Abstract:Nuclear factor 90 (NF90) is a member of an expanding family of double-stranded (ds) RNA-binding proteins thought to be involved in gene expression. Originally identified in complex with nuclear factor 45 (NF45) as a sequence-specific DNA-binding protein, NF90 contains two double stranded RNA-binding motifs (dsRBMs) and interacts with highly structured RNAs as well as the dsRNA-activated protein kinase, PKR. In this report, we characterize the biochemical interactions between these two dsRBM containing proteins. NF90 binds to PKR through two independent mechanisms: an RNA-independent interaction occurs between the N terminus of NF90 and the C-terminal region of PKR, and an RNA-dependent interaction is mediated by the dsRBMs of the two proteins. Co-immunoprecipitation analysis demonstrates that NF90, NF45, and PKR form a complex in both nuclear and cytosolic extracts, and both proteins serve as substrates for PKR in vitro. NF90 is phosphorylated by PKR in its RNA-binding domain, and this reaction is partially blocked by the NF90 N-terminal region. The C-terminal region also inhibits PKR function, probably through competitive binding to dsRNA. A model for NF90-PKR interactions is proposed.
Disinsertion of the inferior oblique muscle for treatment of superior oblique paresis.

Abstract:PURPOSE To examine the safety and efficacy of disinsertion of the inferior oblique muscle in the treatment of long-standing ipsilateral superior oblique paresis. METHODS This retrospective study included 52 patients who underwent disinsertion of the inferior oblique muscle over a 5-year period. Mean age at presentation was 30.8 years (range: 1-70 years). All patients had long-standing unilateral superior oblique paresis and overaction of the ipsilateral inferior oblique muscle. Information recorded included pre- and postoperative vertical deviation, pre- and postoperative Hess chart error scores, reoperation rate, and symptom relief. RESULTS Following surgery, mean vertical deviation was reduced from 12.9 to 4 prism diopters, while mean Hess chart error score decreased from 596 to 258. This procedure alone resulted in satisfactory symptom relief in 84.6% of patients. The only complication was significant undercorrection requiring further surgery in 13.6% of patients and prisms in 1.8%. When further surgery was needed, the procedure of choice was tucking of the affected superior oblique. Recession of the contralateral inferior rectus was carried out when superior oblique tucking was believed to be unsuitable. CONCLUSIONS Disinsertion of the inferior oblique muscle is a safe and effective treatment for the majority of long-standing idiopathic unilateral superior oblique pareses.
A Molecular Imprinting Electrochemical Sensor for L-Tryptophan Determination

Abstract:A molecular imprinting electrochemical sensor for the determination of L-tryptophan was developed.The molecular imprinting polymer(MIP) was fabricated by the polymerization reaction of ethyl silicate(TEOS),phenyltrimethoxysilane(PTMOS) and phenyltrimethoxysilane(MTMOS).The presence and removal of the template molecule of L-tryptophan in the MIP film were confirmed by cyclic voltammetry.The sensor showed an excellent selectivity and a high sensitivity for L-tryptophan detection,with a linear range from 1.0×10-9 to 1.0×10-5 mol/L(r=0.9958) and a limit of detection as low as 1.0×10-12 mol/L.The sensor was successfully applied to the determination L-tryptophan in the serum samples and the recoveries were in the range of 80.0%~104%.

Abstract:A new tryptophan detection sensor utilizing molecularly imprinted ferrocene‐functionalized carbon nanotubes has been rationally designed. It features a broad detection range from 1 μM to 45 μM and from 45 μM to 330 μM, with an ultra‐low detection limit of 0.44 μM for tryptophan. This sensor has successfully detected tryptophan in real samples, including human serum and amino acid oral liquid, yielding excellent results. Molecular imprinting is an advanced technology in the fabrication of novel chemical sensors recently applied, enabling rapid responses and highly selective binding to specific molecules, thereby enabling efficient detection of material constituents. In this study, a new molecularly imprinted electrochemical sensor (MIES) utilizing ferrocene‐functionalized carbon‐nanotubes (Fc‐CNTs) was developed for the determination of tryptophan (Trp). The formation of molecularly imprinted polymer (MIP) involves polymerization in dimethyl sulfoxide (DMSO) using Trp, methacrylic acid (MAA) and ethylene glycol dimethacrylate (EGDMA) as the template molecules, functional monomers, and cross‐linking agents, respectively. Due to the modification of MIPs, the sensor achieves high performance. The sensor demonstrates a linear range from 1 to 45 μM and from 45 to 330 μM, with the limit of detection of 0.44 μM, exceeding that of the majority of sensors reported in the literature. Furthermore, the sensor can effectively detect Trp in real samples, including human serum and amino acid oral liquid. The recovery rate of Trp in human serum samples is 94.2 % to 105.30 %, and in amino acid oral liquid is 94.3 % to 101.68 %. This study provides a promising approach for the effective detection and analysis of tryptophan constituents.

Abstract:In this paper, a tryptophan (Trp) molecularly imprinted chitosan film was prepared on the surface of an acetylene black paste electrode using chitosan as the functional polymer, Trp as the template molecule and sulfuric acid as the crosslinking agent. The surface morphologies of non-imprinted and imprinted electrodes were characterized by scanning electron microscopy (SEM). The formation of hydrogen bonds between the functional polymer and the template molecule was confirmed by infrared spectroscopy. Some factors affecting the performance of the imprinted electrode such as the concentration of chitosan, the mass ratio of chitosan to Trp, the dropping amount of the chitosan-Trp mixture, the solution pH, and the accumulation potential and time were discussed. The experimental results show that the imprinted electrode exhibit good affinity and selectivity for Trp. The dynamic linear ranges of 0.01–4 M, 4–20 M and 20–100 M were obtained by second derivative linear sweep voltammetry, and the detection limit was calculated to be 8.0 nM. The use of the imprinted electrode provides an effective method for eliminating the interference of potentially interfering substances. In addition, the sensor has high sensitivity, reproducibility and stability, and can be used for the determination of Trp in pharmaceutical preparations and human serum samples.

Abstract:L-tryptophan (Trp) is an essential amino acid for humans and plays crucial roles in many metabolic functions. Trp levels can be used for diagnosing different kinds of metabolic disorders and the symptoms associated with those diseases. Herein, a novel, simple and sensitive sensor based on 3D peony-like bimetallic conductive MOFs (Co–Ni-MOFs) was fabricated for the electrochemical determination of Trp. The bimetallic conductive MOFs were synthesized by a facile one-pot hydrothermal process. On account of the synergy between the Ni2+ and Co2+ ions, the bimetallic Co–Ni-MOFs showed excellent electrochemical performance, including good conductivity, large effective surface areas, and high electrocatalytic reactivity toward the oxidation of Trp. Consequently, the Co–Ni-MOFs-modified electrodes obtained a wide linear range from 10 nmol L−1 to 300 μmol L−1 and a low detection limit of 8.7 nmol L−1 (S/N = 3) for Trp. Additionally, the prepared sensor also displayed high selectivity, long-term stability and reproducibility. Moreover, the proposed sensor was successfully applied to determine the levels of Trp in the plasma of mice after cadmium intoxication.

Abstract:Although electrochemical detection based on molecular imprinting polymers (MIP) could dramatically improve the selectivity, the procedure is time-consuming because of the essential incubation step. In addition, current MIP electrochemical detections were not suitable for analysis of microliter-level sample solutions, limiting their applications for real samples. This investigation aims at applying vibration to enhance efficiency of MIP electrochemical detection of 20 μL sample solutions. MIP analysis of Tryptophan (Trp) was used as the model with disposable MIP electrodes prepared by electrochemical polymerization of o-phenylenediamine on carbon ink coated on stainless steel sheets. The MIP electrode was integrated in a 3D-printed analytical device for vibration-enhanced electrochemical detection of Trp. Our results showed that this vibration-enhanced strategy could significantly increase electrochemical responses of Trp at the same incubation time . Such improvement might be attributed to the enhanced mass transfer at the surface of the working electrode brought by vibration. It needs to be emphasized that this strategy is suitable for analysis of sample solutions with the volume of microliters, which is superior to normal stirring in MIP electrochemical detection. Our approach could be successfully utilized for differentiation of Trp in different fruits, opening more opportunities for MIP electrochemical detection of real samples. The enhanced efficiency by vibration could pave foundation for extensive practical MIP detection of sample solutions at the level of microliters.

Abstract:An ordered mesoporous silica (OMS) was synthesized through hydrothermal process. The synthesized material was characterized by different techniques such as X-ray diffraction, nitrogen adsorption. desorption and transmission electron microscope. The electrochemical characteristics of the OMS modified carbon paste electrode (OMS/CPE) were investigated by using cyclic voltammetry and electrochemical impedance spectroscopy. Compared with bare carbon paste electrode (CPE), the OMS/CPE exhibited a larger electrode surface and a faster electron. transfer rate. Electrochemical behaviors and kinetic properties of L-tryptophan (L-Trp) at the OMS/CPE in the presence of sodium dodecylsulphate (SDS) were studied. The results showed that the voltammetric response of L-Trp at OMS/CPE was improved due to the synergistic effect of OMS and SDS. The electrochemical oxidation of L-Trp at OMS/CPE in the presence of SDS was an irreversible process involved two electrons and two protons, and the electrode process was controlled by the adsorption step. Some parameters such as SDS concentration, accumulation time, accumulation potential and pH were optimized. Under optimal conditions, the oxidation peak current was linearly proportional to L-Trp concentration in the range of 8.0x10(-8) to 4.0x10(-6) mol/L,with a detection limit of 7.0x10(-8) mol/L(S/N=3). The proposed method was applied for the determination of L-Trp in oral liquid with the recoveries of 99.6%-102.6%.

Abstract:Tetracycline (TC) residues have a serious impact on human health, sustainable development of animal husbandry and aquaculture. Herein, a novel sensor (Cr-MOF@MIP) for TC detection was developed by modifying molecularly imprinted polymers (MIPs) on the surface of a luminescent metal-organic framework (MOFs). The sensor not only possesses excellent fluorescence stability and specific recognition sites, but also enables selective aggregation of the target. Subsequently, the physicochemical properties of Cr-MOF@MIP, such as morphological structure, crystal shape, surface area, pore size and functional groups, were characterized. Under the optimized conditions, the relative fluorescence intensity of the Cr-MOF@MIP fluorescent sensor decreased gradually and linearly as the TC concentration increased from 0.2 μg/mL to 50 μg/mL. The limit of detection (LOD) of TC was 0.032 μg/mL. In addition, the sensor has higher selectivity for TC compared to non-imprinted polymers (NIPs), with a bursting mechanism of Cr-MOF@MIP mainly attributed to the fluorescence resonance energy transfer. Remarkably, in the complex systems of milk, its specific detection of TC was demonstrated with satisfactory recoveries (90.06–104.34 %). It is expected that the development of Cr-MOF@MIP sensor will provide a new way of thinking for easy, rapid, highly selective and cost-effective analysis in complex samples.

Abstract:Quantitative detection of veterinary drug residues in animal-derived food is of great significance. In this work, a simple and label-free electrochemical aptasensor for the highly sensitive detection of chloramphenicol (CAP) in milk was successfully developed based on a new biosensing method, where the single- or few-layer Ti3C2 MXene nanosheets functionalized via the specific aptamer by self-assembly were used as electrode modifiers for a glassy carbon electrode (aptamer/Ti3C2 MXene/GCE). Differential pulse voltammetry (DPV), electrochemical impedance spectroscopy (EIS), cyclic voltammetry (CV), scanning electron microscopy (SEM), atomic force microscope (AFM), and so on were utilized for electrochemical and morphological characterization. Under the optimized conditions, the constructed aptasensor exhibited excellent performance with a wider linearity to CAP in the range from 10 fM to 1 μM and a low detection limit of 1 fM. Aptamer/Ti3C2 MXene/GCE demonstrated remarkable selectivity over other potentially interfering antibiotics, as well as exceptional reproducibility and stability. In addition, the aptasensor was successfully applied to determine CAP in milk with acceptable recovery values of 96.13% to 108.15% and relative standard deviations below 9%. Therefore, the proposed electrochemical aptasensor is an excellent alternative for determining CAP in food samples.

Abstract:Developing sensitive and effective methods to monitor oxytetracycline residues in food is of great significance for maintaining public health. Herein, a fluorescent sensor (NH2-UIO-66 (Zr)@MIP) based on a molecularly imprinted polymer-coated amino-functionalized zirconium (IV) metal–organic framework was successfully constructed and first used for the ultrasensitive determination of oxytetracycline. NH2-UIO-66 (Zr), with a maximum emission wavelength of 455 nm under 350 nm excitation, was prepared using a microwave-assisted heating method. The NH2-UIO-66 (Zr)@MIP sensor with specific recognition sites for oxytetracycline was then acquired by modifying a molecularly imprinted polymer on the surface of NH2-UIO-66 (Zr). The introduction of NH2-UIO-66 (Zr) as both a signal tag and supporter can strengthen the sensitivity of the fluorescence sensor. Thanks to the combination of the unique characteristics of the molecularly imprinted polymer and NH2-UIO-66 (Zr), the prepared sensor not only exhibited a sensitive fluorescence response, specific identification capabilities and a high selectivity for oxytetracycline, but also showed good fluorescence stability, satisfactory precision and reproducibility. The fabricated sensor displayed a fluorescent linear quenching in the OTC concentration range of 0.05–40 μg mL−1, with a detection limit of 0.012 μg mL−1. More importantly, the fluorescence sensor was finally applied for the detection of oxytetracycline in milk, and the results were comparable to those obtained using the HPLC approach. Hence, the NH2-UIO-66 (Zr)@MIP sensor possesses great application potential for the accurate evaluation of trace oxytetracycline in dairy products.

Abstract:Ciprofloxacin hydrochloride is widely used to treat animal diseases. However, its chronic abuse has caused severe accumulation in animal-derived foods, which has seriously damaged human health. Thus, in this study, a novel molecularly imprinted electrochemical sensor (MIES) was designed for selective and ultra-sensitive detection of ciprofloxacin hydrochloride (CPFX) in milk. The sensor was based on the molecularly imprinted polymers (MIPs) synthesized with poly-pyrrole (PPy) as the functional monomer, AuNPs and S-CoFe-MOFs (CoFe-Metal organic frameworks coated with chitosan) as the Supporting materials, and CPFX as the template molecule. The electrochemical behavior of MIPs/S-CoFe-MOFs/AuNPs/GCE was studied by several electrochemical methods, which showed a wider detection linear range (0.05 - 500 ng & BULL;mL-1) and lower detection limit (0.0033 ng & BULL;mL-1) than those in previous reports. The optimization of experiments confirmed the good selectivity and stability of the sensors--- with the milk sample recovery of 89.04-110.26%, and the relative standard deviations (RSD) of less than 5.5%, which was close to HPLC. In conclusion, the sensor we constructed has a high selectivity and stability potential in detecting CPFX in milk.
Integrated billing mechanisms in the Internet of Things to support information sharing and enable new business opportunities

Abstract:The Internet of Things (IOT) concept and enabling technologies such as RFID offer the prospect of linking the real world of physical objects with the virtual world of information technology to improve visibility and traceability information within supply chains and across the entire lifecycles of products, as well as enabling more intuitive interactions and greater automation possibilities. There is a huge potential for savings through process optimization and profit generation within the IOT, but the sharing of financial benefits across companies remains an unsolved issue. Existing approaches towards sharing of costs and benefits have failed to scale so far. The integration of payment solutions into the IOT architecture could solve this problem. We have reviewed different possible levels of integration. Multiple payment solutions have been researched. Finally we have developed a model that meets the requirements of the IOT in relation to openness and scalability. It supports both hardware-centric and software-centric approaches to integration of payment solutions with the IOT. Different requirements concerning payment solutions within the IOT have been defined and considered in the proposed model. Possible solution providers include telcos, e-payment service providers and new players such as banks and standardization bodies. The proposed model of integrating the Internet of Things with payment solutions will lower the barrier to invoicing for the more granular visibility information generated using the IOT. Thus, it has the potential to enable recovery of the necessary investments in IOT infrastructure and accelerate adoption of the IOT, especially for projects that are only viable when multiple benefits throughout the supply chain need to be accumulated in order to achieve a Return on Investment (ROI). Abstract:Tetracycline residues in animal-derived food pose serious harm to human health, making it demanded to develop simple and sensitive method for detecting tetracycline. Herein, a dual-emission synchronous response fluorescence probe is reported based on amino-functionalized Al-MOF modulated TpTt-COF (donate as NMT). NMT exhibits excellent dual-emission at 455 and 575 nm under single excitation. Tetracycline is sensitively detected through quenching the dual-emission of NMT. NMT specifically recognizes tetracycline through intermolecular hydrogen bonding between -OH/-NH2 of tetracycline and deprotonated O-/-NH-/CN of NMT. A calibration curve is built between the fluorescence ratio and the tetracycline concentration with a detection limit of 0.014 μmol/L. NMT is employed to detect tetracycline in milk, pork and chicken, and displays satisfactory recoveries of 94.39-105.67%, respectively. The optosmart sensor based on NMT and smartphone has been constructed for visually detecting tetracycline. This method provides routes to construct MOF-COF fluorescence probes and has good prospects in food analysis.

Abstract:Molecularly imprinted polymer (MIP)-based optosensing materials capable of direct, reliable, and highly selective detection of small organic analytes in complex aqueous samples hold great promise in many bioanalytical applications, but their development remains a challenging task. Addressing this issue, well-defined hydrophilic "turn-on"-type ratiometric fluorescent MIP microspheres are developed via a versatile and modular strategy based on the controlled/"living" radical polymerization method. Its general principle was demonstrated by the synthesis of red CdTe quantum dot (QD)-labeled silica particles with surface-bound atom transfer radical polymerization (ATRP)-initiating groups via the one-pot sol-gel reaction and their successive grafting of a thin fluorescent 2,4-D (an organic herbicide)-MIP layer (labeled with green organic fluorophores bearing both nitrobenzoxadiazole (NBD) and urea interacting groups) and hydrophilic poly(glyceryl monomethacrylate) (PGMMA) brushes via surface-initiated ATRP. The introduction of PGMMA brushes and rationally selected dual fluorescence labeling (i.e., red CdTe QDs being inert to 2,4-D and green NBD showing fluorescence "light-up" upon binding 2,4-D) onto MIP particles afforded them excellent complex aqueous sample-compatibility (due to their largely enhanced hydrophilicity) and analyte binding-induced "turn-on"-type ratiometric fluorescence changes, respectively. Such advanced MIP particles proved to be promising optosensing materials, which had a detection limit of 0.13 μM and showed obvious fluorescent color change upon binding different concentrations of 2,4-D in the undiluted pure milk. Moreover, they were successfully applied for direct and highly selective quantification of 2,4-D in the undiluted pure goat and bovine milks with good recoveries (97.9%-104.5%), even in the presence of several analogues of 2,4-D.

Abstract:L-tryptophan is an amino acid that significantly impacts metabolic activity in both humans and herbivorous animals. It is also known as a precursor for melatonin and serotonin, and its levels must be regulated in the human body. Therefore, there is a need to develop a cost-effective, simple, sensitive, and selective method for detecting L-tryptophan. Herein, we report the fabrication of an L-tryptophan sensor using a nickel-doped tungsten oxide ceramic-modified electrode. The Ni-WO3 was synthesized using simple strategies and characterized by various advanced techniques such as powder X-ray diffraction, scanning electron microscopy, energy-dispersive X-ray spectroscopy, and photoelectron X-ray spectroscopy. Furthermore, a glassy carbon electrode was modified with the synthesized Ni-WO3 and explored as the L-tryptophan (L-TRP) sensor. Cyclic voltammetry and differential pulse voltammetry were used to investigate the sensing ability of the modified electrode (Ni-WO3/GC). The Ni-WO3/GC exhibited an excellent limit of detection of 0.4 μM with a good dynamic linear range. The Ni-WO3/GC also demonstrated excellent selectivity in the presence of various electroactive molecules. The Ni-WO3/GC also showed decent reproducibility, repeatability, stability, and storage stability. This work proposes the fabrication of novel Ni-WO3/GC for the sensing of L-tryptophan. So far, no report is available on the use of Ni-WO3/GC for the sensing of L-TRP. This is the first report on the use of Ni-WO3/GC for the sensing of L-TRP sensing applications.

Preparation of L -Tryptophan Electrochemical Sensor Based on Graphene Oxide/Carbon Nanotubes Nanocomposite Modified Electrode

Abstract:A single-layer graphene oxide (GO) was prepared by Hummers method, and then mixed with multiwalled carbon nanotubes (MWCNT) in ultrasound bath to form stable GO/MWCNT nanocomposites. A novel L-tryptophan (L-Trp) electrochemical sensor was fabricated based on GO/MWCNT modified glassy carbon electrode. The modified electrode was characterized by transmission electron microscope (TEM), cyclic voltammetry (CV) and alternating current impedance (EIS), and the electrochemical behaviors and kinetic properties of L-Trp on the modified electrode were also investigated. The experimental results showed that a sensitive oxidation peak of L-Trp appeared at GO/MWCNT modified electrode (E-pa = 0.956 V), and the oxidation reaction was an irreversible process containing two electrons and two protons transfer. This electrode process controlled by the adsorption step, and the standard rate constant was 9.613. x 10(-4) cm/s. Trace levels of L-Trp can be determined by this oxidation peak. In phosphate buffer of pH 6.0, the oxidation peak currents were linearly dependent on the L-Trp concentrations in the range of 1.00 x 10(-6) - 1.00 x 10(-4) mol/L with accumulation time of 25 s at 0.600 V and scan rate of 100 mV/s. The correlation coefficient was 0.995 and the detection limit was 3.50 x 10(-7) mol/L. The prepared electrochemical sensor had favorable stability and could be applied to the quick determination of L. Trp in human serum, and the recovery of standard addition was from 97.8% to 104.2%.
