Abstract:In recent years, with the rapid development of installation cultivation of vegetable, many questions which constraint its development appeared. Among them, nematode disease is one of the most important constraint factors, one of which the harm caused by the root-knot nematodes (Meloidogyne spp.) is the most serious. Root-knot nematode is internal parasites settlement in plant roots, which has become one of most governing pathogen threaten to agricultural for its widespread and more parasites. In the past, chemicals were used to control this disease, combined with agricultural control and the use of resistant varieties, but has its limitations. With the advancement of technology and enhanced awareness of environmental protection, biological control has aroused a great attention. This paper reviews the advances of various biological control factors, and important factors influenced the efficiencies of biological control to nematode were analyzed thoroughly, in the last, the application prospect of the biological control were reviewed.
Utilization of Plant Materials for Control of Soybean Cysts Nematode

Abstract:Field experiments were conducted in 2011 and repeated in 2012 to study the nematicidal potentials of acacia, brassica, neem, and seaweed as alternatives to chemical nematicide on population management of Heterodera glycines and growth and yield of soybean. Results obtained showed acacia at 200 g, neem at 450 g, brassica at 1000 g, and seaweed at 2000 g caused the maximum suppression of population of H. glycines and produced the highest yield and growth of soybean. Neem, brassica, acacia, and seaweed caused reduction in H. glycines population by 48.3%, 41.4%, 34.5%, and 28.3%, respectively. Also, neem, brassica, acacia, and seaweed produced increase in soybean pods number by 58.7%, 33.7%, 28%, and seaweed 37.9%, respectively. The results obtained showed the potentialities of these plant materials as alternative to synthetic chemical pesticides. The plant could be utilized in a sustainable farming system to save the environment, conserve the biodiversity, and protect man against harmful effects of chemical nematicides.

Abstract:Plant-parasitic nematodes are one of the most destructive agronomic pests. During several decades, the control of this pest on agricultural crops has depended on chemical pesticides. These chemicals are very toxic with high potential to pollute the environment. Nowadays, the search for substitute products has become a priority. In this sense, the biological control agents have arisen as an environmentally friendly alternative. Different rhizobacterial strains are able to control nematodes improving plant health through the production of secondary metabolites. The aim of this chapter is to review the secondary metabolites produced by rhizospheric bacteria involved in the control of plant-parasitic nematodes. The use of these compounds could help to overcome the problem related with the survival of the biocontrol agents when introduced in new ecosystems. In the same way, the study of the metabolic pathways that lead to the production of these compounds can help to discern the conditions to trigger their production and its consequent activity in the rhizosphere.
Bio-control agents activate plant immune response and prime susceptible tomato against root-knot nematodes

Abstract:Beneficial microorganisms are generally known to activate plant defense against biotic challenges. However, the molecular mechanisms by which activated plants react more rapidly and actively to pests remain still largely unclear. Tomato plants pre-treated with a mixture of beneficial bio-control agents (BCAs), as soil-drenches, were less sensitive to infection of the root-knot nematode (RKN) Meloidogyne incognita. To unravel the molecular mechanisms of this induced resistance against RKNs, we used qRT-PCR to monitor the expression, in tomato roots and leaves, of 6 key defense genes. Gene transcripts were detected until the 12th day after BCA treatment(3, 7, 8, 12 dpt) and3 and 7 days after nematode inoculation of pre-treated plants. Early after BCA treatment, the salicylic acid (SA)-dependent pathogenesis related gene (PR-gene), PR-1b, marker of the systemic acquired resistance (SAR), was systemically over-expressed. Another PR-gene, PR-5, was over-expressed at later stages of BCA-plant interaction, and only in roots. Activation of defense against RKNs was attested by the early up-regulation of 4 genes (PR-1, PR-3, PR-5, ACO) in pre-treated plants after inoculation. Conversely, the expression of the JA/ET-dependent gene JERF3 did not increase after nematode inoculation in primed plants. A catalase gene (CAT)was highly over-expressed by nematode infection, however, this over-expression was annulled at the earliest stages or limited at the later stages of infection toBCA-treated roots. Enzyme activities, such as glucanase and endochitinase, were enhanced in roots of pre-treated inoculated plants with respect to plants left not inoculated as a control. These findings indicate that BCA interaction with roots primes plants against RKNs. BCA-mediated immunity seems to rely on SA-mediated SAR and to be associated with both the activation of chitinase and glucanase enzyme activities and the inhibition of the plant antioxidant enzyme system. Immunity is triggered at the penetration and movements inside the roots of the invading nematode juveniles but probably acts at the feeding site building stage of nematode infection.

Abstract:ABSTRACT The pathogen, Rhizoctonia solani Kuhn is a polyphagous fungus and has a vast potential of attacking a wide range of host plants causing host decay, damping –off, stem rot, crown rot ,stemcanker, root rot, fruit decay and foliage diseases. The stem rot is an emerging devastating disease incarnation rating next in importance to Fusarium wilt with a seedling mortality of 37% under optimum climatic conditions. Because of high incidence and crop importance, the present studywas carried out to manage the disease with application of different plant extracts (botanicals), biofumigant releasing crop residues and biocontrol agents both under in vitro and field conditions. Out of 11 botanicals including two commercial formulations of neem (neemgold, neemazil) testedat 5,10,20,30,40% and 1,2,3,4,5% concentrations respectively revealed that seed extracts of Meliaazedarach and leaf extract of Adhatoda vasica showed maximum inhibition in mycelial growthwithin the range of 44.38 to 44.25% followed by Murraya koenigii (37.77%) and Tagetes erecta(37.62%).The commercial formulations of neem compared to other treatments were foundstatistically superior to inhibits mycelial growth. In general, higher concentration resulted in moreinhibition of the R. solani, above 50 % inhibition was recorded at concentration of 40 per cent, maximum being in neemgold (60.53%) compared to 5% or 10%. Among different crop residues, cauliflower and cabbage were found most effective in reducing the incidence of carnation stem rot to 22.28 and 24.25% from 48.34% registered in control. The plant and flower characters such as plant height, stem length, no. of flowers/plant, flower size/plant and no. of days to first floweringget reduced to 142 compared to 157 days. The average colony forming unit of the pathogen reducedto 22.83x103 from 58.12x103 in one cropping season. Antagonists, Trichoderma viride, T. harzianum and Bacillus subtilis resulted into mycelial inhibition of 65.08, 63.70 and 55.05%. However in field the highest disease control of stem rot was achieved in integrated treatments whenthe rooted cuttings of carnation were pretreated with neemgold or neemazil for 20 minutes as diptreatment and planted in soil amended with T. viride and T. harzianum by giving disease control between 63-73%.
Impacts of Root Metabolites on Soil Nematodes

Abstract:Plant parasitic nematodes cause significant crop damage globally. Currently, many nematicides have been banned or are being phased out in Europe and other parts of the world because of environmental and human health concerns. Therefore, we need to focus on sustainable and alternative methods of nematode control to protect crops. Plant roots contain and release a wide range of bioactive secondary metabolites, many of which are known defense compounds. Hence, profound understanding of the root mediated interactions between plants and plant parasitic nematodes may contribute to efficient control and management of pest nematodes. In this review, we have compiled literature that documents effects of root metabolites on plant parasitic nematodes. These chemical compounds act as either nematode attractants, repellents, hatching stimulants or inhibitors. We have summarized the few studies that describe how root metabolites regulate the expression of nematode genes. As non-herbivorous nematodes contribute to decomposition, nutrient mineralization, microbial community structuring and control of herbivorous insect larvae, we also review the impact of plant metabolites on these non-target organisms.

Abstract:Summary The foundation of most food production systems underpinning global food security is the careful management of soil resources. Embedded in the concept of soil health is the impact of diverse soil‐borne pests and pathogens, and phytoparasitic nematodes represent a particular challenge. Root‐knot nematodes and cyst nematodes are severe threats to agriculture, accounting for annual yield losses of US$157 billion. The control of soil‐borne phytoparasitic nematodes conventionally relies on the use of chemical nematicides, which can have adverse effects on the environment and human health due to their persistence in soil, plants, and water. Nematode‐resistant plants offer a promising alternative, but genetic resistance is species‐dependent, limited to a few crops, and breeding and deploying resistant cultivars often takes years. Novel approaches for the control of phytoparasitic nematodes are therefore required, those that specifically target these parasites in the ground whilst minimizing the impact on the environment, agricultural ecosystems, and human health. In addition to the development of next‐generation, environmentally safer nematicides, promising biochemical strategies include the combination of RNA interference (RNAi) with nanomaterials that ensure the targeted delivery and controlled release of double‐stranded RNA. Genome sequencing has identified more than 75 genes in root knot and cyst nematodes that have been targeted with RNAi so far. But despite encouraging results, the delivery of dsRNA to nematodes in the soil remains inefficient. In this review article, we describe the state‐of‐the‐art RNAi approaches targeting phytoparasitic nematodes and consider the potential benefits of nanotechnology to improve dsRNA delivery.

Abstract:Root-knot nematodes (RKN) ( Meloidogyne spp.) pose a major threat to horticultural crop production globally. Despite yield loss, use of synthetic chemicals to suppress RKN in horticulture crops raised concerns over produce residual toxicity, increased pest tolerance, and environmental chemical persistence. In order to address these issues, sustainable pest management practices are essential to tackle RKN infestation in horticultural crops, while maintaining ecological balance and environmental safety, apart from ensuring nutritional security for humans. The different mechanisms of RKN infestation at molecular level in horticultural crops and their interaction with plants through nematode associated molecular patterns have been discussed. The review emphasizes on sustainable pest management strategies by deciphering the efficiency of phytomolecules in different forms as bionematicides, role of plant signalling molecules in triggering innate immunity of plants and exploring crop wild relatives as resistant rootstocks in imparting resistance against invading nematodes in horticultural crops. Phytomolecules reported as bionematicides against RKNs in horticultural crops includes quercetin, caryophyllene, medicagenic acid, 1,4-naphaloacetic acid, allicin and prodelphinidin along with essential oils (castor and mint), organic acids (humic acid, acetic acid, and maleic acid), volatile organic compounds (dimethyl disulfide) and(methoxymethyl)-benzene. In addition, practical usage of plant bionematicides, challenges and future perspectives for efficient use of phytomolecules as source of resistance and as an ecofriendly approach for management of RKN, including bibliometric analysis of phytochemical based bionematicides has been discussed in detail. The review will help in addressing the research gaps and in developing effective bionematicides from plant source for sustainable pest management in horticultural crops.

Abstract:Plant-parasitic-nematodes represent a major threat to the agricultural production of different crops worldwide. Due to the high toxicity of chemical nematicides, it is necessary to develop new control strategies against nematodes. In this respect, filamentous fungi can be an interesting biocontrol alternative. The genus Trichoderma, mycorrhizal and endophytic fungi are the main groups of filamentous fungi studied and used as biological control agents (BCAs) against nematodes as resistance inducers. They are able to reduce the damage caused by plant-parasitic nematodes directly by parasitism, antibiosis, paralysis and by the production of lytic enzymes. But they also minimize harm by space and resource-competition, by providing higher nutrient and water uptake to the plant, or by modifying the root morphology, and/or rhizosphere interactions, that constitutes an advantage for the plant-growth. Besides, filamentous fungi are able to induce resistance against nematodes by activating hormone-mediated (salicylic and jasmonic acid, strigolactones among others) plant-defense mechanisms. Additionally, the alteration of the transport of chemical defense components through the plant or the synthesis of plant secondary metabolites and different enzymes can also contribute to enhancing plant defenses. Therefore, the use of filamentous fungi of the mentioned groups as BCAs is a promising durable biocontrol strategy in agriculture against plant-parasitic nematodes.

Abstract:Entomopathogenic nematodes are natural enemies and effective biological control agents of subterranean insect herbivores. Interactions between herbivores, plants, and entomopathogenic nematodes are mediated by plant defense pathways. These pathways can induce release of volatiles and recruit entomopathogenic nematodes. Stimulation of these plant defense pathways for induced defense against belowground herbivory may enhance biological control in the field. Knowledge of the factors affecting entomopathogenic nematode behaviour belowground is needed to effectively implement such strategies. To that end, we explore the effect of elicitor, elicitor dose, mechanical damage, and entomopathogenic nematode release distance on recruitment of entomopathogenic nematode infective juveniles to corn seedlings. Increasing doses of methyl jasmonate and methyl salicylate elicitors recruited more entomopathogenic nematodes as did mechanical damage. Recruitment of entomopathogenic nematodes was higher at greater release distances. These results suggest entomopathogenic nematodes are highly tuned to plant status and present a strategy for enhancing biological control using elicitor-stimulated recruitment of entomopathogenic nematodes.

Abstract:Root-knot nematodes (Meloidogyne spp.) are sedentary endoparasites that cause severe economic losses to agricultural crops globally. Due to the regulations of the European Union on the application of nematicides, it is crucial now to discover eco-friendly control strategies for nematode management. Biocontrol is one such safe and reliable method for managing these polyphagous nematodes. Biocontrol agents not only control these parasitic nematodes but also improve plant growth and induce systemic resistance in plants against a variety of biotic stresses. A wide range of organisms such as bacteria, fungi, viruses, and protozoans live in their natural mode as nematode antagonists. Various review articles have discussed the role of biocontrol in nematode management in general, but a specific review on biocontrol of root-knot nematodes is not available in detail. This review, therefore, focuses on the biocontrol of root-knot nematodes by discussing their important known antagonists, modes of action, and interactions.

Abstract:Plant parasitic nematodes (PPNs), such as Meloidogyne spp., Heterodera spp. and Pratylenchus spp., are obligate parasites on a wide range of crops, causing significant agricultural production losses worldwide. These PPNs mainly feed on and within roots, impairing both the below-ground and the above-ground parts, resulting in reduced plant performance. Plants have developed a multi-component defence mechanism against diverse pathogens, including PPNs. Several natural molecules, ranging from cell wall components to secondary metabolites, have been found to protect plants from PPN attack by conferring nematode-specific resistance. Recent advances in omics analytical tools have encouraged researchers to shed light on nematode detection and the biochemical defence mechanisms of plants during nematode infection. Here, we discuss the recent progress on revealing the nematode-associated molecular patterns (NAMPs) and their receptors in plants. The biochemical defence responses of plants, comprising cell wall reinforcement; reactive oxygen species burst; receptor-like cytoplasmic kinases; mitogen-activated protein kinases; antioxidant activities; phytohormone biosynthesis and signalling; transcription factor activation; and the production of anti-PPN phytochemicals are also described. Finally, we also examine the role of epigenetics in regulating the transcriptional response to nematode attack. Understanding the plant defence mechanism against PPN attack is of paramount importance in developing new, effective and sustainable control strategies.

Abstract:AbstractRoot-knot nematodes (RKN) are sedentary parasites of the roots of plants and are considered some of the most damaging pests in agriculture. Since RKN target the root vascular system, they provoke host nutrient deprivation and defective water transport, causing above-ground symptoms of growth stunting, wilting, chlorosis, and reduced crop yields. In Mexico RKN infestations are primarily dealt with by treating with synthetic chemically based nematicides that are preferred by farmers over available bioproducts. However, due to environmental and human health concerns chemical control is increasingly restricted. Biological control of RKNs can help reduce the use of chemical nematicides as it is achieved with antagonistic organisms, mainly bacteria, fungi, other nematodes, or consortia of diverse microorganisms, which control nematodes directly by predation and parasitism at different stages: eggs, juveniles, or adults; or indirectly by the action of toxic diffusible inhibitory metabolites. The need to increase agricultural production and reduce negative environmental impact creates an opportunity for optimizing biological control agents to suppress nematode populations, but this endeavour remains challenging as researchers around the world try to understand diverse control mechanisms, nematode and microbe life cycles, ecology, metabolite production, predatory behaviours, molecular and biochemical interactions, in order to generate attractive products with the approval of local regulatory bodies. Here, we provide a brief review of the biology of the genus Meloidogyne, biological control strategies, and a comparison between chemical and bioproducts in the Mexican market, and guidelines emitted by national agencies to ensure safety and effectiveness of new developments.

Biocontrol of Meloidogyne spp. in Solanum lycopersicum using a dual combination of Bacillus strains

Abstract:Root-knot nematodes (RKNs, Meloidogyne spp.) are obligate plant parasites that constitute a significant pest for agriculture worldwide. They penetrate the plant roots, reducing the uptake of water and nutrients, causing a significant impact on crop yield. One alternative on focus now for nematode management is biological control. Rhizobacteria within the Bacillus genus show multiple modes of action against plant-parasitic nematodes (PPNs) that can act alone or in combination. In this context, we evaluated a dual-strain bacteria combination ( B. paralicheniformi FMCH001 and B. subtilis FMCH002) to reduce nematode infection in tomato plants. We evaluated mortality of larvae from Meloidogyne javanica in vitro , as well as eggs hatching after the treatment. Atraction, penetration, establishment, and reproduction assays in vitro or in pots in tomato plants infected with M. javanica and treated/ untreated with the dual-strain bacteria combination were also performed. Additionally, morphometric parameters comparing giant cells size from galls of treated and untreated plants by using confocal microscopy were also measured. The results showed that this combination of strains has nematicidal properties in the pre-infection phase by decreasing the egg-hatching, juvenile survival, and attractiveness to the roots. Furthermore, nematode establishment, gall formation, and, remarkably, giant cell development was severely impaired after the bacterial treatment, suggesting interference with morphogenetic mechanisms induced by the nematode during GCs development within the plant. Nematode reproduction in tomato plants was reduced independently of the application mode in soil, before or after bacterial treatment. The dual-strain combination was also effective against other PPNs (i.e. Pratylenchus spp.) and in different crops (soybean). Therefore, combining B. paralicheniformis FMCH001 and B. subtilis FMCH002 is an efficient agent for the biological control of Meloidogyne spp. by interfering with different stages of the nematode cycle as a result of multiple modes of action.

Upgrading Strategies for Managing Nematode Pests on Profitable Crops

Abstract:Plant-parasitic nematodes (PPNs) reduce the high profitability of many crops and degrade their quantitative and qualitative yields globally. Traditional nematicides and other nematode control methods are being used against PPNs. However, stakeholders are searching for more sustainable and effective alternatives with limited side effects on the environment and mankind to face increased food demand, unfavorable climate change, and using unhealthy nematicides. This review focuses on upgrading the pre-procedures of PPN control as well as novel measures for their effective and durable management strategies on economically important crops. Sound and effective sampling, extraction, identification, and counting methods of PPNs and their related microorganisms, in addition to perfecting designation of nematode–host susceptibility/resistance, form the bases for these strategies. Therefore, their related frontiers should be expanded to synthesize innovative integrated solutions for these strategies. The latter involve supplanting unsafe nematicides with a new generation of safe and reliable chemical nematicidal and bionematicidal alternatives. For better efficacy, nematicidal materials and techniques should be further developed via computer-aided nematicide design. Bioinformatics devices can reinforce the potential of safe and effective biocontrol agents (BCAs) and their active components. They can delineate the interactions of bionematicides with their targeted PPN species and tackle complex diseases. Also, the functional plan of nematicides based on a blueprint of the intended goals should be further explored. Such goals can currently engage succinate dehydrogenase, acetylcholinesterase, and chitin deacetylase. Nonetheless, other biochemical compounds as novel targets for nematicides should be earnestly sought. Commonly used nematicides should be further tested for synergistic or additive function and be optimized via novel sequential, dual-purpose, and co-application of agricultural inputs, especially in integrated pest management schemes. Future directions and research priorities should address this novelty. Meanwhile, emerging bioactivated nematicides that offer reliability and nematode selectivity should be advanced for their favorable large-scale synthesis. Recent technological means should intervene to prevail over nematicide-related limitations. Nanoencapsulation can challenge production costs, effectiveness, and manufacturing defects of some nematicides. Recent progress in studying molecular plant–nematode interaction mechanisms can be further exploited for novel PPN control given related topics such as interfering RNA techniques, RNA-Seq in BCA development, and targeted genome editing. A few recent materials/techniques for control of PPNs in durable agroecosystems via decision support tools and decision support systems are addressed. The capability and effectiveness of nematicide operation harmony should be optimized via employing proper cooperative mechanisms among all partners.

Abstract:The inhabitation and parasitism of root-knot nematodes (RKNs) can be difficult to control, as its symptoms can be easily confused with other plant diseases; hence, identifying and controlling the occurrence of RKNs in plants remains an ongoing challenge. Moreover, there are only a few biological agents for controlling these harmful nematodes. In this study, Xenorhabdus sp. SCG isolated from entomopathogenic nematodes of genus Steinernema was evaluated for nematicidal effects under in vitro and greenhouse conditions. The cell-free filtrates of strain SCG showed nematicidal activity against Meloidogyne species J2s, with mortalities of > 88% at a final concentration of 10%, as well as significant nematicidal activity against the three other genera of plant-parasitic nematodes in a dose-dependent manner. Thymine was isolated as active compounds by assayguided fractionation and showed high nematicidal activity against M. incognita. Greenhouse experiments suggested that cell-free filtrates of strain SCG efficiently controlled the nematode population in M. incognita-infested tomatoes (Solanum lycopersicum L., cv. Rutgers). In addition, a significant increase in host plant growth was observed after 45 days of treatment. To our knowledge, this is the first to demonstrate the nematicidal activity spectrum of isolated Xenorhabdus species and their application to S. lycopersicum L., cv. Rutgers under greenhouse conditions. Xenorhabdus sp. SCG could be a promising biological nematicidal agent with plant growth-enhancing properties.

Abstract:Control of plant-parasitic nematodes essentially involves use of synthetic nematicides. However, apart from cost, increased concern for the environment has necessitated a reduction in the amount of nematicides used for control. Additionally, there has been an increase in the search for other efficient; ecologically sound and safe control methods. Eggs of root-knot nematode, Meloidogyne incognita (Kofoid & White) Chitwood, were exposed to concentrations of root extracts of Siam weed [Chromolaena odorata (L.) King and Robinson], Neem (Azadirachta indica A. Jass), Castor bean (Ricinus communis L.) and Lemon grass (Cymbopogon citratus L.). Undiluted (100%) concentrations of root extracts of Siam weed and Neem exhibited 100% inhibition of egg hatch and larval mortality, while the 100% concentration of root extracts of Castor bean and Lemon grass exhibited 93 and 95% inhibition of egg hatch and 62 and 75% larval mortality, respectively. Egg hatch inhibition and larval mortality decreased as extracts were diluted. With an increase in exposure time, juvenile mortality increased for the diluted concentrations, but time had no effect on juvenile mortality at the 100% concentration for all of the plant species.

Abstract:The root-knot nematode (RKN) causes significant yield loss in tomatoes. Understanding the interaction of biocontrol agents (BCAs)-nematicides-soil microbiomes and RKNs is essential for enhancing the efficacy of biocontrol agents and nematicides to curb RKN damage to crops. The present study aimed to evaluate the in vitro effectiveness of BACa and nematicide against RKN and to apply the amplicon sequencing to assess the interaction of Bacillus velezensis (VB7) and Trichoderma koningiopsis (TK) against RKNs. Metagenomic analysis revealed the relative abundance of three phyla such as Proteobacteria (42.16%), Firmicutes (19.57%), and Actinobacteria (17.69%) in tomato rhizospheres. Those tomato rhizospheres treated with the combined application of B. velezensis VB7 + T. koningiopsis TK and RKN had a greater frequency of diversity and richness than the control. RKN-infested tomato rhizosphere drenched with bacterial and fungal antagonists had the maximum diversity index of bacterial communities. A strong correlation with a maximum number of interconnection edges in the phyla Proteobacteria, Firmicutes, and Actinobacteria was evident in soils treated with both B. velezensis VB7 and T. koningiopsis TK challenged against RKN in infected soil. The present study determined a much greater diversity of bacterial taxa observed in tomato rhizosphere soils treated with B. velezensis VB7 and T. koningiopsis TK than in untreated soil. It is suggested that the increased diversity and abundance of bacterial communities might be responsible for increased nematicidal properties in tomato plants. Hence, the combined applications of B. velezensis VB7 and T. koningiopsis TK can enhance the nematicidal action to curb RKN infecting tomatoes.

Abstract:Root-knot nematodes are a growing concern for vegetable producers, because chemical nematicides are gradually disappearing. Alternative techniques based on agronomic practices are needed to solve the problem. This review analyzes the most recent studies related to these techniques and their combinations and identifies the most effective ones. Based on an agronomic point of view, the analysis focuses on a description of agricultural factors and practices, rather than on biological processes. Several alternative techniques are considered, including sanitation, soil management, organic amendments, fertilization, biological control and heat-based methods. We analyzed the effects of each practice and interactions among techniques and found large variations among studies. Many practices are only partially effective for nematode control; thus, combining control methods in a systemic analysis presents a challenge. We outline such an ongoing systemic approach and identify key future research studies.
