Abstract:To investigate the effect of hawthorn polyphenols on the physicochemical properties and digestibility of corn starch, different proportions (1%, 2%, 3%, and 4% [w/w]) of hawthorn polyphenol extracts (HPEs) were mixed with corn starch, and their physicochemical properties and digestive properties were characterized by X-ray diffraction, Fourier transform infrared spectroscopy, Rapid Visco Analysis, differential scanning calorimetry, and in vitro/in vivo analysis. Results indicated that small V-type crystal starch tended to be formed in the samples, and the addition of HPEs reduced the viscosity, prolonged the gelatinization temperature of corn starch, and increased the proportion of slowly digestible starch and resistant starch of the corn starch, which accounted for 36.32% +/- 1.05% and 33.32% +/- 4.07%, respectively. Compared with the raw corn starch, the postprandial blood glucose of mice that were administered the hawthorn polyphenols decreased significantly: the blood glucose peak (30 min) decreased from 14.30 +/- 1.52 to 11.77 +/- 1.21 mmol/L. Our study might provide some basic theoretical support for the application of hawthorn polyphenols in healthy starchy food processing.

Abstract:Three different extraction methods were used to extract high-temperature water-extracted pectin (HWp), high-temperature acid-extracted pectin (HAp), and high-temperature alkali-extracted pectin (HALp) from Lycium ruthenicum. The physicochemical properties, structure, and functional properties of three different pectins were studied. The results showed that HWp and HALp can extract rhamnogalacturonan-I (RG-I) from L. ruthenicum better. Through structural feature analysis, HWp and HALp have a branched structure, and HWp has a higher degree of esterification than HAp and HALp. Zeta potential results show that HWp solution is more stable. The thermal analysis results show that the thermal stability is HALp > HAp > HWp. HWp has the highest viscosity. The inhibitory activity results showed that HWp, HAp, and HALp have a certain inhibitory effect on α-glucosidase activity. This study shows the effects of different extraction methods on the properties of L. ruthenicum pectin and aims to provide a theoretical basis for the pharmaceutical and food industries to choose more suitable pectin extraction methods.

Abstract:In this issue of Cancer, Dupont and colleagues report the results of a case-control study relating pathologic findings in benign breast biopsies to the risk breast cancer development in a cohort of women enrolled in the Breast Cancer Detection Demonstration Project (BCDDP).' This is the third large study performed in the United States to assess the relationship between benign breast disease and subsequent breast cancer risk using the same histologic classification system for cate-gorizing benign breast lesions. The first of these studies was a retrospective cohort study of women in Nashville who had one or more benign breast biopsies and for whom long-term follow-up was available.2 The second was a case-control study of a subset of women participating in the Nurses' Health Study (NHS).3 The studies performed by the BCDDP and NHS groups were undertaken in response to a proposal issued by the National Institutes of Health (NIH) to determine if the results obtained in Dupont and Page's initial study of women in Nashville could be duplicated by other pathologists in different cohorts of women.

Abstract:Hawthorn (Crataegus pinnatifida Bunge) has been highlighted as an excellent source of a variety of bioactive polymers, which has attracted increasing research interest. Pectin, as a kind of soluble dietary fiber in hawthorn, is mainly extracted by hot water extraction and ultrasonic or enzymatic hydrolysis and is then extensively used in food, pharmaceutical, and nutraceutical industries. Numerous studies have shown that hawthorn pectin and its derived oligosaccharides exhibit a wide range of biological activities, such as antioxidant activity, hypolipidemic and cholesterol-reducing effects, antimicrobial activity, and intestinal function modulatory activity. As discovered, the bioactivities of hawthorn pectin and its derived oligosaccharides were mainly contributed by structural features and chemical compositions and were highly associated with the extraction methods. Additionally, hawthorn pectin is a potential resource for the development of emulsifiers and gelling agents, food packaging films, novel foods, and traditional medicines. This review provides a comprehensive summary of current research for readers on the extraction techniques, functional characteristics, structure–activity relationship, and applications in order to provide ideas and references for the investigation and utilization of hawthorn pectin and its derived oligosaccharides. Further research and development efforts are imperative to fully explore and harness the potential of hawthorn pectin-derived oligosaccharides in the food and medicine fields.

Abstract:Hawthorn (Crataegus pinnatifida Bge.) contains various active components including polysaccharides, polyphenols, vitamin, phenolic acid etc. Polysaccharides are one of the most critical active components in hawthorn, which exhibits different biological activities such as immunomodulatory antioxidant, anti‐inflammatory, hypoglycemic, and other activities. Hence, the main purpose of this paper is optimize the aqueous two‐phase extraction (UTPE) polysaccharides from hawthorn via response surface methodology coupled genetic algorithm (RSM‐GA) and then evaluated its antioxidant activity through free radicals scavenging experiments. The results show that the optimal extraction conditions to achieve the maximum polysaccharides yield (6.42±0.08)% from hawthorn by UTPE was obtained under the mass fraction of ammonium sulfate of 11%, extraction temperature of 57 °C, liquid‐to‐solid ratio of 33 mL/g, ethanol concentration of 26%, and extraction time of 30 min, and the relative error between the experimental value and the theoretical value was 4.56%. The antioxidant capacity enhanced with the increase of hawthorn polysaccharides (HPs) concentration. However, the antioxidant activity of HPs was weaker than that of ascorbic acid. The results of this study provide a critical material basis for promoting the further research of HPs, and have certain significance for the deep processing and product development of hawthorn. Abstract:The effect of hawthorn berries ripeness on the physicochemical, structural and functional properties of hawthorn pectin (HP) and its potential in sweet cherry preservation were investigated. With the advanced ripeness of hawthorn berries, the galacturonic acid (GalA) content decreased from 59.70 mol% to 52.16 mol%, the molecular weight (Mw) reduced from 368.6 kDa to 284.3 kDa, the microstructure exhibited variable appearance from thick lamella towards porous cross-linked fragment, emulsifying activity and emulsions stability, antioxidant activities, α-amylase and pancreatic lipid inhibitory capacities significantly increased. The heated emulsion stored for 30 d presented higher creaming index and more ordered oil droplets compared to the unheated emulsion. With the extended berries ripeness, the firmness of HP gels remarkably decreased from 225.69 g to 73.39 g, while the springiness increased from 0.78 to 1.16, HP exhibited a superior inhibitory effect in water loss, browning, softening, and bacterial infection in sweet cherries preservation.
Structural characterization and antioxidant activity of pectic polysaccharides from Veronica peregrina L.

Abstract:Pectins are a class of acidic polysaccharides with complex structures. Different pectin molecules are composed of different domains, which have an important impact on their biological activity. In this study, two pectic polysaccharides (WVPP-A2b and WVPP-A3b) with molecular weights of 48.7 × 10 4 and 77.6 × 10 4 kDa, respectively, were isolated from Veronica peregrina L. Structural analysis showed that both contained homogalacturonan (HG), rhamnogalacturonan I (RG-I), and rhamnogalacturonan II (RG-II) domains with a mass ratio of 2.08:2.64:1.00 and 3.87:4.65:1:00, respectively. The RG-I domain contained an arabinogalactan II backbone and arabinans consisting of t-Araf, (1→5)-α-Araf, and (1→3,5)-α-Araf. WVPP-A3b also contained short chains consisting of the [t-α-Araf-(1→5)-α-Araf-(1→] structural unit. WVPP-A3b showed stronger ability to scavenge DPPH, hydroxyl, and ABTS radicals, which was potentially associated with its high content of galacturonic acid and presence of the HG domain. The results provide information for enhancing knowledge of the structure-activity relationship of pectic polysaccharides from V. peregrina and their potential application in the healthcare food field.

Abstract:The health-promoting activities of procyanidin extracts from hawthorn (HPCs) are closely related to their digestive behaviors, absorption, and colonic metabolism, all of which remain unknown for now and thus hinder further exploration. This study aims to explore the dynamic changes of HPCs during in vitro digestion and fermentation, as well as their Caco-2 permeability, focusing mainly on the interaction between gut microbiota and HPCs. The results showed that the digested HPC samples had characteristic absorption peaks at 280 nm, and there were absorption peaks in the stretching vibration zone, including OH and CC on the benzene ring, which suggested that procyanidins were the main components in HPCs after in vitro digestion. Meanwhile, HPCs had the highest stability in the oral phase. However, the total procyanidin content of HPCs decreased during gastrointestinal digestion, and flavan-3-ol dimers and trimers in HPCs are partially degraded into epicatechin. Uptake of epicatechin (4.07 %), procyanidin B2 (2.15 %), and procyanidin B5 (39.44 %) through Caco-2 monolayer was also observed in HPC treatment, while there was still a large portion of procyanidins that was not absorbed. Subsequent fermentation resulted in a decrease in pH along with the production of short-chain fatty acids (SCFAs), mainly due to the degradation and utilization of HPC, as indicated by a reduction of total procyanidins. Furthermore, the HPCs modulated gut microbial populations: down-regulated the abundances of Bacteroides, Fusobacterium, Enterococcus, Parabacteroides, and Bilophila, and up-regulated Escherichia-Shigella, Klebsiella, Turicibacter, Actinobacillus, Roseburia, and Blautia. Ultimately, epicatechin and procyanidin B2, B5 and C1 were converted into phenolic acids through the metabolism of Bacteroides, Sutterella, Butyrobacter and Blautia. 4-ethylbenzoic acid, 4-hydroxyphenylpropionic acid, 3,4-dihydroxyphenyl acetic acid were confirmed as the significant metabolites in the fermentation. These results elucidated the potential mechanisms of HPCs metabolism and their beneficial effects on gut microbiota and colonic phenolic acids production.
[Comparison of metabolic effects of infusions of glucose and glucose substitutes].

Abstract:The side effects of high dosed infusions of glucose and glucose substitutes (fructose, sorbitol, xylitol) in metabolically healthy volunteers have been studied. Infusions of glucose or fructose in high dose cause an increase in blood lactate levels. This effect, to a lesser degree, is observed with sorbitol. No effect, however, was seen with xylitol. Following the termination of the glucose infusions the lactate levels remain elevated, whereas after termination of the fructose infusions the lactate concentration is normalized within a short time. A decrease in the serum phosphate levels was observed for all the substances used. The most active were glucose and xylitol, followed by fructose and sorbitol, respectively. Similarly, all these substances cause an elevation of the serum bilirubin levels, though enzyme activities in serum remain unaffected. Xylitol had the greatest effect on bilirubin levels, however, glucose was also effective. The only side effect caused by the glucose substitutes was the stimulation of uric acid synthesis. In this respect xylitol was most effective. Some of metabolic effects of glucose and glucose substitutes are not yet clearly understood. However, even at very high doses these effects are not to be regarded as true "side-effects".

Abstract:The antioxidant activities of polysaccharides from the fruiting body (PFB), extracellular polysaccharides (EPS) and intracellular polysaccharides (IPS) from Agaricus bitorquis (QuéL.) Sacc. Chaidam ZJU-CDMA-12 in vitro were compared. IPS showed stronger antioxidant activities than PFB and EPS in vitro. Further purification and structure analyses indicated that IPS mainly consisted of three fractions (IPS-I, IPS-II and IPS-III). FT-IR and NMR data indicated that IPS was mainly composed of (1 → 6)-linked α-d-glucose. There are significant differences of antioxidant activities among IPS-I, IPS-II and IPS-III fractions in vitro, and IPS-III showed stronger antioxidant activity than IPS-I and IPS-II. IPS-III also possesses a potent antioxidant ability inside HepG2 cells, and it could protect HepG2 cells from H2O2-induced cytotoxicity by scavenging overproduced cellular ROS and inhibiting SOD, CAT and GSH depletion to weaken lipid peroxidation. These findings suggested that IPS-III could be a novel antioxidant and that it could afford protection against H2O2-induced cytotoxicity and oxidative stress.

Abstract:The antioxidant activity of polysaccharides has attracted tremendous research interest in recent years. In this study, we aimed to investigate the antioxidant properties of Helicteres angustifolia L. polysaccharides (HALP) that was acquired by water extraction and alcohol precipitation using ultrasound-assisted method with decolorization and protein removal. The polysaccharides were then successively purified using chromatography on DEAE-cellulose and Sephadex S-300 columns with two major fractions: HALPs1-1 and HALPs2-1. The physicochemical properties, structural characterization, and antioxidant activities of these fractions were investigated. The results indicated that HALPs1-1 was a glucan with an average molecular weight of 151.70kDa, and HALPs2-1 was composed of glucuronic acid with an average molecular weight of 114.81kDa. Infrared spectroscopies and iodine potassium iodide tests showed that the HALP were acid α-pyranoses. The antioxidant activities of the polysaccharide fractions (HALP, HALPs1-1, and HALPs2-1) were evaluated in vitro. The results suggested that HALPs2-1 had the highest scavenging activities for ABTS, hydroxyl and DPPH radicals than other polysaccharides. Taken together, the results of this study suggested that polysaccharides from Helicteres angustifolia L. could be used as novel potential antioxidants.

Abstract:Dendrobium huoshanense is a famous edible and medicinal herb, and polysaccharides are the main bioactive component in it. In this study, response surface methodology (RSM) combined with a Box–Behnken design (BBD) was used to optimize the enzyme-assisted extraction (EAE), ultrasound–microwave–assisted extraction (UMAE), and hot water extraction (HWE) conditions and obtain the polysaccharides named DHP-E, DHP-UM, and DHP-H. The effects of different extraction methods on the physicochemical properties, structure characteristics, and bioactivity of polysaccharides were compared. The differential thermogravimetric curves indicated that DHP-E showed a broader temperature range during thermal degradation compared with DHP-UM and DHP-H. The SEM results showed that DHP-E displayed an irregular granular structure, but DHP-UM and DHP-H were sponge-like. The results of absolute molecular weight indicated that polysaccharides with higher molecular weight detected in DHP-H and DHP-UM did not appear in DHP-E due to enzymatic degradation. The monosaccharide composition showed that DHPs were all composed of Man, Glc, and Gal but with different proportions. Finally, the glycosidic bond types, which have a significant effect on bioactivity, were decoded with methylation analysis. The results showed that DHPs contained four glycosidic bond types, including Glcp-(1→, →4)-Manp-(1→, →4)-Glcp-(1→, and →4,6)-Manp-(1→ with different ratios. Furthermore, DHP-E exhibited better DPPH and ABTS radical scavenging activities. These findings could provide scientific foundations for selecting appropriate extraction methods to obtain desired bioactivities for applications in the pharmaceutical and functional food industries.

Abstract:Polysaccharides are biological macromolecules that are difficult to absorb into intestinal epithelial cells for exerting activities, whereas the interaction between polysaccharides and gut microbiota might be an alternative method. This study aimed to explore the in vitro digestion of hawthorn polysaccharides (HPS) and their interaction with the gut microbiota. Results showed that the content of reducing sugars increased slightly during gastric digestion. However, no free monosaccharide was detected during the whole simulated digestion process, indicating that HPS was indigestible. The total carbohydrate residue decreased during in vitro fermentation. This result was due to the utilization by the gut microbiota. Meanwhile, short-chain fatty acids were produced due to the utilization of HPS. Notably, HPS could significantly modulate the composition of human gut microbiota; in particular, the relative abundances of Megasphaera, Acidaminococcus and Mitsuokella increased, whereas the relative abundances of Escherichia Shigella and Fusobacterium decreased. It was suggested that HPS could decrease the abundances of harmful intestinal microbiota and regulate the proportion of beneficial bacteria in the intestinal tract. Overall, the beneficial effects of HPS were believed to be related to the gut microbiota and could be used as a potential dietary supplement.

Abstract:Different ratios of hesperetin (HT) were successfully grafted onto pectin from basic water (PB) molecules via free radical-induced reaction. The structure of PB-HT conjugates was characterized by ultraviolet spectroscopy, infrared spectroscopy, X-ray diffraction and scanning electron microscopy. Results indicated that HT was successfully grafted onto pectin molecules, and PB-HT-0.5 showed the highest HT content (103.18 ± 2.76 mg/g). Thermogravimetric analysis indicated that HT crystals showed good thermal resistance and could improve the thermal stability of PB-HT conjugates. Additionally, PB-HT conjugates showed good cytocompatibility and blood compatibility. This study provides a novel and efficient method to synthesize hesperetin-grafted pectin conjugate, which showed potential application in the fields of functional foods in the future.

Abstract:This study performed a comprehensive investigation of Hyperici Perforati Herba polysaccharide (HPHP) regarding the development and optimization of extraction methods, elucidation of structure and characteristics, and determination of antioxidant activities. An ultrasonic-assisted extraction method, which offered advantages in terms of the extraction yield and energy efficiency, was developed by response surface analysis. The following optimum conditions were determined: a crushing degree at 65 mesh, ultrasonic time at 50 min and temperature of 43 °C. Through enzyme-mediated deproteination via the Sevag method, activated carbon depigmentation, and DEAE-52 and Sephadex G-100 column elution, three HPHPs were obtained, and their monosaccharides mainly included mannose, galactose, glucose and arabinose. The molar weights were 8.347, 1.199 and 22.426 kDa, respectively. The HPHP structures were an amorphous aggregate of spherical-like shapes with a rough surface of pores and crevices, which presented characteristic Fourier transform infrared (FT-IR) and nuclear magnetic resonance (NMR) spectra of polysaccharides. Their main glucosidic linkage is the α-type configuration. Moreover, HPHPs exhibited strong scavenging activity for DPPH·, ABTS·+, OH· and O2·− radicals; good ferric reducing power; and effective protection against oxidative damage in human cells. Overall, the results of this work underpinned a fundamental understanding of HPHPs, thus providing a potential antioxidant for further research and development.
Comparative study of HG-type low-ester hawthorn pectin as a promising material for the preparation of hydrogel

Abstract:Homogalacturonan (HG)-type pectin has been considered suitable for the formation of hydrogel, but it is unknown whether the less side chain will limit the properties of hydrogel. The current study successfully obtained low methoxyl HG-type hawthorn pectin (LMHP) from hawthorn by sequential hot water extraction and alkaline de-esterification. The proportion of HG domain, the proportion of rhamnogalacturonan-I (RG-I) domain, molecular weight, and particle size of LMHP were 81.13 %, 4.04 %, 348.43 kDa, and 1386.92 nm, respectively. Compared with commercial citrus pectin (CP), LMHP was more suitable for the preparation of hydrogel. The hydrogel with the densest network structure and relatively stable performance in digestion simulation environment can be obtained as the concentration of LMHP increases to 2.5 %, which is lower than the consumption of commercial citrus pectin. Overall, these findings highlight the potential of hawthorn pectin as a novel hydrogel raw material.

Simultaneously efficient dissolution and structural modification of chrysanthemum pectin: Targeting at proliferation of Bacteroides

Abstract:Pectic polysaccharide is a bioactive ingredient in Chrysanthemum morifolium Ramat. 'Hangbaiju' (CMH), but the high proportion of HG domain limited its use as a prebiotic. In this study, hot water, cellulase-assisted, medium-temperature alkali, and deep eutectic solvent extraction strategies were firstly used to extract pectin from CMH (CMHP). CMHP obtained by cellulase-assisted extraction had high purity and strong ability to promote the proliferation of Bacteroides and mixed probiotics. However, 4 extraction strategies led to general high proportion of HG domain in CMHPs. To further enhance the dissolution and prebiotic potential of CMHP, pectinase was used alone and combined with cellulase. The key factor for the optimal extraction was enzymolysis by cellulase and pectinase in a mass ratio of 3:1 at 1 % (w/w) dosage. The optimal CMHP had high yield (15.15 %), high content of total sugar, and Bacteroides proliferative activity superior to inulin, which was probably due to the cooperation of complex enzyme on the destruction of cell wall and pectin structural modification for raised RG-I domain (80.30 %) with relatively high degree of branching and moderate HG domain. This study provided a green strategy for extraction of RG-I enriched prebiotic pectin from plants.

Abstract:Ethnopharmacologic relevance Crataegus pinnatifida , commonly known as hawthorn, is a plant species with a long history of medicinal use in traditional Chinese medicine. Hawthorn polysaccharides (HP) have gained worldwide attention due to their decent biological activities and potential health benefits. Their excellent antioxidant activity, antitumor activity, immunomodulatory activity, hypoglycemic effect and hypolipidemic effects, intestinal microbiota modulatory activity makes them valuable in the field of ethnopharmacological research. Aim of the study The purpose of the current review is to provide a systematic and comprehensive summary of the latest literatures and put forward the future perspectives on hawthorn polysaccharides in the context of its extraction, purification, structural characteristics and bioactivities. Furthermore, the underlying structure-bioactivity relationship of hawthorn polysaccharides was also explored and discussed. The current review would provide the important research underpinnings and the update the information for future development and application of hawthorn polysaccharides in the pharmaceutical and functional food industries. Materials and methods We use Google Scholar, CNKI, PubMed, Springer, Elsevier, Wiley, Web of Science and other online databases to search and obtain the literature on extraction, isolation, structural analysis and the biological activity of hawthorn polysaccharides published before October 2023. The key words are "extraction", "isolation and purification", "bioactivities", and " Crataegus pinnatifida polysaccharides ". Results Crataegus pinnatifida has been widely used for the treatment of cardiovascular diseases, digestive disorders, inflammatory and oxidative stress in traditional Chinese medicine. Polysaccharides are the key active components of Crataegus pinnatifida which have gained widespread attention. The structure and bioactivity of polysaccharides from Crataegus pinnatifida varies in terms of raw materials, extraction methods and purification techniques. Crataegus pinnatifida polysaccharides possess diverse bioactivities, including antitumor, immunomodulatory, hypoglycemic activity, cardioprotective and antioxidant activities, among others. These biological properties can not only lay firm foundation for the treatment of diverse diseases, but also provide a theoretical basis for the in-depth study of the structure-activity relationship. In addition, the underlying structure-activity relationship is also explored and discussed, and further research and development of hawthorn polysaccharides are also prospected. Conclusion As a natural compound, hawthorn polysaccharides has garnered significant attention and held immense research potential. Hawthorn polysaccharides can be obtained through different extraction methods, including hot water extraction method, ultrasonic extraction method and enzymatic extraction method etc. The structures of hawthorn polysaccharides have also been characterized and reported in numerous studies. Moreover, hawthorn polysaccharides exhibit a wide range of bioactivities, such as the antioxidant activity, the antitumor activity, the immunomodulatory activity, the hypoglycemic effect and the hypolipidemic effect, as well as the intestinal microbiota modulatory activity. These diverse bioactivities contribute to the growing interest in hawthorn polysaccharides and its potential applications. Hawthorn polysaccharides has promising application prospects in various industries, including functional food, pharmaceuticals and biomedical research. Therefore, it is imperative to fully explore and harness the potential of hawthorn polysaccharides in the food and medicine fields.
