Abstract:The detection of sweetness is of great significance due to flavor and health concerns. In this study, an in vivo bioelectronic tongue for sweetness detection is constructed using rat gustatory system and implantable microelectrode array. By recording extracellular potentials in “sweet hotspot” within rat gustatory cortex, sweetener-evoked response patterns are generated by neural decoding algorithms. The results show that sweetener stimulation can cause a significant increase in the power spectral density of the local field potentials signal at 50-80 Hz, and its dynamic response in the time domain can be used for identification and classification of sweetness. Meanwhile, quantitative study shows that power of response signal is dose-dependent and the detection range covers sugar contents of daily beverages. Comparisons with results of human taste panelists also confirm that this in vivo bioelectronic tongue can work across species and has the potential to mimic human taste and evaluate sweetness in practical applications.
Biosensor analysis of natural and artificial sweeteners in intact taste epithelium.

Abstract:Sweeteners are commonly used as food additives in our daily life, which, however, have been causing a number of undesirable diseases since the last century. Therefore, the detection and quantification of sweeteners are of great value for food safety. In this study, we used a taste biosensor to measure and analyze different sweeteners, both natural and artificial sweeteners included. Electrophysiological activities from taste epithelium were detected by the multi-channel biosensors and analyzed with spatiotemporal methods. The longtime signal result showed different temporal-frequency properties with stimulations of individual sweeteners such as glucose, sucrose, saccharin, and cyclamate, while the multi-channel results in our study revealed the spatial expression of taste epithelium to sweet stimuli. Furthermore, in the analysis of sweetener with different concentrations, the result showed obvious dose-dependent increases in signal responses of the taste epithelium, which indicated promising applications in sweetness evaluation. Besides, the mixture experiment of two natural sweeteners with a similar functional unit (glucose and sucrose) presented two signal patterns, which turned out to be similar with responses of each individual stimulus involved. The biosensor analysis of common sweeteners provided new approaches for both natural and artificial sweeteners evaluation.

Abstract:Taste receptor cells are the taste sensation elements expressing sour, salty, sweet, bitter and umami receptors, respectively. There are cell-to-cell communications between different types of cells. Nevertheless, the mechanism of taste sensation and taste information coded by taste receptor cell is not well understood at present and it is a long-standing issue. In order to explore taste sensation and analyze taste-firing responses from another point of view, we present a promising biomimetic taste receptor cell-based biosensor. The temporal firing responses to different tastants are recorded. Meanwhile, we investigate the firing rate and temporal firing of taste receptor cells. The experimental results are consistent with that from patch clamp and molecular biology experiment. Firing rate is dependent on the concentration of stimulus. PCA analysis (principal component analysis) of the temporal firing responses shows that the responses from different types of taste receptor cells can be distinguished. Furthermore, exogenous ATP is applied to mimic the effects of transmitter ATP (adenosine triphosphate) released from type II cells onto type III cells. Both enhanced and inhibitory effects on spontaneous firing are observed. This novel biomimetic hybrid biosensor provides a potential solution to investigate the taste sensation and coding mechanisms in a non-invasive way.
A Biomimetic Taste Biosensor Based on Bitter Receptors Synthesized and Purified on Chip from a Cell-Free Expression System

Abstract:The conventional approaches for the preparation of receptor proteins towards chemical sensing are usually based on the expression of receptors in a heterologous cell system, which suffers from time-consuming, low efficiency, and complicated purification process. To address these limitations, in this study, a bitter receptor, T2R4, was synthesized and purified on chip from a cell-free protein expression system, which can be realized within a few hours. The bitter receptors were utilized as sensitive elements of a chemical biosensor for the detection of specific bitter substances. Quartz crystal microbalance (QCM) devices were used as transducers to monitor the process of sensor preparation and responses to different bitter substances by recoding the crystal resonant frequency shifts. The obtained results show that the dose-response curves of denatonium and crystal resonant frequency shifts are established in a wide linear range of 10 nM to 0.1 mM with a lower detection limit of 5 nM, which is superior to previous reports. Nearly 3 times of enhancement in sensitivity were achieved, which is calculated to be 4.96 kHz/mM. Such on-chip rapid synthesis and purification of receptors serve to achieve increased coupling efficiency and improved sensing performances. The combination of simple and rapid cell-free expression technique provides a very promising approach for the rapid and high-efficient development of receptor-based biosensors for chemical sensing.
A serotonin-sensitive sensor for investigation of taste cell-to-cell communication.

Abstract:Taste receptor cells are the taste sensation elements for sour, salty, sweet, bitter and umami sensations. It was demonstrated that there are cell-to-cell communications between type II (sour) and type III (sweet, bitter and umami) taste cells. Serotonin (5-HT) is released from type III cells, which is the only type of taste cells that has synaptic process with sensory afferent fibers. Then, taste information is transmitted via fibers to the brain. During this process, 5-HT plays important roles in taste information transmission. In order to explore a sensor to detect 5-HT released from taste cell or taste cell networks, we develop a 5-HT sensitive sensor based on LAPS chip. This sensor performs with a detection limit of 3.3×10−13M and a sensitivity of 19.1mV per concentration decade. Upon the stimuli of sour and mix (bitter, sweet and umami) tastants, 5-HT released from taste cells could be detected flexibly, benefit from the addressability of LAPS chip. The experimental results show that the local concentration of 5-HT is around several nM, which is consistent with those from other methods. In addition, immunofluorescent imaging technique is utilized to confirm the functional existence of both type II and III cells in a cluster of isolated taste cells. Different types of taste cells are labeled with corresponding specific antibody. This 5-HT sensitive LAPS chip provides a potential and promising way to detect 5-HT and to investigate the taste coding and information communication mechanisms.
Decoding the Molecular Mechanism of Sweet Taste of Amino Acids by Their Intrinsic Properties and Interactions with Human Sweet Taste Receptor

Abstract:The illustration of the interaction between sweet taste amino acids and sweet taste receptor. The amino, carboxyl, and hydrophobic side chain of amino acids (left lower) represent the essential AH, B, and X glucophores of sweet taste amino acids (left upper), which interact with and activate the human sweet taste receptor (horizontal arrow and right side). The molecular basis of sweet taste toward various amino acids has not been fully elucidated. In this study, by correlation of their sweetness intensities with molecular properties, it is revealed that hydrophobic or dispersion force between side chains and sweet taste receptor (Tas1R2/Tas1R3) plays a prominent role for eliciting sweetness. Furthermore, the interaction modes of 13 amino acids with receptor indicate that amino (AH) and carboxyl (B) glucophores of different amino acids interact with identical residues of receptor, while the hydrophobic side chains (X glucophores) conduct distinct topological orientations with receptor, determining their chirality selection and sweetness intensities. Molecular dynamics analysis shows that sweet d‐amino acids move toward the hydrophobic transmembrane domain during activation with more contact numbers with receptor than nonsweet l‐amino acids, providing a mechanistic elucidation of previous inconsistent sensory evaluation results of specific amino acids. Moreover, by comparison with the interactions of a dipeptide sweetener, aspartame with Tas1R2/Tas1R3 and nonsweet l‐glutamate with class C G protein‐coupled receptors, we conclude that a complementary interaction mode with both hydrophilic and hydrophobic contacts accounts for the molecular origin of their sweet taste, presenting in‐depth insights and valuable guidelines for further molecular designs or modifications of amino acids‐derived sweeteners.

Abstract:Sucrose provides both sweetness and energy by binding to both Venus flytrap domains (VFD) of the heterodimeric sweet taste receptor (T1R2/T1R3). In contrast, non-caloric sweeteners such as sucralose and aspartame only bind to one specific domain (VFD2) of T1R2, resulting in high-intensity sweetness. In this study, we investigate the binding mechanism of various steviol glycosides, artificial sweeteners, and a negative allosteric modulator (lactisole) at four distinct binding sites: VFD2, VFD3, transmembrane domain 2 (TMD2), and TMD3 through binding experiments and computational docking studies. Our docking results reveal multiple binding sites for the tested ligands, including the radiolabeled ligands. Our experimental evidence demonstrates that the C20 carboxy terminus of the Gα protein can bind to the intracellular region of either TMD2 or TMD3, altering GPCR affinity to the high-affinity state for steviol glycosides. These findings provide a mechanistic understanding of the structure and function of this heterodimeric sweet taste receptor.

Abstract:The heterodimeric sweet taste receptor, TAS1R2/1R3, is a class C G protein-coupled receptor (GPCR) that couples to gustducin (Gt), a G protein (GP) specifically involved in taste processing. This makes TAS1R2/1R3 a possible target for newly developing low caloric ligands that taste sweet to address obesity and diabetes. The activation of TAS1R2/1R3 involves the insertion of the GαP C-terminus of the GP into the GPCR in response to ligand binding. However, it is not known for sure whether the GP inserts into the TAS1R2 or TAS1R3 intracellular region of this GPCR dimer. Moreover, TAS1R2/1R3 can also connect to other GPs, such as Gs, Gi1, Gt3, Go, Gq, and G12. These GPs have different C-termini that may modify GPCR signaling. To understand the possible GP dependence of sweet perception, we use molecular dynamic (MD) simulations to examine the coupling of various GαP C20 termini to TAS1R2/1R3 for various steviol glycoside ligands and an artificial sweetener. Since the C20 could interact with the transmembrane domain (TMD) of either TAS1R2 (TMD2) or TAS1R3 (TMD3), we consider both cases. Without any sweetener, we find that the apo GPCR shows similar Go and Gt selectivities, while all steviol glycoside ligands increase the selectivity of Gt but decrease Go selectivity at TMD2. Interestingly, we find that high sweet rebaudioside M (RebM) and RebD ligands show better interactions of C20 at TMD3 for the Gt protein, but low sweet RebC and hydRebM ligands show better interaction of C20 at TMD2 for the Gt protein. Thus, our MD simulation suggests that TAS1R2/1R3 may couple the GP to either 1R2 or to 1R3 and that it can couple other GPs compared to Gt. This will likely lead to multimodal functions producing multiple patterns of intracellular signaling for sweet taste receptors, depending on the particular sweetener. Directing the GP to one of the other may have beneficial therapeutic outcomes.

Abstract:Sweet taste detection is of great significance in fields of disease prevention, food safety inspection, and drug regulation. In this study, a novel biomimetic sensor was constructed for sweet taste detection by compositing graphene oxide, hemin, and 3-aminophenylboronic acid (APBA). First, optimal fabrication of the sensor was investigated by comparing different composition proportions of graphene oxide and APBA functionalized hemin in saccharide measurements. Then, typical substances of natural sugars (i.e., glucose and fructose) and artificial sweeteners (i.e., saccharin and aspartame) were measured with the sensor through optical absorption spectroscopy. The spectra displayed apparent but different absorbance changes in the presence of those two kinds of saccharides, showing different sensing mechanisms of the biomimetic sensor for sugars and sweeteners like sweet receptors. Finally, dose-dependent and specific responses of the sensor were observed by comparing with other taste substances. The results suggested that the sensor can detect and distinguish natural sugars and artificial sweeteners in a biomimetic way. The research provided a low-cost, long-lifetime, and efficient approach for sweet taste detection, indicating its promising applications in fields such as food and beverage analysis. A Research on Sour Sensation Mechanism of Fungiform Taste Receptor Cells Based on Microelectrode Array

Abstract:Taste receptor cells as the fundamental units of taste sensation are not only passive receivers to outside stimulus, but some primary process for the signals and information. In this paper, an innovation on acquisition of taste receptor cells was introduced and larger amount of cells could be obtained. A multichannel microelectrode array (MEA) system was applied in signal recording, which is used in non-invasive, multiple and simultaneous extracellular recording of taste receptor cells. The cells were treated with sour solutions of different pHs, and the relations between concentration of hydrogen and firing rate were observed. Firing rates on pH 7, pH 4 and pH 2 were approximately 1.38 +/- 0.01 (MEAN +/- SE)/s, 1.61 +/- 0.07/s and 2.75 +/- 0.15/s.

Abstract:The sweet taste receptor plays an essential role as an energy sensor by detecting carbohydrates. However, the dynamic mechanisms of receptor activation remain unclear. Here, we describe the interactions between the transmembrane domain of the G protein-coupled sweet receptor subunit, TAS1R3, and allosteric modulators. Molecular dynamics simulations reproduced species-specific sensitivity to ligands. We found that a human-specific sweetener, cyclamate, interacted with the mouse receptor as a negative allosteric modulator. Agonist-induced allostery during receptor activation was found to destabilize the intracellular part of the receptor, which potentially interfaces with the Gα subunit, through ionic lock opening. A common human variant (R757C) of the TAS1R3 exhibited a reduced response to sweet taste, in support of our predictions. Furthermore, histidine residues in the binding site acted as pH-sensitive microswitches to modulate the sensitivity to saccharin. This study provides important insights that may facilitate the prediction of dynamic activation mechanisms for other G protein-coupled receptors.

Abstract:Sweetness in food delivers a delightful sensory experience, underscoring the crucial role of sweeteners in the food industry. However, the widespread use of sweeteners has sparked health concerns. This underscores the importance of developing and screening natural, health-conscious sweeteners. Our study represents a groundbreaking venture into the discovery of such sweeteners derived from egg and soy proteins. Employing virtual hydrolysis as a novel technique, our research entailed a comprehensive screening process that evaluated biological activity, solubility, and toxicity of the derived compounds. We harnessed cutting-edge machine learning methodologies, specifically the latest graph neural network models, for predicting the sweetness of molecules. Subsequent refinements were made through molecular docking screenings and molecular dynamics simulations. This meticulous research approach culminated in the identification of three promising sweet peptides: DCY(Asp-Cys-Tyr), GGR(Gly-Gly-Arg), and IGR(Ile-Gly-Arg). Their binding affinity with T1R2/T1R3 was lower than −15 kcal/mol. Using an electronic tongue, we verified the taste profiles of these peptides, with IGR emerging as the most favorable in terms of taste with a sweetness value of 19.29 and bitterness value of 1.71. This study not only reveals the potential of these natural peptides as healthier alternatives to traditional sweeteners in food applications but also demonstrates the successful synergy of computational predictions and experimental validations in the realm of flavor science.

Synergic Effects in the Activation of the Sweet Receptor GPCR Heterodimer for Various Sweeteners Predicted Using Molecular Metadynamics Simulations

Abstract:The sweet taste is elicited by activation of the TAS1R2/1R3 heterodimer G protein-coupled receptor. This is a therapeutic target for treatment of obesity and metabolic dysfunctions. Sweetener blends provide attractive strategies to lower the sugar level while preserving the attractive taste of food. To understand the synergic effect of various sweetener blend combinations of artificial and natural sweeteners, we carried out our molecular dynamics studies using predicted structures of the TAS1R2/1R3 heterodimer and predicted structures for the sweeteners. We used as a measure of activation the intracellular ionic lock distance between transmembrane helices 3 and 6 of TAS1R3. We find that full synergic combinations [rebaudioside A (Reb-A)/acesulfame K and Reb-A/sucralose] and partial synergic combinations (sucralose/acesulfame K) show significantly more negative changes in the free energy compared to single-ligand cases, while a pair known to be suppressive (saccharin and acesulfame K) shows significantly less changes than for the single-ligand case. This study provides an atomistic understanding of the mechanism for synergy and identifies new combinations of sweeteners to reduce the caloric content for treating diseases.

Abstract:Sweet-tasting protein is a kind of biomacromolecule that has remarkable sweetening power and is regarded as the promising sugar replacer in the future. Some sweet-tasting proteins has been used in foods and beverages. However, the structure and function relationship of these proteins is still elusive, and guidelines for their protein engineering is limited. It is well-known that the sweet-tasting proteins bind to and activate the sweet taste receptor T1R2/T1R3, thus eliciting their sweetness. The “wedge-model” for describing the interaction between sweet-tasting proteins and sweet taste receptor to elucidate their sweetness has been reported. In this perspective article, we revealed that the intramolecular interaction forces in sweet-tasting proteins is directly correlated to their properties (sweetness and stability). This intramolecular interaction pattern, named as “protein sector,” refers to a small subset of residues forming physically connections, which cooperatively affect the function of the proteins. Based on the analysis of previous experimental data, we suggest that “protein sector” of sweet-tasting proteins is pivotal for their sweet properties, which are meaningful guidelines for the future protein engineering.

Abstract:Humans perceive sweet taste via activation of a specific taste receptor expressed at the surface of taste receptor cells located on the tongue and soft palate papillae. The sweet taste receptor functions as an obligate heterodimer, comprising two different class C GPCR subunits. This receptor is unique in that it is activated or modulated by a plethora of ligands from highly diverse chemical classes, from small molecules to peptides and proteins and interacting with topologically distinct sites on each of its subunits. Modulators acting at separate functional domains of the sweet taste receptor can behave as full agonists. However, contrary to observations made with other class C GPCRs such as the metabotropic glutamate receptors and the γ-aminobutyric acid type B receptor (GABAB) receptor, modulators interacting within the allosteric sites in the transmembrane domains of the sweet taste receptor only exert a relatively small effect on the affinity and efficacy of the agonist interacting at the orthosteric binding site located within the Venus fly trap domain (VFD). Newly identified potent and efficacious positive allosteric modulators (PAM)s of the sweet taste receptor rather interact at a site in close proximity to the agonist, within the VFD, display significant probe dependence, and markedly increase the affinity of the orthosteric ligand. Several sweet taste receptor inhibitors have also been characterized. Functional studies reveal a complex relationship between different ligands. Whether the antagonist will be surmountable or insurmountable and will act competitively or non-competitively largely depends on the agonist being studied and the location of its interaction site on the receptor.
Emergence of sweet ligand-protein complexes from seemingly non-sweet conformations

Abstract:Sweet taste perception starts from the interaction of a sweet molecule with residues of the sweet taste receptor (STR). Yet, how the perception of sweet taste emerges from this interaction is still not clear. Here, we reveal a potential mechanism in the initial stages of this cascade. In our previous studies, we observed a 0.6 Pearson correlation coefficient between binding affinity and sweetness intensity. We conducted a series of MD simulations by YASARA Structure, for a total of 1.8 microseconds, to further investigate the mechanism of action of the molecules which we observed to induce considerable sweet taste perception despite having low binding affinity. We observed a gradual expansion in the hydrogen bond (HBond) network around each sweetener in the binding site which involved the water molecules and the residues of the binding pocket. Important HBond and hydrophobic interactions for sweet taste were detected. Some of the residues which we identified to be involved in the HBond network, and also to cause pattern changes in the dynamic cross-correlation matrix, have been known to be parts of the sweet taste signaling pathway. An analysis of the sweeteners' pharmacophore during the MD experiments demonstrated a gradual emergence of the Nofre-Tinti conformation, a proposed determinant in the sweetness intensity, in the ligands despite its initial absence in the ligand–protein complex. In conclusion, these results reveal a novel mechanism for inducing sweet taste and open a new avenue for designing artificial sweeteners, besides the classical hunt for high binding affinity.

In this study, a gene circuit was constructed to couple with the inherent mitogen-activated protein kinase (MAPK) pathway in Saccharomyces cerevisiae to sense sweeteners. The Sst2, Far1, and Ste2 genes were knocked out to amplify MAPK pathway signaling. Then, the C-terminus of the inherent Gpa1 of S. cerevisiae was replaced with the five residues of human taste converting protein so that Gpa1 could interact with T1R2/T1R3 for sweetness signaling. Finally, the human sweetness receptor T1R2/T1R3 was heterologously expressed in engineered S. cerevisiae by N-terminal signal peptide engineering, creating strain D3Gm, which showed the ability to sense sweeteners. This study provided proof of the concept that S. cerevisiae can be engineered to sense sweetness.
