Abstract:Two-stage fermentation can reduce the antinutritional factors (ANFs) in corn-soybean meal (SBM) mixture that interfere with the absorption of nutrients, as well as allergenic proteins that cause diarrhea, but the role of the microbial community in hypoallergenicity and digestibility during the fermentation process remains unclear. Therefore, this study optimized the process of inoculating Bacillus subtilis NCUBSL003 and Lactobacillus acidophilus NCUA06501, and the dynamic changes in physicochemical property and microbial community during two-stage fermentation were investigated. Results showed that the contents of trypsin inhibitor (TI), β-conglycinin and glycinin decreased by 87.28%, 69.02% and 77.72%, respectively, at the end of fermentation. Besides, the trichloroacetic acid soluble protein (TCA-SP), small peptides (SP) and free amino acid (FAA) increased by 6.16, 7.09 and 5.80 folds. Bacillus and Lactobacillus were the main genera during the fermentation process according to results of high-throughput sequencing technology. Notably, according to the results of microbial succession, pathogenic bacteria genera such as Enterococcus , Cronobacter , and Haemophilus parasuis were inhibited effectively which may be due to the lower pH value (4.22) caused by lactic acid (44.85 mg/g). Moreover, the bidirectional orthogonal partial least squares (O2PLS) model analysis indicated the two-stage fermentation could influence ANFs, allergenic protein, pH, SP, and FAA of the corn-SBM mixture effectively. This study suggests that two-stage fermentation can improve the nutrient values and safety of the corn-SBM mixture, and these can contribute to the quality control of plant-based fermented products.
Variations of Soybean Meal and Corn Mixed Substrates in Physicochemical Characteristics and Microbiota During Two-Stage Solid-State Fermentation

Abstract:Corn germ meal (CGM) and corn gluten feed (CGF) are the two main corn byproducts (CBs) obtained from corn starch extraction. Due to their high fiber content, low protein content, and severe imbalance of amino acid, CBs are unable to be fully utilized by animals. In this study, the effect of microorganism, proteases, temperature, solid–liquid ratio, and time on nutritional properties of CB mixture feed (CMF) was investigated with the single-factor method and the response surface method to improve the nutritional quality and utilization of CBs. Fermentation with Pichia kudriavzevii , Lactobacillus plantarum , and neutral protease notably improved the nutritional properties of CMF under the fermentation conditions of 37°C, solid–liquid ratio (1.2:1 g/ml), and 72 h. After two-stage solid-stage fermentation, the crude protein (CP) and trichloroacetic acid-soluble protein (TCA-SP) in fermented CMF (FCMF) were increased ( p < 0.05) by 14.28% and 25.53%, respectively. The in vitro digestibility of CP and total amino acids of FCMF were significantly improved to 78.53% and 74.94%, respectively. In addition, fermentation degraded fiber and provided more organic acids in the CMF. Multiple physicochemical analyses combined with high-throughput sequencing were performed to reveal the dynamic changes that occur during a two-stage solid-state fermentation process. Generally, Ascomycota became the predominant members of the community of the first-stage of fermentation, and after 36 h of anaerobic fermentation, Paenibacillus spp., Pantoea spp., and Lactobacillales were predominant. All of these processes increased the bacterial abundance and lactic acid content ( p < 0.00). Our results suggest that two-stage solid-state fermentation with Pichia kudriavzevii , Lactobacillus plantarum , and protease can efficiently improve protein quality and nutrient utilization of CMF.

Abstract:BackgroundCorn and soybean meal (SBM) are two of the most common feed ingredients used in pig feeds. However, a variety of antinutritional factors (ANFs) present in corn and SBM can interfere with the bioavailability of nutrients and have negative health effects on the pigs. In the present study, two-stage fermentation using Bacillus subtilis followed by Enterococcus faecium was carried out to degrade ANFs and improve the nutritional quality of corn and SBM mixed feed. Furthermore, the microbial composition and in vitro nutrient digestibility of inoculated mixed feed were determined and compared those of the uninoculated controls.ResultsDuring the fermentation process, B. subtilis and lactic acid bacteria (LAB) were the main dominant bacteria in the solid-state fermented inoculated feed, and fermentation produced a large amount of lactic acid (170 mmoL/kg), which resulted in a lower pH (5.0 vs. 6.4) than the fermented uninoculated feed. The amounts of soybean antigenic proteins (β-conglycinin and glycinin) in mixed feed were significantly decreased after first-stage fermentation with B. subtilis. Inoculated mixed feed following two-stage fermentation contained greater concentratioin of crude protein (CP), ash and total phosphorus (P) compared to uninoculated feed, whereas the concentrations of neutral detergent fiber (NDF), hemicellulose and phytate P in fermendted inoculated feed declined (P < 0.05) by 38%, 53%, and 46%, respectively. Notably, the content of trichloroacetic acid soluble protein (TCA-SP), particularly that of small peptides and free amino acids (AA), increased 6.5 fold following two-stage fermentation. There was no difference in the total AA content between fermented inoculated and uninoculated feed. However, aromatic AAs (Phe and Tyr) and Lys in inoculated feed increased, and some polar AAs, including Arg, Asp, and Glu, decreased compared with the uninoculated feed. In vitro dry matter and CP digestibility of inoculated feed improved (P < 0.05) compared with the uninoculated feed.ConclusionsOur results suggest that two-stage fermentation using B. subtilis followed by E. faecium is an effective approach to improve the quality of corn-soybean meal mixed feed.

Abstract:Summary Concentrated monosodium glutamate wastewater (CMGW) generated from the production of monosodium glutamate is an organic wastewater with high concentration of ammonia,chemical oxygen demand, biochemical oxygen demand and SO2-4 and low pH. Discharge of CMGW has raised serious environmental problems,and potential secondary pollution existed even treated with traditional physical and chemical processes. It has already been reported that the richness of nitrogen and carbon makes the recycling of this wastewater possible in the way of microbial fermentation as medium.However,the differences of strain and fermentation purpose require that the medium contains different nutritional compositions with a certain dosage.Bacillus subtilis F-2,isolated from a commercial organic fertilizer,can inhibit the growth of 18 plant pathogenic fungi with varying degrees,especially in F.oxysoporum f.sp niweum with inhibition rate of 93%,which reveal the potential to prepare bio-organic fertilizer under biocontrol.In addition,F-2 also plays an important role in decomposition of organic waste.The study on degradation of food waste has pointed to its dominance in microbial community. Therefore,F-2 as a prospective functional bacteria in agriculture and environment,it is necessary to improve F-2 viable cell production as much as possible.Massive studies were focused on medium optimization of B.subtilis to enhance its cell density in fermentation liquor.However,the main ingredients of medium used in most of these studies were peptone,yeast extracts,beef extracts,soy or corn flour,associated with great cost and excessive consumption of resources. The purpose of this study was to investigate a cost-effective resource utilization of CMGW by increasing the density of B. subtilis F-2 viable cells in cultures. The CMGW medium which was 80-fold diluent of the wastewater,i.e.,12.5 g/L CMGW,was served as the main carbon and nitrogen sources of strain F-2.Contrast experiments of shaking cultures,orthogonal and single factor experiments were applied to find the optimal CMGW medium formula and its culturing condition. The contrast experiments showed that the D(600 nm) value of F-2 suspension and viable cell density of the CMGW medium was significantly higher than that of nutrient broth(NB) medium,and the amino acid content in the CMGW medium decreased significantly after culturing.The optimal formula identifying F-2 by the orthogonal experiment of L1 6(4 3 ×2 6) was as follows:12.5 g/L CMGW,1.0 g/L beef extracts,4.0 g/L peptone,0.5 g/L MnSO4 ·2 H 2 O,0.02 g/L H 3 BO3 ,0.1 g/L FeSO4 ·7 H 2 O and 0.5 g/L MgSO4 ·7 H 2 O.The viable cell density in the optimized CMGW medium was 2.9 and 6.3 times of the CMGW medium and NB medium,respectively. Furthermore,according to the results of single factor experiment,the proper initial pH and culturing temperature of the optimized CMGW medium were screened as pH 6.5 7.5 and 30 35 ℃. In conclusion,the CMGW medium is an ideal substitute for NB medium in the fermentation of B.subtilis F-2,providing a practicable and promising approach to reuse and recover CMGW.Moreover,the CMGW medium is superior to NB medium,and the fermentation effect is even better after the medium and culturing condition are optimized.
[Effects of Panax notoginseng saponins on long-term potentiation in the CA1 region of the rat hippocampus].

Abstract:Panax notoginseng saponins (PNS) are very important extracts from roots of medicinal herb Sanchi Ginseng which is highly regarded in China for its therapeutic ability to meliorate blood-circulation, anti-anoxia, improve memory, and anti-caducity effects. In this study, we used blind whole-cell voltage-clamp recordings to detect the effects of PNS on long-term potentiation (LTP) in the CA1 region of the hippocampus, and investigated the electrophysiological mechanisms underlying potentiating effects of PNS on learning and memory. Wistar rats (3-4 weeks) were decapitated and hippocampal slices (400 microm thick) were cut coronally. Excitatory postsynaptic currents (EPSCs) were recorded by patch clamp technique in whole-cell configuration. The Schaffer collateral/commissural pathway was stimulated by high frequency stimulation (HFS: 100 Hz) pulses to induce LTP. The findings showed that 0.1 - 0.4 g x L(-1) PNS significantly depressed the amplitude of EPSCs (P < 0.05) and had no facilitative effects on LTP of pyramidal neurons located in the CA1 region. PNS in the concentrations of 0.04 - 0.05 g x L(-1) did not appreciably affect the amplitude of EPSCs (P > 0.05) but markedly increased the amplitude of LTP (P < 0.05). In conclusion, 0.04 - 0.05 g x L(-1) PNS could facilitate LTP in the CA1 region of the rat hippocampus and it is reasonable to suggest that this action may contribute to its potentiating effects on learning and memory.
Improving Nutrition Value of Maize-Soybean Meal Based Diet by Microbial Fermentation Combinedwith Multiple-Enzymes

Abstract:The objective of this study was to improve the nutritional value of a corn-soybean meal mix feed by biological fermentation.Bacillus subtilis,Lactobacillus plantarnm,Saccharomyces cerevisiae and several lactic acid bacteria were screened for ferment mixed feed respectively.Compound enzyme preparation was added with fermented strains to decrease the anti-nutritional factors and increase the lactic acid during fermentation.The nutrition value of fermented feed was evaluated by in vitro digestibility of amino acids and animal experiment.The results indicated that:1) Bacillus subtilis BS12 has the greater ability of degrading anti-nutritional factors.Enterococcus faecalis EF can efficiently synthesize lactic acid;2) Mixed fermentation with Bacillus subtilis BS12 and Enterococcus faecalis EF,could get optimal effect by 8∶1 ratio.Soild state fermented corn-soybean meal mixed feed 72 h under 37 ℃ with effective bacteria of 107 CFU/mL,1 × 10％ inoculum size,and 0.50％ compound enzyme preparation,the degradation rate of β-conglycinin could reach to 80.74％ (P ＜ 0.05),glycinin 86.87％ (P ＜ 0.05),neutral detergent fiber 39.36％ (P ＜0.05),Lactic acid content reached 122.57 mmol/kg and pH decreased to 5.0.The vitro digestion analysis suggested that crude protein digestibility increased from 78.46％ to 85.77％,essential amino acid methionine and phenylalanine digestibility increased(P ＜0.05) by 15.15％ and 17.60％,respectively,non-essential amino acid alanine,cysteine and tryptophan increased (P ＜ 0.05) by 10.11％,10.05％ and 9.89％,respectively;3) The average daily gain and feed intake of piglets were increased by substituting fermented feed for 10％ unfermented feed.In conclusion,fermented feed has a good prospect in improving the nutrient digestibility of feed and decreasing antibiotics in animal production.
Optimization of Mixed Solid-state Fermentation of Soybean Meal by Lactobacillus Species and Clostridium butyricum

Abstract:Abstract Soybean meal is the main vegetable protein source in animal feed. Soybean meal contains several anti-nutritional factors, which directly affect digestion and absorption of soy protein, thereby reducing growth performance and value in animals. Fermented soybean meal is rich in probiotics and functional metabolites, which facilitates soybean protein digestion, absorption and utilization in piglets. However, the mixed solid-state fermentation (SSF) conditions of soybean meal remain to be optimized. In this study, we investigated the optimal parameters for SSF of soybean meal by Lactobacillus species and Clostridium butyricum . The results showed that two days of fermentation was sufficient to increase the viable count of bacteria, lactic acid levels and degradation of soybean protein in fermented soybean meal at the initial moisture content of 50%. The pH value, lowering sugar content and oligosaccharides in fermented soybean meal, was significantly reduced at the initial moisture content of 50% after two days of fermentation. Furthermore, the exogenous proteases used in combination with probiotics supplementation were further able to enhance the viable count of bacteria, degradation of soybean protein and lactic acid level in the fermented soybean meal. In addition, the pH value and sugar content in fermented soybean meal were considerably reduced in the presence of both proteases and probiotics. Furthermore, the fermented soybean meal also showed antibacterial activity against Staphylococcus aureus and Escherichia coli . These results together suggest that supplementation of both proteases and probiotics in SSF improves the nutritional value of fermented soybean meal and this is suitable as a protein source in animal feed.

Abstract:Substantial annual economic loss in livestock production is caused by antinutritional factors in soybean meal and corn mixed substrates, which can be degraded by microbial fermentation. Although considerable efforts have been made to explain the effects of fermentation on soybean meal and corn-based feed, the dynamics of the physicochemical characteristics, microbiota, and metabolic functions of soybean meal and corn mixed substrates during solid-state fermentation remain unclear. Here, multiple physicochemical analyses combined with high-throughput sequencing were performed to reveal the dynamic changes that occur during a novel two-stage solid-state fermentation process. Generally, inoculated bacteria rapidly proliferated in the initial 12-h aerobic fermentation (P = 0.002). Notably, most nutritional changes occurred during 12 to 24 h compared to 0 to 12 h. Second-stage anaerobic fermentation increased the bacterial abundance and lactic acid content (P < 0.00). Bacillus spp., Enterococcus spp., and Pseudomonas spp. were predominantly involved in the maturation of the fermented mixed substrates (P < 0.05). Additionally, the available phosphorus exhibited the greatest interaction with the microbial community structure. Cellular processes and environmental information processing might be the main metabolic processes of the microbiota during this fermentation. An in vivo model further evaluated the growth-promoting effects of the fermented products. These results characterized the dynamic changes that occur during two-stage solid-state fermentation and provided potential references for additional interventions to further improve the effectiveness and efficiency of solid-state fermentation of feed. IMPORTANCE Solid-state fermentation (SSF) plays pivotal roles not only in human food but also farm animal diets. Soybean meal (SBM) and corn account for approximately 70% of the global feed consumption. However, the nutritional value of conventional SBM and corn mixed substrates (MS) is limited by antinutritional factors, causing substantial economic loss in livestock production. Although emerging studies have reported that SSF can improve the nutritional value of SBM-based substrates, the dynamic changes in the physicochemical features, microbiota, and metabolic functions of MS during SSF remain poorly understood, limiting further investigation. To provide insights into the dynamics of the physicochemical characteristics and the complex microbiome during the two-stage SSF of MS, multiple physicochemical analyses combined with high-throughput sequencing were applied here. These novel insights shed light on the complex changes that occur in the nutrition and microbiome during two-stage SSF of MS and are of great value for industrial feed-based practices and metabolomic research on SSF ecosystems.

Abstract:Corn gluten meal-corn husk mixes (CCM) are an inexpensive and readily available agricultural by-product. This study explores a novel technique by converting CCM into high-value livestock feed protein sources through fermentation with Aspergillus niger AAX and Lactobacillus fermentum LLS, aiming to sustainably meet future global protein needs. The process of fermentation significantly altered the structural composition of high molecular weight proteins, zein, and dietary fibers. This transformation resulted in a marked elevation in the concentrations of peptides, free amino acids, and polyphenols. The acidic environment produced during fermentation prevented lipid oxidation in CCM, thereby extending its storability. After fermentation, the content of anti-nutritional factors decreased, while its antioxidant capacity increased. In vitro simulated digestion suggested that fermentation improved the digestibility of CCM protein. In vivo animal experiments showed that fermented CCM (FCCM) promoted growth and gut health in chicks. This study provides new insights into the utilization of CCM.

Abstract:BACKGROUND Solid state fermentation (SSF) is a general approach for preparing food and feed, which not only improves nutrition, but also provides prebiotics and metabolites. Although many studies have been conducted on the effects of fermentation on feed substrate, the dynamics of microbiota and metabolites in SSF remain unclear. Here, high-throughput sequencing combined with GC-QTOF-MS technology was used to evaluate the dynamic changes of solid fermented soybean meal and corn mixed matrix inoculated with Bacillus pumilus and Limosilactobacillus fermentum. RESULTS Generally, inoculated bacteria rapidly proliferated, accompanied by the degradation of macromolecular proteins and an increase in the content of SP, TCA-SP, FAA and organic acids. Bacillus, Lactobacillus and Enterococcus dominated the whole fermentation process. 389 non-volatile metabolites and 182 volatile metabolites were identified, including amino acids, organic acids, ketones, aldehydes, furans and pyrazine. Typical non-volatile metabolites such as lactic acid, 4-aminobutanoic acid, L-Glutamic acid, D-Arabinose and volatile metabolites such as 4-ethyl-2-methoxyphenol, 4-Penten-2-ol, 2-Pentanone, 2-ethylfuran, 2-methylhexanoic acid, butanoic acid-ethyl ester was significantly increased in two-stage solid fermentation. However, some adverse metabolites were also produced, such as oxalic acid, acetic acid, tyramine and n-butylamine, which may affect the quality of fermented feed. 16 genera were significantly correlated with differential non-volatile metabolites, while 11 genera were significantly correlated with differential volatile metabolites. CONCLUSION These results characterized the dynamic changes in the process of two-stage solid-state fermentation with Bacillus pumilus and Limosilactobacillus fermentum and provided potential reference for additional intervention on improving the effectiveness and efficiency of solid-state fermentation of feed in furture. Abstract:Corn germ meal (CGM) is one of the major byproducts of corn starch extraction. Although CGM has rich fiber content, it lacks good protein content and amino acid balance, and therefore cannot be fully utilized as animal feed. In this study, we investigated the processing effect of cellulase synergized with Bacillus velezensis on the nutritional value of pretreated CGM (PCGM) in two-stage solid-state fermentation (SSF). High-throughput sequencing technology was used to explore the dynamic changes in microbial diversity. The results showed that compared with four combinations of B. velezensis + Lactiplantibacillus plantarum (PCGM-BL), cellulase + L. plantarum (PCGM-CL),control group (PCGM-CK), and cellulase + B. velezensis + L. plantarum (PCGM-BCL), the fourth combination of PCGM-BCL significantly improved the nutritional characteristics of PCGM. After two-stage SSF (48 h), viable bacterial count and contents of crude protein (CP) and trichloroacetic acid-soluble protein (TCA-SP) all were increased in PCGM-BCL (p < 0.05), while the pH was reduced to 4.38 ± 0.02. In addition, compared with PCGM-BL, the cellulose degradation rate increased from 5.02 to 50.74%, increasing the amounts of short-chain fatty acids (216.61 ± 2.74 to 1727.55 ± 23.00 μg/g) and total amino acids (18.60 to 21.02%) in PCGM-BCL. Furthermore, high-throughput sequencing analysis revealed significant dynamic changes in microbial diversity. In the first stage of PCGM-BCL fermentation, Bacillus was the dominant genus (99.87%), which after 24 h of anaerobic fermentation changed to lactobacillus (37.45%). Kyoto Encylopaedia of Genes and Genomes (KEGG) metabolic pathway analysis revealed that the pathways related to the metabolism of carbohydrates, amino acids, cofactors, and vitamins accounted for more than 10% of the enriched pathways throughout the fermentation period. Concisely, we show that cellulase can effectively improve the nutritional value of PCGM when synergized with B. velezensis in two-stage SSF.

Abstract:In the present study, a 2-stage solid-state fermentation process using followed by was performed with the purpose of improving the nutritional quality of corn and soybean meal (SBM) mixed feed (MF). The matrix of native MF and fermented MF (FMF) was analyzed using scanning electron microscopy to determine external structures after solid-state fermentation. Results of the present study indicated that the FMF had a greater concentration of CP and total P compared with the unfermented feed, whereas the contents of β-conglycinin, glycinin, phytate P, raffinose, stachyose, and NDF in FMF declined by 77.6, 86.7, 41.2, 47.2, 36.6, and 38.9%, respectively. The content of trichloroacetic acid-soluble protein, particularly those of small peptides and free AA, increased more than 3 times after fermentation. Scanning electron microscopy revealed that irregular shapes and a lot of porous structures were observed in the SBM of FMF, the cell wall of corn in FMF was destroyed, and the number of starch granules was decreased after 2-stage fermentation. Two animal experiments were conducted to evaluate the digestibility of CP and P in FMF when fed to pigs. In Exp. 1, 6 barrows (15.20 ± 1.27 kg initial BW) fitted with a T-cannula in the distal ileum were allotted to a replicated 3 × 3 Latin square with 3 diets and 3 periods per square. Two diets containing unfermented MF or FMF as the only source of protein and a N-free diet were formulated. Results indicated that the apparent ileal digestibility (AID) of total AA and Lys in FMF (74.98 and 75.34%) were greater ( < 0.05) than in untreated MF (69.66 and 68.56%). The standard ileal digestibility (SID) of Lys in FMF (82.17%) was also greater ( < 0.05) than in MF (75.91%). In Exp. 2, 16 barrows (17.57 ± 1.08 kg initial BW) were allotted to 2 diets formulated to contain MF or FMF as the sole source of P to determine the digestibility of P. The apparent total tract digestibility and standardized total tract digestibility of P in FMF were 58.14 and 64.72%, respectively. These values were greater ( < 0.01) than in untreated MF (37.11 and 44.89%, respectively). Our results indicate that a solid-state fermentation process using followed by offers an effective approach to improving the quality of corn and SBM MF.

Abstract:The study evaluated the impact of fermentation with Bacillus sp. on the nutritional quality of soybean meal (SBM) and the changes of bacterial community structure during fermentation. High protease-producing strains were screened to degrade SBM macromolecular protein and anti-nutritional factors (ANFs). Unsterilized SBM then underwent an anaerobic solid-state fermentation method to evaluate the effects of fermentation. Results showed that for the nine high-producing protease strains that were screened, acid-soluble protein (ASP) contents in fermented SBM increased, with the highest value found to be 13.48%, which was fermented using strain N-11. N-11 was identified as Bacillus subtilis . N-11 fermentation reduced ANFs such as glycinin and β-conglycinin by 82.38 and 88.32%, respectively. During N-11 fermentation, the bacterial richness and diversity in SBM increased but not significantly. The high-yield protease strain B. subtilis N-11 selected in this experiment improved the nutritional quality of SBM through fermentation, and it can be used for industrial large-scale production.

Abstract:ABSTRACT Soybean meal (SBM) is the most important source of plant protein in animal feeds, containing around 41%–48% crude protein. Nevertheless, 70%–80% of these proteins is allergenic antigens that can have adverse implications for the gastrointestinal well-being of animals, especially to young animals. Microbial fermentation is one of the most cost-effective strategies used to reduce allergenic antigens from plant sources. In this study, we report the isolation and characterization of a novel probiotic Bacillus subtilis “L5” strain from lake mud. L5 demonstrated remarkable temperature tolerance across a broad temperature spectrum, thriving at 25°C, 37°C, and 50°C. In addition, antimicrobial assay revealed that L5 exhibits strong antimicrobial activity against Escherichia coli , effectively reducing or eliminating the growth of Gram-negative bacteria in SBM when fermented with L5. When applied to SBM fermentation, L5 efficiently reduced SBM antinutritional factors such as glycinin, β-conglycinin, trypsin inhibitor, phytic acid, neutral detergent fiber, and acid detergent fiber, which in turn results in an increase in crude protein content and the free amino acid concentration. Our findings on the probiotic and fermentation capabilities of L5 suggest that this novel bacterium has dual functions that make it a strong candidate for improving the nutrient values of feed via its role in fermentation. IMPORTANCE Soybean meal (SBM), containing 41%–48% crude protein, is the most important source of plant protein in animal feeds. Unfortunately, 70%–80% of the proteins in SBM is allergenic antigens including trypsin inhibition, β-conglycinin, and conglycinin, which negatively affect intestine health and function. Microbial solid-state fermentation methods have been applied to animal feeds for decades, to eliminate antinutritional factors. Here, a novel potential probiotic Bacillus subtilis “L5” strain with high enzymatic activity and antimicrobial activity will be a great help to improve the quality and reproducibility of SBM fermentation.

Abstract:BACKGROUD As feeding probiotics, Bacillus subtilis FJAT-4842 and Lactobacillus plantarum FJAT-13737 were employed to improve the nutritional value and reduce the risk of contamination in the solid-state fermentation (SSF) of soybean meal (SBM). RESULT After fermentation with bacterium starters, the crude protein, free amino acid and lactic acid increased, with higher protease and cellulose activity. The addition of Lactobacillus plantarum could increase the crude protein and lactic acid content by 50.1% and 94.9% respectively. After fermentation, the contents of crude fiber and phytic acid decreased by 45.9% and 48.1%. Compared with control treatment, the addition of both B. subtilis FJAT-4842 and L. plantarum FJAT-13737 could effectively elevate the production of FAA and ester. Moreover, the addition of bacterium starter could prevent the production of mycotoxin. Moreover, addition of bacterium starters promoted bacterial diversity of the fermented SBM. Especially, the addition of B. subtilis could reduce the relative content of Staphylococcus. After fermentation of 7 days, lactic acid bacteria including Pediococcus, Weissella and Lactobacillus became the main bacteria in the fermented SBM. CONCLUSION The addition of bacterium starter is benefit to improve the nutritional value and reduce the risk of contamination in the solid state fermentation of soybean. Abstract:The aim of this article is to increase the nutritional value of soybean meal while reducing the content of antinutrients by microbial fermentation of soybean meal with a mixed culture of probiotic microorganisms (Bacillus subtilis, Aspergillus niger, Saccharomyces cerevisiae, Lactiplantibacillus plantarum) at two different hydromodules. The addition of microorganisms increased the content of easily digestible protein and amine nitrogen in fermented soybean meal (30:110 and 30:130, hydromodulus soybean meal:water) while decreasing urease activity (hydromodulus 30:110). The positive effect of microbial fermentation on the mineral composition of soybean meal was demonstrated. The ability of microorganisms in the fermentation process to increase the content of protein, essential amino acids, and macro- and microelements in soybean meal while decreasing anti-nutritional factors opens up possibilities for using this technology to advance animal husbandry.

Integrated Microbiota and Metabolome Analysis to Assess the Effects of the Solid-State Fermentation of Corn–Soybean Meal Feed Using Compound Strains

Abstract:Solid-state fermentation is known to improve plant-based feed nutritional quality; however, the association between microbes and metabolite production in fermented feed remains unclear. We inoculated corn–soybean–wheat bran (CSW) meal feed with Bacillus licheniformis Y5-39, Bacillus subtilis B-1, and lactic acid bacteria RSG-1. Then, 16S rDNA sequencing and untargeted metabolomic profiling were applied to investigate changes in the microflora and metabolites, respectively, and their integrated correlations during fermentation were assessed. The results indicated that trichloroacetic acid soluble protein levels showed a sharp increase, while glycinin and β-conglycinin levels showed a sharp decrease in the fermented feed, as confirmed by sodium dodecyl sulfate–polyacrylamide gel electrophoresis. Pediococcus, Enterococcus, and Lactobacillus were predominant in the fermented feed. Overall, 699 significantly different metabolites were identified before and after fermentation. Arginine and proline, cysteine and methionine, and phenylalanine and tryptophan metabolism were the key pathways, with arginine and proline metabolism being the most important pathway in the fermentation process. By analyzing the correlation between the microbiota and metabolite production, lysyl–valine and lysyl–proline levels were found to be positively correlated with Enterococcus and Lactobacillus abundance. However, Pediococcus was positively correlated with some metabolites contributing to nutritional status and immune function. According to our data, Pediococcus, Enterococcus, and Lactobacillus mainly participate in protein degradation, amino acid metabolism, and lactic acid production in fermented feed. Our results provide new insights into the dynamic changes in metabolism that occurred during the solid-state fermentation of corn–soybean meal feed using compound strains and should facilitate the optimization of fermentation production efficiency and feed quality.

Abstract:<section class="article-section__content"><h3 class="article-section__sub-title section1"> BACKGROUND</h3><p>Fermentation efficiency of thermophiles of <i>Bacillus licheniformis</i> YYC4 and <i>Geobacillus stearothermophilus</i> A75, and mesophilic <i>Bacillus subtilis</i> 10 160 on soybean meal (SBM) was evaluated by examining the nutritional and protein structural changes. </p></section><section class="article-section__content"><h3 class="article-section__sub-title section1"> RESULTS</h3><p>SBM fermenting by <i>B. licheniformis</i> YYC4, <i>B. subtilis</i> 10 160 and <i>G. stearothemophilus</i> A75 increased significantly the crude and soluble protein from 442.4 to 524.8, 516.1 and 499.9 g kg<sup>−1</sup>, and from 53.9 to 203.3, 291.3 and 74.6 g kg<sup>−1</sup>, and decreased trypsin inhibitor from 8.19 to 3.19, 2.14 and 5.10 mg g<sup>−1</sup>, respectively. <i>B. licheniformis</i> YYC4 and <i>B. subtilis</i> 10 160 significantly increased phenols and pyrazines contents. Furthermore, <i>B. licheniformis</i> YYC4 fermentation could produce abundant alcohols, ketones, esters and acids. Surface hydrophobicity, sulfhydryl groups and disulfide bond contents of SBM protein were increased significantly from 98.27 to 166.13, 173.27, and 150.71, from 3.26 to 4.88, 5.03 and 4.21 μmol g<sup>−1</sup>, and from 20.77 to 27.95, 29.53 and 25.5 μmol g<sup>−1</sup> after their fermentation. Fermentation induced red shifts of the maximum absorption wavelength (λmax) of fluorescence spectra from 353 to 362, 376 and 361 nm, while significantly reduced the fluorescence intensity of protein, especially when <i>B. subtilis</i> 10 160 was used. Moreover, fermentation markedly changed secondary structure composition of SBM protein. SDS-PAGE and AFM analyses showed that macro-molecule protein got degraded into small sized protein or peptide during fermentation of SBM . </p></section><section class="article-section__content"><h3 class="article-section__sub-title section1"> CONCLUSION</h3><p><i>B. licheniformis</i> YYC4 fermentation (without sterilization) improved nutrition and protein structure of SBM as <i>B. subtilis</i> 10 160, suggesting its potential application in SBM fermentation industry. </p><p>This article is protected by copyright. Abstract:The improvement in the utilization rate and nutritional value of soybean meal (SBM) represents a significant challenge in the feed industry. This study conducted a 50 kg SBM fermentation based on the 300 g small-scale fermentation of SBM in early laboratory research, to explore the combined effects of lactic acid bacteria (LAB) and acid protease on fermentation quality, chemical composition, microbial population, and macromolecular protein degradation during fermentation and aerobic exposure of SBM in simulated actual production. The results demonstrated that the increase in crude protein content and reduction in crude fiber content were considerably more pronounced after fermentation for 30 days (d) and subsequent aerobic exposure, compared to 3 d. It is also noteworthy that the treated group exhibited a greater degree of macromolecular protein degradation relative to the control and 30 d of fermentation relative to 3 d. Furthermore, after 30 d of fermentation, adding LAB and protease significantly inhibited the growth of undesired microbes including coliform bacteria and aerobic bacteria. In the mixed group, the microbial diversity decreased significantly, and Firmicutes replaced Cyanobacteria for bacteria in both groups’ fermentation.
