Abstract:Cadmium pollution is becoming a serious problem due to its nondegradability and substantial negative influence on the normal growth of crops, thereby harming human health through the food chain. Rhizospheric bacteria play important roles in crop tolerance. However, there is little experimental evidence which demonstrates how various cadmium concentrations affect the bacterial community in wheat fields including rhizosphere microorganisms and nonrhizosphere (bulk) microorganisms. In this study, 16S rRNA amplicon sequencing technology was used to investigate bacterial communities in rhizosphere and bulk soils under different levels of pollution in terms of cadmium concentration. Both the richness and diversity of the rhizosphere microorganism community were higher under nonpolluted soil and very mild and mild cadmium-contaminated soils than compared with bulk soil, with a shift in community profile observed under severe cadmium pollution. Moreover, cadmium at various concentrations had greater influence on bacterial composition than for the nonpolluted site. In addition, redundancy analysis (RDA) and Spearman’s analysis elucidated the impact of exchangeable Cd and total Cd on bacterial community abundance and composition. This study suggests that cadmium imposes a distinct effect on bacterial community, both in bulk and rhizosphere soils of wheat fields. This study increases our understanding of how bacterial communities in wheat fields shaped under different concentrations of cadmium.
Inhibitory effects of Pseudomonas sp. W112 on cadmium accumulation in wheat grains: Reduced the bioavailability in soil and enhanced the interception by plant organs

Abstract:Microorganisms play an important role in heavy metal bioremediation and soil fertility. The effects of soil inoculation with Pseudomonas sp. W112 on Cd accumulation in wheat were investigated by analyzing the transport, subcellular distribution and speciation of Cd in the soil and plants. Pseudomonas sp. W112 application significantly decreased Cd content in the roots, internode and grains by 10.2%, 29.5% and 33.0%, respectively, and decreased Cd transfer from the basal nodes to internodes by 63.5%. Treatment with strain W112 decreased the inorganic and water-soluble Cd content in the roots and increased the proportion of residual Cd in both the roots and basal nodes. At the subcellular level, the Cd content in the root cell wall and basal node cytosol increased by 19.6% and 61.8%, respectively, indicating that strain W112 improved the ability of the root cell wall and basal node cytosol to fix Cd. In the rhizosphere soil, strain W112 effectively colonized and significantly decreased the exchangeable Cd, carbonate-bound Cd and iron-manganese oxide-bound Cd content by 43.5%, 27.3% and 17.6%, respectively, while it increased the proportion of residual Cd by up to 65.2%. Moreover, a 3.1% and 23.5% increase in the pH and inorganic nitrogen content in the rhizosphere soil, respectively, was recorded. Similarly, soil bacterial community sequencing revealed that inoculating with strain W112 increased the abundance of Pseudomonas, Thauera and Azoarcus, which are associated with inorganic nitrogen metabolism, and decreased that of Acidobacteria, which is indicative of soil alkalinization. Hence, root application of Pseudomonas sp. W112 improved soil nitrogen availability and inhibited Cd accumulation in the wheat grains in a two-stage process: by reducing the Cd availability in the rhizosphere soil and by improving Cd interception and fixation in the wheat roots and basal nodes. Pseudomonas sp. W112 may be a suitable bioremediation agent for restoring Cd-contaminated wheat fields.

Abstract:Cadmium contamination poses a significant risk to soil ecosystems in certain parts of the world. Using eco-friendly fertilizers alongside beneficial microorganisms offers a viable solution to mitigate Cd pollution in agricultural soil. This study used an outdoor experiment to evaluate the impact of administering a Cd-immobilizing bacterial (Bacillus) inoculant with two biologically-enriched organic fertilizers (either fermentative edible fungi residue or fermented cow dung) on wheat plants and associated microbial populations in a field contaminated with Cd. The mixed application of fermentative cow dung with the Cd-immobilizing bacterium reduced the effective Cd content of wheat root-soil by 13,0 %. Application of Cd-immobilizing Bacillus inoculant reduced the Cd enrichment of wheat roots by 0.07 mg/kg. Co-application of fermentative cow dung with the bacterial inoculant reduced the Cd enrichment of wheat seeds by 20,0 %. Co-application of the two organic fertilizers could improve some of the nutrients related to wheat and soil fertility; however, the diversity of the soil microbial community changed less and its species richness decreased. Applying the Bacillus inoculant inhibited the growth of native pathogenic bacteria, such as Proteobacteria. Whether administering it with either fermented cow manure or fermented edible fungus residue, the relative abundance of nitrate-reducing bacteria such as Rhodobacter increased, which should promote the soil nitrogen cycle. The main factors influencing soil microbial community structure of wheat plants were pH, available potassium, and available Cd content. Symbiotic network analysis revealed bacterial inoculant and organic fertilizer inoculum further altering the ecological relationships of microbial communities. According to the FAPROTAX functional prediction, Rhodanobacter may play a key role in nitrate respiration in the soil nitrogen cycle. In conclusion, this study provides a comprehensive, timely reference for understanding microbial changes caused by the combined application of this type of bacterial inoculant and organic soil amendments in Cd-contaminated fields.
Wheat-associated Pseudomonas taiwanensis WRS8 reduces cadmium uptake by increasing root surface cadmium adsorption and decreasing cadmium uptake and transport related gene expression in wheat

Abstract:Metal-resistant bacteria can reduce Cd accumulation in plants, but mechanisms underlying this effect are poorly understood. In this study, a highly effective Cd-resistant WRS8 strain was obtained from the rhizoshere soil of Triticum aestivum L. Yangmai-13 and identified as Pseudomonas taiwanensis based on 16S rRNA gene sequence analysis. Strain WRS8 was investigated for its effects on Cd availability and wheat tissue Cd contents and the related mechanisms using a hydroponic culture experiment. In strain WRS8-inoculated solution, the Cd concentration reduced and the pH and cell-adsorbed Cd increased with time. Strain WRS8 increased the wheat root and above-ground tissue dry weights by 11-36% compared to the controls. In strain WRS8-inoculated wheat plants, the Cd contents of the roots and above-ground tissues decreased by 78-85% and 88-94% and the Cd bioconcentration and translocation factors decreased by 78-85% and 46-58% at days 3 and 10, respectively, compared with the controls. The root surface-adsorbed Cd contents increased by 99-121% in the WRS8 strain-inoculated wheat plants at days 3 and 10 compared to the controls. Furthermore, strain WRS8 colonized the wheat root surfaces and interiors and reduced the expression levels of the LCT1 and HMA2 genes involved in Cd accumulation and transport in wheat roots by 46% and 30%, respectively, compared to the controls. In the Cd-contaminated soils, strain WRS8 significantly reduced the available Cd content by 20-24% and increased the pH compared to the controls. These findings showed the important role of strain WRS8 in reducing solution and soil Cd availability and suggested that strain WRS8 reduced the wheat tissue Cd accumulation by increasing root surface Cd adsorption and decreasing wheat root Cd uptake and transport-related gene expression and may provide a new and effective wheat rhizobacteria-enhanced approach for reducing wheat Cd uptake in Cd-polluted environments.

Abstract:The application of phosphorus-solubilizing bacteria is an effective method for increasing the available phosphorus content and inhibiting wheat uptake of heavy metals. However, further research is needed on the mechanism by which phosphorus-solubilizing bacteria inhibit cadmium (Cd) uptake in wheat roots and its impact on the expression of root-related genes. Here, the effects of strain Klebsiella aerogenes M2 on Cd absorption in wheat and the expression of root-related Cd detoxification and immobilization genes were determined. Compared with the control, strain M2 reduced (64.1-64.6%) Cd uptake by wheat roots. Cd fluorescence staining revealed that strain M2 blocked the entry of exogenous Cd into the root interior and enhanced the immobilization of Cd by cell walls. Forty-seven genes related to Cd detoxification, including genes encoding peroxidase, chalcone synthase, and naringenin 3-dioxygenase, were upregulated in the Cd+M2 treatment. Strain M2 enhanced the Cd resistance and detoxification activity of wheat roots through the regulation of flavonoid biosynthesis and antioxidant enzyme activity. Moreover, strain M2 regulated the expression of genes related to phenylalanine metabolism and the MAPK signaling pathway to enhance Cd immobilization in roots. These results provide a theoretical basis for the use of phosphorus-solubilizing bacteria to remediate Cd-contaminated fields and reduce Cd uptake in wheat.

[Effects of Heavy Metal-immobilizing Bacteria on Reducing Wheat Uptake Cd by Regulation Soil Iron Oxides]

Abstract:Iron oxides are widely distributed in soils and have large specific surface areas and strong affinities with heavy metals, and thus play a significant role in the transformation of heavy metals in soils. Pot experiments were conducted to study the effects of heavy metal-immobilizing bacteria Bacillus megaterium N3, Serratia liquefaciens H12, and strain N3 combination with H12 on the absorption of Cd by ZM-3 (Triticum aestivum L.). The effects of different treatments on soil pH, iron oxide content, and relative abundance of iron oxide bacteria, clay particle mineral composition, and aggregates in wheat rhizosphere were also measured. The results showed that strains N3, H12, and N3+H12 significantly (P<0.05) increased the dry weight (36.5%-75.3%) of wheat grains, and significantly (P<0.05) decreased the content of Cd (30.2%-84.9%) in wheat grains. Furthermore, compared with the single application of strain N3 or H12, the application of strain N3 combined with H12 had a stronger ability to reduce the absorption of Cd by ZM-3. The results suggested that strain N3, combined with H12, decreased the uptake of Cd in wheat by:①increasing the pH (from 6.74 to 7.08) of wheat rhizosphere soil; ② increasing the Fe oxide (67.9%) and goethite contents of wheat rhizosphere soil; ③ increasing the relative abundance of iron-oxidizing bacteria (Leptothrix spp. and Gallionella spp.), and ④ increasing the content of small particle size (<0.25 mm) aggregates. The results provide a theoretical basis and technical support for the use of heavy metal-immobilizing bacteria to repair farmland contaminated by heavy metals.
Effects of biochars combined with ferrous sulfate and pig manure on the bioavailability of Cd and potential phytotoxicity for wheat in an alkaline contaminated soil

Abstract:Reducing the content of Cd in wheat grains is necessary for human health, especially in alkaline farmland in northern China where a large area of soils is heavily polluted with Cd. A field experiment was conducted to investigate the effects of wheat straw biochar (WB) and maize straw biochar (MB) combined solely with ferrous sulfate (FeSO4) or combined with FeSO4 and pig manure (PFWB and PFMB) on Cd bioavailability in soil and the phytotoxicity of Cd in wheat in an alkaline contaminated soil. The results showed that the application of these treatments decreased the concentrations of CaCl2-extractable Cd by 32.8-60.5% and increased the soil organic matter by 36.8-106.7% compared with those of the control treatment. The acid-soluble fraction of Cd was decreased by 3.7-16.8% but the residual fraction was increased by 1.7-13.7%. Furthermore, the addition of WB and MB increased the soil pH by 0.29 and 0.16 units, respectively, while the other treatments decreased the soil pH by 0.16-0.37 units. The concentrations of Cd in different wheat tissues were decreased in amended soils, and these decreases were positively correlated with CaCl2-extractable Cd and the acid-soluble fraction of Cd. The ability to accumulate Cd was in the order of root>straw>grain. In addition, there was a downward trend in the bio-concentration and translocation factors under different amendments. The amendments of PFWB and PFMB had better efficiency in the immobilization of Cd in soil and the inhibition of Cd uptake of wheat than WB and MB. In addition, the treatments of PFWB and PFMB increased the wheat yield by 14.3-16.2%, and thus have a great application prospect based on the cost-benefit analysis.
Specific bacterial communities in the rhizosphere of low-cadmium and high‐zinc wheat (Triticum aestivum L.)

Abstract:Microorganisms can modulate the contents of cadmium (Cd) and zinc (Zn) in wheat grains. Increasing the essential nutrient element Zn and decreasing the toxic element Cd in wheat grains can significantly improve human health. To characterize the specific bacterial communities associated with Cd and Zn accumulation in wheat, we conducted a field experiment by planting wheat cultivars differing in their capacity for Cd and Zn accumulation. The grain Cd contents in wheat cultivars YN23 (0.078 mg kg−1), JN17 (0.080 mg kg−1), YN836 (0.081 mg kg−1) and LM2 (0.091 mg kg−1) were significantly lower than those in ZM32 (0.16 mg kg−1). The Zn contents were significantly higher in the grains of JN17 (44.36 mg kg−1), LM2 (42.22 mg kg−1) and ZM32 (43.19 mg kg−1) than YN23 (27.05 mg kg−1) and YN836 (29.70 mg kg−1). On the basis of contents and bio-concentration factors of Cd and Zn in wheat grain, JN17 and LM2 were identified as low-Cd- and high-Zn-accumulating cultivars, YN23 and YN836 were low-Cd- and low-Zn-accumulating cultivars, and ZM23 was a high-Cd- and high-Zn-accumulating cultivar. The relative abundance values of Gemmatimonadaceae, Sphingomonadaceae and Beijerinckiaceae in the rhizospheres of low-Cd cultivars were significantly higher than those of high-Cd cultivars. High-Zn cultivars had higher abundance of Rhodanobacteraceae in the rhizosphere than did low-Zn cultivars. The low-Cd- and high-Zn-accumulating cultivars were enriched in Alphaproteobacteria and Gemmatimonadaceae, and strengthened nitrification function including aerobic_ammonia_oxidation and aerobic_nitrite_oxidation in the rhizosphere soil, thus contributing to the decreased Cd and increased Zn contents in wheat grains. Microbial technology is a promising method to control the contents of Cd and Zn in wheat grains.

Abstract:Phosphorus-solubilizing microorganisms convert insoluble phosphorus in the soil into phosphorus that can be absorbed by plants. Soluble phosphate combines with heavy metals to form precipitation, reducing the content of available heavy metals, thereby reducing the absorption of heavy metals by crops, which plays an important role in the remediation of heavy metal-contaminated soil. The effects of the immobilization of Cd and Pb and the release of PO43- by the phosphorus-solubilizing bacterium Klebsiella sp. M2 were studied through solution culture experiments. In addition, the effects of strain M2 on wheat uptake of Cd and Pb and its microbiological mechanism were also explored through pot experiments. The results showed that strain M2 reduced the concentrations of Cd and Pb and increased the concentration of PO43- in the solution through cell wall adsorption and induced phosphate precipitation. Pot experiments showed that compared to those in the CK group and inactivated strain M2 group, inoculation with live strain M2 significantly increased （123%-293%） the contents of Ca2-P and Ca8-P in rhizosphere soil, decreased the content of DTPA-Cd （34.48%） and DTPA-Pb （36.72%） in wheat rhizosphere soil, and thus hindered the accumulation of Cd and Pb in wheat grains. Moreover, high-throughput sequencing results showed that strain M2 significantly increased the diversity of wheat rhizosphere bacterial communities； increased the relative abundance of Proteobacteria, Gemmatimonadetes, and Bacteroidota in wheat rhizosphere soil； and increased the proportion of heavy metal-immobilizing and phosphorus-promoting bacteria in wheat rhizosphere soil （mainly Sphingomonas, Nocardioides, Bacillus, Gemmatimonas, and Enterobacter）. These bacterial genera played an important role in immobilizing heavy metals and preventing wheat from absorbing heavy metals. These results provide bacterial resources and theoretical basis for the bioremediation of heavy metal-contaminated farmland.
Prevalence and Associated Factors of Tooth Erosion in 8 -12-Year-Old Brazilian Schoolchildren.

Abstract:OBJECTIVE This study estimated the prevalence, distribution and associated factors of tooth erosion in Brazilian schoolchildren. STUDY DESIGN A cross-sectional study was carried out in a representative sample of children aged 8-12-years-old from public and private schools. Six calibrated examiners assessed tooth erosion (O'Sullivan index) and other oral conditions (dental trauma, enamel defects, plaque, dental caries and dental crowding). Socio-demographic and behavioral data were collected using questionnaires. Interviews were conducted in children and in parents. Data were analyzed using Poisson regression model considering the cluster sample (Prevalence Ratio-PR; 95% Confidence Interval-CI). RESULTS A total of 1,210 children participated and were examined. Tooth erosion was observed in 25.1% of the children. Lesions were mostly observed in enamel and less than a half of the buccal surface was affected. The type of school and age were significant associated with higher prevalence of tooth erosion and dental crowding was associated with less probability of tooth erosion significantly. Acidic food or drinks were not associated with dental erosion. CONCLUSION The prevalence of tooth erosion in the studied population was high. Erosive lesions were not severe, confined in enamel. Tooth erosion was associated with age, type of school, socioeconomic level and dental crowding.
Reduced cadmium toxicity in rapeseed via alteration of root properties and accelerated plant growth by a nitrogen-fixing bacterium

Abstract:Cd accumulation in crops has become a global environmental problem because it endangers human health. Screening for microorganisms that can reduce Cd accumulation in crops is a possible measure to address this issue. However, success has been limited, and most previous work did not involve bacteria. In the present study, a strain of N-fixing bacteria ( Burkholderia spp.) that exhibits high levels of Cd tolerance was screened. The ability of this bacterium to reduce Cd in rapeseed was then assessed in sterile hydroponic and open soil culture systems. In the hydroponic system, the Burkholderia inoculum promoted Cd fixation in rapeseed roots and thus reduced Cd enrichment in aboveground edible tissues (leaves). The mechanisms were related to increased activity of pectin methylesterase in root cell walls, and the transformation of the chemical form of root Cd from "active" (NaCl-extracted) to "inert" (HCl-extracted and residual Cd) states. Additionally, Burkholderia accelerated plant growth, thus shortening the period in which the plant is available for Cd absorption. In the soil culture system, Burkholderia also reduced Cd enrichment in rapeseed leaves in the presence of other microorganisms. Thus, the bacterial strain shows potential for broad application for reducing the accumulation of Cd in crops.

Abstract:In recent years, the problem of Cd pollution in paddy fields has become more and more serious, which seriously threatens the safe production of food crops and human health. Using microorganisms to reduce cadmium pollution in rice fields is a green, safe and efficient method, the complicated interactions between the microbes in rice roots throughout the process of cadmium absorption by rice roots are poorly understood. In this investigation, a hydroponic pot experiment was used to examine the effects of bacteria R3 ( Herbaspirillum sp) and T4 ( Bacillus cereus ) on cadmium uptake and the endophytic bacterial community in rice roots. The results showed that compared with CK (Uninoculated bacterial liquid), the two strains had significant inhibitory or promotive effects on cadmium uptake in rice plant, respectively. Among them, the decrease of cadmium content in rice plants by R3 strain reached 78.57-79.39%, and the increase of cadmium content in rice plants by T4 strain reached 140.49-158.19%. Further investigation showed that the cadmium content and root cadmium enrichment coefficient of rice plants were significantly negatively correlated with the relative abundances of Burkholderia and Acidovorax , and significantly positively correlated with the relative abundances of Achromobacter , Agromyces and Acidocella . Moreover, a more complex network of microbes in rice roots inhibited rice plants from absorbing cadmium. These results suggest that cadmium uptake by rice plants is closely related to the endophytic bacterial community of roots. This study provides a reference scheme for the safe production of crops in cadmium contaminated paddies and lays a solid theoretical foundation for subsequent field applications.

Psychoanalytic thought is briefly reviewed and recent studies regarding diagnosis, incidence, etiology, treatment, and prognosis are covered. The Diagnostic and Statistical Manual III is thought to adequately diagnose both adult and childhood depression. Depressive disorder in children is more frequently recognized and important studies are enhancing our knowledge of biologic, genetic, and psychological factors in depression. Soil properties drive the bacterial community to cadmium contamination in the rhizosphere of two contrasting wheat (Triticum aestivum L.) genotypes

Abstract:Cadmium (Cd) bioavailability in the rhizosphere makes an important difference in grain Cd accumulation in wheat. Here, pot experiments combined with 16S rRNA gene sequencing were conducted to compare the Cd bioavailability and bacterial community in the rhizosphere of two wheat (Triticum aestivum L.) genotypes, a low-Cd-accumulating genotype in grains (LT) and a high-Cd-accumulating genotype in grains (HT), grown on four different soils with Cd contamination. Results showed that there was non-significant difference in total Cd concentration among four soils. However, except for black soil, DTPA-Cd concentrations in HT rhizospheres were higher than those of LT in fluvisol, paddy soil and purple soil. Results of 16S rRNA gene sequencing showed that soil type (52.7%) was the strongest determinant of root-associated community, while there were still some differences in rhizosphere bacterial community composition between two wheat genotypes. Taxa specifically colonized in HT rhizosphere (Acidobacteria, Gemmatimonadetes, Bacteroidetes and Deltaproteobacteria) could participate in metal activation, whereas LT rhizosphere was highly enriched by plant growth-promoting taxa. In addition, PICRUSt2 analysis also predicted high relative abundances of imputed functional profiles related to membrane transport and amino acid metabolism in HT rhizosphere. These results revealed that the rhizosphere bacterial community may be an important factor regulating Cd uptake and accumulation in wheat and indicated that the high Cd-accumulating cultivar might improve Cd bioavailability in the rhizosphere by recruiting taxa related to Cd activation, thus promoting Cd uptake and accumulation.

Abstract:Iron plaque is the amorphous and/or crystalline layer of Fe and Mn (hydr)oxides formed on the root surface of wetland plants. It could adsorb and co-precipitate metal(loid)s at the rhizosphere, thus modulating the uptake and accumulation of metal elements in plants. In this study, the Fe(II)/Mn(II)-oxidizing bacteria Burkholderia sp. D416 (D416) and Pseudomonas sp. YGL (YGL) were isolated from Cd-contaminated rice field, both hydroponic experiment and pot experiment were performed to assess the impact of bacterial inoculation on iron plaque formation, elemental content of the plaque, plant dry mass, antioxidant enzyme activity and Cd content in rice plants. The results revealed that inoculation with D416, YGL, and D416+YGL stimulated iron plaque formation on the root surface of the hydroponic rice. The content of C, N, O, Na, Mg, Al, Si, P, S, Cl, K, Fe and Ca in the root plaque were affected by the bacterial inoculation and varied among different plant growth stages. The pot experiment indicated that inoculation with D416 increased the root dry biomass by 58.89%, and the combined inoculation of D416 and YGL increased the dry biomass of root, shoot and grain by 16.89%, 21.66% and 23.26%, respectively. Importantly, YGL inoculation decreased the Cd translocation from root to shoot and from glume to brown rice grain by 50.00% and 50.27%, respectively, and the Cd content in shoot and brown rice grain were decreased by 20.00% and 34.48%, respectively. Taken together, the elemental content of the iron plaque and Cd content in rice plants varied among different plant growth stages and when plants were inoculated with different bacterial strains. YGL dramatically reduced the Cd content in brown rice grain, thus it could potentially be used to reduce Cd content in rice crop grown in Cd-contaminated soils.

Abstract:The harmful effects of cadmium (Cd) on crop quality and the limited effective control methods continue to raise concerns regarding safe agricultural production in Cd-polluted weakly alkaline soils. Past studies were less concerned with the role of fungi, popular in the soil -wheat system, in the Cd transfer from soil to wheat. The current research explored the potential role of rhizosphere fungi in the absorption of Cd in wheat seedlings by applying fungicides, including chlorothalonil and benomyl, into Cd-contaminated weakly alkaline soils. Maximum increase in aboveground fresh -weight, leaf chlorophyll content, and maximum decrease in malondialdehyde were recorded with benomyl at the dose of 150 mg Kg -1 for wheat seedling. Besides, the benomyl at the dose of 150 mg Kg -1 significantly decreased the Cd contents in the stem of wheat seedlings. The wheat seedlings treated with benomyl at a dose of 50 mg Kg -1 exhibited the lowest contents of Cd in their stems and leaves. Moreover, these seedlings had significantly higher levels of zinc and lower levels of calcium in their leaves. The application of fungicides has dramatically changed the fungal communities in rhizosphere soils. Compared to the control group, the application of benomyl resulted in an increase in the population of Rhizopus, a plant -beneficial fungi that is tolerant to Cd. Conversely, it led to a decrease in the population of Fusarium, a fungal pathogen that threatens plants, in the rhizosphere soils. It shows that fungicide could be an effective material in controlling Cd uptake by wheat in a weakly alkaline soil.

Abstract:Applying plant growth-promoting rhizobacteria (PGPR) improves the efficiency of soil-borne disease control and is considered a sustainable practice. However, the effect of PGPR on the fungal community, especially pathogenic fungi and arbuscular mycorrhizal fungi (AMF), remains unclear. In this study, we examined the effects of a compound microbial agent (consisting of Bacillus subtilis HG-15 and Bacillus velezensis JC-K3) on the incidence and yield of wheat under low salt stress, as well as compared the diversity and community composition of the rhizosphere fungal and AMF communities of wheat in the CK (not inoculated bacterial agent) and BIO (inoculated with a bacterial agent) groups. Chlorophyll relative content (SPAD), net photosynthesis rate ( Pn ), transpiration rate ( Tr ), leaf water use efficiency ( WUE L ), grains per spike and wheat yield in the BIO group increased more than in the CK group. The number of diseased plants and disease incidence was observed to be reduced. The relative efficacy reached 79.80%. We classified 1007 fungal operational taxonomic units (OTU) based on Miseq sequencing data: 11 phyla, 173 families, 319 genera, and 521 species. Fifty-four OTUs were classified from the AMF effective sequences, including 1 phylum, 3 families, 3 genera, and 17 species. The inoculation of bacterial agents reduced the relative abundance of pathogen genera such as Gibberella , Fusarium , Cladosporium , and Alternaria in wheat rhizosphere. It increased the relative abundance of AMF species such as Glomus-group-B-Glomus-lamellosu-VTX00193, Glomus-viscosum-VTX00063, and Glomus-Glo2-VTX00280. In addition, pH, EC, exchangeable K, available N, total N, organic matter, and olsen P were the main driving forces for shaping wheat rhizosphere fungi. The pH value was positively correlated with the relative abundance of fungal communities in soil, especially Gibberella , Cladosporium , Fusarium , and Alternaria . In summary, inoculation with Bacillus subtilis HG-15 and Bacillus velezensis JC-K3 affected wheat yield, incidence, rhizosphere soil chemical properties, rhizosphere fungi, and AMF fungal diversity and community. The findings may provide a theoretical foundation and strain support for constructing efficient PGPR-community and clarifying its mechanism of pathogenic bacteria inhibition.

Abstract:Heavy metal ions, in particular cadmium (Cd), have a negative impact on the growth and productivity of major crops, including wheat. The use of environmentally friendly approaches, in particular, bacteria that have a growth-stimulating and protective effect, can increase the resistance of plants. The effects of the pre-sowing seed treatment with the plant growth-promoting endophyte Bacillus subtilis 10-4 (BS) on cadmium acetate (Cd)-stressed Triticum aestivum L. (wheat) growth, photosynthetic pigments, oxidative stress parameters, roots’ lignin content, and Cd ions accumulation in plants were analyzed. The results showed that the tested Cd-tolerant BS improved the ability of wheat seeds to germinate in the presence of different Cd concentrations (0, 0.1, 0.5, and 1 mM). In addition, the bacterial treatment significantly decreased the damaging effects of Cd stress (1 mM) on seedlings’ linear dimensions (lengths of roots and shoots), biomass, as well as on the integrity and permeability of the cell walls (i.e., lipid peroxidation and electrolyte leakage) and resulted in reduced H2O2 generation. The pretreatment with BS prevented the Cd-induced degradation of the leaf photosynthetic pigments chlorophyll (Chl) a, Chl b, and carotenoids. Moreover, the bacterial treatment intensified the lignin deposition in the roots under normal and, especially, Cd stress conditions, thereby enhancing the barrier properties of the cell wall. This manifested in a reduced Cd ions accumulation in the roots and in the restriction of its translocation to the aboveground parts (shoots) of the bacterized plants under Cd stress in comparison with non-bacterized controls. Thus, the pre-sowing seed treatment with the endophyte BS may serve as an eco-friendly approach to improve wheat production in Cd-contaminated areas.
