Abstract:To better understand the bacterial potential in chromium (Cr) bioremediation, a thoroughly survey was performed to investigate the diversity and importance of Cr-resistant and/or -reducing bacterial taxa by genome mining, literature survey and laboratory isolation. Genome mining among 7887 species revealed that 1877 species distributed in 20 phyla contained Cr-resistant gene chrA. Pseudomonas, Bacillus and Vibrio were the three most abundant genera. 81 species affiliated to 4 phyla were found containing Cr(VI) reductase gene. Bifidobacterium, Bordetella and Bacillus were the three most abundant genera. Importantly, genome mining identified 30 genera contained both chrA and Cr(VI) reductase genes, while literature survey showed that 43 genera capable of Cr(VI) resistant and reducing. The two sets taxa have only four genera in overlap. We conducted an isolation work in lab in which nine Cr(VI)-resistant bacteria and one Cr(VI)-reducing Arthrobacter were isolated from xixi wetland sediment. Eight isolates fell in taxa identified by genome mining or literature survey, while Lactococcus represented a novel Cr(VI)-resistant genus, indicating that there are more Cr(VI)-resistant and/or -reducing taxa in the nature to be recognized. We present here the so-far most comprehensive Cr-resistant and/or -reducing bacterial taxa, which provide important information for Cr bioremediation.

Abstract:This study reports electron microscope observations on the process of red cell sequestration and destruction in the spleen and liver of the phenylhydrazine-treated rabbit. Damaged red cells are recognized by virtue of their Heinz bodies, a morphologic manifestation of the oxidative injury which they have sustained. Sequestration, in the spleen, involves the selective accumulation of damaged cells within the vascular spaces of the Billroth cords. Erythrophagocytosis and the intracellular digestion of red cells follows sequestration. More severely injured cells may undergo intravascular hemolysis within the splenic red pulp. In the liver, however, no evidence for the intravascular sequestration of injured red cells is observed. Damaged cells are removed directly from the sinusoidal blood by erythrophagocytosis. The selectivity of spleen and liver for red cells subjected to different degrees of injury is discussed in terms of the observed differences in the vascular architecture of the two organs.
Characterization and molecular insights of a chromium-reducing bacterium Bacillus tropicus

Abstract:Environmental pollution from metal toxicity is a widespread concern. Certain bacteria hold promise for bioremediation that converts toxic chromium into a less harmful form, promoting environmental cleanup. In this study, we report the isolation and detailed characterization of a highly chromium-tolerant bacterium, Bacillus tropicus CRB14. The isolate is capable of growing on 5000 mg/l Cr (VI) in LB agar plate while on 900 mg/l Cr (VI) in LB broth with an 86.57% reduction ability within 96 hours of culture. It can also tolerate high levels of As, Cd, Co, Fe, Zn, and Pb. The plant growth-promoting potential of the isolate was demonstrated by a significant activity of nitrogen fixation, phosphate solubilization, IAA, and siderophore production. Whole-genome sequencing revealed that the isolate lacks plasmids for Cr resistance, suggesting genes reside on its chromosome. The presence of the chrA gene points towards Cr (VI) transport, while the absence of ycnD suggests alternative reduction pathways. The genome harbors features like genomic islands and CRISPR-Cas systems, potentially aiding adaptation and defense. Analysis suggests a robust metabolism, potentially involved in Cr detoxification. Notably, genes for siderophore and NRP-metallophore production were identified. Whole Genome Sequencing (WGS) data also provides the basis for molecular validation of various genes. Findings from this study highlight the potential application of Bacillus tropicus CRB14 for bioremediation while plant growth promotion can be utilized as an added benefit.

Abstract:PurposeElectroplating industries are the main sources of heavy metals, chromium, nickel, lead, zinc, cadmium and copper. The highest concentrations of chromium (VI) in the effluent cause a direct hazards to human and animals. Therefore, there is a need of an effective and affordable biotechnological solution for removal of chromium from electroplating effluent.MethodsBacterial strains were isolated from electroplating effluent to find out higher tolerant isolate against chromate. The isolate was identified by 16S rDNA sequence analysis. Absorbed chromium level of bacterium was determined by inductively coupled plasma-atomic emission spectrometer (ICP-AES), atomic absorption spectrophotometer (AAS), scanning electron microscope (SEM), transmission electron microscope (TEM) and energy dispersive X-ray analysis (EDX). Removal of metals by bacterium from the electroplating effluent eventually led to the detoxification of effluent confirmed by MTT assay. Conformational changes of functional groups of bacterial cell surface were studied through Fourier transform infrared spectroscopy.ResultsThe chromate tolerant isolate was identified as Bacillus cereus. Bacterium has potency to remove more than 75% of chromium as measured by ICP-AES and AAS. The study indicated the accumulation of chromium (VI) on bacterial cell surface which was confirmed by the SEM-EDX and TEM analysis. The biosorption of metals from the electroplating effluent eventually led to the detoxification of effluent. The increased survivability of Huh7 cells cultured with treated effluent also confirmed the detoxification as examined by MTT assay.ConclusionIsolated strain B. cereus was able to remove and detoxify chromium (VI). It would be an efficient tool of the biotechnological approach in mitigating the heavy metal pollutants.

Abstract:Tannery wastewater treatment using three isolated bacteria has been studied separately and in combination in the present study. The removal of COD, BOD and Chromium were evaluated. The high salt and Cr tolerant bacteria designated as strain-I showed maximum efficiency in COD and Cr removal from the tannery wastewater of Kolkata, India compared to other two strain-II and strain-III. In combination treatment by all three bacteria showed additive effect in waste degradation. From the results it can be said that the bacteria present in the tannery effluents and nearby soil have significant potential in treatment of leather industrial wastewater.

Abstract:Cr(VI) represents a serious threat to human health, living resources and ecological system as it is persistent, carcinogenic and toxic, whereas, Cr(III), another stable oxidation state of Cr, is less toxic and can be readily precipitated out of solution. The conventional methods of Cr(VI) removal from wastewaters comprise of chemical reduction followed by chemical precipitation. However, these methods utilize large amounts of chemicals and generate toxic sludge. This necessitates the need for devising an eco-technological strategy that would use the untapped potential of the biological world for remediation of Cr(VI) containing wastewaters. Among several viable approaches, biotransformation of Cr(VI) to relatively non-toxic Cr(III) by chromium resistant bacteria offers an economical- and environment-friendly option for its detoxification. Various studies on use of Cr(VI) tolerant viable bacterial isolates for treatment of Cr(VI) containing solutions and wastewater have been reported. Therefore, a detailed account of mechanisms and processes involved in bioreduction of Cr(VI) from solutions and wastewaters by bacterial isolates are the focus of this review article in addition to a discussion on toxicity of Cr(VI) on bacterial strains and various factors affecting Cr(VI) bioreduction.

Abstract:Two native bacterial strains, FY1 and WZ2, that showed high chromium(VI)-reducing ability were respectively isolated from electroplating and tannery effluent-contaminated sites and identified as Bacillus and Arthrobacter. The objective of the present study was to evaluate their potential for future application in soil bioremediation. The results showed that both Bacillus sp. FY1 and Arthrobacter sp. WZ2 were tolerant to 1000mg L-1 Cr(VI) and capable of reducing 78-85% and 75-82% of Cr(VI) (100-200mg L-1) within 24h, respectively. The Cr(VI) reduction rate decreased with increasing levels of Cr(VI) concentration (200-1000mg L-1). The optimum pH, temperature, and inoculum concentration for Cr(VI) reduction were found to be between pH 7.0 and 8.0; 30 and 35 degrees C; and 1x 10(8) cells ml(-1), respectively. Further evidence for the bioremediation potential of Bacillus sp. FY1 and Arthrobacter sp. WZ2 was provided by the high capacity to reduce 100, 200, and 500mg kg(-1) Cr(VI) in contaminated soil by 83-91%, 78-85%, and 71-78% within 7days, respectively. These findings demonstrated the high potential of Bacillus sp. FY1 and Arthrobacter sp. WZ2 for application in future soil bioremediation.

Abstract:Chromium, extensively used in various industries, poses significant challenges due to its environmental impact. The threat of Cr(VI) causes critical concerns in aquatic ecosystems as a consequence of the fluidity of water. The conventional approach for the treatment of effluents containing Cr(VI) is reducing Cr(VI) to low-noxious Cr(III). This research is related to a Gram positive bacterium newly isolated from tannery effluent under aerobic conditions. To characterize functional groups on the isolate, Fourier transform infrared spectroscopy was utilized. The effect of different factors on Cr(VI) bioreduction was investigated, including temperature, initial Cr(VI) concentration, acetate concentration, and Tween 80 surfactant. Under optimal conditions (37 °C and 0.90 g/L sodium acetate), the bioreduction rate of the isolate, identified as Lactococcus lactis AM99, achieved 88.0 % at 300 mg/L Cr(VI) during 72 h (p < 0.05). It was observed that Cr(VI) bioreduction was enhanced by the acetate in both the quantity and intensity, while Tween 80 had no impact on the reaction. The strain AM99 exhibited remarkable characteristics, notably a marginal decrease in growth at elevated concentrations of hexavalent chromium and an exceptional potential to reduce Cr(VI) even at very low biomass levels, surpassing any prior findings in the associated research. Furthermore, The isolate could tolerate 1400 mg/L Cr(VI) in a solid medium. These distinctive features make the isolate a promising and well-suited candidate for remediating Cr(VI)-polluted environments. Additionally, the impact of biogenic extracellular polymer produced by the strain AM99 on reduction was examined at different temperatures.

Abstract:Chromium contamination from tannery wastewater poses a significant threat to both the environment and public health, making its removal from effluents crucial before discharge into surface water. This study explores the effectiveness of activated bio-adsorbents made from cost-effective and sustainable sawmill waste Ficus benghalensis (banyan) bark and Bambusa vulgaris (bamboo) for removing Cr (III) from tannery wastewater. The easy accessibility and unique porous structure of these biomaterials make them strong candidates for Cr (III) pollution mitigation. Through carbonization and surface-functionalization via single and two-step methods, the single-step synthesized activated bio-adsorbent exhibited a superior chromium removal rate, achieving 94 % for activated carbon derived from banyan bark (ACBB), compared to 66 % for the two-step process (ACBB*). Single-step synthesis was chosen due to shorter carbonization time, practically viable, and better surface performance of materials. A similar pattern was observed for bamboo-derived activated carbon (ACB), with the one-step process yielding an 82 % removal rate, in contrast to 43 % for the two-step process (ACB*). The bio-absorbents were characterized using XRD, FTIR, BET, SEM, TGA, and zeta potential analysis. Batch process optimization reveals that ACBB and ACB demonstrate maximum chromium removal efficiencies of 92 % and 76 %, respectively, under specific pH, dosage, and temperature conditions. The adsorbents adhere to the Freundlich isotherm model, indicating multilayer adsorption, and follow pseudo-second-order kinetics, suggesting chemisorption. The maximum adsorption capacities were 278.44 mg/g for ACBB and 212.99 mg/g for ACB, which are considerably higher than the findings of previous researchers for similar materials. Thermodynamic analysis reveals that Cr (III) adsorption is spontaneous and endothermic for ACBB, while it is spontaneous and exothermic for ACB. Furthermore, the adsorbents maintain efficacy over three regeneration cycles with less than a 10 % reduction in performance. This research can facilitate efficient Cr (III) removal in industrial-scale tannery wastewater treatment due to its reusability and high adsorption performance, while also guiding future investigations. The potential of these eco-friendly bio-adsorbents for scalable, industrial tannery wastewater treatment offers a sustainable solution for chromium removal.

Abstract:AbstractContinuous discharge of untreated wastewater with high chromium contents from tanning industries causes severe environmental pollution in different aquatic systems as well as poses a variety of health risks. To reduce harmful chromium pollution in various water bodies, it is imperative to develop a method that is both affordable and environmentally benign. In the present study, chromium was successfully removed from tannery effluents using the roots of the water hyacinth (Eichhornia Crassipes). Chromium status in wastewater bodies before and after the biosorption processes was evaluated using standard spectrophotometric techniques such as UV–Vis and atomic absorption spectroscopy (AAS). The metal removal capacity of water hyacinth roots was determined by varying the biosorbent dosages, temperature, and starting chromium ion concentrations. The maximum removal of chromium was found 95% from dilute tannery wastewater and 72% of chromium was extracted directly from raw tannery effluents by using different quantities of water hyacinth. The Langmuir and Freundlich type isotherms were used to study the chromium biosorption process, and the results were found to be highly consistent with the Langmuir monolayer biosorption type isotherm. The results of kinetic investigation showed that the removal of chromium by water hyacinth roots followed a pseudo-second-order kinetic with a R2 of 0.99618. The biosorption of chromium on water hyacinth roots was exothermic and spontaneous which were confirmed from the evaluation of thermodynamic parameters such as ΔG°, ΔH°, and ΔS°. The present research findings revealed that this naturally available plant could be utilized as very promising, environmentally friendly, and cost-effective biosorbent for removing chromium species from raw as well as diluted industrial wastewater systems.

Abstract:Hexavalent chromium is resistant to degradation and harmful toxic substance to environment and community health. Physicochemical treatment methods are demanding high cost, used large quantities of chemicals & energy, release large amount of secondary toxic degradants. Mycoremediation is an eco-friendly alternative treatment method. The main objective of this research is to isolate and characterize chrome (VI) tolerant fungi from farm soil & industry effluent for mycoremedation purpose. The screening and isolation of yeast was carried out on potato dextrose agar media. PDA and broth assay test for fungi tolerance to hexavalent chromium at different concentration, temperature and pH was evaluated. Fungi species was identified biochemically using Biolog Microstation depending on carbon utilization and chemical sensitivity test. The result revealed that 10 yeast species was identified with full ID from effluent waste and farm soil based on their probability ≥ 75% and similarity index ≥ 0.5 as well as their Cr (VI) tolerance ability up to 2500 ppm. These are Yarrowia lipolytica (100%, 0.7), Cryptococcus luteolus( 100%, 0.64), Rhodotorula aurantiaca A(100%, 0.62), Ustilago maydis (100%, 0.58) Trichosporon beigelii B (100%, 0.51), Cryptococcus terreus A (100%, 0.62), Zygosaccharomyces bailii (98%, 0.65), Nadsoniafulvenscens (90%, 0.62), Schizoblastosporonstarkeyihenricii (89%, 0.56), Endomycopsis vivi (84%, 0.62 ), Rhodotorula pustula (Sim, 0.59). Two yeast species Yarrowia lipolytica and Nadsoniafulvenscens show the highest growth mean Optical density (OD) measure 0.74 ± 0.2 & 0.60 ± 0.2 respectively at pH 7 & 25 °C. The highest tolerance index (mm) was recorded by Schizoblastosporon starkey henricii 0.3067 ± 0.152. Cr (VI)-tolerance ability of these yeast strains used in the development of chromium-bioremediation technologies provide an alternative option for chromium sequestration after HPLC analysis& molecular characterization.

Abstract:Heavy metal pollution is a direct consequence of the extensive utilization of heavy metals in various industrial processes. The persistence and nondegradability of heavy metals cause them to bioaccumulate in nature, and when they come in direct contact with the pristine environment, they not only contaminate it severely but also pose dire consequences to the health of all living forms on earth, including humans. Chromium (Cr) is one of the heavy metals which has been extensively used in various industrial processes such as mining, alloy manufacturing, tanning of hides and skins, pigment production, etc. However, it is regarded as a priority pollutant due to its highly toxic, teratogenic, mutagenic, and carcinogenic nature, and the U.S. Environmental Protection Agency (EPA) also categorized it into group “A” human carcinogen. In contrast to water-soluble hexavalent chromium (Cr6+), its reduced form, trivalent chromium (Cr3+), is relatively benign and readily precipitated at environmental pH. Thus, bioremediation of Cr6+ through microorganisms including bacteria, yeast, and algae provides a promising approach to decontaminate a metal-polluted environment. This review describes an overview of the microbial reduction of Cr6+, resistance mechanism, and the antioxidant profiling exhibited by these microorganisms when exposed to Cr6+. It also describes the pilot-scale study of the successive use of bacterial, fungal, and algal strains and the subsequent use of microbially purified water for the cultivation of plant growth. Abstract:Rapid industrialization and human activities, including the unregulated use of agrochemicals, fossil fuel combustion, and sewage sludge disposal, have led to significant contamination of soils and waterways with heavy metals. These metals are non-biodegradable and persist in the environment, necessitating remediation to prevent their leaching or movement into other environmental segments and to enable their extraction. Heavy metal pollution is becoming more prevalent over time, reducing the potential for healthy food production from natural resources. These metals are usually toxic, leading to various abnormalities in plants, animals, and humans. Toxic heavy metal ions include chromium ions, particularly hexavalent ions (Cr(VI)) produced by various industrial processes like metallurgy, petroleum refractory, electroplating, mineral extraction, tanning factories, textile, paper pulp, etc. reached in to water bodies can cause detrimental effects on aquatic life, plants, microorganisms, and humans. Although chromium (Cr) (VI) is recognized as a dangerous form, it can be bioremediated by living cells to become (Cr) (III), which is less harmful. Bacterial Community Shift in Two Sectors of a Tannery Plant and its Cr (VI) Removing Potential

Abstract:The bacterial community, analyzed by both 16S rRNA gene-based clone library and cultivation methods, changed in two different sectors of a tannery plant. Ten different bacterial phyla were detected through the clone library. The bacterial community of the tanning waste consisted of Gammaproteobacteria (34.8%), Bacteroidetes (20.9%), Betaproteobacteria (12.8%), Alphaproteobacteria (7.0%), Firmicutes (7.0%), Deinococcus-Thermus (7%), Planctomycetes (4.7%), Verrucomicrobia (2.3%), Epsilonproteobacteria (1.2%), Spirochaetes (1.2%), and that of the settling tank sludge consisted of Firmicutes (38.8%), Betaproteobacteria (27.1%), Gammaproteobacteria (11.8%), Bacteroidetes (9.4%), Alphaproteobacteria (4.7%), Deltaproteobacteria (3.5%), Actinobacteria (2.4%), Synergistes (1.2%). The tanning waste accommodated higher biodiversity than the downstream primary settling tank did. Cultivation conditions seriously influenced the culturability and isolation of bacterial strains: different levels of nutrients and Cr (VI) and temperatures resulted in isolation of different bacterial strains, which showed different removal of Cr (VI) (from 1.2% to 99.1%). In addition, supplementation of nutritious LB medium enhanced the Cr (VI) culturability of bacterial isolates. Several bacteria detected as operational taxonomy units (OTUs) by culture-independent analysis were isolated, yet many OTUs did not have their relative isolates. Among the isolates, Pseudochrobactrum, Shewanella, Marinobacter, Microbacterium, Psychrobacter and Agrococcus strains showed considerable Cr (VI) removal efficiencies and good potentials for Cr (VI) pollution treatment.

Abstract:Agricultural fields are polluted with Cr (VI) due to the excessive use of industrial effluent for irrigation purposes. The current study was focused on evaluating and comparing the impact of chromate stress on wheat growth with distilled water and industrial effluent, with and without inoculation of bacteria. All chromate-resistant bacteria (UT8, UT25, AKR2, LM3, LM8, and NY2) isolated from polluted environments proficiently resisted K2CrO4 (1500 µg/ml). Staphylococcus simulans UT8 significantly produced plant-growth-promoting compounds such as 1-aminocyclopropane-1-carboxylate (ACC) deaminase (365.05 nmol α-ketobutyrate mg/protein/h), indole acetic acid (IAA) production (77.14 µg/ml), and hydrogen cyanide (HCN). Strain UT8 showed an increment in the germination of seeds (38.47%), and root and shoot length (95.19 and 119.29%, respectively) under chromate stress (350 µg/ml) as compared to uninoculated controls. Similarly, UT8 inoculated seedlings also exhibited remarkable augmentation in the plant height (49.18%), tiller number/plant (87.96%), and grain yield/pot (108.96%) under chromate as compared to inoculum free treatment (control). Scanning electron micrographs of S. simulans UT8 treated seedlings root exhibited enhanced colonization in the presence of Cr (VI). Chromate removal potential of strain UT8 and the competence to secrete active phytohormones make it among the agronomically potent microorganisms to enhance wheat growth in metal-polluted soils.

Abstract:Background In this investigation, heavy metal-resistant microorganisms from tannery effluent were isolated, characterized, and identified. Also, to study their phytotoxicity towards Macrotyloma uniflorum . The characterized bacterium, identified as Stenotrophomonas rhizophila , showed different resistance levels and sensitivity to too many antibiotics tested Methods In the current investigation, nine aerobic strains of bacteria from the tannery effluent were identified. Based on the tests looked at, such as the lowest inhibitory concentration, the relatively heavy metal analysis showed the maximum percentage of degradation of Na 2 HAsO 4 was 91.6 %, Hg was 91 %, and Cr was 78.66 %, respectively. Furthermore, the IAA generation exhibited greater tolerance and stimulated plant development. The isolated bacteria showed MARI against the evaluated antibiotics at a rate of 30 %. Significant findings On the other hand, the study found that inoculation with metal-tolerant bacteria dramatically enhanced plant growth and metal accumulation and protected plants from the damaging effects of heavy metals. The selected bacteria shield plants from the growth-inhibiting effects of heavy metals (Cd and Cr). According to these results, such metal-tolerant bacteria that promote plant growth are valuable resources exploited to create bio-inoculants that would increase the effectiveness of heavy metal removal. Abstract:Abstract Excessive industrialization and anthropogenic activities have resulted in widespread prevalence of heavy metals including hexavalent chromium in the environment. In addition to toxic properties, Cr(VI) possesses high stability and mobility, which in total makes it included in the list of priority heavy metals; thus it needs to be managed urgently. Among different methods available for remediation of Cr(VI), bioremediation is considered as one of the sustainable methods which could effectively be utilized for controlling Cr(VI) pollution. In this aspect, the treatment of Cr(VI)-containing wastewater originating from industries is noteworthy. The present review thus is an attempt to present a systematic overview dealing with studies on remediation of hexavalent chromium by using microorganisms and their application in treatment of Cr(VI)–containing industrial wastewaters. Various factors affecting the Cr(VI) removal and methods to enhance the bio-treatment are highlighted, which might act as a basis for researchers developing Cr(VI) bioremediation techniques.

New Plant Growth-Promoting, Chromium-Detoxifying Microbacterium Species Isolated From a Tannery Wastewater: Performance and Genomic Insights

Abstract:Hexavalent chromium [Cr(VI)], widely generated by tannery activities, is considered among the most toxic substances and causes a serious damage for the environment and for human health. Interestingly, some microorganisms have a potential of bioremediation of chromium-contaminated wastewaters and soils through the reduction of Cr(VI) (soluble and harmful form) into Cr(III) (stable and non-toxic form). Here, we present the full genome sequence of a novel heavy-metal-resistant, plant growth-promoting bacterium (PGPB), Microbacterium metallidurans TL13, which was isolated from a Tunisian leather industry. The strain TL13 was resistant to many heavy metals, such as chromium, copper, nickel, cobalt, and arsenic. The 50% TL13 growth inhibitory concentration (IC50) values of HgCl2, CoCl2, K2Cr2O7, CuSO4, NiCl2, FeSO4, and Na2HAsO4 are 368, 445, 676, 1,590, 1,680, 4,403, and 7,007 mg/L, respectively, with the following toxicity order: HgCl2 > CoCl2 > K2Cr2O7 > CuSO4 > NiCl2 > FeSO4 > Na2HAsO4. This new strain was also able to promote the growth of the hybrid tomato (Elika F1) under chromium metal stress. Its whole genome sequence length was estimated to be 3,587,460 bp (3,393 coding sequences) with a G + C content of 70.7%. Functional annotation of the genome of TL13 revealed the presence of open reading frames (ORFs) involved in adaptation to metal stress, such as the chromate transport protein, cobalt–zinc–cadmium resistance protein, copper resistance protein, copper responsive transcriptional regulator, multidrug resistance transporters, arsenical resistance operon repressor, arsenate reductase, arsenic resistance protein, mercuric resistance operon regulatory protein, mercuric ion reductase, and organomercurial lyase. Moreover, genes for the production of glutathione peroxidase, catalase, superoxide dismutase, and thioredoxin reductase, which confer a higher tolerance to oxidative/metal stresses, were identified in TL13 genome. In addition, genes for heat shock tolerance, cold shock tolerance, glycine-betaine production, mineral phosphate solubilization, ammonia assimilation, siderophores, exopolysaccharides, polyketides, and lytic enzymes (cellulase, chitinase, and proteases) production that enable bacteria to survive biotic/abiotic stress and to promote plant growth and health were also revealed. Based on genome analysis and experimental approaches, strain TL13 appears to have evolved from various metabolic strategies and could play a role in ensuring sustainable environmental and agricultural systems.
