Abstract:In this study, surimi products rich in lipids were prepared by using myofibril protein (MP) emulsion gel as carriers. The MP emulsion gel (MP concentration, c = 1.5%, oil fraction, o = 0.68) was prepared by one-step homogenization. The emulsion gel maintained a high elastic modulus (G ') after heating and freezing treat-ment. Confocal laser scanning microscopy revealed that the structure of the emulsion gel was a hybrid network consisting of polymers of cross-linked MP and aggregated protein-stabilized emulsion (W/O/W multiple struc-tures) droplets. The double emulsification of the emulsion gel and MP stabilized the oil droplets in the surimi product, preventing water and oil from leaching out. The microstructure also showed smaller gaps between MPs with increased porosity, while oil droplets were stably embedded in the surimi gel matrix. Moreover, adding MP emulsion gel significantly reduced the surimi gel strength compared to adding oil directly (p < 0.05).

Abstract:The gel strength of kamaboko obtained from walleye pollock (Theragra chalcogramma) was much reduced when the surimi paste was incubated at 60°C for 2h. The proteinase inhibitor, E-64, could enhance the gel strength and, at 200μg/g of surimi, appeared to be the most effective. Amino acid analysis of the corresponding kamaboko showed that E-64 suppressed the release of peptides. Electrophoretograms revealed the inhibition of MHC degradation by E-64. Actomyosin was extracted and subjected to Sepharose 6B gel filtration to obtain the actomyosin non-binding cathepsin L (Lmix). Studies of substrate specificity and the effect of activators and inhibitors confirmed that the thiol–cysteine enzyme obtained was crude cathepsin L. Its high heat-stability indicated its strong hydrolytic ability. Lmix, at 0.6unit/g of surimi, greatly decreased walleye pollock kamaboko gel strength from 112 to 27.8g/cm2 when incubated at 60°C for 2h. The degradation was effectively inhibited by E-64 at 200μg/g of surimi and the gel strength of the corresponding kamaboko was increased to 302g/cm2. The above results suggested that cathepsin L contributed to the modori phenomena in kamaboko processed from walleye pollock surimi.
The fabrication and characterisation of surimi gel modified by konjac glucomannan with the different hydrolysis degree

Abstract:The gel performance of surimi-based products is indispensable technological attribute responsible for their high popularity. Here, surimi gels with tunable gel properties were successfully fabricated based on the double network theory. Konjac glucomannan (KGM) hydrolysed for various periods (0, 5, 15, 30, 60 and 90 min) was introduced into surimi systems. Corresponding structural characterisations including FTIR, XRD, SDS-PAGE and grafting degree assessment, were conducted to confirm the presence of double crosslinking reactions. Furthermore, the gel strength, dynamic rheology, thermal stability and in vitro digestibility of the surimi gels were investigated. Red shifts in the amide A band and an increase in relative intensity indicated that hydrogen bonding existed in the complex systems, according to the FTIR spectra. The XRD patterns suggested that the crystalline structures were weakened by interfering with the protein chain arrangement due to the Maillard reaction. In addition, SDS-PAGE and o-phthalaldehyde (OPA) assays also confirmed that myofibrillar protein and hydrolysed KGM were partially linked by covalent bonding. The SK-2 group showed the greatest mechanical strength and lowest digestibility amongst all groups due to its dense hierarchical structure. This work may provide inspiration for the design and development of surimi gels with improved gel properties via microarchitectural modulation.

Abstract:<p>This study aimed to compare heat-induced gelation in <em>Litopenaeus vannamei</em> and Alaska pollock surimi. Gelation differs in shrimp surimi and surimi upon heating. Texture profile analysis revealed that the shrimp surimi was more elastic and harder than surimi. Frequency and creep and recovery tests indicated that the differences in gelation account for the higher levels of cross-linkage, elasticity, and hardness in shrimp surimi than in surimi. The temperature sweep test revealed two peaks for the shrimp surimi curve and only one peak for the surimi curve in response to heat treatment, representing protein denaturation peaks. Gel electrophoresis revealed that paramyosin (PM) is primarily responsible for these differences and the two peaks in the temperature sweep. Furthermore, the β-sheet content increased in both heated shrimp surimi and surimi in comparison with unheated groups, according to Fourier transform infrared spectroscopy. However, the α-helix content increased in shrimp surimi but decreased in surimi relative to the unheated groups. Therefore, gelation differed between the two materials, and the shrimp surimi structure was more stable than that of surimi. Scanning electron microscopy revealed the post-heating microstructure of shrimp surimi was denser than that of surimi. Fractal dimension calculations confirmed that shrimp surimi gel has a higher degree of cross-linkage than surimi gel because of paramyosin. These findings helped elucidate the heat-induced gelation process in shrimp surimi and develop more products in accordance with the features of shrimp surimi.</p>

Abstract:Solid-fermentation is a promising way of producing novel surimi-based food. In the present study, changes in physicochemical characteristics, protein hydrolysis, and textual properties of surimi during the fermentation, including pehtze preparation, salting, and ripening stages, were studied. The protease produced by Actinomucor elegans led to continuous hydrolysis of surimi protein, reflected from the high value of protease activity which reached 69.34 U/g after pehtze preparation stage, the decrease of protein bands intensity in sodium dodecyl sulfate polyacrylamide gel electrophoresis image especially for bands of 200, 43, and 36 KDa, and the increasing contents of free amino acids and trichloroacetic acid-soluble nitrogen. Hardness, springiness, and cohesiveness of the surimi all decreased during pehtze preparation stage, increased after salting, and decreased again after ripening, while adhesiveness kept increasing until the ripening stage started. Scanning electron microscopy images showed a dense and soft microstructure of surimi after ripening. The final surimi product showed high overall acceptability and no Enterobacteriaceae was detected.

Abstract:Several kinds of emulsified surimi gels were prepared from different quality levels of Alaska Pollack surimi, and the relationship between the emulsifying stability (ES) of myofibrillar protein and the properties of the emulsified surimi gels was investigated. Fish oil emulsified into surimi gels enhanced the breaking strength, but this was decreased by denaturation of the surimi protein, and the rate of enhanced gel-forming ability with emulsification decreased with decreasing ES. Expressible drip also decreased with emulsification; however, increasing amounts of lipid in the expressible drip were separated out from the gel upon protein denaturation of the source surimi. Scanning electron microscopy revealed that the shape of fish oil particles became irregular and some voids caused by oil leakage were observed with increasing storage period of source surimi. The results suggested that improvement in gel properties of the emulsified surimi gels was correlated with ES as well as the level of protein denaturation.

Abstract:It is well known that aquatic products such as fish and shellfish, when stored for a long period of time under inappropriate conditions, can suffer from muscle softening. This phenomenon is mainly caused by endogenous proteases, which are activated during heating and accelerates the degradation of myofibrillar proteins, directly leading to weaker gels and poorer water retention capacity. This paper reviews the changes in fish proteins during storage after death and the factors affecting protein hydrolysis. A brief overview of the extraction of protease inhibitors, polysaccharides and proteins is given, as well as their mechanism of inhibition of protein hydrolysis in surimi and the current status of their application to improve the properties of surimi.

Abstract:To investigate the cross-linking degree on the in vitro gastrointestinal digestion and absorption properties of surimi gel, three types of surimi gels with low, moderate, and high cross-linking degrees were prepared, and then in vitro digestion models (static and dynamic) and a Caco-2 cell monolayer model combined with LC-MS/MS were used to do peptidomic analysis of digestive and absorbed juices. The results showed that an increase in cross-linking degree promoted the release of peptides after gastrointestinal digestion. These peptides originated from the myosin head and rod, the rod was the main digestion region. More potential bioactive peptides from intestinal digestive juice could be transported through the intestinal epithelium. Compared with static digestion, dynamic digestion digested surimi gels more thoroughly, especially during gastric digestion. This study provides a theoretical basis and guidance for the production of surimi products with higher nutritional value and the in vitro digestion methods of gelatinous foods.

Abstract:Variations in the cold gelation of shrimp surimi at low temperatures significantly affect their gel properties and secondary processing. In this study, the physical properties and protein conformation of shrimp surimi at 4 °C storage were investigated for fresh shrimp (F0) and shrimp frozen for 4/8 months (F4/F8). Gel hardness and frequency tests indicated that F0 maintained a sol state, whereas F4 and F8 exhibited elastic gel characteristics, and a higher degree of crosslinking occurred in F8 during storage. Gel electrophoresis revealed that the myosin heavy chain underwent polymerization in F4 and F8 during cold storage. The Raman spectra showed that F4 and F8 exhibited lower α-helix and higher β-sheet contents after storage. The intensity of C–H bending peaks and the ratio of tyrosine doublet bands also decreased significantly in F4 and F8. The changes in the secondary and tertiary structures in F8 were more notable than those in F4. Scanning electron microscopy confirmed the formation of the most compact gel network in F8. These results helped clarify the gelation process of shrimp surimi prepared from fresh and frozen states at low temperatures and contributed to regulate the gelation properties of shrimp surimi products.
Improvement of gelation properties of Penaeus vannamei surimi by magnetic field-assisted freezing in combination with curdlan

Abstract:Traditional methods of freezing and thawing may harm the quality of meat products. In order to reduce the negative impact of freezing on surimi products, the magnetic field-assisted freezing method is combined with various curdlan ratios to enhance the gelation characteristics of Penaeus vannamei surimi in this study. The results showed that the magnetic field-assisted freezing technique significantly improved the quality of thawed surimi compared with soaking freezing (SF), whereas the addition of curdlan further improved the gelation properties, and the gel strength, water-holding capacity, textural properties, whiteness, and G' value were significantly improved when its content was increased to 0.6 %. However, excessive amounts of curdlan interfered with protein covalent cross-linking, leading to a decrease in gel quality. Additionally, the addition of magnetic field and curdlan encouraged the shift of the α-helix to the random coil and β-sheet transition, which stimulated the growth of myofibril molecules, exposed the hydrophobic groups and thiols, improved protein-molecule interactions, and promoted systematic gathering of proteins, leading to the formation of the microstructure of dense and small pores. It also resulted in a drop in water release, an increase in the proton density and a shift in the water condition from free water to more immobile water, which had higher sensory qualities. These effects together resulted in a reduction in thawing and cooking loss to 11.41 % and 13.83 %, respectively. These results also help to clarify the gelation process of shrimp surimi and help to regulate the gelation characteristics of shrimp surimi products.

Abstract:The object of this study is to investigate the effects of sodium alginate (SA) and CaCl 2 on the physicochemical characteristics of shrimp surimi by studying gel strength, water holding capacity (WHC), as well as by texture profile analysis (TPA). In addition, the mechanism was analyzed through chemical interactions, protein secondary structure, and microstructure. The results showed that with the addition of different concentrations of SA and CaCl 2 to the shrimp surimi, the gel quality firstly increased and then decreased with the increase of SA and CaCl 2 concentrations. The highest values of WHC , breaking force and gel strength were obtained with the addition of 1.2% SA or 0.1% CaCl 2 . When SA and CaCl 2 were used in concert, the group containing 1.2% SA and 0.15% CaCl 2 had the highest gel strength with the densest three-dimensional network structure of the gel. In addition, the results of chemical interaction analyses showed that hydrogen and ionic bonds were the main chemical bonds of shrimp surimi sol, while shrimp surimi gel mainly consisted of hydrophobic and disulfide bonds. The incorporation of SA and CaCl 2 resulted in a significant increase in hydrophobic interactions and disulfide bonding, which could effectively improve the gel properties of shrimp surimi.

Abstract:High resistance to heating treatments is a prerequisite for ready-to-eat (RTE) surimi products. In this study, emulsion-formulated surimi gels were prepared, and the effects of oil types and emulsification degrees on the thermal stability of surimi gel were investigated. The results showed the gel properties of surimi gels were modulated by oil types and emulsification degrees. In detail, the rising pre-emulsification ratio caused the increase of the emulsifying activity index (EAI) and decrease of emulsifying stability index (ESI) for both emulsions. The larger droplet sizes of perilla seed oil than soybean oil may be responsible for their emulsifying stability difference. The gel strength, water retention, dynamic modulus and texture properties of both kinds of surimi gels displayed a firstly increased and then decreased tendency with the rising pre-emulsification ratios. The peak values were obtained as perilla seed oil emulsion with emulsification ratio of 20% group (P1) and soybean oil emulsion with emulsification ratio of 40% group (S2), respectively. Anyway, all emulsion gels showed higher thermal stability than the control group regardless of oil types. Similar curves were also obtained for the changes of hydrogen bond, ionic bond and hydrophobic interactions. Overall, perilla seed oil emulsion with emulsification ratio of 20% (P1 group) contributed to the improved thermal stability of surimi gels.

Abstract:ABSTRACT: The effects of shrimp head protein hydrolysate (SHPH) from three species of shrimp (northern pink shrimp [Pandalus eous], endeavour shrimp [Metapenaeus endeavouri], black tiger shrimp [Penaeus monodon]) on gel forming ability and protein denaturation of lizardfish surimi during frozen storage at −25°C were evaluated. The quality of lizardfish surimi with 5% (dried matter) of any of the three SHPH or sodium glutamate (Na‐Glu) was examined in terms of gel strength, whiteness, Ca‐ATPase activity and the amount of unfrozen water, comparing with those of surimi without additive as the control. The residual Ca‐ATPase activity and gel strength of surimi with SHPH were higher than those of the control throughout 180 days of frozen storage, regardless of shrimp species. The highest effect was found in surimi with Na‐Glu. The gel strength and Ca‐ATPase activity found a high positive correlation. The addition of SHPH to surimi also increased the amount of unfrozen water by approximately 1.29–1.36 fold higher than the control, however kamaboko gels of the control was significantly whiter. From these results, freeze‐induced denaturation of lizardfish muscle protein could be lessened by the addition of SHPH, resulting in a high gel strength and Ca‐ATPase activity.

Abstract:Fermentation conditions and microorganisms were determined, based on acid production, glucose concentration as carbohydrate source. Inoculation levels to obtain a stable shrimp waste silage were also determined. Shrimp waste ensilation was an efficient method of preservation, allowing the recovery of chitin and another added-value products such as pigments, proteins and enzymes. From the various lactic acid bacteria tested, Lactobacillus pentosus and Lactobacillus sp. (B2) were the best lactic acid producers, although small quantities of acetic acid were detected in samples inoculated with Lactobacillus pentosus. Therefore B2 was chosen for the analysis of glucose consumption as well as for the determination of optimum inoculation levels. The best results were obtained at 10% (w/w wet basis) and 5% (v/w wet basis) respectively. Presence of starters and initial glucose concentration were critical factors in the fermentation of shrimp waste. High initial glucose and starter concentrations reduced the time and increased the amount of lactic acid produced. The fermentation pattern changed during ensilation from hetero to homofermentative. Shrimp waste ensilation prevented the growth of spoilage microorganisms keeping their microbial counts steady and pH values within the acid region.

Abstract:One-step fermentation, inoculated with Lactobacillus fermentum (L. fermentum) in shrimp by-products, was carried out to obtain chitin and flavor protein hydrolysates at the same time. The fermentation conditions were optimized using response surface methodology, resulting in chitin with a demineralization rate of 89.48%, a deproteinization rate of 85.11%, and a chitin yield of 16.3%. The surface of chitin after fermentation was shown to be not dense, and there were a lot of pores. According to Fourier transform infrared spectroscopy and X-ray diffraction patterns, the fermented chitin belonged to α-chitin. More than 60 volatiles were identified from the fermentation broth after chitin extraction using gas chromatography–ion transfer spectrometry analysis. L. fermentum fermentation decreased the intensities of volatile compounds related to unsaturated fatty acid oxidation or amino acid deamination. By contrast, much more pleasant flavors related to fruity and roasted aroma were all enhanced in the fermentation broth. Our results suggest an efficient one-step fermentation technique to recover chitin and to increase aroma and flavor constituents from shrimp by-products.

Abstract:Decapterus maruadsi surimi products were prepared using the thermal treatment methods of boiling (BOI), steaming (STE), back-pressure sterilization (BAC), roasting (ROA), microwaving (MIC), and frying (FRI), respectively. The effect of glutamine transaminase (TGase) addition was also investigated. The moisture distribution, water retention, microstructure, color, fracture constant, protein secondary structure, chemical forces, and flavor components of each sample were determined. The differences in gel and favor characteristics between D. maruadsi surimi products caused by thermal treatment methods were analyzed. The results showed that BOI, STE, and FRI had the largest protein secondary structure transitions and formed dense gel structures with high fracture constant. The kinds of flavour components in BOI and STE were completer and more balanced. The high temperature treatment available at BAC and FRI (110 °C and 150 °C) accelerated the chemical reaction involved in flavor formation, which highlighted the flavor profiles dominated by furans or esters. The open thermal treatment environments of ROA, MIC, and FRI gave them a low moisture content and water loss. This allowed the MIC to underheat during the heat treatment, which formed a loose gel structure with a low fracture coefficient. The addition of TGase enhances the gel quality, most noticeably in the ROA. The aldehyde content of the FRI was enhanced in the flavor characteristic. The effect of adding TGase to enhance the quality of the gel is most evident in ROA. It also substantially increased the content of aldehydes in FRI. In conclusion, different heat treatments could change the gel characteristics of surimi products and provide different flavor profiles. The gel quality of BOI and STE was consistently better in all aspects.

Abstract:Traditional ice is usually employed to preserve food freshness and extend shelf life. However, ice cannot bear repeated freeze - thaw cycles during the transportation and retailing process, resulting in microbial cross-contamination and spoilage of foods. Herein, succinoglycan riclin was oxidated (RO) and crosslinked with gelatin (Ge), the Ge-RO cryogels were prepared via Schiff base reaction and three freeze - thaw cycles. The Ge-RO cryogels showed improved storage modulus (G') and thermal stability compared with pure gelatin hydrogel. The polymer framework of Ge-RO gels exhibited stable properties against ice crystals destructions during nine freeze - thaw treatments. During the storage and repeated freeze - thaw treatments of shrimps, Ge-RO cryogels exhibited a remarkable preservation effect on shrimps, and their freshness was evaluated using an electronic nose technique equipped with ten sensors. The results demonstrated that the shrimp muscle preserved in ice generated off-odors and resulted in high sensor responses. The sensor responses were reduced sharply of shrimps preserved in cryogels. Moreover, 1H NMR-based metabolomics analysis revealed that shrimps in Ge-RO cryogels group reversed the metabolic perturbations compared with the traditional ice group, the metabolic pathways were related to energy metabolism, nucleotide metabolism, and amino acid metabolism, which provide new clues to the freshness of shrimps. Furthermore, RO exhibited superior antimicrobial activity against E. coli and S. aureus microorganisms. Thus, the crosslinked cryogels are potentially applicable to food preservation, offering sustainable and reusable solutions against traditional ice.
