Abstract:Acid soil is a worldwide problem to plant production. Acid toxicity is mainly caused by a lack of essential nutrients in the soil and excessive toxic metals in the plant root zone. Of the toxic metals, aluminum (Al) is the most prevalent and most toxic. Plant species have evolved to variable levels of tolerance to aluminum enabling breeding of high Al-tolerant cultivars. Physiological and molecular approaches have revealed some mechanisms of Al toxicity in higher plants. Mechanisms of plant tolerance to Al stress include: 1) exclusion of Al from the root tips, and 2) absorbance, but tolerance of Al in root cells. Organic acid exudation to chelate Al is a feature shared by many higher plants. The future challenge for Al tolerance studies is the identification of novel tolerance mechanisms and the combination of different mechanisms to achieve higher tolerance. Molecular approaches have led to significant progress in explaining mechanisms and detection of genes responsible for Al tolerance. Gene-specific molecular markers offer better options for marker-assisted selection in breeding programs than linked marker strategies. This paper mainly focuses on recent progress in the use of molecular approaches in Al tolerance research.

Abstract:The non-essential metalloid arsenic (As) is widely distributed in soil and underground water of many countries. Arsenic contamination is a concern because it creates threat to food security in terms of crop productivity and food safety. Plants exposed to As show morpho-physiological, growth and developmental disorder which altogether result in loss of productivity. At physiological level, As-induced altered biochemistry in chloroplast, mitochondria, peroxisome, endoplasmic reticulum, cell wall, plasma membrane causes reactive oxygen species (ROS) overgeneration which damage cell through disintegrating the structure of lipids, proteins, and DNA. Therefore, plants tolerance to ROS-induced oxidative stress is a vital strategy for enhancing As tolerance in plants. Plants having enhanced antioxidant defense system show greater tolerance to As toxicity. Depending upon plant diversity (As hyperaccumulator/non-hyperaccumulator or As tolerant/susceptible) the mechanisms of As accumulation, absorption or toxicity response may differ. There can be various crop management practices such as exogenous application of nutrients, hormones, antioxidants, osmolytes, signaling molecules, different chelating agents, microbial inoculants, organic amendments etc. can be effective against As toxicity in plants. There is information gap in understanding the mechanism of As-induced response (damage or tolerance response) in plants. This review presents the mechanism of As uptake and accumulation in plants, physiological responses under As stress, As-induced ROS generation and antioxidant defense system response, various approaches for enhancing As tolerance in plants from the available literatures which will make understanding the to date knowledge, knowledge gap and future guideline to be worked out for the development of As tolerant plant cultivars.
Arsenic transport, detoxification, and recent technologies for mitigation: A systemic review

Abstract:Arsenic (As) is an acute toxic metalloid that affects plant growth and development. As is found in the environment in organic and inorganic forms, but arsenite As(III) and arsenate As(V) are the most prevalent forms that negatively impact the plants. Roots exposed to As can easily absorb it mainly through transporters that carry vital mineral nutrients. As reach the food chain via crops irrigated with As-polluted water and exerts a negative impact. Even at low levels, As exposure disrupts the regular functioning of plants by generating a high level of reactive oxygen species (ROS) results into oxidative damage, and disruption of redox system. Plants have built-in defence mechanisms to combat this oxidative damage. The development of a food crop with lower As levels is dependent upon understanding the molecular process of As detoxification in plants, which will help reduce the consumption of As-contaminated food. Numerous genes in plants that may provide tolerance under hazardous conditions have been examined using genetic engineering techniques. The suppression of genes by RNA interference (RNAi) and CRISPR-Cas 9 (CRISPR associated protein 9) technology revealed an intriguing approach for developing a crop that has minimal As levels in consumable portions. This study aims to present current information on the biochemical and molecular networks associated with As uptake, as well as recent advances in the field of As mitigation using exogenous salicylic acid (SA), Serendipita indica and biotechnological tools in terms of generating As-tolerant plants with low As accumulation.

Abstract:This review explains the transport, mobility, resistance and detoxification of toxic metalloid arsenic (As) in plants. Arsenic is ubiquitously present in Earth’s crust; however, numerous human interventions such as rapid industrialization use of As-based pesticides, insecticides and discharge of industrial wastes in water bodies leads to cumulative increase in As in the environment and has become a global challenge. Arsenic exists in different organic and inorganic forms, but inorganic forms such as pentavalent arsenate (AsV) and trivalent arsenite (AsIII) are more toxic and actively taken up by plants. Its toxicity is marked by generation of reactive oxygen species (ROS) that are capable of degrading various biomolecules of the cellular systems. To keep the ROS under the limit, plants have an array of enzymatic antioxidants such as superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), ascorbate peroxidase (APX), glutathione reductase (GR) and glutathione-S-transferase (GST); and non-enzymatic antioxidant like ascorbate, proline, and cysteine. Contrary to this, As-hyper-accumulator plants survive under high concentration of As through the strenuous action of Asv reduction into AsIII followed by the vacuolar compartmentalization of complex or inorganic As. Hence, this review focuses on the potential sources of As in the environment, its speciation and toxicity, and tolerance strategies in plants.

Abstract:Current research studies on arsenic in the plant and soil system reveal that arsenic toxicity to plant species is strongly influenced by many factors including its nature (organic or inorganic), speciation (i.e. As (V); arsenite, arsenate, As (III)), solubility, concentration, the type of plant species, and other soil variables affecting arsenic bioaccumulation in plants. Therefore, As (V) or As (III) toxicity, uptake, and detoxification vary among plant species. Herein, the origins and spread of As pollution and As speciation in soil are briefly examined. Further, the numerous physiological and molecular mechanisms contributing to As(V) and As(III) toxicity, absorption, and detoxification processes in plants are investigated in detail. The role of several enzymatic (glutathione reductase, ascorbate peroxidase superoxide dismutase, and catalase) and non-enzymatic (phosphorus, phytochelatins, proline, glutathione, salicylic acid, and nitric oxide) substances under As (V/III) stress have been explained using a theoretical framework that illustrates the toxicity and translocation pathways of this metalloid in plants. This review focuses on two main topics: (i) the role of different molecules in modifying As-induced toxicities in plants and (ii) the role of nanoparticles in reducing As stress in plants. Significant research gaps that need to be addressed in order to improve our scientific knowledge of As in soil-plant systems are also highlighted.

Abstract:Arsenic is a ubiquitous toxic metalloid abundant in Earth’s crust. It is of major concern with respect to its increased accumulation in soils, in the food chain, or in drinking water. This chapter will focus on recent progress on the mechanisms of its uptake, toxicity, and detoxification in microbes and in planta. Due to widespread occurrence in nature, both microbes and plants have evolved a wide range of tolerance and detoxification mechanisms such as reduced uptake, immobilization, chelation, reduction/oxidation, methylation, and efflux. Among microbes, the ars operon is a well-characterized genetic system for arsenic detoxification. The mechanisms proposed for metal detoxification and hyperaccumulation within the plant involve chelation of the metal cation by ligands and binding with thiol groups or sequestration of metals away from sites or metabolism in the cytoplasm, notably into the vacuole or cell wall. Finally, this chapter will also shed light on hyperaccumulators and mechanisms of hyperaccumulation.

Abstract:Arsenic is one of the most potent carcinogens in the biosphere, jeopardizing the health of millions of people due to its entrance into the human food chain through arsenic-contaminated waters and staple crops, particularly rice. Although the mechanisms of arsenic sensing are widely known in yeast and bacteria, scientific evidence concerning arsenic sensors or components of early arsenic signaling in plants is still in its infancy. However, in recent years, we have gained understanding of the mechanisms involved in arsenic uptake and detoxification in different plant species and started to get insights into arsenic perception and signaling, which allows us to glimpse the possibility to design effective strategies to prevent arsenic accumulation in edible crops or to increase plant arsenic extraction for phytoremediation purposes. In this context, it has been recently described a mechanism according to which arsenite, the reduced form of arsenic, regulates the arsenate/phosphate transporter, consistent with the idea that arsenite functions as a selective signal that coordinates arsenate uptake with detoxification mechanisms. Additionally, several transcriptional and post-translational regulators, miRNAs and phytohormones involved in arsenic signaling and tolerance have been identified. On the other hand, studies concerning the developmental programs triggered to adapt root architecture in order to cope with arsenic toxicity are just starting to be disclosed. In this review, we compile and analyze the latest advances toward understanding how plants perceive arsenic and coordinate its acquisition with detoxification mechanisms and root developmental programs.

Abstract:Arsenic (As) contamination of drinking water and groundwater used for irrigation can lead to contamination of the food chain and poses serious health risk to people worldwide. To reduce As intake through the consumption of contaminated food, identification of the mechanisms for As accumulation and detoxification in plant is a prerequisite to develop efficient phytoremediation methods and safer crops with reduced As levels. Transcriptome, proteome, and metabolome analysis of any organism reflects the total biological activities at any given time which are responsible for the adaptation of the organism to the surrounding environmental conditions. As these approaches are very important in analyzing plant As transport and accumulation, we termed "Arsenomics" as approach which deals transcriptome, proteome, and metabolome alterations during As exposure. Although, various studies have been performed to understand modulation in transcriptome in response to As, many important questions need to be addressed regarding the translated proteins of plants at proteomic and metabolomic level, resulting in various ecophysiological responses. In this review, the comprehensive knowledge generated in this area has been compiled and analyzed. There is a need to strengthen Arsenomics which will lead to build up tools to develop As-free plants for safe consumption.

Abstract:Arsenic (As) is a widespread carcinogenic pollutant. Phytoremediation is the most suited technology for alleviating the As contamination of soil. In this review, we have discussed the uptake mechanism and the associated transporters for different As species. Glutathione, phytochelatins, metallothionins, and secondary metabolites play important role in As detoxification and enhancing tolerance. The roles of MAPK signaling and calcium signaling are highlighted in the perception of As stress along with phytohormones signaling in stress tolerance. Furthermore, transcription factors involved in regulation of gene expression under As stress are discussed. High-throughput sequencing has reduced the time duration and enhanced the knowledge regarding understanding the molecular mechanism of phytoremediation. The role of CRISPR/Cas9 and synthetic genes in context to phytoremediation is discussed. We have provided a holistic understanding of the present knowledge about phytoremediation in the context of mechanisms of the As uptake and tolerance. A complete understanding of the phytoremediation process is essential for As-risk mitigation and will help in augmenting its efficiency and true potential.

Abstract:Arsenic (As)-contaminated soil and groundwater lead to contamination of the food chain and pose serious risk to human health worldwide. Arsenic accumulation, particularly inorganic arsenic, in food crops leads to potential health problems including cancer in populations. Elucidation of the mechanisms for As transport and metabolism is a prerequisite to develop safer crops with reduced As levels and efficient phytoremediation methods. Arsenate is taken up by plants through the phosphate transporters and arsenite and undissociated methylated As species through the nodulin 26-like intrinsic (NIP) and plasma membrane intrinsic (PIP) aquaporin channels. Arsenate is readily reduced to arsenite in planta, which is detoxified by complexation with thiol-rich peptides such as phytochelatins and/or vacuolar sequestration. Production of reactive oxygen species is induced by As exposure which can lead to the production of antioxidant metabolites and several enzymes involved in antioxidant defense. Nitrogen and sulfur assimilation pathways, oxidative carbon metabolism, and amino acid and protein relationships are also affected by As exposure. Plant-based remediation strategies either through natural or genetically modified plants for cleaning up of the As-contaminated sites is a cost-effective, green-clean technology that has been variously used for As containment. In the present chapter, the comprehensive knowledge generated in the area of arsenic transporters, its metabolism, and phytoremediation from contaminated soil has been discussed.

Abstract:Metalloid arsenic (As) is not a necessary element for plants, but its excessive accumulation is toxic to plants, and it also poses a great health risk to humans via the food chain. Plants absorb and metabolize As through a variety of processes. Arsenate in the form of As5+ is absorbed by phosphate transporters, but methylated As and As3+ enter plant tissues mainly through aquaporin channels. Various strategies and practices have been proposed and applied to alleviate As toxicity or reduce As accumulation in plants, but an efficient and environment-friendly approach has yet to be developed. This review comprehensively explores As sources and uptake mechanisms, as well as the interactions of phosphorus (P) and As in their uptake, transportation and influences on plant growth and physiological activities. This comprehensive review covers the transport, metabolism, and tolerance processes that plants exhibit in response to As stress and the addition of P. In addition, we also present recent advances in reducing As toxicity and accumulation by improving P nutrition, manipulating P transporter genes and optimizing the plant microbial community. Finally, the future research directions and main challenges are briefly discussed.

Abstract:Environmental contamination with arsenic (As) is a global environmental, agricultural and health issue due to the highly toxic and carcinogenic nature of As. Exposure of plants to As, even at very low concentration, can cause many morphological, physiological, and biochemical changes. The recent research on As in the soil-plant system indicates that As toxicity to plants varies with its speciation in plants (e.g., arsenite, As(III); arsenate, As(V)), with the type of plant species, and with other soil factors controlling As accumulation in plants. Various plant species have different mechanisms of As(III) or As(V) uptake, toxicity, and detoxification. This review briefly describes the sources and global extent of As contamination and As speciation in soil. We discuss different mechanisms responsible for As(III) and As(V) uptake, toxicity, and detoxification in plants, at physiological, biochemical, and molecular levels. This review highlights the importance of the As-induced generation of reactive oxygen species (ROS), as well as their damaging impacts on plants at biochemical, genetic, and molecular levels. The role of different enzymatic (superoxide dismutase, catalase, glutathione reductase, and ascorbate peroxidase) and non-enzymatic (salicylic acid, proline, phytochelatins, glutathione, nitric oxide, and phosphorous) substances under As(III/V) stress have been delineated via conceptual models showing As translocation and toxicity pathways in plant species. Significantly, this review addresses the current, albeit partially understood, emerging aspects on (i) As-induced physiological, biochemical, and genotoxic mechanisms and responses in plants and (ii) the roles of different molecules in modulation of As-induced toxicities in plants. We also provide insight on some important research gaps that need to be filled to advance our scientific understanding in this area of research on As in soil-plant systems.

Abstract:Arsenic (As) is a carcinogen and it is ubiquitous in the environment. Its accumulation in plants can enter the food chain, thereby causing adverse impacts on human health. Based on studies using Xenopus oocytes, engineered microbes, and model plants, aquaporins play key roles in arsenite (AsIII) passage across the cell membrane. Herein, we summarized the roles of aquaporins in AsIII uptake, transport, and accumulation. The coupling action between polar-localized aquaporins and positive transporters probably explains the efficient As transport mediated by bi-directional channels. The expression of aquaporins is regulated at both transcriptional and post-transcriptional levels via microRNAs. However, in response to As, the proteins can be modified through phosphorylation and be degraded via the ubiquitin 26S proteasome system. Further, a proposed model comprising reactive oxygen species, Ca, Ca-dependent kinases, and transcriptional factors is constructed to reveal the signaling pathways and aquaporins' responses to As in model plants. We also provided strategies to identify and investigate putative aquaporins in plants. Identification and modification of aquaporins' residues critical for their substrate selectivity, and optimization of their expression pattern by employing appropriate promoters can be employed to reduce As accumulation in crops. Based on the characteristics of their functions and expression patterns, it is promising to employ aquaporins for genetic-engineering of crops to reduce As content and increase food safety.

Abstract:Cadmium (Cd) and arsenic (As) are two highly toxic heavy metals and metalloids that coexist in many situations posing severe threats to plants. Our investigation was conducted to explore the different regulatory mechanisms of ryegrass ( Lolium perenne L.) responding to individual and combined Cd and As stresses in hydroponics. Results showed that the ryegrass well-growth phenotype was not affected by Cd stress of 10 mg·L -1 . However, As of 10 mg·L -1 caused rapid water loss, proline surge, and chlorosis in shoots, suggesting that ryegrass was highly sensitive to As. Transcriptomic analysis revealed that the transcription factor LpIRO2 mediated the upregulation of ZIP1 and YSL6 that played an important role in Cd tolerance. We found that the presence of As caused the overexpression of LpSWT12 , a process potentially regulated by bHLH14, to mitigate hyperosmolarity. Indoleacetic acid (IAA) and abscisic acid (ABA) contents and expression of their signaling-related genes were significantly affected by As stress rather than Cd. We predict a regulatory network to illustrate the interaction between transporters, transcription factors, and signaling transduction, and explain the antagonism of Cd and As toxicity. This present work provides a research basis for plant protection from Cd and As pollution.

Abstract:Arsenic is a ubiquitous metalloid in the biosphere, and its origin can be either geogenic or anthropic. Four oxidation states (-3, 0, +3 and + 5) characterize organic and inorganic As- compounds. Although arsenic is reportedly a toxicant, its harmful effects are closely related to its chemical form: inorganic compounds are most toxic, followed by organic ones and finally by arsine gas. Although drinking water is the primary source of arsenic exposure to humans, the metalloid enters the food chain through its uptake by crops, the extent of which is tightly dependent on its phytoavailability. Arsenate is taken up by roots via phosphate carriers, while arsenite is taken up by a subclass of aquaporins (NIP), some of which involved in silicon (Si) transport. NIP and Si transporters are also involved in the uptake of methylated forms of As. Once taken up, its distribution is regulated by the same type of transporters albeit with mobility efficiencies depending on As forms and its accumulation generally occurs in the following decreasing order: roots > stems > leaves > fruits (seeds). Besides providing a survey on the uptake and transport mechanisms in higher plants, this review reports on measures able to reducing plant uptake and the ensuing transfer into edible parts. On the one hand, these measures include a variety of plant-based approaches including breeding, genetic engineering of transport systems, graft/rootstock combinations, and mycorrhization. On the other hand, they include agronomic practices with a particular focus on the use of inorganic and organic amendments, treatment of irrigation water, and fertilization.

Abstract:This study investigates the role of the phenylpropanoid pathway in arsenic (As) contaminated rice genotypes under natural conditions, exploring the intricate relationship between genetic regulation and physiological adaptation. Differential approaches adapted by rice genotypes to counteract As exposure are elucidated here through analysis of enzyme activities and related gene expression patterns, docking simulations, and nutrient dynamics. Enzymatic analysis from the phenylpropanoid pathway revealed significant variations across rice genotypes, with Mini mansoori exhibiting notably higher activity levels of key enzymes (PAL, C4H, 4CL, CHI, DFR and F3H) compared to Sampoorna and Pioneer. Additionally, the gene expression profiling unveiled differential responses, with Mini mansoori and Pioneer demonstrating higher expression of genes ( OsPAL, OsCHS, OsCHI, OsF3H, OsF3′H, OsFLS, OsDFR , and OsLAR ) associated with As resistance and tolerance, compared to Sampoorna. Enrichment analysis emphasized the involvement of cinnamic acid biosynthesis and related pathways. Molecular docking depicted certain proteins, such as Os4CL, OsFLS, OsDFR, and OsLAR susceptible to As binding, potentially affecting enzymatic activity. Ionomic analysis unveiled that Mini mansoori maintained higher levels of essential nutrients such as Na, Ca, P, Mn, Mg, and Zn in grains. However, this contrasted with Pioneer and Sampoorna, which experienced nutrient imbalance likely due to higher As accumulation. Chlorophyll fluorescence analysis depicted that Mini mansoori and Pioneer maintained better photosynthetic efficiency under As toxicity compared to Sampoorna. Moreover, network analysis highlights the critical role of Mg and Na interaction with essential phenolics and flavonoids, in combating the stress. Harnessing this understanding, targeted breeding effort could yield As-resistant rice varieties with enhanced nutrient and flavonoid contents, addressing both food safety and malnutrition in affected regions.

Abstract:Arsenic (As) toxicity is an escalating problem; however, information about the metabolic events controlling the varied pattern of As accumulation in rice genotypes within their natural environment is still lacking. The present study is thus an advancement in unravelling the response of such rice genotypes. Soil-water-rice samples were analyzed for As accumulation using ICP-MS. Furthermore, we implemented metabolomics through LC-MS/MS and UHPLC to identify metabolic signatures regulating As content by observing the metalloid's composition in rice agrosystem. Results showed that rice genotypes differed significantly in their levels of metabolites, with Mini mansoori and Pioneer having the highest levels. Mini mansoori contained least As which might have been regulated by Ala, Ser, Glu, Phe, Asn, His, Ile, Lys, Gln, Trp, Tyr, chlorogenic, p-coumaric, trans-ferulic, rutin, morin, naringenin, kampferol, and myricetin, while Asp, Arg, Met, syringic, epigalocatechin, and apigenin contributed to the greater As acclimatization ability of Pioneer. Multivariate tools separated the rice genotypes into two major clusters: Pioneer-Mini mansoori and Damini-Sampoorna-Chintu. KEGG identified three major metabolic pathways (aminoacyl-tRNA, phenylpropanoid, and secondary metabolites biosynthesis route) linked with As tolerance and adaptation mechanisms in rice. Overall, these two genotypes symbolize their As hostile and accommodating attitudes probably due to the accumulated metabolites and the physicochemical attributes of the soil-water. Thus, thorough understanding of the metabolic reactions to As may facilitate the emergence of As tolerant/resilient genotypes. This will aid in the selection of molecular markers to cultivate healthier rice genotypes in As-contaminated areas.

Abstract:Heavy metals are important environmental pollutants and many of them are toxic even at very low concentrations. Pollution of the biosphere with heavy metals has accelerated dramatically since the industrial revolution. Metal toxicity is one of the major abiotic stresses leading to hazardous effects in plants. Plants are susceptible to heavy metal toxicity and respond to avoid detrimental effects in a variety of different ways. Several studies revealed the additive and coordinative involvement of heavy metal transporters, ligands such as phytochelatins and metallothioneins, antioxidant, glyoxalase systems, heat shock proteins, proline, polyamines, salicylic acid, nitric oxide and miRNAs. They alleviate heavy metal stress by many different ways including restriction of uptake and transport of metals and through symbiotic association between plants and ectomycorrhizas or rhizobacteria. The aim of this review is to integrate a recent understanding of physiological and biochemical mechanisms of heavy metal induced plant stress responses based on the findings of current plant molecular biological research. This paper also describes recent developments of transgenic plants with special reference to phytochelatin synthases and metallothionein genes for improving plant metal tolerance and further applications in phytoremediation. Undoubtedly, isolation and overexpression of phytochelatin synthase and metallothionein genes have fostered some understanding of their role in metal stress tolerance. However, single genes are unlikely to help in the amelioration of heavy metal stress tolerance. Therefore, constructs with combinations of PC synthesis genes like γ-glutamylcysteine synthase (GCS), glutathione synthase (GS), ATP sulfurylase and serine acetyl transferase may be important besides many transcription factors associated with metal stress tolerance for developing an efficient phytoremediation technology useful for removal of heavy metals from soils.

Abstract:Arsenic poses a global threat to living organisms, compromising crop security and yield. Limited understanding of the transcriptional network integrating arsenic-tolerance mechanisms with plant developmental responses hinders the development of strategies against this toxic metalloid. Here, we conducted a high-throughput yeast one-hybrid assay using as baits the promoter region from the arsenic-inducible genes ARQ1 and ASK18 from Arabidopsis thaliana, coupled with a transcriptomic analysis, to uncover novel transcriptional regulators of the arsenic response. We identified the GLABRA2 (GL2) transcription factor as a novel regulator of arsenic tolerance, revealing a wider regulatory role beyond its established function as a repressor of root hair formation. Furthermore, we found that ANTHOCYANINLESS2 (ANL2), a GL2 subfamily member, acts redundantly with this transcription factor in the regulation of arsenic signaling. Both transcription factors act as repressors of arsenic response. gl2 and anl2 mutants exhibit enhanced tolerance and reduced arsenic accumulation. Transcriptional analysis in the gl2 mutant unveils potential regulators of arsenic tolerance. These findings highlight GL2 and ANL2 as novel integrators of the arsenic response with developmental outcomes, offering insights for developing safer crops with reduced arsenic content and increased tolerance to this hazardous metalloid.
