Abstract:Microbial recurrent infections and antimicrobial resistance have motivated the researchers to explore bioactive compounds as safe alternative antimicrobials to target pathogenic microorganisms. Interest in finding new biologically active substances to replace conventional antibiotics in aquatic feed has rapidly increased, since the misuse or overuse of antibiotics produced antibiotic resistance. Glycerol monolaurate (GML), has attracted attention of researchers due to its growth-promoting and immunomodulatory potential by exerting beneficial effects on the gut microbiota and host health. This study aimed to evaluate the effects of dietary GML using as a feed additive on the growth performance, blood lipid profile, hepatic lipid deposition, body composition, digestive enzymes, serum anti-inflammatory activities, and gut microbiota of zebrafish. The study’s findings showed that diet supplemented with 750 mg kg−1 of GML, significantly improved the growth performance, feed utilization and intestinal lipase activity, as well as body crude lipid content without inducing hepatic fat accumulation. GML supplementation also promoted the serum anti-inflammatory potential by elevating the level of TGF-β1 and IL-10. Moreover, dietary supplementation of GML positively restructured the intestinal microbial ecology by improving the relative abundance of several favorable bacteria, including Cetobacterium, Shewanella, and Vibrio at the genus level. Conclusively, study’s findings suggest that GML supplementation contributes in improving the growth, digestive enzymes activities, anti-inflammatory potential, and gut microbiota, indicating GML promising potential as a feed additive in aquaculture nutrition.
Effects of Partial Substitution of Fish Meal with Soybean Products and Chicken Meal on Growth, Antioxidant Capacity and Intestinal Microbiota of Penaeus Monodon

Abstract:The aim of this experiment was to investigate the effects of the partial substitution of fish meal with soybean products and chicken meal on the growth performance, antioxidant capacity and intestinal microbiota of Penaeus monodon. A total of 450 healthy, consistent shrimp were randomly divided into five groups, with three replicates per group and 30 shrimp per replicate. The proportion of fish meal substituted with soybean products and chicken meal in the five feed groups was 0% (FM), 40% (40SC), 60% (60SC), 80% (80SC) and 100% (100SC). The experiment lasted for 8 weeks. The results showed that, compared to the FM group, the 40SC and 60SC groups had a decrease in WG and SR, but there was no significant difference (p > 0.05). In contrast, compared to the FM group, the FCR in the 100SC group was significantly increased (p < 0.05), while there was no significant difference among the FM and 40SC, 60SC and 80SC groups (p > 0.05). Compared to the FM group, the ACP in the 80SC and 100SC groups significantly increased (p < 0.05), while the 40SC and 60SC groups had no significant difference (p > 0.05). The AKP in the 100SC group was significantly higher than that in the FM group (p < 0.05), while there were no significant differences among the other four groups (p > 0.05). There were no significant differences in T-AOC and T-SOD among all the treatment groups (p > 0.05). The next-generation sequencing of the intestinal microbiota showed that Proteobacteria was the most abundant phylum in the five groups, accounting for 37.67%, 66%, 40%, 40% and 43.33%, respectively. Compared to the FM group, the Fusobacteriota in the other four groups decreased significantly (p < 0.05). The functional prediction of FAPROTAX indicated that no functional components were observed which are harmful to the body. Considering the effects on growth performance, antioxidant capacity and intestinal microbiota, it is feasible to use soybean products and chicken meal to replace 60% of fish meal in the feed of P. monodon.

Abstract:An eight-week feeding trial was conducted to examine the impact of dietary supplementation with glycerol monolaurate (GML) on juvenile black sea bream. A basal diet was formulated containing 24% fish meal, while five additional diets were prepared, each supplemented with varying levels of GML: GML1 (0.01%), GML2 (0.02%), GML3 (0.04%), GML4 (0.08%), and GML5 (0.16%). Triplicate tanks were randomly allocated to each diet, each containing 20 fish with an initial weight of 1.55 ± 0.05 g. By the trial’s end, the GML3 group displayed a notably higher final body weight (FBW), weight gain (WG), specific growth rate (SGR), and protein efficiency ratio (PER) compared to the other groups (p < 0.05), but the FCR was significantly higher in the control group. However, no significant differences were observed in the MFI, PPV, CF, HSI, IPF, VSI, or SR among the groups (p > 0.05). Regarding the proximate compositions of the dorsal muscle and whole body, no substantial differences were observed across the groups (p > 0.05). Additionally, there were no significant variations in digestive enzyme activity (p > 0.05), serum immune, or biochemical parameters in the midgut and hindgut among the treatment groups. But in the serum immune response IgM, C3 and C4 were significantly higher in the GML3 group as compared to the other groups (p < 0.05). However, the GML3 group exhibited significantly greater fore-intestinal villus height, crypt depth, villus height per crypt depth, and the number of goblet cells per villus compared to the other groups (p < 0.05). Overall, GML supplementation, particularly GML3, significantly improved growth indicators like the final body weight and intestinal morphology. While certain parameters remained unaffected, these findings suggest GML’s potential as a beneficial dietary supplement in fish diets.

Abstract:Simple Summary The multienzyme complex is composed of one or several single enzymes as the main component, mixed with other single enzyme preparations, or obtained by the fermentation of one or more microorganisms, and an the exogenous enzyme mixture with multiple functions of a single enzyme. Supplementation with exogenous enzyme preparations offsets the negative effects of removing antibiotics from animal diets. The multienzyme complex supplemented in aquatic feed can enhance the secretion and activity of endogenous digestive enzymes in aquatic animals. Meanwhile, exogenous digestive enzymes can also help aquatic animals to decompose some anti-nutrients, improve the utilization and digestibility of feed, and promote animal growth. The present study evaluated the impact of dietary multienzyme complex (MEC) supplementation on growth performance, digestive enzyme activity, histomorphology, serum metabolism and hepatopancreas glycometabolism in snakeheads (Channa argus). A total of 600 fish (initial weight, 69.70 +/- 0.30 g) were randomly divided into four groups. Four diets were formulated: (1) control (basic diet); (2) E1 (400 U kg(-1) amylase, 150 U kg(-1) acid protease, 1900 U kg(-1) neutral protease and basic diet); (3) E2 (800 U kg(-1) amylase, 300 U kg(-1) acid protease, 3800 U kg(-1) neutral protease and basic diet); and (4) E3 (1200 U kg(-1) amylase, 450 U kg(-1) acid protease, 5700 U kg(-1) neutral protease and basic diet). The results show that the E2 group increased the specific growth rate, weight gain rate and the final body weight, as well as decreasing the blood urea nitrogen, alanine aminotransferase and triglyceride. The mRNA levels and activities of digestive enzymes and key glucose metabolism enzymes in the hepatopancreas were enhanced in snakeheads fed the MEC. Meanwhile, moderate MEC diet (E2 groups) supplementation improved digestive tract morphology, increased the glycogen in the hepatopancreas and the lipids in the dorsal muscle. Moreover, plasma metabolomics revealed differential metabolites mainly involved in amino acid metabolism. These findings suggest that dietary supplementation with the MEC improved growth performance, digestive tract morphology, gene expression and the activity of digestive enzymes, enhanced the glycolysis-gluconeogenesis and amino acid metabolism of snakeheads, and the optimal composition of the MEC was group E2.

Abstract:Four experimental diets were formulated to evaluate the effect of coated lysine (Lys) and methionine (Met) supplemented in mixture protein diets of juvenile black sea bream, Acanthopagrus schlegelii. Fish meal (FM) was the main protein source in control diet, and 21.2% FM was replaced by the mixture of soybean protein concentrate, meat and bone meal, and monosodium glutamate protein in replacement diets with coating Lys and Met were suppled at none (0%, 0%), low (1.7%, 1.1%) and high (5.0%, 3.4%) level (designated as NLM, LLM and HLM diet, respectively). Each diet was fed to triplicate groups of 20 fish (initial mean weight, 4.13 +/- 0.16 g) twice a day to apparent satiation for 8 weeks, in a flow-through system. Results showed that amino acid supplementation significantly improved weight gain and specific growth rate in these mixture protein diets groups, and the fish fed with HLM diet showed comparable growth with the control group. Feed efficiency ratio (FER), protein efficiency ratio (PER), and crude protein and Met content in dorsal muscle were significantly affected by diet amino acid level. The highest apparent digestibility coefficients appeared in HLM group but the values showed insignificant difference with the control group. Serum activities of aspertate aminotransferase and alanine aminotransferase in control group were lower than those of in other treatments. Fish fed with NLM and LLM diets had higher triglycerides concentration than those of fed the HLM and FM diets. Furthermore, the feed cost of the various feed treatments were evaluated, and the three test diets were all lower than the control diet. The present study indicated that 21.2% FM in black sea bream diet can be successfully replaced by mixture protein with sufficient coated Lys (5.0%) and Met (3.4%) supplemented.

Abstract:Glycerol monolaurate (GML) is a chemical compound derived from the combination of lauric acid and glycerol, exhibiting potent antimicrobial properties. An eight-week feeding trial was conducted to evaluate the effects of dietary supplementation of Glycerol Monolaurate (GML) on juvenile black sea bream. The control diet included 24% fish meal, while five additional diets were formulated with increasing GML concentrations: 0.01% (GML1), 0.02% (GML2), 0.04% (GML3), 0.08% (GML4), and 0.16% (GML5). Each diet was assigned to triplicate tanks. At the end of the experiment, serum immune and antioxidant in liver were non-significantly different among the groups. The relative gene expression of IGF-1 and GH in liver was statistically higher in the GML3 treated. The relative gene expression of IL-1, TGF, TNF-1, CCK, CLDN1, CASS-3 and GPR41 in intestine were statistically higher in the GML3 group. The photomicrographs of transmission electron microscope of the intestine were also studied. The GML supplemented groups had longer brush borders. The cells seemed to be joined by an intact junctional complex without visible intercellular spaces. The observed variations in phyla, class, and genus did not demonstrate statistical significance. Proteobacteria, Cyanobacteria, Firmicutes and Actinobacteria in the phylum, and Oxyphotobacteria, Bacteroidia, Holophagae and Negativicutes in the class, Brevundimonas, and Achromobacter in the genera were all substantially increased with higher levels of GML supplementation. Based on these results, it is suggested that glycerol monolaurate (GML) may serve as a potential dietary supplement for promoting different gene expression and intestinal health in black sea bream.
Improvement of hybrid grouper (Epinephelus fuscoguttatus ♀ × E. lanceolatus ♂) by enzyme-digested poultry by-product: Growth performance, amino acid and peptide transport capacity, and intestinal morphology

Abstract:Background At present, fish meal (FM) resources are in short supply, and competition for food between humans and animals is becoming increasingly critical. Finding non-grain protein sources that can replace FM is the key to solving the rapid development of aquaculture. Methods Seven trial diets were prepared with 0 g/kg (EP0), 30 g/kg (EP3), 60 g/kg (EP6), 90 g/kg (EP9), 120 g/kg (EP12), 150 g/kg (EP15), and 180 g/kg (EP18) of enzyme-digested poultry by-product meal (EPBM) by replacing of FM. A total of 630 hybrid groupers ( Epinephelus fuscoguttatus ♀ × E. lanceolatus ♂) were equally portioned into 21 tanks. At 8:00 and 16:00 each day, groupers were fed until they were full for a cumulative period of 8 weeks. Results The results showed that 30 g/kg of EPBM significantly increased the rates of weight gain and special growth ( P < 0.05). Significantly higher activities of serum glutamic pyruvic transaminase, glutamic oxaloacetic transaminase, catalase, and superoxide dismutase were observed in the EP3 group ( P < 0.05). The categories and numbers of the top 10 dominant bacteria in the phylum and genus levels were not significantly influenced by feed ( P > 0.05). In the proximal intestine and distal intestine, there were significantly higher expressions of SNAT3, LAAT1, CAT2 , and CAT1 in the EP3 group compared with the EP0 group ( P < 0.05). In the EP3 group, the expressions of PepT1, LAAT1, B 0, + AT , and CAT2 were significantly increased in MI than those in all other groups (except the EP0 group, P < 0.05). Conclusion When FM was replaced by 30 g/kg of EPBM, growth performance, antioxidant capacity, and the ability to transport amino acids and peptides of hybrid grouper were significantly improved.

Abstract:Introduction As the aquaculture industry intensifies to enhance production efficiency and capacity, the risk of disease outbreaks in high-density systems, such as those for largemouth bass, has escalated. This necessitates the exploration of novel strategies for disease prevention and control. Studies have shown that Atractylodes macrocephala polysaccharide (AMP) possesses the functions of promoting growth and enhancing immune capacity, making it a potential feed additive in animal production. Methods This study aimed to assess the impact of AMP on the growth performance, serum biochemical indices, and intestinal flora structure of largemouth bass. A total of 360 healthy largemouth bass (mean weight: 15.25 ± 1.29 g) were randomly assigned to four groups, each with three replicates. The groups were fed diets supplemented with 0% (Group D, control group), 0.4% (Group A), 0.8% (Group B), or 1.2% (Group C) AMP for 42 days. Results The results showed that the addition of an appropriate amount of AMP in the feed significantly improved the weight gain rate (WGR) and specific growth rate (SGR) of largemouth bass, reduced the feed conversion ratio (FCR), and enhanced growth performance. AMP reduced the levels of AST and ALT, indicating a hepatoprotective effect on largemouth bass, with significant differences from the control group (P < 0.05). AMP also improved the intestinal microbiota composition of largemouth bass, positively affecting intestinal health. The analysis of intestinal microbiota revealed 1,288 amplicon sequence variants (ASVs) in the intestine of largemouth bass in Group B, dominated by Firmicutes and Bacteroidota. In contrast, Group C (1.2%) had 920 ASVs, with Fusobacteriota and Firmicutes as the major components, while the relative abundance of Firmicutes was lower, showing significant differences from the control Group D. The functional analysis of intestinal microbiota based on KEGG showed significant differences (P < 0.05) among the four groups in Carbon metabolism, Biosynthesis of amino acids, and Metabolic pathways. Discussion The study concludes that the inclusion of AMP at 0.8% – 1.2% in the feed can enhance the growth performance and intestinal health of largemouth bass, offering a promising strategy for disease prevention and control in intensive aquaculture settings.

Abstract:The present study investigated the effects of Astragalus membranaceus extract (AME) on growth performance, immune response, and energy metabolism of juvenile largemouth bass (Micropterus salmoides). Seven diets containing 0%, 0.1%, 0.2%, 0.3%, 0.4%, 0.5%, and 0.6% AME (Con, AME0.1, AME0.2, AME0.3, AME0.4, AME0.5, and AME0.6 groups) were formulated and fed to M. salmoides for 8 weeks. Final body weight (FBW), feed intake (FI), weight gain (WG), and specific growth rate (SGR) were all significantly higher in AME0.4 group than in Con group (P < 0.05). Feed conversion rate (FCR) was significantly improved in AME0.5 group compared with Con group (P < 0.05). Whole-body crude protein contents were significantly increased in AME0.2 group (P < 0.05). Whole-body crude lipid contents were significantly lower in AME0.2 and AME0.3 groups, while muscle lipid was upregulated by dietary AME (P < 0.05). Hepatic malondialdehyde (MDA) contents were significantly lowered in AME0.3 and AME0.4 groups, and catalase (CAT) activities were significantly increased in AME0.1 and AME0.2 groups (P < 0.05). Plasma aspartate aminotransferase (AST) level was significantly lowered in AME0.5, and AME0.6 groups, and alanine aminotransferase (ALT) level was lowered in AME0.5 groups (P < 0.05). Plasma triglyceride was declined in AME0.6 group, and glucose was decreased by 0.3%-0.5% AME (P < 0.05). Significantly higher hepatocyte diameter, lamina propria width, and submucosal layer thickness were recorded in AME0.6 groups, while the longest villi height was obtained in AME0.2 and AME0.3 groups (P < 0.05). The mRNA expression levels of insulin-like growth factor 1 (igf1) revealed the growth-promoting effect of AME. The anti-inflammatory and antiapoptotic effects of AME were demonstrated by transcription levels of interleukin 8 (il-8), tumor necrosis factor-alpha (tnf-a), caspase, B-cell lymphoma-xl (Bcl-xl), bcl-2 associated x (Bax), and bcl-2-associated death protein (Bad). The transcription levels of lipid metabolism and gluconeogenesis related genes, including acetyl-CoA carboxylase alpha (acc1), fatty acid synthase (fasn), fatty acid binding protein 1 (fabp1), phosphoenolpyruvate carboxykinase 2 (pepck2), and glucose-6-phosphatase catalytic subunit 1a (g6pc), were reduced by AME treatment, while the levels of glycolysis-related genes, including glucokinase (gck) and pyruvate kinase (pk), were the highest in AME0.2 and AME0.3 groups (P < 0.05). According to polynomial regression analysis of SGR, WG, FCR, whole-body crude lipid, MDA, and ALT, the optimal AME supplementation level was estimated to be 0.320%-0.429% of the diet. These results provided insights into the roles of AME in regulating immunity and metabolism, which highly indicated its potential as immunostimulants and metabolic regulators in diverse aquatic animals.
Dietary Antimicrobial Peptides Improve Intestinal Function, Microbial Composition and Oxidative Stress Induced by Aeromonas hydrophila in Pengze Crucian Carp (Carassius auratus var. Pengze)

Abstract:There is increasing evidence for the potential use of antimicrobial peptides as dietary supplements and antibiotic substitutes. In this study, we analyzed the differential effects of varying levels of antimicrobial peptides on the intestinal function and intestinal microbial and disease resistance of Pengze crucian carp. Approximately 630 experimental fishes were randomized in the control group (G0: 0 mg/kg) and in five groups supplemented with different doses of AMPs (G1: 100 mg/kg, G2: 200 mg/kg, G3: 400 mg/kg, G4: 800 mg/kg, and G5: 1600 mg/kg) and were fed for ten weeks. Three replicates per group of 35 fish were performed. The results showed that AMPs promoted intestinal villus development and increased intestinal muscular thickness (p < 0.05) and goblet cell abundance. The enzymatic activities of all groups supplemented with AMPs were effectively improved. AMP supplementation significantly enhanced the activities of antioxidant enzymes and digestive enzymes in the intestines of G3 animals (p < 0.05). Compared with G0 animals, AMP-supplemented animals regulated the expression of intestinal immune-related genes and exhibited significant differences in the G3 animal group (p < 0.05). The abundance of intestinal Firmicutes and Bacteroidetes increased in the AMP-supplemented groups, but the Firmicutes/Bacteroidetes ratio was lower than that in the G0 group. AMP supplementation also decreased the abundance of Fusobacterium while increasing the proportion of Actinobacteria (p < 0.05). After Aeromonas hydrophila infection, the expression levels of anti-inflammatory factors in the intestinal tract of G3 animals were significantly upregulated, and the level of the proinflammatory factor was decreased (p < 0.05). The intestinal Cetobacterium levels of G3 animals were significantly increased (p < 0.01), while the Proteobacteria levels were decreased, and the intestinal goblet cell proliferation was significantly lower than that of G0 animals (p < 0.05). This indicates that groups supplemented with AMPs have better disease resistance than the G0 group and can rapidly reduce the adverse effects caused by inflammatory response. Taken together, the present results suggest that AMP supplementation can improve intestinal function and intestinal microbial and pathogen resistance in Pengze crucian carp.

Abstract:The cultivation of largemouth bass ( Micropterus salmoides ), a species of significant economic value in aquaculture, has experienced notable growth recently. However, the deterioration of water quality seriously affects the metabolic responses of M. salmoides . While compound microbial agent (CMA) is widely utilized for ecological rehabilitation and water filtration, its application in M. salmoides has not been reported. Here, based on physio-biochemical tests and 16S rRNA sequencing, we investigated the effects of CMA (yeast, Bacillus subtilis , and lactic acid bacteria) on the water quality within the recirculating aquaculture system, along with physiological indices and gut microbiota of M. salmoides . Compared to the control and single microbial agent (yeast), CMA treatment improved the water quality by improving the dissolved oxygen and delaying the increase of pH, total nitrogen, total phosphorus, ammonia nitrogen, and nitrite. The 16s rRNA gene sequencing revealed that the water treated with CMA exhibited elevated levels of chao1, Shannon, Pd, and a larger population of dominant bacterial. Besides, higher values of ACE, chao1, Shannon, and OTU level, and lower Simpson index were found in CMA treated M. salmoides samples, suggesting that CMA treatment enhanced the species richness and diversity of gut microbiota of M. salmoides . Furthermore, CMA treatment hindered the generation and proliferation of harmful bacteria, such as the Mycoplasma mobile 163K species and the Erysipelotrichaceae family, which was associated with enhanced antioxidant enzymatic activity and decreased MDA level in both the serum and liver. These findings shed light on the essential roles of CMA in M. salmoides culturing and introduce an innovative approach to enhance the aquatic environment.

Abstract:The effects of diets fermented with compound probiotics, namely Lactobacillus acidophilus, Limosilactobacillus reuteri and Lactiplantibacillus plantarum, on the growth performance, physiological and biochemical indexes, fatty acid composition and intestinal health of juvenile largemouth bass (Micropterus salmoides) were investigated. Three hundred healthy juvenile M. salmoides (5.29 ± 0.02 g) were selected and randomly divided into two groups with triplicates for each. The basic diet was set as the control group (CON), and fermentation of the basic diet with a mixed bacterial solution (1.8 × 109 cfu/mL, L. acidophilus:L. reuteri:L. plantarum = 1:1:1) was set as the fermentation group (FER). Fish were hand fed to satiation for 56 days and two-thirds of the culture water was renewed every 3 days. The results showed that feed intake of fish in the FER group was significantly lowered, thereby increasing feed efficiency (FE) and protein efficiency (PER) (p < 0.05). Serum alanine aminotransferase (ALT) activity was significantly decreased, and catalase (CAT) activity was significantly increased in the FER group (p < 0.05). The liver superoxide dismutase activity (SOD) was significantly enhanced, and intestinal trypsin was significantly increased in the FER group (p < 0.05). Being fed with the fermentation diet significantly increased the content of n-3 polyunsaturated fatty acids (PUFA), docosahexaenoic acid (DHA) and the n-3/n-6 PUFAs ratios in the liver (p < 0.05). Intestinal histology showed that villus height and width of the intestine and the number of goblet cells were significantly increased in the FER group (p < 0.05). Those fed with fermentation diets had limited diversity of gut microbiota. Compared to the CON group, the relative abundance of Aeromonas decreased significantly (p < 0.05), while the relative abundance of Fusobacteria, Cetobacteria and Lactobacillusis in FER increased greatly in the gut microbiota of the FER group. In conclusion, fermented feed with the three probiotics effectively improved the feed utilization and antioxidant capacity, promoted digestion and absorption of dietary protein, improved the ability of synthesize DHA and n-3 PUFAs in the liver and reduced the abundance of pathogenic bacteria in the gut. Therefore, the present research provided a new way of co-fermented feed with three probiotics for the aquaculture of M. salmoides.

[Examination of the pleural effusion and its interpretation].

Abstract:Pleural effusion appears in the course of respiratory diseases and disorders of other organs. Examination of the effusion enables differentiation between exudate or transudate. It allows to estimate the severity of the disease, helps to choose the way of treatment and assesses its effects. In most cases Light's criteria are sufficient for differentiation between exudate and transudate. If the patient has an exudative pleural effusion additional tests are indicated to determine the etiology of the effusion. Laboratory tests which are useful in the further diagnostics of pleural effusion include: bacterial cultures, cytological and biochemical tests.

Abstract:This study aimed to investigate the effects of fishmeal (FM) replacement by rice protein concentrate (RPC) with a xylooligosaccharides supplement on the growth performance, antioxidant capability and immune response against Aeromonas hydrophila in blunt snout bream (Megalobrama amblycephala). Fish (46.85 ± 0.34 g) were randomly assigned to one of 6 diets, namely the control diet (containing FM), the RPC diet (FM replaced by RPC) and RPC diet supplemented with 0.5, 1.5, 2.3 and 3% XOS respectively, for 8 weeks. After the feeding trial, fish were challenged by Aeromonas hydrophila for 96 h with the blood and liver sample obtained at 48 and 96 h respectively. The results showed that the final weight, weight gain and protein efficiency ratio of fish fed RPC diet were significantly (P < 0.05) lower than that of the control group, whereas the opposite was true for FCR. However, the supplement of 1.5% XOS remarkably (P < 0.05) improved these parameters compared to the control diet. Plasma total iron binding capacity of fish fed the RPC diet showed little difference (P > 0.05) with that of the control group, but it enhanced significantly (P < 0.05) with the supplement of 1.5% XOS compared to the control group. After bacterial infection, plasma lysozyme (LYM), complement 3, complement 4, myeloperoxidase (MPO), acid phosphatase (ACP) and alkaline phosphatase activities, as well as immunoglobulin M, levels all increased significantly (P < 0.05) with the maximum value is attained at 48 h, then they decreased significantly (P < 0.05) with further increasing time at 96 h. Similar results were also observed in liver superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX) activities as well as malondialdehyde (MDA) content. Regarding dietary treatment, these parameters of fish fed RPC diet showed little difference (P > 0.05) with those fed the control diet but were significantly enhanced (P < 0.05) when RPC diet was supplemented with 1.5-2.3% XOS, except for hepatic (MDA) content which showed an opposite trend compared to the control group. After 96 h of challenge, the relative percentage survival (RPS) of fish fed XOS was significantly higher (P < 0.05) than that of fish offered the control and RPC diet. In addition, significant (P < 0.05) interactions between dietary XOS and sampling time were also observed in plasma LYM, MPO, ACP, and hepatic SOD, CAT, GPX, MDA, as well as RPS. In conclusion, this study indicated that dietary supplementation of 1.5%XOS could significantly improve the growth performance, antioxidant capability, innate immunity and A. hydrophila resistance of blunt snout bream fed diets with FM replaced by RPC.

Abstract:Mannan oligosaccharides (MOS) have been studied and applied as a feed additive, whereas their regulation on the growth performance and immunity of aquatic animals lacks consensus. Furthermore, their immunoprotective effects on the freshwater fish Megalobrama amblycephala have not been sufficiently studied. Thus, we investigated the effects of dietary MOS of 0, 200, and 400 mg/kg on the growth performance, non-specific immunity, intestinal health, and resistance to Aeromonas hydrophila infection in juvenile M. amblycephala . The results showed that the weight gain rate of juvenile M. amblycephala was not significantly different after 8 weeks of feeding, whereas the feed conversion ratio decreased in the MOS group of 400 mg/kg. Moreover, dietary MOS increased the survival rate of juvenile M. amblycephala upon infection, which may be attributed to enhanced host immunity. For instance, dietary MOS increase host bactericidal and antioxidative abilities by regulating the activities of hepatic antimicrobial and antioxidant enzymes. In addition, MOS supplementation increased the number of intestinal goblet cells, and the intestine was protected from necrosis of the intestinal folds and disruption of the microvilli and junctional complexes, thus maintaining the stability of the intestinal epithelial barrier. The expression levels of M. amblycephala immune and tight junction-related genes increased after feeding dietary MOS for 8 weeks. However, the upregulated expression of immune and tight junction-related genes in the MOS supplemental groups was not as notable as that in the control group postinfection. Therefore, MOS supplementation might suppress the damage caused by excessive intestinal inflammation. Furthermore, dietary MOS affected the richness and composition of the gut microbiota, which improved the gut health of juvenile M. amblycephala by increasing the relative abundance of beneficial gut microbiota. Briefly, dietary MOS exhibited significant immune protective effects to juvenile M. amblycephala , which is a functional feed additive and immunostimulant.

Abstract:Effects of antimicrobial peptides (AMP) added to diets on aquatic animal health and body function are influenced by multiple factors such as animal species, initial body weight, the dosage of AMP and feeding duration. However, there is limited knowledge on the relationship between these factors and the body function of aquatic animals. Here, we aimed to perform multiple meta-analyses to investigate the effects of dietary AMP on growth performance (feed conversion ratio [FCR], specific growth rate [SGR]), enzyme activity (superoxide dismutase activity [SOD], lysozyme activity [LSA]), disease resistance (cumulative survival rate [CSR], the expression of immune-related genes [GENE]) and the abundance of gut microbiota (MICRO) from a pool of empirical studies. Additionally, the dose-effect model was applied to determine the optimal AMP dose, initial body weight and feeding duration to maximize body function. To conduct the meta-analyses, we included 34 publications that estimated 705 effect sizes across 21 fish, 2 shrimp and 2 shellfish species. The results confirmed that the inclusion of AMP in the diet can significantly improve SGR, SOD, LSA, CSR and GENE and decrease FCR for aquatic animals. Interestingly, our findings implied a slight positive effect of AMP on MICRO albeit with a limited number of studies available on fish gut microbial communities. Although no significant linear or quadratic relationship was predicted by meta-regression, the dose-effect indicated that the optimal AMP doses for FCR, SGR, SOD and LSA were 707.5, 750.0, 1,050.0 and 937.5 mg/kg, respectively. Taken together, fish with an initial body weight of 30 g could be fed with a dose of 600 to 800 mg/kg for 2 mo when AMP-supplemented diets were applied in aquaculture, which can effectively improve body function and health while lowering aquafeed costs. In addition, more studies should address fish gut microbiota to delimitate the influence of dietary AMP on MICRO in the future.
Dection of Direct Versus Indirect Effects: Were Experiments Long Enough?

Abstract:To evaluate the hypothesis that indirect effects generally take much longer to become evident in manipulative studies of community regulation than do direct effects and thus may often be missed, I studied the effect of experiment duration in a survey of marine intertidal interaction webs. Contrary to expectation, indirect effects appeared either simultaneously with direct effects or shortly after direct effects were evident. While experiment durations varied greatly, on average most direct and indirect effects became statistically significant within the first 20%–40% of the total experiment duration. Further, the duration of most experiments appeared sufficient so that most indirect effects that would be generated by the manipulation could be observed. On average, a period of “constancy” (i.e., of no further change) lasting roughly 20%–60% of the total experiment duration occurred after the last indirect effect was observed. Experiment duration did not vary with web species richness, which suggests no tendency to perform manipulations for more (or less) time in more complex webs. The number of indirect effects per species did not increase with increasing experiment duration, nor did the number of longer interaction chains (four species vs. three species), which suggests no trends for increased complexity of indirect effects with longer experiments. Ecological theory states that, in interaction webs whose dynamics are imperfectly known, indirect effects may compromise the predictability of species manipulations. However, empirical results suggest that, despite incomplete knowledge of indirect effects, community dynamics may be more predictable than expected.

Abstract:In this paper, the application of adaptive system identification based on genetic algorithm is realized. A high order unknown plant is modeled with a lower order adaptive finite impulse response (FIR) filter. Also, a performance comparison has been made between the conventional gradient based least mean square (LMS) algorithm and genetic algorithm (GA). Simulation results show that the genetic algorithm based design method has a better performance in terms of convergence speed and error performance.

Abstract:In order to investigate the effect of bacterium Bacillus amyloliquefaciens on growth performance,digestive enzymes activities,serum proteins levels and liver superoxide dismutase( SOD) activity in juvenile black seabream( Sparus macrocephalus),this paper reared the fish with 7 g body weight in a small cement pool of an indoor microwater circulation system and fed diets containing Bacillus amyloliquefaciens at a dose of 0,0. 025%,0. 05%,and 0.1%,respectively. After 56 d,the results showed that the bacterium led to improve weight gain rate( WGR) and specific growth rate( SGR) in juvenile black seabream. The WGR and SGR were significantly higher in juvenile black seabream fed by diet supplemented with 0.05% of Bacillus amyloliquefaciens than that of the fish fed by control diet.The intestinal protease and lipase activities in the fish fed by diet supplemented with Bacillus amyloliquefaciens were significantly higher than that of the control group. The total serum protein levels of juveniles fed with 0.025% Bacillus dietary supplement were significantly higher than that of the control group,among them the globulin and albumin levels were extremely increased. In addition,the intestinal amylase activity and hepatic SOD activity were also obviously higher in the 0. 1% Bacillus amyloliquefaciens dietary supplement group than that of the control. These results indicated that optimal dose of Bacillus amyloliquefaciens administration could increase the intestinal digestive enzyme activity,improve antioxidant ability in juvenile black seabream,and promote their growth performance.
