Abstract:For the diagnosis and therapy of small cell lung cancer (SCLC), the accurate and sensitive determination of neuron-specific enolase (NSE) content is crucial. This work outlines a dual-quenching electrochemiluminescence resonance energy transfer (ECL-RET) immunosensor based on the double quenching effects of iron base metal organic frameworks (FeMOFs) loaded with small sized CuO nanoparticles (FeMOFs-sCuO) towards CoPd nano -particles (CoPdNPs) enhanced porous g-C3N4 (P-C3N4-CoPdNPs). To be specific, we prepared a porous g-C3N4 (P-C3N4) which has a rich porous structure, and significantly increased the specific surface area and the number of reaction sites of P-C3N4. Meanwhile, the CoPdNPs were loaded onto P-C3N4 to improve the ECL luminescence property of P-C3N4/K2S2O8 system through acting as a coreaction accelerator. In addition, the ultraviolet-visible (UV-vis) absorption spectra of FeMOFs and small sized CuO nanoparticles (sCuO) showed considerable overlap with the ECL emission spectra of P-C3N4 appropriately. Therefore, FeMOFs with high specific surface area were prepared and well combined with sCuO to effectively dual-quenching the ECL emission of P-C3N4 based on resonance energy transfer. Hence, a new type ECL-RET couple made up of P-C3N4-CoPdNPs (donor) and FeMOFs-sCuO (acceptor) were developed for the first time. A certain amount of P-C3N4-CoPdNPs, Ab1, BSA, NSE were modified layer by layer onto the electrode surface. Then FeMOFs-sCuO-Ab2 bioconjugates was incubated through the immune recognition binding. As a result, a sandwich-type ECL biosensor was manufactured suc-cessfully for NSE immunoassay. Under optimal experimental conditions, the limit of detection (LOD) and the limit of quantitation (LOQ) of the prepared ECL sensor for NSE analysis was 20.4 fg mL-1 and 7.99 fg mL-1, respectively, with the relative standard deviation (RSD) of 1.68%. The linear detection range was 0.0000500-100 ng mL-1. The studied immunosensor had satisfactory sensitivity, specificity and reproducibility, manifesting the suggested sensing strategy might offer a good technical means and theoretical basis for the sensitivity analysis of NSE and has a potential application in clinical diagnosis analysis.

Abstract:A quenching electrochemiluminescence (ECL) immunosensor for neuron-specific enolase (NSE) detection was constructed by the resonance energy transfer between Ag@Ce2Sn2O7 (energy donor) and Ag@Cu2O (energy acceptor). The novel luminophore, Ag@Ce2Sn2O7, was synthesized by complexing Ce2Sn2O7 nanocubes with Ag concave-convex nanospheres. It exhibits a stronger ECL emission and a lower potential required for the emissions than pure Ce2Sn2O7. The Ag@Cu2O core-shell structure is composed of a dense Ag core inside and a loose Cu2O shell that provide a large specific surface area and the protection for the independence and integrity of the Ag core. The UV-Vis absorption of Ag@Cu2O overlaps the ECL emission of Ag@Ce2Sn2O7, and thus it can act as a quencher to effectively quench the ECL signal of Ag@Ce2Sn2O7. The great application potentials of the immunosensor in the NSE detection is demonstrated with the good linearity in the wide concentration range of 25 fg/ mL - 100 ng/mL, and the low detection limit of 8.6 fg/mL. Our work has provided a reference for the immunosensor development of other biomarkers.
Dual-signal Electrochemiluminescence Immunosensor for Neuron-specific Enolase Detection Based on "Dual-Potential" Emitter Ru(bpy)32+ Functionalized Zinc-Based Metal-Organic Frameworks.

Abstract:Neuron-specific enolase (NSE) is the preferred marker for monitoring small cell lung cancer and neuroblastoma. We devised a dual-signal ratiometric electrochemiluminescence (ECL) sensing strategy for sensitive detection of NSE. In this work, Ru (bpy)32+ functionalized zinc-based metal-organic framework (Ru-MOF-5) nanoflowers (NFs) with plentiful carboxyl groups provide an excellent biocompatible sensing platform for the construction of immunosensor. Importantly, Ru-MOF-5 NFs possess stable and efficient "dual-potential" ECL emission of cathode (-1.5 V) and anode (1.5 V) in the existence of co-reactant K2S2O8. Simultaneously, the cathode ECL emitter ZnOAgNPs are employed as the secondary antibody marker, whose participation amplify the cathode ECL signal as well attenuate the anode ECL emission of Ru-MOF-5 NFs. By monitoring the ECL dual-signal of -1.5 V and 1.5 V and calculating their ratios, a ratiometric strategy of quantified readout proportional is implemented for the proposed immunosensor to precise analyze NSE. Based on optimization conditions, the ECL immunosensor displays the wide linear range of 0.0001 ng/mL to 200 ng/mL and the minimum detection limit is 0.041 pg/mL. The "dual-potential" ratiometric ECL immunosensor effectively reduces system error or background signal by self-calibration from both emissions and improves detection reliability. The dual-signal ratiometric strategy with satisfactory reproducibility and stability provides further development possibilities for other biomolecular detection and analysis.

Abstract:Electrochemiluminescence (ECL) is a widely applied technique for trace detection and analysis, but the stability of luminophores and external environmental factors strongly influence the analysis results. Herein, a low-potential ECL sensing platform based on enhanced nanomaterial catalysis for the traditional system luminol-H 2 O 2 is proposed for the detection of neuron specific enolase (NSE). In this strategy, Au nanoparticles modified with three-dimensional graphitic phase carbon nitride (3DCNA) were used as a solid platform for antibodies (Ab 2 ), which not only solved the problem of accumulation between traditional two-dimensional nanosheets but also had good biocompatibility, ensuring the accuracy of the analysis results. CoSn(OH) 6 , benefiting from its inherently higher catalytic activity and greater number of adsorption sites on perovskite hydroxide, can catalyze the oxidation reduction reaction of H 2 O 2 to improve the ECL behavior of luminol and lower the applied voltage to reduce damage to biological activity. Owing to these exceptional properties, this proposed sandwich ECL immunosensor was explored for the primary diagnosis of SCLC by detecting NSE with a detection limit of 0.18 pg mL –1 . It exhibited satisfactory linearity in the range of 0.0005 ng mL –1 to 100 ng mL –1 with favorable stability, excellent specificity, and good reproducibility. This study provides an important reference for low-potential ECL with promising application prospects in actual clinical analysis and detection.

Abstract:As a specific biomarker, neuron-specific enolase (NSE) is an essential clinical indicator for diagnosing small cell lung cancer. In this paper, a sandwich-type electrochemical immunosensor was designed for the quantitative detection of NSE. AuPt nanoblock spherical nanoarchitectonics (AuPt NSNs), a bimetallic nanoparticle with a rugged morphology, were utilized as the substrate, which could enhance the electronic conduction and increase the immobilization capacity of the primary antibody (Ab1). Moreover, through a simple hydrothermal method, Au/CuxO@CeO2 was prepared as a spiny core-shell nanocube with cerium dioxide (CeO2) and gold nanoparticles (Au NPs) loading. The combination of Cu2O, CuO, and CeO2 showed favorable catalytic activity toward hydrogen peroxide (H2O2). Furthermore, the deposition of Au NPs on the spiny surface structure enhanced the specific surface area and biocompatibility, thereby rendering it more effective for loading the second antibody (Ab2). As the label material, the Au/CuxO@CeO2 achieved signal amplification and sensitive detection with the immunosensor. Under optimal conditions, the designed immunosensor possessed a broad linear range of 50 fg mL-1 to 100 ng mL-1 and a limit of detection of 31.3 fg mL-1, along with satisfactory performance in sensitivity, selectivity, and stability.

Abstract:An electrochemical immunosensor for detection of neuron specific enolase (NSE) was designed by immobilizing NSE covalently functionalized single-walled carbon nanotubes (NSE-SWNTs) on a glassy carbon electrode. The NSE-SWNTs not only enhanced electrochemical signal but also presented abundant antigen domains for competitive immunological recognition to anti-NSE primary antibody and then gold nanoprobes labeled with alkaline phosphatase conjugated secondary antibody (AP-anti-IgG/AuNPs). The AP-anti-IgG/AuNPs exhibited highly catalytic activity toward enzyme substrate and significantly amplified the amperometric signal for target molecule detection. Based on the dual signal amplification of SWNTs and gold nanoprobe, the immunosensor could response down to 0.033 ng mL(-1) NSE with a linear range from 0.1 ng mL(-1) to 2 mu g mL(-1), and showed acceptable precision and reproducibility. The designed immunosensor was amenable to direct quantification of target protein with a wide range of concentration in complex clinical serum specimens. The assay results were in a good agreement with the reference values. The proposed electrochemical immunosensor provided a pragmatic platform for convenient detection of tumor markers in clinical diagnosis. (C) 2012 Elsevier B.V. All rights reserved.

Abstract:Enzyme-free catalytic amplification is of great significance for sensitive label-free electrochemical immunosensors. In this study, an enzyme-free catalytic amplification based label-free amperometric immunosensor was developed for sensitive detection of neuron-specific enolase (NSE) by use of a AuPd nanoparticle–multiwalled carbon nanotube (AuPd-MWCNT) composite, ferrocenecarboxaldehyde (Fc-CHO), and chitosan hybrid hydrogel. The intrinsic virtues of chitosan not only resulted in bioactivity of the attached antibodies and made the other component of the immunosensor easier to fix on the electrode, but also imparted abundant binding sites to the hydrogel to condense Fc-CHO to achieve the initial signal amplification. Fc-CHO, which served as an electroactive species to generate a redox response, also exhibits excellent electrocatalytic activity toward H2O2. AuPd-MWCNT composite, with enhanced peroxidase-like catalytic activity, could catalyze H2O2 to accelerate electron transfer. When H2O2 was present in the detection solution, synergetic catalysis of Fc-CHO and AuPd-MWCNT composite toward H2O2 was achieved, thus realizing enzyme-free signal amplification. On the basis of this enzyme-free signal amplification, the electrochemical immunosensing platform provided a wide linear range from 1 pg mL-1 to 100 ng mL-1, a low detection limit of 0.483 pg mL-1, and high sensitivity of 7.22 μA (log10CNSE)-1. Abstract:Lung cancer is a high-mortality cancer related to the concentration of neuron-specific enolase (NSE). In this work, a sandwich-type photoelectrochemical (PEC) immunosensor was constructed for ultrasensitive detection of NSE, which is based on iron trioxide/indium zinc cadmium sulfide (Fe2O3/Cd-ZnIn2.2Sy) as a sensing platform and Ag-modified polyaniline (Ag@PANI) as a signal amplification label. The 1D Fe2O3 porous nanorods with a large specific surface area were synthesized by calcination of Fe-MIL-88A and etching of NaOH. To improve the photocurrent response, the 3D architecture Cd-ZnIn2.2Sy was combined with the 1D Fe2O3 porous nanorods to form a 1D Fe2O3/3D Cd-ZnIn2.2Sy heterostructure. Specifically, the Fe2O3/Cd-ZnIn2.2Sy heterostructure with a good energy level matching (the two can form a stepped energy level matching, which accelerates the transfer rate of electrons) can improve the separation efficiency of electron-hole pairs (e-/h+) under visible light irradiation, which enhances the photocurrent response. Ag@PANI has a strong electron transport capability and can be used as a secondary antibody marker for the signal amplification of the immunosensor. The sensor exhibits a good linear detection range of 100 fg/mL to 100 ng/mL with a low detection limit of 33.5 fg/mL. Moreover, the constructed sandwich-type PEC immunosensor shows good performance and possesses excellent specificity, selectivity, and stability over a period of 4 weeks for NSE detection. With these excellent properties, the immunosensor can be extended to analyze and diagnose other disease biomarkers.
A signal amplifying photoelectrochemical immunosensor based on the synergism of Au@CoFe2O4 and CdS/NiCo2O4 for the sensitive detection of neuron-specific enolases

Abstract:Neuron-specific enolase (NSE), a marker of small cell lung cancer, is over-expressed in patient serum. To reduce the threat of lung cancer to human health and life, a rapid and effective method for the sensitive detection of NSE is necessary. In this study, a sandwich-type photoelectrochemical (PEC) immunosensor for the rapid and sensitive detection of NSE was constructed. To provide a powerful and stable photocurrent signal, the bandgap-matched CdS/NiCo2O4 was used as the substrate material. In addition, Au@CoFe2O4, which can form a step electron transfer mode with NiCo2O4, was proposed for the first time as a second antibody (Ab2) marker to amplify photoelectric signals. The constructed signal-amplified PEC immunosensor showed good linearity in the range of 1 pg/mL − 100 ng/mL with a detection limit as low as 0.24 pg/mL (S/N=3). The constructed PEC immunosensor has shown excellent stability, selectivity, reproducibility, and repeatability. It showed recoveries in the range of 98% − 105% and relative standard deviations (RSD) within 5% in sample analysis. The method proposed in this study is expected to be more widely used in the future for the detection of other disease markers as well as environmental pollutants.

Abstract:The preparation of highly efficient electrochemiluminescence (ECL) illuminants is an effective method to improve the sensitivity and repeatability of ECL immunoassay. In this study, we prepared an ECL immunoassay for efficient and sensitive detection of neuron-specific enolase (NSE) by linking carboxylated Ru(bpy)32+ to an iron-based metal-organic framework (NH2-MIL-88 (Fe)) via an amide bond as an ECL signal probe. NH2-MIL-88 (Fe) possesses a large number of amino groups that can catalyze the co-reactant S2O82-, which generates abundant reaction intermediates SO4•- around Ru(dcbpy)32+, reduces the loss of material transport and energy transfer between SO4•- and Ru(dcbpy)32+, and significantly enhances the ECL signal. We used polyaniline-intercalating vanadium oxide (PVO) nanosheets as the substrates to capture NSE owing to the large specific surface area and extraordinary conductivity of the nanosheets. Similarly, PVO nanosheets also possess abundant amino groups, which can act as co-reaction promoters to catalyze the reaction of S2O82- to SO4•-, enhancing the ECL signal of the immunoassay. Therefore, we constructed a dual-enhanced ECL immunoassay with Ru(dcbpy)32+/NH2-MIL-88 (Fe) and PVO as the signal probe and substrate, respectively, which exhibited excellent sensitivity and selectivity for detecting NSE. This study offers an effective strategy for ultrasensitive detection of trace proteins using ECL immunoassays.

Abstract:In this study, a signal-on type PEC immunosensor was constructed to detect neuron-specific enolase (NSE) via Z-scheme WO3/NiCo2O4 p-n heterojunction with cactus-like structure used as photoactive materials and MnxCd1-xS⊃Au NPs (MCS⊃Au NPs) as signal labels. Firstly, Z-scheme WO3/NiCo2O4 heterojunction could accelerate the separation efficiency of carriers and well-matched photoactive materials may promote charge migration, which resulted in WO3/NiCo2O4 generating strong and stable current. In addition, Z-scheme WO3/NiCo2O4 heterojunction directly grown on the surface of FTO via hydrothermal method facilitated the preparation of PEC immunosensor with outstanding stability. Secondly, an efficient signal amplification strategy was proposed by MnxCd1-xS⊃Au NPs incubating with signal antibody (Ab2). On the one hand, the well-matched energy levels of MnxCd1-xS with WO3/NiCo2O4 boosted the photo-generated electrons transferred to the electrode; on the other hand, the LSPR effect of Au may convert thermion to photocurrent to achieve signal amplification. Based on the above strategies, a PEC immunosensor with outstanding reproducibility and stability was obtained for sensitive detection of NSE. Under the optimum experimental conditions, current response range of the constructed signal amplification PEC sensor to NSE was 0.1 pg/mL ∼50 ng/mL and the detection limit was 0.07 pg/mL (S/N = 3). After the application tests in the detection of actual samples, the feasibility of the prepared PEC immunosensor with excellent selectivity, high sensitivity and satisfactory reproducibility was verified and the satisfactory results were obtained.

Abstract:Neuron-specific enolase (NSE) is a specific marker for small cell carcinoma (SCLC). Sandwich-type electrochemical immunosensors are powerful for biomarker analysis, and the electrocatalytic activity of the signal amplification platform and the performance of the substrate are critical to their sensitivity. In this work, N atom-doped graphene functionalized with hollow porous Pt-skin Ag-Pt alloy (HP-Ag/Pt/NGR) was designed as a dual signal amplifier. The hollow porous Pt skin structure improves the atomic utilization and the larger internal cavity spacing significantly increases the number of electroactive centers, thus exhibiting more extraordinary electrocatalytic activity and durability for H2O2 reduction. Using NGR with good catalytic activity as the support material of HP-Ag/Pt, the double amplification of the current signal is realized. For the substrate, polypyrrole-poly(3,4-ethylenedioxythiophene) (PPy-PEDOT) nanotubes were synthesized by a novel chemical polymerization route, which effectively increased the interfacial electron transfer rate. By coupling Au nanoparticles (Au NPs) with PPy-PEDOT, the immune activity of biomolecules is maintained and the conductivity is further enhanced. Under optimal conditions, the linear range was 50 fg mL-1 - 100 ng mL-1, and the limit of detection (LOD) was 18.5 fg mL-1. The results confirm that the developed immunosensor has great promise for the early clinical diagnosis of SCLC.

Abstract:Silver nanoparticles (Ag·NPs) show promising advantages in electrochemiluminescence (ECL) owing to their favorable optical properties and biocompatibility. However, their susceptibility to oxidation and degradation in the presence of air adversely affects ECL intensity. In this study, we employed a sandwich sensing platform using silica-coated silver nanoparticles (Ag@SiO 2 ) as a novel luminescent material and cerium dioxide (CeO 2 ) as an ECL signal quencher for sensitive neuro-specific enolase (NSE) detection. The core-shell structure protected Ag NPs within the silica (SiO 2 ) layer, enhancing their ECL luminescence properties by reducing external environmental influence and preventing Ag NPs aggregation. Amino-functionalized CeO 2 efficiently diminished Ag@SiO 2 ECL emission through electron transfer, resulting in a "signal-off" detection mode with high sensitivity and accuracy. The detection limit reached 1.66 fg/mL, and the detection range spanned from 100 fg/mL to 500 ng/mL, showcasing a powerful biomolecule detection strategy.

Abstract:Silver nanoparticles (Ag·NPs) show promising advantages in electrochemiluminescence (ECL) owing to their favorable optical properties and biocompatibility. However, their susceptibility to oxidation and degradation in the presence of air adversely affects ECL intensity. In this study, we employed a sandwich sensing platform using silica-coated silver nanoparticles (Ag@SiO2) as a novel luminescent material and cerium dioxide (CeO2) as an ECL signal quencher for sensitive neuro-specific enolase (NSE) detection. The core-shell structure protected Ag NPs within the silica (SiO2) layer, enhancing their ECL luminescence properties by reducing external environmental influence and preventing Ag NPs aggregation. Amino-functionalized CeO2 efficiently diminished Ag@SiO2 ECL emission through electron transfer, resulting in a "signal-off" detection mode with high sensitivity and accuracy. The detection limit reached 1.66 fg/mL, and the detection range spanned from 100 fg/mL to 500 ng/mL, showcasing a powerful biomolecule detection strategy.

Abstract:In this work, based on electrochemiluminescence (ECL) technology and self-assembled portable disease detection chips, a bioactivity-maintained sensing platform was developed for the quantitative detection of neuron-specific enolase. First, we prepared Fe3O4@MoS2 nanocomposites as an efficient catalyst to accelerate the reduction of persulfate (S2O82-). Specifically, abundant sulfate radicals (SO4•-) were generated because of cyclic conversion between Fe2+ and Fe3+. Meanwhile, MoS2 nanoflowers with a high specific surface area could not only load more Fe3O4 but also solve its agglomeration problem, which greatly improved the catalytic efficiency. Moreover, a biosensor chip was constructed by standard lithography processes for disease detection, which had good sensitivity and portability. According to the above strategies, the developed portable sensing platform played the part of promoting the practical application of bioanalysis in early tumor screening and clinical diagnosis. In addition, we developed a short peptide ligand (NARKFYKG, NAR) to avoid the occupation of antigen binding sites by specifically connecting to Fc fragments in antibodies. Thus, the binding efficiency of antibodies and the activity of biosensors were improved due to the introduction of NAR.

Abstract:A novel quenching electrochemiluminescence (ECL) immunosensor for trace detection of neuron-specific enolase (NSE) was developed. Doping CdS with Zn2+ to obtain ZnCdS not only significantly improves the original properties of CdS, but also can be loaded onto the surface of g-C3N4 to form a special complex structure, which effectively improves the ECL performance of g-C3N4. Rh0.6Ru0.4 alloys with flower-like structures were prepared by simple hydrothermal synthesis, with excellent electrical conductivity and catalytic performance. Due to its special flower-like structure with large specific surface area, Ag nanoparticles can be uniformly loaded on its surface to form Rh0.6Ru0.4@Ag composites. Through spectral comparison, it is found that Rh0.6Ru0.4@Ag has strong UV-vis absorption in the ECL emission wavelength range of g-C3N4, which can generate resonance energy transfer, thereby effectively quenching the ECL signal. The immunosensor has a wide detection range with a detection limit of 16 fg/mL (S/N=3), good stability and biocompatibility, and has great potential in clinical applications.

Abstract:In view of the importance of quantification of neuron-specific enolase (NSE), an electrochemical NSE immunosensor was developed. The sandwich voltammetric immunosensor utilized vinyl-functionalized crystalline covalent organic framework (COFTAPT-Dva) modified electrode to load lots of Ab1 via thiol-ene "click" reaction as matrix. A crystalline cationic EB-COF:Br was used to load Au nanoparticles (AuNPs) and H3[PMo12O40] (PMo12) as immunoprobe. The AuNPs with the size of about 30 nm were firstly grown on EB-COF:Br and then a large number of electroactive PMo12 were uniformly assembled on AuNPs/EB-COF:Br via ion exchanging reaction. The AuNPs not only facilitated the bonding of Ab2 based on Au–S bond, but also improved performance of Ab2/AuNPs/EB-COF:PMo12 immunoprobe. The sensitivity of sandwich electrochemical immunosensor could be primarily amplified based on loaded abundant PMo12. Secondary sensitivity amplification of immunosensor could be achieved by using PMo12 to catalyze ascorbic acid. The linear range of sandwich voltammetric immunosensor based on current change of differential pulse voltammetry is 500 ± 36 fg mL −1 - 100 ± 8 ng mL −1 . Thanks to the dual sensitivity amplification strategy, the sensitivity is as high as 54.06 ± 3.2 μA cm −2 /lg(cNSE/ng mL −1 ), and the detection limit is as low as 166 ± 10.8 fg mL −1 . It proves that it is completely feasible to amplify sensitivity of sandwich voltammetric immunosensors using polyoxometalate-COF and its catalytic substrate.

Restricted-Enhanced Electrochemiluminescence Biosensor of Low Excitation Potential Carbon Quantum Dots Driven by Magnetic Metal-Organic Frameworks for Sensitive Immunoassay of Neuron Specific Enolase

Abstract:Electrochemiluminescence (ECL) of highly efficient and non-toxic carbon quantum dots (CQDs) plays an important role in immunoassays. However, the high absolute value of the excitation potential and poor controllability make precise detection of low levels difficult. In this work, novel CQDs with low excitation potential were reported to be used for the ultra-sensitive immunoassay of neuron-specific enolase (NSE). It is noteworthy that the narrow bandgap CQDs exhibit a trigger potential of -0.5 V and show excellent applicability of ECL signal at an excitation potential of -0.8 V. This property eliminates side reactions and electrode passivation and significantly improves ECL efficiency. Based on this, the magnetic zeolite-imidazole framework (MZIF) nanoreactor with co-reaction acceleration was employed as a carrier for the in-situ encapsulation of CQDs. The constrained-enhanced ECL sensor platform demonstrated the ability to achieve stable and controllable signal output. A sandwich-type biosensor was constructed by introducing copper sulfide (Cu7S4) as a dual-mechanism quencher, with NSE serving as the target analyte. Moreover, the electrochemical detection method based on the differential pulse voltammetry (DPV) properties of Cu7S4, was successfully validated against ECL. The sensors constructed on magnetic electrodes exhibited a bimodal signal output of ECL and DPV, encompassing a wide detection range (0.001 ng/mL - 500 ng/mL) and low detection limit (0.18 pg/mL for ECL and 0.15 pg/mL for DPV). This design offers a novel approach to understanding the efficient emission of carbon dots and provides a solution for the sensitive detection of biomarkers in clinical research.
