Abstract:Two new p-methoxyphenyl-type derivatives cytchloramol (1) and cytoxazinanone (2), along with six known compounds (3-8) were identified from the chemical investigations of a saline lake actinomycete, Streptomyces sp. XZB32. The structures of the new compounds were elucidated by extensive NMR spectroscopic analysis, HRESIMS data, GIAO (gauge-including atomic orbitals) NMR, specific optical rotation (SOR) and electronic circular dichroism (ECD) calculations. Cytotoxicity evaluation of the two new compounds showed that compound 1 exhibited significant activity against HCT-116 and MDA-MB-231 human cancer cell line with IC50 values of 2.7 ± 0.07 µM and 1.54 ± 0.14 µM, respectively.

Abstract:Four new C-21 steroidal glycoside, stemucronatosides D (1), E (2), F (3), and G (4) were isolated from the roots of Stephanotis mucronata. Their structures were determined on the basis of chemical evidence and extensive spectroscopic methods including one-dimensional and two-dimensional NMR. These isolated compounds were assayed for their immunological activities in vitro against concanavalin A (Con A)- and lipopolysaccharide (LPS)-induced proliferation of mice splenocytes. Compounds 1, 2, and 4 showed immunosuppressive activities in a dose-dependent manner, while compound 3 showed immunomodulating activities.

Abstract:Strain HM190, a moderate halophile, was isolated from rhizosphere soil of the mangroveKandelia obovatain Fugong village, China. The 16S ribosomal RNA (rRNA) gene sequence and the results of phylogenetic analysis revealed that strain HM190 belonged to the genusStreptomycesand had the highest sequence similarity of 99.79% toStreptomyces heilongjiangensisNEAU-W2(T). The complete genome of strain HM190 comprised 7,762,826 bp in a linear chromosome with 71.97% G + C content. According to antiSMASH analysis, a total of 30 biosynthetic gene clusters (BGCs) were predicted to be involved in secondary metabolism, 12 of which were responsible for the production of polyketide- and non-ribosomal peptide-derived secondary metabolites. Gene cluster 5 was responsible for macrolide biosynthesis in a strain-specific 126,331-bp genomic island belonging to the left-arm region. Combined genomics-metabolomics analysis led to the discovery of three 22-membered macrolides (compounds1-3). Their structures were elucidated by using spectroscopic techniques including high-resolution electrospray ionization mass spectroscopy (HRESIMS) and nuclear magnetic resonance (NMR). The absolute configurations of compounds1-3were determined by the X-ray single crystal diffraction and NMR data analysis. All three compounds displayed moderate cytotoxic activities toward tumor cell lines HepG2, A549, and HCT116.

Abstract:Stephanotis mucronata (Blanco) Merr. has been used for rheumatoid arthritis in Chinese folk herb medicine. Guided by bioactive test, a novel potent immunosuppressive C(21) steroidal glycoside stemucronatoside K (SMK) was isolated from this plant. Its structure was elucidated on the basis of the chemical evidence and extensive spectroscopic methods. We investigated the immunosuppressive effects of SMK in vitro and in vivo. SMK significantly suppressed concanavalin A (Con A)- and lipopolysaccharide (LPS)-stimulated splenocyte proliferation in vitro in a concentration-dependent manner. ICR mice were immunized subcutaneously with OVA on the first day and administered intraperitoneally with SMK at the doses of 2.5, 5, and 10 mg/kg once daily for 10 days. At 24 h after the last administration, mitogen- and OVA-stimulated splenocyte proliferation, the levels of cytokines from splenocytes, and specific antibody titers in serum were measured. SMK significantly inhibited Con A-, LPS- and OVA-induced splenocyte proliferation in the immunized mice in a dose-dependent manner. OVA-specific IgG, IgG1, and IgG2b antibody titers were significantly reduced by SMK compared with the control group. SMK also significantly decreased OVA-induced interleukin-2 (IL-2), interferon-gamma (IFN-gamma), and IL-4 production from splenocytes in the OVA-immunized mice. These results demonstrated that SMK could suppress the cellular and humoral immune response in mice. This study provided evidence to understand the therapeutic effects of S. mucronata and an immunosuppressive natural product compound to further researches to be developed as immunosuppressant.
21,24-Cyclolanostanes revisited: Structural revision and biological evaluation

Abstract:Chemical fractionation of the EtOH extract of a medicinal macro fungus, Inonotus obliquus, afforded an array of lanostane-type triterpenoids (1-11) including two new ones (1 and 8). The structures of these compounds were determined on the basis of spectroscopic analyses, single crystal X-ray crystallography of 3-6 and biosynthetic considerations. With the confirmatory structural information provided by X-ray diffraction analysis in hand, several previously reported 21,24-cyclolanostanes, such as inonotsutriols A-C and (20R,21S,24S)-21,24-cyclopenta-3β,21,25-trihydroxylanosta-8-ene, were structurally corrected. In addition, the NMR data of other types of 21,24-cyclo triterpenoids were also re-examined and structural revisions were thus suggested. Compounds 2, 6 and 8 showed significant cytostatic effects against a panel of tumor cell lines with IC50 values ranging from 7.80 to 18.5 μM. Further assays established that compound 2 exerted promising in vitro anti-breast cancer potential by inhibiting the proliferation and migration of 4T1 cells.

Abstract:A novel staurosporine derivate, streptomholyrine A (1), along with 6 known compounds were identified from the rice-based solid fermentation of marine-derived Streptomyces sp. ZS-A121. The planar structure and absolute configuration of streptomholyrine A were elucidated using a combination of 1D, 2D NMR, HRESIMS data analysis, chemical transformation, ECD and NMR calculations. Screening of all these compounds revealed their cytotoxic activity against HCT-116 cell lines, with IC50 values ranging from 0.012 to 11.67 mu M, except for the known 1H-indole-3-hydroxyacetyl, which showed no inhibition activity. Furthermore, streptomholyrine A, along with two known staurosporine derivatives, k252d and staurosporine, exhibited activities against Candida albicans, with MICs of 12.5, 25.0 and 50.0 mu g/ml, respectively.

Abstract:Phytochemical investigations on the ethanol extract of the whole plant of Euphorbia maculata Linn. Resulted in the identification of 16 lanostane-related triterpenoids, including 11 undescribed ones, namely spiromaculatols A–C ( 1 – 3 ) and euphomaculatoids A–H ( 4 – 11 ). The structural determinations of the previously undescribed ones ( 1 – 11 ) were elucidated based on the interpretation of comprehensive spectroscopic data, quantum chemical calculation, as well as X-ray crystallographic experiments. Spiromaculatols A–C ( 1 – 3 ) possess a rare spirobi [indane] skeleton, which was biosynthetically derived from the 7 (8 → 9)- abeo bond migration of lanostane precursors. The biological activity of compounds 1 – 3 , 5 , 7, and 12 – 13 displayed inhibitory effect on the release of NO in an LPS-activated RAW264.7 cells model. Molecular mechanism study indicated that the most potent spiromaculatol C ( 3 ) can reduce the nuclear translocation of NF- κ B p65 and decrease the transcriptional expressions of its downstream pro-inflammatory mediators.

Abstract:Celastrol, a friedelane-type triterpenoid isolated from the genus Triperygium, possesses antitumor, anti-inflammatory, and immunosuppressive activities. A total of 42 celastrol derivatives (1a–1t, 2a–2l, and 3a–3j) were synthesized and evaluated for their immunosuppressive activities. Compounds 2a–2e showed immunosuppressive effects, with IC50 values ranging from 25 to 83 nM, and weak cytotoxicity (CC50 > 1 μM). Compound 2a, with a selectivity index value 31 times higher than that of celastrol, was selected as a lead compound. Abstract:Thirteen steroids ( 1 – 13 ), including two undescribed steroids, acrocalysterol C ( 1 ) and acrocalysterol D ( 2 ), were isolated from cultures of the fungus Acrocalymma cycadis using the OSMAC approach. Compounds 1 and 2 represent highly oxygenated steroids that contain an epoxy fragment. Through NMR and HRESIMS analyses, quantum chemical calculations, and electronic circular dichroism (ECD) experiments, their structures and absolute configurations were characterized. Then, the in vitro cytotoxicity of each compound was evaluated. Compounds 4 and 8 - 10 exhibited remarkable cytotoxicities against HeLa cells. Obviously, compound 10 exhibited twofold greater activity against HeLa than positive control, with IC 50 values of 13.2 ± 1.23 μ M. Additionally, compound 2 presented moderate potent cytotoxicity activities against three human cancer cells, comparable to the positive control. Thus, the bioactive metabolites of the strain A. cycadis may serve as a novel resource for development.

Abstract:Two novel triterpenoid saponins, colqueleganoids A (1) and B (2), with the first methyl-30 incorporated 6/6/6/6-cyclized carbon skeleton (named colquelegane), were isolated from the root of Colquhounia elegans. Their structures including absolute configuration were determined by spectroscopic methods and X-ray crystallographic analyses. Abstract:Cyclopiumolides A (1) and B (2), first representatives of two novel biosynthetic related 13-membered macrolides featuring an uncommon verrucosidinol unit condensed with a spiculisporic acidic moiety, were identified from the fungus Penicillium cyclopium SD-413, which was obtained from the deep-sea sediments collected in the East China Sea. The structures of cyclopiumolides A (1) and B (2) were identified on the basis of extensive NMR spectroscopic and mass spectrometric data analysis. Their relative and absolute configurations were determined by quantum mechanical calculations of ECD spectra comparing with that of experimental curves and by DP4 + NMR data calculations. Compounds 1 and 2 exhibited significant cytotoxic potencies against the tumor cell lines SF126, FaDu, and TE-1 with IC50 values ranging from 5.86 to 17.05 μM. The inhibition modes and binding sites of 1 and 2 were inspected using molecular docking simulations.

Abstract:An unusual pyridine-containing sesterterpenoid, leucosceptrodine (1), and five new nor-leucosceptrane sesterterpenoids including bisnor- (C23, 2), tetranor- (C21, 3) and pentanor- (C20, 4−6) skeletons, were isolated from the leaves of Tibetan Leucosceptrum canum. Their structures including the absolute configurations were determined by extensive spectroscopic analyses and quantum chemical calculations. Single crystal of one epimer (5) was crystallized from a pair of inseparable epimers, and its structure including absolute configuration was determined by X-ray crystallographic analysis. Immunosuppressive activity of compounds 1−4 with different potencies through inhibiting the secretion of cytokines TNF-α and IL-6 in LPS-induced macrophages RAW264.7 was observed.

Abstract:Seven new 14-membered macrolides, pestalotioprolides C (2), D-H (4-8), and 7-O-methylnigrosporolide (3), together with four known analogues, pestalotioprolide B (1), seiricuprolide (9), nigrosporolide (10), and 4,7-dihydroxy-13-tetradeca-2,5,8-trienolide (11), were isolated from the mangrove-derived endophytic fungus Pestalotiopsis microspora. Their structures were elucidated by analysis of NMR and MS data and by comparison with literature data. Single-crystal X-ray diffraction analysis was used to confirm the absolute configurations of 1, 2, and 10, while Mosher's method and the TDDFT-ECD approach were applied to determine the absolute configurations of 5 and 6. Compounds 3-6 showed significant cytotoxicity against the murine lymphoma cell line L5178Y with IC50 values of 0.7, 5.6, 3.4, and 3.9 μM, respectively, while compound 5 showed potent activity against the human ovarian cancer cell line A2780 with an IC50 value of 1.2 μM. Structure-activity relationships are discussed. Coculture of P. microspora with Streptomyces lividans caused a roughly 10-fold enhanced accumulation of compounds 5 and 6 compared to axenic fungal control.

Abstract:Lithocarpins E‐G, featuring a rare naturally‐occurring highly oxygenated tenellone‐macrolide skeleton, were isolated from the culture extract of a marine‐derived fungus Phomopsis lithocarpus FS508. Their structures were fully elucidated by NMR and MS spectroscopic analysis and electronic circular dichroism calculations. Compounds 1‐3 were evaluated for their in vitro cytotoxicities against HepG2, MCF‐7, SF‐268, as well as A549 cell lines, among which compound 1 exhibited inhibitory activity against HepG2 cells with an IC50 value of 6.3 μM. The further mechanistic investigation demonstrated that lithocarpin E could cause the apoptosis of HepG2 cells through activation of p‐ERK, Bax and caspase‐3 gene expression.This article is protected by copyright. Abstract:Six undescribed compounds, including four undescribed ecdysteroids (cyathsterones A-D) and two undescribed phenolic glycosides (cyathglucosides A-B), were isolated from the roots of Cyathula officinalis Kuan. Their structures were based on chemical analyses, NMR spectroscopic evidence, DP4+ calculations, and hydrolysis products. All compounds inhibited NO release in lipopolysaccharide (LPS)-induced RAW 264.7 macrophages in vitro. Among them, cyathsterone A showed the strongest inhibitory effects. Moreover, cyathsterone A has been shown to inhibit the release of the proinflammatory cytokines TNF-α, IL-6, and IL-1β in LPS-induced RAW 264.7 macrophages in vitro. Further studies found that cyathsterone A present concentration-dependent suppression of the protein expression of iNOS and COX-2 in LPS-stimulated RAW 264.7 cells in vitro and exerted anti-inflammatory activity via the NF-κB signalling pathway.

Function and Ultrastructure of Platelets of Neonates: Enhanced Ristocetin Aggregation of Neonatal Platelets

Abstract:We have investigated platelet morphology and function in human maternal‐newborn pairs. Fibrinogen concentration and factor‐VIII activity in plasma were also determined. Our results showed that, compared to maternal platelets, neonatal platelets were poorly responsive to adenosine diphosphate, adrenaline and collagen. Uptake of labelled serotonin by neonatal and maternal platelets was the same, but release of the radioactivity was reduced in the former. Phagocytic activity of neonatal platelets, demonstrated with latex particles, was similar to that of maternal platelets. Although the ultrastructure of neonatal platelets approximated that of maternal platelets, immature appearing platelets were occasionally found in the neonatal samples. An unanticipated finding was that platelet aggregation induced by ristocetin was more vigorous in neonatal than in maternal platelet‐rich plasma samples. Furthermore, neonatal plasma, which had lower fibrinogen and factor‐VIII content than maternal plasma, facilitated maternal platelet aggregation by ristocetin and showed a greater ability than maternal plasma to promote ristocetin‐induced aggregation of platelets of a patient with von Willebrand's disease. These results indicate that the plasma of neonates contains large quantities of the ristocetin‐dependent platelet aggregation factor (RAF), probably more than is in maternal plasma, despite the higher levels of factor‐VIII procoagulant activity in the latter. Thus, in the newborn, there is a clear dissociation between factor‐VIII clotting activity and the RAF activity.
Discovery, Total Synthesis and Key Structural Elements for the Immunosuppressive Activity of Cocosolide, a Symmetrical Glycosylated Macrolide Dimer from Marine Cyanobacteria.

Abstract:A new dimeric macrolide xylopyranoside, cocosolide (1), was isolated from the marine cyanobacterium preliminarily identified as Symploca sp. from Guam. The structure was determined by a combination of NMR spectroscopy, HRMS, X-ray diffraction studies and Mosher's analysis of the base hydrolysis product. Its carbon skeleton closely resembles that of clavosolides A-D isolated from the sponge Myriastra clavosa, for which no bioactivity is known. We performed the first total synthesis of cocosolide (1) along with its [α,α]-anomer (26) and macrocyclic core (28), thus leading to the confirmation of the structure of natural 1. The convergent synthesis featured Wadsworth-Emmons cyclopropanation, Sakurai annulation, Yamaguchi macrocyclization/dimerization reaction, α-selective glycosidation and β-selective glycosidation. Compounds 1 and 26 potently inhibited IL-2 production in both T-cell receptor dependent and independent manners. Full activity requires the presence of the sugar moiety as well as the intact dimeric structure. Cocosolide also suppressed the proliferation of anti-CD3-stimulated T-cells in a dose-dependent manner.

Abstract:A phytochemical investigation of the dichloromethane soluble fraction of the ethanolic extract obtained from the roots of Marsdenia tenacissima led to the discovery of the sixteen undescribed pregnane C21 steroids (1-16) and isolation of eleven known C21 steroidal analogues (17-27). Their chemical structures were elucidated by one- and two-dimensional nuclear magnetic resonance spectroscopy and, high resolution-electrospray ionization mass spectrometry and their absolute configurations were determined using electronic circular dichroism or single-crystal X-ray diffraction. The in vitro anti-proliferative effects of 1-16 were evaluated against HepG2 (human hepatocellular cancer), A549 (lung cancer), and MCF-7 (human breast cancer) cell lines. Even though some of them showed moderate cytotoxic activities, marsectohexol derivative 12 exhibited significant cytotoxicity against A549 cells with an IC50 value of 5.2 μM.

Abstract:Five new ergostanes, penicisteroids D-H (1-5), were isolated from the liquid culture of the deep-sea-derived fungus Penicillium granulatum MCCC 3A00475, along with 27 known compounds. The structures of the new steroids were established mainly on the basis of extensive analysis of 1D and 2D NMR as well as HRESIMS data. Moreover, the absolute configurations of 1 were confirmed unambiguously by the single-crystal X-ray crystallography. Compounds 2 and 4-7 showed moderate antiproliferative effects selectively against 12 different cancer cell lines with IC50 values of around 5 M. Compounds 2 and 6, potent RXR binders with Kd values of 13.8 and 12.9 M, respectively, could induce apoptosis by a Retinoid X Receptor (RXR)--dependent mechanism by regulating RXR transcriptional expression and promoting the poly-ADP-ribose polymerase (PARP) cleavage. Moreover, they could inhibit proliferation by cell cycle arrest at the G0/G1 phase.
