Abstract:Gut microbiota dynamically participate in diverse physiological activities with direct impact on the host's health. A range of factors associated with the highly complex intestinal flora ecosystem poses challenges in regulating the homeostasis of microbiota. The consumption of live probiotic bacteria, in principle, can address these challenges and confer health benefits. In this context, one of the major problems is ensuring the survival of probiotic cells when faced with physical and chemical assaults during their intake and subsequent gastrointestinal passage to the gut. Advances in the field have focused on improving conventional encapsulation techniques in the microscale to achieve high cell viability, gastric and temperature resistance, and longer shelf lives. However, these microencapsulation approaches are known to have limitations with possible difficulties in clinical translation. In this Perspective, we present a brief overview of the current progress of different probiotic encapsulation methods and highlight the contemporary and emerging single-cell encapsulation strategies using nanocoatings for individual probiotic cells. Finally, we discuss the relative advantages of various nanoencapsulation approaches and the future trend toward developing coated probiotics with advanced features and health benefits.

Enhanced Viability of Probiotics (pediococcus Pentosaceus Li05) by Encapsulation in Microgels Doped with Inorganic Nanoparticles

Abstract:The major reduction in probiotic viability that occurs during food storage and gastrointestinal transit limits their potential health benefits. Microencapsulation is one of the most effective methods to protect probiotics from various harsh conditions. In this study, a model probiotic (Pediococcus pentosaceus Li05) was encapsulated in an alginate-gelatin microgels in the absence and presence of magnesium oxide (MgO) nanoparticles (NPs). The morphology and surface properties of the encapsulation systems were characterized by transmission electron microscopy (TEM) and atomic force microscopy (AFM), which showed that both the probiotics and NPs were successfully incorporated into spherical microgels. The viability of the probiotic was evaluated after exposure to different conditions: long-term storage in an aerobic environment; heat treatment; and gastrointestinal transit. Encapsulation of the probiotics significantly enhanced their viability under these different conditions. Probiotics encapsulated in MgO-loaded microgels were more stable than free bacterial cells or those encapsulated in microgels alone: less than 2 log(10) CFU reduction after 40 min incubation in gastric fluids versus 5 log(10) CFU reduction in the first 10 min for free cells. The SEM images indicated that the NPs may lead to enhanced probiotic viability by filling pores inside the microgels, which may have inhibited the ability of oxygen and hydrogen ions to access the probiotics. Moreover, the MgO NPs neutralized the hydrogen ions in the gastric fluids, thereby reducing acid-induced degradation of the probiotics. These results demonstrate that MgO-loaded microgels may be a promising encapsulation and delivery system for improving the efficacy of orally administered probiotics by protecting them from the harsh conditions during storage and in the gastrointestinal tract.

Abstract:Gut microbes are closely associated with most human health. When ingested orally, probiotics can effectively regulate the composition and quantity of human intestinal microorganisms, which is beneficial to human health. However, probiotics will be affected by the harsh environment of the digestive tract during the in vivo transportation process, and ensuring the viability of probiotics is a great challenge. Probiotic encapsulating technology provides an effective solution to this problem. The introduction of extreme temperatures, large probiotic microcapsule sizes and the difficulty in controlling probiotic microcapsule particle sizes mean that traditional microcapsule encapsulation methods have some limitations. From traditional microcapsule technology to the bulk encapsulation of probiotics with nanofibers and nanoparticles to the recent ability to wear nano “armor” for a single probiotic through biofilm, biological membrane and nanocoating. Emerging probiotic nanoagents provides a new conceptual and development direction for the field of probiotic encapsulation. In this review, we presented the characteristics of encapsulated probiotic carrier materials and digestive tract transport systems, we focused on the encapsulation systems of probiotic nanoagents, we analyzed the shortcomings and advantages of the current agent encapsulation systems, and we stated the developmental direction and challenges for these agents for the future.

Alginate Nanoencapsulated Synbiotic Composite of Pomegranate Peel Phytogenics and Multi-Probiotic Species as a Potential Feed Additive: Physicochemical, Antioxidant, and Antimicrobial Activities

Abstract:A synbiotic composed of alginate nanoencapsulated prebiotic (pomegranate peel phytogenics) and multi-species probiotics (Lactococcus lactis, Lactobacillus plantarum, Lactobacillus paracasei, and Saccharomyces cerevisiae) has been developed as a potential eco-friendly alternative to antibiotics. The physicochemical properties of the encapsulated synbiotic were evaluated, and its gastric and storage tolerance, as well as its antioxidant and antimicrobial activity, were tested and compared to that of the non-encapsulated synbiotic (free synbiotic). The results showed that the prebiotic pomegranate peel ethanolic extract contained seven phenolic compounds, with cinnamic being the most abundant (13.26 µL/mL). Sodium alginate-CaCl2 nanocapsules were effective in encapsulating 84.06 ± 1.5% of the prebiotic's phenolic compounds and 98.85 ± 0.57% of the probiotics. The particle size of the alginate-CaCl2 nanoencapsulated synbiotic was 544.5 nm, and the polydispersity index and zeta potential values were 0.593 and -12.3 mV, respectively. Thermogravimetric analysis showed that the alginate-CaCl2 nanoencapsulated synbiotic had high thermal stability at high temperatures, with only 2.31% of its weight being lost within the temperature range of 70-100 °C. The count of viable probiotics in the nanoencapsulated synbiotic was significantly higher than that in the free synbiotic after exposure to gastric acidity and storage for six months at room temperature. The percent inhibition values of the nanoencapsulated synbiotic and ascorbic acid (as a standard antioxidant) were comparable and significantly greater than those of the free synbiotic. The half-maximal inhibitory concentrations (IC50) of the nanoencapsulated synbiotic and ascorbic acid were significantly lower than those of the free synbiotic (3.96 ± 0.42 µg/mL and 4.08 ± 0.79 µg/mL for nanoencapsulated synbiotic and ascorbic acid, respectively, vs. 65.75 ± 2.14 µg/mL for free synbiotic). The nanoencapsulated synbiotic showed the highest significant antimicrobial activity against Escherichia coli (ATCC 8739). Both the nanoencapsulated and free synbiotics showed antimicrobial activity against Staphylococcus aureus (ATCC 6538), similar to that of gentamicin, although the nanoencapsulated synbiotic showed significantly higher inhibition activity compared to the free synbiotic. The nanoencapsulated synbiotic showed antimicrobial activity comparable to gentamicin against Pseudomonas aeruginosa (ATCC 90274), whereas the free synbiotic showed the least antimicrobial activity (p < 0.05). Both synbiotics showed significantly higher antimicrobial activity against Salmonella typhi (ATCC 6539) than gentamicin. Both synbiotics showed antifungal activity against Aspergillus niger and Aspergillus flavus, with a stronger effect observed for the nanoencapsulated synbiotic. However, the activity of both synbiotics was significantly lower than that of fluconazole (an antifungal drug).

Abstract:In the present study, encapsulation of Lactiplantibacillus plantarum (L.p) was performed using chitosan and casein through calcium phosphate intercrossing. Chitosan and casein both considered as non-toxic and biocompatible food derived components with intrinsic antioxidant properties. Layer by layer strategy was performed for deposition of modified cross-linked chitosan along with casein as the novel protective layers on the surface of probiotics. After confirmation of successful encapsulation, the viability and antioxidant activity of encapsulated L.p was evaluated. The results showed enhanced survival and antioxidant activity of encapsulated L.p compared to free bacteria in simulated digestive conditions. The survival of free and encapsulated L.p was respectively 1.38 ± 0.29 log cfu/ml and 6.99 ± 0.12 log cfu/ml in SGF and 8.54 ± 0.05 log cfu/ml and 7.25 ± 0.23 log cfu/ml in SIJ after 2 h of incubation. HPLC analysis was also used to investigate the detoxification activity of probiotics toward Aflatoxin M1 and obtained results showed encapsulated bacteria could significantly reduce aflatoxin M1 (68.44 ± 0.5 %) compared to free bacteria (43.76 ± 0.54 %). The results of this research suggest that the chitosan/casein mediated encapsulation of L.p with layer-by-layer technology is an effective method to improve the survival and antioxidant properties of probiotics with enhanced detoxification of AFM1.

Abstract:Nanotechnology is one of the highly evolving fields of research having immense potential in various fields of healthcare sectors. The very advent of nanotechnology lies in its ability to serve as a targeted drug delivery system. The introduction of a new branch namely bionanotechnology has further expanded the scope, especially in the diagnostics and treatment of various diseases. Probiotics being a natural source with a plethora of beneficial properties have been investigated actively in recent days. Probiotics administered into the digestive system have been shown to promote gut health by increasing the microbial balance in the gut. However, the bioavailability of such administered probiotics remains a major concern. These probiotics are protected through microencapsulation techniques, which encapsulate them in small capsules. Several nanoparticles with varied dimensions, forms, surfaces and composites have recently been investigated for probiotic microencapsulation. This has been used for various therapeutic applications, such as drug delivery. This review gives an insight on various materials and strategies used for probiotic encapsulation. The main aim of this review is to give a perception of the different types of methods of probiotic encapsulation. This review implies the significance of probiotics and subsequent active release in the gastrointestinal system. Different sections of this review paper, on the other hand, may offer up new opportunities for comprehensive research in the field of microencapsulation for boosting probiotic viability and also talks about the various encapsulating materials that has been employed. This review emphasizes more perceptions about the ongoing and imminent techniques for encapsulating probiotics.

Abstract:Percutaneous coronary intervention (PCI) via a transradial approach has become feasible for the treatment of chronic total occlusion (CTO). However, abnormalities such as anomalous origin coronary arteries represent the most technically challenging cases for interventional cardiologists. In this case, we describe a CTO in a rare anomalous right coronary artery (RCA) that originated from a high anterior takeoff with downward direction; transradial recanalization was the second attempt at RCA CTO and was successfully performed by employing a double-anchoring technique with an antegrade and retrograde approach.

Abstract:The increasing recognition of the potential advantages beyond nanoencapsulation of probiotics gained great attention owing to effective properties. Hence, we provided the most in-depth look into the influence of nanoformulated multi strain probiotics; BLB-NPs comprising Bacillus subtilis ATCC19659, Lactobacillus plantarum ATCC8014 and Bifidobacterium bifidum ATCC29521 on growth performance, antioxidant status and intestinal immunity supporting the defense against Salmonella Typhimurium (S. Typhimurium) challenge in broilers chickens. A total of 2,800 one-day-old male Ross 308 boiler chicks were divided into 7 groups; 1 control without additives, 3 probiotics [fed control diets mixed with B. subtilis, L. plantarum and B. bifidum (BLB) at concentrations of 1 × 104 (BLBI), 1 × 106 (BLBII) and 1 × 108 (BLBIII) CFU /kg diet, respectively] and 3 nanoencapsulated probiotics [fed control diets supplemented with BLB loaded nanoparticles (BLB-NPs) at concentrations of 1 × 104 (BLB-NPsI), 1 × 106 (BLB-NPsII) and 1 × 108 (BLB-NPsIII) CFU /kg diet, respectively]. All previous groups were challenged at d 22 of age with S. Typhimurium. Birds fed BLB-NPs II and III exhibited better weight gain and FCR simultaneously with upregulation in nutrients transporters genes (LAT-1, PepT-1, CAT-1 and SGLT1) even after S. Typhimurium challenge. Upregulation of immmune related genes (IL-1β, IL-6, IL-8, MyD88, NF-kB, CCL20, CXCLi2, TLR-2, TLR-4 and SOCS1) was prominently subsided in BLB-NPsIII fed group. The strengthening ability of BLB-NPs for broilers' intestinal barriers was evidenced by augmented expression of JAM, MUC-2, occludin and FABP-2 genes, diminished S. Typhimurium counts and suppressed its virulence related genes (HilA and SopD) with restored histopathological pictures of cecum. Notably, post dietary inclusion of higher levels of BLB-NPsIII, the abundance of beneficial Biofidobacterium and Lactobacillus species was dominated over harmful E. coli ones. Birds fortified with BLB-NPs displayed potent antioxidant potential signified by boosting serum and intestinal antioxidant markers alongside reducing oxidative ones. Overall, the abovementioned positive outcomes of BLB-NPs encouraged their potential application in poultry feed to attain superior performance and elicit protective immunity against S. Typhimurium infection.

Abstract:Probiotics are of increasing interest for their potential health benefits, but their survival and adhesion in the harsh gastrointestinal environment remain a concern. This study explored a single-cell encapsulation technique to enhance probiotic survival and adhesion in the gastrointestinal tract. We encapsulated probiotics in curcumin-loaded liposomes, further coated them with polymers using layer-by-layer techniques. The coated probiotics were evaluated for survival in simulated gastrointestinal conditions, adhesion to colonic mucus, and scavenging of reactive oxygen species (ROS). The results showed that multi-layer encapsulation increased probiotic size at the nanoscale, enhancing their survival in simulated gastrointestinal conditions. Upon reaching the colon, the shedding of the coating coincided with probiotic proliferation. Additionally, the coated probiotics exhibited increased adhesion to colonic mucus. Moreover, the coating acted as a protective barrier for effectively scavenging reactive oxygen radicals, ensuring probiotic survival in inflammatory environments. This study combines the synergistic effects of probiotics and curcumin, underscoring the promise of single-cell encapsulation techniques in improving the efficacy of probiotics for addressing colitis-related diseases.

Abstract:Alzheimer's disease, a progressive neurodegenerative disorder marked by cognitive decline, affects millions worldwide. The presence of amyloid plaques and neurofibrillary tangles in the brain is the key pathological feature, leading to neuronal dysfunction and cell death. Current treatment options include pharmacological approaches such as cholinesterase inhibitors, as well as non-pharmacological strategies like cognitive training and lifestyle modifications. Recently, the potential role of probiotics, particularly strains, such as Lactobacillus and Bifidobacterium , in managing neurodegenerative diseases through the gut–brain axis has garnered significant attention. Probiotics can modulate inflammation, produce neurotransmitters, and support neuronal health, potentially slowing disease progression and alleviating symptoms, such as stress and anxiety. Optimizing the pharmacotherapeutic effects of probiotics is critical and involves advanced formulation techniques, such as microencapsulation and nanoencapsulation. Microencapsulation employs natural or synthetic polymers to protect probiotic cells, enhancing their viability and stability against environmental stressors. Methods like extrusion, emulsion, and spray-drying are used to create microcapsules suited for various applications. Nanoencapsulation, on the other hand, operates at the nanoscale, utilizing polymeric or lipid-based nanoparticles to improve the bioavailability and shelf life of probiotics. Techniques, such as nanoprecipitation and emulsification, are employed to ensure stable nanocapsule formation, thereby augmenting the therapeutic potential of probiotics as nutraceutical agents. This study delves into the essential formulation aspects of microencapsulation and nanoencapsulation for beneficial probiotic strains, aimed at managing Alzheimer's disease by optimizing the gut–brain axis. The insights gained from these advanced techniques promise to enhance probiotic delivery efficacy, potentially leading to improved health outcomes for patients suffering from neurodegenerative disorders.

Abstract:The establishment of the health-promoting benefits of probiotics is challenged by the antimicrobial bio-barriers throughout the host’s gastrointestinal (GI) tract after oral administration. Although microencapsulation has been frequently utilised to enhance the delivery of probiotics, microcapsules of sub-100 μm were found to be ineffective and therefore questioned as an effective delivery vehicle for viable probiotics despite the sensory advantage. In this study, four probiotics strains were encapsulated in chitosan-coated alginate microcapsules of sub-100 μm. Only a minor protective effect was observed from this original type of microcapsule. In order to enhance the survival of these probiotics, sucrose, a metabolisable sugar, and lecithin vesicles were added to the wall material. Both of the ingredients could be readily encapsulated with the probiotics, and protected them from stresses in the simulated GI fluids. The metabolisable sugar effectively increased the survival of the probiotics in gastric acid, mainly through energizing the membrane-bound F1F0-ATPases. The lecithin vesicles proved to alleviate the bile salt stress, and hence notably reduced the viability loss at the elevated bile salt concentrations. Overall, three out of the total four probiotics in the reinforced sub-100 μm microencapsules could significantly survive through an 8-h sequential treatment of the simulated GI fluids, giving less than 1-log drop in viable count. The most vulnerable strain of bifidobacteria also yielded a viability increase of 3-logs from this protection. In conclusion, the sub-100 μm microcapsules can be a useful vehicle for the delivery of probiotics, as long as suitable protectants are incorporated in the wall matrix.

Abstract:The viability of probiotic cells decreases during passage through the gastrointestinal tract. The process of probiotics encapsulation with sodium alginate and chitosan polymers was carried out to protect the Lactobacillus plantarum in adverse conditions. Lactobacillus plantarum was entrapped in sodium alginate/chitosan (SA/BChi) and sodium alginate/nano-chitosan (SA/NChi) wall materials. Encapsulating L. plantarum with SA/BChi and SA/NChi resulted in a high encapsulation efficiency % of ~ 86.41 to 91.09%. In addition, coating bacteria cells in encapsulants improved the survivability of the cells under the simulated gastrointestinal fluids by ~ 52.61% in SA/Chi and 58.04% in SA/NChi compared to 29% for unencapsulated forms. Probiotic beads under field emission-scanning electron microscopy (FE-SEM) were morphologically compact with a cracked appearance of SA/NChi beads. The Fourier transform-infrared spectroscopy (FT-IR) and differential scanning calorimetry (DSC) showed vigorous electrostatic interaction between polymers, as well as the high melting points, which corroborate the previous investigations in the field for using SA/BChi or SA/NChi as a promising encapsulating agent for ameliorating the survivability of probiotics under harsh conditions. The distinctive properties possessed by the two coatings make them excellent candidates for use as polymeric carriers in probiotic delivery systems.

Molecular Design and Property Prediction for a Series of Novel Dicyclic Cyclotrimethylene Trinitramines (RDX) Derivatized as High Energy Density Materials.

Abstract:Quantum chemistry calculations and thermodynamics methods were carried out to screen out novel high energy density materials (HEDMs) from several new derivatives with dicyclic structures of Cyclotrimethylene trinitramine (RDX). Their volumes, densities, heats of formation, detonation properties and impact sensitivities have been calculated with thermodynamics methods under DFT B3LYP 6-31++g (d, p) level and all of these compounds exhibit good performance as HEDMs. Especially, R4 has given outstanding values as a potential HEDM. Its crystal density (2.07 g/cm(3)), heat of detonation (1.67 kJ/g), detonation velocity (10051m/s), and detonation pressure (48.5 GPa) are even higher than those of CL-20 while its impact sensitivity (h50, 16 cm) remains a relative safety value. The results indicate that the derivative work in common explosives is a good strategy which can design novel HEDMs with high energetic properties and low sensitivity. And furthermore, some mature processes can be used to synthesize them.

Abstract:Chitosan is an amino-polysaccharide made of glucosamine and N-acetyl-D-glucosamine. Owing to its biocompatible, biodegradable, and nontoxic nature, chitosan is considered as biomaterial, and these unique properties attested that chitosan has greater potential for biological applications. As cationic molecule, chitosan interacts with negatively charged gastrointestinal (GI) mucosal surface and hence is considered as potent mucoadhesive. Therefore chitosan-based encapsulation techniques provide better viability of probiotic microorganisms and protecting the latter in food products and at GI tract. In recent eons, chitosan or chitosan-based nanocomposites are considered as an attractive supportive matrix for enzyme immobilization because of the presence of reactive groups like amino and hydroxyl which supports long-term reusability of the immobilized biocatalyst. The current assignment highlights the recent research and cutting-edge strategies regarding chitosan-based nanomaterials in the field of probiotics encapsulation and enzyme immobilization.
Antioxidant Activity and Probiotic Properties of Lactic Acid Bacteria

Abstract:Oxidative stress, which can cause imbalance in the body by damaging cells and tissues, arises from the immoderate production of reactive oxygen species (ROS)/reactive nitrogen species (RNS). Therefore, external supplements having antioxidant activity are required for reducing oxidative stress. In our study, we investigated DPPH and ABTS radical scavenging ability, and the inhibition effect on the nitric oxide (NO) production of 15 food-derived bacterial strains in LPS-activated RAW264.7 cells. Among these LAB strains, eight strains with an excellent inhibition effect on NO production were selected through comparisons within the same genera. Moreover, the selected strains, including Leuconostoc mesenteroides MG860, Leu. citreum MG210, Pediococcus acidilactici MG5001, P. pentosaceus MG5078, Weissella cibaria MG5090, Levilactobacillus brevis MG5306, Latilactobacillus curvatus MG5020, and Latilactobacillus sakei MG5048 diminished the inducible nitric oxide synthase (iNOS)/cyclooxygenase-2 (COX-2) expression. In addition, the stability and adhesion ability of the eight LAB strains in the gastrointestinal tract were determined. In conclusion, the selected strains have potential as new probiotics with antioxidant effects.
Nanocarrier-mediated probiotic delivery: a systematic meta-analysis assessing the biological effects

Abstract:Probiotics have gained a significant attention as a promising way to improve gut health and overall well-being. The increasing recognition of the potential health advantages associated with functional food products, leading to a specific emphasis on co-encapsulating probiotic bacteria and bioactive compounds within a unified matrix. To further explore this concept, a meta-analysis was performed to assess the effects of probiotics encapsulated in nanoparticles. A comprehensive meta-analysis was conducted, encompassing 10 papers published from 2017 to 2022, focusing on the encapsulation of probiotics within nanoparticles and their viability in various gastrointestinal conditions. The selection of these papers was based on their direct relevance to the research topic. Random-effect models were used to aggregate study-specific risk estimates. In the majority of studies, it was observed that nano-encapsulated nanoparticles showed improved viability over time compared to their free state counterparts. At various time intervals, the odds ratios (OR) with 95% confidence intervals (CI) were estimated using fixed and random effect models. At 0 min, the OR (95%CI) was 2.79 (2.79; 2.80) and 2.38 (2.14; 2.64) for. At 30 and 60 min observation was at similar rate of 2.23 (2.23; 2.24) and 2.05 (1.73; 2.43). However, at 90 min it was 1.39 (1.39; 1.39) and 1.66 (1.29; 2.14) and at 120 min 2.41 (2.41; 2.42) and 2.03 (1.63; 2.52). Overall evaluation of encapsulation revealed an improvement in probiotic bacterial viability in simulated the gastrointestinal environments.

Antioxidant and Antibacterial Effects of Potential Probiotics Isolated from Korean Fermented Foods

Abstract:A total of sixteen bacterial strains were isolated and identified from the fourteen types of Korean fermented foods that were evaluated for their in vitro probiotic potentials. The results showed the highest survivability for Bacillus sp. compared to Lactobacillus sp. in simulated gastric pH, and it was found to be maximum for B. inaquosorum KNUAS016 (8.25 ± 0.08 log10 CFU/mL) and minimum for L. sakei KNUAS019 (0.8 ± 0.02 log10 CFU/mL) at 3 h of incubation. Furthermore, B. inaquosorum KNUAS016 and L. brevis KNUAS017 also had the highest survival rates of 6.86 ± 0.02 and 5.37 ± 0.01 log10 CFU/mL, respectively, in a simulated intestinal fluid condition at 4 h of incubation. The percentage of autoaggregation at 6 h for L. sakei KNUAS019 (66.55 ± 0.33%), B. tequilensis KNUAS015 (64.56 ± 0.14%), and B. inaquosorum KNUAS016 (61.63 ± 0.19%) was >60%, whereas it was lower for L. brevis KNUAS017 (29.98 ± 0.09%). Additionally, B. subtilis KNUAS003 showed higher coaggregation at 63.84 ± 0.19% while B. proteolyticus KNUAS001 found at 30.02 ± 0.33%. Among them, Lactobacillus sp. showed the best non-hemolytic activity. The highest DPPH and ABTS radical scavenging activity was observed in L. sakei KNUAS019 (58.25% and 71.88%). The cell-free supernatant of Lactobacillus sp. considerably inhibited pathogenic growth, while the cell-free supernatant of Bacillus sp. was moderately inhibited when incubated for 24 h. However, the overall results found that B. subtilis KNUAS003, B. proteolyticus KNUAS012, L. brevis KNUAS017, L. graminis KNUAS018, and L. sakei KNUAS019 were recognized as potential probiotics through different functional and toxicity assessments.
