Abstract:Objective To investigate the protein expression level of laminin 332α3 chain (LNα3) and the correlation of its expression pattern with pancreatic carcinoma. Methods The expression level and pattern of LNα3 were detected by immuno-histochemistry with EnVision method in 65 pancreatic carcinoma specimens and adjacent normal tissues. The correlation of LNα3 expression and its pattern with clinicopathologic features and prognosis was analyzed. Results The positive rate of LNα3 was significantly higher in 65 pancreatic carcinoma specimens (100.0%) than that in adjacent normal tissues (1.6%, P<0.01). The single cytoplasmic expression type (SCT) was presented in 42 cases and the cytoplasmic/mesenchymal coexpression type(CMT) was in 23 cases. The different expression patterns of LNα3 were associated with hepatic metastasis and vascular in-vasion and bile duct invasion (P<0.05 or 0.01) and the CMT type indicated a higher risk. The overal survival of patients was sig-nificantly correlated with hepatic metastasis, T stages, duodenal invasion and tumor location(P<0.05 or 0.01). Tumor location and T stage were independent predictors of overal survival (P<0.05). The tumor location and higher clinical stages were correlated with poor prognosis, while hepatic metastasis and invasion to serosa and duodenal invasion were not the independent predictors (P>0.05). Conclusion The expression of LNα3 is closely correlated with pancreatic carcinoma and its invasion and metastasis, especial y the CMT type can be used as an important prognostic index.
P-STAT3 and IL-17 in Tumor Tissues Enhances the Prognostic Value of CEA and CA125 in Patients with Lung Adenocarcinoma

Abstract:Aim: The present study aimed to examine the capability of p- signal transducer and activator of transcription (STAT)3 and interleukin-17 (IL-17), along with two known tumor markers carcinoembryonic antigen (CEA) and carbohydrate antigen 125 (CA125), for disease prognosis. Moreover, the associations among biomarkers and clinicopathological parameters were evaluated to uncover the potential mechanisms responsible for their correlations with lung adenocarcinoma (LAD) prognosis. Methods: Five LAD-related parameters were used in the study: CEA, CA125, STAT3, p-STAT3, and IL-17. Spearman and chi-square correlation tests were used to explore the relationships between some clinicopathological variables and parameter expression levels and the associations among these five parameters. Results: The disease-specific survival decreased with the positive expression of CEA, CA125, p-STAT3, and IL-17, with no significant difference in the expression level of STAT3. Combinations of p-STAT3 and IL-17, CEA and p-STAT3, CEA and IL-17, CA125 and p-STAT3, and CA125 and IL-17 had higher predictive values in LAD prognosis. The correlation analyses indicated the synergic activities of STAT3, p-STAT3, and IL-17 and the coordinated expression of CEA, CA125, p-STAT3, and IL-17. The tumor-node-metastasis (TNM) stage significantly correlated with the levels of CA125 and p-STAT3. Conclusions: Elevated levels of CEA, CA125, p-STAT3, and IL-17 alone and/or combinations of p-STAT3 and IL-17, CEA and p-STAT3, CEA and IL-17, CA125 and p-STAT3, and CA125 and IL-17 were recommended as the prognostic predictors of unfavorable clinical outcomes in patients with postoperative LAD. Also, p-STAT3 and IL-17 combined with CA125 and CEA helped in predicting the overall survival of patients with LAD and informing the TNM stage.

Abstract:Pancreatic ductal adenocarcinoma (PDA) is a worldwide health problem. Early diagnosis and assessment may enhance the quality of life and survival of patients. The present study investigated the potential correlations between the gene and protein expression of laminin-332 (LM-332 or laminin-5) and clinicopathological factors as well as evaluating its influence on the survival of patients with PDA. The expression of LM-332 subunit mRNAs in pancreatic carcinoma specimens from 37 patients was investigated by reverse transcription-quantitative polymerase chain reaction (RT-qPCR) analysis. Using immunohistochemical methods, the protein expressions of the three chains of LM-322 (LNα3, LNβ3 and LNγ2) were determined in 96 pancreatic carcinoma specimens, for association analysis with clinicopathological characteristics from patient data. The results of the prognosis analysis of three mRNAs expression datasets were validated in The Cancer Genome Atlas datasets. RT-qPCR results indicated that the overall relative values of LNα3 and LNγ2 mRNAs were increased in pancreatic carcinoma compared with the control. In immunostaining analyses LNα3 and LNγ2 expression was observed in all tumor tissues from the 96 patient samples. The expression levels of LNα3, LNβ3 and LNγ2 were associated with each other. LNα3 and LNγ2 positivity was significantly associated with differentiation, depth of invasion and advanced stage (P<0.05). The samples were classified into three groups: Basement membrane (B) type, cytoplasmic (C) type and mixed (M) type, according to their LNγ2 immunohistochemical expression patterns. The B type correlated significantly with differentiation (P=0.010) and the M type was significantly associated with hepatic metastasis (P=0.031). Patients with B-type LNγ2 demonstrated significantly better outcomes than patients with the C or M type (P=0.012 and P=0.003, respectively). Overexpression of the α3, β3 and γ2 chains of LM-332 may serve an important role in the progression and prognosis of PDA.

Abstract:Background:Laminin-332 (LM-332, formerly termed laminin-5) is a heterotrimeric glycoprotein that promotes cellular adhesion and migration. The heterotrimer consists of an 3, a 3, and a 2 chain. The aim of this investigation was to clarify the clinicopathologic significance of laminin-3323 (LN3) chain expression and determine its influence on survival in pancreatic ductal adenocarcinoma.Materials and Methods:Quantitative real-time polymerase chain reaction was used to validate and detect the expression of LN3 mRNA in 37 pancreatic carcinoma tissue specimens and non-neoplastic pancreatic tissue samples. In addition, the protein expression of LN3 was detected by immunohistochemistry methods in 96 pancreatic carcinoma specimens and 90 non-neoplastic pancreatic tissues. We analyzed the association between immunohistochemically detected LN3 expression in pancreatic ductal adenocarcinoma and clinicopathologic characteristics. Survival curves were completed using the Kaplan-Meier method and compared using log-rank analysis.Results:Quantitative real-time polymerase chain reaction indicated that the relative value of LN3 mRNA was 1.4271.554 and 1.423 +/- 1.439 by 2(-(Ct)) in pancreatic carcinoma and non-neoplastic pancreatic tissues, respectively, values that were not statistically associated (P=0.991). Immunostaining for LN3 was expressed in all patients with pancreatic ductal adenocarcinoma. LN3 expression was related to differentiation (P=0.000) and advanced stage (P=0.034). Tumors with low expression of LN3 had a survival advantage compared with tumors that had high expression of LN3 (P=0.016). Multivariate analysis indicated that location is an independent predictor of overall survival, whereas other clinicopathologic characteristics such as tumor size, duodenal invasion, differentiation, extent of invasion, hepatic metastasis, and expression of LN3 were not.Conclusion:Our results suggest that LN3 expression may play a key role in the progression and prognosis of pancreatic ductal adenocarcinoma.
Wnt3a Expression Is Associated with Epithelial-Mesenchymal Transition and Impacts Prognosis of Lung Adenocarcinoma Patients.

Abstract:Background: Epithelial-mesenchymal transition (EMT) plays an important role in the invasion and migration during cancer metastasis. Wnt3a is one of the ligands in canonical Wnt/beta-catenin signaling pathway, which contributes to the carcinogenesis and progression of lung cancer cell lines. The aim of this study was to evaluate the association between Wnt3a and EMT-related proteins (E-cadherin and N-cadherin), and to further investigate its impact on prognosis of lung adenocarcinoma patients. Methods: A total of 147 lung adenocarcinoma patients were included and their clinicopathological characteristics were collected in this retrospective study. The expression levels of Wnt3a, E-cadherin and N-cadherin in post-surgery cancerous and adjacent normal tissues were assessed by immunohistochemistry. The association between Wnt3a and EMT-related proteins and their prognostic values were systematically evaluated. HCC827 and PC9 cell lines were treated with Wnt3a to detect the expression of EMT-related and Wnt/beta-catenin signaling-associated proteins, as well as the in vitro migration and invasion abilities. Results: High Wnt3a expression level was significantly associated with low E-cadherin (P<0.001) and high N-cadherin (P<0.001) expression levels in lung adenocarcinoma tissues. Besides, high Wnt3a level predicted poorer lung adenocarcinoma survival by univariate Cox analysis (P=0.001), while the multivariate result was not significant (P=0.355). Subgroup analysis suggested that the prognostic value of Wnt3a expression level was significant in stage T1-T2 (log rank P=0.003) and stage N0 (log rank P=0.031) patients. The multivariate Cox analysis suggested N-cadherin was an independent prognostic factor for lung adenocarcinoma patients (P=0.012). After including these markers into a nomogram, the Harrell's C-index of the nomogram was 0.755. The decision-curve analysis of our nomogram performed net benefit at the threshold probability from 21.6% to 82.0%, and the current model had a better prognostic value than TNM-classification with a lower Akaike information criterion (AIC) value of 166.54. In vitro experiments suggested that Wnt3a could regulate EMT-related proteins and promotes in vitro invasion and migration abilities. Conclusions: Wnt3a could regulate EMT-related proteins and promote the migration and invasion process of lung adenocarcinoma. Although its value as an independent prognostic factor was limited, the combined model suggested good prognostic performance for lung adenocarcinoma patients.
Correlation of LAMA3 with onset and prognosis of ovarian cancer

Abstract:Correlation of laminin subunit α3 (LAMA3) gene with onset and prognosis of ovarian cancer was investigated. In total, 210 ovarian cancer patients who received surgical resection in West China Second Hospital from March 2011 to March 2013 were randomly selected, and another 160 non-ovarian cancer patients who needed ovariectomy were also selected. The relative expression of LAMA3 gene was compared via quantitative polymerase chain reaction (qPCR) in carcinoma tissues, para-carcinoma tissues and non-carcinoma normal tissues in ovarian cancer patients. The methylation level was compared among the above three tissue types. The correlation between the mutation site rs12373237 in LAMA3 gene and onset was analyzed. The expression of laminin in ovarian cancer was detected using immunohistochemistry. Moreover, the 5-year survival rate after operation was recorded and the survival curve was plotted. The expression level of LAMA3 was lower in carcinoma tissues than those in normal tissues and para-carcinoma tissues (P<0.05). The methylation degree was lower in para-carcinoma tissues and normal tissues than that in carcinoma tissues (P<0.05). The CC homozygous mutation of rs12373237 was highly correlated with the onset of ovarian cancer (OR=4.333, P=0.028). The expression of LAMA3 was classified via immunohistochemistry, and the number in high-expression group (63.8%) was larger than that in low-expression group (36.2%) (P<0.05). According to the analysis of 5-year survival rate, the recurrence-free survival rate and overall survival rate in LAMA3 high-expression group were significantly higher than those in LAMA3 low-expression group (P<0.05). The expression level and base mutation of LAMA3 gene can change the level of laminin, which have a certain influence on the onset and prognosis of ovarian cancer.

Apobec3g Expression is Correlated with Poor Prognosis in Colon Carcinoma Patients with Hepatic Metastasis

Abstract:Increased expression of apolipoprotein B mRNA-editing enzyme catalytic polypeptide-like 3G (APOBEC3G) in human primary colorectal tumors and hepatic metastasis has been detected. However, the clinical relevance of APOBEC3G in colon carcinoma hepatic metastasis remains uncertain. The aim of this study was to assess the prognostic value of APOBEC3G in colon carcinoma patients with hepatic metastasis after hepatic resection. APOBEC3G expression was evaluated by immunohistochemistry in paraffin-embedded primary colon carcinoma and paired hepatic metastasis tissues from 136 patients with liver metastasis from colon carcinoma that underwent hepatic resection. The relation between APOBEC3G expression and clinicopathologic factors and long-term prognosis in these 136 patients was retrospectively examined. The prognostic significance of negative or positive APOBEC3G expression in colon carcinoma hepatic metastasis was assessed using Kaplan-Meier survival analysis and log-rank tests. Positive expression of APOBEC3G was correlated with liver metastasis of colon cancer. Univariate analysis indicated significantly worse overall survival (OS) for patients with a positive APOBEC3G expression in colon carcinoma hepatic metastasis than for patients with a negative APOBEC3G expression. Multivariate analysis showed positive-APOBEC3G in colon carcinoma hepatic metastasis to be an independent prognostic factor for OS after hepatic resection (P = 0.000). Positive expression of APOBEC3G was statistically significantly associated with poor prognosis of colon carcinoma patients with hepatic metastasis. APOBEC3G could be a novel predictor for poor prognosis of colon carcinoma patients with hepatic metastasis after hepatic resection.
Effect of LAMA4 on Prognosis and Its Correlation with Immune Infiltration in Gastric Cancer

Abstract:Background: Laminin alpha 4 (LAMA4) is widely distributed in the basement membranes of various tissues. It can regulate cancer cell proliferation and migration. We investigated the effects of LAMA4 in gastric cancer (GC). Methods: LAMA4 expression patterns were analyzed in GC using the Gene Expression Omnibus (GEO), Gene Expression Profiling Interactive Analysis (GEPIA), and UALCAN. Correlations between LAMA4 expression and clinicopathological characteristics were evaluated using data from The Cancer Genome Atlas (TCGA). The survival analysis was examined using the Kaplan-Meier plotter and GEPIA and ascertained by multivariate Cox analysis. Genetic alterations and DNA methylation of LAMA4 were analyzed using cBioPortal and MethSurv. LinkedOmics was applied to identify coexpressed genes of LAMA4. The association between LAMA4 and infiltration of immune cells was explored using Tumor Immune Estimation Resource (TIMER) and GEPIA. Results: LAMA4 was highly expressed in GC, and its upregulation significantly correlated with T classification (P = 0.040). LAMA4 expression was an independent risk factor for overall survival (OS, P = 0.033). Patients with genetic alterations of LAMA4 showed a significantly better disease-free survival (DFS, P = 0.022). Ten CpG sites of LAMA4 were significantly associated with prognosis in GC. The functions of LAMA4 and coexpression genes were mainly involved in extracellular matrix (ECM) receptor interaction. LAMA4 expression significantly correlated with infiltration of macrophages (P < 0.001), CD4+ T cells (P < 0.001), and dendritic cells (P < 0.001). Furthermore, LAMA4 expression was significantly associated with markers of M2 and tumor-associated macrophages (TAMs). Conclusion: LAMA4 expression was linked to GC prognosis and immune cell infiltration, indicating its potential use as a prognostic biomarker and therapeutic target.
A pan-cancer analysis of prognostic significance and immunological role of lysosomal-associated membrane protein 3

Abstract:Lysosomal dysfunction can drive carcinogenesis. Lysosomal-associated membrane protein 3 (LAMP3), is a member of the Lysosome Associated Membrane Proteins and is involved in the malignant phenotype such as tumour metastasis and drug resistance, while the mechanisms that regulate the malignant progression of tumour remain vague. Our study aims to provide a more systematic and comprehensive understanding of the role of LAMP3 in the progression of various cancers by various databases.We explored the role of LAMP3 in pan-cancer using The Cancer Genome Atlas (TCGA) and Genotype-Tissue Expression (GTEx) database. Multiple online web platforms and software were used for data analysis, including HPA, TIMER, TISIDB, GEPIA, UALCAN, Kaplan-Meier plotter, DAVID and TIGER. The immunohistochemistry was used to quantify the LAMP3 and PD-L1 expression levels in cancer.High LAMP3 expression was found in most cancers and differentially expressed across molecular and immune subtypes. The expression of LAMP3 was involved in the immune-associated processes of Antigen processing and presentation, Th17 cell differentiation, Th1 and Th2 cell differentiation, and the immune-associated pathways of T cell receptor and B cell receptor signalling pathways in most cancers. It also correlated with genetic markers of immunomodulators in various cancers. LAMP3 and PD-L1 expression in BRCA and HNSC tissues was higher than that in corresponding adjacent normal tissues by immunohistochemistry. There is a significant correlation between the expression of LAMP3 and PD-L1.Our study elucidates that LAMP3 has different expression patterns and genetic alteration patterns in different tumours. It is a potential biomarker for immune-related cancer diagnosis, prognosis and efficacy prediction.
Laminin 332 expression levels predict clinical outcomes and chemotherapy response in patients with pancreatic adenocarcinoma

Abstract:Poor outcomes and chemotherapy resistance for patients with pancreatic adenocarcinoma (PAAD) are a challenge worldwide, and new or improved prognostic biomarkers are urgently required. Individual laminin family members have been established as cancer-associated markers, predicting patient outcomes in many cancer types, including PAAD. Here, we used multiple modalities including RNAseq and gene chip, and genomic and proteomic data to examine the relationships of all laminin genes in PAAD with clinical outcomes. These analyses identified that LAMA3, LAMB3, and LAMC2 expression levels are increased at the mRNA and protein levels in PAAD tumours with evidence of co-regulation. Increased expression of all three genes was associated with decreased promoter methylation status, TP53 mutations, and altered receptor tyrosine kinase (RTK) pathways. Clinically, high LAMA3, LAMB3, and LAMC2 transcript abundance was each related to an advanced histological grade. Moreover, high expression of these genes individually predicted poor patient survival, while a signature of combined high expression of LAMA3, LAMB3, and LAMC2 was a stronger predictor of patient outcomes than each gene alone. Interestingly, cell lines with high expression of LM332 chains were not sensitive to the commonly used PAAD chemotherapy drugs paclitaxel and gemcitabine; however, increased sensitivity was evident for erlotinib, afatinib, gefitinib, and cetuximab epidermal growth factor (EGFR) RTK inhibitors. To explore possible mechanisms, we investigated co-expressed genes, identifying eight hub genes, namely, GJB3 , ITGB6 , SERPINB5 , GPRC5A , PLEK2 , TMPRSS4 , P2RY2 , and TRIM29 , which are co-expressed with all three of LAMA3, LAMB3, and LAMC2. Of these, only SERPINB5 provided a stronger predictive value than the laminin-encoding genes. Together, these multiple integrated analyses suggest that the combined expression of LM332 is a useful prognostic biomarker for PAAD and could help patient stratification and therapeutic selection.

Liver-Metastasis-Related Genes Are Potential Biomarkers for Predicting the Clinical Outcomes of Patients with Pancreatic Adenocarcinoma.

Abstract:It is widely acknowledged that metastasis determines the prognosis of pancreatic adenocarcinoma (PAAD), and the liver is the most primary distant metastatic location of PAAD. It is worth exploring the value of liver-metastasis-related genetic prognostic signature (LM-PS) in predicting the clinical outcomes of PAAD patients post R0 resection. We collected 65 tumors and 165 normal pancreatic data from The Cancer Genome Atlas (TCGA) and the Genotype-Tissue Expression project (GTEx), respectively. Differentially expressed genes (DEGs) between primary tumor and normal pancreatic samples were intersected with DEGs between primary tumor samples with liver metastasis and those without new tumor events. The intersected 45 genes were input into univariate Cox regression analysis to identify the prognostic genes. Thirty-three prognostic liver-metastasis-related genes were identified and included in least absolute shrinkage and selection operator (LASSO) analysis to develop a seven-gene LM-PS, which included six risk genes (ANO1, FAM83A, GPR87, ITGB6, KLK10, and SERPINE1) and one protective gene (SMIM32). The PAAD patients were grouped into low- and high-risk groups based on the median value of risk scores. The LM-PS harbored an independent predictive ability to distinguish patients with a high-risk of death and liver metastasis after R0 resection. Moreover, a robust prognostic nomogram based on LM-PS, the number of positive lymph nodes, and histologic grade were established to predict the overall survival of PAAD patients. Besides, a transcription factor-microRNA coregulatory network was constructed for the seven LM-PS genes, and the immune infiltration and genomic alterations were systematically explored in the TGCA-PAAD cohort.
Comprehensive Analysis of the Expression and Prognosis for Laminin Genes in Ovarian Cancer

Abstract:Survival is low in ovarian cancer (OC). Most OC patients demonstrate advanced metastases, and recurrence is common. Dysregulation of laminin interactions is associated with cancer development. However, it is unknown whether laminin subunits can be considered as biomarkers for OC diagnosis, prognosis, and treatment. We used cBioPortal, GEO, ONCOMINE, GEPIA, Human Protein Atlas, Kaplan-Meier Plotter, TIMER, and Metascape to determine the associations among laminin expression, prognosis, and immune cell infiltration in OC. LAMA5, LAMB3, and LAMC2 mRNAs and LAMA3, LAMB1/B2/B3, and LAMC1/C2 proteins were overexpressed in OC tissues compared with normal ovaries. LAMA4, LAMB1, and LAMC1 mRNA upregulation was positively correlated with worse overall survival (OS) and progression-free survival (PFS) in OC. Elevated LAMA2 and LAMC2 mRNA expression levels were related to better PFS or OS, respectively. The results speculated that LAMA5 could potentially be a good prognostic factor in OC. Its expression proves valuable for predicting OS in patients diagnosed with stage IV and grade 3 OC and PFS in patients diagnosed with all OC stages or grades. LAMB3 and LAMC2 expression was correlated with platinum resistance development. ROC analysis of laminins in OC sets revealed that LAMA2/A4/A5, LAMB1/B2/B3, and LAMC2 could be used to differentiate between malignant tumors and non-neoplastic tissues. LAMA1/A5 and LAMC1 were significantly and negatively correlated with various tumor immune infiltrates (TILs), especially with dendritic cells, CD8 + T cells or neutrophil. LAMA4 and LAMB1 might be associated with tumor purity in OC. Overall, LAMA5 and LAMC1 could help predict OC survival and diagnosis and might be deemed important OC oncogenes.

Abstract:The poor prognosis of patients with pancreatic ductal adenocarcinoma (PDAC) is partially attributed to the invasive and metastatic behavior of this disease. Laminin subunit beta-3 (LAMB3) encodes one of the three subunits of LM-332, an extracellular matrix protein secreted by cultured human keratinocytes. In addition, LAMB3 is involved in the invasive and metastatic abilities of some types of cancer, including colon, pancreas, lung, cervix, stomach, and prostate cancer, but the role and mechanism of LAMB3 in PDAC have not been previously determined. Herein, we tentatively investigated the role of LAMB3 in the malignant biological behavior of PDAC. In this study, we demonstrated that LAMB3 is upregulated in PDAC. Inhibition of LAMB3 abrogated the tumorigenic outcomes of PI3K/Akt signaling pathway activation, including those involving cell cycle arrest, cell apoptosis, proliferation, invasion and migration in vitro, and tumor growth and liver metastasis in vivo. Our results showed that LAMB3 could mediate cell cycle arrest and apoptosis in PDAC cells and alter the proliferative, invasive, and metastatic behaviors of PDAC by regulating the PI3K/Akt signaling pathway. LAMB3 may be a novel therapeutic target for the treatment of PDAC in the future.

Abstract:Background Recently, increasing evidence has indicated that platelet-activating factor acetylhydrolase 1b catalytic subunit 3 (PAFAH1B3) plays an important role in several cancers. However, its role in lung adenocarcinoma (LUAD) has not been reported until now. Methods The expression of PAFAH1B3 in LUAD was determined by using the Gene Expression Profiling Interactive Analysis (GEPIA) database and real-time PCR (RT–PCR), western blot and immunohistochemical (IHC) analyses. A chi-square test was used to investigate the correlation between PAFAH1B3 expression and clinical parameters. Cox regression and Kaplan–Meier analysis were performed to analyze the prognostic value of PAFAH1B3. The CCK-8 assay, clone formation assay, transwell invasion assay and flow cytometry were conducted to detect cell proliferation, clone formation, invasion and the cell cycle. The xenograft tumor model was constructed to explore the function of PAFAH1B3 in vivo . Western blot and IHC analyses were performed to detect epithelial-to-mesenchymal transition (EMT)-related markers. Immune Cell Abundance Identifier (ImmuneCellAI) and IHC analyses were used to analyze the effect of PAFAH1B3 on immune cell infiltration. Results Our study showed that the expression of PAFAH1B3 was upregulated in LUAD tissues and cells compared with noncancerous tissues and cells. Additionally, the results indicated that the expression of PAFAH1B3 was positively correlated with distant metastasis, TNM stage and poor clinical outcome and it was an independent prognostic risk factor for LUAD. In addition, silencing PAFAH1B3 suppressed cell proliferation, colony formation, and invasion and increased the cell population in the G0-G1 phases in vitro . Furthermore, our results showed that knockdown of PAFAH1B3 increased the epithelial marker E-cadherin level and decreased the mesenchymal marker N-cadherin level in vitro and in vivo . We also proved that PAFAH1B3 downregulation inhibited tumorigenesis and neutrophil infiltration in the xenograft tumor model. Conclusion Our studies indicate that PAFAH1B3, a prognostic risk factor, promotes proliferation, invasion and EMT and affects immune infiltrates in LUAD.

Abstract:We designed this study to identify potential diagnostic and prognostic markers for lung adenocarcinoma (LUAD). Four gene expression profiles were downloaded from GEO and merged into a training cohort, and those genes that were differentially expressed between LUAD and normal samples were selected. We performed LASSO regression, SVM-RFE, and ROC curve analyses, and external validations were conducted using the GSE115002 dataset, TCGA + GTEx datasets, and tissue microarrays (TMAs) of 56 patients with LUAD from our hospital. Subsequent analyses included survival analyses, Cox regression, CIBERSORT, CCK-8, colony formation, and flow cytometry analyses. We observed that the expression of SH3GL2, CAV1, and GPC3 was reduced in LUAD samples in comparison to normal samples. Therefore, these genes were identified as diagnostic markers for LUAD, and validated using the GSE115002 and TCGA + GTEx datasets (AUC > 0.86 for all three). Notably, lower GPC3 expression levels were associated with worse prognoses. And GPC3 is an independent prognostic factor for LUAD; TMA analysis confirmed low GPC3 expression in tumors. Moreover, GPC3 overexpression inhibited cell viability and clonality but not cell apoptosis. In conclusion, we infer that GPC3 could serve as both a diagnostic and prognostic marker for LUAD.

Abstract:Background and Objectives: The present study evaluated the potential role of lysophosphatidic acid receptors (LPARS) in processes leading to local invasiveness and metastasis in Chinese pancreatic carcinoma. Methods: Real-time reverse-transcriptase polymerase chain reaction and Western blot analysis were used to detect expression of LPARS in tumour and adjacent non-tumour tissues from patients with surgically resected pancreatic carcinoma. Surgical specimens from 50 patients were examined for relative expression of each receptor’s messenger RNA (mRNA) and protein. Findings were analyzed for correlations with tumour size, pathologic classification, clinical stage, and infiltration of capsule and lymphonodi. Results: Increased levels of mrna of LPARS (LPAR1 ≈ LPAR3 < LPAR2) were found in the pancreatic cancer tissues examined. Low levels of transcripts for LPAR1, LPAR2, and LPAR3 receptors were detectable in adjacent non-tumour tissues. The difference in LPAR1 protein expression between tumour and adjacent non-tumour tissues does not seem significant, but the signals of LPAR2 expression in pancreatic cancer tumour tissues were significantly amplified compared with those in adjacent non-tumour tissues. Tumour and adjacent non-tumour tissues both weakly expressed LPAR3 protein with no statistical difference. However, expression of LPAR1, LPAR2, and LPAR3 showed an obvious correlation with infiltration of capsule cells, surrounding lymphonodi, and specific histopathologic features. Conclusions: Lysophosphatidic acid receptor is a promising indicator for pancreatic cancer, and our findings suggested that LPAR2 might be a potential target for clinical treatment of pancreatic cancer.

Abstract:AIMS:Recent studies showed that muscarinic acetylcholine receptor 3 (M3), as a muscarinic acetylcholine receptor family member that plays an important role in normal physiological function, is engaged in cancer progression. However, the role of M3 in pancreatic ductal adenocarcinoma (PDAC) is not known. The aim of this study is to investigate the expression and prognostic value of M3 in patients with PDAC.MATERIALS AND METHODS:The localization and expression of M3 in PDAC were examined by immunohistochemistry. VAChT was employed to detect parasympathetic nerve fibers in the corresponding M3 PDAC tissues. The correlation between M3 expression and patients' survival was assessed by Kaplan-Meier analysis.RESULTS:M3 was discovered predominantly localized in the cell cytoplasm and expressed in all specimens of PDAC patients. Significant correlation was noted between increased M3 intensity and high grade of PDAC (P<0.01), more lymph node metastasis (P<0.01) as well as shorter patient overall survival (P<0.01). Morphologically, cells with high M3 expression were more frequently located at the invasive tumor front/tumor budding cells, metastatic lymph nodes and parasympathetic nerve fibers.CONCLUSION:High expression of M3 is a prognostic marker for PDAC.

Abstract:Pancreatic cancers carry a poor prognosis and are the 4th leading cause of cancer deaths in the United States. At time of diagnosis these cancers may already have lymph node involvement or metastatic spread, precluding the possibility of curative surgery and ensuring poorer prognosis. The 5-year survival rate of resectable pancreatic cancer is approximately 20% compared to less than 5% in unresectable cancers. L1 cell adhesion molecule (L1CAM) is a transmembrane glycoprotein that is not expressed in normal pancreas, but its expression in surgically-resected pancreatic cancers has been shown to correlate with prognosis, lymph node involvement, and metastatic disease. Prognostic markers of pancreatic cancer found before surgical resection with endoscopic ultrasound-guided-fine -needle aspiration (EUS-FNA) may guide patient treatment. This study aims to explore the clinical significance of L1CAM in pancreatic cancers diagnosed by EUS-FNA.
