Abstract:The title compound, C30H28N6O6S4 center dot CHCl3, is a cytotoxic and antibacterial compound which was isolated from ethyl acetate extracts of Amanita flavorubescens Alk. affected by Verticillium sp. The molecule is a dimer with two epidithio-dioxopiperazine nuclei, the two halves being related by an approximate twofold axis. The two five-membered rings are cis-fused. The crystal structure is stablized by a hydrogen-bond network involving both OH groups and the carbonyl group.

Abstract:Alstoscholarisine K, an indole alkaloid with eight chiral carbons and featuring a novel 6/5/6/6/6/6/6/5 octacyclic architecture, was found to be specific to the gall-infected leaves of Alstonia scholaris. Its structure was elucidated by spectroscopy, computational analysis, and single-crystal X-ray diffraction. The unusual highly fused cage-like pyrrolo[1,2-a]pyrimidine structure with an additional −C4N unit is possibly derived from a combination of monoterpenoid indole and polyamine pathways. Abstract:Phytochemical investigation on the stems of Alstonia scholaris (L.) R. Br. led to the isolation and identification of 30 natural products, including sixteen alkaloids (1–16), nine triterpenoids (17–25), two phenols (26–27), and three lignans (28–30). Their structures were established by comprehensive spectroscopic analyses together with the comparison with reported data. Compound 1 was a new indole alkaloid. Compound 24 was firstly isolated from Apocynaceae family, compounds 22–23 and 26–30 were obtained from the genus Alstonia for the first time, and this is the first report of the presence of 11 and 16 in A. scholaris. The chemotaxonomic significance of these isolated compounds was also discussed.

Abstract:A new 2-quinolone alkaloid, iso-oligophyline (1), and two very unusual C34 terpenoids, proposed names ravespanol (2) and ravespanone (3), along with two known compounds, β-sitosterol (4), and methyl linoleate (5), were isolated from the leaf extract of Ravenia spectabilis engl. Methyl linoleate constitutes the first report of isolation from this species. We have already reported the isolation of atanine (6), oligophyline (7), ravenoline (8), and arborinine (9) from the plant. Based on nuclear magnetic resonance (NMR) spectroscopy and mass spectrometric analysis, the structure of the isolated chemicals was determined. The crude fractions and four compounds (6,7,8 and 9) were evaluated for a cytotoxicity study on a panel of six human stomach cancer cell lines (SCL, SCL-6, SCL-37'6, SCL-9, K-3, N21) by MTT assay. Among the plant extracts and isolated compounds, petroleum ether fraction and compound 7 exhibited the highest cytotoxic activity against SCL and SCL-6 cells, where the IC50 values were 17.9 and 16.56 μM, respectively.

Abstract:2‐hydroxy-1-naphthaldehyde and 3,4-diaminotoluene were reacted with bis(acetylacetonato) oxovanadium to yield a novel NNOO -type chelate complex, VOL. The prepared complex is characterized by employing elemental analysis, electronic and infrared spectra. The expected structure was crystallized and identified by using the single-crystal X-ray diffraction method. The VOL complex crystallizes in the monoclinic system with the P2 1 /c space group. Additionally, we affected a comprehensive analysis of VO V , VO IV and VO III complexes focusing on their structural, electronic, and optical properties using DFT and TD-DFT calculations in gas. The computed parameters closely matched the experimental data, demonstrating good agreement. The electrochemical behavior of the VOL complex, investigated on a glassy carbon electrode (GCE) by means of cyclic voltammetry (CV), revealing a metal-centered one-electron redox systems corresponding to the VO IV /VO III and VO V /VO IV at -1206 and 547 mV/SCE respectively. The in vitro antibacterial activities of the free ligand H 2 L and the corresponding VOL complex were evaluated against Gram-positive strains, Staphylococcus aureus and Streptococcus pyogenes , and Gram-negative strains, Escherichia coli, Proteus mirabilis and Klebseilla pneumoniae . The biological screening revealed that the VOL complex exhibits more potent activity than the free ligand.

Abstract:Two compounds (1 and 2) have been isolated from the stem bark of Dysoxylum binectariferum (Roxb.) Hook. f. ex Bedd. Compound 1 is rohitukine [5,7-dihydroxy-2-methyl-8-[4-(3-hydroxy-1- methyl)-piperidinyl]-4H-1-benzopyran-4-one], a known compound that has anti-inflammatory and immunomodulatory activities. Compound 2 is a new alkaloid, named rohitukine N-oxide, the structure of which was elucidated on the basis of spectral evidences and comparison with the 1H NMR data of rohitukine.

Abstract:In this work, we synthesized and confirmed the structure of several alkaloid N-oxides using mass spectrometry and Fourier-transform infrared spectroscopy. We also investigated their reduction mechanisms using voltammetry. For the first time, we obtained alkaloid N-oxides using an oxidation reaction with potassium peroxymonosulfate as an oxidant. The structure was established based on the obtained fragmentation mass spectra recorded by LC-Q-ToF-MS. In the FT-IR spectra of the alkaloid N-oxides, characteristic signals of N-O group vibrations were recorded (bands in the range of 928 cm−1 to 971 cm−1), confirming the presence of this functional group. Electrochemical reduction studies demonstrated the reduction of alkaloid N-oxides at mercury-based electrodes back to the original form of the alkaloid. For the first time, the products of the electrochemical reduction of alkaloid N-oxides were detected by mass spectrometry. The findings provide insights into the structural characteristics and reduction behaviors of alkaloid N-oxides, offering implications for pharmacological and biochemical applications. This research contributes to a better understanding of alkaloid metabolism and degradation processes, with potential implications for drug development and environmental science.

Abstract:Monomeric alkannin and shikonin (A/S) are potent pharmaceutical substances with a wide spectrum of biological activity and comprise the active ingredients for several pharmaceutical preparations. Therefore, the determination of the impurities, degradation products or byproducts in alkannin and shikonin samples is of great importance. Oligomeric alkannin and shikonin are formed during biosynthesis of these bioactive secondary metabolites in Boraginaceaous root plants, during tissue culture production of A/S, during alkaline hydrolysis of A/S esters and also thermal treatment of A/S. In the present study, a dimeric alkannin/shikonin compound was isolated by size exclusion chromatography from alkannin and shikonin commercial samples and its structure was determined by one- and two-dimensional NMR spectroscopy. The structure of the most abundant oligomeric species in these samples, a dimeric naphthoquinone, was established for the fi rst time, indicating that coupling of the side chain of one naphthoquinone unit with the aromatic ring of a second naphthoquinone leads to dimer formation. This type of coupling allows further oligomerization by leaving one isohexenyl side chain available at the second monomer unit.

Abstract:Two new monoterpenoid indole alkaloids, named alstoscholines A and B (1 and 2), along with three known ones (3–5) were isolated from the stems of Alstonia scholaris. The new structures including their absolute configurations were elucidated on the basis of HRMS and NMR spectroscopic data and computational calculation methods. Compounds 1 and 2 exhibited moderate inhibitory effects on LPS-induced nitric oxide (NO) production in RAW 264.7 macrophages.

Abstract:Five unusual alkaloids featuring a pyrrolo[1,2- a ]quinolone skeleton (pyrroloquinolones B–F, 1 – 5 ) were isolated from the ethanol extract of the whole plant of Vernonia glabra (Steetz) Vatke, along with sixteen known compounds. Their structures were established by means of spectroscopic (1D and 2D NMR, UV, IR, and ECD) and high resolution mass spectrometric techniques as well as by comparison of their spectroscopic data with those reported in the literature. The ethanol extract and some isolated compounds were assessed for their antibacterial activity against four bacterial strains. The extract was significantly active against Staphylococcus aureus ATCC1026 and S. epidermidis ATCC35984 (MIC = 64 μg/mL). All the tested compounds showed moderate activity against S. epidermidis (16 ≤ MIC ≤ 64 μg/mL). Furthermore, this is the first report on tricyclic pyrrolo[1,2- a ]quinolone alkaloids from a plant source. A biosynthetic pathway for the formation of these compounds is also proposed.

Abstract:Veratrazine A (1), a steroidal alkaloid with a unique 6/5/5 triheterocyclic scaffold as the side chain, was isolated from Veratrum stenophyllum, and its structure was established via spectroscopic analyses and X-ray diffraction. Operative Treatment of Nonunion of Surgical Neck Fractures of the Humerus

Abstract:&NA; Seventeen patients who had undergone operative treatment for painful nonunion of a surgical neck fracture of the proximal humerus were evaluated an average of 6.3 years (range, 2‐11 years) from the time of surgery to determine clinical and radiographic outcome. Ten patients underwent open reduction and internal fixation with tension band wiring and Rush rods, and 7 underwent proximal humeral hemiarthroplasty. The average age of the patients was 60 years old. After surgery, pain, function, and range of motion improved for both groups. University of California Los Angeles scores improved from an average of 4.4 to 22 points in the internal fixation group and from 4.8 to 21.4 in the hemiarthroplasty group. All 17 patients were eventually able to perform activities of daily living independently. No patient in either group was able to return to his or her preinjury level of work. Nonunion persisted in 2 patients who had undergone internal fixation, and avascular necrosis developed in 2 others. Eight of 10 patients in the internal fixation group required removal of the Rush rods for treatment of symptomatic mechanical impingement. Painful nonunion of a surgical neck fracture of the proximal humerus is a difficult problem to manage operatively. Pain relief and functional improvement was similar in patients who underwent either procedure. However, 11 of 17 patients had fair or poor ultimate outcomes.

Abstract:An unprecedented new limonoid-based alkaloid, granatoine (1), and a new phragmalin, xylocarpin L (2), along with xyloccensin Y were isolated as minor components from the fruits of the Chinese mangrove plant Xylocarpus granatum. Their structures were elucidated by extensive spectroscopic data (1D and 2D NMR) analysis. A possible biogenetic pathway for granatoine was also depicted.

Abstract:A newstemona alkaloid, tuberostemonone (C22H31NO6), has been isolated from the roots ofstemona tuberosa Lour, and its molecular structure determined by MS, IR, NMR spectra and X-ray crystallography (R=0.046 for 1746 observed Mo-Kα data). Besides retaining the characteristicstemona skeleton, which is composed of anα-methyl-γ-lactone and anN-methylene azepine ring linked at the γ-position of this lactone, the tuberostemonone molecule possesses a nine-membered azacycle which is hitherto not found in other knownstemona alkaloids.

Abstract:Tabernaesine J (1), an unprecedented vincamine‐type indole alkaloid with 6/5/6/6/6/5/5 heptacyclic‐ring system, as well as one new biogenetically related vincamine‐type indole alkaloid (2), were isolated from leaves of Tabernaemontana pachysiphon. Their structures were established by a combination of HRESIMS, NMR, single crystal X‐ray diffraction, and ECD calculation. The unprecedented penta‐lactone ring in tabernaesine J (1) was postulated to be derived from acetyl‐CoA enolate anion. Alkaloid 2 combined with fluconazole has the potential to overcome fluconazole resistance in Candida albicans. Tabernaesine J (1), a novel vincamine‐type indole alkaloid with 6/5/6/6/6/5/5 heptacyclic‐ring system, was isolated and identified from leaves of Tabernaemontana pachysiphon. The unprecedented penta‐lactone ring might be derived from acetyl‐CoA enolate anion from a biogenetic point of view.

Vitiquinolone--a quinolone alkaloid from Hibiscus vitifolius Linn

Abstract:Phytochemical investigations of the powdered root of Hibiscus vitifolius Linn. (Malvaceae) was extracted successively with n-hexane and chloroform. Analysis of the n-hexane extract by GC-MS led to the identification of twenty-six components by comparison of their mass spectra with GC-MS library data. A novel quinolone alkaloid, vitiquinolone (5) together with eight known compounds viz. β-Amyrin acetate (1), n-octacosanol (2), β-Amyrin (3), stigmasterol (4), xanthyletin (6), alloxanthoxyletin (7), xanthoxyletin (8) and betulinic acid (9) were isolated from chloroform extract by column chromatography over silica gel. The structure of vitiquinolone was established on the basis of spectroscopic methods including UV, IR, 1D, 2D NMR and ESI-MS. The known compounds were identified on the basis of their physical and spectroscopic data as reported in the literature.

Abstract:Chemical examination of an arctic actinomycete Streptomyces nitrosporeus resulted in the isolation of two new alkaloids named nitrosporeusines A (1) and B (2), an unprecedented skeleton containing benzenecarbothioc cyclopenta[c]pyrrole-1,3-dione. Their structures were determined through extensive spectroscopic analyses in association with X-ray single crystal diffraction. Both 1 and 2 exhibited inhibitory activities against the H1N1 virus in MDCK cells.
