Abstract:The concentration of butyric acid in the intestine increased with the increase in the content of fermentable dietary fibre; however, the potential physiological impact of a high dose of butyric acid on fish has not been sufficiently studied. The aim of this study was to investigate the effect of two dosages of butyric acid on the growth and health of the liver and intestine of the largemouth bass (Micropterus salmoides). Sodium butyrate (SB) was added to the diet at 0 g/kg (CON), 2 g/kg (SB2), and 20 g/kg (SB20), and the juvenile largemouth bass were fed to apparent satiation for 56 days. No significant difference was observed in the specific growth rate or hepatosomatic index among the groups ( P > 0.05 ). The concentration of β-hydroxybutyric acid in the liver, the activities of alanine aminotransferase, aspartate aminotransferase, and alkaline phosphatase, and the concentrations of triglyceride and total cholesterol in serum increased significantly in the SB20 group compared to the CON group ( P < 0.05 ). The relative expression of fas, acc, il1b, nfkb, and tnfa in the liver of the SB20 groups was also significantly higher than that of the CON group ( P < 0.05 ). The above indicators in the group SB2 had similar change tendencies. The expression of nfkb and il1b in the intestine of both the SB2 and SB20 groups was significantly downregulated compared with that in the CON group ( P < 0.05 ). The size of hepatocytes was enlarged, and the intracellular lipid droplets and the degree of hepatic fibrosis were increased in the SB20 group compared to the CON group. There was no significant difference in intestinal morphology among the groups. The above results indicated that neither 2 g/kg nor 20 g/kg SB had a positive effect on the growth of largemouth bass, while a high dosage of SB induced liver fat accumulation and fibrosis.

Abstract:The aim of this study was to investigate the effects of sodium butyrate (SB) supplementation on growth performance, antioxidant enzyme activities, inflammatory factors, and hypoxic stress in largemouth bass ( Micropterus salmoides ). Diets were supplemented with different doses of SB at 0 (SB0), 0.5 (SB1), 1.0 (SB2) and 2.0 (SB3) g/kg. The hypoxic stress experiment was performed after 56 days of culture. The results showed that compared with the SB0 group, the final body weight, weight gain rate and protein deposition rate of the SB3 group were significantly increased ( P <0.05), while FCR was significantly decreased ( P <0.05). The contents of dry matter, crude lipids, and ash in the SB2 group were significantly higher than those in the SB0 group (P<0.05). The urea level was significantly decreased ( P <0.05), and the glucose content was significantly increased ( P <0.05) in the SB supplement group. Compared with the SB0 group, the SB2 group had significant reductions in the levels of serum triglyceride, cholesterol, elevated-density lipoprotein cholesterol, and low-density lipoprotein ( P <0.05), and significant reductions in the levels of liver alkaline phosphatase and malondialdehyde ( P <0.05). The total antioxidant capacity of the SB1 group was higher than that of other groups ( P <0.05). Compared with the SB0 group, the mRNA expression of TLR 22, MyD 88, TGF-β 1, IL -1 β and IL -8 in the SB2 group significantly decreased ( P <0.05). The cumulative mortality rate was significantly decreased in the SB2 and SB3 groups in comparison with that in the SB0 group after three hours of hypoxic stress ( P <0.05). In a 56-day feeding trial, SB enhanced largemouth bass growth by increasing antioxidant enzyme activity and inhibiting TLR 22- MyD 88 signaling, therefore increasing cumulative mortality from hypoxic stress in largemouth bass.

Abstract:This study assessed the effects of feed carbohydrate content on intestinal physical barrier and immunity in juvenile largemouth bass (Micropterus salmoides). Triplicate groups of juvenile fish (4.1 ± 0.2 g) were fed low (LCD, 7%), medium (MCD, 12%) and high (HCD, 17%) carbohydrate diets for eight weeks. Gut histology revealed the slight infiltration of inflammatory cells and moderate loss of mucous membrane layer in HCD group. Expression of ZO1, occluding, and claudin7 genes and epidermal growth factor receptor (EGFR) gene were significantly decreased in HCD group indicating the impairment of tight junction and epithelial cell regeneration. The results showed the significant (P < 0.05) reduction of antioxidant capacity in HCD group compared to LCD. Furthermore, expression of intestinal ERS-related genes such as IRE1, Eif2α, GRP78, CHOPα and CHOPβ in HCD group was significantly higher than the LCD group. In addition, HCD induced the up-regulated expression of inflammatory (IL-8, IL-1β, TNFα and COX2) and apoptosis (TRAF2, bax, casepase3, caspase8 and casepase9) related genes in fish intestine. The data generated in this study clearly demonstrated that HCD induced ERS and oxidative stress, which promoted intestinal inflammation and apoptosis in juvenile largemouth bass.

Abstract:Four experimental diets were formulated to evaluate the effect of coated lysine (Lys) and methionine (Met) supplemented in mixture protein diets of juvenile black sea bream, Acanthopagrus schlegelii. Fish meal (FM) was the main protein source in control diet, and 21.2% FM was replaced by the mixture of soybean protein concentrate, meat and bone meal, and monosodium glutamate protein in replacement diets with coating Lys and Met were suppled at none (0%, 0%), low (1.7%, 1.1%) and high (5.0%, 3.4%) level (designated as NLM, LLM and HLM diet, respectively). Each diet was fed to triplicate groups of 20 fish (initial mean weight, 4.13 +/- 0.16 g) twice a day to apparent satiation for 8 weeks, in a flow-through system. Results showed that amino acid supplementation significantly improved weight gain and specific growth rate in these mixture protein diets groups, and the fish fed with HLM diet showed comparable growth with the control group. Feed efficiency ratio (FER), protein efficiency ratio (PER), and crude protein and Met content in dorsal muscle were significantly affected by diet amino acid level. The highest apparent digestibility coefficients appeared in HLM group but the values showed insignificant difference with the control group. Serum activities of aspertate aminotransferase and alanine aminotransferase in control group were lower than those of in other treatments. Fish fed with NLM and LLM diets had higher triglycerides concentration than those of fed the HLM and FM diets. Furthermore, the feed cost of the various feed treatments were evaluated, and the three test diets were all lower than the control diet. The present study indicated that 21.2% FM in black sea bream diet can be successfully replaced by mixture protein with sufficient coated Lys (5.0%) and Met (3.4%) supplemented.

Abstract:Bile acid (BA) has been reported to improve growth performance and play an important role in lipid metabolism of fish. Five diets were formulated to investigate the effects of dietary bile acid (chenodeoxycholic acid) supplementation on growth performance, antioxidant capacity, lipid metabolism and intestinal health of juvenile largemouth bass (Micropterus salmoides). Fish (18.35 ± 0.05 g) were randomly fed five diets: the positive control diet (10.87% lipid, C), high fat diet (18.08% lipid, HF), and HF diets supplemented with 300, 600 and 900 mg/kg chenodeoxycholic acid (HFC1, HFC2 and HFC3, respectively). After 9 weeks of feeding experiment, the weight gain (WG) and special growth rate (SGR) were significantly lower in the fish fed diet HF compared with those fed diet C (P < 0.05). Compared with the group C, there were no significant differences in WG and SGR of fish fed high fat diet with 900 mg/kg chenodeoxycholic acid (P > 0.05). Viscerosomatic index (VSI) and intraperitoneal fat ratio (IPF) were significantly higher in fish fed high fat diets than those fed the control diet (P < 0.05). The crude lipid and protein contents in muscle showed no significant differences among fish fed diets with high dietary lipid (P > 0.05). With increasing supplementation of BA in diets, the crude lipid of muscle gradually decreased. Triglyceride (TG), malondialdehyde (MDA) in plasma and MDA in liver were higher in HF group than control group, addition of BA could decrease those contents effectively and increase the gene expression of SOD and GSH-Px. Lipidprotein transport and cholesterol synthesis related mRNA levels were significantly increased in fish fed high fat diets containing BA. The supplementation of 900 mg/kg BA significantly increased the gene expression of apolipoprotein B (ApoB-100), sterol 26-hydroxylase (CYP27α) and liver X receptor (LXR) (P < 0 0.05). High-fat diet significantly increased the pro-apoptotic gene expression, while anti-apoptotic gene B-cell lymphoma-2 (Bcl-2) and B-cell lymphoma-xl (Bcl-xl) mRNA levels were significantly increased with the increasing dietary BA (P < 0.05). In addition, supplemental BA in high-fat diet increased the height of intestinal fold and decreased lipid accumulation in liver. These results suggested that dietary BA supplementation could improve the digestion and absorption of lipids, antioxidant capacity and intestinal health, thereby attenuating the adverse effect induced by high fat diet of largemouth bass.

Abstract:Starch is an inexpensive feed ingredient that has been widely used in fish feed. However, starch utilization by carnivorous fish is limited and excess starch is detrimental to the health of the organism. High starch diets often lead to liver damage, but the effects on the intestine are often overlooked. Therefore, in this study, two isonitrogenous and isolipidic semi-pure diets (NC: 0% α-starch, HC: 22% α-starch) were formulated and fed to largemouth bass ( Micropterus salmoides) for 45 days. The effects of the high starch diet on the intestine of largemouth bass were comprehensively investigated by intestinal microbiota, histopathology, ultrastructural pathology, and enzymology analyses. Feeding the HC diet did not affect the growth of largemouth bass during the experimental period. However, the high starch diet led to a reduction in the diversity and abundance of intestinal microbiota in largemouth bass, with a significant increase in the abundance of harmful bacteria ( Aeromonas ) and a decrease in the abundance of beneficial bacteria ( Clostridium , Lactobacillus , and Bifidobacterium ). Feeding the HC diet caused the development of enteritis, with goblet cell hyperplasia, epithelial necrosis and detachment and inflammatory cell infiltration, and leading to enlarged apical openings and mitochondrial damage in goblet cells. Long-term feeding of the HC diet inhibited intestinal α-amylase activity. changes in the intestinal microbiota, such as an increase in Aeromonas and a decrease in Clostridium , Lactobacillus , and Bifidobacterium , may be closely related to the development of enteritis. Therefore, adding these beneficial bacteria as probiotics may be an effective way to prevent damage to the intestine of largemouth bass from a high carbohydrate diet. Our results suggest reducing the amount of starch added to the largemouth bass diets. This study provides a reference for protecting the largemouth bass gut during modern intensive culture.

Abstract:The present study was conducted to investigate the effects of dietary inclusion of protein hydrolysates on growth performance, digestive enzyme activities, protein metabolism, and intestinal health in larval largemouth bass (Micropterus salmoides). The experimental feeding trial presented in this study was based on five isonitrogenous and isolipidic diets formulated with graded inclusion levels of protein hydrolysates, and it showed that protein hydrolysates improved growth performance, reduced larval deformity rate, and increased the activity of digestive enzymes, including pepsin and trypsin. Gene expression results revealed that the supplementation of protein hydrolysates upregulated the expression of intestinal amino acid transporters LAT2 and peptide transporter 2 (PepT2), as well as the amino acid transporters LAT1 in muscle. Dietary provision of protein hydrolysates activated the target of rapamycin (TOR) pathway including the up-regulation of TOR and AKT1, and down-regulation of 4EBP1. Additionally, the expression of genes involved in the amino acids response (AAR) pathway, ATF4 and REDD1, were inhibited. Protein hydrolysates inhibited the transcription of some pro-inflammatory cytokines, including IL-8 and 5-LOX, but promoted the expression of anti-inflammatory cytokines TGF-beta and IL-10. The 16S rRNA analysis, using V3-V4 region, indicated that dietary protein hydrolysates supplementation reduced the diversity of the intestine microbial community, increased the enrichment of Plesiomonas and reduced the enrichment of Staphylococcus at the genus level. In summary, protein hydrolysates have been shown to be an active and useful supplement to positively complement other protein sources in the diets for largemouth bass larvae, and this study provided novel insights on the beneficial roles and possible mechanisms of action of dietary protein hydrolysates in improving the overall performance of fish larvae.

Abstract:Abstract A short-term 2-week (2w) and long-term 8-week (8w) feeding trial was conducted to investigate the effects of low-starch (LS) and high-starch (HS) diets on the growth performance, metabolism and liver health of largemouth bass (Micropterus salmoides). Two isonitrogenous and isolipidic diets containing two levels of starch (LS, 9·06 %; HS, 13·56 %) were fed to largemouth bass. The results indicated that HS diet had no significant effects on specific growth rate during 2w, whereas significantly lowered specific growth rate at 8w. HS diet significantly increased hepatic glycolysis and gluconeogenesis at postprandial 24 h in 2w. The hepatosomatic index, plasma alkaline phosphatase, total bile acid (TBA) levels, and hepatic glycogen, TAG, total cholesterol, TBA, and NEFA contents were significantly increased in the HS group at 2w. Moreover, HS diet up-regulated fatty acid and TAG synthesis-related genes and down-regulated TAG hydrolysis and β-oxidation-related genes. Therefore, the glucolipid metabolism disorders resulted in metabolic liver disease induced by HS diet at 2w. However, the up-regulation of bile acid synthesis, inflammation and energy metabolism-related genes in 2w indicated that largemouth bass was still in a state of ‘self-repair’ response. Interestingly, all the metabolic parameters were returned to homoeostasis, with up-regulation of intestinal glucose uptake and transport-related genes, even hepatic histopathological analysis showed no obvious abnormality in the HS group in 8w. In conclusion, HS feed induced short-term acute metabolic disorder, but long-term metabolic adaptation to HS diet was related to repairing metabolism disorders via improving inflammatory responses, bile acid synthesis and energy metabolism. These results strongly indicated that the largemouth bass owned certain adaptability to HS diet.

Abstract:Abstract Background: The intestine is the first line of defense against bacteria, toxins and antigens presents in the lumen of the instine. Aeromonas veronii is an opportunistic pathogen that has been related to mortality in fish. However, the protective mechanisms of intestinal damage caused by Aeromonas veronii are poorly defined. Thus, the objetive of this study was to evaluate the influence of dietary valine (Val) on growth, antioxidant capacity, and endoplasmic reticulum stress-mediated apoptosis in intestines of the juvenile largemouth bass Micropterus punctulatus after infection with Aeromonas veronii (A. veronii). Methods: A total of 720 fish (37.99 ± 0.01 g) were randomly allocated to six groups with three replicates each group, with different levels of Val (11.0, 14.0, 17.0, 20.1, 23.1, and 26.1 g/kg diet, respectively) for 77 d, and then challenged with A. veronii. Results: In the present study, we demonstrated that dietary Val: (1) increased the percent weight gain (PWG), specific growth rate (SGR), feed intake (FI), feed efficiency (FE), protein efficiency ratio (PER), protein production value (PPV), lipid production value (LPV), and ash production value (APV) (P< 0.05); (2) improved survival rate (SR) and alleviated intestinal injury; (3) reduced the levels of reactive oxygen species (ROS), malondialdehyde (MDA), and protein carbonyl (PC) (P< 0.05), and improved the intestinal antioxidant capacity by enhancing the mRNA levels of Cu/Zn superoxide dismutase (SOD1), catalase (CAT), glutathione-S-transferase (GST), glutathione peroxidase (GPx), glutathione reductase (GR), and γ-glutamyl cysteine ligase catalytic subunit (GCLC) (P< 0.05); (4) increased nuclear NFE2-related factor 2 (Nrf2) mRNA and protein levels (P< 0.05); (5) reduced intestinal DNA fragments, and down-regulated the mRNA levels of 78 KD glucose regulated protein (GRP78), activating transcription factor 6 (ATF6), inositol requiring enzyme-1 (IRE1), protein kinase RNA-dependent-like endoplasmic reticulum kinase (PERK), eukaryotic initiation factor 2α (eIF2α), activating transcription factor 4 (ATF4), C/EBP homologous protein (CHOP), cysteine-aspartic protease 9 (caspase 9), and cysteine-aspartic protease 3 (caspase 3), while up-regulating that of B-cell lymphoma-2 (Bcl2) (P< 0.05); (6) decreased phosphorylation of PERK (p-PERK) and phosphorylation of eIF2α (p-eIF2α), caspase 9, and caspase 3 protein levels. Conclusion: Dietary optimal Val level enhanced the growth performance and the intestinal antioxidant capacity via the Nrf2 signaling pathway after infection with A. veronii. Furthermore, this study is the first to demonstrate the role of Val in reducing ERS-mediated apoptosis through modulation of the PERK/eIF2α signaling pathway. Finally, Quadratic regression analysis of the SGR indicated that the dietary Val requirement of largemouth bass (37.99-162.92 g) was 21.50 g/kg diet, corresponding to 46.64 g/kg protein.

Abstract:We investigated the effects of dietary tannic acid (TA) supplementation of a high-carbohydrate diet on growth, feed utilization, whole-body proximate composition, serum biochemical indicators, antioxidant capacity, digestive enzyme activity, and liver and intestinal health of juvenile largemouth bass, Micropterus salmoides (initial mean weight: 8.08 ± 0.08 g). Five diets were prepared, including a positive control (dietary carbohydrate level, 16%, LC0), a negative control (dietary carbohydrate level, 21%, HC0), and three TA-supplementation diets based on the negative control diet with TA addition at 200, 400, and 800 mg/kg, respectively. After 8 weeks of feeding, the results showed that compared with the LC0 diet, 400-800 mg/kg dietary TA significantly improved the survival rate of largemouth bass (P < 0.05) while significantly reducing its weight-gain rate and specific growth rate (P < 0.05). Compared with the HC0 diet, 400 mg/kg dietary TA significantly increased serum catalase activity (P < 0.05), and significantly decreased serum malondialdehyde, liver glycogen, lightness (L ∗), and yellowness (b ∗) (P < 0.05). Moreover, compared with the HC0 diet, 200-400 mg/kg dietary TA effectively improved the vacuolation of hepatocytes caused by the high-carbohydrate diet and reduced the occurrence of intestinal epithelial cell vacuolation and necrosis. In turn, 800 mg/kg dietary TA significantly inhibited protease activity in the pyloric caecum and intestine (P < 0.05). In conclusion, dietary supplementation with TA inhibited protease activity, which resulted in decreased growth performance in largemouth bass. However, it was also found that 200-400 mg/kg TA enhanced the antioxidant capacity of largemouth bass in the case of the high-carbohydrate diet, reduced liver glycogen levels, and improved liver and intestinal health. Finally, it should be noted that, when the dietary TA level exceeded 800 mg/kg, TA appeared to play a pro-oxidation role in the liver, which may cause oxidative stress in the liver.
Dietary berberine alleviates high carbohydrate diet-induced intestinal damages and improves lipid metabolism in largemouth bass (Micropterus salmoides)

Abstract:High carbohydrate diet (HCD) causes metabolism disorder and intestinal damages in aquaculture fish. Berberine has been applied to improve obesity, diabetes and NAFLD. However, whether berberine contributes to the alleviation of HCD-induced intestinal damages in aquaculture fish is still unclear. Here we investigated the effects and mechanism of berberine on HCD-induced intestinal damages in largemouth bass ( Micropterus salmoides ). We found dietary berberine (50 mg/kg) improved the physical indexes (VSI and HSI) without affecting the growth performance and survival rate of largemouth bass. Importantly, the results showed that dietary berberine reduced the HCD-induced tissue damages and repaired the barrier in the intestine of largemouth bass. We observed dietary berberine significantly suppressed HCD-induced intestinal apoptosis rate (from 31.21 to 8.35%) and the activity level of Caspase3/9 ( P < 0.05) by alleviating the inflammation ( il1 β, il8 , tgf β, and IL-6, P < 0.05) and ER stress ( atf6 , xbp1 , perk , eif2 α, chopa , chopb , and BIP, P < 0.05) in largemouth bass. Further results showed that dietary berberine declined the HCD-induced excessive lipogenesis (oil red O area, TG content, acaca , fasn , scd , ppar γ, and srebp1 , P < 0.05) and promoted the lipolysis ( hsl , lpl , cpt1a , and cpt2 , P < 0.05) via activating adenosine monophosphate-activated protein kinase (AMPK, P < 0.05) and inhibiting sterol regulatory element-binding protein 1 (SREBP1, P < 0.05) in the intestine of largemouth bass. Besides, we also found that dietary berberine significantly promoted the hepatic lipid catabolism ( hsl , lpl , cpt1a , and cpt2 , P < 0.05) and glycolysis ( pk and ira , P < 0.05) to reduce the systematic lipid deposition in largemouth bass fed with HCD. Therefore, we elucidated that 50 mg/kg dietary berberine alleviated HCD-induced intestinal damages and improved AMPK/SREBP1-mediated lipid metabolism in largemouth bass, and evaluated the feasibility for berberine as an aquafeed additive to enhance the intestinal function of aquaculture species.

Abstract:Yinchenhao Decoction (YD), a Chinese herbal medicine, has been traditionally used for treatment of metabolic liver diseases. A 10-week feeding trail was carried out to examine the effects of YD supplementation in a high carbohydrate diet (HCD) on liver histopathology, immune response, disease resistance, and expression of genes associated with endoplasmic reticulum stress, autophagy, apoptosis, necroptosis and inflammation in juvenile largemouth. A diet containing 9% carbohydrate was used as a low carbohydrate diet (LCD), and a HCD was formulated to contain 18% carbohydrate and supplemented with 0, 0.5, 1, 2 or 4% YD (HCD, HCD+0.5YD, HCD+1YD, HCD+2YD and HCD+4YD). Triplicate groups of fish (5.6 ± 0.2 g) were feed the test diets to visual satiety for 10 weeks. The highest survival rate after Nocardia seriolae challenge was recorded for the HCD+4YD group. YD application led to reduced ACP, AKP, AST and ALT activities. HCD-induced cells swelling, ruptured cell membrane, migrated nuclei and increasing inflammatory cells in hepatocytes were mitigated by YD addition. Moreover, YD decreased the expressions of pro-inflammation genes (TNF-α, IL-1β, IL-8, hepcidin1, NF-κB, COX2, CD80 and CD83) and increased the mRNA levels of anti-inflammation genes (IL-10 and IKBα). The mode of liver cell death was preferably changed to programed apoptosis rather than uncontrolled necroptosis by application of YD in HCD. Furthermore, the expression of UPR genes (IRE1, Eif2α, ATF6, XBP1 and GRP78/Bip) and autophagy genes (LC3-2, BNIP3 and P62) was increased by YD supplementation. In summary, our results demonstrated that YD addition in HCD enhances UPR, autophagy and programed apoptosis maintaining the homeostasis, and decreases uncontrolled necroptosis and inflammation, ultimately leading to improved immune response in largemouth bass.

Abstract:A 60-d feeding trial was conducted to explore the potential regulatory effects of dietary Clostridium butyricum cultures (CBC) supplementation in high-carbohydrate diet (HCD) on carbohydrate utilisation, antioxidant capacity and intestinal microbiota of largemouth bass. Triplicate groups of largemouth bass (average weight 35·03 ± 0·04 g), with a destiny of twenty-eight individuals per tank, were fed low-carbohydrate diet and HCD supplemented with different concentration of CBC (0 %, 0·25 %, 0·50 % and 1·00 %). The results showed that dietary CBC inclusion alleviated the hepatic glycogen accumulation induced by HCD intake. Additionally, the expression of hepatic ampkα1 and insulin signaling pathway-related genes (ira, irb, irs, p13kr1 and akt1) increased linearly with dietary CBC inclusion, which might be associated with the activation of glycolysis-related genes (gk, pfkl and pk). Meanwhile, the expression of intestinal SCFA transport-related genes (ffar3 and mct1) was significantly increased with dietary CBC inclusion. In addition, the hepatic antioxidant capacity was improved with dietary CBC supplementation, as evidenced by linear decrease in malondialdehyde concentration and expression of keap1, and linear increase in antioxidant enzyme activities (total antioxidative capacity, total superoxide dismutase and catalase) and expression of antioxidant enzyme-related genes (nrf2, sod1, sod2 and cat). The analysis of bacterial 16S rRNA V3-4 region indicated that dietary CBC inclusion significantly reduced the enrichment of Firmicutes and potential pathogenic bacteria genus Mycoplasma but significantly elevated the relative abundance of Fusobacteria and Cetobacterium. In summary, dietary CBC inclusion improved carbohydrate utilization, antioxidant capacity and intestinal microbiota of largemouth bass fed HCD.

Abstract:This study is aimed at identifying the effects of dietary fiber on gut health, as well as the association between that understanding and fiber consumption in fish. A total of 300 juvenile largemouth bass (micropterus salmoides, initial average weight: 15.38 ± 0.16g) were randomly divided into three treatment groups (4 replicates per group). Fish were fed with isoproteic and isolipidic diets containing 0% (low fiber, LF), 4% (moderate fiber, MF) and 8% (high fiber, HF) soybean fiber, respectively. The intestine and intestinal content of test fish per treatment group after 56 days of treatment were sampled. The results showed that the anterior intestinal sections had normal histological architecture, and no considerable damage or inflammation was observed in any histological section from all subjects examined. Curiously, fish fed the MF diet had better histological alterations than the other treatments. Meanwhile, the intestinal antioxidant capacity in the MF group was significantly promoted when compared to the other groups, as well as up-regulated expression of antioxidant-related genes including sod, cat and gpx with increasing dietary fiber concentrations. Importantly, the administrations of MF diet remarkably elevated largemouth bass innate immune parameters include intestinal inducible nitric oxide synthase (iNOS) activity, nitric oxide (NO) and total protein content. Similarly, dietary administrations of fiber down-regulated notablely the expression of pro-inflammatory cytokines including IL-8, IL-1β and TNFα, whereas up-regulated tolerogenic cytokine IL-10 and TGF-β1 mRNA levels. In addition, dietary fibers also modulated the community structure of the intestinal microbiota by significantly altering bacterial diversity. Dietary supplemental fibers regulated intestinal microbiota in largemouth bass, characterized by a reduced abundance of Fusobacteria along with increased abundances of Proteobacteria and Firmicutes. Taken together, the present results suggested that moderate fiber supplementation was beneficial to promoting intestinal health status of fish through antioxidant and anti-inflammatory effects, which could be at least partially responsible by the modulation of gut microbial composition.

Abstract:A 28 day feeding trial was conducted to investigate the growth performance, immune response and intestinal microbiota of laminarin (LAM) supplemented diets in juvenile largemouth bass (Micropterus salmoides). Four hundred and eighty fish (initial average weight: 0.72 ± 0.04 g) were randomly divided into four groups (40 fish per tank with three replicates in each group) Four diets were prepared with LAM supplementation at the doses of 0 (control), 5 g Kg−1 (LL), 10 g Kg−1 (ML) and 15 g Kg−1 (HL), respectively. No significant difference in the specific growth rate (SGR) and hepatosomatic index (HSI) was observed in fish among the four groups, or in the lipid and ash content of fish flesh. In addition, fish in the LL group exhibited much higher antioxidant capacity (p 0.05). Dietary LAM at the dose of 5 g Kg−1 inhibited the transcription of interleukin-1β (il-1β) and tumor necrosis factor-α (tnf-α), while promoting the expression of transforming growth factor-β (tgf-β) in fish intestine. Moreover, the beneficial intestinal bacteria Bacteroide, Comamonas and Mycoplasma abundance significantly increased in fish from the LL group, while the content of opportunistic pathogens Plesiomonas, Aeromonas and Brevinema in fish of the HL group was substantially higher than the control group. Overall, the appropriate dose of supplemented LAM in the diet was 5 g Kg−1, while an excessive supplementation of LAM in the diet led to microbial community instability in largemouth bass.

Abstract:This research revealed that low-salinity environments have positive effects on largemouth bass (Micropterus salmoides), with the study finding that low-salinity levels can have beneficial impacts on this species. A 8-week experiment was conducted using four salinity gradients S0, S1, S2, S3 (0, 2, 4, 6 psu), and biochemical indicators and 16S rRNA sequencing were analyzed. Results showed relatively good growth performance at S0 and S1, and no significant antioxidant stress response at S3. Na+–K+–ATPase activity was significantly lower in the S2 group. The diversity showed no significant differences between the treatment groups. However, with increasing salinity, the proportion of potentially pathogenic bacteria in freshwater decreased significantly while that in the seawater increased. Indicator species analysis showed a significantly higher abundance of positive microbial species in the 4 psu group. In conclusion, although growth performance decreased at 4 psu, antioxidant damage was relatively small and disease risk was significantly reduced, indicating a positive effect on the largemouth bass.

Abstract:Zinc is essential for normal growth and reproduction in all animals and plays a crucial role in many biological processes. The present study aimed to compare the intervention effects of zinc on intestinal health in a high lipid diet or high starch diet. Seven iso-nitrogenous (∼520 g kg-1) diets were formulated containing a positive control diet (115 g kg-1 lipid + 115 g kg-1 starch + 20 mg kg-1 Zn), three high starch diets (HS, 166 g kg-1 starch) and three high lipid diets (HL, 182 g kg-1 lipid), with 0 (HS-LZn, HL-LZn), 20 (HS-MZn, HL-MZn) and 150 (HS-HZn, HL-HZn) mg kg-1 Zn being supplemented. High starch diet and high lipid diet promoted feed efficiency, as evidenced by the lower feed conversion ratio. Three-way factorial ANOVA analysis showed high starch diet (166 g kg-1) significantly decreased final body weight and weight gain compared to the normal starch level (115 g kg-1). Diamine oxidase in serum significantly increased in diets HS-LZn and HL-LZn. In addition, distal intestinal mucosal fold damage and inflammatory infiltration were observed in the HS-LZn, HS-HZn, HL-LZn and HL-HZn groups. Fish fed HL diets (HL-LZn, HL-MZn, HL-HZn) showed lower expressions of claudin 5 and claudin 34, and higher IgD and IgM. Diets HL-LZn and HL-MZn significantly up-regulated C4 and C7. Proinflammatory cytokines including il8, il1β and tnfα significantly up-regulated in diet HL-LZn, even higher than the HS-LZn. Intestinal microbial composition indicated the abundance of Cetobacterium in HL-LZn was significantly higher than the control and HL-MZn diets. Similarly, LEfSe showed that Cetobacterium (P = 0.039) significantly enriched in the HL-LZn group. This study clarified high energy diet induced intestinal damage, which can be alleviated by zinc.
