Abstract:Much is known about molecular mechanisms by which animals detect pathogenic microbes, but how animals sense beneficial microbes remains poorly understood. The roundworm Caenorhabditis elegans is a microbivore that must distinguish nutritive microbes from pathogens. We characterized a neural circuit used by C. elegans to rapidly discriminate between nutritive bacteria and pathogens. Distinct sensory neuron populations responded to chemical cues from nutritive Escherichia coli and pathogenic Enterococcus faecalis , and these neural signals are decoded by downstream AIB interneurons. The polyamine metabolites cadaverine, putrescine, and spermidine produced by E. coli activate this neural circuit and elicit positive chemotaxis. Our study shows how polyamine odorants can be sensed by animals as proxies for microbe identity and suggests that, hence, polyamines might have widespread roles brokering host-microbe interactions.

The marine worm Capitella teleta is sensitive to neurochemical manipulation, as revealed via a novel high-throughput behavioural tool

Abstract:Anthropogenic impacts have led to increases in contaminants in marine habitats. The presence of compounds such as pharmaceuticals, personal care products, and pesticides is a concern, as these compounds have been shown to act as neurochemicals in aquatic organisms. However, screens of the multitude of chemicals found environmentally have yet to be carried out. Capitella teleta are marine annelid polychaete worms that live in the sediment of estuarian environments, acting as ecosystem engineers for marine habitats. This study aimed to develop a behaviour tool to identify neurochemical pathways involved in controlling locomotion that could be applied to screen the impacts of toxicants in the environment using this key invertebrate species at multiple life stages. Adult and juvenile life stages (2 weeks post-emergence) of Capitella teleta were observed in Petri dishes (adults only) or 6-well plates (adults and juveniles), and conserved behavioural responses were isolated, such as their velocity, distance travelled, the time to reach the edge of the arena, and time spent at the periphery. We exposed juvenile and adult worms to nicotine (acetylcholine agonist) as a proof of concept. We noted similar disruptions to locomotion at both life stages, with low doses of nicotine stimulating movement and higher doses reducing locomotion. From here, we exposed juvenile worms to fluoxetine (serotonin reuptake inhibitor), phenobarbital (GABA agonist), and apomorphine (dopamine agonist). The behaviour of juvenile worms can be altered by exposure to fluoxetine, phenobarbital, and apomorphine. Fluoxetine and phenobarbital exposure reduces movement at high doses, but fluoxetine influences the amount of time at the periphery of the arena. Apomorphine produced modest changes in locomotion compared to other chemicals tested. Genome searching, as well as transcriptomics, were used to confirm the presence of neurochemical pathways in Capitella teleta. Our results demonstrate that Capitella teleta is a viable model for behavioural work, that several neurochemical pathways contribute to locomotory behaviour, and that this assay may be useful as a screen for contaminants found in marine habitats.

Abstract:Animals are exposed to many microbes in their environment, some of which have been shown to colonize various tissues including the intestine. The composition of the intestinal microbiota affects many aspects of the host's physiology and health. Despite this, very little is known about whether host behavior contributes to the colonization. We approach this question in the nematode C. elegans , which feeds on bacteria and also harbors an intestinal microbiome. We examined the behavior of C. elegans towards CeMbio, a simplified microbiome consisting of twelve strains that represent the bacteria found in the animal's natural environment. We observed that C. elegans raised on E. coli shows a strong preference for three members of CeMbio ( Lelliottia amnigena JUb66 , Enterobacter hormaechei CEent1, and Pantoea nemavictus BIGb0393) compared to E. coli . Previously, these three bacterial strains have been shown to support faster C. elegans development time than E. coli OP50 and are low colonizers compared to eight other members of CeMbio. We then used gas chromatography coupled to mass spectrometry to identify that these three bacteria release isoamyl alcohol, a previously described C. elegans chemoattractant. We suggest that C. elegans seeks bacteria that release isoamyl alcohol and support faster growth.

Abstract:BACKGROUND: The lack of new anthelmintic agents is of growing concern because it affects human health and our food supply, as both livestock and plants are affected. Two principal factors contribute to this problem. First, nematode resistance to anthelmintic drugs is increasing worldwide and second, many effective nematicides pose environmental hazards. In this paper we address this problem by deploying a high throughput screening platform for anthelmintic drug discovery using the nematode Caenorhabditis elegans as a surrogate for infectious nematodes. This method offers the possibility of identifying new anthelmintics in a cost-effective and timely manner.METHODS/PRINCIPAL FINDINGS: Using our high throughput screening platform we have identified 14 new potential anthelmintics by screening more than 26,000 compounds from the Chembridge and Maybridge chemical libraries. Using phylogenetic profiling we identified a subset of the 14 compounds as potential anthelmintics based on the relative sensitivity of C. elegans when compared to yeast and mammalian cells in culture. We showed that a subset of these compounds might employ mechanisms distinct from currently used anthelmintics by testing diverse drug resistant strains of C. elegans. One of these newly identified compounds targets mitochondrial complex II, and we used structural analysis of the target to suggest how differential binding of this compound may account for its different effects in nematodes versus mammalian cells.CONCLUSIONS/SIGNIFICANCE: The challenge of anthelmintic drug discovery is exacerbated by several factors; including, 1) the biochemical similarity between host and parasite genomes, 2) the geographic location of parasitic nematodes and 3) the rapid development of resistance. Accordingly, an approach that can screen large compound collections rapidly is required. C. elegans as a surrogate parasite offers the ability to screen compounds rapidly and, equally importantly, with specificity, thus reducing the potential toxicity of these compounds to the host and the environment. We believe this approach will help to replenish the pipeline of potential nematicides.

Are Dieting and Dietary Inadequacy a Second Hit in the Association with Polycystic Ovary Syndrome Severity?

Abstract:Background The composition of the diet is of increasing importance for the development and maturation of the ovarian follicles. In Polycystic Ovary Syndrome (PCOS) healthy dietary interventions improve the clinical spectrum. We hypothesized that dieting and diet inadequacy in the reproductive life course is associated with impaired programming of ovarian follicles and contributes to the severity of the PCOS phenotype. Methods and Findings To determine associations between the use of a self-initiated diet and diet inadequacy and the severity of the PCOS phenotype, we performed an explorative nested case control study embedded in a periconception cohort of 1,251 patients visiting the preconception outpatient clinic. 218 patients with PCOS and 799 subfertile controls were selected from the cohort and self-administered questionnaires, anthropometric measurements and blood samples were obtained. The Preconception Dietary Risk Score (PDR score), based on the Dutch dietary guidelines, was used to determine diet inadequacy in all women. The PDR score was negatively associated to cobalamin, serum and red blood cell folate and positively to tHcy. PCOS patients (19.9%), in particular the hyperandrogenic (HA) phenotype (22.5%) reported more often the use of a self-initiated diet than controls (13.1%; p = 0.023). The use of an inadequate diet was also significantly higher in PCOS than in controls (PDR score 3.7 vs 3.5; p = 0.017) and every point increase was associated with a more than 1.3 fold higher risk of the HA phenotype (adjusted OR 1.351, 95% CI 1.09–1.68). Diet inadequacy was independently associated with the anti-Müllerian Hormone (AMH) concentration (β 0.084; p = 0.044; 95% CI 0.002 to 0.165) and free androgen index (β 0.128; p = 0.013; 95% CI 0.028 to 0.229) in PCOS patients. Conclusions The use of a self-initiated diet and diet inadequacy is associated with PCOS, in particular with the severe HA phenotype. This novel finding substantiated by the association between diet inadequacy and AMH needs further investigation.

Abstract:DEHP (Di-ethyl hexyl phthalate), an ubiquitous chemical constituent of plasticizer, cosmetics etc. poses different abnormal neurological and behavioral responses in eukaryotic model organism C. elegans (Caenorhabditis elegans). Based upon such reports, we hypothesized that due to exposure of DEHP, the larval growth of C. elegans was altered and animals went into dauer at high concentrations of exposure at 8.26 ppm. Using chemosensory assay, the orientation of repulsion (− 0.1) of the nematodes were determined with exposure of DEHP at 4.78 ppm and 8.26 ppm. We show that N2 wild type worms display a preferential food choice and search for favorable food, and alter two modes of locomotion (forward and backward) that measures the maximum bending rate of an adult worm. Thermo sensation also plays a crucial role in determining the toxicity level in C. elegans at 15 °C, 20 °C, and 25 °C. Understanding the chemosensory pattern towards a specific chemical for C. elegans, the recent work could shed light in exploring the molecular mechanism involved in the tactile movement of the worms against a specific chemical, thereby making worms a potential biological sensing platform for external stimuli perception.

Abstract:Chemotaxis in the nematode Caenorhabditis elegans has basically been examined using conventional assay methods. Although these can be problematic, for example, in their use of anesthesia, the method has never been improved. We propose a pond assay for the sensory systems (PASS) of C. elegans as a novel population-based method of behavioral analysis. The test solution is injected into a recess(es) formed on agar and the response of C. elegans to its odor and/or taste is examined. Once C. elegans individuals fall into recesses (ponds) filled with liquid, they cannot return to a solid medium. In this way, the animals are trapped with certainty without the use of anesthesia. The anesthesia used to keep animals in the attractant area in conventional chemotaxis assays is no longer required, allowing pure evaluation of the attractant or repellent response to specific substances. Furthermore, the assay itself can be greatly streamlined because the preparation can be completed simply by providing a recess(es) and filling the liquid. The present paper reports the detailed method and effectiveness of the novel PASS.

Abstract:Abstract Chemosensory neurons translate perception of external chemical cues, including odorants, tastants, and pheromones, into information that drives attraction or avoidance motor programs. In the laboratory, robust behavioral assays, coupled with powerful genetic, molecular and optical tools, have made Caenorhabditis elegans an ideal experimental system in which to dissect the contributions of individual genes and neurons to ethologically relevant chemosensory behaviors. Here, we review current knowledge of the neurons, signal transduction molecules and regulatory mechanisms that underlie the response of C. elegans to chemicals, including pheromones. The majority of identified molecules and pathways share remarkable homology with sensory mechanisms in other organisms. With the development of new tools and technologies, we anticipate that continued study of chemosensory signal transduction and processing in C. elegans will yield additional new insights into the mechanisms by which this animal is able to detect and discriminate among thousands of chemical cues with a limited sensory neuron repertoire.

Volatile Sex Pheromone Extraction and Chemoattraction Assay in Caenorhabditis elegans

Abstract:Chemical communication is vital in organismal health, reproduction, and overall well-being. Understanding the molecular pathways, neural processes, and computations governing these signals remains an active area of research. The nematode Caenorhabditis elegans provides a powerful model for studying these processes as it produces a volatile sex pheromone. This pheromone is synthesized by virgin females or sperm-depleted hermaphrodites and serves as an attractant for males. This protocol describes a detailed method for isolating the volatile sex pheromone from several C. elegans strains (WT strain N2, daf-22, and fog-2) and C. remanei. We also provide a protocol for quantifying the male chemotaxis response to the volatile sex pheromone. Our analysis utilizes measurements such as chemotaxis index (C.I.), arrival time (A.T.), and a trajectory plot to compare male responses under various conditions accurately. This method allows for robust comparisons between males of different genetic backgrounds or developmental stages. Furthermore, the experimental setup outlined here is adaptable to investigating other chemoattraction chemicals.

Abstract:Aggregation is a spatial distribution pattern where individuals can be grouped through interaction with particular signals or cues [e.g., chemical substances]. Numerous triatomine species exhibit attraction and aggregation around both conspecific and heterospecific feces. It remains unclear whether compounds released by feces function as signals (pheromones or synomones) or cues. Employing a bioassay that mimics field conditions, we investigated the response of Triatoma pallidipennis bugs to volatiles present in its feces and blends of these compounds. Our findings indicate that short-chain aldehydes, namely nonanal, octanal, heptanal, and hexanal, attract fifth-instar nymphs. Furthermore, insects responded to individual compounds as well as secondary, ternary, and quaternary blends. The most attractive blend consisted of all four aldehydes. Additionally, quaternary blends at various compound ratios attracted fifth-instar nymphs of T. phyllosoma and T. longipennis . We discuss the potential roles of these compounds as signals or cues and explore their application as bait in control programs.

Abstract:Animals can move towards or away from an odorant. Such chemotaxis has been used as a paradigm for learning when coupled with pre-exposure to the sensed odorant. Here we develop an assay for the nematode that avoids the typical use of chemical or physical immobilization when measuring the response of worms to odorants. Using two sets of rectangular arenas that are oriented such that worms in one set must move in the opposite direction to worms in the other set for the same response, we found that unfed worms show reproducible movement towards the odorants butanone and benzaldehyde, and away from the odorant nonanone. In addition to the use of opposing orientations to control for gradients of unknown cues outside the arena, we introduce a measure of dispersal to control for locomotion defects and unknown cues within the arena. Since this assay avoids the use of paralytics or physical constraints, it is useful for the analysis of graded responses to a variety of chemicals and the discovery of underlying molecular mechanisms. Using this setup, we found that pre-exposure of unfed worms to butanone to induce an association of starvation with butanone resulted in different extents of such associative learning during different trials – from no learning to learned avoidance. Given this variation in associative learning despite the artificially controlled lab setting, we speculate that in dynamic natural environments such learning might be rare and highlight the challenge in discovering evolutionarily selected mechanisms that could underlie learning in the wild.

Abstract:The understanding of force interplays between an organism and its environment is imperative in biological processes. Noticeably scarce from the study of C. elegans locomotion is the measurement of the nematode locomotion forces together with other important locomotive metrics. To bridge the current gap, we present the investigation of C. elegans muscular forces and locomotion metrics (speed, amplitude and wavelength) in one single assay. This assay uses polydimethylsiloxane (PDMS) micropillars as force sensing elements and, by variation of the pillar arrangement, introduces microstructure. To show the usefulness of the assay, twelve wild-type C. elegans sample worms were tested to obtain a total of 4665 data points. The experimental results lead to several key findings. These include: (1) maximum force is exerted when the pillar is in contact with the middle part of the worm body, (2) C. elegans locomotion forces are highly dependent on the structure of the surrounding environment, (3) the worms' undulation frequency and locomotion speed increases steadily from the narrow spacing of 'honeycomb' design to the wider spacing of 'lattice' pillar arrangement, and (4) C. elegans maintained their natural sinusoidal movement in the microstructured device, despite the existence of PDMS micropillars. The assay presented here focuses on wild type C. elegans, but the method can be easily applied to its mutants and other organisms. In addition, we also show that, by inverting the measurement device, worm locomotion behaviour can be studied in various substrate environments normally unconducive to flexible pillar fabrication. The quantitative measurements demonstrated in this work further improve the understanding of C. elegans mechanosensation and locomotion.

Abstract:Throughout history, humans have relied on plants as a source of medication, flavoring, and food. Plants synthesize large chemical libraries and release many of these compounds into the rhizosphere and atmosphere where they affect animal and microbe behavior. To survive, nematodes must have evolved the sensory capacity to distinguish plant-made small molecules (SMs) that are harmful and must be avoided from those that are beneficial and should be sought. This ability to classify chemical cues as a function of their value is fundamental to olfaction, and represents a capacity shared by many animals, including humans. Here, we present an efficient platform based on multi-well plates, liquid handling instrumentation, inexpensive optical scanners, and bespoke software that can efficiently determine the valence (attraction or repulsion) of single SMs in the model nematode, . Using this integrated hardware-wetware-software platform, we screened 90 plant SMs and identified 37 that attracted or repelled wild-type animals, but had no effect on mutants defective in chemosensory transduction. Genetic dissection indicates that for at least 10 of these SMs, response valence emerges from the integration of opposing signals, arguing that olfactory valence is often determined by integrating chemosensory signals over multiple lines of information. This study establishes that is an effective discovery engine for determining chemotaxis valence and for identifying natural products detected by the chemosensory nervous system.

Behavioral Preferences of Solenopsis Invicta Buren to Essential Oils and Active Compounds from Amiaceae Plants

Abstract:The red imported fire ant (RIFA) ( Solenopsis invicta Buren) has caused serious ecological and public safety problems in invasive areas. While extensive use of pesticides to control RIFA may negatively affect human health and environment, essential oils (EOs) of Lamiaceae possessing aromatic smell and multiple biological activities are potential alternatives of chemical pesticides. In this study, we evaluated the behavioral preferences of different castes of RIFA towards the EOs from five species of Lamiaceae plants ( Salvia miltiorrhiza , Perilla frutescens , Pogostemon cablin , Schizonepeta tenusfolia, Mentha canadensis ), and then investigated the active compounds in these EOs that trigger insect behavioral preferences by gas chromatography-electroantennographic detection (GC-EAD). The results of behavioral experiment indicated that three EOs strongly altered the behavior of RIFA. Notably, all concentrations of P. frutescens EO showed a significant repellent activity on virgin queen ants, with a repellent rate of 62.50% at the lowest concentration of 2 mu g/ml. The repellent effect of S. tenusfolia EO against small worker ants reached a very significant difference between control and treatment groups, with the highest repellent rate of 85.00% observed at 1000 mu g/ml. Save for the lowest concentration 2 mu g/ml, other doses of M. canadensis EO showed a remarkable repellent effect on small worker ants, demonstrating a positive dose -effect relationship (with repellent rates of 77.50% and 65.00% at 1000 and 8 mu g/ml, respectively). A total of 39 EAD-active compounds were detected in these EOs, and 12 of them were subsequently evaluated for behavioral preferences of RIFA. Our findings indicated that six compounds (menthone, menthol, caryophyllene oxide, pulegone, 3-methylcyclopentanol, eugenol) consistently triggered behavioral preferences in worker ants. Meanwhile, six compounds, including borneol, eugenol, phenylethyl alcohol, perilla ketone, 3-methylcyclopentanol, alpha-guaiene, were found to significantly alter the behavioral selectivity of virgin queen ants. These results may help to develop application value of selected Lamiaceae plants EOs and contribute to the exploration of new green control methods for invasive pests.

Abstract:Associative learning is a critical survival trait that promotes behavioral plasticity in response to changing environments. Chemosensation and mechanosensation are important sensory modalities that enable animals to gather information about their internal state and external environment. However, there is a limited amount of research on these two modalities. In this paper, a novel PDMS–agar hybrid microfluidic device is proposed for training and analyzing chemical–mechanical associative learning behavior in the nematode Caenorhabditis elegans. The microfluidic device consisted of a bottom agar gel layer and an upper PDMS layer. A chemical concentration gradient was generated on the agar gel layer, and the PDMS layer served to mimic mechanical stimuli. Based on this platform, C. elegans can perform chemical–mechanical associative learning behavior after training. Our findings indicated that the aversive component of training is the primary driver of the observed associative learning behavior. In addition, the results indicated that the neurotransmitter octopamine is involved in regulating this associative learning behavior via the SER-6 receptor. Thus, the microfluidic device provides a highly efficient platform for studying the associative learning behavior of C. elegans, and it may be applied in mutant screening and drug testing.

Often, the decisions underlying these behavioral programs are mediated by volatile compounds in the animal's environment, which they detect and discriminate with specialized olfactory neurons along their body. Caenorhabditis (C.) elegans senses chemical stimuli with neurons located in the head and the tail of the animal, which mediate either attractive or aversive behaviors. How conflicting stimuli are processed in animals navigating different chemical gradients is poorly understood. Here, we conceived, created, and capitalized on a novel microfluidic device to enable automated and precise stimulation of head and tail neurons, either simultaneously or sequentially, while reading out neuronal activity in sensory and interneurons using genetically encoded calcium indicators. We achieve robust and programmable chemical pulses through the modulation of inlet pressures. To evaluate the device performance, we synchronized the flow control with microscopy data acquisition and characterized the flow properties in the fabricated devices. Together, our design has the potential to provide insight into the neural circuits and behavior of C. elegans simulating the experience of natural environments.

Abstract:Pathogen avoidance behaviour has been observed across animal taxa as a vital host-microbe interaction mechanism. The nematode Caenorhabditis elegans has evolved multiple diverse mechanisms for pathogen avoidance under natural selection pressure. We summarise the current knowledge of the stimuli that trigger pathogen avoidance, including alterations in aerotaxis, intestinal bloating, and metabolites. We then survey the neural circuits involved in pathogen avoidance, transgenerational epigenetic inheritance of pathogen avoidance, signalling crosstalk between pathogen avoidance and innate immunity, and C. elegans avoidance of non- Pseudomonas bacteria. In this review, we highlight the latest advances in understanding host-microbe interactions and the gut-brain axis.

Navigational Signals for Insect and Slug Parasitic Nematodes: The Role of Ascorbate-Glutathione System and Volatiles Released by Insect-Damaged Sweet Pepper Roots

Abstract:This study of underground multitrophic communication, involving plant roots, insects, and parasitic nematodes, is an emerging field with significant implications for understanding plant-insect-nematode interactions. Our research investigated the impact of wireworm (Agriotes lineatus L. [Coleoptera: Elateridae]) infestations on the ascorbate-glutathione system in sweet pepper (Capsicum annuum L.) plants in order to study the potential role in root-exudate-mediated nematode chemotaxis. We observed that an A. lineatus infestation led to a decrease in leaf ascorbate levels and an increase in root ascorbate, with corresponding increases in the glutathione content in both roots and leaves. Additionally, a pigment analysis revealed increased carotenoid and chlorophyll levels and a shift towards a de-epoxidized state in the xanthophyll cycle. These changes suggest an individual and integrated regulatory function of photosynthetic pigments accompanied with redox modifications of the ascorbate-glutathione system that enhance plant defense. We also noted changes in the root volatile organic compound (VOC). Limonene, methyl salicylate, and benzyl salicylate decreased, whereas hexanal, neoisopulegol, nonanal, phenylethyl alcohol, m-di-tert-butylbenzene, and trans-β-ionone increased in the roots of attacked plants compared to the control group. Most notably, the VOC hexanal and amino acid exudate cysteine were tested for the chemotaxis assay. Nematode responses to chemoattractants were found to be species-specific, influenced by environmental conditions such as temperature. This study highlights the complexity of nematode chemotaxis and suggests that VOC-based biological control strategies must consider nematode foraging strategies and environmental factors. Future research should further explore these dynamics to optimize nematode management in agricultural systems.

Abstract:Neurological hazard assessment of industrial and pesticidal chemicals demands a substantial amount of time and resources. Caenorhabditis elegans is an established model organism in developmental biology and neuroscience. It presents an ideal test system with relatively fewer neurons (302 in hermaphrodites) versus higher-order species, a transparent body, short lifespan, making it easier to perform neurotoxic assessment in a time and cost-effective manner. Yet, no regulatory testing guidelines have been developed for C. elegans in the field of developmental and adult neurotoxicity. Here, we describe a set of morphological and behavioral assessment protocols to examine neurotoxicity in C. elegans with relevance to cholinergic and dopaminergic systems. We discuss the homology of human genes and associated proteins in these two signaling pathways and evaluate the morphological and behavioral endpoints of C. elegans in the context of published adverse outcome pathways of neurodegenerative diseases. We conclude that C. elegans neurotoxicity testing will not only be instrumental to eliminating mammalian testing in neurological hazard assessment but also lead to new knowledge and mechanistic validation in the adverse outcome pathway framework.
