Abstract:Stress can lead to various health problems. Exposure to stress is linked to several diseases including cancer, hypertension, diabetes, mental disorders, and heart attacks. Stress stimulates several pathways that produce free radicals, which increases oxidative stress. This results in functional and structural damage to organs, such as the brain, which is highly sensitive to oxidative stress Vitamin E is a naturally occurring potent antioxidant used for various purposes. The main purpose of the current study was to evaluate how vitamin E protects the brain and to what extent it affects antioxidant levels in rats subjected to two hours per day of immobilization stress, the form of stress with the strongest effect. The rats were immobilized by folding their limbs in and wrapping them in netting to prevent movement, while allowing their tails to be extended. The rats were then hung upside down. The effect of vitamin E was tested by intraperitoneally injecting rats with 40 mg/kg of vitamin E daily. Oxidative stress parameters were determined at the completion of the experiment. A dramatic decrease in malondialdehyde (MDA) levels and an increase in catalase (CAT), including glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD), has been shown by vitamin E in the brain. The effects of vitamin E were significantly higher in the acute and chronic stress groups than in the control group. Vitamin E can decrease oxidative stress in stressed rats, indicating that it exerts therapeutic and protective effects owing to its antioxidant properties.
The Dynamic Changes of Behavioral, Immune and Sympathetic Responses to Chronic Stress in Rats

Abstract:Previous studies have shown that emotional stress induced by randomly giving an empty water bottle to rats during watering periods for 14 consecutive days activated both the hypothalamic-pituitary-adrenal axis and the sympathetic nervous system, leading to the increased blood levels of corticosterone and norepinephrine. It also elicited attacking and exploring behaviors, suppressed the level of specific anti-ovalbumin IgG antibody production which was negatively related to norepinephrine level. However, three day empty bottle presentation had no significant effect on anti-OVA antibody production. These results suggest that the duration of stressors determines the consequences of body reactions induced by stress, at least, humoral immune responses. In order to examine whether the duration of stress exerts an accumulative effect or an adaptive effect on body reactions, the present study was designed to investigate the dynamic changes in behavioral, immune and neuroendocrine responses of rats that were subjected to chronic emotional stress with different durations. Male Wistar rats were divided into four groups: (1) emotional stress (induced by randomly giving an empty water bottle to rats at set watering time)(n=19); (2) physiological stress (given neither empty water bottles nor water during the same watering periods as for emotional stress group) (n=19); (3) control 1 (allowed free access to water during watering periods)(n=15); and (4) control 2 (given water ad libitum during the whole experimental periods)(n=15). Behavioral changes, humoral immune responses and the reaction of sympathetic nervous system were measured and compared during different days(14,21 and 28 days) of stress. The behaviors observed included attacking, exploring and grooming. Norepinephrin levels in plasma were determined using high-performance liquid chromatography. Serum levels of IgG antibodies to Ovalbumin were determined by enzyme linked immunosorbent assay. It was found that emotional stress stably induced aggressive behavior of rats over the whole experimental period. Compared with controls, rats subjected to 14,21 or 28 days of stress showed decrease in the level of specific anti-OVA antibody to a similar degree. Both 14-day and 28-day stress activated the sympathetic nervous system, leading to the increased blood levels of norepinephrine. However, a decline of norepinephrine level was also obvious from 14-day to 28-day stress. The body weight showed an early decrease during the first 14-day stress period but it was gradually recovered after prolonged stress exposure. Taken together, these results demonstrated different adaptive processes of behavior, immune, neuroendocrine and physical responses to chronic stress, and may be helpful for further understanding the complex relationships among various stress responses.
High-fat diet effects on metabolic responses to chronic stress

Abstract:CONTEXT: High-fat diets and chronic stress are prevalent risk factors for various chronic diseases in modern societies.OBJECTIVE: This study investigated the effect of high-fat diet on glucose-related metabolic responses to chronic foot-shock stress.MATERIALS AND METHODS: Male rats were divided into high-fat diet (containing 54.21% saturated and 44.89% unsaturated fatty acids) and normal diet groups and then into stress and non-stress subgroups. The diets were applied for 5 weeks, and stress was induced during the last week of the diet course. Plasma levels of metabolic parameters, HOMA-IR index, intra-abdominal fat weight, and islets' insulin secretion were assessed.RESULTS: High-fat diet increased abdominal fat weight and plasma leptin, and insulin levels in response to stress without affecting HOMA-IR index and islets' insulin secretion.CONCLUSIONS: High proportion of unsaturated fat may not lead to deleterious metabolic responses; however combined with chronic stress has a synergistic and adverse effect on visceral adiposity and results in elevated plasma leptin.

Abstract:We studied the effect of separate and combined influence of chronic forced physical activity reduction and high-fat and high-carbohydrate diet containing cholesterol on some indicators of carbohydrate, lipid, and cholesterol metabolism in growing male Wistar rats. Used combination of factors simulating a sedentary lifestyle and unhealthy diet did not have a synergistic effect on the selected biomarkers. On the contrary, the effect was antagonistic: body weight and appetite decreased and insulin resistance increased. The obtained results indicate certain prospects of hypercholesterolemia model using in preclinical studies of specialized food products to optimize the diet of individuals with disorders of carbohydrate and lipid metabolism.

[Influence of multiple insufficiency of vitamin D and B group vitamins, calcium and magnesium in the diet of rats on micronutrient sufficiency and plasma biochemical indicators]

Abstract:The lack of vitamins D, B group, calcium and magnesium is common for the diet of the Russian population. It has been previously demonstrated that that the elimination of B vitamin deficiency is a necessary condition for the implementation of vitamin D biological functions. The aim of the research was to assess the effect of a combined deficiency of vitamins D and B group, calcium and magnesium in the diet of rats on biomarkers of micronutrient sufficiency and plasma biochemical indicators. Material amd methods. Male Wistar rats with an initial body weight of 66±1 g were randomly assigned to one of five groups. The rats of the 1st group (Control) were fed a standard semi-synthetic diet (SD) for 28 days. The combined deficit of vitamins D and B group, calcium and magnesium in rats of four experimental groups was caused by a 5-fold decrease in their content in the vitamin mixture and 2-fold decrease in their content in mineral mixture of the SD for 23 days. Over the next 5 days, the rats of 2nd group (+В+D+Ca+Mg) were fed a diet replenished for all missing vitamins and minerals, the rats of 3rd group (-В+D+Ca+Mg) were fed a diet with continued deficiency of B group vitamins, the rats of the 4th group (+В+D-Ca-Mg) were fed a diet with continued lack of calcium and magnesium, the diet of the 5th group (-B-D-Ca-Mg) was not replenished. Vitamins B1 and B2 in lyophilized liver and brain and urine, riboflavin in plasma and 4-pyridoxic acid in urine were determined by fluorimetric methods, 25(OH)D in plasma was determined by ELISA, the level of vitamins A and E in blood plasma and lyophilized liver, of vitamin E in whole brain - by HPLC. Biochemical parameters of blood plasma were determined using a biochemical analyzer. Results. In rats of the 5th group (-B-D-Ca-Mg), there were 3.4-fold increase (p<0.05) in iron plasma level, 1.7-fold elevation (p<0.10) in alkaline phosphatase activity, and 1.8-fold decrease (p<0.05) in alanine aminotransferase (ALT) activity on the background of the increase in α-tocopherol blood plasma level by 26.7% (p<0.05) and liver content by 2.0 fold (p<0.05) relative to the indicators in animals who hadn't passed the deficiency of micronutrients (Control) and / or from the 2nd group (+B+D+Ca+Mg). The lack of B group vitamins in the diet, which persisted during the correction of vitamin D, calcium and magnesium deficiency in rats of the 3rd group (-B+D+Ca+Mg), inhibited the recovery of diagnostically significant biochemical parameters of blood plasma (namely, an increased level of glucose, iron, triglycerides, cholesterol, α-tocopherol, increased alkaline phosphatase activity and reduced ALT activity) to the level in animals of the control group and/or rats fed the diet replenished for all missing micronutrients (+B+D+Ca+Mg). Conclusion. Combined deficiency of several micronutrients led to changes in biochemical blood parameters. Reduced intake of calcium and magnesium during the correction of the lack of vitamins D and B group in the diet can have a negative impact on vitamin B2 status. Even under normal dietary vitamin E intake the combined deficiency of several other micronutrients affected the metabolism of this vitamin (increased levels of vitamin E in the liver and blood plasma of animals). Chronic combined alimentary deficit of B vitamins, calcium and magnesium, which is characteristic in the diet of the Russian population, reduces vitamin D bioavailability, which justifies the expediency of using vitamin-mineral complexes.

Abstract:Objective To explore the effect of chronic compound stress on serum adiponectin level and AdipoR2 expression in rats.Methods Forty male Sprague-Dawley rats were randomly divided into two groups:the stress group underwent chronic compound stress and the control group was fed normally.After 8 weeks of stress or feeding,the serum adiponectin,blood glucose ,insulin resistance and HbA1 C,HDL-C,LDL-C,triglyceride and total cholesterol were measured.Results ①After 8 weeks of stress,the score of open-field behaviors and 24-h urocortisol were significantly different between the stress and control groups (32.0 ± 8.6 vs.52.0 ± 12.7,5.8 ± 0.4 vs.(5.3 ±0.1 ) ng/L,repectively; P ＜0.05 or 0.01 ) ;②After 8 weeks of stress,the adiponectin,total cholesterol ,triglyceride,HDL-C,InIR and HbA1 C were significantly different between the two groups (P ＜ 0.05 or 0.01 ) ;③After 8 weeks of stress,there were significant difference on the expression of AdipoR2mRNA and protein between the two groups (P ＜0.01 ) ,with the expression of AdipoR2mRNA of 0.67 ±0.04 and 1.00 ±0.11 and the expression of protein of 0.73±0.02 and 1.00 ± 0.04 in the stress and control group,respectively.Adiponectin was negatively related with HbA1 C,LDL-C and IR ( r = - 0.26,- 0.88 and - 0.37,P ＜ 0.05 or 0.01 ),and positively related with HDL-C ( r =0.78 ,P ＜0.01 ).Conclusions In rats,chronic compound stress decreases the level of serum adiponectin and the expression of AdipoR2 in liver.

Abstract:Objective:To Explore the effects of chronic stress on glucose metabolism in rats.Methods:Forty male Sprague-Dawley rats were randomly divided into stress and control groups according to body weight,with twenty in each.The different weight,open-field experience,OGTT,insulin resistance and B cell function in rats in different groups were investigated.Results:Compared with the control group,the body weight,open-field experience,glucose tolerance,insulin,insulin resistance,B cell function and urinary cortisol level of the stress group were significantly differente.Conclusion:The study suggests that chronic mild stress contribute to abnormal glucose tolerance and induce insulin resistance.

Abstract:We studied the effect of Selank administered intraperitoneally in doses of 100, 300, and 1000 μg/kg to male Wistar rats 15 min prior to restraint stress on the content of aminotransferases and total protein concentration in blood serum and intensity of free radical oxidation in the liver. Under conditions of acute restraint stress, Selank in doses of 100 and 300 μg/kg decreased catalase and superoxide dismutase activities and malondialdehyde concentration and increased total antioxidant activity in the liver homogenate. Administration of Selank in a dose of 1000 μg/kg reduced the content of aminotransferases in blood serum, decreased superoxide dismutase activity in the liver, and increased total antioxidant activity. Under conditions of chronic stress, Selank in all doses produced similar effects: reduced superoxide dismutase activity and malondialdehyde concentration in the liver tissue and AST activity in the serum. The other parameters remained unchanged.

Abstract:Analysis of literature data and the results of our own research show that even with intensive rearing of animals with the use of balanced feeding and keeping them in accordance with the technology - it is almost impossible to avoid stress. And especially, excessive man-made load on agroecosystems also has a negative impact on animal life. Factors that cause a decrease in immune status and the emergence of immune pathology in animals include: industrial technology of animal husbandry, chemicalization in crop and livestock production, man-made pressure, dietary imbalance in nutrients and biologically active substances. This increases the impact on the body of heavy metals, which displace nutrients from body tissues, in particular trace elements and replace them in metabolic processes, which is a potential prerequisite for the development of oxidative stress. The mechanism of development of stress reaction of an organism is closely connected with activity of POL (lipid peroxidation) and depression of antioxidant potential. Under these conditions, the ability of the organism to mobilize protective and adaptive capabilities in response to the action of negative factors becomes especially important. Keeping productive animals in adverse conditions, unbalanced feeding are components of immobilization stress, which reduces their productivity, affects reproductive function, metabolic and functional disorders, reducing nonspecific and specific resistance of the organism (Fedoruk & Kravtsiv, 2003; Velychko, 2008; Velychko, 2011). Under conditions of man-caused load on the environment, respectively, and animals – it is promising to develop effective methods for regulating the activity of the enzymatic system of antioxidant protection with the help of biologically active substances, in particular trace elements. The results of research confirm that this has a positive effect on the formation of productive and adaptive properties of animals. The system of antioxidant protection is a necessary part of non-specific reactions of the organism, a component of the processes of its adaptation to environmental conditions, a component of normal life, a factor in maintaining homeostasis. Widespread immunodeficiency and elucidation of the main links of their pathogenesis have exacerbated the problem of regulating disorders of the immune system. Therefore, knowledge of the patterns of formation of the immune status of animals, especially in the early postnatal period, is important in the development of methods for the correction of immunodeficiency, antioxidant protection.
Effects of two types of restraint stress on spontaneous behavior of Sprague-Dawley and Lewis rats

Abstract:The purpose of this study was to evaluate the action of two types of stressors in Sprague-Dawley (S-D) and Lewis (LEW) rats differing in their hypothalamic-pituitary-adrenal axis activity on locomotion and rearing in an open space. Exposure to restraint immobilization alone (IMO) or this immobilization combined with cold water (22 degrees C) immersion (IMO+C) lasted for 1 h and started 2 or 5 h before the test. To evaluate the acute and persisting effects of both stressors, four trials were performed in one-week intervals; rats were exposed to the stressors in trial 1 and 3. While in LEW rats both acute stressors produced reduction of locomotion and rearing in all stressed groups, S-D rats responded with a decrease of both parameters only after IMO+C presented 2 h before testing. Neither strain displayed a changed performance one week after the first stress exposure. One week after the second stress exposure rats of both strains exhibited a tendency to an increase of both parameters reaching the significance in several experimental groups. The findings indicate: a) the IMO+C produced stronger behavioral alteration than IMO alone; b) the behavioral responses to stressors were more pronounced in LEW compared to S-D strain; c) change from the reduction of activity to its increase may be interpreted as bi-directional manifestation of long-term effects of immobilization stress.
Effect of acute and chronic stress on memory impairment and hippocampal oxidative stress following global cerebral ischemia in adult male rats

Abstract:Background and purpose: Stress, especially immobility stress, is quite common and one of the most important and influential risk factors in neurological disorders. This study aimed to investigate the effect of acute and chronic immobility stress on the level of cortical and hippocampal oxidative stress indicators and memory impairment following global cerebral ischemia. Experimental approach: In this study, 48 male Wistar rats were randomly divided into 6 groups: 1, sham (S); 2, sham-acute stress (SSA); 3, sham-chronic stress (SSC); 4, ischemia (IS); 5, ischemia-acute stress (ISA); 6, ischemia-chronic stress (ISC). The Morris water maze (MWM) test was performed 14 days after surgery, and cortisol levels and oxidative stress factors such as malondialdehyde MDA and total thiol were measured. Findings/results: In the MWM test, the time to find the platform (latency time) in the ISC and IS groups significantly increased compared to the S group. The time spent in the target quarter in these two groups was significantly reduced compared to the S group on the day of the probe. The results showed a significant increase in cortisol levels and malondialdehyde concentration in the ISA, ISC, and IS groups compared to the S group, but there was no significant difference in total thiol concentration. No significant difference was observed in the level of oxidative stress factors in the cortex. Conclusion and implication: Chronic immobility stress could reduce antioxidant factors in the hippocampus and exacerbate memory impairment caused by global ischemia.
Experimental study of the relationship between chronic stress and somatic symptoms

Abstract:Objective To study the possible effects of chronic stress on somatic symtoms through observing the gastric motility of rats under chronic stress. Methods Seventy-five Sprague Dawley(SD) male rats were randomly divided into two groups as chronic stress group(n=50) and normal control(n=25).The chronic stress group underwent water immersion restraint stress 1 hour everyday for 21 consecutive days,and the raising conditions were similar for two groups.Observation index:(1) The changes of bodyweight and food intake of the rats after chronic stress;(2) Stool obscure blood test of the rats;(3) The contents of monoamine transmitters and related metabolites(5-HT,5-HIAA,NA,MHPG,DA and HVA)in the cerebrospinal fluid after chronic stress;(4) Evaluation of cognitive ability by water maze test;(5) The measurement of the velocity of gastric emptying by using phenol red test. Results(1) Before the chronic stress,there was no significant difference in the body weight(g)(258.27±10.72 vs 260.97±10.29,P>0.05) and the food intake(g)(26.63±0.82 vs 25.99± 0.43,P>0.05) between the two groups;after the chronic stress,the body weight of the stress group was significantly lower than the control(289.72±9.84 vs 392.48±10.81,P<0.001),and the food intake was significantly smaller than that of the control(19.72±1.99 vs 29.22±1.45,P<0.01);(2) The stool obscure blood test of the two groups was negative;(3) Comparing with the control,the contents of 5-HT and 5-HIAA of the stress group were significantly lower(14.80±2.50 vs 34.5±2.50,P<0.001 and 68.36±5.34 vs 114.15±6.13 respectively,P<0.001),while the contents of DA,NA and HVA were higher,but with no statistical significance;(4) There was a significant difference in time consumption for the rats to swim out of the maze between the two groups in water maze test(the first day: 57.2±12.01 vs 47.68±10.13,P<0.01;the second day: 58.44±14.13 vs 46.88±10.48,P<0.01),but there was no significant difference in the error frequency between the two groups in water maze test(the first day:6.22±3.26 vs 5.88±3.38,P=0.403,the second day: 6.4±3.31 vs 5.72±3.37,P=0.304);(5) The gastric residual phenol red rate of the stress group was significantly higher than that of the control(33.52±10.00 vs 21.66±6.22,P<0.005). Conclusions Chronic stress can cause a decrease gastric motility of rat without disorders of cognitive function.

Abstract:Oxidative stress is caused by an imbalance between the production of harmful oxygen molecules and the body's ability to repair their detrimental implications, leading to reduced growth rates, increased disease susceptibility, and decreased reproductive performance in animals. Vitamin A, comprising retinol, retinal, and retinoic acid, is crucial for normal growth, reproduction, and vision. Vitamin A possesses antioxidant properties by directly scavenging reactive oxygen species, boosting antioxidant enzyme activity, and promoting antioxidant defence mechanisms. Numerous studies have shown that livestock with adequate levels of vitamin A in their diet experience reduced oxidative stress compared to those with vitamin A deficiency. Moreover, vitamin A supplementation can mitigate oxidative stress in animals exposed to stressful conditions like heat stress. Adequate vitamin A status in livestock through dietary interventions and improved animal management practices can significantly benefit animal health and well-being. However, further research is still needed to optimize dosing strategies and fully understand the relationship between vitamin A and oxidative stress in different animal species and production systems. Therefore, continued research efforts are essential to fully harness the potential of vitamin A as an effective tool for mitigating oxidative stress and improving animal welfare.

Chronic orthostatic and antiorthostatic restraint induce neuroendocrine, immune and neurophysiologial disorders in rats

Abstract:The tail-cast suspension rat model has been developed in ground laboratories interested in space physiology for extensive study of mechanisms causing the pathophysiological syndrome associated with space flights. We used individually-caged male rats to explore the effects of acute and chronic (7d) orthostatic restraint (OR) and head-down anti-orthostatic restraint (AOR) on a series of physiological variables. The acute restraint study showed that (1) the installation of the OR device induced an acute reaction for 2 days, with a substantial rise in ACTH (x2) and CORT (x6), and that (2) the head-down tilt from OR to AOR induced (i) within 10 min and lasting 60 min a 2-fold rise in the intra-cerebro-ventricular pressure (Picv) monitored with an icv telemetric recording system, which receded to normal between 60 and 120 min; and (ii) within 30 min a short-lived 4-fold rise in plasma ACTH and CORT levels. Chronic OR induced (1) the suppression of the diurnal ACTH/CORT rhythm, with increased mean levels, especially for ACTH, (2) a degraded circadian locomotor activity rhythm manifested by a significant reduction in the spectral power of the 24h periodicity and a concomitant emergence of shorter (ultradian) periodicities, (3) an associated, but less pronounced alteration of the diurnal rhythm in body temperature; and (4) a marked increase in baseline plasma levels of IL-1 beta and an increased reactivity in cytokine release following an E. coli endotoxin (LPS) challenge. AOR induced (1) a similar obliteration of the circadian ACTH/CORT rhythm, (2) the loss of close correlation between ACTH and CORT, (3) a generalized increase in baseline plasma IL-1 beta levels and (4) more extensive degradation of the circadian periodicity for both locomotor activity and, to a lesser extent, body temperature, replaced by dominant spectral powers for ultradian periodicities (3 to 10h). In conclusion, both experimental paradigms--but AOR more than OR--caused a blockade of the circadian rhythmicity of major physiological variables, the loss of normal correlations between ACTH and CORT, and inflammatory-immune hyperreactivity. These pathophysiological disorders may all be parts of a complex chronic stress syndrome.
Alterations of serum biochemical parameters and tyrosine phosphorylation in kidney and liver of chronic stress-induced rats

Abstract:Abstract Chronic stress (CS) can contribute to dysfunction in several organs including liver and kidney. This study was performed to investigate the changes in serum biochemistry, histological structure, as well as in localization of tyrosine phosphorylated proteins (TyrPho) and Heat shock protein 70 (Hsp-70) in liver and kidney tissues of CS rats induced by two stressors (restrained and force swimming) for 60 consecutive days. Samples of blood, liver, and kidney were collected from adult male Sprague–Dawley rats in each group. Our results showed that serum biochemical parameters including corticosterone, blood sugar, urea nitrogen, creatinine, cholesterol, triglyceride, HDL-C, LDL-C, ALT, AST, alkaline phosphatase in CS group were significantly different from that in normal group in both liver and kidney tissues. Although histological structure was not changed. TyrPho expression was significantly increased in liver lysate but significantly decreased in kidney. Hsp-70 expression in liver increased whereas in kidney decreased. In conclusion, CS can induce changes in liver and kidney functions.

Abstract:We studied stress-induced organ and hormonal responses in behaviorally active and passive rats against the background of immunization with glutamate-BSA conjugate. The relative weight of the adrenal glands after immobilization was lower in rats immunized with the conjugate in comparison with non-immunized animals. The weight of the adrenal glands in behaviorally active rats decreased in parallel with the decrease in blood corticosterone. In behaviorally active and passive rats immunized with the conjugate, ulcer formation in the stomach was slightly intensified after immobilization. It was hypothesized that immunization with glutamate-BSA conjugate suppresses activity of the hypothalamic-pituitary-adrenal feedback mechanism underlying the production of glucocorticoid hormones, which is manifested in slightly increased ulceration due to attenuation of the gastroprotective action of corticosterone under stress.
INFLUENCE OF VITAMIN E ON THE HAEMODYNAMIC EFFECTS OF INTRAGASTRIC CHROMIUM INTAKE AND LIPID PEROXIDATION IN EXPERIMENTAL RATS

Abstract:Purpose: To examine the possibility of using vitamin E for the correction of free radical oxidation disorders and haemodynamic effects of potassium bichromate. Methods: Experiments were performed in 90 male Wistar rats; the study included 9 experimental groups. Vitamin E and sunflower oil (control) were injected daily intragastrically through an atraumatic tube into the stomach. At the end of the experimental period (30 days and 60 days), the main parameters of systemic hemodynamics and values of lipid peroxidation were measured. Differences between groups were analyzed using Student’s t-test. Results: The analysis of basic parameters of systemic haemodynamics showed that PO administration of potassium bichromate promoted an increase in mean arterial pressure due to the increase in normalized peripheral vascular resistance (systemic vascular resistance, SVR). At the same time there was a decrease in the stroke volume index. The haemodynamic effects of chromium were more pronounced with prolonged administration. Parameters of systemic hemodynamics in combined administration of potassium bichromate and sunflower oil changed similarly to the effects of combined administration of the metal and vitamin E, but changes were less pronounced. A decrease in SVR and the restoration of the stroke volume index were observed. The isolated administration of the metal for thirty days resulted in an increase in lipid peroxidation (LPO) accompanied by a stimulation of catalase and superoxide dismutase activity. During prolonged (2 months) isolated administration of potassium bichromate, a more pronounced increase in LPO was observed along with the depletion of the antioxidant system. The prevention of chromium intoxication using vitamin E during the first month of administration reduced lipoperoxidation phenomena accompanying increased antioxidant defense activity. These dynamics persisted after two months of the combination of vitamin E and potassium bichromate. Conclusions: The intragastric administration of potassium bichromate for 30 and 60 days resulted in arterial hypertension in a rat model. The administration of toxic doses of potassium bichromate to laboratory animals may lead to the activation of lipid peroxidation. The administration of vitamin E may attenuate the haemodynamic effects of chromium intoxication and the activation of lipid peroxidation.
Blood Oxidative Stress Modulates Alveolar Bone Loss in Chronically Stressed Rats

Abstract:We aimed to investigate the effects of chronic stress (CS) on experimental periodontitis (EP) in rats. For this, 28 Wistar rats were divided into four groups: control, ligature-induced experimental periodontitis (EP), chronic stress (CS; by physical restraint model) and CS+EP (association of chronic stress and ligature-induced periodontitis). The experimental period lasted 30 days, including exposure to CS every day and ligature was performed on the 15th experimental day. After 30 days, the animals were submitted to the behavioral test of the elevated plus maze (EPM). Next, rats were euthanized for blood and mandible collection in order to evaluate the oxidative biochemistry (by nitric oxide (NO), reduced-glutathione activity (GSH), and thiobarbituric acid reactive substance levels (TBARS)) and alveolar bone characterization (by morphometric, micro-CT, and immunohistochemistry), respectively. The behavioral parameters evaluated in EPM indicated higher anxiogenic activity in the CS and CS+EP, groups, which is a behavioral reflex of CS. The results showed that CS was able to change the blood oxidative biochemistry in CS and CS+EP groups, decrease GSH activity in the blood, and increase the NO and TBARS concentrations. Thus, CS induces oxidative blood imbalance, which can potentialize or generate morphological, structural, and metabolic damages to the alveolar bone.
