Abstract:Plant volatile emissions can recruit predators of herbivores for indirect defense and attract pollinators to aid in pollination. Although volatiles involved in defense and pollinator attraction are primarily emitted from leaves and flowers, respectively, they will co-evolve if their underlying genetic basis is intrinsically linked, due either to pleiotropy or to genetic linkage. However, direct evidence of co-evolving defense and floral traits is scarce. We characterized intraspecific variation of herbivory-induced plant volatiles (HIPVs), the key components of indirect defense against herbivores, and floral volatiles in wild tobaccoNicotiana attenuata. We found that variation of (E)-beta-ocimene and (E)-alpha-bergamotene contributed to the correlated changes in HIPVs and floral volatiles amongN. attenuatanatural accessions. Intraspecific variations of (E)-beta-ocimene and (E)-alpha-bergamotene emissions resulted from allelic variation of two genetically co-localized terpene synthase genes,NaTPS25andNaTPS38, respectively. Analyzing haplotypes ofNaTPS25andNaTPS38revealed that allelic variations ofNaTPS25andNaTPS38resulted in correlated changes of (E)-beta-ocimene and (E)-alpha-bergamotene emission in HIPVs and floral volatiles inN. attenuata. Together, these results provide evidence that pleiotropy and genetic linkage result in correlated changes in defenses and floral signals in natural populations, and the evolution of plant volatiles is probably under diffuse selection.
The Rapidly Evolving Associations among Herbivore Associated Elicitor-Induced Phytohormones in Nicotiana.

Abstract:In response to herbivore attack, plants perceive herbivore associated elicitors (HAE) and rapidly accumulate jasmonic acid (JA) and other phytohormones, which interact in complex ways, such as the crosstalk between JA and salicylic acid (SA). Although recent studies have shown that HAE-induced individual phytohormones can be highly specific among closely related species, it remains unclear how conserved and specific the relationships among HAE-induced phytohormones are. Here we analyzed the correlations among 4 different phytohormones, JA, JA-isoleucine (JA-Ile), SA, and abscisic acid (ABA) in 6 closely related Nicotiana species that were induced by 3 different HAEs. Our results showed that while no clear association between ABA and other phytohormones were found, the positive association between JA and JA-Ile is mostly conserved among closely related Nicotiana species. Interestingly, the association between JA and SA are highly variable and can be regulated by different HAEs.

Abstract:Abstract It is firmly established that plants respond to biotic and abiotic stimuli by emitting volatile organic compounds (VOCs). These VOCs provide information on the physiological status of the emitter plant and are available for detection by the whole community. In the context of plant–plant interactions, research has focused mostly on the defence-related responses of receiver plants. However, responses may span hormone signalling and both primary and secondary metabolism, and ultimately affect plant fitness. Here we present a synthesis of plant–plant interactions, focusing on the effects of VOC exposure on receiver plants. An overview of the important chemical cues, the uptake and conversion of VOCs, and the adsorption of VOCs to plant surfaces is presented. This is followed by a review of the substantial VOC-induced changes to receiver plants affecting both primary and secondary metabolism and influencing plant growth and reproduction. Further research should consider whole-plant responses for the effective evaluation of the mechanisms and fitness consequences of exposure of the receiver plant to VOCs.

Abstract:Plants emit volatile compounds that can act as a communication method to insects, neighboring plants and pathogens. Plants respond to leaf and root damage by herbivores and pathogens by emitting these compounds. The volatile compounds can deter the herbivores or pathogens directly or indirectly by attracting their natural enemies to kill them. The simultaneous damage of plants by herbivores and pathogens can influence plant defense. The induced plant volatiles can also make neighboring plants ready for defense or induce defense in parts distant from the damaged area of the same plant. Belowground root herbivory can alter the defense response to aboveground leaf herbivory. In addition, most plants normally emit volatile compounds from their flowers that directly attract foraging mutualistic insects for nectar, which in turn perform the very important function of pollination for subsequent reproduction. The volatile compounds emitted from the floral and vegetative parts of plants belong to three main classes of compounds: terpenoids, phenylpropanoids/benzenoids, and C6-aldehydes (green-leaf volatiles). The volatile phytohormones methyl salicylate and methyl jasmonate serve as important signaling molecules for communication purposes, and interact with each other to optimize the plant defense response. Here we discuss and integrate the current knowledge on all types of communication between plants and insects, neighboring plants and pathogens that are mediated through plant volatiles.

Abstract:Abstract Plants enhance their growth in the presence of particular soil bacteria due to volatile compounds affecting the homeostasis of plant growth hormones. However, the mechanisms of volatile compound signaling and plant perception has been unclear. This study identifies the bioactive volatile 3-octanone as a plant growth stimulating volatile, constitutively emitted by the soil bacterium Streptomyces coelicolor grown on a rich medium. When 3-octanone is applied to developing Arabidopsis thaliana seedlings, a family-wide induction of the Kelch-repeat F-box genes known as KISS ME DEADLY ( KMD ) subsequently alters auxin/cytokinin homeostasis to promote the growth of lateral roots and inhibit the primary root. Loss of function of the KMD family or other alterations of auxin/cytokinin homeostasis suppresses the volatile-induced growth response. This reveals a function of KMD s in the pathway of microbial volatile perception and plant growth responses. Significance Statement Volatiles from soil microbes are profound stimulators of plant growth. This work identifies for the first time a plant hormone signaling regulator, the gene family KISS ME DEADLY ( KMD ), to be an early essential step in plant growth promotion by a soil bacterial volatile, 3-octanone. The KMD -regulated gene network alters the tissue sensitivity balance for the growth hormones auxin and cytokinin, modifying root growth rate and architecture. Previously, the Kelch repeat F-box gene family of KMD s have been shown to be important down-regulators of both positive cytokinin signaling and phenylpropanoid biosynthesis, but upstream cues were unknown. This report places the KMD family regulation of plant growth and defense into its biotic context.

Abstract:<span>One of the most important challenges for individual plants is coexistence with their neighbors. To compensate for their sessile lifestyle, plants developed complex and sophisticated chemical systems of communication among each other. Site-specific biotic and abiotic factors constantly alter the physiological activity of plants, which causes them to release various secondary metabolites in their environments. Volatile organic compounds (VOCs) are the most common cues that reflect a plant's current physiological status. In this sense, the identity of its immediate neighbors may have the greatest impact for a plant, as they share the same available resources. Plants constantly monitor and respond to these cues with great sensitivity and discrimination, resulting in specific changes in their growth pattern and adjusting their physiology, morphology, and phenotype accordingly. Those typical competition responses in receivers may increase their fitness as they can be elicited even before the competition takes place. Plant–plant interactions are dynamic and complex as they can include many different and important surrounding cues. A major challenge for all individual plants is detecting and actively responding only to "true" cues that point to real upcoming threat. Such selective responses to highly specific cues embedded in volatile bouquets are of great ecological importance in understanding plant–plant interactions. We have reviewed recent research on the role of VOCs in complex plant–plant interactions in plant-cross kingdom and highlighted their influence on organisms at higher trophic levels.</span>

Abstract:SummaryThe ability to predict future risks is essential for many organisms, including plants. Plants can gather information about potential future herbivory by detecting volatiles that are emitted by herbivore‐attacked neighbors. Several individual volatiles have been identified as active danger cues. Recent work has also shown that plants may integrate multiple volatiles into their defense responses. Here, I discuss how the integration of multiple volatiles can increase the capacity of plants to predict future herbivore attack. I propose that integration of multiple volatile cues does not occur at the perception stage, but may through downstream early defense signaling and then be further consolidated by hormonal crosstalk. Exploring plant volatile cue integration can facilitate our understanding and utilization of chemical information transfer.

Abstract:Plants communicate with insects and other organisms through the release of volatile organic compounds (VOCs). Using Boolean operators, we retrieved 1093 articles from the Web of Science and Scopus databases, selecting 406 for detailed analysis, with approximately 50% focusing on herbivore-induced plant volatiles (HIPVs). This review examines the roles of VOCs in direct and indirect plant defense mechanisms and their influence on complex communication networks within ecosystems. Our research reveals significant functions of VOCs in four principal areas: activating insect antennae, attracting adult insects, attracting female insects, and attracting natural enemies. Terpenoids like α-pinene and β-myrcene significantly alter pest behavior by attracting natural enemies. β-ocimene and β-caryophyllene are crucial in regulating aboveground and belowground interactions. We emphasize the potential applications of VOCs in agriculture for developing novel pest control strategies and enhancing crop resilience. Additionally, we identify research gaps and propose new directions, stressing the importance of comparative studies across ecosystems and long-term observational research to better understand VOCs dynamics. In conclusion, we provide insights into the multifunctionality of VOCs in natural ecosystems, their potential for future research and applications, and their role in advancing sustainable agricultural and ecological practices, contributing to a deeper understanding of their mechanisms and ecological functions.

Abstract:Plants release distinct blends of herbivore-induced plant volatiles (HIPVs) upon herbivore attack. HIPVs have long been known to influence the behavior of herbivores and natural enemies. In addition, HIPVs can act as physiological regulators that induce or prime plant defenses. Recent work indicates that the regulatory capacity of HIPVs may extend to herbivore immunity: herbivores that are exposed to HIPVs can become more resistant or susceptible to parasitoids and pathogens. While the mechanisms of HIPV-mediated plant defense regulation are being unraveled, the mechanisms underlying the regulation of herbivore immunity are unclear. Evidence so far suggests a high degree of context dependency. Here, we review the mechanisms by which HIPVs regulate plant defense and herbivore immunity. We address major gaps of knowledge and discuss directions for future mechanistic research to facilitate efforts to use the regulatory capacity of HIPVs for the biological control of insect pests.

Abstract:Petunias have trumpet-shaped flowers with fused petals that form a long corolla tube and an expanded corolla limb at the front. Flower buds are elongated before anthesis and harbour a similarly elongated immature pistil. Upon anthesis, the corolla limb opens and the stigma matures. Terpene synthase 1 (TPS1) is expressed in the corolla tube and synthesizes sesquiterpenes that are required for stigma maturation after anthesis. This suggests that a volatile signal emerges from the tubes, accumulates in flower buds and is perceived by developing stigmas upon anthesis. However, a receptor for such a signal had not yet been identified. To identify such a receptor, these researchers performed RNA sequencing with tps1 mutant and wild-type stigmas before and after anthesis. Only expression of PhKAI2ia — one of the four KAI2 paralogues in petunia — was found to be increased in tps1 stigmas. Knockdown of PhKAI2ia by RNA interference mimics the small stigma and reduced seed number phenotypes of tps1 mutants, while — on the other hand — terpenoid emission is not affected. Gas phase complementation assays showed that (−)-germacrene D (but not its enantiomer, (+)-germacrene D) can complement the defects of stigmas in tps1 -mutant tubes. Limited proteolysis mass spectrometry showed conformational changes of PhKAI2ia when exposed to (−)-germacrene D (but not to its enantiomer) and displacement assays showed that hydrolysis activity of PhKAI2ia is inhibited in a (−)-germacrene-D dose-dependent way. In the presence of (−)-germacrene D (but not its enantiomer), PhKAI2ia can interact with PhMAX2; this affects PhSMAX1 degradation, which suggests that signalling components that are downstream of PhKAI2ia are related to the canonical karrikin and strigolactone signalling pathways.

Abstract:Plant volatile organic compounds are the most abundant and structurally diverse plant secondary metabolites. They play a key role in plant lifespan via direct and indirect plant defenses, attracting pollinators, and mediating various interactions between plants and their environment. The ecological diversity and context‐dependence of plant–plant communication driven by volatiles are crucial elements that influence plant performance in different habitats. Plant volatiles are also valued for their multiple applications in the food, flavor, pharmaceutical, and cosmetics industries. In the current review, we summarize recent advances that have elucidated the functions of plant volatile organic compounds as mediators of plant interaction at community and individual levels, highlighting the complexities of plant receiver feedback to various signals and cues. This review emphasizes volatile terpenoids, the most abundant class of plant volatile organic compounds, highlighting their role in plant adaptability to global climate change and stress‐response pathways that are integral to plant growth and survival. Abstract:Field studies have shown that plants growing next to herbivore-infested plants acquire higher resistance to herbivore damage. This increased resistance is partly due to regulation of plant gene expression by volatile organic compounds (VOCs) released by plants that sense environmental challenges such as herbivores. The molecular basis for VOC sensing in plants, however, is poorly understood. Here, we report the identification of TOPLESS-like proteins (TPLs) that have VOC-binding activity and are involved in VOC sensing in tobacco. While screening for volatiles that induce stress-responsive gene expression in tobacco BY-2 cells and tobacco plants, we found that some sesquiterpenes induce the expression of stress-responsive genes. These results provided evidence that plants sense these VOCs and motivated us to analyze the mechanisms underlying volatile sensing using tobacco as a model system. Using a pulldown assay with caryophyllene derivative-linked beads, we identified TPLs as transcriptional co-repressors that bind volatile caryophyllene analogs. Overexpression of TPLs in cultured BY-2 cells or tobacco leaves reduced caryophyllene-induced gene expression, indicating that TPLs are involved in the responses to caryophyllene analogs in tobacco. We propose that unlike animals, which use membrane receptors for sensing odorants, a transcriptional co-repressor plays a role in sensing and mediating VOC signals in plant cells.

Abstract:Clinical isolates of Neisseria gonorrhoeae were tested for growth inhibition by various quinolones. One acrosoxacin-resistant isolate was also resistant to several fluorinated quinolones with 7-heterocyclic substitution, but, inhibitory concentrations were not elevated for quinolones substituted with a methyl group in the corresponding position (nalidixic acid:position 7, S-25930:position 8) or unsubstituted (flumequine:position 8). Isolates resistant to flumequine demonstrated elevated inhibitory concentrations for all quinolones examined.

Abstract:Abstract Plants respond to environmental stimuli via the release of volatile organic compounds (VOCs), and neighboring plants constantly monitor and respond to these VOCs with great sensitivity and discrimination. This sensing can trigger increased plant fitness and reduce future plant damage through the priming of their own defenses. The defense mechanism in neighboring plants can either be induced by activation of the regulatory or transcriptional machienery, or it can be delayed by the absorbing and storing of VOCs for the generation of an appropriate response later. Despite much research, many key questions remain on the role of VOCs in interplant communication and plant fitness. Here, we reviewed recent research on the VOCs induced by biotic (i.e., insects and pathogen) and abiotic (i.e., cold, drought and salt) stresses, elucidated the biosynthesis of stress-induced VOCs in tea plants. Our focus was on the role of stress-induced VOCs in complex ecological environments. Particularly, the roles of VOCs under abiotic stress were highlighted. Finally, we discuss pertinent questions and future research directions for advancing our understanding of plant interactions via VOCs.

Sustained defense response via volatile signaling and its epigenetic transcriptional regulation

Abstract:Plants perceive volatiles emitted from herbivore-damaged neighboring plants to urgently adapt or prime their defense responses to prepare for forthcoming herbivores. Mechanistically, these volatiles can induce epigenetic regulation based on histone modifications that alter the transcriptional status of defense genes, but little is known about the underlying mechanisms. To understand the roles of such epigenetic regulation of plant volatile signaling, we explored the response of Arabidopsis (Arabidopsis thaliana) plants to the volatile β-ocimene. Defense traits of Arabidopsis plants toward larvae of Spodoptera litura were induced in response to β-ocimene, through enriched histone acetylation and elevated transcriptional levels of defense gene regulators, including ethylene response factor genes (ERF8 and ERF104) in leaves. The enhanced defense ability of the plants was maintained for 5 d but not over 10 d after exposure to β-ocimene, and this coincided with elevated expression of those ERFs in their leaves. An array of histone acetyltransferases, including HAC1, HAC5, and HAM1, were responsible for the induction and maintenance of the anti-herbivore property. HDA6, a histone deacetylase, played a role in the reverse histone remodeling. Collectively, our findings illuminate the role of epigenetic regulation in plant volatile signaling.
Plant Host Finding by Parasitic Plants A New Perspective on Plant to Plant Communication

Abstract:Plants release airborne chemicals that can convey ecologically relevant information to other organisms. These plant volatiles are known to mediate a large array of, often complex, interactions between plants and insects. It has been suggested that plant volatiles may have similar importance in mediating interactions among plant species, but there are few well-documented examples of plant-to-plant communication via volatiles, and the ecological significance of such interactions has been much debated. To date, nearly all studies of volatile-mediated interactions among plant species have focused on the reception of herbivore-induced volatiles by neighboring plants. We recently documented volatile effects in another system, demonstrating that the parasitic plant Cuscuta pentagona uses volatile cues to locate its hosts. This finding may broaden the discussion regarding plant-to-plant communication, and suggests that new classes of volatile-meditated interactions among plant species await discovery.

Abstract:Plants monitor multiple environmental cues, such as light and temperature, to ensure they germinate at the right time and place. Some specialist plants, like ephemeral fire-following weeds and root parasitic plants, germinate primarily in response to small molecules found in specific environments. Although these species come from distinct clades, they use the same HYPOSENSITIVE TO LIGHT/KARRIKIN INSENSITIVE 2 (HTL/KAI2) signalling pathway, to perceive different small molecules suggesting convergent evolution on this pathway. Here, we show that HTL/KAI2 signalling in Arabidopsis thaliana bypasses the light requirement for germination. The HTL/KAI2 downstream component, SUPPRESSOR OF MAX2 1 (SMAX1) accumulates in the dark and is necessary for PHYTOCHROME INTERACTING FACTOR 1/PHYTOCHROME INTERACTING FACTOR 3-LIKE 5 (PIF1/PIL5) to regulate hormone response pathways conducive to germination. The interaction of HTL/KAI2 and light signalling may help to explain how specialist plants like ephemeral and parasitic weeds evolved their germination behaviour in response to specific environments. Specialist plants have convergently evolved to use the same family of receptors to perceive different small molecules in lieu of necessary germination cues, like light. Although light is necessary for the germination of many generalist plants such as the model organism Arabidopsis thaliana , we show that activating the conserved receptor in Arabidopsis promotes germination in the absence of light. We show that the downstream component of the AtKAI2 signalling pathway is stable in the dark which explains its ability to negatively regulate germination. Additionally, we show that activating the receptor in the dark can modulate the gene expression profile by regulating key components of the light signalling pathway. Our study highlights how specialist plants may recycle components of conserved signaling pathways to thrive in new environments.

Fungal volatile organic compounds: mechanisms involved in their sensing and dynamic communication with plants

Abstract:Microbial volatile organic compounds (MVOCs) are mixtures of gas-phase hydrophobic carbon-based molecules produced by microorganisms such as bacteria and fungi. They can act as airborne signals sensed by plants being crucial players in triggering signaling cascades influencing their secondary metabolism, development, and growth. The role of fungal volatile organic compounds (FVOCs) from beneficial or detrimental species to influence the physiology and priming effect of plants has been well studied. However, the plants mechanisms to discern between FVOCs from friend or foe remains significantly understudied. Under this outlook, we present an overview of the VOCs produced by plant-associate fungal species, with a particular focus on the challenges faced in VOCs research: i ) understanding how plants could perceive FVOCs, ii ) investigating the differential responses of plants to VOCs from beneficial or detrimental fungal strains, and finally, iii ) exploring practical aspects related to the collection of VOCs and their eco-friendly application in agriculture.
