Abstract:Minors et al. (1991) recently published the first phase response curve (PRC) for single light pulses in human subjects, adding substantially to earlier measurements by Honma and Honma (1988). Another PRC, using multiple pulses, has been published by Czeisler et al. (1989). There is an apparent discrepancy among the different PRCs now available. In particular, the PRC of Minors et al. (1991) shows weak (type 1) phase resetting (Winfree, 1980), whereas Czeisler et al. (1989) have claimed strong (type 0) phase resetting in humans. This apparent inconsistency calls for a better understanding of the effects of multiple versus single pulses. Abstract:OBJECTIVE:To analyze the chemical components of the essential oil extracted from the seeds of Myristica fragrans (nutmeg) processed by different methods (steamed with water steam, roasted with flour, sauted with flour, roasted with talcum powder, roasted with loess, and roasted with bran) and to provide quality control foundations in the sciences.METHOD:The essential oil was extracted by steam distillation and separated with GC capillary column. The relative content of every compound was determined with area normalization method and the structures were elucidated by GC-MS technique.RESULT:Fifty-eight to one hundred and four of chromatographic peaks were detected, among them seventy-six compounds accounting for 98.32% to 99.99% of the total essential oil in nutmeg were identified, which were composed of 69.15% to 97.24% for monoterpenoids and 2.06% to 25.51% for aromatic compounds of the total essential oil, respectively.CONCLUSION:It was shown that monoterpenoids and their derivatives were main composition, and aromatic compounds were secondary composition in the total essential oil of nutmeg grows in Indonesia and processed by different traditional methods on the basis of theory of traditional Chinese medicine. In addition, it was suggested that we should be careful to use processed nutmeg owing to contain safrole and a-asarone induced genetoxicity in animals and mutagenicity in the Ames Salmonella assay, and myristicin and elemicin induced narcotism in human. The processed method roasted with bran for nutmeg may be better and will be developed.
GC-MS Analysis of Essential Oil from the Seeds of Myristica Fragrans Houtt in Indonesia

Abstract:Objective: To analyze the chemical constituents of the essential oil extracted from the seeds of Myristica fragrans Houtt.(nutmeg) in Indonesia and to provide qualities control scientific foundations.Methods: The essential oil was extracted by steam distillation and separated with GC capillary column.The relative content of every compound was determined by normalization method and the chemical compositions were evaluated by GC-MS.Results: Ninety-three chromatographic peaks were isolated and sixty-one compounds were identified,accounting for 97.62% of the total essential oil in nutmeg.Conclusion: The essential components of nutmeg in Indonesia is monoterpenoids and it's derivatives,accounting for 78.87% of the total essential oil.And phenolic compounds were the secondary composition,accounting for 16.79%.The essential oil contains safrole,which can induce genetoxicity and mutagenicity in animal,and myristicin and elemicin which can induce illusion.So,it must be careful when Myristica fragrans Houtt was used.
Determination of Nonylphenol in Selected Foods and Identification of Single Isomers in a Coffee Sample by Comprehensive Two-Dimensional Gas Chromatography-Time of Flight Mass Spectrometry

Abstract:Nonylphenols (NPs) are a degradation product of nonylphenol ethoxylate surfactants, which occur in many samples from the environment, food, and human organism. NPs are persistent and toxic and exhibit estrogenic activity. One of the challenges in understanding the estrogenic effects of NPs is the large number of isomers that may exist in different samples. After analyzing selected foods from different countries by high-performance liquid chromatography coupled with tandem mass spectrometry, NPs were detected in 8 of the 12 food samples. The concentration of NPs, based on fresh weight, ranged from 0.5 to 18.9 μg kg −1 . The highest NP contents were found in coffee samples, and therefore an isomer-specific analysis was carried out by comprehensive two-dimensional gas chromatography coupled with time-of-flight mass spectrometry (GCxGC-TOF-MS). Fourteen pure isomers of NP were used as standards for cross-referencing the NP isomers. We identified the two main NP isomers in the coffee samples as 4-[3-ethyl-1,1-dimethylpentyl]-phenol and 4-[1-ethyl-1,3-dimethylpentyl]-phenol or NP 128 and NP 111 (Juelich nomenclature), respectively. The GCxGC-TOF-MS system with a liquid nitrogen cryogenic modulator used in this study is suitable for the isomer-specific analysis of NP and can be applied and further optimized for future routine analysis in food control.

Abstract:Clinopodium nepeta subsp. glandulosum (Req.) Govaerts (Lamiaceae), is traditionally used as antispasmodic, diaphoretic, stimulant, and tonic medicinal herbs and as mint-like spice used in various culinary recipes. Nonetheless, there is still a dearth of studies to validate its biological properties. The total phenolic and flavonoid contents, scavenging activity of DPPH and ABTS radicals, ferric reducing antioxidant power (FRAP), total antioxidant via phosphomolybdenum assay, metal ion chelating capacity of different extracts ( n- hexane, dichloromethane, ethyl acetate, ethanol, ethanol/water and water) of C. nepeta were determined. The water extract contained the highest phenolic contents (51.47 ± 0.62 mg GAE/g) and ethanol/water extract yielded largest amount of flavonoid contents (43.86 ± 0.63 mg RE/g). The DPPH free radical scavenging activity of the extracts was in the order of ethanol/water > water > ethanol > ethyl acetate > dichloromethane > n-hexane. The enzyme inhibition of the different extracts was also investigated. Ethanol/Water extract demonstrated the highest AChE inhibitory capacity (2.66 mg GALAE/g) while ethyl acetate extract revealed high BChE inhibitory activity (3.29 mg GALAE/g). Exhaustive 2D-NMR investigation has revealed the presence of acacetin derivatives, ursolic acid along with and 2,4-di- t -butylphenol. These findings revealed that extracts of C. nepeta could be used as a promising therapeutic agent against free radicals, brain enzymes including AChE and BChE and in the treatment of hyperpigmentation as a skin whitening ingredient as well as in the treatment and management of diabetes. However, further studies are required to determine the safety profiles of these extracts.

Abstract:The aril and seed of nutmeg, Myristica fragrans Houtt. (Myristicaceae), hold significant value in various industries globally. Our preliminary research found two morphological variations: a globose shape and an oval shape. Due to these different characteristics, the safety of consumers is of primary concern. Thus, authentication and comparative pharmacological and toxicity analyses are necessary. In this study, pharmacognostic and advanced phytochemical analyses, DNA barcoding, cytotoxicity, and the anti-nitric oxide production of commercial Thai nutmeg were examined. Via morphologic examinations and TLC fingerprinting, all the sampled aril and seed were categorized into globose and oval-shaped groups. The results of HPLC, GC-MS, and LC-MS/MS experiments revealed distinct differences between these groups. The DNA barcoding of the trnH-psbA region using the BLAST method and neighbor-joining tree analyses confirmed the globose nutmeg as M. fragrans and the oval-shaped variant as M. argentea. A comparison was then carried out between the potential toxicity and anti-inflammatory capabilities of M. fragrans and M. argentea. Cytotoxicity tests on HaCaT, 3T3-L1, Caco-2, HEK293, and RAW264.7 were performed using both methanolic extracts and volatile oil from the arils and seeds of both species. This study concludes that blending or substituting these two species maintains their therapeutic integrity without posing safety concerns.

Abstract:This work aimed to isolate the essential oil from the nutmeg ( Myristica fragrans Houtt.) seed, to determine the qualitative and quantitative chemical composition of the obtained essential oil, as well as to examine its antioxidant and antimicrobial activity. The essential oil was obtained by Clevenger-type hydrodistillation from the disintegrated nutmeg seed (hydromodulus 1:15 m/V, extraction time 180 minutes). The qualitative and quantitative chemical composition of the nutmeg essential oil was determined by GC/MS and GC/FID analysis. Twenty-five compounds were identified in the nutmeg essential oil and the most abundant was monoterpene hydrocarbon sabinene (42.3 %). The antioxidant activity was spectrophotometrically examined by using DPPH assay. The nutmeg essential oil showed a good antioxidant activity after incubation (EC 50 = 1.35 ± 0.003 mg/ml). The antimicrobial activity of the nutmeg essential oil was examined by using a disc diffusion method on Gram-positive bacteria (Staphylococcus aureus, Bacillus cereus, B. luteus, Listeria monocytogenes), Gram-negative bacteria (Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, Proteus vulgaris) and fungus (Candida albicans). The essential oil showed good antifungal and antibacterial activity. The anticandidal activity of the essential oil (28 mm) was comparable to the activity of conventional antifungal drug nystatin (17 mm). Of the tested Gram-positive bacteria, the most sensitive was B. luteus with the inhibition zone diameter of 23 mm, while the tested Gram-negative bacteria showed lower sensitivity. The isolated nutmeg essential oil has the potential for use in the pharmaceutical, chemical and food industry as a natural antioxidant and an antimicrobial agent.

Abstract:Culinary spices and herbs have been used to impart a characteristic flavour and aroma in food due to their appealing fragrance. Recently, bioactive compounds from herbs, especially phenolics, have gained much attention due to their potential health outcomes. The aim of this study was to characterize and quantify the phenolic compounds from 10 widely used Australian-grown herbs (oregano, rosemary, bay, basil, sage, fenugreek, dill, parsley, mint and thyme). For this purpose, liquid chromatography mass spectrometry (LC-MS) was used for the complete profiling of polyphenolic compounds and quantification of abundant phenolic compounds was completed with high-performance liquid chromatography—photodiode array detection (HPLC-PDA). Polyphenols from Australian-grown herbs were estimated through total phenolic content (TP), total flavonoids (TF) and total tannins (TT) along with their in-vitro antioxidant activities. Oregano and mint were estimated with the highest value of TP (140.59 ± 9.52 and 103.28 ± 8.08 mg GAE/g, milligram gallic acid equivalent/gram) while rosemary and mint had the highest TF (8.19 ± 0.74 and 7.05 ± 0.43 mg QE (quercetin equivalent)/g). In this study, eighty-four (84) phenolic compounds were screened and confirmed through LC-MS/MS by comparing their masses and fragmentation pattern with published libraries. The results of this study validate the use of these herbs as bioactives and their positive impact on human health.

Abstract:In recent years, nutmeg (Myristica fragans Houtt.) has attracted considerable attention in the field of phytochemistry due to its diverse array of bioactive compounds. However, the potential application of nutmeg as a biorational for crop protection has been insufficiently explored. This study investigated the constituents of a nutmeg hydroethanolic extract via gas chromatography-mass spectrometry and vibrational spectroscopy. The research explored the extract's activity against phytopathogenic fungi and oomycetes, elucidating its mechanism of action. The phytochemical profile revealed fatty acids (including tetradecanoic acid, 9-octadecenoic acid, n-hexadecanoic acid, dodecanoic acid, and octadecanoic acid), methoxyeugenol, and elemicin as the main constituents. Previously unreported phytochemicals included veratone, gelsevirine, and montanine. Significant radial growth inhibition of mycelia was observed against Botrytis cinerea, Colletotrichum acutatum, Diplodia corticola, Phytophthora cinnamomi, and especially against Fusarium culmorum. Mode of action investigation, involving Saccharomyces cerevisiae labeled positively with propidium iodide, and a mutant strain affected in ERG6, encoding sterol C-24 methyltransferase, suggested that the extract induces a necrotic type of death and targets ergosterol biosynthesis. The evidence presented underscores the potential of nutmeg as a source of new antimicrobial agents, showing particular promise against F. culmorum.

Abstract:Eugenol is a phenolic aromatic compound obtained mainly from clove oil. Due to its known antibacterial, antiviral, antifungal, anticancer, anti-inflammatory and antioxidant properties, it has long been used in various areas, such as cosmetology, medicine, and pharmacology. However, high concentrations can be toxic. A dose of 2.5 mg/kg body weight is regarded as safe. This paper reviews the current state of knowledge regarding the activities and application of eugenol and its derivatives and recent research of these compounds. This review is based on information concerning eugenol characteristics and recent research from articles in PubMed. Eugenol remains of great interest to researchers, since its multidirectional action allows it to be a potential component of drugs and other products with therapeutic potential against a range of diseases.

Abstract:Natural products, known for their medicinal properties since antiquity, are continuously being studied for their biological properties. In the present study, we analyzed the composition of the volatile preparations of essential oils of the Greek plants Ocimum basilicum (sweet basil), Mentha spicata (spearmint), Pimpinella anisum (anise) and Fortunella margarita (kumquat). GC/MS analyses revealed that the major components in the essential oil fractions, were carvone (85.4%) in spearmint, methyl chavicol (74.9%) in sweet basil, trans-anethole (88.1%) in anise, and limonene (93.8%) in kumquat. We further explored their biological potential by studying their antimicrobial, antioxidant and antiproliferative activities. Only the essential oils from spearmint and sweet basil demonstrated cytotoxicity against common foodborne bacteria, while all preparations were active against the fungi Saccharomyces cerevisiae and Aspergillus niger. Antioxidant evaluation by DPPH and ABTS radical scavenging activity assays revealed a variable degree of antioxidant potency. Finally, their antiproliferative potential was tested against a panel of human cancer cell lines and evaluated by using the sulforhodamine B (SRB) assay. All essential oil preparations exhibited a variable degree of antiproliferative activity, depending on the cancer model used, with the most potent one being sweet basil against an in vitro model of human colon carcinoma.

A Novel GC-MS Bioanalytical Method for Natural Borneol and Its Application in Investigating Natural Borneol Distribution in Mice

Abstract:One of the major goals of our group is to understand the meridian tropism of natural borneol (NB). As a first step, this study served two purposes: to develop a GC-MS bioanalytical method for NB, and with the aid of this method to investigate NB distribution in mice. Mouse tissue samples, spiked with naphthalene as internal standard, were homogenated with NS and extracted using hexane. The extract supernatant was analyzed in GC-MS with conditions of DB-5MS capillary column (0.25mm×30m×0.25μm), electron impact (EI) ionization with energy of 70 eV and multiplier voltage of 1100 V, and Selected Ion Monitor (SIM, m/z 95 for camphor and NB, whereas m/z 128 for naphthalene) detection mode. The calibration curve was linear in the range from 22 ng/mL to 22 μg/mL with a regression equation Y= 0.000486X + 0.00036, R2=0.999 (n=3). The limit of detection was 10 ng/mL. The lower limit of quantitation was 22 ng/mL. The within-day and inter-day precisions were 3.72∼6.11% and 7.29∼17.6%, respectively. The relative recovery was 85.95∼107.93% and extraction recovery was 93.04∼103.03%. This bioanalytical method has sufficient recovery and is accurate and reliable for quantitating NB in tissues. The tissue distribution up to 150 min after a single oral dose of NB was investigated in mice. This compound was detected in most tissues. The liver had the highest concentration, followed by kidney, heart, brain, and others. Tissue concentration-time curves had double or triple peaks. The tissue distribution pattern suggested that NB may attribute to the cardio-system.
GC-MS Analysis of Essential Oil Constituents from Rhizome and Root of Notopterygium Incisum

Abstract:Aim To analyse the chemical constituents of the essential oil extracted from the rhizome and root of Notopterygium incisum Ting ex H.T. Chang, providing scientific basis for quality control. Methods The total essential oil was extracted by water-steam distillation and separated by capillary gas chromatography (GC). The components were determined by normalization method, and identified by GC-MS. Results GC-MS exhibited 242 peaks and 83 compounds were identified, accounting for 75.77% of the total essential oil. Conclusion In the total essential oil of the rhizome and root of N. incisum, monoterpenes and sesquiterpenes accounted for 13.63% and 67.93%, respectively, in which (1S)-β-pinene (1.67%), 3-carene (1.05%), limonene (1.22%), and 1S-endo-bornyl acetate (1.68%) as the monoterpenes and its derivatives, and (+)-β-elemene (6.78%), sativene (1.54%), α-caryophyllene (2.64%), germacrene D (1.67%), eudesma-4 (14),11-diene (2.36%), α-selinene (2.42%), δ-cadinene (1.55%), 3,7,11-trimethyl-2,6,10-dodecatrien-1-ol (1.03%), (±)-elemol (5.18%), (-)-spathulenol (1.40%), guaiol (3.81%), dehydroxy-isocalamendiol (1.06%), γ-eudesmol (1.05%), α-eudesmol (7.97%), bulnesol (3.09%), and carotol (2.30%) as the sesquiterpenes and its derivatives were main components. In addition, isopropyl trans-cinnamate was the maximum compound (11.3%) of the total essential oil.

Abstract:In the current investigation, a comprehensive analysis was carried out on essential oils (Eos) extracted from six aromatic plant species, namely Rosmarinus officinalis, Pelargonium graveolens, Thymus vulgaris, Origanum vulgare, Laurus nobilis, and Aloysia citrodora. An exploration was conducted into the chemical composition using Gas Chromatography‐Mass Spectrometry (GC/MS), antioxidant properties assessed through DPPH, ABTS, CUPRAC, FRAP, MCA, and PBD assays, ecotoxicological impacts evaluated via allelopathy and the Daphnia magna heartbeat test, as well as bio‐pharmacological effects including anticancer activity and gene expression analysis. Results revealed strong antioxidant activity in all essential oils, with T. vulgaris EO (2748.00 mg TE/g) and O. vulgare EO (2609.29 mg TE/g) leading in CUPRAC assay. R. officinalis EO showed the highest α‐amylase inhibition at 1.58 mmol ACAE/g, while O. vulgare EO excelled in α‐glucosidase inhibition at 1.57 mmol ACAE/g. Additionally, cytotoxic effects were evaluated on human colorectal cancer (HCT116) cells. A. citrodora, O. vulgare, and R. officinalis EOs were found the most potent anticancer, as also witnessed by their higher modulatory effects on the gene expression of BAX and Bcl‐2. Collectively, the present data highlight the importance to implement the knowledge and to valorise the chain of aromatic plants.

Abstract:Nutmeg (Myristica fragrans), a popular spice and traditional medicine worldwide, is susceptible to fungal and mycotoxin contamination. Hence, it is important to monitor the residual levels of multiple mycotoxins in nutmeg and related products. However, analysts are continuously faced with the challenges of quantifying mycotoxin concentrations in complex matrices such as spices and traditional Chinese medicines. To develop a simple and sensitive mycotoxin analysis method encompassing the total workflow from sample preparation to quantitative analysis, a simple, robust, and sensitive method for the simultaneous identification and quantification of 21 mycotoxins in nutmeg and related products was described. The method is based on sample preparation by modified QuEChERS extraction and dispersive solid-phase extraction (dSPE) cleanup followed by ultrahigh-performance liquid chromatography-triple quadrupole tandem mass spectrometry (UPLC-MS/MS). Matrix-matched calibration was applied for the quantification of the investigated mycotoxins. The limits of detection of the target analytes ranged from 0.04 μg/kg to 6.0 μg/kg. Recovery rates ranged from 60.49% to 97.46% at three spiking levels, with relative standard deviations<13%. This method was applied to 45 samples of nutmegs, mace and related products. Three nutmeg samples and one nutmeg powder were contaminated with aflatoxins with residue levels of 1.76 ~ 3.39 μg/kg for AFB1 and 2.51 ~ 8.77 μg/kg for AFs (the sum of AFB1, AFB2, AFG1, AFG2 and AFM1). No mycotoxins were detected in mace. All of the contamination levels were below the regulatory maximum residue limits suggested by the European Commission and China.

Abstract:Phenanthrenes and their derivatives have biological relevance owing to their antimicrobial, antioxidant, and cytotoxic effects on cancer cells. They can be efficiently analyzed through ultrahigh-performance liquid chromatography coupled to tandem high-resolution mass spectrometry (UHPLC-MS/HRMS). Herein, we first studied the unique fragmentation behavior of phenanthrenes based on direct infusion MS/HRMS analysis. As a newly described phenomenon, "organ pipe distribution", we found a structural connection linking their unique fragmentation pattern to serial H radical losses. The bonds responsible for this behavior were identified through quantum chemical calculations using a stepwise approach. Furthermore, the chromatographic aspect of this study was enhanced by developing, validating, and applying a new unscheduled targeted UHPLC-MS/HRMS method for quantifying phenanthrenes in Juncus compressus herb. Targeted compounds were efficiently separated within 4 min upon utilizing the Accucore C30 column, and the unscheduled targeted analytical approach afforded five new isomers. Compounds 1 (effususol), 3 (dehydroeffusol), and 6 (7-hydroxy-1-methyl-2-methoxy-5-vinyl-9,10-dihydrophenanthrene) had their linearity limits determined within 10-5000 nM, and Compounds 2 (effusol), 4 (juncusol), 5 (effususin A), and 7 (compressin A) within 25-5000 nM. The coefficients of variation for precision ranged from 1.4 % to 15.2 %. The obtained matrix effects and accuracy values were also within acceptable ranges. Compounds 2 (effusol) and 3 (dehydroeffusol) were present in both methanolic and dichloromethanolic extracts of Plants 1 and 3 at the highest concentrations. Furthermore, the relationship between phenanthrene fingerprints, obtained through ANOVA statistical analysis of quantitative data, and the geographical location of herbs was also established.

Abstract:Intensive plasmapheresis was performed on 7 alloimmunized pregnant women. About 60% of the plasma volume was removed each day, Monday to Friday, with pauses over the weekends. This treatment resulted in drops in immunoglobulin levels to about 1/3 of the pre-treatment concentrations, whereas the haemostatic parameters studied remained within normal limits. Thus, generally the treatment (as performed in this study) does not seem to induce changes that predispose for thrombotic or haemorrhagic complications. However, in one of the patients, antithrombin III became progressively depleted over the weeks of treatment. Therefore it is suggested that the patients be screened for their haemostatic profile prior to and after a few weeks of plasmapheresis treatment.
LC-MS-based metabolite profiling of methanolic extracts from the medicinal and aromatic species Mentha pulegium and Origanum majorana

Abstract:Introduction: There has been increasing interest dedicated to the phenolic compounds with a view to their antioxidant and healthy properties. Recent studies have focused on plants from the Lamiaceae family with special interest in phenolic compounds antioxidant potential. Objective: The metabolite profile of methanolic extracts from two Lamiacea medicinal plants was investigated. Materials and methods: Mentha pulegium and Origanum majorana methanolic extracts were analysed using reversed-phase ultra-high-performance liquid chromatography (RP-UHPLC) coupled to electrospray ionisation quadrupole time-of-flight mass spectrometry (ESI-QTOF-MS) detection in the negative ion mode. Results: A total of 85 metabolites were characterised from different families, such as organic acids and derivatives, amino acids and derivatives, nucleosides, phenolic compounds as well as other polar metabolites, by using the MS and MS/MS information provided by the QTOF-MS. However, the total phenols and flavonoids were also quantified spectrophotometrically and they registered higher amounts in Mentha pulegium than in Origanum majorana extract. Gallocatechin was the major compound in M. pulegium extract whereas quercetin dimethyl ether, jaceidin and dihydrokaempferide were the major ones in O. majorana extract. Conclusion: The distribution of phenolic compounds in the methanolic extract showed a variation among studied plants. Mentha pulegium can be considered as a source of gallocatechin.
