Abstract:In this study, surimi products rich in lipids were prepared by using myofibril protein (MP) emulsion gel as carriers. The MP emulsion gel (MP concentration, c = 1.5%, oil fraction, o = 0.68) was prepared by one-step homogenization. The emulsion gel maintained a high elastic modulus (G ') after heating and freezing treat-ment. Confocal laser scanning microscopy revealed that the structure of the emulsion gel was a hybrid network consisting of polymers of cross-linked MP and aggregated protein-stabilized emulsion (W/O/W multiple struc-tures) droplets. The double emulsification of the emulsion gel and MP stabilized the oil droplets in the surimi product, preventing water and oil from leaching out. The microstructure also showed smaller gaps between MPs with increased porosity, while oil droplets were stably embedded in the surimi gel matrix. Moreover, adding MP emulsion gel significantly reduced the surimi gel strength compared to adding oil directly (p < 0.05).

Abstract:The study presented the effects of modulating the hydrophobic interaction and disulfide bond on the properties of myofibrillar protein (MP) emulsion gels at high temperature (95 degrees C) and the differentiation on the contri-bution of non-covalent (hydrophobic interaction) and covalent intermolecular interactions (disulfide bond) to the properties of interfacial protein films were also determined. The hydrophobic interactions among MP were modulated by the addition of octenyl succinic anhydride (OSA), and the disulfide bonds were modulated by the SH/SS exchange reactions mediated by GSH. The results showed that the MP emulsion gel properties at 95 degrees C were improved by modulating the hydrophobic interaction or disulfide bonds, and the dynamic interfacial adsorption of MP and dissipation quartz crystal microbalance experiments showed the interfacial adsorption pattens of protein were also changed. In addition, the hydrophobic interactions putted emphasis on improving the gel matrix, whereas the disulfide bonds focused on increasing the stiffness of interfacial protein films.

Abstract:This study investigated the emulsifying properties of a hot water extracted polysaccharide (SCMP) derived from liquid fermentation mycelium of Schizophyllum commune in stabilizing oil-in-water (o/w) emulsions. SCMP demonstrated the ability to stabilize o/w emulsions (referred to as SCMPe) for extended periods (>21 days) even at low concentrations (2 g/L) and increased the viscosity and elasticity of the emulsion system. Through confocal laser-scanning microscope visualization, it was observed that SCMP created a physical barrier on the surface of the oil droplets, contributing to emulsion stability. Furthermore, SCMPe demonstrated excellent tolerance to varying concentrations of Na+ (0.1-0.5 mol/L) and Ca2+ (0.01-0.05 mol/L) as well as a wide pH range (3.0-9.0). Moreover, increasing SCMP concentration effectively inhibited lipid oxidation and improved the stability of beta-carotene and curcumin in the emulsion during storage. A detailed analysis of SCMP's composition and structure was done to better understand its emulsifier properties, revealing SCMP to be composed of two /3-glucans with a rigid chain and high molecular weight. These results suggest that SCMP holds great potential an efficient stabilizer in the food industry, particularly in enhancing emulsion stability.

Abstract:Mannosylerythritol lipid-A (MEL-A) is one of natural glycolipid biosurfactants with extensive applications into food and cosmetics field. This study focused on the effect of MEL-A on physicochemical properties of myofibrillar protein (MP) from chicken and explained the underlying mechanism between biosurfactant-protein interaction from molecular level. Specifically, the mixed MP-MEL-A emulsions were prepared and characterized by rheological properties, protein structure alternations and emulsifying and foaming properties. The study showed that the addition of MEL-A promoted the formation of smaller droplet size, resulting from the electrostatic interaction and hydrophobic interaction inside the emulsion. Further investigations of SEM and SDS-PAGE indicated the protein aggregation phenomenon induced by MEL-A. With the increase of MEL-A content, the apparent viscosity, elasticity and sulfhydryl content increased significantly. Moreover, the decrease of alpha-helix and the increase of beta-sheet in protein secondary structure proved that MEL-A leaded to the formation of protein network for instance in gel. Besides, the addition of MEL-A also enhanced the emulsifying and foaming properties of emulsions, 1.5% of which seems to be the best choice. These results implied the great potential of MEL-A applied into meat gel product industry.

Abstract:SummaryThis study investigated the effects of direct current magnetic field (DC‐MF) treatment time (1, 3, 5, 8 h) on properties of porcine myofibrillar protein (MP). Gel water‐holding capacity increased from 83.40% to 87.20% when DC‐MF‐treatment time changed from 1‐h to 8‐h. The 3‐h treatment time of DC‐MF was found to promote MP unfolding, rearrangement and aggregation, leading to the loss of total sulfhydryl, the increase of reactive sulfhydryl, surface hydrophobicity, turbidity as well as the formation of MP clusters and the greater degree of crosslinking as compared with 0‐h treatment, thus a firmer and more ordered MP gel network for trapping more water. However, excessive DC‐MF treatment (8‐h) weakened DC‐MF effect on MP aggregation as well as gel network and texture. This study has shed light on the effects of DC‐MF treatment time on MP properties and provides useful information for the application of DC‐MF in the food industry.

Abstract:ABSTRACT The beneficial effects of social support on well-being have been shown in various studies. Less is known about factors which constrain or enhance the availability of social support. The present study profiles social support among women veterans and attempts to identify factors which enhance or constrain perceptions of support and social integration for women of different military eras. Data are derived from a national sample of women veterans who had at least one VA out-patient visit during a one year time period. Twenty percent of women veterans report having no one to depend on. Social support is lowest among Vietnam and Post-Vietnam era women. Chronic strains (such as having problems with relatives, housing, and paying bills) are important factors which are associated with levels of perceived support. Factors related to loneliness/alone status appear to be associated with constraints in group activities and perceived support. This research illustrates the importance of examining factors which constrain and enhance supportive activities and relationships.
Insight into the mechanism of enhancing myofibrillar protein gel hardness by ultrasonic treatment combined with insoluble dietary fiber from oat

Abstract:The present study investigated the effects of ultrasonic treatment combined with insoluble dietary fiber (IDF) from oat on gelation and structural characteristics of myofibrillar protein (MP). Ultrasonic treatment combined with IDF promoted the deployment of the MP chain and significantly increased ( P < 0.05) the solubility of MP. The samples treated with 120 W of ultrasonic power combined with IDF (U120 + IDF) formed a more continuous and dense gel microstructure, resulting the water-holding capacity (WHC) reached maximun values of 0.88 g/g. Moreover, compared with the U80 and U120 groups, MP treated by 80 W of ultrasonic power combined with IDF (U80 + IDF) and U120 + IDF groups had a higher β-sheet content of 46% and 40%, respectively. The gel hardness and chewiness of the U120 + IDF group reached maximum values of 2.14 N and 7.09 mJ, respectively. Ultrasonic treatment combined with IDF enhanced the texture and WHC of MP gel by expanding the secondary structure and increasing protein solubility. In conclusion, the U120 + IDF group had the highest value of gel hardness, chewiness, and a three-dimensional network structure.

Abstract:This study aimed to explore how pulsed electric field (PEF) treatment affects the structural, physicochemical, and emulsification properties of porcine-derived myofibrillar proteins (MPs). Increasing PEF treatment induced partial polarization and protein unfolding, resulting in notable denaturation that affected both the secondary and tertiary structures. PEF treatment also improved the solubility and emulsification ability of MPs by reducing their pH and surface hydrophobicity. Confocal laser scanning microscopy confirmed the effective adsorption of MPs and PEF-treated MPs at the oil/water interface, resulting in well-fabricated Pickering emulsions. A weak particle network increased the apparent viscosity in short-term PEF-treated Pickering emulsions. Conversely, in emulsions with long-term PEF-treated MP, rheological variables decreased, and dispersion stability increased. These results endorse the potential application of PEF-treated porcine-derived MPs as efficient Pickering stabilizers, offering valuable insights into the creative use of PEF for enhancing high-quality meat products, meeting the increasing demand for clean-label choices.

Abstract:This work aimed to investigate the combined effect of ultrasound (US) treatment and κ -carrageenan (KC) addition on the gelling properties and rheological behaviors of myofibrillar protein (MP). Without US treatment, the KC incorporation promoted the gel strength and water-holding capacity (WHC) of MP gels. These properties were further improved by 20 min US treatment with gel strength of 98.61 g and WHC of 79.87 %, which was mainly attributed to changes associated with hydrophobic interactions and disulfide bonds and the transformation from α-helix to β-sheet in MP gels. In addition, US treatment for 20 min effectively resulted in a more homogeneous polymer distribution of the MP-KC mixed system, leading to lower particle size and the largest G′ and G′′ values of the MP-KC mixed gels. However, longer US treatment times (30, 40 and 50 min) rendered lower gel strength, WHC, storage modulus and loss modulus of MP-KC mixed gels, which was mainly due to the formation of loose and disordered gel structures. Our present results indicated that the application of US to MP for an intermediate treatment time (20 min) combined with KC provides a potential and novel strategy to promote the gel qualities of heat-induced MP gels.

Abstract:In this study, we systematically investigated the emulsifying capabilities of myofibrillar protein (MP)- and MP peptide (MPP)-based conjugates synthesized through intensification techniques: water bath (WB), microwave, ultrasound, and the combined ultrasound-microwave (UM) methods. Compared with WB, microwave, and ultrasound treatments, the combined UM treatment greatly promoted the glycation reaction because ultrasound and microwave mutually reinforced modification effects. The resultant conjugate structure tended to unfold with more flexible conformation and homogeneous morphology. Moreover, the emulsifying properties of conjugates developed with single and combined ultrasound-assisted glycation displayed substantial improvement, and pre-hydrolysis further enhanced these performances, as observed in the Principal Component Analysis as well. Remarkably, MPP grafted by maltodextrin with the assistance of a combined UM field produced the smallest and most uniform emulsion system, positioning it as the most efficient emulsifier among all the fabricated glycoconjugates. Our study highlighted the potential of synergistically applying ultrasound and microwave techniques to develop a well-performance glycation with an ideal conjugate structure, in which they would be associated into a strong film that provided the robust physical barrier, creaming stability, heat retention, and oxidation resistance. These findings offered a basis for better utilizing complex ultrasonic technology to develop novel and improved MP-based food products.

Abstract:The real-time structural changes of the molecular space conformation of myofibrillar protein microgels (MPM) after heat treatment (90 °C, 30 min) were analyzed by molecular dynamics simulation, and the structural properties and changes of MPM at the oil-water interface were analyzed by the combination of Raman spectroscopy and molecular dynamics simulation. The shift in the oil ratio had a major impact on the transformation of disulfide bonds within the protein molecule. Simultaneously, it caused tryptophan and tyrosine residues (I850 cm-1/ I850 cm-1 > 1) to become exposed, increasing the locations of amino acid residues in the protein that interact with the oil phase. HIPE with different oil phases influenced the change in spatial structural conformation of MPM, and there was a flexible structural change in the molecular space. The HIPE system, which was stabilized by 3.0 wt% MPM and 0.75 oil phase, exhibited a thixotropic recovery of >70 % and the highest elastic modulus G' (822.14 Pa) based on the rheological behavior. It is expected to provide a theoretical basis for the development and utilization of high internal phase emulsion stabilized by microgel protein in food industry.

Abstract:In this study, the effect of chickpea dietary fiber (CDF) concentration (0%, 0.4%, 0.8%, 1.2%, 1.6%, and 2.0%) on emulsion gel properties of myofibrillar protein (MP) was investigated. It was found that the emulsifying activity index (EAI) and emulsifying stability index (ESI) of MP increased with the increasing content of CDF. Moreover, the water- and fat-binding capacity (WFB), gel strength, storage modulus (G'), and loss modulus (G") of MP emulsion gel also increased with increasing content of CDF. When the concentration of CDF was 2%, the most significant improvement was observed for EAI, breaking force, and WFB (p < 0.05); the three-dimensional gel network structure of the MP emulsion gel was denser and the pore diameter was smaller. The T21 relaxation time of emulsion gel decreased while the PT21 increased significantly with the increasing content of CDF, suggesting that the emulsion gel with CDF had a better three-dimension network. The addition of CDF led to an increased content of β-sheet and reactive sulfhydryl and increased surface hydrophobicity of MP, thus improving the gel properties of the MP emulsion gel. In conclusion, the addition of CDF improved the functional properties and facilitated the gelation of the MP emulsion, indicating that CDF has the potential to improve the quality of emulsified meat products.

Abstract:The emulsions prepared by three non-meat proteins, sodium caseinate (SC), soy protein isolate (SPI) and egg white protein (EPI), were individually added to the continuous phase of myofibrillar protein (MP) sol to form MP composite gels to simulate meat products. The research aimed to investigate the effects of Transglutaminase (TGase) on the physicochemical properties, microstructure and water phase distribution of non-meat protein emulsion MP composite gels. The results of this study revealed that TGase played a crucial role in forming a tight gel network structure in the composite gels. This enhanced their ability to retain water and improved their overall gel strength. Additionally, TGase increased the gel formation temperature of myofibrillar proteins. Electrophoresis analysis showed that when catalyzed by TGase, there was a lighter band compared to those not catalyzed by TGase. This indicated that the addition of TGase facilitated cross-linking interactions between meat proteins and non-meat proteins in the composite gels. Furthermore, microscopy observations demonstrated that composite gels treated with TGase exhibited a more uniform microstructure. This could be attributed to an acceleration in relaxation time T2. The uniform network structure restricted the movement of water molecules in the gel matrix, thereby improving its water-holding capacity. Overall, these findings highlight how incorporating non-meat proteins into myofibrillar systems can be effectively achieved through enzymatic treatment with TGase. Such modifications not only enhanced important functional properties but also contributed towards developing alternative meat products with improved texture and moisture retention abilities.

Abstract:This study aimed to improve the gel properties of emulsion gels prepared by physical complex of fish protein and polysaccharide. The effects of Konjac glucomannan (KGM) on the structure, emulsibility of Nemipterus virgatus myofibrillar protein (MP), and the gel properties of heat-induced MP emulsion gel were investigated. The results indicated KGM modification facilitated the unfolding of MP, exposing more tryptophan and hydrophobic groups, thereby improving the wettability and emulsifying activity of MP/KGM mixtures. The interactions between MP and KGM molecules were mainly non-covalent hydrogen bonding and hydrophobic interaction. The MP/KGM mixtures exhibited superior adsorption properties at the oil-water interface, particularly with 0.5 % KGM addition, thus forming small and evenly distributed oil droplets. Besides, the curves of viscosity, G' and G" of MP/KGM emulsions climbed continuously with the increased KGM concentration. The presence of KGM, especially the medium amount (0.5 %), was conducive to the translation of α-helix into β-sheet and the formation of more dense and ordered gel network structure, thereby enhancing the gel properties of MP/KGM emulsion gels.

Abstract:The effects of insoluble dietary fiber (IDF) on myofibrillar protein (MP) emulsion gelations were investigated through a model system. Steady shear tests indicated an increase of molecular interaction/friction in composite solutions with IDF inclusion. It was noted that IDF addition prevented oil dispersion and resulted in the average enlargement oil globules volume diameter (d4,3) from 145.25 (control) to 200 μm (2% IDF), in emulsified systems. In dynamic rheology, the final G′ of the emulsion gelations significantly increased from 203 (control) to 462 Pa (2% IDF), while the T21 relaxation time of immobilized water, in emulsion gels containing 2% IDF (202.44 ms), was significantly shorter than in control gels (244.25 ms). In addition, the water- and fat-binding capabilities, and the gel strength of emulsion gelations, significantly increased with IDF addition. The aforementioned results indicate that IDF addition could promote the formation of a stronger gel network. As a result, the MP gel microstructure was further studied. The MP gel microstructure of gels containing IDF had reduced formation of interconnected water with a gel network possessing more integrity and overall compactness, especially 2% IDF addition. In conclusion, IDF addition was shown to play an important role in the physicochemical properties of low-fat emulsion gels through the enlargement of oil particle size and through the modification of the overall MP gel network.

Abstract:This study investigated the gelling and structural properties of composite gels prepared with myofibrillar protein (MP) and different concentrations of thermo-reversible curdlan gels (TRC) and thermo-irreversible curdlan gels (TIRC). The gel strength, water holding capacity, and whiteness of MP gels were significantly increased with increasing TRC and TIRC content (P < 0.05). Dynamic rheological testing upon temperature sweeping indicated that the G' and G '' values of MP were significantly enhanced with added TRC or TIRC, which were positively related to the lower particle size of composite MP sols. Moreover, the presence of TRC or TIRC changed the secondary structure of MP gels, as well as altered the tertiary structure of MP sols. The micrographs of MP gels indicated that TRC prompted the formation of more dense and stable protein matrix. Our results revealed that TRC or TIRC had distinctive effects on the gelling properties and secondary structure of MP.
Enhancement of hydrogen bonds between proteins and polyphenols through magnetic field treatment: Structure, interfacial properties, and emulsifying properties

Abstract:In food systems, proteins and polyphenols typically coexist in a non-covalent manner. However, the inherent rigid structure of proteins may hinder the binding sites of polyphenols, thereby limiting the strength of their interaction. In the study, magnetic field (MF) treatment was used to enhance non-covalent interactions between coconut globulin (CG) and tannic acid (TA) to improve protein flexibility, enhancing their functional properties without causing oxidation of polyphenols. Based on protein structure results, the interaction between CG and TA caused protein structure to unfold, exposing hydrophobic groups. Treatment with a MF, particularly at 3 mT, further promoted protein unfolding, as evidenced by a decrease in α-helix structure and an increase in coil random. These structural transformations led to the exposure of the internal binding site bound to TA and strengthening the CG-TA interaction (polyphenol binding degree increased from 62.3 to 68.2%). The characterization of molecular forces indicated that MF treatment strengthened hydrogen bonding-dominated non-covalent interactions between CG and TA, leading to improved molecular flexibility of the protein. Specifically, at a MF treatment at 3 mT, CG-TA colloidal particles with small size and high surface hydrophobicity exhibited optimal interfacial activity and wettability (as evidenced by a three-phase contact angle of 89.0°). Consequently, CG-TA-stabilized high internal phase Pickering emulsions (HIPPEs) with uniform droplets and dense gel networks at 3 mT. Furthermore, the utilization of HIPPEs in 3D printing resulted in consistent geometric shapes, uniform surface textures, and distinct printed layers, demonstrating superior printing stability. As a result, MF treatment at 3 mT was identified as the most favorable. This research provides novel insights into how proteins and polyphenols interact, thereby enabling natural proteins to be utilized in a variety of food applications.
Elevated PrPC Expression Predisposes to Increased HSV-1 Pathogenicity

Abstract:PrPC is a ubiquitously expressed glycophos-phatidylinositol-linked cell-surface glycoprotein found primarily in neural tissue. Although its normal function has not been established, there is evidence suggesting that PrPC is involved in cell signalling and cellular homeostasis. This suggests that variation in neuronal expression levels of this protein contributes towards pathogenicity induced by neurotropic agents. We have investigated the pathological response to infection with herpes simplex virus type-1 (HSV-1) in strains of mice that express different levels of PrPC. Prnp−/− mice fail to express PrPC due to an interruption in the open reading frame of the Prnp gene, whilst tg19 and tga20 mice express approximately 5 and 10 times more PrPC protein, respectively, than wild-type animals. Mice that express normal or increased levels of PrPC protein were more susceptible to acute HSV-1 infection than Prnp−/− mice. Following ear pinna inoculation with HSV-1 SC16, the order of susceptibility was tga20>tg19>wild-type>Prnp−/−. This trend was reversed when latent virus was assessed. Prnp−/− mice expressed significantly higher levels of latency-associated transcript-positive neurons in various tissues when compared with wild-type, tg19 and tga20 mice. Collectively, our data show that acute HSV-1 replication proceeds more efficiently in neuronal tissue that expresses PrPC protein and lends support to the view that this protein is involved in regulation of neurotropic viral pathogenesis. This suggests that interference of PrPC expression, or possible biochemical pathways associated with its function, may serve as an effective means of limiting the pathogenesis of acute HSV-1 infection.
