First Report of Neofusicoccum parvum Causing Fruit Rot on Psidium guajava in China

Abstract:Guava (Psidium guajava L.), a nutrient-rich and economically significant fruit, is extensively cultivated in southern China. In six continuous years (from 2018 to 2023), dark-purple rotted guava with sunken lesion were observed on guava trees (variety 'Zhenzhu', aged over 5 years) in Dongguan and Panyu districts, Guangdong Province. Annually, the incidence of fruit rot disease in the affected fields reached 30% to 50% and significantly reduced the yield and quality of harvest guava. The initial symptoms on the epicarp of the fruits were black, needle-like dots that rapidly spread, causing partial or complete fruit rot within two to three weeks. To identify the causative agent, six symptomatic fruits were collected from two different orchards. Samples of 0.5 cm³ were excised from the lesion margins of each fruit. These samples were surface-sterilized with 70% ethanol for 30 seconds, followed by 0.2% NaClO for 2-3 minutes, and rinsed in sterile water three times. The samples were then cultured on potato dextrose agar (PDA) at 25°C for five days. This process yielded eight fungal isolates with similar morphological. Initially, the colonies were white with dense aerial mycelium becoming dark gray after 4 to 5 days. The mycelia were septate and branched. No spores were observed on PDA. To induce spore formation, the isolates were cultured on water agar for 20 days. This process led to the production of hyaline, aseptate, ellipsoid conidia, which were thin-walled, smooth-surfaced, and measured 3.7-5.1 × 1.6-2.2 μm (n = 100). Three isolates, including at least one from each orchard (Np1, Np2, Np3), were selected for further analysis. Genomic DNA was extracted using Axygen MAG-FRAG-I-50 (Axygen Bio-Tek). The internal transcribed spacer of rDNAs (ITS), beta-tubulin (tub2), the nuclear ribosomal large subunit (LSU), and translation elongation factor 1-α (tef1-α) gene regions were amplified using the primers ITS1/4, Bt2a/Bt2b, LR5/LR0R, and EF1-728F/EF1-986R (Golzar and Burgess 2011) and sequenced. Sequence analysis using MEGA 7.0 (Kumar et al., 2018) revealed 100% similarity among the isolates. BLAST searches of the ITS, tub2, LSU, and tef1-α sequences (accession nos. MN907637, MT537938, MT528156, MT537939) showed the highest nucleotide similarities (99.24 to 100%) to Neofusicoccum parvum strains (Crous et al. 2006). A phylogenetic tree was constructed with MEGA 7.0 based on the nucleotide sequence tub2 using the maximum likelihood method. Pathogenicity tests on 10 healthy guava fruits using mycelium-inoculated and control fruits confirmed the causative agent. The inoculated fruits, maintained at 25°C under a 12-h light/dark cycle, exhibited symptoms identical to the field infections within four to seven days, while control fruits remained symptomless. The fungus, reisolated from the inoculated fruits, was morphologically identical to the original isolates, fulfilling Koch's postulates. In conclusion, based on molecular, morphological, and pathogenic analysis, N. parvum as the causal agent of the fruit rot disease on guava. Previously, N. parvum has been reported in association with fruit rot on Eriobotrya japonica and Juglans regia (Zhai et al. 2019; Chen et al. 2019). To our knowledge, this is the first report of N. parvum affecting guava in China.
Bacterial Fruit Rot of Apricot Caused by Burkholderia Cepacia in China

Abstract:An unreported disease of apricot was observed in orchards in Zhejiang province, China. Symptoms started as water soaked lesions on the fruit surface. Later, water-soaked areas developed and spread to the entire fruit, resulting in soft rot of the whole fruit. The causal organism isolated from symptomatic fruits was identified as Burkholderia cepacia based on its biochemical and physiological characteristics and confirmed by the cellular fatty acid composition and Biolog data as well as 16S rRNA gene sequence analysis. The bacterial isolates caused similar symptoms when inoculated onto fruits of apricot. In addition, European plum, Japanese plum, nectarine and kiwifruit were susceptible to the B. cepacia pathogen. However, the B. cepacia pathogen failed to cause any visible symptoms when it was inoculated onto 16 other fruits. This is the first report of a bacterial disease of apricot caused by B. cepacia in China.

In July 2018, an unknown disease occurred on 5 to 10% of 5,000 postharvest Cuiguan pear fruit (P. pyrifolia) after 15 days in our laboratory storage room (20°C, 85 to 90% relative humidity) in NanChang City (28.68° N, 115.85° E), Jiangxi Province. Initial symptoms were shallow black spots on the fruit surface that later enlarged into circular brown lesions with water-soaked margins and a lesion diameter of 20 to 40 mm. White hyphae grew on the pear lesions and, slowly, infected fruit became soft and necrotic. To isolate the suspected pathogen, small sections (5 × 3 mm) of diseased fruit were sterilized with 75% ethanol for 30 s followed by 0.1% mercuric chloride solution for 5 min, rinsed three times with sterile water, and then transferred onto potato dextrose agar (PDA) and incubated at 28°C for 5 days. Finally, five isolates were obtained and purified on PDA. The isolates produced abundant aerial mycelium, which was initially white and then became purple over time at 28°C. The average growth of mycelium was 5.4 mm per day (n = 5). On carnation leaf agar (Fisher et al. 1982), macroconidia were three to five septa, moderately curved to straight, and 28.7 to 53.2 × 3.2 to 5.3 µm in size (38.4 × 4.2 µm on average, n = 25). On PDA, the microconidia were almost straight, aseptate, and 6.3 to 11.4 × 2.1 to 4.0 µm (8.3 × 2.9 µm on average, n = 25), and no chlamydospores were observed. The morphological features were consistent with Fusarium proliferatum (Leslie and Summerell 2006). In addition, molecular phylogenetic analyses were used to define the isolate identity. The universal primers ITS1/ITS4 (White et al. 1990) and F. proliferatum-specific primers PRO1/PRO2 (Mulè et al. 2004) were used to amplify the internal transcribed spacer (ITS)-5.8S ribosomal DNA region and calmodulin gene. The PCR amplicons (accession nos. MK158221 and MK144327) aligned with GenBank showed 100% similarity with F. proliferatum (accession nos. KU891521.1 and LT841258.1). Thus, the pathogen was identified as F. proliferatum based on morphological and molecular characteristics. Pathogenicity tests were conducted by inoculating nine disease-free pear fruit after disinfesting in 75% ethanol. A prepared spore suspension (10 μl, 1.0 × 106 spores/ml) was dropped on the wounds (a depth of 1 mm with a 1-mm diameter) on the pear surface, which were made by a sterilized needle. Sterile water (10 μl) was dropped on the wounded sites of nine pear fruits as negative controls, and 10 μl of a prepared spore suspension was dropped onto nine nonwounded control pears. All fruits were incubated in a growth chamber at 30/26°C (day/night, 12-h photoperiod, 90% relative humidity). After 3 days, the first symptom of shallow black spots was observed on nine wounded inoculated pears, and after 5 days mean lesion diameter was 19.8 mm, whereas the symptom of nonwounded pears was mild. The water controls remained healthy. To complete Koch’s postulates, F. proliferatum was again isolated from infected pears. To our knowledge, this is the first report of F. proliferatum causing rot on Cuiguan pear in China and around the world. First Report of Phytophthora Nicotianae Causing Blight of Dendrobium Candidum in Zhejiang Province, China

Abstract:Dendrobium candidum is a traditional Chinese medicinal herb, used raw or processed for health-care products in recent years. Due to the high economic value, there is huge demand and over-collecting of this plant, and hence the resources of wild D. candidum are endangered. In order to meet the increasing demands, tissue cultured seedlings are grown on large scale fields by local enterprises. Since 2001, a root and basal stem rot and die-back disease has been observed on D. candidum in Yiwu city, Zhejiang province, China. In the field, 1-year-old affected transplanted seedlings demonstrate typical symptoms of Phytophthora root and basal stem rot. Symptom progression is acropetal and plants die within a few days. Tap roots are completely rotted with black-brown to black sunken lesions frequently extending up the stem. In contrast, 2- to 3-year-old infected plants only have die-back symptoms with almost no root and basal stem rot. Typical black-brown lesions are observed on the tender top parts of a plant, causing top wilt and defoliation. Dendrobium candidum blight has not been observed in the wild. Five isolates of a Phytophthora species from diseased material were initially identified as Phytophthora nicotianae using morphological features (Stamps et al., 1990; Erwin & Ribeiro, 1996). No oospores were observed in the tissue. Mating types were determined by pairing known P. nicotianae tester strains. Oospores were produced following pairing tester strains and all isolates proved to be of the A2 mating type. PCR amplification of genomic DNA using the primers ITS6 and ITS4 generated sequences of approximately 890 bp. The sequences were submitted to the GenBank database (Acc. No. EF140984–88). Sequence analysis showed that the ITS sequences obtained were identical with others obtained from P. nicotianae (Estevez de Jensen et al., 2006), thus confirming the identity of the pathogen. Mycelial plugs of the five Phytophthora isolates taken from agar medium, were inoculated into the basal stem of 2-month-old transplanted seedlings grown in pasteurized potting substance (mainly chipped pine bark) and to the top parts (new shoots) of 2-year-old plants. Plants were grown in the greenhouse at 25°C, where they developed typical disease symptoms after 7 days. Non-inoculated controls did not develop disease symptoms. The pathogenic isolate was re-isolated from all diseased plants. Isolates have been deposited at the Biotechnology Institute, Zhejiang University. This is first record of phytophthora blight of D. candidum in China.

First Report of Fusarium Concentricum As a Causal Agent of Fusarium Leaf Blotch on Pecan (carya Illinoinensis) in Southeast China.

Abstract:The economically important nut crop pecan (Carya illinoinensis (Wangenh.) K. Koch) is seriously affected by increasing incidence of fungal disease worldwide (Xiao et al 2021). The top leaves of the pecan variety 'Pawnee' in the orchard of Zhejiang A&F University, Zhejiang, China were damaged by massive dark brown plaques in summer to autumn 2021. The causal agent was isolated from leaves with target plaques following the steps: sterilized with 70% alcohol (30 s × 2), rinsed with sterilized water (3 ×) before and after 5% sodium hypochlorite (30 s), excised the plaques, and placed on PDA medium at 28℃ in a dark incubator for 3-d. The mycelium on the edge of each colony was transferred to fresh SNA medium in dark for 2 weeks to induce conidia formation. A few conidia-germinated mycelia were transferredand inoculated on new plates containing fresh PDA medium to obtain the purified cultures. Koch's postulates were applied to validate the pathogenicity of the purified isolates. Non-woundedly healthy leaves (disinfected with 5% sodium hypochlorite) of 'Pawnee' seedlings were inoculated with 5 mm 7-d old purified cultures. Dark-brown spots appeared on the leaves 2 days post inoculation at 25℃. The spots became larger accompanied by partially cracking and slight deformation of inoculated leaves from day 2 to day 4, while the control leaves remained asymptomatic. A re-isolated strain ZJ-6 from these infected leaves was identified as the pathogenic isolate with the same symptom as the previous one. Morphologically, aerial mycelia of the pathogenic isolate ZJ-6 cashmere and white. The reverse of colony orange. The edge of the colony appeared gradually thinner, the aerial mycelia loose and flocky, and the matrix mycelium whitened. Hyphae were septate, translucent with smooth wall and 1.47-7.14 µm in width. Microconidia (n = 20) obovoid to fusoid, mainly with 0-septate, 4.45-7.78×4.79-16.25 µm. Macroconidia (n = 20) sickle, mainly with 3-5 septa, 5.56-10.28×56.67-114.54 µm. Simultaneous of monophialidic and polyphialidic conidiophores. Conidiophore width 1.47-3.68 µm, slightly smaller than vegetative hyphae. The morphological characteristics matched with previous descriptions of Fusarium species (Nirenberg and O'Donnell 1998; Wang et al 2013). The identity of ZJ-6 was confirmed by phylogenetic reconstruction using the concatenated sequences of the ATP citrate lyase (ACL1), Calmodulin (CaM), the internal transcribed spacer (ITS) rDNA region, ribosomal RNA gene (LSU), the largest subunit of DNA-dependent RNA polymerase II (RPB1), partial translation elongation factor-1 alpha (TEF) and β-Tubulin (TUB). To this end, the genomic DNA of ZJ-6 was extracted by the M5 hipermix-MF859 (Mei5 Biotechnology) and submitted to GenBank under the accession numbers OP933646, OP933647, OP925890, OP925889, OP933396, OP933648, and OP933397, respectively. The obtained sequences of ZJ-6 were used for nucleotide BLAST against thetandard databases, respectively, and the strains with sequence identification values above 98% were selected to construct multiple alignment for building a phylogenetic tree. This analyses allowed the identification of ZJ-6 as Fusarium concentricum Nirenberg & O'Donnell, a species with few reports that can cause serious damage to the fruits and branches of other hosts (Hasan et al 2020; Huda-Shakirah et al 2020; Wang et al 2013). This is the first report of pathogenic F. concentricum on pecan in Southeast China that caused no harvest of infected plants.

Perinatal transmission of Trichomonas vaginalis: a case report.

Abstract:BACKGROUND Trichomonas vaginalis infection is associated with increased risks of adverse pregnancy outcome. Perinatal transmission of T vaginalis from an infected, untreated, pregnant woman to her female neonate can occur. CASE A 22-year-old woman, gravida 3, para 2, with untreated trichomoniasis had an uncomplicated spontaneous vaginal delivery of a healthy female infant. At day of life 19 the newborn was diagnosed and treated for trichomoniasis. No evidence was found consistent with child sexual abuse. CONCLUSION Perinatal transmission of T vaginalis occurs rarely. Neonatal infection has medical and psychosocial implications. This outcome, albeit rare, should be a consideration in the decision regarding antenatal treatment of T vaginalis infection.
First Report of Peach Brown Rot Caused by Monilinia fructicola in Central and Western China

Abstract:Brown rot of peach (Prunus persica) in China has been reported to be caused by at least three Monilinia species (1). In the present study, peaches with symptoms resembling brown rot caused by Monilinia species were collected from commercial orchards in the northwestern province of Gansu in August 2010, the southwestern province of Yunnan in July 2011, and in the central province of Hubei in July 2012. Affected fruit showed the typical symptoms of brown rot with zones of sporulation. Fungal isolates were single-spored and cultured on potato dextrose agar (PDA). Colonies showed grayness with concentric rings of sporulation after incubation at 25°C in the dark. Mean mycelial growth of isolates YHC11-1a and YHC11-2a from Yunnan, GTC10-1a and GTC10-2a from Gansu, and HWC12-14a and HWC12-23a from Hubei, was 4.6 ± 0.4 and 7.5 ± 0.7 cm after 3 and 5 days incubation, respectively. Conidia were lemon shaped and formed in branched monilioid chains, and the mean size was 9.3 (6.7 to 11.5) × 12.5 (7.9 to 17.8) μm, which was consistent with the characteristics of Monilinia fructicola (1,2). The species identification was confirmed by sequencing of the ribosomal ITS sequences. The ribosomal ITS1-5.8S-ITS2 region was amplified from each of the six isolates using primers ITS1 and ITS4 (3). Results indicated that the ITS sequences of these isolates were identical and showed the highest similarity (100%) with M. fructicola ITS sequences from isolates collected in China (GenBank Accession Nos. The pathogen was also confirmed to be M. fructicola based on the detection of an M. fructicola- specific band (534 bp) using a PCR-based molecular tool developed for distinguishing Chinese Monilinia species affecting peach (1). Pathogenicity was tested on surface-sterilized, mature peaches (Shui Mi Tao) with representative isolates. Fruits were holed at three equidistant positions to a depth of 5 mm using a sterile cork borer. Mycelial plugs (5 mm in diameter) from the periphery of a 4-day-old colony of each isolate were placed upside down into each hole, control fruits received water agar. After 3 days of incubation at 22°C in a moist chamber, inoculated fruits developed typical brown rot symptoms while control fruits remained healthy. Pathogens from the inoculated fruit were confirmed to be M. fructicola based on morphological characteristics. To our knowledge, this is the first report of occurrence of M. fructicola in Gansu, Yunnan, and Hubei provinces, thousands of kilometers away from eastern China where occurrence of peach brown rot caused by M. fructicola has been confirmed (2,4). Bacterial Leaf Spot of Plum Caused by Sphingomonas spermidinifaciens in Guangxi, China

Abstract:Plum (Prunus salicina L.) is a traditional fruit in Southern China and is ubiquitous throughout the world. In August 2021, leaves of plum trees showed water-soaking spots and light yellow-green halos with incidence exceeding 50% in Babu district in Hezhou, Guangxi (N23°49'-24°48', E111°12'-112°03'). To isolate the causal agent, three diseased leaves collected from three different trees growing in different orchards were cut into 5 mm × 5 mm pieces, disinfected with 75% ethanol for 10 sec, 2% sodium hypochlorite for 1 min and rinsed three times in sterile water. The diseased pieces were ground in sterile water and then kept static for about 10 min. Ten-fold serial dilutions in water were prepared and 100 µL of each dilution from 10-1 to 10-6 were plated on Luria-Bertani (LB) Agar. After incubation at 28℃ for 48 h, the proportion of isolates with similar morphology was 73%. Three representative isolates (GY11-1, GY12-1 and GY15-1) were selected for further study. The colonies were non-spore-forming, yellow, round, opaque, rod shaped, convex with smooth and bright neat edges. Biochemical test results showed that the colonies were strictly aerobic and gram-negative. The isolates were able to grow on LB agar containing 0-2% (w/v) NaCl and could utilize glucose, lactose, galactose, mannose, sucrose, maltose and rhamnose as a carbon source. They displayed a positive reaction for H2S production, oxidase, catalase and gelatin, but negative for starch. Genomic DNA of the three isolates was extracted for amplification of the 16S rDNA with primers 27F and 1492R. The resulting amplicons were sequenced. Additionally, five housekeeping genes atpD, dnaK, gap, recA, and rpoB of the three isolates were amplified using the corresponding primer pairs and sequenced. The sequences were deposited in GenBank (16S rDNA, OP861004-OP861006; atpD, OQ703328-OQ703330; dnaK, OQ703331-OQ703333; gap, OQ703334-OQ703336; recA, OQ703337-OQ703339; and rpoB, OQ703340-OQ703342). The isolates were identified as Sphingomonas spermidinifaciens based on the phylogenetic tree inferred by maximum-likelihood using MegaX 7.0 of the concatenated six sequences (multilocus sequence analysis, MLSA) compared with sequences from different Sphingomonas type strains . Pathogenicity of the isolates was tested on healthy leaves of the two-year-old plum plants in a greenhouse. The leaves were wounded by a sterilized needle and sprayed with bacterial suspensions prepared in PBS (Phosphate buffer saline) at OD600=0.5. PBS buffer solution was used as negative control. Each isolate was used to inoculate on 20 leaves per plum tree. The plants were covered with plastic bags to maintain high humidity. Dark brown-to-black lesions were observed on leaves 3 days post incubation at 28℃ with constant light. The average diameter of lesions was 1 cm after seven days, but the negative controls were symptomless. Bacteria reisolated from the diseased leaves were the same as the ones used for inoculation on the basis of morphological and molecular identification, fulfilling Koch's postulates. Plant disease caused by a Sphingomonas species has been reported on mango, pomeand Spanish melon. However, this is the first report of S. spermidinifaciens causing leaf spot disease of plum in China. This report will help to develop effective disease control strategies in the future.
First Report of Brown Rot Caused by Monilia yunnanensis on Sweet Cherry in China

Abstract:Chinese cherry industry has developed rapidly over the past few years, with the planting acreage continuously expanding, from Shandong province to Liaoning, Shaanxi, Hebei, Sichuan etc. Monilia spp. are the most important causal agents of brown rot of cherry, to date, M. fructicola, M. mumecola, and M. fructigena were reported to cause brown rot of cherry in China (Chen et al. 2013; Yin et al. 2014; Liu et al. 2012). In May 2023, fruit of sweet cherry cultivar 'Hongdeng' (Prunus avium L.) with symptoms resembling brown rot were collected from Tongchuan City, Shaanxi Province. Conidia on diseased tissues were spread on a water agar (WA, 1.5% agar and distilled water) medium and isolated with a glass needle under a professional single spore separation microscope (Wuhan Heipu Science and Technology Ltd., Wuhan, China). If no conidia were present, fruit pieces (5 × 5 mm) at the intersection of healthy and diseased tissues were surface sterilized with a sodium hypochlorite solution (1%) for 30 s and washed three times in sterilized water, followed by 75% ethanol for 30 s, then washed three times in sterilized water. After the tissue pieces were dried, they were placed on potato dextrose agar (PDA; 200 g of potato, 20 g of dextrose, and agar at 20 g/L) and incubated at 22 °C for about twenty days to produce spores and then single spore isolation was carried out. Thirty single-spore isolates were obtained and all were morphologically similar. The isolates produced white-gray colonies with even margins and concentric rings of sporogenous mycelium after 3 days incubation, and abundant black-colored stromata on the PDA medium after 15 days of incubation at 22°C. Conidia were one-celled, hyaline, ellipsoid to lemon shape (14.12 × 10.37 μm), with 1-2 germs which is similar to M. yunnanensis on peach. The genomic DNA of the isolates was extracted as described previously (Chi et al. 2009). The pathogen identity was confirmed by multiplex PCR which resulted in a 237bp amplicon, which is diagnostic of M. yunnanensis (Hu et al. 2011). Further sequencing of the internal transcribed spacer (ITS) region 1 and 2 and 5.8S gene (accession number: OR192774) indicated 100% identity with that of M. yunnanensis isolates (accession numbers: MW355895, ON024742). The average daily growth of mycelium on PDA at 22°C was 11.44 mm. Koch's postulates were fulfilled by inoculating 20 mature sweet cherry fruits of cv. 'Van' with mycelial plugs in a drilled hole. After 3 days of incubation at 22℃ in an airtight plastic tray with wet paper, the inoculated fruit developed typical brown rot symptoms. The developing spores on inoculated fruit were confirmed to be M. yunnanensis based on ITS sequence. All control fruit inoculated with a PDA plug remained healthy. M. yunnanensis was first reported as the causal agent of brown rot of peach in China (Hu et al. 2011). Later studies demonstrated that it is also pathogen on other fruits, e.g. hawthorn (Zhao et al. 2013), plum (Yin et al. 2015), apricot (Yin et al. 2017), apple, and pear in China (Zhu et al. 2016). To our knowledge, this is the first report of cherry brown fruit rot caused by M. yunnanensis, indicating the high risk of this species to cherry production, and effective strategies must be taken to prevent the possible control failure in practice.
First Report of Fruit Rot of Sweet Cultivars of Japanese Plum caused by Alternaria alternata, A. arborescens, and A. tenuissima in Chile

Abstract:During the last two seasons, an unusual fruit rot was observed in four orchards of sweet Japanese plum (Prunus salicina) cultivars located in the Chilean Central Valley (30°00'S, 70°42'W). The incidence was 5% in Black Majesty, 4% in Red Lyon, and 6% in Sweet Mary cultivars in 2020. Fruits in the field showed a firm, dehydrated, and slightly sunken rot on the blossom end, along with rough and irregular epidermis in the affected area. Internally, the fruit flesh appeared light to dark-brown or olive-green. Symptomatic fruits (n=119) were superficially disinfected (75% ethanol) and, pieces of the pericarp (3 x 3 mm) were removed and placed on potato dextrose agar (PDA). Isolates of Alternaria spp. were obtained and 9 of these were selected for identification. Colonies were dark olive to gray-brown with white margins, small, catenulate and muriform conidia, produced in single or branched conidiophores. Isolates produced brown to golden-brown, ovoid, ellipsoidal to obclavate conidia with dimensions of 19.7 to 26.7 × 10.0 to 11.9 μm with two to four transverse and zero to three longitudinal septa on 0.05× PDA (Pryor and Michailides 2002) after 7 d at 20°C under 10/14 h light/dark cycles. A molecular analysis was performed by sequencing the nuclear genes RNA polymerase II subunit (RPB2), plasma membrane ATPase (ATP), and the calmodulin (Cal) gene using primers RPB2-5F2/fRPB2-7cR, ATPDF1/ATPDR1, and CALDF1/CALDR1, respectively (Lawrence et al. 2013; Woudenberg et al. 2013). A BLAST search revealed the presence of Alternaria spp. with a 99% to 100% identity with the reference sequences of A. alternata (JQ905182, JQ671874, JQ646208), A. arborescens (JQ646487, JQ671880, JQ646214), and A. tenuissima (JQ811961, JQ811989, JQ646209). Maximum parsimony phylogenetic analysis confirmed the identifications. Sequences were deposited in GenBank as numbers MW514249 to MW514257, MT872324 to MT872332, and MT872314 to MT872322 for RPB2, ATP, and Cal sequences, respectively. All these Alternaria isolates were deposited in the Colección Chilena de Recursos Genéticos Microbianos - INIA, Chillán Chile (RGM3069 to RGM3077). Pathogenicity of A. alternata (n=4), A. arborescens (n=3) and, A. tenuissima (n=2) was tested in Red Lyon plum fruits. Plums were disinfected in 1% sodium hypochlorite for 2 min, rinsed in sterile distilled water for 1 min and dried on absorbent towels in a laminar flow hood. Then, the plums were wounded on the blossom end with a sterile needle (1 x 0.5 mm), inoculated with 10 µl of a conidial suspension (106 conidia/ml), wrapped with Parafilm and maintained in a humid chamber (>95% relative humidity). An equal number of fruits wounded and inoculated with sterile water were used as a control. After 7 days at 20°C, all inoculated fruits developed a dark-brown firm rot with lesion lengths of 24.4 (±3.0) mm, 19.6 (±0.7) mm, and 16.8 (±2.4) mm for A. alternata, A. arborescens and A. tenuissima, respectively. A. alternata was the most aggressive species (P < 0.001). Control fruits remained asymptomatic. Koch's postulates were fulfilled after the re-isolating the causal agent from the border of the lesions. Leaf spots and fruit rots caused by Alternaria isolates have been reported in stone fruits, including plums (Kim et al. 2005; Long et al. 2021; Moosa et al., 2019; Yang et al. 2014). To our knowledge, this is the first report of A. alternata, A. arborescens, and A. tenuissima associated with fruit rot in sweet Japanese plum cultivars in the field, in Chile.
First Report of Fusarium pernambucanum Causing Fruit Rot of Melon in China

Abstract:In May of 2020, November of 2021 and May of 2022, a preharvest fruit rot with white mycelia was observed inside and outside of the fruits of thick skin muskmelon (Cucumis melo L.) growing in about ten greenhouses (each greenhouse had about 320 muskmelons) with disease incidence of 70% in Ningbo, Zhejiang Province of China. In order to identify the causal agent, plant tissues from the margin of the symptomatic tissue were sterilized for 1 min with 1% sodium hypochlorite (NaClO), 2 min with 75% ethyl alcohol, rinsed in sterile distilled water three times (Zhou et al 2019), and then placed on potato dextrose agar (PDA) plates containing streptomycin sulfate (100 μg/mL) at 25℃ for 4 days. Only Fusarium colonies were isolated from all the plant tissues. The growing hyphae were transferred to new PDA plates using the hyphal tip method, putative Fusarium colonies were purified by single-sporing. Six fungal isolates (Fi-1~6) were obtained. The average radial mycelial growth rate of Fusarium isolate Fi-3 was 4.6 mm/day at 25℃ in the dark on PDA, and like other five isolates. The colonies are abnormal, producing lots of aerial hyphae, each isolate was white to light orange. Isolate Fi-3 produced macroconidia with 4 to 6 septa, tapered with pronounced dorsiventral curvature and measured 21 to 30 μm long 4 to 5 μm wide on Spezieller Nährstoffarmer Agar (SNA) medium at 25℃ for 10 days (Leslie and Summerell 2006), but polyphialides and chlamydospores were still not available for 30 days. The pathogen species was further identified by translation elongation factor-1 alpha (EF-1α) sequencing. The EF-1α of six isolates were sequenced, and their EF-1α sequences were 100% identical to each other, and the sequence of strain Fi-3 was deposited in GenBank with accession no. OL782040 and was also compared with sequences in the FUSARIUM-ID database (Geiser et al. 2004), which indicated that it was 100% identical to those of F. pernambucanum strain NRRL 32864 (GenBank accession GQ505613), F. pernambucanum strain LC7040 (GenBank accession MK289626), and F. pernambucanum strain LC12149 (GenBank accession MK289588) within the Fusarium incarnatum - F. equiseti species complex 17 (FIESC17). Two phylogenetic trees were established based on the TEF1-α sequences of Fi-1~6 and other Fusarium spp., Fi-1~6 was clustered with the sequences of F. pernambucanum within the FIESC17. Thus, both morphological and molecular criteria supported identification of the strain as F. pernambucanum. A pathogenicity test was conducted to verify Koch's postulates, mycelium agar plugs (6 mm in diameter) were removed from the colony margin of a 3-day-old culture of strain Fi-3, healthy melon fruits were surface-sterilized with 70% ethanol and rinsed twice with sterile-distilled water. Then, the melons were wounded using a sterile inoculating needle to stab and inoculated by a mycelium agar plug of strain Fi-3 on the wound sites. 5 fruits were inoculated in each treatment, and a mycelium-free PDA plug was used as a negative control, repeated 3 times, at 25℃ with high relative humidity for 10 days. The results show disease symptoms similar to those naturally infected fruits on all inoculated melon fruits. The fungus re-isolated from the diseased fruits, showed the same colony morphology as the original isolate. Koch's postulates were repeated three times with the same results. Strain Fi-3 inoculated fruits without wounding remained healthy. To our knowledge, this is the first report of fruit rot of melon caused by F. pernambucanum in China.

Abstract:Pomegranate (Punica granatum) is an important fruit crop for therapeutic and food applications. In June 2022, brown spots were observed on the fruit surface of pomegranate cultivar named Guangyan in Mengzi (23°20'6''N,103°25'5''E), Yunnan, China. The early spots appeared as circular or irregular lesions, measuring 1~1.5 mm in diameter. They were light brown with a clear boundary between disease and healthy tissues. Over time, these spots developed into polygonal lesions covering the entire fruit surface. Eventually, the diseased fruits decayed, and more than 50% of fruits were infected in pomegranate orchards. The tissues from the interface between health and disease were cut down, immersed in 75% ethanol for 15 s, then 5% NaOCl disinfecting for 2 min, washed three times with sterile water, and the PDA cultured at 26 °C in an incubator under dark conditions. Twenty-five samples were collected for pathogen isolation, ten fungal isolates were obtained by single spore germination, and these isolates had similar morphological characters. The colonies were white with 81 mm diameter at 7 days of incubation, containing undulate edges with dense aerial mycelium. After 14 days, the black conidiomata formed superficially, gathering into black droplets. Conidiogenous cells were hyaline, short, and filiform. Conidia were fusiform, straight or slightly curved, and comprised five cells, 24.12 to 34.53 (x̄=29.78) μm × 4.21 to 12.15 (x̄=8.68) μm (n=50). The three median cells were 13.13 to 25.22 μm (x̄=18.54), dark brown, whose septa and periclinal walls were darker than the other two cells. The apical cells showed two to four appendages, 12.31 to 29.15 (x̄=21.56) μm. Only a single appendage was found on the basal cell, 2.34 to 7.16 μm. Based on morphological features, these isolates were identified as Neopestalotiopsis clavispora (Maharachchikumbura et al., 2012, 2014). Molecular identification of isolate YNSL-3 was performed by amplification and sequencing of ITS4/ITS5, BT2A/ BT2B and EF1-728F/EF-2, respectively (White et al. 1990, Glass et al.1995, Carbone et al. 1999, O'Donnell et al. 1998). These base sequences were deposited in GenBank with accession numbers OQ891378 (ITS), OR088917 (Tef) and OR513439(Tub), respectively. BLAST searches of the sequences revealed 100% (478/478 bp), 100% (484/484 bp), and 94.67% (426/450 bp) homology with those of N. clavispora NM16311a from GenBank (LC209216, LC209220, and LC209221), respectively. Phylogenetic analysis (IQ-TREE) by maximum likelihood method showed that the isolate YNSL-3 was clustered with N. clavispora. The pathogenicity was tested with the isolate of YNSL-3, YNSL-5 and YNSL-8 by detached assay. The fruit surface of pomegranate cultivar Guangyan was wounded with a sterilized needle. The mycelial blocks (5mm2) of isolates cultured on PDA for 7 days were attached to the points of inoculation. Controls were inoculated with sterile PDA agar. All inoculated fruits were maintained in a growth chamber at 26°C with 75% relative humidity. The test was performed thrice. The brown lesions were observed after 7 days, whereas the controls showed no symptoms. The same pathogens reisolated were identical to the original isolates based on morphological characterization and molecular analyses. N. clavispora could cause different diseases in many plants (Rajashekara et al. 2023, Loredana et al. 2020). To our knowledge, this is the first report of fruit brown spot on Punica granatum caused by N. clavispora in China. This finding will help improve management strategies against the fruit brown spots on P. granatum in China.

First Report of Fruit Rot on Plum Caused by Monilinia fructicola at Alcalá del Río (Seville), Southwestern Spain

Abstract:Monilinia fructicola, causal agent of brown rot, is one of the most important fungal pathogens of stone fruit. In the summer of 2011, Japanese plum fruit of 'Larry Ann' (Prunus salicina Lindl) showing symptoms of fruit rot disease were detected and collected from trees in an experimental field at Alcalá del Río (Seville), southwestern Spain. Fruit rot lesions were brown, sunken, and covered with grayish brown tufts or pustules. The majority of infected fruit became dry and mummified on the trees after 30 days. Symptoms were similar to those caused by three Monilinia species, M. laxa, M. fructigena, and M. fructicola (2). Pieces of infected tissue, previously disinfested in 0.6% NaOCl, were placed on potato dextrose agar (PDA) amended with 50 μg of streptomycin per liter and incubated at 22°C with a 12-h photoperiod for 15 days. The isolates produced abundant, grayish white mycelium, which after sporulation became hazel in color, and colonies displayed concentric rings. Colonies produced scarce conidia, which were arranged in branched, monilioid chains. Conidia were one celled, hyaline, ellipsoid to lemon shaped, and measured 15.42 ± 1.91 × 8.02 ± 0.9 μm. The morphological data and growth rates match the description of M. fructicola (Winter) Honey (2-4). Fungal identification was confirmed by PCR using genomic DNA extracted from the mycelia of pure cultures. The DNA was amplified with a common reverse primer and three specific forward primers obtained from a sequence-characterized, amplified region that distinguishes between M. fructicola, M. fructigena, and M. laxa. The size of the amplified fragment (a product of 535 bp) fit with the one described for M. fructicola (2). To confirm the pathogenicity of the isolate, mature 'Larry Ann' and 'Sungold' plum fruits (six fruits per cultivar) were inoculated by placing an agar plug from the edge of an actively growing colony on PDA directly on the fruit surface. After 5 days of incubation, typical brown rot symptoms developed on inoculated fruit and the fungus was successfully reisolated, thus fulfilling Koch's postulates. No symptoms appeared on control fruit. To our knowledge, this is the first report of M. fructicola on plums in southwestern Spain. M. fructicola is a quarantined pathogen in Europe and has been reported on imported apricot and nectarine (1) and peach in several European countries (3,4). References: (1) E. Bosshard et al. First Report of Postharvest Fruit Rot on Citrus reticulata Blanco Caused by Fusarium concentricum in China

Abstract:Nanfeng tangerine (Citrus reticulata Blanco) is highly regarded for its nutritional and economic value. In January 2022, an unknown fruit rot was observed on Nanfeng tangerine fruits harvested from Nanfeng County (27.22 °N, 116.53 °E), Fuzhou City, Jiangxi Province after 70 days in storage (25 °C, 90% relative humidity). The disease mostly started from the pedicel or a wound. Symptoms initiated with dark brown lesions that rapidly expanded between the fruit center and pulp capsule causing total fruit rot. The surface of symptomatic fruit was sterilized with 75% ethanol for 30 s and 2% NaClO for 30 s. Small diseased tissue pieces (2 mm2) between diseased and healthy tissues were placed on potato dextrose agar (PDA) and put in an incubator (25 ± 1 °C) for 3 days. The representative isolate NFMJ-1 was subcultured onto PDA using single-spore purification. Colonies on PDA were light yellow to white, with abundant flocculent aerial hyphae. Microconidia were oval, obovoid to allantoid, 0 septate, occasionally 1 septate, 4.07 to 17.53 × 1.69 to 3.56 μm (average=7.40 μm × 2.55 μm, n=50). Macroconidia were slender, with a beaked apical cell and a foot-shaped basal cell, 3 to 5 septate, 22.99 to 81.12 × 2.34 to 3.81 μm (average=45.04 μm × 3.12 μm, n=50). According to morphological characteristics, the isolate was tentatively identified as Fusarium sp. (Leslie and Summerell 2006). To confirm the identification, the internal transcribed spacer (ITS), translation elongation factor 1-alpha (TEF), RNA polymerase II second largest subunit (RPB2), beta-tubulin gene (TUB2), and calmodulin gene (CaM) sequences were amplified with primers ITS1/ITS4 (Gardes et al. 1993), TEF1/TEF2 (O'Donnell et al. 2010), RPB2-5f2/RPB2-7cr (Liu et al. 1999), Bt2a/Bt2b (Glass and Donaldson 1995), and CL1C/CL2C (Weir et al. 2012), respectively. The obtained sequences (ON184033, ON212051, ON212052, ON212053, ON212054) showed homology with F. concentricum ITS (MW016417.1; 514/514 bp), TEF (MK609902.1; 667/667 bp), RPB2 (LC631461.1; 941/972 bp), TUB2 (MT942588.1; 331/337 bp), and CaM (MK609916.1; 558/597 bp). A phylogenetic analysis of concatenated ITS-RPB2-TEF sequences was performed by MEGA7.0 with the maximum likelihood and Kimura 2-parameter model, revealing that the isolate was placed in the F. concentricum clade. To confirm pathogenicity, 36 healthy tangerine fruits were surface sterilized with 75% alcohol, then 18 disinfected fruits were wounded with sterile needles and 18 remained unwounded. Half of the wounded and un-wounded fruits were inoculated with 10 μL of a conidial suspension (1.0 × 106 conidia/ml) of isolate NFMJ-1 cultured for 7 days on PDA. Half of the wounded and un-wounded fruits were mock-inoculated with sterile water as controls. After incubation in an incubator (25 ± 1°C, 90% relative humidity) for 7 days, the wounded fruits inoculated with F. concentricum showed similar symptoms to the original diseased fruits, while the mock-inoculated fruits were asymptomatic. The pathogenicity test was repeated three times. The pathogen was re-isolated from the wound-inoculated fruits and identified as F. concentricum by morphological and molecular analysis, completing Koch's postulates. F. concentricum has been reported as a pathogen of Podocarpus macrophyllus (Dong et al. 2022), Capsicum annuum (Wang et al. 2013) and Zea mays (Du et al. 2020) in China. This is the first report of fruit rot caused by F. concentricum on Citrus reticulata in China. Appropriate prevention and control measure of the pathogen need to be developed to preserve marketability of this economically important citrus fruit.
First Report of Colletotrichum Plurivorum Causing Anthracnose on Pecan (carya Illinoinensis) in China

Abstract:Pecan (Carya illinoinensis) is an economically important nut crop worldwide (Xiao et al 2021). Anthracnose symptoms were found on pecan fruits and leaves in plantations in Anhui and Jiangsu provinces, China in August 2019. Irregular, dark brown or black spotted lesions firstly appeared on the surface and inside of fruits, and spread to all leaves. The symptoms resulted in 30% to 50% leaf drop and nearly a half of fruit decay in almost all trees of the susceptible cv. Wichita. The causal agent were isolated from fruits with target symptoms following the steps: surface disinfected with 75% ethanol (2×, 30 s), rinsed with sterile deionized water (3×), ~ 0.5 cm small fragments of the fruits excised and plated on potato dextrose agar (PDA) medium and incubated at 28 °C in dark for 3-d. Mycelium of each colony was picked and incubated on fresh PDA at 25 °C with a 12-hour light/dark cycle for 6-d to induce conidia formation. One 5-mm hyphal plug produced from each single spore isolate was transferred onto fresh PDA to obtain the pure cultures. Koch's postulates was employed for pathogenicity determination of the isolates. Non-wounded healthy leaves from seedlings of the disease susceptible cv. Pawnee were disinfected with 1% NaClO and inoculated with 5-mm 5-d hyphal of each isolate at 25 ℃. Tiny lesion spots were visible on the leaves after 2 days post inoculation (DPI) with isolate W-6 (the only pathogenic one among all isolates), and expanded over time until to the leaves withered, while the control leaves and leaves inoculated with other isolates remained asymptomatic. The pathogenicity of W-6 were confirmed using leaves and fruits of living Pawnee trees growing in Linglong Mountain Plantation, Lin'an, Hangzhou, China (119⁰38'51″E, 30⁰12'39″N, elevation: 119m). Three experimental replicates were conducted separately with three bio-replicates for all pathogenetic testing. The same symptoms were observed on both detached leaves and leaves and fruits of living trees.. The colony of W-6 have round cottony mycelium with complete edges and showed the fastest growth rate 3 - 4 DPI. After 7 DPI, white aerial mycelium turned yellowish brown and formed Acervulus in the mycelium. Conidia (n=50) one-celled, 12.0 - 20.0 μm × 3.5 to 6.0 μm width. Hyaline cylindrical with slightly rounded ends and two or three large guttulate at the centre. Most Acervulus dark brown and slightly irregular in shape, 12.70 × 18.79 μm (n=10). Setae were dark brown in color with average length around 34.10 μm (n=10). These characteristics matched previous descriptions of Colletotrichum orchidearum species complex, including C. plurivorum (Damm et al 2019). The identity of W-6 was confirmed by multi-locus phylogenetic analysis using the internal transcribed spacer (ITS) rDNA region and partial sequences of the conserved genes glyceraldehyde-3-phosphate dehydrogenase (GAPDH), actin (ACT), beta-tubulin 2 (TUB2), and chitin synthase (CHS). The sequences of W-6 were used for Basic Local Alignment Search Tool (BLAST) against NCBI GenBank and the sequences with 100% identity to that of W-6 were achieved, respectively. The concatenated sequences of the ACT-CHS-GAPDH-ITS-TUB2 was used for building a phylogenetic tree. The molecular analyses allowed the identification of the pathogen as C. plurivorum. It was known that 9 of the 11 Colletotrichum species causing pecan anthrax worldwide were reported in southern China (Brenneman 1989; Oh et al 2021). This is the first report of C. plurivorum as causal agent of pecan in China.
