Abstract:The importance of maize (Zea mays L.) to global agriculture, world economy, and food security is widely known and increasing. Current maize breeding programs are deeply integrated with recent and rapid technological advances in genome sequencing, computational biology, and new genotyping and phenotyping technologies. Transformation and genome editing capabilities are a central hub to an array of advanced molecular and breeding approaches to crop improvement. Tissue culture and somatic embryogenesis play essential and central roles in maize transformation biology. Synergistic applications of maize transformation, advanced genomics, and genome editing provide a potent interdependent triad for functional genomics research and advanced molecular breeding. Implementation of advanced capabilities to transform maize and conduct genome editing will profoundly influence the dynamics of global agriculture ushering in a new era of varietal development and molecular breeding. With over 60.9 Mha planted in 2019 alone, biotech maize accounts for 31% of the world's maize production. Up to 10% higher yields are achieved using new varieties generated using genetic modification technologies compared to similar conventional varieties. By extension, the impact new varietal releases developed through genome editing will likely be more significant. Advances in transformation and genome editing technologies will facilitate an even wider applicability for the development of new varieties with increasingly complex traits. The introduction of biochemical pathways and the use of synthetic biology have become increasingly more attainable. The future is of genotype independent maize transformation and genome editing, as the working platform will impact world agriculture, global food security, and plant science well into the future.

Abstract:This investigation characterizes a prominent nuclear matrix protein isolated from tissue cultured mouse lens epithelial cells. The nuclear matrix protein was isolated using a modified Penman technique. Total nuclear matrix proteins were further separated by SDS‐polyacrylamide gel electrophoresis. The SDS‐PAGE profile of the nuclear matrix proteins displayed a prominent doublet band at 60 kDa region. Nonequilibrium 2D gel electrophoresis revealed that this protein is a basic nuclear protein. This 60 kDa protein was further characterized by comparing its internal peptide amino acid sequence with known protein sequence using the BLAST technique, and this study demonstrated that 60 kDa nuclear matrix protein displays significant sequence similarity with Xenopus Laevis heat shock transcription factor. We also raised antibodies against 60 kDa nuclear matrix protein. Immunofluorescence, studies showed that this 60 kDa nuclear matrix protein preferably decorates nucleus, and puncted pattern of fluorescence suggest presence of this protein in the discrete areas of the nucleus. Heat shock transcription factors upregulate synthesis of heat shock proteins and many of these protein act as molecular chaperones. Thus, presence of a nuclear matrix protein with significant sequence similarity with heat shock transcription factor suggests sustained heat shock protein synthesis in the mouse lens cells. J. Cell. Abstract:Regenerable, embryogenic haploid cell suspensions were initiated and established from type II pollen calluses of two selected Chinese maize genotypes (No 592 Y and 592.A2 LY). The induction frequency of friable, embryogenic callus (type II) was highly dependent on three factors: genotype, medium, cold pretreatment, and on their interactions. Repeated callus and cell selection during the culture procedure led to stable haploid suspensions consisting of fine clusters each containing 20–50 cells. The selected cell lines were able to maintain their morphogenic ability during long-term subculture (2 years). Protoplasts were successfully isolated from subcultured, friable, embryogenic pollen calluses and cultured on N6BM and N6K media using a feeder layer, obtained from 2-day-old suspension culture. Healthy plants were regenerated from protoplast-derived calluses.
Whole Transcriptome Profiling of Maize during Early Somatic Embryogenesis Reveals Altered Expression of Stress Factors and Embryogenesis-Related Genes

Abstract:Embryogenic tissue culture systems are utilized in propagation and genetic engineering of crop plants, but applications are limited by genotype-dependent culture response. To date, few genes necessary for embryogenic callus formation have been identified or characterized. The goal of this research was to enhance our understanding of gene expression during maize embryogenic tissue culture initiation. In this study, we highlight the expression of candidate genes that have been previously regarded in the literature as having important roles in somatic embryogenesis. We utilized RNA based sequencing (RNA-seq) to characterize the transcriptome of immature embryo explants of the highly embryogenic and regenerable maize genotype A188 at 0, 24, 36, 48, and 72 hours after placement of explants on tissue culture initiation medium. Genes annotated as functioning in stress response, such as glutathione-S-transferases and germin-like proteins, and genes involved with hormone transport, such as PINFORMED, increased in expression over 8-fold in the study. Maize genes with high sequence similarity to genes previously described in the initiation of embryogenic cultures, such as transcription factors BABY BOOM, LEAFY COTYLEDON, and AGAMOUS, and important receptor-like kinases such as SOMATIC EMBRYOGENESIS RECEPTOR LIKE KINASES and CLAVATA, were also expressed in this time course study. By combining results from whole genome transcriptome analysis with an in depth review of key genes that play a role in the onset of embryogenesis, we propose a model of coordinated expression of somatic embryogenesis-related genes, providing an improved understanding of genomic factors involved in the early steps of embryogenic culture initiation in maize and other plant species.

Abstract:Efficient methods for in vitro propagation, regeneration, and transformation of plants are of pivotal importance to both basic and applied research. While being the world’s major food crops, cereals are among the most difficult-to-handle plants in tissue culture which severely limits genetic engineering approaches. In maize, immature zygotic embryos provide the predominantly used material for establishing regeneration-competent cell or callus cultures for genetic transformation experiments. The procedures involved are demanding, laborious and time consuming and depend on greenhouse facilities. We have developed a novel tissue culture and plant regeneration system that uses maize leaf tissue and thus is independent of zygotic embryos and greenhouse facilities. We report here: (i) a protocol for the efficient induction of regeneration-competent callus from maize leaves in the dark, (ii) a protocol for inducing highly regenerable callus in the light, and (iii) the use of leaf-derived callus for the generation of stably transformed maize plants.

Abstract:Maize ( Zea mays ssp. mays ) is a popular genetic model due to its ease of crossing, well-established toolkits, and its status as a major global food crop. Recent technology developments for precise manipulation of the genome are further impacting both basic biological research and biotechnological application in agriculture. Crop gene editing often requires a process of genetic transformation in which the editing reagents are introduced into plant cells. In maize, this procedure is well-established for a limited number of public lines that are amenable for genetic transformation. Fast-Flowering Mini-Maize (FFMM) lines A and B were recently developed as an open-source tool for maize research by reducing the space requirements and the generation time. Neither line of FFMM were competent for genetic transformation using traditional protocols, a necessity to its status as a complete toolkit for public maize genetic research. Here we report the development of new lines of FFMM that have been bred for amenability to genetic transformation. By hybridizing a transformable maize genotype high Type-II callus parent A (Hi-II A) with line A of FFMM, we introgressed the ability to form embryogenic callus from Hi-II A into the FFMM-A genetic background. Through multiple generations of iterative self-hybridization or doubled-haploid method, we established maize lines that have a strong ability to produce embryogenic callus from immature embryos and maintain resemblance to FFMM-A in flowering time and stature. Using an Agrobacterium -mediated standard transformation method, we successfully introduced the CRISPR-Cas9 reagents into immature embryos and generated transgenic and mutant lines displaying the expected mutant phenotypes and genotypes. The transformation frequencies of the tested genotypes, defined as the numbers of transgenic event producing T1 seeds per 100 infected embryos, ranged from 0 to 17.1%. Approximately 80% of transgenic plants analyzed in this study showed various mutation patterns at the target site. The transformable FFMM line, FFMM-AT, can serve as a useful genetic and genomic resource for the maize community.

Abstract:Summary The ability of plant somatic cells to dedifferentiate, form somatic embryos and regenerate whole plants in vitro has been harnessed for both clonal propagation and as a key component of plant genetic engineering systems. Embryogenic culture response is significantly limited, however, by plant genotype in most species. This impedes advancements in both plant transformation‐based functional genomics research and crop improvement efforts. We utilized natural variation among maize inbred lines to genetically map somatic embryo generation potential in tissue culture and identify candidate genes underlying totipotency. Using a series of maize lines derived from crosses involving the culturable parent A188 and the non‐responsive parent B73, we identified a region on chromosome 3 associated with embryogenic culture response and focused on three candidate genes within the region based on genetic position and expression pattern. Two candidate genes showed no effect when ectopically expressed in B73, but the gene Wox2a was found to induce somatic embryogenesis and embryogenic callus proliferation. Transgenic B73 cells with strong constitutive expression of the B73 and A188 coding sequences of Wox2a were found to produce somatic embryos at similar frequencies, demonstrating that sufficient expression of either allele could rescue the embryogenic culture phenotype. Transgenic B73 plants were regenerated from the somatic embryos without chemical selection and no pleiotropic effects were observed in the Wox2a overexpression lines in the regenerated T0 plants or in the two independent events which produced T1 progeny. In addition to linking natural variation in tissue culture response to Wox2a, our data support the utility of Wox2a in enabling transformation of recalcitrant genotypes.

Abstract:A novel, efficient maize genetic transformation system was developed using Agrobacterium -mediated transformation of embryo explants from mature seeds. Seeds from field grown plants were sterilized and crushed to isolate embryo explants consisting of the coleoptile, leaf primordia, and shoot apical meristem which were then purified from the ground seed bulk preparation. The infection of relevant tissues of seed embryo explants (SEEs) by Agrobacterium was improved by the centrifugation of the explants. Transgenic plants were obtained by multiple bud induction on high cytokinin media, followed by plant regeneration on hormone-free medium. Three different selectable markers ( cp4 epsps, aadA , and nptII ) were successfully used for producing transgenic plants. Stable integration of transgenes in the maize genome was demonstrated by molecular analyses and germline transmission of the inserted transgenes to the next generation was confirmed by pollen segregation and progeny analysis. Phenotypic evidence for chimeric transgenic tissue was frequently observed in initial experiments but was significantly reduced by including a second bud induction step with optimized cytokinin concentration. Additional improvements, including culturing explants at an elevated temperature during bud induction led to the development of a revolutionary system for efficient transgenic plant production and genome editing. To our knowledge, this is the first report of successful transgenic plant regeneration through Agrobacterium -mediated transformation of maize mature SEEs. This system starts with mature seed that can be produced in large volumes and the SEEs explants are storable. It has significant advantages in terms of scalability and flexibility over methods that rely on immature explants.

Abstract:The regenerative capacity of the embryonic callus, a complex quantitative trait, is one of the main limiting factors for maize transformation. This trait was decomposed into five traits, namely, green callus rate (GCR), callus differentiating rate (CDR), callus plantlet number (CPN), callus rooting rate (CRR), and callus browning rate (CBR). To dissect the genetic foundation of maize transformation, in this study multi-locus genome-wide association studies (GWAS) for the five traits were performed in a population of 144 inbred lines genotyped with 43,427 SNPs. Using the phenotypic values in three environments and best linear unbiased prediction (BLUP) values, as a result, a total of 127, 56, 160, and 130 significant quantitative trait nucleotides (QTNs) were identified by mrMLM, FASTmrEMMA, ISIS EM-BLASSO, and pLARmEB, respectively. Of these QTNs, 63 QTNs were commonly detected, including 15 across multiple environments and 58 across multiple methods. Allele distribution analysis showed that the proportion of superior alleles for 36 QTNs was <50% in 31 elite inbred lines. Meanwhile, these superior alleles had obviously additive effect on the regenerative capacity. This indicates that the regenerative capacity-related traits can be improved by proper integration of the superior alleles using marker-assisted selection. Moreover, a total of 40 candidate genes were found based on these common QTNs. Some annotated genes were previously reported to relate with auxin transport, cell fate, seed germination, or embryo development, especially, GRMZM2G108933 (WOX2) was found to promote maize transgenic embryonic callus regeneration. These identified candidate genes will contribute to a further understanding of the genetic foundation of maize embryonic callus regeneration.

Abstract:The traditional breeding approaches brought a lot of success in sorghum improvement in the last few decades. However, identification and transfer of novel genes in sorghum for productivity and resistance to biotic and abiotic stresses through contemporary genetic engineering tools have potential to enhance overall sorghum productivity. Producing transgenic plants in vitro of sorghum, being a recalcitrant, is a highly challenging task. Efficient and stable genetic transformation for generating transgenic plants is of great importance for developing a successful and commercially viable event. Agrobacterium-mediated genetic transformation of plants is the most widely used among all known transformation methods. In recent days, the progress in Agrobacterium-mediated transformation of sorghum has led to the emergence of various transgenic technologies, including overexpression of recombinant protein, knockdown of targeted gene through RNAi, and targeted genome editing for genetic improvement. However, the use of modern genome editing tools such as meganucleases (MgNs), zinc finger nucleases (ZFNs), transcription activator-like effector nucleases (TALENs), and clustered regularly interspaced short palindromic repeats (CRISPR) and CRISPR-associated endonuclease (Cas) has been slowly picking up the pace. Genetic improvement in sorghum through tissue culture-dependent genetic transformation and advantages of modern genetic tools for improving the targeted traits were discussed in this chapter.
A Short History and Perspectives on Plant Genetic Transformation

Abstract:Plant genetic transformation is an important technological advancement in modern science, which has not only facilitated gaining fundamental insights into plant biology but also started a new era in crop improvement and commercial farming. However, for many crop plants, efficient transformation and regeneration still remain a challenge even after more than 30 years of technical developments in this field. Recently, FokI endonuclease-based genome editing applications in plants offered an exciting avenue for augmenting crop productivity but it is mainly dependent on efficient genetic transformation and regeneration, which is a major roadblock for implementing genome editing technology in plants. In this chapter, we have outlined the major historical developments in plant genetic transformation for developing biotech crops. Overall, this field needs innovations in plant tissue culture methods for simplification of operational steps for enhancing the transformation efficiency. Similarly, discovering genes controlling developmental reprogramming and homologous recombination need considerable attention, followed by understanding their role in enhancing genetic transformation efficiency in plants. Further, there is an urgent need for exploring new and low-cost universal delivery systems for DNA/RNA and protein into plants. The advancements in synthetic biology, novel vector systems for precision genome editing and gene integration could potentially bring revolution in crop-genetic potential enhancement for a sustainable future. Therefore, efficient plant transformation system standardization across species holds the key for translating advances in plant molecular biology to crop improvement.

Abstract:Summary Mutations play a pivotal role in shaping the trajectory and outcomes of a species evolution and domestication. Maize (Zea mays) has been a major staple crop and model for genetic research for more than 100 yr. With the arrival of site‐directed mutagenesis and genome editing (GE) driven by the Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR), maize mutational research is once again in the spotlight. If we combine the powerful physiological and genetic characteristics of maize with the already available and ever increasing toolbox of CRISPR‐Cas, prospects for its future trait engineering are very promising. This review aimed to give an overview of the progression and learnings of maize screening studies analyzing forward genetics, natural variation and reverse genetics to focus on recent GE approaches. We will highlight how each strategy and resource has contributed to our understanding of maize natural and induced trait variability and how this information could be used to design the next generation of mutational screenings.

Somatic embryogenesis and optimization of regeneration system from immature embryos in maize inbred lines

Abstract:Maize production and productivity is on a sharp decline due to abiotic and abiotic stresses, therefore, an efficient regeneration protocol is an important tool that can contribute to maize improvement and gene-function studies to improve food security for the ever-growing population. The objective of this study was to optimize a regeneration system for CML 444 inbred line with CML 442 maize inbred line used as a reference. Callus was generated by incubation of immature embryos in Murashige and Skoog (MS) medium with vitamins supplemented with 0 - 4 g L-1 of 2, 4-D hormones, 900 mg L-1 proline, 250 mg L-1casein hydrolysate and 10 mg L-1 of filter sterilized AgNO3, 30 g L-l of sucrose and 3 g L-1 gelrite. Somatic embryo maturation was achieved by transferring 6-week old callus to MS medium with vitamins prepared as previously in callus induction with 60 g L-1 of sucrose and zero plant growth regulators (PGR). Shoot initiation was conducted in MS medium with vitamins supplemented with BAP, NAA at varied concentrations and a 0 mg L-1 control. Plants at a 3-leaf stage that had not rooted were transferred to MS media with vitamins with IBA at a concentration of 0 - 0.3 mg L–l. The 2, 4-D rates were significantly (p≤0.001) different for callus onset and callus induction. The genotype × rate interaction effects showed that 0 and 2 g L-1 2, 4-D had the lowest and highest mean, respectively in both lines during onset and induction of callus. The lines had significant (p≤0.001) effects on shooting induction, however, their means were not significantly different. Similarly, the means for the hormones were not significantly different for shooting induction. The lines, IBA rate and their interaction were significantly (p≤0.05) different for rooting induction. The means for the lines were significantly different for rooting induction in different IBA rates. Conversely, the mean for the IBA rates was significantly different for rooting induction. This study found that plant growth regulators rates during the callus induction stage play a key role during regeneration. This protocol was a success and could provide a fundamental platform for future transformation in this line.

Abstract:Maize (Zea mays) is one of the most important cereal crops to secure food supplies in the world. A growing world population requires new approaches to facilitate and accelerate maize breeding. As a powerful tool, the CRISPR/Cas system has revolutionized the fields of functional genomics and crop breeding. Compared to cross breeding and transgenic breeding, genome-editing breeding based on CRISPR/Cas will bring new opportunities in rapidly cultivating novel maize varieties with desired characteristics. Here, we review the applications of the CRISPR/Cas9 system in maize improvement. We also discuss the potential of new CRISPR/Cas technologies, such as base editors, prime editors, and multiplex genome editing toward future maize breeding. In addition, challenges for genome-editing breeding are discussed.
The production of callus capable of plant regeneration from immature embryos of numerous Zea mays genotypes

Abstract:In the summer of 1983, immature embryos from 101 selfed inbred lines and germplasm stocks of Zea mays L. were examined for their ability to produce callus cultures capable of plant regeneration (regenerable cultures) using a medium with which some limited success had previously been obtained. Forty-nine of the genotypes (49%) produced callus which visually appeared similar to callus previously cultured and shown to be capable of plant regeneration. After five months, 38 of these genotypes were alive in culture and plants were subsequently regenerated from 35 (92%) of them. No correlation was observed between plant regeneration and callus growth rate, the vivipary mutation (genes vp1, 2, 5, 7, 8 and 9), or published vigor ratings based on K+ uptake by roots. When F1 hybrid embryos were cultured, 97% of the hybrids having at least one regenerable parent also produced callus capable of plant regeneration. No regenerable cultures were obtained from any hybrid lacking a parent capable of producing a regenerable callus culture.In the summer of 1984, immature embryos from 218 additional inbred lines and germplasm stocks were plated and examined for their ability to produce regenerable callus cultures on media containing altered micronutrient concentrations, 3,6-dichloro-o-anisic acid (dicamba), glucose, and elevated levels of vitamin-free casamino acids and thiamine. Of these genotypes 199 (91%) produced callus that was regenerable in appearance. In the 1984 study, plant regeneration was noted in many commercially important inbreds, including B73, Mo17, B84, A632, A634, Ms71, W117, H993H95 and Cm105. Thus tissue-culture techniques are now available to obtain callus cultures capable of plant regeneration from immature embryos of most maize genotypes.

Abstract:Abstract Genome-editing tools such as the clustered regularly interspaced short palindromic repeats/Cas9 (CRISPR/Cas9) system have become essential tools for increasing the efficiency and accuracy of plant breeding. Using such genome-editing tools on maize, one of the most important cereal crops of the world, will greatly benefit the agriculture and the mankind. Conventional genome-editing methods typically used for maize involve insertion of a Cas9-guide RNA expression cassette and a selectable marker in the genome DNA; however, using such methods, it is essential to eliminate the inserted DNA cassettes to avoid legislative concerns on gene-modified organisms. Another major hurdle for establishing an efficient and broadly applicable DNA-free genome-editing system for maize is presented by recalcitrant genotypes/cultivars, since cell/tissue culture and its subsequent regeneration into plantlets are crucial for producing transgenic and/or genome-edited maize. In this study, to establish a DNA-free genome-editing system for recalcitrant maize genotypes/cultivars, Cas9-gRNA ribonucleoproteins were directly delivered into zygotes isolated from the pollinated flowers of the maize-B73 cultivar. The zygotes successfully developed and were regenerated into genome-edited plantlets by co-culture with phytosulfokine, a peptide phytohormone. The method developed herein made it possible to obtain DNA- and selectable-marker-free genome-edited recalcitrant maize genotypes/cultivars with high efficiency. This method can advance the molecular breeding of maize and other important cereals, regardless of their recalcitrant characteristics.

Abstract:Recent findings call for the critical overview of some incorrectly used plant cell and tissue culture terminology such as dedifferentiation, callus, totipotency, and somatic embryogenesis. Plant cell and tissue culture methods are efficient means to preserve and propagate genotypes with superior germplasm as well as to increase genetic variability for breading. Besides, they are useful research tools and objects of plant developmental biology. The history of plant cell and tissue culture dates back to more than a century. Its basic methodology and terminology were formulated preceding modern plant biology. Recent progress in molecular and cell biology techniques allowed unprecedented insights into the underlying processes of plant cell/tissue culture and regeneration. The main aim of this review is to provide a theoretical framework supported by recent experimental findings to reconsider certain historical, even dogmatic, statements widely used by plant scientists and teachers such as "plant cells are totipotent" or "callus is a mass of dedifferentiated cells," or "somatic embryos have a single cell origin." These statements are based on a confused terminology. Clarification of it might help to avoid further misunderstanding and to overcome potential "terminology-raised" barriers in plant research.

Abstract:This review charts the major developments in the genetic manipulation of plant cells that have taken place since the first gene transfer experiments using Ti plasmids in 1983. Tremendous progress has been made in both our scientific understanding and technological capabilities since the first genetically modified（GM）crops were developed with single gene resistances to herbicides, insects, viruses, and the silencing of undesirable genes. Despite opposition in some parts of the world, the area planted with first generation GM crops has grown from 1.7 Mhm~2 in 1996 to 179.7 Mhm~2 in 2015.The toolkit available for genetic modification has expanded greatly since 1996 and recently Nobel Laureates have called on Greenpeace to end their blanket opposition,and plant scientists have urged that consideration be given to the benefits of GM crops based on actual evidence. It is now possible to use GM to breed new crop cultivars resistant to a much wider range of pests and diseases, and to produce crops better able to adapt to climate change.The advent of new CRISPR-based technologies makes it possible to contemplate a much wider range of improvements based on transfer of new metabolic pathways and traits to improve nutritional quality, with a much greater degree of precision. Use of GM, sometimes in conjunction with other approaches, offers great opportunities for improving food quality, safety, and security in a changing world.
