Abstract:The wastewater from animal farms in China is threatening the quality and security of local water environment. In the laboratory scale, a traditional SBR (A/O) and a denitrifying phosphorus-accumulating organisms (DNPAO) sequencing batch reactor (SBR) in biologically removing nutrients and organic matters from swine manure were investigated in this study. When piggery wastewater was directly treated with a traditional SBR, the efficiency was low with unstable working state, the reduction of TN, TP and BOD5 reached 89.08%, 86.04% and 93.88%, respectively. While it performs a good effect in treating high density of piggery wastewater using DNPAO SBR technology, with the help of low-density aeration, two-step of influent feeding and two circulations of anaerobic-anoxic-anaerobic-anoxic (An/Ax2), the reduction of TN, TP and BOD5 reached the maximums of 93.95%, 99.26% and 99.93%, respectively. The specific operation of (An/Ax2) SBR contributed both low concentrations and limited export loads of NO3-. The temporal curves of ORP, pH and DO over time could be indicators for simultaneous removals of nitrogen and phosphorus along with biodegradation of organic matter during this specific SBR running. A pilot-scale investigation was recommended to further testify the feasibility and efficiency of this kind of SBR under the real-world condition.

Abstract:Wastewater from animal farms in China is threatening the quality and security of the local water environment. A traditional sequencing batch reactor (SBR) and a reformed SBR integrated with two-step feeding and low-intensity aeration at laboratory scale were investigated in this study for biological removal of nutrients and organic matter from swine wastewater. A low efficiency and poor stability were found under the traditional SBR, and the reduction of total nitrogen (TN), total phosphorus (TP) and biochemical oxygen demand (BOD5) reached 89.1, 86.0 and 93.9%, respectively. When the wastewater was treated with the reformed SBR under the running cycle of anaerobic/anoxicanaerobic/anoxic process, the reductions in TN, TP, and BOD5 reached a maximum of 94.0, 99.3 and 99.9%, respectively. The curves of oxidation reduction potential (ORP) and nutrients over time during operation of the reformed SBR could reflect the activity of denitrifying phosphorus-accumulating organisms (DNPAOs).

Abstract:A new multi-point inflow pre-anoxic/oxic/anaerobic/anoxic/oxic (A1/O2/A3/A4/O5) sludge-membrane coupling process and pilot plant were developed and designed to solve the problem of nitrogen and phosphorus removal of low carbon and nitrogen (C/N) ratio domestic sewage in southern China. The removal effect and transformation rule of organic matter, nitrogen, and phosphorus in the system were studied by changing the distribution ratio of multi-point influent. The average C/N ratio of the influent was 2.09 and the influent distribution ratio was 1:1. When the temperature was 16–25 °C, the average concentrations of chemical oxygen demand (COD), ammonia nitrogen (NH4+- N), total nitrogen (TN), and total phosphorus (TP) in the effluent were 21.31 (±2.65), 0.60 (±0.24), 12.76 (±1.09), and 0.34 (±0.05) mg/L, respectively, and their average removals are 87.3 (±1.2)%, 98.7 (±0.4)%, 74.1 (±1.3)%, and 88.1 (±0.4)% respectively. When the low temperature was 12–15 °C, the average removals were 78.6 (±1.1)%, 90.5 (±1.3)%, 73.7 (±1.13)%, and 86.6 (±1.7)%, respectively. Compared with the traditional anaerobic/anoxic/aerobic (A2O) process under the same conditions, the TN removal was increased by 15.4%, and the TP removal was increased by 22.2%. This system has obvious advantages in treating wastewater with low C/N ratio, thereby solving the problem wherein the effluent of biological phosphorus removal from low C/N ratio domestic sewage was difficult when it was lower than 0.5 mg/L.

Partial Nitrification and Enhanced Biological Phosphorus Removal in a Sequencing Batch Reactor Treating High-Strength Wastewater

Abstract:Complex and high levels of various pollutants in high-strength wastewaters hinder efficient and stable biological nutrient removal. In this study, the changes in pollutant removal performance and microbial community structure in a laboratory-scale anaerobic/aerobic sequencing batch reactor (SBR) treating simulated pre-fermented high-strength wastewater were investigated under different influent loading conditions. The results showed that when the influent chemical oxygen demand (COD), total nitrogen (TN), and orthophosphate (PO43--P) concentrations in the SBR increased to 983, 56, and 20 mg/L, respectively, the COD removal efficiency was maintained above 85%, the TN removal efficiency was 64.5%, and the PO43--P removal efficiency increased from 78.3% to 97.5%. Partial nitrification with simultaneous accumulation of ammonia (NH4+-N) and nitrite (NO2--N) was observed, which may be related to the effect of high influent load on ammonia- and nitrite-oxidising bacteria. The biological phosphorus removal activity was higher when propionate was used as the carbon source instead of acetate. The relative abundance of glycogen accumulating organisms (GAOs) increased significantly with the increase in organic load, while Tetrasphaera was the consistently dominant polyphosphate accumulating organism (PAO) in the reactor. Under high organic loading conditions, there was no significant PAO-GAO competition in the reactor, thus the phosphorus removal performance was not affected.
Nutrients removal from low C/N actual municipal wastewater by partial nitritation/anammox (PN/A) coupling with a step-feed anaerobic-anoxic-oxic (A/A/O) system

Abstract:In this study, a novel combined strategy was successfully established by partial nitritation/anammox (PN/A) within a step-feed A/A/O process integrated with fixed-biocarriers to treat municipal sewage for 200 days. The excellent nutrients removal performance of this system compared with national level of discharging standard were achieved: low total inorganic nitrogen (7.1 mg/L) and phosphorus (0.3 mg/L) in the effluent with the influent (51.1 and 4.2 mg/L) at C/N ratios of 3.4 ± 0.5, mainly attributed to the stable PN (oxic zone) and subsequently anammox effect (anoxic zone). Nitrogen mass balance indicated that anammox contribution in anoxic zones to nitrogen loss could be up to 42% at stable phase. Therefore, aeration and carbon cost could be greatly reduced under low DO, low C/N and aerobic hydraulic retention time (HRT) of 7.4 h condition. The low DO and anammox bacteria retention in anoxic chambers promoted the washout of NOB and combination of anammox and partial nitritation process. During long-term operation, the activity of AOB effectively maintained while that of NOB drastically reduced to 0.1 mg N / g MLSS / h resulting in high and stable nitrite accumulation ratios (about 90%). The achievement of partial nitritation was mainly due to low DO (0.4–0.5 mg/L) and effective retention of anammox bacteria even with a low temperature (14.5 °C). Notedly, anammox activity gradually increased both on the biocarriers and in the flocs while a higher anammox abundance was observed on the biocarriers (2.48%) than that in suspend flocs (0.03%). As above, this study indicated that the novel combined strategies could be applicable to mainstream anammox, and a pilot-scale reactor will be established to verify and promote the industrial application of mainstream anammox.

Abstract:Stringent wastewater discharge standards require wastewater treatment plants (WWTPs) to focus on enhancing nitrogen (N) and phosphorus (P) removal efficiency. Increasing sludge concentration by regulation of sludge retention time (SRT) would enhance wastewater treatment loads. However, phosphorus-accumulating organisms (PAOs) would be outcompeted by glycogen-accumulating organisms (GAOs) under long SRT, leading to a collapse of P removal. In this work, pilot-scale anaerobic-oxic-anoxic (AOA) and anaerobic-anoxic-oxic (AAO) systems with long SRT (30 d) were parallelly established for actual urban wastewater treatment. The results indicated that sludge reflux ratio, temperature, and C/N ratio significantly impact N and P removal performance of AOA and AAO systems with long SRT, and removal efficiency of AOA system significantly exceeded that of AAO system. AOA system with long SRT achieved the optimal performance at sludge reflux ratio of 200%, temperature of 25 °C, and C/N ratio of 8, with COD, NH4+-N, TN, and PO43--P removal ratio of 92.80 ± 2.24%, 97.38 ± 0.89%, 88.97 ± 2.47%, and 94.33 ± 3.27%, respectively. In addition, compared to AAO system, AOA system could save 23.08% of the aeration volume. This work highlighted that AOA system with long SRT included multiple coupled nitrogen and phosphorus removal pathways, such as autotrophic/heterotrophic nitrification, anoxic/oxic denitrification, endogenous denitrification, and denitrifying phosphorus removal. Among these, the synergistic effect of endogenous denitrification and denitrifying phosphorus removal driven by internal carbon sources contributed to satisfactory nitrogen and phosphorus removal efficiency in AOA system with long SRT.

Abstract:In case of no advanced treatment units and supplement of carbon sources with high dose, removal of TN remains as challenge for wastewater treatment plants. In this study, a 67.5 m 3 /day Anoxic-Oxic-Anoxic/ Sequencing Batch Fixed-biofilm and Activated Sludge Reactor (AOA-SBFASR) was developed and operated for 150 days. During the trial, removal efficiencies of COD, NH 4 + -N and TN reached 92.55 %, 98.75 %, and 75.64 %, respectively. Reconfiguration of microbial community was observed during the trial, and the abundances of nitrifying bacteria, denitrifying bacteria and refractory organic degrading bacteria were increased, indicating that AOA-SBFASR might has great potentials for enhanced nitrogen removal and refractory organics conversion and utilization. The analysis of nitrogen metabolism showed that the reduced nitrogen was oxidized through complete nitrification pathway and the nitrogen reduction was mainly performed by pathways of denitrification and dissimilatory nitrate reduction.

Abstract:Biological phosphorus removal using an anaerobic–aerobic sequencing batch reactor (SBR) in a low temperature can be difficult to remove, and aeration always accounts for nearly half of the total electricity costs at many wastewater treatment plants. In this study, a pilot-scale anaerobic–critical aerobic SBR (A–CA SBR) was developed for synthetic domestic wastewater. More importantly, the phase, whose concentration of diffused oxygen was controlled at 1.0–1.5 mg/L, was defined as a critical aerobic phase, which reduced expenses during the operation. To be specific, half of the ammonia was removed within 10 days and no NO3−–N was accumulated during the process. From the SEM and metagenome analysis, Rhodocyclus, Zooglea, Dechloromonas, and Simplicispira had the ability to remove phosphorus and NO3−–N simultaneously, which proved the existence of a potential double-layer sludge structure under an A–CA operational condition. All of the results disclose that the pilot-scale A–CA SBR is a reliable manipulation strategy for phosphorus removal under low temperatures, which can hopefully apply to practical wastewater remediation.

Abstract:A two-sludge anaerobic/anoxic/oxic + nitrification system with simultaneous nitrogen and phosphorus removal was studied for enhanced low-strength wastewater treatment. After 158 days of operation, excellent NH 4 + -N, chemical oxygen demand (COD) and PO 4 3 − -P removal (99.0 %, 90.0 % and 92.0 %, respectively) were attained under a low carbon/nitrogen ratio of 5, resulting in effluent NH 4 + -N, COD and PO 4 3 − -P concentrations of 0.3, 30.0 and 0.5 mg/L, respectively. The results demonstrate that the anaerobic/anoxic/oxic sequencing batch reactor (A 2 -SBR) and nitrification sequencing batch reactor (N-SBR) had favorable denitrifying phosphorus removal and nitrification performance, respectively. High-throughput sequencing results indicate that the phosphate-accumulating organisms Dechloromonas (1.1 %) and Tetrasphaera (1.2 %) were enriched in the A 2 -SBR , while the ammonia-oxidizing bacteria Nitrosomonas (7.8 %) and the nitrite-oxidizing bacteria Nitrospira (18.1 %) showed excellent accumulation in the N-SBR Further analysis via functional prediction revealed that denitrification is the primary pathway of nitrogen metabolism throughout the system. Overall, the system achieved low carbon and high efficiency nutrient removal.

Abstract:Previous researches have demonstrated that biological phosphorus removal from wastewater could be induced by oxic/extended-idle (O/EI) regime. In this study, an anoxic period was introduced after the aeration to realize biological nutrient removal. High nitrite accumulation ratio and polyhydroxyalkanoates biosynthesis were obtained in the aeration and biological nutrient removal could be well achieved in oxic/anoxic/extended-idle (O/A/EI) regime for the wastewater used. In addition, nitrogen and phosphorus removal performance in O/A/EI regime was compared with that in conventional anaerobic/anoxic/aerobic (A(2)/O) and O/EI processes. The results showed that O/A/EI regime exhibited higher nitrogen and phosphorus removal than A(2)/O and O/EI processes. More ammonium oxidizing bacteria and polyphosphate accumulating organisms and less glycogen accumulating organisms containing in the biomass might be the principal reason for the better nitrogen and phosphorus removal in O/A/EI regime. Furthermore, biological nutrient removal with O/A/EI regime was demonstrated with municipal wastewater. The average TN, SOP and COD removal efficiencies were 93%, 95% and 87%, respectively.
Enhanced wastewater treatment with an AnF-AAO system for improved internal carbon source utilization

Abstract:The main challenge in removing nutrients from municipal wastewater in China is the lack of available carbon sources. While hydrolysis acidification tanks can improve wastewater biodegradability by effectively utilizing internal carbon sources, high sludge concentrations are difficult to control in traditional tank variants. In this study, an innovative anaerobic filter (AnF) hydrolysis acidification reactor composed of a continuously stirred tank reactor (CSTR) and cloth media filter was designed to regulate and maintain high sludge concentrations in the hydrolysis acidifier. The reactor was used as a pretreatment unit for the anaerobic/anoxic/oxic (AAO) units and combined into an AnF-AAO system to explore the effectiveness of internal carbon source utilization in wastewater. The results indicate that as the sludge concentration in the hydrolysis acidifier increased, the hydrolysis and acidification processes became more efficient. The optimal sludge concentration was 40 g/L, which significantly increased the production of soluble chemical oxygen demand and volatile fatty acids. Above this concentration, the efficiency decreased. Compared to traditional AAO processes, the AnF-AAO system achieved superior total nitrogen and phosphorus removal with shorter hydraulic retention times and reduced sludge production by a significant amount of 35%. Due to its capacity for enhancing internal carbon source utilization, the AnF-AAO system constitutes a promising approach for sustainable urban wastewater treatment.

Abstract:PCN bacteria capable of heterotrophic-aerobic nitrogen removal was successfully applied for bioaugmented treatment of municipal wastewater in a pilot-scale SBR. At an appropriate COD/N ratio of 8, the bioaugmentation system exhibited stable and excellent carbon and nutrients removal, the averaged effluent concentrations of COD, NH4(+)-N, TN and TP were 20.6, 0.69, 14.1 and 0.40 mg/L, respectively, which could meet the first class requirement of the National Municipal Wastewater Discharge Standards of China (COD<50 mg/L, TN<15 mg/L, TP<0.5 mg/L). Clone library and real-time PCR analysis revealed that the introduced bacteria greatly improved the structure of original microbial community and facilitated their aerobic nutrients removal capacities. The proposed emerging technology was shown to be an alternative technology to establish new wastewater treatment systems and upgrade or retrofit conventional systems from secondary-level to tertiary-level.
NOB Suppression in Pilot-Scale Mainstream Nitritation-Denitritation System Coupled with MBR for Municipal Wastewater Treatment

Abstract:The high energy consumption associated with biological treatment of municipal wastewater is posing a serious impact and challenge on the current global wastewater industry and is also inevitably linked to the issue of global climate change. To tackle such an emerging situation, this study aimed to develop strategies to effectively suppress nitrite oxidizing bacteria (NOB) in pilot-scale mainstream nitritation-denitritation system coupled with MBR for municipal wastewater treatment. The results showed that stable nitrite shunt was achieved, while more than 90% of COD and NH4+-N removal were obtained via nitritation-denitritation in the pilot plant fed with real municipal wastewater. Through adjusting aeration intensity in MBR in combination with the integrated control of dissolved oxygen (DO), sludge retention time (SRT) and sludge return ratio, NOB was successfully suppressed with a nitrite accumulation rate (NAR) of more than 80%.
Advanced nutrient removal and external sludge reduction: Demonstration in a pilot-scale sequencing batch reactor

Abstract:Although the addition of excess sludge fermentation products to improve nutrient removal from sewage is cost-effective, its application has rarely been demonstrated. In this study, the external sludge was first collected and fermented under a sludge retention time of 10 days, then introduced into SBR with a 1:15 sewage ratio. The results revealed a gradual increase in the nitrite accumulation ratio to 34.7% in the SBR at the end of the oxic stage after 64 days of adding fermented sludge products. In addition, the average effluent total nitrogen and phosphorous decreased to 7.3 and 0.5 mg/L, corresponding to removal efficiencies of 86.7% and 95.5%, respectively. On the other hand, the use of the fermented sludge products as external organic carbon sources in the SBR increased the external sludge reduction ratio to 42.5%. High-throughput sequencing demonstrated that the increase in the endogenous denitrifier community, polyphosphate-accumulating organisms, and fermentation bacteria were the main factors contributing to the increase in nutrient removal and excess sludge reduction. The economic evaluation indicated that the operational cost of the pilot-scale system saves 0.011$/m3 of sewage treated. PRACTITIONER POINTS: Fermented sludge addition effectively enhanced nutrient removal in pilot-scale SBR. Average effluent TN and PO4 3- -P decreased to 7.3 and 0.5 mg/L, respectively. Highest external sludge reduction rate was 42.5% in pilot-scale reactor. Sewage treatment cost can save 0.011$/m3 under advanced nutrient removal.
Comparison of nitrogen removal efficiency and microbial characteristics of modified two-stage A/O, A 2 /O and SBR processes

Abstract:The low temperature of sewage in north China results in low performance of biological treatment at municipal wastewater treatment plants (MWTPs), especially in biological nitrogen removal. A modified two-stage A/O process with an embedded biofilm was proposed to enhance nitrogen removal. The operation performance of a pilot test was compared with an A 2 /O and SBR process at existing MWTPs to investigate the resistance to low temperature. The microbial communities for the three processes were compared based on the metagenomics results of 16sDNA high-throughput sequencing from activated sludge. The modified A/O resulted in a higher average removal of COD (90.12%) than A 2 /O (85.23%) and SBR (83.03%), especially of small-molecule organic compounds (< 500 Da) and macromolecular refractory organics (> 5 k Da); the TN removal rate of A2/O, SBR and the modified A/O was also increased from 74.47%, 70.63% and 78.46%, respectively. High-throughput sequencing revealed increased microbial diversity and an abundance of denitrifying functional bacteria was observed in the modified A/O process at low temperatures. The abundance of nitrite oxidation bacteria (NOB) including Nitrosomonas and Nitrospira , the amount was 1.76% and 2.34% in modified A/O, respectively, whereas NOB only accounted for 1.82% in A 2 /O and 1.35% in SBR.

Abstract:Acclimatization of short-cut denitrifying polyphosphate accumulating organisms (SDPAOs), metabolic mechanism, and operating parameters were analyzed to investigate the performance of the anaerobic/anoxic sequencing batch reactor (A 2 SBR) process. The high-throughput sequencing technology was employed to explore the microbial community structures of activated sludge systems. The experimental results illustrated that SDPAOs were successfully enriched with three-phase inoculation for 36 days. The removal rates of TP and NO 2 − -N were 93.22% and 91.36%, respectively, under the optimal parameters of a pH of 7.5, an SRT of 26 days, a temperature of 24 °C and a COD of 200.00 mg·L −1 using acetate as the carbon source. In the anaerobic stage, 82.20% external carbon source was converted into 88.78 mg·g −1 PHB, and the removal rate of NO 2 − -N in the anoxic stage was characterized by ΔNO 2 − -N/ΔPHB, anoxic ΔP/ΔPHB effective was 0.289, which was higher than anaerobic ΔP/ΔCOD effective of 0.203. Ignavibacterium and Povalibacter with significant phosphorus removal ability were the dominant bacterial genera. The nitrogen and phosphorus removal could be realized simultaneously in an anaerobic/anoxic sequencing batch reactor. Therefore, this study provided an important understanding of the removal of nitrogen and phosphorus from low-carbon nitrogen wastewater. GRAPHICAL

Abstract:An anaerobic/oxic/anoxic continuous plug-flow biorereactor was established to derive stable advanced nitrogen removal of oligotrophic domestic wastewater by setting a sludge dual-reflux system and a mixed liquid cross-flow system, while extending the hydraulic retention time in anoxic section. The effluent total inorganic nitrogen was 7.9 ± 2.2 mg N/L, with removal efficiency of 84 ± 3.9%. Results of nitrogen balance calculations indicated that the contribution of simultaneous nitrification and denitrification to total inorganic nitrogen loss in oxic region was 15% during stable stage, and the total inorganic nitrogen removal by endogenous-denitrification and enhanced exogenous-denitrification in the anoxic region was 39.9%. Prolongation of hydraulic retention time in anoxic segment is the critical reason for enhancing endogenous-denitrification, and cross-flow system is an important measure to improve exogenous-denitrification. This study provides new insights into bridging the gap between energy-saving and high-level nitrogen removal from municipal wastewater with low carbon to nitrogen ratios.

Abstract:To improve the efficiency of nitrogen removal from pharmaceutical wastewater, wastewater from traditional Chinese medicine was treated in an anaerobic sequencing batch reactor (ASBR) combined with a modified sequencing batch biofilm reactor (SBBR). The chemical oxygen demand (COD) and total nitrogen (TN) contents were 3,750 ± 50 mg/L and 210 ± 10 mg/L, respectively. After 99 days of start-up and domestication, the COD, NH 4 + -N, and TN contents in the effluent were 230 ± 10 mg/L, 1 ± 0.5 mg/L, and 5 ± 3 mg/L, respectively, and the removal efficiencies reached more than 93.5, 99, and 96%, respectively. Among these results, the COD removal efficiency in traditional Chinese medicine wastewater with an ASBR reached more than 85%, and the effluent and raw water were mixed to adjust the C/N ratio in the SBBR influent. The initial operation mode of the improved SBBR was anaerobic–aerobic–anoxic. When the C/N ratio in the influent was adjusted to 5, the simultaneous nitrification and denitrification (SND) in the aerobic section was gradually enhanced, and the endogenous denitrification (ED) in the anoxic section gradually decreased. In conclusion, deep denitrification of the system was achieved only through SND, and the running time of the cycle was shortened from the initial 24 to 4.6 h. High-throughput sequencing analysis showed that the relative abundances of Bacteroidetes and Proteobacteria in the system were 39.69 and 37.34%, respectively. The content of Firmicutes with denitrification in the system was also high, accounting for 5.17%. At the genus level, the bacteria with denitrification functions in the system were mainly Thauera and unidentified_Sphingobacteriales , accounting for 5.67 and 1.66% of the system, respectively. In addition, there was heterotrophic nitrification–aerobic denitrification (HN-AD) activated in the system, including Denitratisoma , Paracoccus , and Pseudomonas . The total relative abundance of these bacteria was 0.612%. Their existence may be one of the reasons for the good effect of SND in this system.

Abstract:Advanced nitrogen removal cannot be achieved through the conventional biological nitrogen removal process, which requires higher carbon sources and aeration energy. The proposal of intermittent aeration in the aerobic chambers offered an innovative approach to enhance nitrogen removal in low carbon-to-nitrogen ratio (C/N) municipal sewage, using a plug-flow reactor with anaerobic/aerobic/anoxic (AOA) process. Due to the effective utilization of internal carbon sources through the intermittent aeration, the total inorganic nitrogen removal efficiency (NRE) increased to 77.9 ± 3.2 % with the mean aerobic hydraulic retention time of only 3.2 h and a low C/N of 3.3 during the operation of 210 days. Polyhydroxyalkanoates dominated the nitrogen removal in this AOA system, accounting for 48.0 %, primarily occurring in the alternant aerobic/anoxic chambers. Moreover, the microbial community structure remained unchanged while the NRE increased to 77.9 %. This study provided an efficient and economic strategy for the continuous plug-flow AOA process.

Abstract:The nitrogen removal performance of a pilot-scale biosystem was significantly improved via partial denitrification/anammox (PD/A) in real sewage with low COD/N ratio. The modified pilot plant was designed as an anaerobic/anoxic/oxic (AAO) reactor combined with a biological aerated filter. The inoculation of biocarriers into anaerobic and anoxic zones enhanced anammox and the nitrogen removal performance. Despite a COD/N ratio of 3.1, effluent total inorganic nitrogen decreased from 17.1 to 9.8 mg N/L. The anoxic unit developed as the PD/A hotspot, which was associated with the enrichment of Ca. Brocadia (2.00%) and partial denitrification functional groups (OLB14, 13.50%; Thauera, 5.45%) in the anoxic-carrier biofilms and contributed 34.1% towards total nitrogen removal. Besides improving the PD/A process, enhanced denitrifying dephosphatation was simultaneously realized, suggesting that the integration of PD/A into this modified system is a promising approach to enhance nutrient removal of low COD/N wastewater.
