Abstract:OBJECTIVE:To investigate the change of behavior, as well as the plasticity of somatosensory cortex after whisker trimming.METHODS:SD rats were divided into 4 groups. Group A is the normal control group; group B: bilateral vibrissotomy on the second postnatal day; group C: unilateral right vibrissotomy on the second postnatal day; group D: right unilateral whisker trimmed during 1-5 days after birth, and leave untreated after the 5th postnatal day. Their body weight, length of the left D2 whiskers was measured on the 30th postnatal day. At the same time, the changes of their behavior (including the slit-detection test, the home exploring behavior and thigmotaxis test) were also recorded on the 30th postnatal day. Cytochrome oxydase histochemistry (CO reaction)was applied to study the development and arrangement of barrel cortex.RESULTS:In the slit-detection test, control rats could find and get into the right slit very quickly. The rats in group B could get into the slit only if their noses touched the slit. The rats in group C couldn't identify the slit by right face, but if they turned their body and touched the slit with the left whiskers, they could get into the slit very quickly. The behavior of rats in group D was similar to that in group C. The time spent for finding out the right slit of the rats in group A, B, C was obviously longer than that of group A (P < 0.01, P < 0.05, P < 0.01). In the exploring behavior and thigmotaxis test, the time for left thigmotaxis, right thigmotaxis and total thigmotaxis of rats in group B was longer than that of control animals. The time for right thigmotaxis of group C was significantly shorter than that of group A (P < 0.05). Both the weight of the rats and the length of left D2 whiskers of rats in all the four groups had no significant difference. CO reaction showed that the barrels became smaller, the septum was not clear, the arrangement of the barrels was not tidy in the mice whose right whiskers were trimmed from 2-30 days after birth.CONCLUSION:Deafferentation doesn't change the body weight and length of the whiskers left. But the stimulation of whiskers is important for rodent especially in thigmotaxis and exploring behavior. Deafferentation can also induce the plastic change of barrel cortex.

Abstract:Rats were trained to orient to a visual cue presented in either visual field. An asymmetry in reaction time showed that they used one eye to control responses to both visual fields. Cortical removal contralateral to this dominant eye produced a severe and permanent response deficit. Cortical removal ipsilateral to the dominant eye caused a mild and temporary change. Thus, in the rat, there is a functional lateralization that is similar to that seen in humans.
Antioxidative activities of white rose flower extract and pharmaceutical advantages of its hexane fraction via free radical scavenging effects.

Abstract:In this study, we determined the antioxidant activities of two different solvent fractions(butanol and hexane) obtained from white Rosa rugosa flowers by employing various assays such as 2,2-diphenyl-1-picrylhydrazyl hydrate (DPPH), 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) radical scavenging activity, and nitric oxide (NO) scavenging and inhibition activity in S-nitroso-N-acetylpenicillamine (SNAP) in the RAW264.7 model. In addition, more advanced antioxidant assays were conducted, including lipid peroxidation, hydroxyl radical-mediated oxidation, DNA fragmentation, apoptosis, and cell growth. The results revealed that the hexane fraction, which contained a significant amount of polyphenols and volatile components, had excellent antioxidant potency and could scavenge free radicals of DPPH and ABTS. Interestingly, the hexane fraction inhibited lipid peroxidation to almost the same degree as a chemical antioxidant. In the NO assay, the hexane fraction effectively scavenged free radicals at all dose ranges and is expected to inhibit NO production in mammalian cells. The hexane fraction effectively prevented oxidative damage, which was induced by Cu2+/H2O2, to target proteins at lower concentrations (>1 microg x mL(-1)). The DNA fragmentation and the cell-level assays suggest that the hexane fraction may play a crucial role in inhibiting peroxynitrite and H2O2 attack. Based on the findings described in this study, the hexane fraction holds promise for use as a novel pharmaceutical antioxidant.
Handedness in Alzheimer’s disease: A systematic review

Abstract:Handedness has traditionally been employed as a proxy of brain lateralization in research. Alzheimer’s disease (AD) manifests as a neurodegenerative disorder characterized by impairments across various neuropsychological functions, including visuospatial and language, many of which exhibit lateralization in the human brain. While previous studies have investigated the relationship between AD and handedness, findings have been inconsistent. This article aims to provide an up-to-date overview of studies investigating hand preference in AD and the subtypes, specifically early- and late-onset AD. Through a synthesis of these studies, we conclude that handedness currently lacks utility as a diagnostic biomarker for AD and its subtypes, and this is further supported by the meta-analytic results based on data from over 10,000 AD patients. We emphasize the necessity for future research endeavors, particularly those leveraging advanced neuroimaging techniques to explore the role of brain asymmetry in AD
Species- and concentration-dependent differences of acetyl- and butyrylcholinesterase sensitivity to physostigmine and neostigmine

Abstract:Previous and more recent studies show that cholinesterase inhibitors (ChE-Is) are an important possibility for therapeutic intervention in Alzheimer's Disease, sepsis and other inflammatory syndromes. ChE-Is maintain high levels of acetylcholine (ACh) determining beneficial effects on the disease process. Despite numerous efforts to identify the appropriate choice of agents and dose of ChE-Is, a common protocol regarding concentration- and species-dependent differences in inhibitory potency (IC 50) of clinical relevant ChE-Is is still not available. To evaluate the in vitro sensitivity of Acetyl- and Butyrylcholinesterase (AChE, BChE), we compared the concentration-response effects of physostigmine and neostigmine on cholinesterases in whole blood from rat and human. A spectrophotometrical test system based on in vitro Ellman's reagent has been used to determine the kinetic properties of clinical relevant ChE-Is. In vitro, the enzyme activity of human AChE and BChE was inhibited in a concentration-dependent manner until a residual activity of 4-6% for AChE and 20-30% for BChE (IC 50 human AChE: 0.117 ± 0.007 μM physostigmine, 0.062 ± 0.003 μM neostigmine; IC 50 human BChE: 0.373 ± 0.089 μM neostigmine; 0.059 ± 0.012 μM physostigmine). The inhibition curve of rat BChE in contrast showed no concentration-dependency for physostigmine and neostigmine (87% residual activity even at high inhibitor concentrations). Rat AChE was inhibited in a concentration-dependent manner until a residual activity of 53%. The results suggest that cholinesterases from human and rat show marked species- and inhibitor-dependent differences in sensitivity to physostigmine and neostigmine. Knowledge of such differences may be critical in assessing the possible therapeutic effects of ChE-Is in both species and may guide researchers in the optimal design of future experiments regarding the application of ChE-Is.

Abstract:Acetylcholinesterase, butyrylcholinesterase, carboxylesterase, and paraoxonase activities in human, mouse, and rat blood were measured. The proportions of these enzymes activities differed significantly. In humans, the most significant were cholinesterase activities, while in rats and mice the contribution of carboxylesterase activity was the greatest. High arylesterase activity of paraoxonase was observed in all cases. Species-specific differences should be taken into consideration when carrying out preclinical trials on rodents for optimization of the pharmacokinetic characteristics of drugs containing complex ester groups.

Abstract:Recent studies have suggested a functional link between cortical cholinergic output and attentional task demands, whereby acetylcholine (ACh) release is regulated according to the outcome of ongoing behaviour. To explore this hypothesis we measured ACh efflux in the rat medial prefrontal cortex (mPFC) during between-session manipulations of the cognitive demands of an attentional task. Rats were trained to detect visual stimuli in a five-choice serial reaction time task (5-CSRTT) which involves sustained and divided attention. Following habituation to tethering and implantation with a microdialysis probe in the mPFC, rats were tested in the 5-CSRTT for three consecutive days, with different lengths of stimulus duration. During performance of the 5-CSRTT we measured robust, reproducible, task-related increases in ACh release in the mPFC across all sessions. Variations of the stimulus duration from the standard 0.5 s resulted in the predicted behavioural effects (reductions and increases in choice accuracy with 0.25 s and 5 s, respectively), but there was no evidence of either greater changes in ACh release in the more demanding condition or smaller changes in the less demanding condition. By contrast, in the session with 5-s stimulus duration there was a positive correlation between prefrontal cortical ACh efflux and the total number of trials completed. In summary, the present study shows that ACh efflux in the rat mPFC is increased during performance of a 5-CSRTT, but has found no evidence to support a specific relationship between cholinergic cortical output and attentional performance.
Changes in cardiovascular autonomic control induced by chronic inhibition of acetylcholinesterase during pyridostigmine or donepezil treatment of spontaneously hypertensive rats

Abstract:Chronic treatment with acetylcholinesterase inhibitors may be a promising therapeutic strategy for treatment of cardiovascular diseases. The aim of our study was to analyze the changes in blood pressure (BP) and heart rate (HR) during 14 days of treatment with two different acetylcholinesterase inhibitors – pyridostigmine (PYR) having only peripheral effects or donepezil (DON) with both peripheral and central effects. In addition, we studied their effects on the cardiovascular response to restraint stress and on sympathovagal control of HR in normotensive Wistar-Kyoto rats (WKY) and spontaneously hypertensive rats (SHR). SHR were characterized by elevated BP and increased low-frequency component of systolic BP variability (LF-SBPV), but their cardiac vagal tone and HR variability (HRV) were reduced compared with WKY. Chronic treatment with either acetylcholinesterase inhibitor decreased HR and increased HRV in both strains. PYR treatment slightly decreased BP and LF-SBPV in the dark phase of the day. Neither drug significantly altered BP response to stress, but PYR attenuated HR increase during restraint stress. Regarding sympathovagal balance, acute methylatropine administration caused a greater increase of HR in WKY than in SHR. Chronic PYR or DON treatment enhanced HRV and HR response to methylatropine (vagal tone) in WKY, whereas PYR but not DON treatment potentiated HRV and vagal tone in SHR. In conclusion, vagal tone was lower in SHR compared with WKY, but was enhanced by chronic PYR treatment in both strains. Thus, chronic peripheral, but not central, acetylcholinesterase inhibition has major effects on HR and its variability in both normotensive and hypertensive rats.

Abstract:Cholinesterase (ChE) inhibitors are under consideration to be used in the treatment of cardiovascular pathologies. A prerequisite to advancing ChE inhibitors into the clinic is their thorough characterization in the heart. The aim here was to provide a detailed analysis of cardiac ChE to understand their molecular composition, localization, and physiological functions. A battery of biochemical, microscopic, and physiological experiments was used to analyze two known ChE, acetylcholinesterase (AChE) and butyrylcholinesterase (BChE), in hearts of mutant mice lacking different ChE molecular forms. Overall, AChE activity was exceeded by BChE, while it was localized mainly in the atria and the ventricular epicardium of the heart base. AChE was anchored by ColQ in the basal lamina or by PRiMA at the plasma membrane and co-localized with the neuronal marker TUJ1. In absence of anchored AChE, heart rate was unresponsive to a ChE inhibitor. BChE, the major ChE in heart, was detected predominantly in ventricles, presumably as a precursor (soluble monomers/dimers). Mice lacking BChE were more sensitive to a ChE inhibitor. Nevertheless, the overall impact on heart physiology was subtle, showing mainly a role in cholinergic antagonism to the positive inotropic effect of β-adrenergic stimulation. Our results help to unravel the mechanisms of ChE in cardiovascular pathologies and provide a foundation to facilitate the design of a novel, more effective pharmacotherapies, which may reduce morbidity and mortality of patients with various heart diseases.

Abstract:Lesions of the basal forebrain (BF) cortical cholinergic system impair performance on a rodent five-choice visual attentional task. This study examines the effects on the same task of selective depletion of acetylcholine from the prefrontal cortex (PFC) using 192 IgG-saporin, the cholinergic immunotoxin. Rats were trained to detect brief visual stimuli, either presented unpredictably both temporally and spatially to increase attentional load, or under less demanding conditions where stimuli were temporally and spatially predictable. Following training, 192 IgG-saporin (50 ng or 100 ng/infusion) or its vehicle was infused bilaterally into the ventromedial PFC. The 100 ng lesion group exhibited post-operatively a transient increase in perseveration, specifically when the visual stimuli were temporally unpredictable. A vigilance decrement, as well as a reinstatement of perseverative responding occurred in both lesion groups under conditions of enhanced attentional load, specifically with high target frequency sustained over many trials. Lesioned subjects were also more impulsive with increased anticipatory errors. Systemic administration of the muscarinic receptor antagonist scopolamine further dissociated the groups with attentional accuracy in the 100 ng group decreasing relative to shams. These findings are consistent with an important modulatory influence of PFC function by BF cholinergic neurons, particularly during increased attentional demand.
Cholinergic modulation of auditory steady-state response in the auditory cortex of the freely moving rat

Abstract:As disturbance in auditory steady-state response (ASSR) has been consistently found in many neuropsychiatric disorders, such as autism spectrum disorder and schizophrenia, there is considerable interest in the development of translational rat models to elucidate the underlying neural and neurochemical mechanisms involved in ASSR. This is the first study to investigate the effects of the non-selective muscarinic antagonist scopolamine and the cholinesterase inhibitor donepezil (also in combination with scopolamine) on ASSR. We recorded the local field potentials through the chronic microelectrodes implanted in the auditory cortex of freely moving rat. ASSRs were recorded in response to auditory stimuli delivered over a range of frequencies (10-80 Hz) and averaged over 60 trials. We found that a single dose of scopolamine produced a temporal attenuation in response to auditory stimuli; the most attenuation occurred at 40 Hz. Time-frequency analysis revealed deficits in both power and phase-locking to 40 Hz. Donepezil augmented 40-Hz steady-state power and phase-locking. Scopolamine combined with donepezil had an enhanced effect on the phase-locking, but not power of ASSR. These changes induced by cholinergic drugs suggest an involvement of muscarinic neurotransmission in auditory processing and provide a rodent model investigating the neurochemical mechanism of neurophysiological deficits seen in patients.
The Cerebral Cortex of the Rat and Visual Attentional Function: Dissociable Effects of Mediofrontal, Cingulate, Anterior Dorsolateral, and Parietal Cortex Lesions on a Five-Choice Serial Reaction Time Task

Abstract:Dissociable effects of bilateral excitotoxic lesions of different regions of the rat neocortex, including medial prefrontal and anterior cingulate cortices, were investigated in a five-choice serial reaction time task that provides several indices of the accuracy and speed of attentional function. Whereas medial prefrontal cortical lesions impaired performance of the task as revealed by a reduction in choice accuracy, an increase in the latency to respond correctly to the visual target and enhanced perseverative responding, lesions of the anterior cingulate cortex specifically increased premature responding. By contrast, lateral frontal cortical lesions did not significantly disrupt baseline performance of the task, but rather increased the latency to respond correctly to the visual target during various behavioral manipulations, for example, when the length of the intertrial interval was varied unpredictably and during interpolation of distracting bursts of white noise. Lesions of the parietal cortex failed to disrupt any aspect of task performance investigated. These behavioral effects in the five-choice task were compared with the effect of these same lesions on acquisition and retention of a one-trial passive avoidance task. The main finding from this paradigm was that lesions of the lateral frontal cortex produced a significant disruption to the retention of passive avoidance, which stands in marked contrast to the successful retention observed by animals of the other lesion groups. In addition, this pattern of results reveals that the 'disinhibitory' effect of cingulate cortex lesions are relatively specific to the five-choice attentional task. Finally, the results of the present study are compared with the findings of previous experiments using the five-choice task, which have examined the effect of selective manipulations of the ascending noradrenergic, cholinergic, dopaminergic, and serotonergic projections. In particular, the deficits in attentional function observed following cholinergic lesions of the nucleus basalis magnocellularis appear to be attributable to cholinergic denervation of the medial frontal cortex. These results are discussed in terms of the role of parallel distributed neural systems within the neocortex that mediate continuous attentional performance in the rat.

Abstract:Acetylcholinesterase and butyrylcholinesterase are the only two known cholinesterases. Acetylcholinesterase plays an important part in cholinergic system. It terminates neurotransmission by hydrolysis of transmitter acetylcholine. The role of butyrylcholinesterase is not well understood. It is able to detoxify several compounds such as cocaine, succinylcholine, and so forth. The current review is focused on the application of cholinesterases in biorecognition. Cholinesterases are important markers in the body. Butyrylcholinesterase activity in plasma can serve as a liver function test or specific marker for sensitivity to myorelaxants or liver carcinoma. Both cholinesterases can serve as markers of poisoning by some neurotoxic compounds. Nerve agents (sarin, soman, tabun, VX), some Alzheimer disease drugs (galantamine, huperzine, donepezil, rivastigmine), pesticides (carbofuran, trichlorfon, paraoxon, malaoxon), and natural toxins (aflatoxin, pyridostigmine) can act as inhibitors of butyrylcholinesterase and/or acetylcholinesterase. Devices filled with immobilized cholinesterases can be used for the assay of the aforementioned toxins. In this review, methods for examination of cholinesterases activity in the body and in analytical devices are described. Applications, types of diagnosis, and assays are described as well.

Abstract:Responses of extracellular acetylcholine (ACh) release and blood flow in the cerebral cortex in the parietal lobe following focal electrical stimulation of the nucleus basalis of Meynert (NBM) ipsilateral to the parietal cortex were compared in healthy adult (6-8 months) and aged (27-28 months) Fischer-344 rats anesthetized with halothane. The focal electrical stimulation of the NBM produced an increase in the ACh release and blood flow in the parietal cortex in both the adult and aged rats. The increased responses of ACh and blood flow observed in the healthy, adult rats were well maintained in the aged rats.

Abstract:Previous investigations aimed at determining the role of corticopetal cholinergic afferents in cognition have relied upon human psychopharmacological studies, neuropsychological analyses of Alzheimer's patients, or psychopharmacological manipulations and excitotoxic lesions in animals. Unfortunately, each approach has its limitations. The interpretation of neuropsychological data relies upon correlations of post-mortem assessments of cholinergic degeneration that may be quite temporally distant from the time of cognitive assessment. In contrast, the use of animals allows direct manipulations of the cholinergic system and the establishment of causal relationships between acetylcholine and cognitive function but is limited by the selectivity of the toxins and drugs available to manipulate the system. The recent introduction of immunotoxins to lesion cortical cholinergic pathways with greater selectivity has allowed the effective testing of these hypotheses of cholinergic functions in cognition. Previous neuropsychological, psychopharmacological and excitotoxic lesion data are reviewed and compared to results produced using the more selective immunotoxins to provide an update to the current hypotheses of the role of corticopetal cholinergic afferents in cognitive function. Additionally, the conceptual and methodological cost and benefits of the methods of infusion used to produce lesions with these immunotoxins is assessed.
Cortical neural activity during responses to mechanical perturbation: Effects of hand preference and hand used

Abstract:Handedness, as measured by self-reported hand preference, is an important feature of human behavioral lateralization that has often been associated with hemispheric specialization. We examined the extent to which hand preference and whether the dominant hand is used or not influence the motor and neural response during voluntary unimanual corrective actions. The experimental task involved controlling a robotic manipulandum to move a cursor from a center start point to a target presented above or below the start. In some trials, a mechanical perturbation of the hand was randomly applied by the robot either consistent or against the target direction, while electroencephalography (EEG) was recorded. Twelve left-handers and ten right-handers completed the experiment. Left-handed individuals had a greater negative peak in the frontal event-related potential (ERP) than right-handed participants during the initial response phase (N150) than right-handed individuals. Furthermore, left-handed individuals showed more symmetrical ERP distributions between two hemispheres than right-handed individuals in the frontal and parietal regions during the late voluntary response phase (P390). To the best of our knowledge, this is the first evidence that demonstrates the differences in the cortical control of voluntary corrective actions between left-handers and right-handers.

Differences in home cage behavior and endocrine parametres in rats of four strains

Abstract:Objective: The majority of studies that involve behavioral testing in different rat strains is based on measuring behavioral responses to stressful situations in a novel environment. Very little is known on the spontaneous behavior in rat strains. The aim of the present study was to compare home cage behavior and basal hormone levels in two outbred rat strains, Sprague-Dawley (SD) and Wistar as well as two inbred strains, Lewis and Spontaneously Hypertensive Rats (SHR). Methods: Twenty-eight male rats from four strains of rats (n=7/strain) were used in this study. Behavior of each rat in his home cage was recorded by a video camera for 45 minutes during the dark phase of the day. The parameters considered included rearing, jumps, ambulation, grooming, feeding/drinking and no movements. Blood plasma was analyzed for aldosterone, plasma renin activity (PRA), adrenocorticotropin hormone (ACTH) and corticosterone by specific radioimmunoassays. Results: One-way ANOVA revealed significant inter-strain differences in counts of jumps and rearing. In comparison with Sprague-Dawley and Wistar rats, Lewis rats displayed significantly more jumps and rearing. Statistical analysis showed significant inter-strain differences in the levels of aldosterone and of plasma renin activity. The highest levels of aldosterone were found in Lewis rats. Plasma renin activity was significantly lower in SHR than in Sprague-Dawley rats. Correlation analysis failed to reveal any relationships between behavioral and endocrine parameters. Significant strain differences were observed also in relative weights of the spleen, adrenals and thymus. Conclusion: Plasma renin activity and basal mineralocorticoid secretion did not show parallel pattern if compared among different rat strains. Locomotor activity in the home cage, which could represent general activity to be considered in evaluating emotional responses, was highest in the Lewis rats.
Studies on the Behavior and Pathology of the Cerebellar Ataxia Rat Model

Abstract:Aim: To evaluate the cerebellar ataxia rat model which was induced by injecting kainic acid (KA) into unilateral fastigial nucleus (FN). Methods: With KA injected into the left cerebellar fastigial nucleus of rat, the ataxia model was induced. KA-treated rats were tested by balance beam test and Rota-rod test to investigate their behavioral change, then the injecting site of KA and the pathological change of cerebellums were observed under light microscope. Results: Compared with sham-operated rats, KA-treated ones performed worse in balance beam test and Rota-rod test, the scores in balance beam test and the latency on Rota-rod test decreased significantly (P<0.05), and the time to cross the balance beam increased significantly (P<0.05). The injected site was accurate and neurons in the left cerebellar fastigial nucleus were damaged after KA injected. During the following one month observation, the animals showed excellent survivability. Conclusion: The KA-treated cerebellar ataxia rat provided a suitable model for the following studies in therapies.
Histone Hyperacetylation as a Response to Global Brain Ischemia Associated with Hyperhomocysteinemia in Rats

Abstract:Epigenetic regulations play an important role in both normal and pathological conditions of an organism, and are influenced by various exogenous and endogenous factors. Hyperhomocysteinemia (hHcy), as a risk factor for several pathological conditions affecting the central nervous system, is supposed to alter the epigenetic signature of the given tissue, which therefore worsens the subsequent damage. To investigate the effect of hHcy in combination with ischemia-reperfusion injury (IRI) and histone acetylation, we used the hHcy animal model of global forebrain ischemia in rats. Cresyl violet staining showed massive neural disintegration in the M1 (primary motor cortex) region as well as in the CA1 (cornu ammonis 1) area of the hippocampus induced by IRI. Neural loss was significantly higher in the group with induced hHcy. Moreover, immunohistochemistry and Western blot analysis of the brain cortex showed prominent changes in the acetylation of histones H3 and H4, at lysine 9 and 12, respectively, as a result of IRI and induced hHcy. It seems that the differences in histone acetylation patterns in the cortical region have a preferred role in pathological processes induced by IRI associated with hHcy and could be considered in therapeutic strategies.

Abstract:The location of AChE in the brains of Polyrhachis vicina Roger by histochemistry is reported.The results exhibit that the distribution of AChE are widespread.They mainly locate in the deuterencephalon,ganglion subpharyngeale,deutocerebrum,then the medial complex,calyxes of mushroom body,optic lobes,the pedicels,and less in lobes of mushroom body.The comparision of AChE positive reaction in different grades of Polyrhachis vicina Roger display that there are fewer positive reaction of AChE in the optic lobes and olfactory lobes of worker than male′s and female′s,the distribution of AChE in the medial complex of female has stronger reaction than other′s.These results indicate that cholinergic nerves join in the transmission of olfactory sensation and olfactory sensation information in the brain of Polyrhachis vicina Roger;Acording to the clinical diagnosis criteria,the calyxes of mushroom bodies belong to the input regions of sensory information,the pedunculus and lobes are the output regions of the mushroom bodies′s feedback message.
