Abstract:Nanozymes are nanomaterials with mimicked enzymatic activity, whose catalytic activity can be designed by changing their physical parameters and chemical composition. With the development of biomedical and material science, artificially created nanozymes have high biocompatibility and can catalyze specific biochemical reactions under biological conditions, thus playing a vital role in regulating physiological activities. Under pathological conditions, natural enzymes are limited in their catalytic capacity by the varying reaction conditions. In contrast, compared to natural enzymes, nanozymes have advantages such as high stability, simplicity of modification, targeting ability, and versatility. As a result, the novel role of nanozymes in medicine, especially in tumor therapy, is gaining increasing attention. In this review, function and application of various nanozymes in the treatment of cancer are summarized. Future exploration paths of nanozymes in cancer therapies based on new insights arising from recent research are outlined.

Abstract:Nanozymes are nanoscale materials that display enzyme-like properties, which have been improved to eliminate the limitations of natural enzymes and further broaden the use of conventional artificial enzymes. In the last decade, the research and exploration of nanozymes have attracted considerable attention in the chemical and biological fields, especially in the fields of biomedicine and tumor therapy. To date, plenty of nanozymes have been developed with the single or multiple activities of natural enzymes, including peroxidase (POD), catalase (CAT), superoxide dismutase (SOD), glucose oxidase (GOx). Tumor-characteristic metabolites can be transformed into toxic substances under the catalysis of nanozymes to kill tumor cells. However, the therapeutic effects of nanozymes greatly depend on their catalytic activity, which displays a lot of differences in vitro and in vivo. Moreover, the complex tumor environment (low pH, high H2O2 and GSH concentration, hypoxia, etc.) plays an important role in affecting their catalytic activity. Besides, the uncontrollable catalysis of nanozymes may lead to the destruction of normal tissues. To solve these problems, researchers have exploited several imaging methods to monitor the reaction processes during catalysis, including optical imaging methods (fluorescence and chemiluminescence), photoacoustic imaging, and magnetic resonance imaging. In this review, we have summarized the development of tumor treatment using nanozymes in recent years, along with the current imaging tools to monitor the catalyzing activity of nanozymes. Representative examples have been elaborated on to show the current development of these imaging tools. We hope this review will provide some instructive perspectives on the development of nanozymes and promote the applications of imaging-guided tumor therapeutics.

Abstract:Abstract Since the discovery of enzyme-like activity of Fe 3 O 4 nanoparticles in 2007, nanozymes are becoming the promising substitutes for natural enzymes due to their advantages of high catalytic activity, low cost, mild reaction conditions, good stability, and suitable for large-scale production. Recently, with the cross fusion of nanomedicine and nanocatalysis, nanozyme-based theranostic strategies attract great attention, since the enzymatic reactions can be triggered in the tumor microenvironment to achieve good curative effect with substrate specificity and low side effects. Thus, various nanozymes have been developed and used for tumor therapy. In this review, more than 270 research articles are discussed systematically to present progress in the past five years. First, the discovery and development of nanozymes are summarized. Second, classification and catalytic mechanism of nanozymes are discussed. Third, activity prediction and rational design of nanozymes are focused by highlighting the methods of density functional theory, machine learning, biomimetic and chemical design. Then, synergistic theranostic strategy of nanozymes are introduced. Finally, current challenges and future prospects of nanozymes used for tumor theranostic are outlined, including selectivity, biosafety, repeatability and stability, in-depth catalytic mechanism, predicting and evaluating activities.

Abstract:Artificial nanoscale enzyme-mimics (nanozymes) are promising functional alternatives to natural enzymes and have aroused great interest due to their inherent <i>in vivo</i> stability, affordability, and high catalytic ability. Iron-based nanozymes are one of the most investigated synthetic nanomaterials with versatile enzyme-like catalytic properties and have demonstrated remarkable relevance to a variety of biomedical applications, especially biocatalytic therapy against tumor indications. Nevertheless, despite the recent advances in biology and nanotechnology, the therapeutic performance of iron-based nanozymes <i>in vivo</i> is still limited by technical issues such as low catalytic efficiency and lack of tumor specificity. In this mini review, we briefly summarized the representative studies of iron-based nanozymes, while special emphasis was placed on the current challenges and future direction regarding the therapeutic implementation of iron-based nanozymes for the development of advanced tumor therapies with improved availability and biosafety.

Abstract:Natural enzymes play an important role to support the regular life activities of the human body. However, the application conditions of natural enzymes are harsh and there are limitations in their use. As artificial enzymes, nanozymes possess the substrate specificity of natural enzymes. Due to the advantages of low cost, good stability and strong catalytic properties, nanozymes hold a wide range of applications in the fields of sensing, chemical, food and medicine. Some of the more common ones are noble metal nanozymes, metal oxide nanozymes and carbon-based nanozymes. Among them, metal oxide nanozymes have attracted much attention because of their decent fixity, exceedingly good physicochemical properties and other advantages. Today, malignant tumors pose a great danger to the human body and are a serious threat to human health. However, traditional treatments have more side effects, and finding new treatment modalities is particularly important for tumor treatment. For example, enzyme therapy can be used to catalyze reactions in the body to achieve tumor treatment. Nanozymes can exert enzymatic activity and effectively treat malignant tumors through catalysis and synergy, and have made certain progress. This paper reviews the detection and application of metal oxide nanozymes in tumor detection and treatment in recent years and provides an outlook on their future application and development.

Abstract:Nanozymes, possessing enzyme-like characteristics, have emerged as valuable nanomaterials in biomedical applications owing to their advantages over natural enzymes. Since nanozymes exhibit several vital and excellent catalysis activities to mediate multiple oxidation-reduction reactions that can further induce changes of cancer cells and tumor microenvironment, their applications in cancer theranostics have attracted widespread attention. Thus, this scientometric analysis aims to offer a comprehensive overview of the advancements and burgeoning trends in this field. The comprehensive assessment of scientometric analysis encompasses focal areas of inquiry, emergent research paradigms, and bibliometric profiles. The study analyzes the annual distribution of publications and citations, the geographical distribution across countries and institutions, categorizations by subject matter, contributions from academic journals, authorship patterns, and cited references. Drawing upon the findings of our scientometric evaluation coupled with an integrative review of pertinent contemporary literature, comprehensive analyses of this domain have been executed. First, the classification of nanozymes used in cancer diagnosis and treatment is reviewed. Succeeding that, various nanozyme-based approaches for both in vitro and in vivo cancer detection are outlined, supplemented by relevant examples. Subsequently, distinct nanozymebased treatment strategies are delineated, including catalytic activity-based strategies, such as ROS generation, GSH depletion, starvation therapy, oxygen supply, and TAE remodeling and immunogenic cell death, and nanostructure-based non-catalytic strategies, such as drug delivery, photodynamic and sonodynamic effects, photothermal and magnetic hyperthermia effects, prodrug activation, and ion imbalance. Finally, seven prevalent treatment modalities (catalytic therapy combined with chemodynamic therapy, immunotherapy, chemotherapy, radiotherapy, photothermal therapy or magnetic hyperthermia therapy, photodynamic therapy, and sonodynamic therapy) are categorized and elaborated on. In conclusion, this comprehensive review aims to inspire innovative research endeavors in related domains and accelerate progress in nanozyme-mediated cancer diagnostics and therapeutics.

Abstract:Nanozymes, which combine enzyme-like catalytic activity and the biological properties of nanomaterials, have been widely used in biomedical fields. Single-atom nanozymes (SANs) with atomically dispersed metal centers exhibit excellent biological catalytic activity due to the maximization of atomic utilization efficiency, unique metal coordination structures, and metal-support interaction, and their structure-activity relationship can also be clearly investigated. Therefore, they have become an emerging alternative to natural enzymes. This review summarizes the examples of nanocatalytic therapy based on SANs in tumor diagnosis and treatment in recent years, providing an overview of material classification, activity modulation, and therapeutic means. Next, we will delve into the therapeutic mechanism of SNAs in the tumor microenvironment and the advantages of synergistic multiple therapeutic modalities (e.g., chemodynamic therapy, sonodynamic therapy, photothermal therapy, chemotherapy, photodynamic therapy, sonothermal therapy, and gas therapy). Finally, this review proposes the main challenges and prospects for the future development of SANs in cancer diagnosis and therapy.

These properties make nanozymes highly suitable for large-scale production. In recent years, the convergence of nanomedicine and nanocatalysis has highlighted the potential of nanozymes in diagnostic and therapeutic applications, particularly in tumor therapy. Despite these advancements, the clinical translation of nanozymes remains hindered by the lack of designs tailored to specific tumor characteristics, limiting their effectiveness in targeted therapy. This review addresses the mechanisms by which nanozymes induce cell death in various tumor types and emphasizes the key design considerations needed to enhance their therapeutic potential. By identifying the challenges and opportunities in the field, this study aims to provide a foundation for future nanozyme development, ultimately contributing to more precise and effective cancer treatments.

Abstract:Malignant tumors, one of the worst-case scenarios within human health problems, are now posing an increasing threat to the well-being of the global population. At present, the treatment of malignant tumors mainly includes surgery, radiotherapy, chemotherapy, immunotherapy, etc. Radiotherapy and chemotherapy are often applied to inoperable tumors, and some other tumors after surgery as important adjuvant therapies. Nonetheless, both radiotherapy and chemotherapy have a series of side effects, such as radiation-induced lung injury, and chemotherapy-induced bone marrow suppression. In addition, the positioning accuracy of radiotherapy and chemotherapy is not assured and satisfactory, and the possibility of tumor cells not being sensitive to radiation and chemotherapy drugs is also problematic. Nanozymes are nanomaterials that display natural enzyme activities, and their applications to tumor therapy have made great progress recently. The most studied one, platinum nanozyme, has been shown to possess a significant correlation with radiotherapy sensitization of tumors as well as photodynamic therapy. However, there are still several issues that limited the usage of platinum-based nanozymes in vivo . In this review, we briefly summarize the representative studies regarding platinum nanozymes, and especially emphasize on the current challenges and the directions of future development for platinum nanozymes therapy.

Abstract:Nanozymes, as a type of nanomaterials with enzymatic catalytic activity, have demonstrated tremendous potential in cancer treatment owing to their unique biomedical properties. However, the heterogeneity of tumors and the complex tumor microenvironment pose significant challenges to the in vivo catalytic efficacy of traditional nanozymes. Drawing inspiration from natural enzymes, scientists are now using biomimetic design to build nanozymes from the ground up. This approach aims to replicate the key characteristics of natural enzymes, including active structures, catalytic processes, and the ability to adapt to the tumor environment. This achieves selective optimization of nanozyme catalytic performance and therapeutic effects. This review takes a deep dive into the use of these biomimetically designed nanozymes in cancer treatment. It explores a range of biomimetic design strategies, from structural and process mimicry to advanced functional biomimicry. A significant focus is on tweaking the nanozyme structures to boost their catalytic performance, integrating them into complex enzyme networks similar to those in biological systems, and adjusting functions like altering tumor metabolism, reshaping the tumor environment, and enhancing drug delivery. The review also covers the applications of specially designed nanozymes in pan-cancer treatment, from catalytic therapy to improved traditional methods like chemotherapy, radiotherapy, and sonodynamic therapy, specifically analyzing the anti-tumor mechanisms of different therapeutic combination systems. Through rational design, these biomimetically designed nanozymes not only deepen the understanding of the regulatory mechanisms of nanozyme structure and performance but also adapt profoundly to tumor physiology, optimizing therapeutic effects and paving new pathways for innovative cancer treatment.

Abstract:Over the past decade, there has been a growing emphasis on investigating the role of immunotherapy in cancer treatment. However, it faces challenges such as limited efficacy, a diminished response rate, and serious adverse effects. Nanozymes, a subset of nanomaterials, demonstrate boundless potential in cancer catalytic therapy for their tunable activity, enhanced stability, and cost-effectiveness. By selectively targeting the metabolic vulnerabilities of tumors, they can effectively intensify the destruction of tumor cells and promote the release of antigenic substances, thereby eliciting immune clearance responses and impeding tumor progression. Combined with other therapies, they synergistically enhance the efficacy of immunotherapy. Hence, a large number of metabolism-regulating nanozymes with synergistic immunotherapeutic effects have been developed. This review summarizes recent advancements in cancer immunotherapy facilitated by nanozymes, focusing on engineering nanozymes to potentiate antitumor immune responses by disturbing tumor metabolism and performing synergistic treatment. The challenges and prospects in this field are outlined. We aim to provide guidance for nanozyme-mediated immunotherapy and pave the way for achieving durable tumor eradication.

Abstract:Nanotechnology is a new technology for observing and manipulating atoms and molecules in the range of 0.1-100 nm. The development of nanotechnology provides a new way for biomedicine. Based on the unique physical and chemical properties of nanomaterials, the novel biosensor based on bio-nanotechnology can be used for early diagnosis and therapy of tumors. Nowadays, the application of nanomedicine is mainly focused on the following points: tissue engineering and regenerative medicine, the accurate diagnosis and real-time monitoring of the disease, novel imaging and analysis techniques, and multifunctional drug delivery system and targeting therapy. Nanotechnology will provide effective means for accurate diagnosis and treatment of cancer. This article reviews the new progress of nanotechnology in cancer diagnosis and treatment.
Multifaceted nanozymes for synergistic antitumor therapy: A review

Abstract:Although various cancer therapies have been developed in the last decade, much of their anti-tumor effects are restricted by the tumor microenvironment complexity, such as the hypoxia in photodynamic therapy and sonodynamic therapy, and the poor penetration of radiosensitizers in radiotherapy. The development of multifunctional nanozymes that combines the physiochemical properties of nanomaterials ( e.g. special optical transformation properties) and catalytic activities of enzymes (mainly including superoxide dismutase, catalase, peroxidase, and oxidase) can provide promising weapons for enhancing antitumor efficiency. Importantly, some types of nanozymes can also act as ideal nanocarriers for improving therapeutic reagent delivery, especially in tumor immunotherapy. In this review, we summarize the recent advances in multifunctional nanozymes with an emphasis on their applications in catalytic therapy combined with immunotherapy, phototherapy, sonodynamic therapy, radiotherapy, and chemotherapy. We believe this review will provide theoretical and technical support for the subsequent development of nanozyme-based antitumor reagents and inspire novel therapeutic strategies for efficient tumor therapy.

Abstract:Studies from past years have observed various enzymes that are artificial, which are issued to mimic naturally occurring enzymes based on their function and structure. The nanozymes possess nanomaterials that resemble natural enzymes and are considered an innovative class. This innovative class has achieved a brilliant response from various developments and researchers owing to this unique property. In this regard, numerous nanomaterials are inspected as natural enzyme mimics for multiple types of applications, such as imaging, water treatment, therapeutics, and sensing. Nanozymes have nanomaterial properties occurring with an inheritance that provides a single substitute and multiple platforms. Nanozymes can be controlled remotely via stimuli including heat, light, magnetic field, and ultrasound. Collectively, these all can be used to increase the therapeutic as well as diagnostic efficacies. These nanozymes have major biomedical applications including cancer therapy and diagnosis, medical diagnostics, and bio sensing. We summarized and emphasized the latest progress of nanozymes, including their biomedical mechanisms and applications involving synergistic and remote control nanozymes. Finally, we cover the challenges and limitations of further improving therapeutic applications and provide a future direction for using engineered nanozymes with enhanced biomedical and diagnostic applications.

Abstract:In recent years, with the development of nanomaterials, a slice of nanomaterials has been demonstrated to possess high catalytic activity similar to natural enzymes and counter the dilemmas including easy inactivation and low yield natural of enzymes, which are labeled as nanozymes. The catalytic activity of nanozymes could be easily regulated by size, structure, surface modification and other factors. In comparison with natural enzymes, nanozymes featured with a more stable structure, economical preparation and preservation, diversity of functions and adjustable catalytic activity, thus becoming the potentially ideal substitute for natural enzymes. Generally, the are mainly three types containing metal oxide nanozymes, noble metal nanozymes and carbon-based nanozymes, owing various applications in biomedical, energy and environmental fields. In this review, to summarize the recent representative applications of nanozymes, and potentially explore the scientific problems in this field at the same time, we are going to discuss the catalytic mechanisms of diverse nanozymes, with the emphasis on their applications in the fields of tumor therapy, anti-inflammatory and biosensing, hoping to help and guide the future development of nanozymes.

Abstract:Nanozymes, as an emerging class of enzyme mimics, have attracted much attention due to their adjustable catalytic activity, low cost, easy modification, and good stability. Researchers have made great efforts in developing and applying high-performance nanozymes. Recently, transition-metal-based nanozymes have been designed and widely developed because they possess unique photoelectric properties and high enzyme-like catalytic activities. To highlight these achievements and help researchers to understand the research status of transition-metal-based nanozymes, the development of transition-metal-based nanozymes from material characteristics to biological applications is summarized. Herein, we focus on introducing six categories of transition-metal-based nanozymes and highlight their progress in biomarker sensing and catalytic therapy for tumors. We hope that this review can guide the further development of transition-metal-based nanozymes and promote their practical applications in cancer diagnosis and treatment.

Abstract:As the next generation of artificial enzymes, nanozymes have shown unique properties compared to its natural counterparts, such as stability in harsh environment, low cost, and ease of production and modification, paving the way for its biomedical applications. Among them, tumor catalytic therapy mediated by the generation of reactive oxygen species (ROS) has made great progress mainly from the peroxidase-like activity of nanozymes. Fe3O4 nanozymes, the earliest type of nanomaterial discovered to possess peroxidase-like activity, has consequently received wide attention for tumor therapy due to its ROS generation ability and tumor cell killing ability. However, inconsistent results of cytotoxicity were observed between different reports, and some even showed the scavenging of ROS in some cases. By collectively studying these inconsistent outcomes, we raise the question whether surface modification of Fe3O4 nanozymes, either through affecting peroxidase activity or by affecting the biodistribution and intracellular fate, play an important role in its therapeutic effects. This review will go over the fundamental catalytic mechanisms of Fe3O4 nanozymes and recent advances in tumor catalytic therapy, and discuss the importance of surface modification. Employing Fe3O4 nanozymes as an example, we hope to provide an outlook on the improvement of nanozyme-based antitumor activity.

Abstract:Nanoenzymes, as a new generation of artificial enzymes with highly effective enzyme-like properties, have come into focus and have achieved great progress. Considering the enormous potential of nanoenzymes in the field of disease diagnosis and treatment, such as biological detection, anti-inflammatory, anti-oxidative damage and cancer treatments, etc., we firstly summarize the wide range of bio-applications of different nanoenzymes in these fields. In this review, we discuss the factors influencing the properties of nanoenzymes such as pH and temperature, surface modification and doping of other elements. In addition, we introduce five kinds of nanoenzymes and their bio-applications in various diseases. The current review will make researchers better understand the current state of the development of nanoenzymes and may be beneficial for new breakthroughs.

Abstract:Nowadays, nanozymes have not only been used as biosensors in the detection field, but also their application prospects in disease treatment have been explored. Numerous nanomaterials have similar catalytic activities such as peroxidase, oxidase, catalase and superoxide dismutase, and they can be used for antibacterial, anticancer and antioxidant therapy. Although there have been many studies on the application of nanozymes in the therapeutic field, the current nanozyme-based systems often lack targeting and ignore the harm to the surrounding normal tissues. Although promising, the biosafety of nanomaterials has always been the concern of researchers. To improve the treatment effect and reduce toxic and side effects, precision treatment has become the key. At present, a few studies have modified targeted molecules on nanozymes to achieve precise targeting through specific interaction with surface overexpression factors of bacteria or cells. Combined with the catalysis of nanozymes, the targeted treatment of diseases can be achieved. This review summarizes the current research of nanozyme systems in targeted antibacterial, anticancer and antioxidant applications. At the same time, the challenges and development prospects of nanozyme-based targeted therapy system are summarized. It is expected that this work will provide new ideas and new directions for the precise treatment of nanozymes.

Abstract:DNAzyme-based gene therapy has received great interest in the treatment of cancer and other diseases. Despite its considerable potential in clinical applications, the in vivo therapeutic efficacy of DNAzymes is still unsatisfactory, mainly due to challenges such as poor stability, inadequate catalytic activity, and insufficient target specificity. With the rapid development of nucleic acid chemistry and nanomedicine, substantial efforts have been dedicated in recent years to designing DNAzyme-based nanoplatforms with enhanced treatment efficacy. In addition, extensive combinations of DNAzymes with other treatment strategies have led to many synergistic therapies with improved therapeutic effects. This review focuses on the advance of DNAzyme-based nanoplatforms in cancer treatment. It first summarizes the solutions proposed in recent years to address each of the key challenges in DNAzyme-based tumor therapy, from the perspectives of DNA sequence design, the construction of organic/inorganic nanosystems, and regulation by environmental factors. Next, it reviews the different categories of combined treatment strategies using DNAzymes in cancer therapy. Finally, future trends as well as suggestions in the field are discussed in detail.
