Abstract:Pampus belongs to the Perciformes, Stromateoidei, and Stromateidae and are widely distributed throughout the Indian Ocean and the Western Pacific. In this study, we employed next-generation sequencing to generate comprehensive genomic data for Pampus punctatissimus and Pampus echinogaster and subsequently compared the obtained genomic information with the published data of Pampus argenteus . P. echinogaster was identified to be the largest genome size (602 Mb), heterozygosity (1.38%), and repetitive sequence ratio (33.06%), while similar genome sizes between P. punctatissimus (540 Mb) and P. argenteus (550 Mb) were observed. A total of 402,921 and 628,298 SSRs were identified for P. punctatissimus and P. echinogaster , respectively. Additionally, 1,876, 638, and 394 single-copy homologous genes were identified from P. punctatissimus , P. echinogaster , and P. argenteus , respectively. The KEGG enrichment results of the unique single-copy genes to the three species were significantly different in genetic information processing, protein processing, and metabolic functions. The complete mitochondrial genomes of P. punctatissimus and P. echinogaster were successfully assembled, and the resulting phylogenetic relationships align with the currently reported taxonomic status of species in the Stromateoidei. Additionally, population size analysis using PSMC revealed that P. punctatissimus experienced one bottleneck effect, while P. echinogaster underwent two bottleneck events during the Pleistocene Glacial Epoch.

Abstract:Penaeus monodon is a crucial economic crustacean globally and a focal point of aquatic research. Germplasm resources provide a diverse food source for humans, meeting various nutritional needs; they also maintain plant and animal gene banks, enhancing resilience, adaptability, and promoting ecological balance in terms of genetic diversity. To accurately assess the germplasm resources of P. monodon, we conducted a comprehensive investigation. We analyzed muscle characteristics, nutrient content, amino acids, fatty acids, and genetic diversity using whole-genome sequencing in six P. monodon groups. The results showed that the crude protein content of the six groups of P. monodon was 21.17–24.60 g/100 g, crude fat content was 0.60–0.80 g/100 g, essential amino acid content was 5.44–6.41 g/100 g, total amino acid content was 15.68–18.63 g/100 g, and total fatty acid content was 354.78–587.43 mg/100 g. Genetic differentiation index (Fst) values ranged from 0.029 to 0.604 for six groups. PmSZ has a good genetic condition. There was no significant difference in crude fat in conventional nutrients, and the difference in biochemical indexes T-AOC, CAT, and SOD was not significant (P > 0.05). The noteworthy point is that the analysis of genetic diversity reveals a moderate level of genetic diversity. PmDF and PmZH exhibit the highest degree of differentiation, while PmRP and PmZH show the lowest degree of differentiation. Our survey results establish a critical foundation for comprehending and harnessing the germplasm resources essential for selectively breeding robust P. monodon populations.
Species diversity and distribution of genus Pampus (Pelagiaria: Stromateidae) based on global mitochondrial data

Abstract:Due to the highly similar external morphology of the Pampus species, misidentifications frequently occur and hinder the understanding of the taxonomy and species distributions of the genus. In this study, we generated 271 mitochondrial sequences and obtained 1,226 sequences from the public databases to understand the species diversity and distributions of the genus Pampus . Most phylogenetic analyses and species delimitations congruently concluded seven valid species within the genus Pampus (i.e., P. argenteus , P. candidus , P. chinensis , P. cinereus , P. liuorum , P. minor , and P. punctatissimus ). Sequences of P. argenteus are reported through the northern South China Sea to the Japan Archipelago, which covered the type locality of P. echinogaster . Sequences of P. echinogaster in the public databases are all identified as P. argenteus , suggesting that the species should be synonymized with P. argenteus . Furthermore, sequences of P. griseus were identical to our P. cinereus data and, therefore, should be treated as a synonym of the latter. Inference on divergence time and ancestral distribution implied that the genus Pampus originated in the central Indo-Pacific region around 8.35–11.33 million years ago (the late Miocene), associated with the rise of the Indonesian–Australian Archipelago biodiversity hotspot. The divergence between P. cinereus and P. liuorum dated back to 1.20–1.72 million years ago, which might be related to glacial isolation during the Mid-Pleistocene transition.

Abstract:Pomfrets (genus Pampus ), a highly commercial fishery resource distributed in the Indo-Western Pacific that includes Lessepsian migrants, have witnessed a series of systematic reforms. In this study, based on comprehensive sampling spanning type localities and coevals in the Northern Indian Ocean, the cryptic and valid species Stromateus griseus is resurrected from the synonymy and re-described as Pampus griseus (New Combination) based on 35 specimens from the Bay of Bengal, corroborated by a molecular analysis, which indicated a confined distribution of the species. The Bayesian phylogeny of the genus was reconstructed, incorporating redressed barcodes (582 nucleotides) and concatenated mitochondrial gene sequence data (1,822 nucleotides) generated from the recorded species P . candidus , P . chinensis and the neophyte along with sequences from GenBank entrusting the latest literature. The phylograms differed in topology as for seven valid species, and the one predicated on the concatenated data erected a highly supported polytomous clade for the P . cinereus complex ( P . griseus , P . cinereus , and P . candidus ) which shares synapomorphies. Pampus argenteus and P . minor , together, formed a sister clade to the rest. Climate-driven vicariant events during glacial epochs and the Indo-Pacific Barrier effect can be the drivers behind the Indian and Pacific Ocean sister lineages in P . chinensis . A multivariate analysis isolated the cryptic species from its congeners. This article portrays the systematics revision of genus Pampus with an integrative taxonomic approach compiling distinctive molecular, morphological, and anatomical features, revised key for species identification, taxonomic archives of Indian stromateids, and winds up with specific remarks.

Abstract:The parrot grunt fish, Pomadasys perotaei , has a limited distribution in the Eastern Atlantic Ocean and is an important species in marine capture fisheries across several West African countries. Despite its ecological and economic significance, the mitogenomic information for this species is lacking. This study utilized next-generation sequencing to generate the de novo mitogenome of P. perotaei from Eastern Atlantic. The resulting mitogenome is 16,691 base pairs and includes 13 protein-coding genes (PCGs), 22 transfer RNAs, two ribosomal RNAs, and an AT-rich control region (CR). Most of the PCGs exhibit nonsynonymous (Ka) and synonymous (Ks) substitution rates of less than '1', indicating strong negative selection across haemulid fishes. The control region of Pomadasys species contains four conserved domains, as seen in other teleost's, with polymorphic nucleotides that can be used to study population structures through the amplification of short mitochondrial gene fragments. Additionally, Bayesian phylogenetic analysis based on PCGs revealed a non-monophyletic clustering pattern of Pomadasys within the haemulid matrilineal tree. Overall, the structural characterization and phylogenetic analysis enhance our understanding of the genetic composition and evolutionary history of Pomadasys species from the Indo-West Pacific and Eastern Atlantic Oceans.

Abstract:Pampus argenteus (Euphrasen, 1788) is one of the major fishery species in coastal China. Pampus argenteus has a highly specialized morphology, and its declining fishery resources have encouraged massive research efforts on its aquacultural biology. In this study, we reported the first high-quality chromosome-level genome of P. argenteus obtained by integrating Illumina, PacBio HiFi, and Hi-C sequencing techniques. The final size of the genome was 518.06 Mb, with contig and scaffold N50 values of 20.47 and 22.86 Mb, respectively. The sequences were anchored and oriented onto 24 pseudochromosomes based on Hi-C data corresponding to the 24-chromatid karyotype of P. argenteus . A colinear relationship was observed between the P. argenteus genome and that of a closely related species ( Scomber japonicus ). A total of 24,696 protein-coding genes were identified from the genome, 98.9% of which were complete BUSCOs. This report represents the first case of high-quality chromosome-level genome assembly for P. argenteus and can provide valuable information for future evolutionary, conservation, and aquacultural research.

Abstract:Despite the critical role of mitochondrial genomes (mitogenomes) in species identification and evolutionary studies in the genus Parupeneus , current resources are inadequate, given the species richness. Although previous studies have suggested a complex evolutionary history, the detailed mitogenomic variations and their implications remain largely unexplored. Therefore, we sequenced and assembled the mitogenomes of P . barberinoides , P . barberinus , P . biaculeatus , P . crassilabris , P . cyclostomus , P . heptacanthus , P . multifasciatus , and P . chrysopleuron , to enrich the molecular data and provide novel insights into the genetic diversity, evolutionary dynamics and phylogenetics of the family Mullidae. Our analysis revealed a novel gene rearrangement in P . chrysopleuron , Cytb - T - P -CR- Q - I - F -12S- V -16S- ND1-M - ND2 , which differed from the conventional sequence of Cytb - T - P -CR- F -12S- V -16S- ND1 - I - Q - M - ND2 observed in other species. In the novel rearrangement, four non-coding regions are inserted between ND1 and M , Q and I , I and ψ M (tRNA- Met pseudogene), ψ M and F . We assume that two tandem duplication/random loss events occur in the CR and IQM , making the entire sequence longer than that in other Parupeneus species. The phylogenetic results indicated that Mullidae formed a sister group relationship with the family Dactylopteridae, contradicting previous studies that identified a sister group relationship between Mullidae and Callionymoidei. The genera Parupeneus and Mullus formed a sister group, and discrepancies were found in the topological structure of the interspecies relationships within the genus Parupeneus compared with those reported by previous studies. Through combined phylogenetic and mitochondrial structural analysis, we found that phylogenetic topology is closely related to mitochondrial structural abnormalities. This study not only expands the mitogenomic dataset available for Mullidae but also underscores the importance of mitochondrial DNA studies in resolving taxonomic ambiguities and understanding the evolutionary history of marine fishes. Our study contributes to the ongoing research on marine fish taxonomy, mitogenomics, and evolutionary biology by providing new insights into the genetic diversity of marine ecosystems.

Unraveling the Phylogenetic Relationships and Taxonomic Status of a Puzzling Freshwater Mussel Genus Inversidens (Bivalvia, Unionidae) through Multilocus Phylogeny and Mitochondrial Phylogenomics

Abstract:Accurate phylogenetic reconstructions are crucial for comprehending the evolutionary histories, reproductive traits, and ecological habits of organisms. The subfamily Gonideinae of freshwater mussels is currently thought to include eight tribes. However, due to inadequate taxon sampling and molecular data, the assignment of the freshwater mussel genus Inversidens at the tribe level has been unstable. Additionally, the lack of phylogenetic data has hindered efforts to understand the basic biology and implementation of conservation efforts of Inversidens rentianensis, an endemic species to China. Here, we first present the complete mitochondrial genome of Inversidens rentianensis and offer a detailed description of its anatomical morphology. Based on DNA sequence data from five genes (COI, ND1, 16S rRNA, 18S rRNA, and 28S rRNA) and complete mitochondrial genomes, we investigated the phylogenetic position of Inversidens using various analytical methods. Both the concatenated five-gene and mitogenome datasets strongly supported that Inversidens classified to the tribe Gonideini in Gonideinae and formed a basal clade within the tribe Gonideini. Molecular dating analysis suggested that Inversidens originated during the mid-Cretaceous era (102.73 Mya, 95% highest posterior density HPD = 72.22 ‐ 137.03 Mya) and underwent diversification in the Late Paleogene era (37.92 Mya, 95% HPD = 20.39 ‐ 60.59 Mya). Moreover, based on the Quantitative Assessment of Species for Conservation (QASCP), Inversidens rentianensis is ranked as second priority, providing valuable insights for its management and conservation efforts. Taken together, this study provides a comprehensive understanding of the systematic position and evolutionary history of Inversidens within the currently accepted subfamily Gonideinae classification framework. These findings establish a solid foundation for future investigations on the ecology, reproductive behavior patterns, and conservation biology of this taxonomic group.

Abstract:The Amur sleeper Perccottus glenii (Perciformes, Gobioidei, Odontobutidae) is well known as an invasive fish in the river basins of Eastern and Central Europe, but its genetic background is unavailable across its native habitats in northeast Asia. In this study, we used the mitochondrial cytochrome b gene by sampling 19 populations of P. glenii across its native distributional areas of Liaohe and Amur River basins to explore its evolutionary history. Phylogenetic analyses identified three major clades within P. glenii, among which Clade A and Clade B were co-distributed in the Liaohe and Amur River basins, and Clade C was restricted to the latter. Molecular dating showed that the splits of Clades A, B and C have happened in the late Early-early Middle Pleistocene and the most recent common ancestors of these clades have been presented in the late Middle-early Late Pleistocene. The P. glenii showed very high levels of genetic structure among populations (ΦST = 0.801), probably due to the characters of its life histories with very limited dispersal ability. The admixture of different clades in some populations of P. glenii probably reflects historical secondary contact. These findings indicate that Pleistocene climatic oscillation and river capture were major determinants for genetic variations and evolutionary history of the P. glenii.
Complete Mitochondrial Genome of Four Peristediidae Fish Species: Genome Characterization and Phylogenetic Analysis

Abstract:The systematic revision of the family Peristediidae remains an unresolved issue due to their diverse and unique morphology. Despite the popularity of using mitochondrial genome research to comprehensively understand phylogenetic relationships in fish, genetic data for peristediid fish need to be included. Therefore, this study aims to investigate the mitochondrial genomic characteristics and intra-family phylogenetic relationships of Peristediidae by utilizing mitochondrial genome analysis. Therefore, this study aims to investigate the phylogenetic relationship of Peristediidae by utilizing mitochondrial genome analysis. The mitochondrial genome of four species of Peristediidae (Peristedion liorhynchus, Satyrichthys welchi, Satyrichthys rieffeli, and Scalicus amiscus) collected in the East China Sea was studied. The mitochondrial gene sequence lengths of four fish species were 16,533 bp, 16,526 bp, 16,527 bp, and 16,526 bp, respectively. They had the same mitochondrial structure and were all composed of 37 genes and one control region. Most PCGs used ATG as the start codon, and a few used GTG as the start codon. An incomplete stop codon (TA/T) occurred. The AT-skew and GC-skew values of 13 PCGs from four species were negative, and the GC-skew amplitude was greater than that of AT-skew. All cases of D-arm were found in tRNA-Ser (GCT). The Ka/Ks ratio analysis indicated that 13 PCGs were suffering purifying selection. Based on 12 PCGs (excluding ND6) sequences, a phylogenetic tree was constructed using Bayesian inference (BI) and maximum likelihood (ML) methods, providing a further supplement to the scientific classification of Peristediidae fish. According to the results of divergence time, the four species of fish had apparent divergence in the Early Cenozoic, which indicates that the geological events at that time caused the climax of species divergence and evolution.

Abstract:Catillopecten is a small genus of deep-sea glass scallops, but its diversity is poorly known in many parts of the world ocean. We described C. margaritatus n. sp. (Pectinoidea: Propeamussiidae), and performed morphological analyses and DNA sequencing, and estimated the divergence time of scallops based on samples collected from Haima cold seep in the South China Sea. Morphologically, the new species can be distinguished from congeneric species by its large shell size, relatively small auricle length, absence of without monocrystal aerials, presence of longitudinal radial ridges on the left valve, and the alternated rounded striae and distal and proximal growth lines of prisms on the right valve. Anatomically, this new species can be distinguished from C. vulcani by its anteriorly located auriculate gills, compared to the centrally located lamellar gills of the latter, and the different locations of the pericardium. Sequence comparison and phylogenetic analysis based on the 18S rRNA fragments supported the placement of the new species in Catillopecten . We also report the mitogenome of C. margaritatus n. sp. as the only reported mitogenome of the family Propeamussiidae, which differs from those of other scallops substantially in gene order arrangement. Divergence time estimation revealed that Propeamussiidae and Pectinidae diverged in the early Carboniferous, while Catillopecten and Parvamussium diverged during the late Cretaceous to early Eocene. Finally, we presented a key to the species of Catillopecten .

Abstract:Pimelodidae is a family of large‐sized catfishes consisting of 116 valid species divided into 30 genera distributed in the Neotropical region. Despite recent advancements in understanding the pimelodid phylogeny, several internal relationships have remained unclear and have received limited support from various phylogenetic analyses. We provide a new genomic dataset comprising 55 species representing 25 genera of Pimelodidae and related siluriform families Aspredinidae, Auchenipteridae, Cetopsidae, Doradidae, Diplomystidae, Heptapteridae, Loricariidae, and Pseudopimelodidae. Our phylogenetic analyses support the monophyly of Pimelodidae, as well as the existence of four major lineages: the Steindachneridion clade, the Leiarius + Phractocephalus clade, and two major subclades herein classified as Sorubiminae and Pimelodinae. Our topology resolves Phractocephalus sister to Leiarius differing from previous phylogenies. In Sorubiminae, Zungaro is sister to two large clades: one with Sorubim, Sorubimichthys, and Pseudoplatystoma, and the other with Hemisorubim, Platysilurus, Platystomatichthys, Brachyplatystoma, and Hypophthalmus. In Pimelodinae, we found the Pimelodus ornatus clade, the calophysines, and the clade with Exallodontus, Propimelodus, and Pimelodus altissimus closer to the Pimelodus group. Parapimelodus, Duopalatinus, Iheringichthys, and Bergiaria are included within the Pimelodus group. The Pimelodus group contains four main components: (1) Duopalatinus peruanus, (2) the trans‐Andean Pimelodus grosskopfii; (3) Pimelodus maculatus clade with species from the Brazilian Shield in southeastern Brazil; and (4) Pimelodus blochii clade composed by species from the Amazon, Orinoco, Paraná‐Paraguay, Parnaíba, São Francisco and coastal drainages of the Guiana Shield. The P. maculatus clade is composed by phenotypically diverse species, and the P. blochii clade houses species morphologically similar that appear nested within the widely distributed P. blochii.

Abstract:The species in the genus Pristipomoides are commercial fishes with high economic value. There are few studies on the phylogenetic relationship within the genus Pristipomoides at present. mtDNA has the characteristics of maternal inheritance, close gene arrangement, and a high evolutionary rate, which is an excellent tool to study the phylogeny of the species. In this study, the mitochondrial genomes of five species in the genus Pristipomoides were sequenced using the Sanger sequencing method and analyzed for their features. The mitochondrial genome length of the five species ranged from 16,499 to 16,530 bp. The start codon was ATG\GTG, and the stop codon was TAA\TAG\T--\AGG. The ratio of Ka and Ks for protein-coding genes ranged from 0 to 0.117, suggesting a strong purifying selection acting on the 13 protein-coding genes (PCGs). The gene with the highest diversity was nd2 (NADH dehydrogenase subunit 2), suggesting the highest evolutionary rate. Phylogenetic analysis of five Pristipomoides species with other species of Lutjanidae was conducted using maximum likelihood (ML) and Bayesian Inference (BI). The results showed that P. zonatus and P. auricilla were closely related to P. argyrogrammicus and P. sieboldii, respectively, and P. filamentosus and P. multidens clustered together. Furthermore, A. rutilans is deeply nested within the Pristipomoides genus, indicating a close phylogenetic relationship with the species in the Pristipomoides genus. Based on this evidence, we suggest that A. rutilans should be classified under the Pristipomoides genus and recommend a revision in its taxonomy. The molecular data and phylogenetic analysis provided in this study would be helpful for the species identification and phylogenetic studies of the family Lutjanidae.

Abstract:Order Acipenseriformes contains 27 extant species distributed across the northern hemisphere, including so-called "living fossil" species of garfish and sturgeons. Previous studies have focused on their mitochondrial genetics and have rarely used nuclear genetic data, leaving questions as to their phylogenetic relationships. This study aimed to utilize a bioinformatics approach to screen for candidate single-copy nuclear genes, using transcriptomic data from sturgeon species and genomic data from the spotted gar, Lepisosteus oculatus. We utilized nested polymerase chain reaction (PCR) and degenerate primers to identify nuclear protein-coding (NPC) gene markers to determine phylogenetic relationships among the Acipenseriformes. We identified 193 nuclear single-copy genes, selected from 1850 candidate genes with at least one exon larger than 700 bp. Forty-three of these genes were used for primer design and development of 30 NPC markers, which were sequenced for at least 14 Acipenseriformes species. Twenty-seven NPC markers were found completely in 16 species. Gene trees according to Bayesian inference (BI) and maximum likelihood (ML) were calculated based on the 30 NPC markers (20,946 bp total). Both gene and species trees produced very similar topologies. A molecular clock model estimated the divergence time between sturgeon and paddlefish at 204.1 Mya, approximately 10% later than previous estimates based on cytochrome b data (184.4 Mya). The successful development and application of NPC markers provides a new perspective and insight for the phylogenetic relationships of Acipenseriformes. Furthermore, the newly developed nuclear markers may be useful in further studies on the conservation, evolution, and genomic biology of this group.

Abstract:Trois types de crises partielles complexes (CPS) ont été décrits d'après le comportement observable au début de la crise. Les CPS type I débutent par une fixité du regard avec immobilité et ont été correlées avec l'existence d'un foyer temporal. Les CPS type 2 ne débutent pas par un regard fixe et une immobilité et ont été correlées avec un foyer extratemporal. Un 3éme type de CPS, la syncope du lobe temporal a été correlée à des foyers temporaux mésiaux bilatéraux. Nous avons étudié l'utilité de cette classification des CPS pour la prédication des résultats du traitement chirurgical en révisant les observations avec nos patients présentant des CPS rebelles au traitement médical et opérés par excision du foyer épileptogène. 46 patients consécutifs sontéudiés dont le foyer épileptogène a été démontréêtre temporal dans 41 cas et frontal dans 5. Les 5 sujets ayant un foyer frontal avaient des CPS type 2. Sur les 41 avec foyer temporal, 20 avaient des CPS type 1 et 21 des CPS type 2. 20 des 26 patients avec CPS type 2 et 18 des 20 patients avec CPS type 1 ont eu une évolution post‐opératoire bonne ou excellente. Nos résultats suggérent que les patients ayant un foyer frontal ont des CPS type 2, mais l'inverse n'est pas vrai. De plus, le type des CPS n'est pas correlé avec l'évolution post‐opératoire, puisqu'il n'y a pas de différence significative de catégorie d'évolution post‐opératoire entre les types de CPS.

Abstract:Pampus argenteus is a highly commercial marine fish whose population is declining sharply. Here, we generated a female P. argenteus genome, spanning 536.33 Mb with contig N50 of 1.79 Mb; 24070 genes (99.50% of 24,182) were functionally annotated. To improve quality of it, we assembled a 553.79 Mb genome of male fish with contig N50 of 24.75 Mb through HiFi and ultra-long ONT sequence technologies; 550.82 Mb were anchored onto 24 gap-free chromosomes; 22,892 genes (98.1% of 23,346) were functionally annotated; the QV value was 51.55 with 98.9% of BUSCO and 99.39% coverage of Illumina reads. Finally, we compared this genome with previous published one, revealing 37,301 SVs. 52.82 Mb and 18.05 Mb SDs were characterized in our and published assemblies, respectively, and 48.96 Mb PURs were constructed. Thus, this genome assembly exhibits excellent completeness, continuity and accuracy comparing to the published one, which can be current preferred reference genome. Overall, these works help aquaculture and wild resources recovery of P. argenteus and provide a valuable genetic resource for study.

Extensive mitogenomic heteroplasmy and its implications in the phylogeny of the fish genus Megalobrama

Abstract:Megalobrama is China's most economically valuable fish genera. Four species make up this genus: M. amblycephala (MA), M. terminalis (MT), M. pellegrini (MP), and M. hoffmanni (MH). Many researchers have investigated the genetic relationship of Megalobrama based on mitochondrial DNA (mtDNA) and discovered that the branches of the phylogenetic tree for MT and MP are intertwined. We hypothesized that this occurs because mitogenomic heteroplasmy is overlooked when working with mtDNA, which causes MP and MT positions to intersect in phylogenetic trees. To eliminate the influence of nuclear mitochondrial DNA fragments (NUMTs) before analyzing mitogenomic heteroplasmy, we used PLastZ to identify NUMTs, which were then removed from the samples for the subsequent heteroplasmy analysis. Using the heteroplasmy caller icHET, we discovered 126, 339, 135, and 203 heteroplasmic variants in six MA, MT, MP, and MH samples. We reconstructed the Megalobrama fish genus's phylogenetic tree using the RY coding method and rejecting the third position on codons, which improved the performance of the phylogenetic tree by increasing the ratio of treeness to relative component variability from 100.02 ± 1.76 to 688.59 ± 190.56. Despite this, the RY coding method cannot alter the intersection of MP and MT positions in phylogenetic trees. We hypothesize that gene flow between MT and MP leads to intertwining mtDNA-based phylogenetic trees. In conclusion, our findings on the mitogenomic heteroplasmy of Megalobrama provide new insights into mtDNA-based phylogenetic studies.

Abstract:Pimelodus is the most speciose genus of the family Pimelodidae, and is amply distributed in the Neotropical region. The species-level taxonomy and phylogenetic relationships within this genus are still poorly resolved, however. These taxonomic problems and the general lack of data have generated major uncertainties with regard to the identification of specimens from different localities. In the present study, we applied a single-locus species delimitation approach to identify the MOTUs found within the genus Pimelodus and provide sound evidence for the evaluation of the species richness of this genus in the different river basins of the Neotropical region. The study was based on the analysis of sequences of the mitochondrial COI gene of 13 nominal species, which resulted in the identification of 24 consensus MOTUs. Only six nominal species were recovered as well-defined molecular entities by both the traditional barcoding analysis and the molecular delimitation methods, while the other seven presented cryptic diversity or persistent taxonomic uncertainties. The lineages identified from the Parnaíba ecoregions, Amazonas Estuary and Coastal Drainages may represent a much greater diversity of Pimelodus species than that recognized currently, although a more detailed study of this diversity will be necessary to provide a more definitive classification of the genus.
