Abstract:This paper investigated the feasibility for pasteurizing raw (100 %) pomegranate juice in a commercial scale pulsed electric field (PEF) processing system. The juice was processed at 35 and 38 kV/cm for 281 μs at 55 °C with a flow rate of 100 L/h. Effect of PEF processing on microbial stability, color, °Brix, pH, sediment, antioxidant activity, total phenolic content, anthocyanin, and sensory properties after the treatments and during storage at 4 °C for 12 weeks were studied and compared to those of thermally processed juice. PEF treatments significantly ( p < 0.05) inhibited the growth of total aerobic bacteria, which remained at <2.5 log colony-forming units (CFU)/ml during the 12-week storage. No yeast and mold were detected (<0.69 log CFU/ml) in the PEF-treated juices during storage up to weeks 10 and 12, which is similar to the thermally processed juice. There were no significant differences in pH and °Brix values between the PEF processed juice and unprocessed juice. PEF processing did not alter the contents of total phenolics and anthocyanin as compared to unprocessed juice. PEF processing had significantly ( p < 0.05) less impact on the color of pomegranate juice than thermal processing. PEF-treated juice had the same consumer satisfaction scores as the unprocessed juice, which were significantly ( p < 0.05) higher than thermally processed juice samples. There was no significant difference between the two PEF treatments in all results. This study demonstrated that PEF technology extended microbial shelf-life and preserved the major quality and nutritional characteristics of pomegranate juice, and hence, is technically feasible for commercialization in the juice industry.

Abstract:This study was conducted to investigate the effectiveness of pulsed electric fields (PEFs), pulsed UV light (PL), and antimicrobial packaging (AP), either individually or combined, for inactivating bacteria and maintaining the quality of fruit juices. Apple juice samples were inoculated with Escherichia coli K-12 or native molds and yeasts (M&Y), treated with bench scale PEF and/or PL processing systems, and stored in glass jars with antimicrobial caps containing 10 mu L of carvacrol. The reductions in microbial populations and the physicochemical properties of juice samples were determined after treatments and during storage at 10 degrees C. The treatments were PL (5 to 50 s at 1.04 J/cm(2)/s), PEF (19, 23, and 30 kV/cm), PEF followed by PL (PEF+PL), PL followed by PEF (PL+PEF), and PEF+PL+AP. PEF treatments at 19 to 30 kV/cm (PEF19, PEF23, and PEF30) resulted in 2.0-, 2.6-, and 4.0-log reductions of E. coli, respectively, and PL treatments for 10 to 50 s (PL10, PL20, PL30, PL40, and PL50) resulted in 0.45-, 0.67-, 0.76-, 2.3-, and 4.0-log reductions, respectively. No significant differences (P > 0.05) were found between the combined PL20+PEF19 and PEF19+PL20 treatments; both treatments reduced E. coli K-12 populations to nondetectable levels (> 5-log reductions) after 7 days. Both PEF+PL and PEF+PL+AP treatments achieved > 5-log reductions of M & Y; however, juice samples subjected to PEF+PL+AP treatment had lower M&Y counts (2.9 log units) than did samples subjected to PEF+PL treatment (3.9 log units) after 7 days. No significant differences (P > 0.05) in pH, acidity, and total soluble solids were found among all samples after treatments. Increased PL treatment times reduced color a* and b* values, total phenolics, and carotenoids. This study provides information valuable to juice processors for consideration and design of nonthermal pasteurization of juice products. HIGHLIGHTS Combined treatments had additive or synergistic antimicrobial effects. Combined treatments achieved.5-log reductions of E. coli and molds and yeasts. Combined treatments did not significantly affect pH, acidity, or total soluble solids. Combined treatments slightly impacted color, carotenoids, and phenolic compounds.

Abstract:Pomegranate juice was processed using bench top (7.2L/h flow rate, 35kV/cm field strength, 72µs total treatment time) and pilot scale (100L/h flow rate, 35kV/cm field strength, 281µs total treatment time) continuous pulsed electric field (PEF) processing systems. The treated juice was packaged in PET bottles or PET bottles coated with potassium sorbate and sodium benzoate, and stored at 4°C for 84days. Samples were assessed every 7days for total aerobic bacteria and yeast and mold. Untreated juice had less than one week of shelf-life, while untreated juices in antimicrobial bottles had 56days. Juices treated with PEF alone had a shelf-life of 21days (bench scale) and over 84days (pilot scale). Juices treated with PEF and stored in antimicrobial bottles had a shelf-life over 84days for both scale tests, which significantly extended the microbiological shelf-life of pomegranate juice.

Abstract:Pulsed electric field (PEF) processing of juice has been intensively studied with benchtop scale experiments. However, there is still limited information regarding critical factors to be considered for PEF efficacy in microbial reduction with PEF processing during pilot or commercial scale production of juice. In the present study, continuous benchtop (3.6-7.2 L/h) PEF processing systems with co-field treatment chambers and bipolar square waveform pulses were used and simulated production conditions were tested for pomegranate juice. Microbial reductions of Escherichia coli, as affected by PEF process conditions (field strength, pulse width, pulse frequency, total treatment time, input energy), production conditions (flow rate, juice holding time and temperature), and juice properties (pH, conductivity, particulate), were investigated. Flow rate, PEF process parameters, production conditions, type of target microorganism, and properties of juice significantly affected microbial reductions by PEF treatments. E. coli ATCC 35218, a non pathogenic surrogate bacterium, exhibited higher resistance to PEF treatments than E. coli 0157:H7 and E. coli K12 in pomegranate juice. Increase of a single PEF parameter (field strength, pulse width, pulse frequency, total treatment time, or energy input) is insufficient to achieve maximum microbial reduction. Optimal PEP treatment conditions for maximum microbial reduction depend on multiple factors including PEF processing parameters, production conditions and product properties. This study demonstrates that scale-up and validation studies in a specific PEF system for specific products are very important and necessary before successful commercial application of this novel technology is possible. Abstract:Effects of PEF processing (25 kV/cm for 200 mus at 1 Hz) on enzymes and quality-related parameters of carrot juice were investigated during 28 days storage at 4degC and compared with effects of traditional heat pasteurization at 95degC for 15 sec in this paper. PEF treatment inactivated 37.35% and 52.14% of the activity of PME and PPO in carrot juice respectively, and the Activity of enzymes remained significantly constant during 28 days storage at 4degC (p > 0.05). PEF-treated carrot juice retained better turbidity than thermal-pasteurized juice, accompanying with insignificant changes in color indices (L, a, b, DeltaE and browning index) throughout storage. PEF treatment decreased the non-enzymatic browning reaction, and kept browning index almost stable throughout storage. The pH value of juice had no significant change after PEF treatment, and remained almost constant during storage time. PEF treatment caused an 8% decrease of the carotene content, and another 6.2% reduction of carotene content was observed after stored at 4degC for 28 days.

Abstract:Effects of pulsed electric fields (PEF) treatment (0–547μs and 0–40kV/cm) on physicochemical properties of soybean protein isolates (SPI) were studied. Solubility, surface free sulfhydryls (SHF) and hydrophobicity of SPI dispersions (20mg/ml) increased with the increment of the PEF strength and treatment time at constant pulse width 2μs, pulse frequency 500 pulse per second (pps) and sample flow rate (1ml/s). When the PEF strength and treatment time were above 30kV/cm and 288μs, solubility, surface SHF, and hydrophobicity of SPI decreased due to denaturation and aggregation of SPI by hydrophobic interactions and disulfide bonds. Size-exclusion chromatography and laser light scattering analyses demonstrated further that stronger PEF treatment-induced dissociation, denaturation and reaggregation of SPI. Circular dichroism analysis showed that PEF treatment did not produce significant secondary structure changes of SPI. These results suggested that controlled PEF could be applied to process liquid food including soybean protein ingredient and to modify their structure and function in order to get desired products.

Abstract:The aim of this study was to evaluate the effects of ultra-high pressure (UHP, 600 MPa/2 min), thermal pasteurization (TP, 95 °C/1 min) and ultra-high temperature (UHT, 115 °C/5 s) sterilization on the color, sensory evaluation, microorganisms, physicochemical characteristics and nutritional components of freshly-squeezed lettuce juice (FLJ). Results showed that three sterilization methods demonstrated desirable inactivation effects on total aerobic bacteria, yeast and mold, and there were no significant changes in the main nutritional components, including ash, protein, carbohydrate and total dietary fiber. However, UHT and TP significantly affected the color of FLJ from bright green to light brown and made chlorophyll, β-carotene and vitamins (VE, VC, VK1, VB6, VB12, and folic acid) contents markedly decreased. By contrast, UHP maintained the original color, fresh-like sensory qualities, vitamins, and carotene of FLJ to the greatest extent. Our results provide a promising application of UHP in the large-scale processing of FLJ.

Abstract:ABSTRACT The antimicrobial activity and preservative effect of Ceylon spinach (Basella alba L.) seed protein extract on sturgeon fillets stored at 4°C were investigated in this study. The results showed that Ceylon spinach seed protein extract (CSSPE) had inhibitory effect on 33 microorganisms, including 1 fungus and 32 bacteria (25 gram-negative bacteria and 7 gram-positive bacteria). In addition, CSSPE inhibited bacterial growth (total viable counts, Pseudomonas, Aeromonas and psychrotrophs), suppressed ATP degradation and retained good-quality characteristic of sturgeon fillets stored at 4°C. The shelf life of 30 mg/mL CSSPE treated fillets was around 7 days according to the results of total viable count, which extended around 2 days compared with that of the control group. The stability of CSSPE was also evaluated, and the results showed that CSSPE had excellent stability under UV irradiation, monovalent metal ions, surfactant and protease treatment. CSSPE could maintain high antimicrobial activity within the range of pH 2–8 and 12–13 and its thermal denaturation temperature was 70°C. The results suggested that CSSPE had the potential to be developed as a preservative for aquatic products. Graphical Abstract

Abstract:The aim of this study was to produce concentrated orange juice (OJ) powders by the application of two new techniques, namely micro-wet milling (MWM) and vacuum spray drying (VSD) process. MWM produced OJ with smaller particle sizes (55.0±1.05μm) than conventional methods and increased the nutritional and antioxidant properties of the concentrated juice over the commercial OJ. VSD process was conducted at low temperature (40–60°C) using superheated steam (200°C) as a heating medium and maltodextrin (13DE) as a carrier. The effects of VSD on physicochemical and antioxidant properties of MWM OJ powders produced with four different weight ratios of juice solids to maltodextrin solids; 60:40, 50:50, 40:60 and 30:70 were investigated. The obtained powders were analyzed for moisture content, water activity, bulk density, tapped density, particle density, porosity, and particle size and distributions and microstructure of the particles. The quality in respect to the physical properties of OJ powders was improved except color parameter with increases of maltodextrin solids. The VSD powders retained a higher amount of ascorbic acid, total phenolic content (TPC) and total flavonoid content (TFC) than spray drying. MWM orange powder with overall good quality in terms of color, yield, ascorbic acid, TFC, TPC and DPPH activity was successfully produced by VSD.

Abstract:The effect of microchip-pulsed electric field (MPEF) and high temperature short time sterilization (HTST) technologies on microbiological, physicochemical properties and flavor of freshly squeezed blueberry juice were compared. Under the MPEF treatment at 350 V, total plate count, total yeast and molds were completely inactivated, meanwhile, VitaminC, anthocyanin, total phenols, titratable acid and color had no significantly changes. During storage of 30 days at 4 °C, VitaminC decreased by 11.52% and 26.78% after MPEF and HTST treatments, all treated juices showed to be microbiologically safe, whereas MPEF can protect nutritionally related properties and color of blueberry juice samples more efficiently. Besides, relatively small changes of juice in odor, taste and volatile compounds concentration was observed by MPEF treatment. Hence, application of MPEF may be an ideal alternative treatment to HTST processing in order to ensure longer shelf life, higher functional and nutritional values of blueberry juice.

Abstract:Vegetable beverages are a convenient strategy to enhance the consumption of horticultural commodities, with the possibility of being fortified with plant by-products to increase functional quality. The main objective was to develop a new veggie beverage from broccoli stalks and carrot by-products seasoned with natural antioxidants and antimicrobial ingredients. Pasteurization, Ultrasound (US), and High Hydrostatic Pressure (HHP) and their combinations were used as processing treatments, while no treatment was used as a control (CTRL). A shelf-life study of 28 days at 4 °C was assayed. Microbial load, antioxidant capacity, and bioactive compounds were periodically measured. Non-thermal treatments have successfully preserved antioxidants (~6 mg/L ΣCarotenoids) and sulfur compounds (~1.25 g/L ΣGlucosinolates and ~5.5 mg/L sulforaphane) throughout the refrigerated storage, with a longer shelf life compared to a pasteurized beverage. Total vial count was reduced by 1.5–2 log CFU/mL at day 0 and by 6 log CFU/mL at the end of the storage in HHP treatments. Thus, the product developed in this study could help increase the daily intake of glucosinolates and carotenoids. These beverages can be a good strategy to revitalize broccoli and carrot by-products with high nutritional potential while maintaining a pleasant sensory perception for the final consumer.

Abstract:Pulsed electric field (PEF) is an innovative, non-thermal technology for food preservation with many superiorities. However, the sub-lethally injured microorganisms caused by PEF and their recovery provide serious food safety problems. Our study examined the effects of pH, temperature and natural preservatives (tea polyphenols and natamycin) on the recovery of PEF-induced, sub-lethally injured Saccharomyces cerevisiae cells, and further explored the bactericidal effects of the combined treatments of PEF with the pivotal factors in cantaloupe juice. We first found that low pH (pH 4.0), low temperature (4 °C), tea polyphenols and natamycin inhibited the recovery of injured S. cerevisiae cells. Then, the synergistic effects of PEF, combined with cold-temperature storage (4 °C), a mild treatment temperature (50 and 55 °C), tea polyphenols or natamycin, on the inactivation of S. cerevisiae in cantaloupe juice were evaluated. Our results showed that the combination of PEF and heat treatment, tea polyphenols or natamycin enhanced the inactivation of S. cerevisiae and reduced the level of sub-lethally injured cells. Moreover, PEF combined with 55 °C heat treatment or tea polyphenols was applied for cantaloupe juice. In the practical application, the two combined PEF methods displayed a comparable inactivation heat pasteurization ability, prolonged the shelf life of juice compared with PEF treatment alone, and better preserved the physicochemical properties and vitamin C levels of cantaloupe juice. These results provide valuable information to inhibit the recovery of PEF-injured microbial cells and shed light on the combination of PEF with other factors to inactivate microorganisms for better food preservation.

Abstract:Strawberry juice, which is rich in nutrients and charming flavor, is favored by consumers. To explore whether multi-mode thermosonication (MTS) can ensure the quality stability of strawberry clear juice (SCJ) during storage, the effects of microbial inhibition, enzyme activity, and physicochemical properties of SCJ pretreated by MTS were evaluated during storage at 4, 25, and 37 °C in comparison with thermal pretreatment (TP) at 90 °C for 1 min. The MTS, including dual-frequency energy-gathered ultrasound pretreatment (DEUP) and flat sweep-frequency dispersive ultrasound pretreatment (FSDUP), were conducted at 60 °C for 5 and 15 min, respectively. Results showed that the total phenols, flavonoids, anthocyanins, ascorbic acid, and DPPH free radical scavenging ability of SCJ decreased during the storage period. The control sample of SCJ was able to sage for only 7 days at 4 °C based on the microbiological quality, while the FSDUP and DEUP group extended the storage period up to 21 and 14 days, respectively. The polyphenol oxidase in SCJ pretreated by MTS did not reactivate during the storage period. The MTS remarkably (p < 0.05) reduced the color deterioration, browning degree, and nutrient degradation during the storage period. Moreover, the FSDUP group exhibited the maximum shelf life with a minimum loss of quality, demonstrating that it was the most suitable processing method for obtaining high-quality SCJ. It can be concluded that the MTS has the potential to inhibit enzymatic browning, inactivating microorganisms, preserve original quality attributes, and prolong the shelf life of SCJ.

Effects of pulsed light and sanitizer wash combination on inactivation of Escherichia coli O157:H7, microbial loads and apparent quality of spinach leaves

Abstract:The purpose of this study was to investigate the efficacy of pulsed light (PL), a new formula of sanitizer (HEN) consisting of hydrogen peroxide, EDTA and Nisin, as well as synergy of PL and HEN sanitizer (PL-HEN) wash in inactivating E. coli O157:H7 on spinach. The treatment effect on microbial loads and apparent quality during 13 days storage at 4 °C was also determined. A bacterial cocktail containing three strains of E. coli O157:H7 was used as inoculum based on their association with produce-related outbreaks. Spinach leaves were spot inoculated on surface before treating with PL (1-63J/cm2), HEN sanitizer wash (2 min) or their combinations. PL inactivation was influenced significantly at low doses. Treatment dose of 15.75 J/cm2, equivalent to 15 s intense PL treatment, was found optimal above which adverse quality effect was evident. The optimal PL dose resulted 2.7 log CFU/g reduction of E. coli O157:H7 while a rapid 2 min wash in sanitizer formulation HEN, provided comparatively low, 1.8 log CFU/g, reduction of the pathogen. Two different sequences of PL and HEN treatment combinations were tested. In PL-HEN treatment, inoculated leaves were first treated at optimal PL dose (15.75 J/cm2) followed by 2 min immersion in HEN whereas in HEN-PL treatment, leaves were first washed in HEN before PL exposure. HEN-PL treatment indicated a compound inactivation activity (4.6 logs reduction) while PL-HEN treatment indicated a strong synergistic inactivation as E. coli cells were not detectable after treatment indicating >5 log reduction. The PL-HEN treatment not only significantly reduced spoilage microbial populations on spinach but also slowed their growth during storage. Furthermore, the visual and firmness quality of spinach were not significantly affected by the PL-HEN treatment. Overall, our results demonstrate that integrated PL-HEN technology can be used to enhance microbial safety of spinach.

Abstract:This study evaluated the stability of strawberry pulp subjected to three factors, pasteurisation (pasteurised and unpasteurised), freezing method (static air and forced air) and storage time (0, 2, 4 and 6 months). Pasteurisation favoured vitamin C retention during storage but enhanced the total loss of phenolics without affecting anthocyanin levels. Freezing by forced air was more effective in retaining phenolics during the first 4 months of storage, although the freezing method did not affect the anthocyanin levels. Processing and storage reduced the levels of individual phenolics. Freezing by forced air was more effective than static air in retaining antioxidant activity of the pulp. Polyphenol oxidase and peroxidase enzyme levels were relatively stable and independent of pasteurisation, freezing and storage time. Even after 6 months of frozen storage, strawberry pulp is a significant source of nutrients and bioactive compounds and retains high antioxidant capacity independent of pasteurisation and freezing method.

Abstract:The worldwide market for vegetable and fruit juices stands as a thriving sector with projected revenues reaching to $81.4 billion by 2024 and an anticipated annual growth rate of 5.27% until 2028. Juices offer a convenient means of consuming bioactive compounds and essential nutrients crucial for human health. However, conventional thermal treatments employed in the juice and beverage industry to inactivate spoilage and pathogenic microorganisms, as well as endogenous enzymes, can lead to the degradation of bioactive compounds and vitamins. In response, non-thermal technologies have emerged as promising alternatives to traditional heat processing, with pulsed electric field (PEF) technology standing out as an innovative and sustainable choice. In this context, this comprehensive review investigated the impact of PEF on the microbiological, physicochemical, functional, nutritional, and sensory qualities of vegetable and fruit juices. PEF induces electroporation phenomena in cell membranes, resulting in reversible or irreversible changes. Consequently, a detailed examination of the effects of PEF process variables on juice properties is essential. Monitoring factors such as electric field strength, frequency, pulse width, total treatment time, and specific energy is important to ensure the production of a safe and chemically/kinetically stable product. PEF technology proves effective in microbial and enzymatic inactivation within vegetable and fruit juices, mitigating factors contributing to deterioration while maintaining the physicochemical characteristics of these products. Furthermore, PEF treatment does not compromise the content of substances with functional, nutritional, and sensory properties, such as phenolic compounds and vitamins. When compared to alternative processing methods, such as mild thermal treatments and other non-thermal technologies, PEF treatment consistently demonstrates comparable outcomes in terms of physicochemical attributes, functional properties, nutritional quality, and overall safety.
Effects of konjac glucomannan/pomegranate peel extract composite coating on the quality and nutritional properties of fresh-cut kiwifruit and green bell pepper

Abstract:The effects of an edible coating, based on konjac glucomannan (KG) incorporated with pomegranate peel extracts (PE), on the physicochemical and nutritional properties of fresh-cut kiwifruit and green bell pepper during storage were investigated. The optimal extract time (40.6 min), temperature (54.5 °C), and ultrasound power (255.5 W) with response surface method, provided a high total antioxidant activity (TAA) of (92.31 ± 1.43)%. Fresh-cut kiwifruit and green bell pepper were coated by dipping using five treatments (distilled water, ascorbic acid, KG, PE, KG + PE), packed into polymeric film and stored for 8 days at 10 °C. Distilled water treatment was used as control. KG + PE treatment resulted in the highest total soluble solid and titratable acidity in fresh-cut kiwifruit, while the maximum firmness in fresh-cut green bell pepper. The weight loss was both effectively decreased in samples treated with KG or KG + PE. All samples treated with KG + PE had significantly higher contents of chlorophyll, ascorbic acid, total phenolic and TAA than others. Moreover, the KG + PE group had the lowest counts of microorganisms in all samples. KG coating incorporated with PE was proved to be efficient in maintaining the physico-chemical and nutritional properties of fresh-cut kiwifruit and green bell pepper during low temperature storage compared with control.
