Abstract:Due to their biodegradability and biocompatibility, chitosan-based hydrogels have great potential in regenerative medicine, with applications such as bacteriostasis, hemostasis, and wound healing. However, toxicity and high cost are problems that must be solved for chitosan-based hydrogel crosslinking agents such as formaldehyde, glutaraldehyde, and genipin. Therefore, we developed a biocompatible yet cost-effective chitosan-based hydrogel system as a candidate biomaterial to prevent infection during wound healing. The hydrogel was fabricated by crosslinking chitosan with dialdehyde chitosan (CTS–CHO) via dynamic Schiff-base reactions, resulting in a self-healable and injectable system. The rheological properties, degradation profile, and self-healable properties of the chitosan-based hydrogel were evaluated. The excellent antibacterial activity of the hydrogel was validated by a spread plate experiment. The use of Live/Dead assay on HEK 293 cells showed that the hydrogel exhibited excellent biocompatibility. The results demonstrate that the newly designed chitosan-based hydrogel is an excellent antibacterial wound dressing candidate with good biocompatibility.
A Novel Conductive Antibacterial Nanocomposite Hydrogel Dressing for Healing of Severely Infected Wounds

Abstract:Wound infections are serious medical complications that can endanger human health. Latest researches show that conductive composite materials may make endogenous/exogenous electrical stimulation more effective, guide/comb cell migration to the wound, and subsequently promote wound healing. To accelerate infected wound healing, a novel medical silver nanoparticle-doped conductive polymer-based hydrogel system (Ag NPs/CPH) dressing with good conductivity, biocompatibility, and mechanical and antibacterial properties was fabricated. For the hydrogel dressing, Ag NPs/CPH, polyvinyl alcohol (PVA), and gelatin were used as the host matrix materials, and phytic acid (PA) was used as the cross-linking agent to introduce conductive polyaniline into the matrix, with antibacterial Ag NPs loaded via impregnation. After a series of analyses, the material containing 5 wt% of PVA by concentration, 1.5 wt% gelatin, 600 μL of AN reactive volume, and 600 μL of PA reactive volume was chosen for Ag NPs/CPH preparation. XPS and FTIR analysis had been further used to characterize the composition of the prepared Ag NPs/CPH. The test on the swelling property showed that the hydrogels had abundant pores with good water absorption (≈140% within 12 h). They can be loaded and continuously release Ag NPs. Thus, the prepared Ag NPs/CPH showed excellent antibacterial property with increasing duration of immersion of Ag NPs. Additionally, to evaluate in vivo safety, CCK-8 experiments of HaCat, LO2 and 293T cells were treated with different concentrations of the Ag NPs/CPH hydrogel soaking solution. The experimental results showed the Ag NPs/CPH had no significant inhibitory effect on any of the cells. Finally, an innovative infection and inflammation model was designed to evaluate the prepared Ag NPs/CPH hydrogel dressing for the treatment of severely infected wounds. The results showed that even when infected with bacteria for long periods of time (more than 20 h), the proposed conductive antibacterial hydrogel could treat severely infected wounds.

Abstract:The preparation of hydrogel wound dressing materials with high antibacterial activities and good biosafety simultaneously can be relatively challenging. Aiming at solving this crucial problem, a multi-functional chitosan/carboxymethyl chitosan/silver nanoparticles (CTS/CMCTS/AgNPs) polyelectrolyte composite physical hydrogel is constructed by in situ photoreduction of silver ions with α-hydroxy of CMCTS and semi-dissolution acidification sol-gel transition (SD-A-SGT) methods using natural polymers with no chemical reductant involved. The XRD, TEM and IR characterizations reveal a highly uniform structure of the composite hydrogel with good mechanical strength. The composite hydrogel also exhibits good self-healing ability, and the self-healing mechanism of the composite hydrogel prepared by natural polymers with high similarity is put forward on the basis of molecular dynamics simulation. The AgNPs gives the hydrogel excellent antibacterial activities against both S. aureus and P. aeruginosa., while the preparation method with no chemical reductant and crosslinking agent required endows the hydrogel high biocompatibility. The great application potentials of CTS/CMCTS/AgNPs polyelectrolyte composite hydrogel are demonstrated with its promoting effects on the healing of P. aeruginosa infected wounds as the wound dressing. It is a breakthrough in achieving hydrogels with desired biosafety and antibacterial activities simultaneously. The composite hydrogel is found with great application potentials as wound dressing.
Histological findings and immunohistochemical surfactant protein A (SP-A) expression in asphyxia: its application in the diagnosis of drowning.

Abstract:The histopathological alterations that permit the diagnosis of death by asphyxia are very unspecific, although pulmonary alterations are of great importance in this respect. The postmortem diagnosis of drowning, particularly, continues to be one of the most difficult in forensic pathology. The aim of this study is to jointly evaluate microscopic findings and immunohistochemical surfactant protein A (SP-A) expression in the upper and lower lobes of lungs in different causes of death, and their possible application to the diagnosis of drowning. We studied 120 cadavers from subjects with a mean age of 48.73 years (SD 19.45; range 2-86 years), and with a mean post-mortem interval of 30 hours (SD 39.59; range 3-216 hours). According to the scene, cause and circumstances of death, and autopsy findings, cases were classified into groups as follows: (a) drowning (n=47); (b) other asphyxia (n=44) and (c) other causes (n=29). In the upper and lower lobes of lungs, histological studies of H&E staining and immunohistochemical surfactant protein A expression were made. The presence and severity of congestion, haemorrhage and oedema, together with immunohistochemical SP-A expression, may have a diagnostic value in differentiating asphyxia and drowning from other causes of death, and drowning from other types of asphyxia. Our findings suggest that both lobes should be investigated to establish the diagnosis, although the findings in the upper lobe might be the most important for differentiating the exact cause of death.

Abstract:Interactive wound dressings combining healing and antimicrobial potentials, besides ensuring patient compliance with a recognized wound care service gained considerable interest recently. Both hydrogel spray dried microparticles (HMP) and soft hydrogel (G) were prepared. The bio-inspired combinatory platform included natural bio-macromolecules namely: chitosan (CS) and collagen (COL) with wound healing enhancement and connective tissue building capabilities cross linked with the natural genipin (GN) to build a three dimensional structured matrix. The optimized plain hydrogel obtained by a box behnken design (BBD) program (G) scored maximum swelling and porosity. The network was hosted with green synthesized cefotaxime sodium (cef.Na) AgNPs reduced by the anabolic folic acid (FA). Both hydrogels exhibited good antimicrobial activity against gram +ve and -ve bacteria. The wound healing activity, evaluated in injured rats, showed >98 % and complete wound closure after two and three weeks respectively. Oxidative stress minimization was proved by the estimation of biochemical markers malondialdehyde (MDA) and superoxide dismutase (SOD) levels at the wound site.

Abstract:Wound infection resulting in delayed wound healing and wound deterioration remains a clinical challenge. Recently, multifunctional hydrogel dressing was a promising strategy which has attracted wide attention in preventing wound infection and promoting wound healing. In this study, a hybrid hydrogel made of gelatin (GL), tannic acid (TA), oxidized sodium alginate (OSA), and zinc oxide nanoparticles (ZnO NPs) was prepared mainly by double network cross-linking approach, named tannic acid-gelatin/oxidized sodium alginate/zinc oxide (TA-GL/OSA/ZnO). The composite hydrogels exhibited improved mechanical properties, which provided by TA modified the structure of GL network, Schiff base reaction between GL and OSA, and the strengthening effect of ZnO NPs. Meanwhile, the composite hydrogel showed high antibacterial activity against Staphylococcus aureus (S. aureus) (97.8 % ± 0.9 %) and Escherichia coli (E. coli) (96.6 % ± 1.2 %), attributed to the synergistic effect of TA and ZnO NPs. Furthermore, benefiting from the good antioxidative properties of TA, the sustain-released Zn2+ with the good capability to kill bacteria, and promoting the regeneration of skin epithelial tissues in BALB/c mice constantly, the multifunctional hydrogel had a significant therapeutic effect on wound healing and broad application prospects.

Abstract:Bacterial infections, excessive reactive oxygen species (ROS) accumulation and a persistent inflammatory response severely impede the wound healing process. In this study, we developed a novel multifunctional hydrogel dressing (LCPN) based on lipoic acid modified chitosan (LAMC), polypyrrole nanoparticles (PPy NPs) and nicotinamide mononucleotide (NMN) for accelerated healing of infected wounds. The synthesized LCPN hydrogel has several properties. Modification of lipoic acid significantly enhances the water solubility of chitosan making it easier to dissolve and absorb wound secretions. Interestingly, owing to the breaking and restructuring of disulfide bonds, LCPN hydrogel can be quickly bonded under UV light without relying on photoinitiators. In addition, the incorporation of PPy NPs not only enhances the electrical conductivity of LCPN hydrogel, but also confers photothermal antimicrobial capability to LCPN hydrogel. More importantly, the sustained release of NMN in LCPN hydrogel can significantly enhance cell proliferation, migration and antioxidant capacity, which is conducive to accelerated wound healing. In vitro and in vivo experiments have shown that LCPN hydrogel has excellent biocompatibility and the ability to promote wound healing. Therefore, the prepared multifunctional hydrogel is expected to be used as a novel dressing to accelerate wound healing.

Abstract:Wound healing involves a sophisticated biological process that relies on ideal conditions to advance through various stages of repair. Modern wound dressings are designed to imitate the natural surroundings around cells and offer properties such as moisture regulation, strength, and antimicrobial defense to boost healing. A recent research project unveiled a new type of gelatin (Gel)/dextran (Dex) hydrogels, linked through Diels-Alder (D-A) reactions, loaded with silver nanoparticles (Ag-NPs) for cutting-edge wound treatment. Gel and Dex were chemically modified to form the hydrogels via the D-A reaction. The hydrogels were enriched with Ag-NPs at varying levels. Thorough analyses of the hydrogels using methods like NMR, FT-IR, and SEM were carried out to assess their structure and nanoparticle integration. Rheological tests displayed that the hydrogels had favorable mechanical attributes, particularly when Ag-NPs were included. The hydrogels demonstrated controlled swelling, responsiveness to pH changes, and were non-toxic. Testing against E. coli showcased the strong antibacterial activity of the nanocomposite hydrogels in a concentration-dependent manner. This investigation showcased the promise of these bioactive nanocomposite hydrogels in promoting speedy wound healing by maintaining a moist environment, offering an antimicrobial shield, and ensuring mechanical support at the wound site.

Abstract:Abstract At present, the development trend of dressing materials is being multifunctional for convenient and long-term nursing care process of some complicated wounds. Here, basing on the theory of wound moist healing, an injectable and self-healing hydrogel comprising of collagen (COL), chitosan (CS) and oxidation modified Konjac glucomannan (OKGM), which acts as a macromolecular cross-linker to construct dynamic Schiff-base bonds was smartly designed. The strategy of introducing the silver nanoparticles (Ag NPs) into the COL–CS–OKGM hydrogel matrix achieved a markedly enhanced antibacterial activity derived from the synergistical effect between the Ag+ and the mild photothermal efficacy of Ag NPs, which also improved the local capillary blood circulation of the wound area to further facilitate wound healing process. The excellent syringeability and self-healing behaviors endowed the COL–CS–OKGM–Ag hydrogel with self-adapting ability for the wounds with irregular and large area needing frequent applying and changing without secondary injuries. In vitro and in vivo evaluations verified that so-designed COL–CS–OKGM–Ag hydrogel also with hemostatic performance is a promising multifunctional dressing for the treatments of infected wound with not only good biocompatibility and convenient use, but also with desired regenerative healing prognoses benefited from hydrogel moist environment and physiotherapy.

Abstract:Skin wounds are susceptible to infection, leading to severe inflammatory reactions that can progress to chronic wounds, ultimately causing significant physical and mental distress to the patient. In this study, we propose an injectable composite hydrogel achieved through one-pot gelation of oxidized xyloglucan (OXG), cationic polyamide ε-poly-l-lysine (EPL), and surface amino-rich silicon nanoparticles (SiNPs). OXG exhibits commendable anti-inflammatory properties and provides crosslinking sites. SiNPs serve as mechanically reinforced crosslinkers, facilitating the construction of a dynamic Schiff base network. SiNPs significantly reduced the gelation time to 3 s and tripled the storage modulus of the hydrogels. Additionally, the combination of EPL and SiNPs demonstrated synergistic antimicrobial activity against both S. aureus and E. coli. Notably, the hydrogel effectively halted liver bleeding within 30 s. The hydrogel demonstrated outstanding shear-thinning and self-healing properties, crucial considerations for the design of injectable hydrogels. Furthermore, its efficacy was evaluated as a wound dressing in a mouse model with S. aureus infection. The results indicated that, compared to commercial products, the hydrogel exhibited a shorter wound healing time, decreased inflammation, thinner epithelium, increased hair follicles, enhanced neovascularization, and more substantial collagen deposition. These findings strongly suggest the promising potential of the proposed hydrogel as an effective wound dressing for the treatment of infected wounds.

Abstract:Infections from bacteria and fungi, along with ongoing inflammation in chronic wounds, can prolong the healing process and potentially lead to cancers. Hydrogel dressings, when used as wet dressings, have shown promise in accelerating wound healing and minimizing further wound damage. We developed a hydrogel dressing incorporating natural small molecules, specifically Lipoic acid and Magnolol, to reduce reliance on hazardous organic solvents and heavy metal ions. This development employed an environmentally friendly and sustainable approach, contributing to energy conservation. The fabricated hydrogel underwent testing to evaluate its injectability, antioxidant properties, sustained drug release, in vitro degradability, and effectiveness in healing infected wounds. Notably, the hydrogel exhibited low cytotoxicity, enhancing cell proliferation at concentrations below 62.5 mg/L. It also demonstrated anti-inflammatory and antifungal properties, alongside substantial antibacterial qualities that facilitate the healing of bacterial-infected wounds. Furthermore, the material’s recyclability has been established. Multiple studies have validated the efficacy of this multifunctional, recycled hydrogel, underscoring its potential as a sustainable dressing for treating infected wounds.

Abstract:Burns and scalds often result in deep wounds that challenge adequate debridement and inflammation control using traditional sheet-like hydrogel dressings. Herein, we developed an antibacterial, injectable, and self-healing hydrogel (ADCM@Ag) by employing carboxymethyl chitosan (CMCS) for in situ green reduction of silver ions and utilizing a spontaneous Schiff base reaction with aldehyde-functionalized dextran (AD). SEM analysis revealed a porous structure within the hydrogel. Swelling and enzymatic degradation assays demonstrated that ADCM@Ag hydrogel possesses excellent fluid absorption capacity and biodegradability. Mechanical tests indicated good mechanical properties, allowing the hydrogel to withstand external forces when applied to animal wounds. The hydrogel exhibited good injectability, shape adaptability, and self-healing capability. Cell experiments showed that the ADCM@Ag hydrogel avoided the cytotoxicity caused by high concentrations of silver ions and had good cell compatibility. Antimicrobial assays showed that ADCM@Ag exhibited potent bactericidal effects against Gram-negative and Gram-positive bacteria, achieving at least 85% killing efficacy. Collectively, ADCM@Ag hydrogel has good potential for wound dressing applications.

Abstract:Hydrogel dressings with similar structural characteristics to the extracellular matrix and tunable physicochemical properties have become promising candidates for wound healing. However, the fabrication of an ideal hydrogel dressing with low-cost, good biocompatibility, excellent hemostatic capacity, potent and broad-spectrum antibacterial activity remains a huge challenge. Herein, a lignocellulose-based nanocomposite hydrogel (ATC/SA/PVA) is fabricated by simply mixing Ag nanoparticles loaded, tannic acid-decorated lignocellulose nanofibrils with sodium alginate and polyvinyl alcohol. Based on the dynamic borate ester bonds and multiple weak hydrogen bonds, the fabricated hydrogel exhibits excellent flexibility and self-healing performance. Its highly porous structure endows the gel excellent blood and tissue exudates absorption ability. Interestingly, the release behavior of Ag nanoparticles from hydrogel displays pH dependence, which can facilitate the accumulation of Ag nanoparticles at the wound site, thereby accelerating the process of wound healing. In vitro antibacterial assay demonstrates the potent antibacterial ability of hydrogel against both Gram-positive (S. aureus) and negative bacteria (E. coli). More importantly, in vivo investigations reveal that such hydrogel can effectively accelerate tissue regeneration and wound healing with no obvious adverse effects. All these results suggest that this nanocomposite hydrogel would be a promising candidate to accelerate wound healing.
Efficient Antibacterial Polyphosphazene Material with Potential to Prominent Wound Healing

Abstract:Reducing the toxicity of silver-based antibacterial agents used in gels and improving their biocompatibility are of great significance in the development of wound dressings. Thus, a highly crosslinked organic–inorganic hybrid porous polyphosphazene nanospheres (PRV-HMSs) were prepared via precipitation polymerization. The structures and properties were investigated. The results indicated that PRV-HMSs have uniform particle size about 500 nm, excellent thermal stability, high silver loading (26.5 wt.%) and remarkable sustained-release properties after loading AgNO3 (up to 2 weeks). The antibacterial effects were investigated with zone of inhibition testing, minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), Live/Dead bacterial staining assay and bacterial growth curve assays. The silver-loaded nanospheres have excellent bactericidal effects for E. coli or S. aureus and an average bactericidal rate of 94.02% and 94.67% in the 120 hour long-acting antibacterial test. The cytotoxicity and laser confocal immunofluorescence staining experiments indicate that the simple combination of nanospheres and agar not only give low cytotoxicity, but also significantly promote the activation of macrophages, which showing potential application in the wound dressings based on hydrogel.

Abstract:Hydrogels containing antimicrobial materials have emerged as attractive platforms for wound treatment in the past decade due to their favorable bio-mimicking properties, excellent modulation of bacterial infection, and ability to minimize bacterial resistance. Herein, a hybrid combination of polyvinyl alcohol (PVA), hyperbranched poly L-lysine (L), tannic acid decorated AgNPs (AgTA NPs), loaded with Allantoin (Alla) is used to fabricate PLAg-Alla hydrogel dressing via the freeze-thaw method without use of any chemical cross-linker. The PLAg-Alla hydrogel possesses a great structure, is biodegradable, and safe, and exhibits high antibacterial potential, all required for efficient wound healing. The incorporation of AgTA and poly L-lysine (L) within the hydrogel contributes to the enhancement of antibacterial ability, as well as effectively promoting the wound healing. This hybrid hydrogel possessed favorable physicochemical features, robust antibacterial properties, and accelerated wound healing in vivo as promising dressing for the clinical application.

Abstract:Clinical application of antibiotic-free agents like silver nanoparticle-derived materials remains a critical challenge due to their limited long-term antibacterial activity and potential system toxicity. Herein, a highly biocompatible Ag nanocluster-reinforced hydrogel with enhanced synergistic antibacterial ability has been developed. Specifically, bioactive curcumin was incorporated into lysozyme-protected ultrasmall Ag nanoclusters (LC-AgNCs) and further integrated with sodium alginate (Sa) hydrogel (LC-AgNCs@Sa) through multiple interaction forces. Due to the synergistic antibacterial activity, LC-AgNCs could effectively kill both S. aureus and E. coli bacteria with a concentration down to 2.5 μg mL-1. In-depth mechanism investigations revealed that the bactericidal effect of LC-AgNCs lies in their bacterial membrane destruction, reactive oxygen species (ROS) production, glutathione depletion and prooxidant-antioxidant system disruption ability. Curcumin can mediate the intracellular ROS balance to protect NIH 3T3 cells from oxidative stress and improve the biocompatibility of LC-AgNCs@Sa. LC-AgNCs@Sa with long-term antibacterial ability can effectively protect the wound from bacterial invasion in vivo, and significantly accelerate the wound healing process due to their distinctive functions of inhibiting inflammatory factor (TNF-α) production, promoting collagen deposit and facilitating re-epithelization. This study provides a new, versatile strategy for the design of high-performance antibacterial dressing for broad infectious disease therapy.

Abstract:The ever-growing challenges of traditional antibiotic therapy and chronic wound healing have created a hot topic for the development and application of new antimicrobial agents. Silver nanoclusters (Ag NCs) with ultrasmall sizes (<2 nm) and antibacterial effects are promising candidates for next-generation antibiotics, particularly against multi-drug resistant strains. However, the biosafety in the clinical application of Ag NCs remains suboptimal despite some existing studies of Ag NCs for biomedical applications. Considering this, an ultrasmall Ag NC with excellent water solubility was synthesized by a two-phase ligand-exchange method, which exhibits broad-spectrum antibacterial performance. The minimum inhibitory concentrations of Ag NCs against MRSA , S. aureus , P. aeruginosa and E. coli were evaluated as 50, 80, 5 and 5 μg mL −1 , respectively. Furthermore, a carbomer hydrogel was prepared to be incorporated into the Ag NCs for achieving excellent biocompatibility and biosafety. In vitro experiments demonstrate that the Ag NC-gel exhibits good antibacterial properties with lower cytotoxicity. Finally, in vivo experiments suggest that this ultrasmall Ag NC functionalized with the hydrogel can serve as an effective and safe antimicrobial agent to aid in wound healing.

Abstract:Anti-infection and neovascularization at the wound site are two vital factors that accelerate diabetic wound healing. However, for a wound healing dressing, the two functions need to work at different sites (inner and outer), giving big challenges for dressing design. In this study, we fabricated a novel sodium alginate/chitosan (SA/CS) Janus hydrogel dressing by the assembly of SA hydrogel loaded with silver nanoparticles (AgNPs) and CS hydrogel impregnated with l -arginine loaded sodium alginate microspheres (ArgMSs) based on electrostatic interactions to combine the two functions. The outer SA-AgNP hydrogel could prevent infection while avoiding the deposition of AgNPs in the wound site, and the inner CS-ArgMS hydrogel on the wound surface could realize the sustained release of l -arginine and promote vascular regeneration. The composition, morphology and swelling/degradation of the SA-AgNP/CS-ArgMS hydrogel were characterized systematically. l -Arginine release behavior has been tested and SA-AgNP/CS-ArgMS hydrogel has been confirmed for excellent biocompatibility. Antibacterial and angiogenesis assays demonstrated the antibacterial and angiogenesis characteristics of the SA-AgNP/CS-ArgMS hydrogel. Finally, in vivo diabetic wound healing assay demonstrated that the SA-AgNP/CS-ArgMS hydrogel could significantly accelerate re-epithelialization, granulation tissue formation, collagen deposition and angiogenesis, thereby resulting in enhanced diabetic wound healing
