potential use in sustainable agricultural practices. The identified dihydropyranopyridine bicyclic class of herbicides were demonstrated to have in vitro inhibitory activity against the plant specific MoA FAT as well as showing promising control of a variety of weed species, particularly grass weeds in greenhouse trials on levels competitive with commercial standards. © 2024 The Author(s). The Insecticidal Activity of Secondary Metabolites Produced by Streptomyces sp. SA61 against Trialeurodes vaporariorum (Hemiptera: Aleyrodidae)

Abstract:Trialeurodes vaporariorum Westwood poses a significant threat to vegetable and ornamental crops in temperate zones, resulting in notable reductions in yield and substantial economic burdens. In order to find compounds with high insecticidal activity against T. vaporariorum, five compounds were isolated and identified from the crude extract of Streptomyces sp. SA61. These include three new polyketides, named strekingmycins F-H (1-3); one new diterpenoid, named phenalinolactone CD8 (4); and one known compound, strekingmycin A (5). Their structures were analyzed using high-resolution electrospray ionization mass spectrometry and one-dimensional and two-dimensional nuclear magnetic resonance spectroscopy data and by comparing them with previously reported data. The insecticidal activities of compounds 1-5 against T. vaporariorum were evaluated. Among them, compound 5 exhibited the highest insecticidal activity, with an LC50 of 6.949 mg/L against T. vaporariorum at 72 h using the leaf-dip method. Lower insecticidal activities were found in compounds 1-4, with LC50 values of 22.817, 19.150, 16.981 and 41.501 mg/L, respectively. These data indicate that strekingmycin could be a potential candidate for a novel insecticide to control T. vaporariorum.
Susceptibility of Tetranychus urticae to the Alkaloidal Extract of Zanthoxylum schreberi Bark: Phenotypic and Biochemical Insights for Biotechnological Exploitation

Abstract:Tetranychus urticae Koch, a phytophagous mite, is one of the most significant crop pests globally. The primary method employed for controlling T. urticae involves chemical means, utilizing synthesized products, posing the risk of developing resistance. The urgency for novel strategies integrated into pest management programs to combat this mite is becoming increasingly imperative. Botanical pesticides emerge as a promising tool to forestall arthropod resistance. Among these, extracts from Rutaceae plants, abundant in bioactive specialized metabolites, have demonstrated potential as insecticides and miticides. In this study, various concentrations of alkaloidal extracts sourced from the bark of Zanthoxylum schreberi J.F.Gmel. (Rutaceae) were evaluated against T. urticae adult females. Furthermore, the extract’s combination with three distinct commercial acaricides (i.e., chlorfenapyr, cyflumetofen, and abamectin) was also assessed for this mite. Chemical characterization of the extract via LC-MS allowed for the annotation of various compounds related to ten benzylisoquinoline-derived alkaloids. The extract, both alone and in combination with commercial insecticides, yielded varying responses, inducing over 40% mortality at 2% w/w, demonstrating a 90% repellency rate at the same concentration, and exerting a moderate impact on fecundity. These treatments extended beyond phenotypic responses, delving into the biochemical effects on treated T. urticae females through an exploration of the impact on four enzymes, i.e., acetylcholinesterase (AChE), glutathione S-transferase (GST), esterases (GE), and P450-like monooxygenases (PMO). Employing consensus docking studies and in vitro enzymatic evaluations, it was discovered that the Z. schreberi-derived extract and its constituents significantly affected two key enzymes, AChE and GST (IC50 < 6 μM), which were associated with the phenotypic observations of T. urticae females. The evaluation of alkaloid-rich botanicals showcases promising potential as a relevant biotechnological strategy in addressing mite-related concerns, offering a pathway toward innovative and sustainable pest management solutions.

Abstract:To reduce the risk of resistance development, a novel fungicide with dual specificity is demanded. Trehalose is absent in animals, and its synthases, trehalose-6-phosphate synthase (TPS) and trehalose-6-phosphate phosphatase (TPP), are safe fungicide targets. Here, we report the discovery of a dual-specificity inhibitor of MoTps1 (Magnaporthe oryzae Tps1, TPS) and MoTps2 (M. oryzae Tps2, TPP). The inhibitor, named A1-4, was obtained from a virtual screening and subsequent surface plasmon resonance screening. In in vitro assays, A1-4 interacts with MoTps1 and MoTps2-TPP (MoTps2 TPP domain) and inhibits their enzyme activities. In biological activity assays, A1-4 not only inhibits the virulence of M. oryzae on host but also causes aggregation of conidia cytosol, which is a characteristic phenotype of MoTps2. Furthermore, hydrogen/deuterium exchange mass spectrometry assays support the notion that A1-4 binds to the substrate pockets of TPS and TPP. Collectively, A1-4 is a promising hit compound for the development of safe fungicide with dual-target specificity.
Effects of surfactins, Bacillus lipopeptides, on the behavior of an aphid and host selection by its parasitoid

Abstract:Background: Lipopeptides extracted from the Bacillus genus are emerging biopesticides, especially in protecting crops against phytopathogens. Among the three main families of lipopeptides, surfactins have been identified as having insecticidal properties against several insect orders. However, the sublethal effects of these promising biopesticides on insect pests and their natural enemies remain largely unknown. The aim of this study was to evaluate the effects of surfactins topically applied on black bean aphid Aphis fabae mortality. First, the effects of surfactins on aphid mortality were determined by delivering increasing concentrations to adults and nymphs. Second, the sublethal effects of surfactins on locomotor activity and feeding behavior of surviving aphids were evaluated using the electropenetrography method. Finally, the effect of host exposure to surfactins on host selection behavior by Aphidius matricariae parasitoid females was analyzed. Results: Four surfactins concentrations were studied (0.5, 1, 2.5 and 5 g L-1 ). There was concentration-dependent mortality in response to surfactins at 24 h after treatment. Surfactins impacted aphid behavior when delivered at 1 g L-1 by inducing a greater locomotor activity and a reduction in feeding activity. By contrast, at the third trophic level, exposure of aphid hosts to surfactins did not affect behaviors leading to host recognition and acceptance by parasitoid females. Conclusion: This study highlighted the consequences of aphid exposure to surfactins in the context of bottom-up regulation. Although surfactins could directly impact aphid behavior, they had no apparent consequences on the host selection behavior exhibited by parasitoid wasps.
A first-in-class dimethyl 2-acetamido terephthalate inhibitor targeting Conyza canadensis SHMT1 with a novel herbicidal mode-of-action

Abstract:Introduction: Herbicide application is a highly efficiency method of weed control that boots agricultural output and assures food security. The development of novel herbicides focuses on improved bioactivity and new modes of action. The amino acid biosynthesis was validated as a promising novel mode of action for herbicidal compounds. However, the amino acid biosynthesis enzyme remains largely unexplored for herbicidal targets. Objectives: Serine hydroxymethyl transferase (SHMT) is an essentialenzyme in the photorespiratory cycle. The study aims to explore Conyza canadensis SHMT1 (CcSHMT1) as a promising target for herbicide discovery. Methods: Structure determination of CcSHMT1 was resolved by X-ray crystallography. Virtual screening docking experiments were performed with Glide version 5.5. Novel derivatives of dimethyl 2-acetamido terephthalate were further designed, synthesized, and bioassay. The druggability of the inhibitor was evidenced by ultrastructural changes in mitochondria, in vivo and vitro enzyme activity assays, and genetics analysis. Results: CcSHMT1 has a typical PLP-dependent enzyme 3D structure. The dimethyl 2-acetamido terephthalate-containing compounds had herbicidal activity. Dimethyl 2-(2-(4-(2-(4-bromo-2-chlorophenoxy) acetyl)piperazin-1-yl)acetamido) terephthalate (Compound 9ay, EC50 = 193.8 g a.i./ ha) exhibited the highest herbicidal activity on tested weed among the synthesized compounds. Compound 9ay had no obvious adverse effect on the growth of maize and honeybees. Compound 9ay was verified to target CcSHMT1 as an herbicide candidate. Conclusion: A first-in-class CcSHMT1 inhibitor that could be developed as a potent herbicide with a new mode of action and provide an avenue for discovering novel inhibitors of pyridoxal-5-phosphate-dependent enzymes.

Abstract:Introduction: Spodoptera frugiperda is an omnivorous agricultural pest which is great dangerous for grain output. Methods: In order to investigate the effects of potential trehalase inhibitors, ZK-PI-5 and ZK-PI-9, on the growth and development of S. frugiperda , and to identify new avenues for S. frugiperda control, we measured the content of the trehalose, glucose, glycogen and chitin, enzyme activity, and gene expression levels in trehalose and chitin metabolism of S. frugiperda . Besides, their growth and development were also observed. Results: The results showed that ZK-PI-9 significantly reduced trehalase activity and ZK-PI-5 significantly reduced membraned-bound trehalase activity. Moreover, ZK-PI-5 inhibited the expression of SfTRE2, SfCHS2, and SfCHT , thus affecting the chitin metabolism. In addition, the mortality of S. frugiperda in pupal stage and eclosion stage increased significantly after treatment with ZK-PI-5 and ZK-PI-9, which affected their development stage and caused death phenotype (abnormal pupation and difficulty in breaking pupa). Discussion: These results have provided a theoretical basis for the application of trehalase inhibitors in the control of agricultural pests to promote future global grain yield.
