Abstract:Circulating microRNAs (miRNAs) are effective cancer biomarkers that are highly stable in body fluids enabling noninvasive detection. Portable, low-cost and efficient biosensing systems is crucial for early on-site diagnosis, especially in resource-limited settings. Present work deals with the development of a smartphone-based biosensing system for electrochemical detection of circulating microRNA-21 (miR-21) biomarker in saliva as a proof-of-concept. The smartphone-based biosensing system composed of a disposable screen-printed biosensor modified with reduced graphene oxide/gold (rGO/Au) composite, a circuit board for detection and a Bluetoothenabled smartphone equipped with specially designed Android application. The hybridization between miR-21 target and ssDNA probe on the rGO/Au-modified electrode resulted in the decrease in peak current with increase in miR-21 target concentration. The smartphone-based biosensing system showed comparable performance with commercial electrochemical workstation, for miR-21 detection in the concentration range of 1 x 10(-4) M to 1 x 10(-1)(2) M with correlation coefficient (r(2)) of 0.99. In addition to good selectivity in discriminating mismatched and non-target sequences, the biosensing system also showed acceptable recovery rate ranging from 96.2%-107.2 % in spiked artificial saliva. The smartphone-based biosensing system is envisioned to be able to realize rapid and highly sensitive detection of circulating miRNA biomarker in body fluid for point-of-care testing in remote areas.

Abstract:MicroRNA-21 (miRNA-21) is a significant biomarker for the development and progression of diverse cancers but is present in relatively low concentrations. Detecting such low-abundance molecules accurately can be challenging, especially in early-stage cancers where the concentration may be even lower. Herein, a self-calibration biosensing platform based on 3D novel MNPs-IL-rGO-AuNPs nanocomposites was successfully established for the ultrasensitive detection of miRNA-21. Duplex-specific nuclease (DSN) was introduced to recognize perfectly matched duplexes and trigger target recycling, enhancing the specificity and sensitivity of the biosensor. DSN-assisted target recycling, in conjunction with magnetic separation enrichment and high-performance MNPs-IL-rGO-AuNPs, collectively formed a multiple-signal amplification strategy. The obtained biosensor could output dual signals in both electrochemical and fluorescent modes, enabling self-correcting detection to enhance the accuracy. The obtained dual-mode biosensor prepared exhibited a wide detection range from 5 fM to 100 nM with a remarkably low LOD of 1.601 fM. It accomplished the sensitive evaluation of miRNA-21 in total RNA extracted from various human cancer cell lines and normal cell lines. Additionally, the greatly satisfactory outcomes in the analysis of human serum samples suggested that the proposed biosensor was a powerful screening candidate in early clinical diagnosis of cancer.

Abstract:MicroRNA-21 (miRNA-21) is a class of small non-coding RNAs that play a crucial role in the regulation of post- transcriptional gene expression. By leveraging the principle of base complementary pairing, the detection of miRNA-21 can be achieved through its analogue, MicroDNA-21 (miDNA-21), which possesses an identical sequence. Utilizing screen-printed carbon electrodes (SPCE), a conducting polymer (polypyrrole, Ppy), and metal nanocomposites (gold-platinum nanoparticles, Au@Pt NPs) were sequentially electrodeposited to create an electrochemical biosensor for the detection of miDNA-21. This biosensor employed ferrocyanide/ferricyanide (Fe (CN) 6 3-/4- ) as the electrochemical signaling probe, and its structure was elucidated via field emission scanning electron microscopy (FESEM) and Energy Dispersive Spectrometer (EDS). Quantitative detection was facilitated through differential pulse voltammetry (DPV), yielding a concentration range and linear fitting equation of 10.00-7.00 x 10.00- 14 mol/L: Delta I = 0.5143 lgc + 9.191, R2 = 0.9986. The limit of detection (LOD) was determined to be 2.90 x 10-15 mol/L, with a sensitivity of 9.42 mu A/mu mol/L & sdot;cm2. Subsequent performance assessments confirmed the sensor's excellent selectivity, reproducibility, and stability. Evaluation using the standard addition method, with normal human serum as the matrix, yielded recoveries ranging from 98.97 % to 102.70 %, and relative standard deviation (RSD) values below 1.00%. These results demonstrate the method's viability for practical application in detecting miRNA in human serum.

Abstract:AIM to explore Hong Kong Chinese women's experiences and perceptions of elective caesarean section performed under regional anaesthesia. DESIGN an exploratory study using a qualitative approach. Data were collected by tape-recorded in-depth interviews two to five days after caesarean section. PARTICIPANTS a purposive sample of 18 Hong Kong Chinese women having an uncomplicated, elective caesarean section under successful regional anaesthesia. SETTING a post-caesarean ward of a University affiliated District General Hospital in Hong Kong. KEY FINDINGS the three categories relating to the overall theme of Consciousness during Surgery were: Interacting with others, Experience of birthing, and Awareness of the environment. IMPLICATIONS FOR PRACTICE the experiences of women, related to the general theme and categories cited above, affected their feelings of security and fulfillment. On the whole, women were satisfied with the regional anaesthesia, preferring to remain conscious throughout the surgery. They appreciated being able to interact with others and listening to music, but needed information regarding what was happening. Many were distressed by the sight and noise of surgical instruments, the narrowness of the operating table, the operating theatre lamps and the coldness of the theatre. Recommendations for practice are made relating to these issues.
An Ultrasensitive Electrochemical Biosensor Integrated by Nicking Endonuclease-Assisted Primer Exchange Reaction Cascade Amplification and DNA Nanosphere-Mediated Electrochemical Signal-Enhanced System for MicroRNA Detection

Abstract:Specific and sensitive microRNAs (miRNAs) detection is essential to early cancer diagnosis. The development of these technologies including functional nuclease-mediated target amplification and DNA nanotechnology possesses tremendous potential for the high-performance detection of miRNAs in the accurate diagnosis of disease. In this study, we have established an ultrasensitive electrochemical biosensor by combining nicking endonuclease-assisted primer exchange reaction (PER) cascade amplification with a DNA nanosphere (DNS)-mediated electrochemical signal-enhanced system for the detection of miRNA-21 (miR-21). The cascade amplification is initiated by a nicking endonuclease that can cleave specific DNA substrates and highly amplify translation of the target to single-stranded DNA fragments (sDNA). Then, the PER cascade is powered by strand-displacing polymerase and generates a large amount of nascent single stranded connector DNA (cDNA) via sDNA triggering of the dumbbell probe (DP), thus achieving the cascade amplification of the target. Finally, the DNS loaded with plenty of electroactive substances can be captured on the electrode via cDNA for further enhancing the electrochemical signal and highly sensitive detection of miR-21. The proposed electrochemical biosensor exhibits a wide detection range of 1 aM to 0.1 nM and a low detection limit of 0.58 aM. The excellent selectivity allows the biosensor to discriminate miR-21 from other miRNAs, even the one base-mismatched sequence. Moreover, the practicability of the biosensor is investigated by analyzing miR-21 in human serum and cancer cell lysate. Therefore, our proposed nicking endonuclease-assisted PER cascade amplification strategy provides a powerful platform for the early detection of miRNA-related disease and molecular diagnosis.

Abstract:Because microRNAs (miRNAs) are biological indicators for the diagnosis, treatment, and monitoring of tumors, cancers, and other diseases, it is significant to develop a rapid, sensitive, and reliable miRNA detection platform. In this study, based on miRNA-21 detection, DNA-a with a 3′ end overhang and Texas Red fluorophore-labeled 5′ end was designed, which reacts with miRNA-21 and hybridizes with exonuclease III (Exo III), where the part connected to miRNA-21 is hydrolyzed, leaving a-DNA. At the same time, miRNA-21 is released to participate in the following reaction, to achieve cyclic amplification. a-DNA reacts with DNA-b conjugated to gold nanoparticles to achieve fluorescence quenching, with the quenching value denoted as F; additionally, after adding DNA-d and linked streptavidin immunomagnetic beads (SIBs), fluorescence recovery was achieved using DNA-c, with the recovered fluorescence recorded as F0. By comparing the difference in the fluorescence (F0 − F) between the two experiments, the amount of DNA-a hydrolyzed to produce a-DNA was established to determine the target miRNA-21 content. Under optimized conditions, by comparing the changes in the fluorescence signal, the developed strategy shows good sensitivity and repeatability, with a detection limit of 18 pM, good discriminative ability and selectivity, and promise for the early diagnosis of breast and intestinal cancers.
Toehold-mediated strand displacement coupled with single nanoparticle dark-field microscopy imaging for ultrasensitive biosensing

Abstract:Highly sensitive detection of biomarkers is essential for disease prevention and early diagnosis. Herein, a highly sensitive strategy was proposed for microRNA-21 (miRNA-21) detection by the incorporation of programmable toehold-mediated strand displacement (TMSD) and dark-field microscopy imaging. Firstly, efficient and specific TSMD was carried out via hybridization between the substrate strand (Sub) and two short probe strands (P1, P2). Then, miRNA-21 could specifically hybridize with Sub due to the toehold that existed on its tail, which triggered the amplification with the help of the assist strands, and forming a large number of Sub-assist double-stranded DNA (dsDNA). This process realized the targeted highly specific recognition of miRNA-21 and the amplification of the trace target to high-output dsDNA. Additionally, as glucose oxidase (Gox) was modified on the end of the assist strands in advance, hydrogen peroxide was generated after adding glucose to the system, which further etched gold-silver core-shell nanocubes (Au@Ag NCs). As a result, the size of Au@Ag NCs decreased and the scattering intensity reduced simultaneously. The scattering intensity reduction value of Au@Ag NCs has a linear relationship with miRNA-21 concentration in the range of 1.0 to 100.0 fM with a limit of detection of 1.0 fM. Finally, the proposed method has been successfully demonstrated for the determination of miRNA-21 in lung cancer cell A549 lysate.

Abstract:Herein, a novel ratiometric electrochemical biosensor with methylene blue (MB) as the only one signal tag was proposed for highly reliable and ultrasensitive detection of microRNA-21 (miRNA-21) under the assistance of an intelligent target-induced dual signal amplification (T-DSA). First, a small amount of target miRNA-21 could produce abundant mimic targets DNA S1 and Zn2+ through target-induced recycle and acid dissolution, respectively. Then, S1 triggered rolling circle amplification (RCA) to generate functional DNA nanospheres (DSP) encoded by DNAzyme and substrate sequence for loading numerous signal tag MB with a remarkable electrochemical signal (signal on), and the Zn2+ cofactor mediated the nonviolent DNAzyme-catalyzed cleavage of DSP to sharply release MB with obviously reduced electrochemical responses (signal off). Impressively, our strategy could controllably load and release the only signal tag MB through the well-designed DSP to effectively avoid the false positive responses caused by the non-ideal upright state of DNA probes connected to electrodes in traditional distance-dependent signal adjustment ratiometric strategies with two different signal tags. Meanwhile, with the aid of innovative T-DSA recycle and RCA-produced functional DSP, the detection sensitivity of this sensing platform was significantly improved. Abstract:MicroRNAs (miRNAs) have emerged as essential biomarkers for disease diagnosis, and several techniques are available to determine type 2 diabetes (T2D) relevant miRNAs. However, detecting circulating miRNAs can be challenging due to their small size, low abundance, and high sequence similarity, often requiring sensitive detection approaches combined with additional amplification processes. Laser-induced fluorescence (LIF) is a classic analytical method suitable for sensitively detecting trace amounts of nucleotide acid. Duplex-specific nuclease (DSN)-mediated amplification recently gained attention due to its catalytic activity based on target recycling, demonstrating a promising approach for miRNA amplification. This work developed a novel N-annulated perylene fluorescent dye to create a biosensor to analyze the miRNA (miR-223) relevant to T2D. The amine-reactive fluorescent dye assists the amidation reaction for nucleotide labeling, giving the oligonucleotide probe a high fluorescence quantum yield and sufficient water solubility. By combining the locked nucleic acid (LNA) modified oligonucleotide fluorescent probe to enhance the stability of LNA/RNA hybrids, thereby improving the DSN-mediated target miR-223 recycling for signal amplification, the proposed biosensor can highly selectively determine miR-223 with a limit of detection (LOD, S/N = 3) of 9.5 pM. When applied to real-world samples, the biosensor demonstrated its potential to distinguish between T2D patients and healthy controls.
Enzyme-Assisted Colorimetric Salivary miR-21 Detection by Functional Gold Nanoparticles

Abstract:Nano, Ahead of Print. miRNA-21 (miR-21) is a potential biomarker for the monitoring of diseases through its expression levels. Simple, portable and sensitive miR-21 detection of is advantageous for health monitoring in Point of Care Testing (POCT). Gold nanoparticles (AuNP) as excellent colorimetric sensors are widely used in the POCT. However, their low sensitivity is a limitation of their clinical use. Herein, we developed an AuNPs-based miR-21 assay with enzyme-assisted amplification reaction to construct the colorimetric platform capable of detecting as low as 0.1[math]nM. In this platform, template ssDNA as a signal molecule could hybridize with ssDNA-modified AuNPs to generate the color reaction. The target miR-21 specifically hybridized to the template ssDNA, which was then cleaved by exonuclease III (Exo III) to release the target miR-21. As a trigger, miR-21 catalyzed the degradation of the template ssDNA to amplify the signal by Exo III. By hybridizing miR-21 and template ssDNA in the presence of Exo III, R-21 induced a significant decrease in the level of template ssDNA to reduce the aggregation of AuNPs. There is a clear color difference in the presence/absence of miR-21 in the assay. In this assay, the optimal concentration of templated ssDNA and Exo III were 100[math]nM and 0.06[math]U/[math]L in a 100[math][math]L detection system. The LoD for UV–Vis spectrum and colorimetric reaction were 0.1[math]nM and 0.5[math]nM, respectively. The detection system has good selectivity and can be used to detect miR-21 in the simulated saliva. It has great potential for application in biomedical research as well as in clinical diagnostics.

A tutorial on using hidden Markov models for phoneme recognition

Abstract:A phoneme is a contrastive unit in the sound system of a particular language that helps us distinguish between meanings of words from a set of similar sounds corresponding to it pronounced in one or more ways, depending on the configuration of the articulators like air flow, teeth, tongue, lip and vocal chord movements. In this paper we explain in detail the process of phoneme recognition using hidden Markov models (HMM), implemented with HTK (hidden Markov model toolkit) developed by Cambridge University. An HMM is a statistical model that describes a probability distribution over a number of possible sequences. HTK is widely used in speech recognition research worldwide and aims at manipulating and building hidden Markov models. This paper follows a 3-state HMM to give an insight into the training and testing phases of phoneme recognition. We were able to achieve up to 100% accuracy in recognizing the phonemes, and thus strongly believe that a 3-state HMM ideally suits our requirements. We also explore the features of various accents, evaluating their cepstral coefficients and perform time and frequency domain analysis.

Abstract:MicroRNAs (miRNAs) play an important regulatory role in tumor progression and have been emerging as promising biomarkers for early diagnosis of breast cancer. Herein, we developed a dual-signal surface-enhanced Raman scattering (SERS) biosensor for the quantitative detection of miRNA 21 (miR-21) by utilizing a tightly ordered spindle-shaped gold (SAu) array with 5, 5'-dithio-bis-(2-nitrobenzoic acid) (DTNB) molecule as an internal standard. Meanwhile, 6-carboxyl-Xrhodamine (ROX) labeled hairpin DNA H1 was linked to the SAu array by Au–S bonds generating another SERS signal. In the presence of miR-21, H1 was turned on with a decrease in SERS signal of ROX, while the SERS signal of DTNB remained stable. The SAu array as substrate provides a large area of hot spots, and the dual-signal can avoid background interference and achieve signal self-calibration. By the dual-signal SERS biosensor, target miR-21 could be detected over a broad linear range (0.1−10 5 pM) with a low detection limit (0.046 pM) without signal amplification strategy. Thanks to the SAu array and dual-signal output, the sensor showed high sensitivity, good signal reproducibility, excellent specificity, and high detection accuracy. When this sensor was applied to the miR-21 assay in human serum samples, its performance was comparable to quantitative reverse transcription polymerase chain reaction (qRT-PCR), indicating promising applications of the developed sensor for cancer diagnostics.

Abstract:A highly sensitive surface-enhanced Raman scattering (SERS) biosensor has been developed for the detection of microRNA-21 (miR-21) using an isothermal enzyme-free cascade amplification method involving catalytic hairpin assembly (CHA) and hybridization chain reaction (HCR). The CHA reaction is triggered by the target miR-21, which causes hairpin DNA (C1 and C2) to self-assemble into CHA products. After AgNPs@Capture captures the resulting CHA product, the HCR reaction is started, forming long-stranded DNA on the surface of AgNPs. A strong SERS signal is generated due to the presence of a large amount of the Raman reporter methylene blue (MB) in the vicinity of the SERS "hot spot" on the surface of AgNPs. The monitoring of the SERS signal changes of MB allows for the highly sensitive and specific detection of miR-21. In optimal conditions, the biosensor exhibits a satisfactory linear range and a low detection limit for miR-21 of 42.3 fM. Additionally, this SERS biosensor shows outstanding selectivity and reproducibility. The application of this methodology to clinical blood samples allows for the differentiation of cancer patients from healthy controls. As a result, the CHA-HCR amplification strategy used in this SERS biosensor could be a useful tool for miRNA detection and early cancer screening.

Abstract:Abstract A simple carbon nanodot–based electrogenerated chemiluminescence biosensor is described for sensitive and selective detection of microRNA-21 (miRNA-21), a biomarker of several pathologies including cardiovascular diseases (CVDs). The photoluminescent carbon nanodots (CNDs) were obtained using a new synthesis method, simply by treating tiger nut milk in a microwave reactor. The synthesis is environmentally friendly, simple, and efficient. The optical properties and morphological characteristics of the CNDs were exhaustively investigated, confirming that they have oxygen and nitrogen functional groups on their surfaces and exhibit excitation-dependent fluorescence emission, as well as photostability. They act as co-reactant agents in the anodic electrochemiluminescence (ECL) of [Ru(bpy) 3 ] 2+ , producing different signals for the probe (single-stranded DNA) and the hybridized target (double-stranded DNA). These results paved the way for the development of a sensitive ECL biosensor for the detection of miRNA-21. This was developed by immobilization of a thiolated oligonucleotide, fully complementary to the miRNA-21 sequence, on the disposable gold electrode. The target miRNA-21 was hybridized with the probe on the electrode surface, and the hybridization was detected by the enhancement of the [Ru(bpy) 3 ] 2+ /DNA ECL signal using CNDs. The biosensor shows a linear response to miRNA-21 concentration up to 100.0 pM with a detection limit of 0.721 fM. The method does not require complex labeling steps, and has a rapid response. It was successfully used to detect miRNA-21 directly in serum samples from heart failure patients without previous RNA extraction neither amplification process. Graphical abstract

Abstract:MicroRNA-21 (miR-21) is a promising diagnostic biomarker for breast cancer screening and disease progression, thus the method for the sensitive and selective detection of miR-21 is vital to its clinical diagnosis. Herein, we develop a novel method to quantify miR-21 levels as low as attomolar sensitivity by a target-triggered triple isothermal cascade amplification (3TICA) strategy. An ingenious unimolecular DNA template with three functional parts has been designed: 5'-fragment as the miR-21 recognition unit, middle fragment as the miR-21 analogue amplification unit, and 3'-fragment as the 8-17 DNAzyme production unit. Triggered by miR-21 and accompanied by polymerase-nicking enzyme cascade, new miR-21 analogues autonomously generated for the successive re-triggering and cleavage process. Simultaneously, the 8-17 DNAzyme-contained sequence could be exponentially released and activated for the second cyclic cleavage toward a specific ribonucleotide (rA)-contained substrate, inducing a remarkably amplified generation of HRP-mimicking DNAzyme in the presence of hemin. Finally, the amperometric technique was used to record the catalytic reduction current of 3,3',5,5'-tetramethylbenzidine (TMB) in the presence of H2O2. The increase in the steady-state current was proportional with the increase of the miR-21 concentration from 1 aM to 100 pM. An ultra-low detection limit of 0.5 aM with an excellent selectivity for even discriminating differences between 1-base mismatched target and miR-21 was achieved. This simple and cost-effective 3TICA strategy is promising for the detection of any short oligonucleotides, simply by altering the target recognition unit in the template sequence.

Abstract:The development of a convenient and efficient assay using miRNA-21 as a lung cancer marker is of great importance for the early prevention of cancer. Herein, an electrochemical biosensor for the detection of miRNA-21 was successfully fabricated under blue light excitation using click chemistry and photocatalytic atom transfer radical polymerization (photo-ATRP). By using hairpin DNA as a recognition probe, the electrochemical sensor deposits numerous electroactive monomers (ferrocenylmethyl methacrylate) on the electrode surface under the reaction of photocatalyst (fluorescein) and pentamethyldiethylenetriamine, thereby achieving signal amplification. This biosensor is sensitive, precise and selective for miRNA-21, and is highly specific for RNAs with different base mismatches. Under optimal conditions, the biosensor showed a linear relationship in the range of 10 fM ∼1 nM (R 2 = 0.995), with a detection limit of 1.35 fM. Furthermore, the biosensor exhibits anti-interference performance when analyzing RNAs in serum samples. The biosensor is based on green chemistry and has the advantages of low cost, specificity and anti-interference ability, providing economic benefits while achieving detection objectives, which makes it highly promising for the analysis of complex samples.

Abstract:The latest in a series of speculations concerning the origins of HIV a Rolling Stone magazine article claiming that AIDS may have been introduced through the polio vaccines has touched off angry responses from vaccine experts including several who claim to have been misrepresented by the article. In an article entitled "The Origin of AIDS" found in the current issue of Rolling Stone free-lance writer Tom Curtis hypothesizes that AIDS first entered humans in Africa as a contaminant of an oral polio vaccine widely administered in the late 1950s a vaccine formulated at Philadelphias Wistar Institute by Hilary Koprowski. Curtis speculates that a still-undiscovered simian form of HIV may have contaminated Koprowskis polio vaccine developed from monkey kidney cell cultures. Some researchers say that the articles claim is not so far-fetched considering that simian viruses were found to contaminate some early polio vaccines. Most researchers however dismiss the theory as far too speculative arguing that no supporting evidence exists. Both the Wistar and the Center for Disease Control (CDC) have come out against the theory. Koprowski maintains that the existing data refutes Curtiss hypothesis since in preparing the vaccine he used wild macaques from the Philippines and India and no wild macaques have ever tested positive for a lentivirus. In response to Koprowskis claim Curtis quoted the chief of CDCs retrovirus branch Thomas Folks who offered a scenario in which the monkey kidney cells might have become infected. Folks however has angrily accuses Curtis of selective editing. Others interviewed by Curtis have also claimed to have been misrepresented. Despite the rejections the Wistar Institute has announced that it will form a committee to evaluate the theory.

Abstract:miRNAs regulate several life activities and they have emerged as biomarkers in cancer diagnosis. Pursuing highly sensitive sensing platforms for detecting miRNAs, signal amplification strategies are the most frequently utilized means. In this research, a click chemistry-mediated variable temperature amplification (CCVTA) biosensor was developed for the highly selective and sensitive detection of miRNA-21. Sensing depended on enzyme-free amplification and copper-free type click chemistry, which greatly simplified the pre-treatment procedure and the conditions for sensing. In addition, the amplification products were used to output two signals, which are electrochemical and colorimetric modes. The dual-mode biosensor respectively achieved a detection limits of 0.57 and 17.19 fmol/mL, as well as excellent selectivity, stability, and applicability. It can be expected that CCVTA-biosensor sparkle with attractive prospects in the sensing field.

Abstract:With the emergence of microRNA (miRNA) as a key player in early clinical disease diagnosis, development of rapidly sensitive and quantitative miRNA detection methods are imperative. Herein, a label-free SERS assay coupled with duplex-specific nuclease (DSN) signal amplification strategy was proposed for facilely ultrasensitive and quantitative analysis of miRNA-21. Firstly, magnetic beads assembled with excessive capture DNA were utilized to hybridize the target miRNA-21. These DNA-RNA heteroduplexes were cleaved by DSN to generate small nucleotide fragments into the supernatant and the miRNA-21 released and rehybridized another DNA, going to the next DSN cycle. Consequently, numerous of small nucleotide fragments of capture DNA were released from magnetic beads and the miRNA-21 signal was transferred and amplified by the SERS signals of total phosphate backbones which are abundant in nucleotide. Furthermore, iodide-modified Ag nanoparticles (AgINPs) was employed to generate a strong and reproducible SERS signal. The proposed method displayed excellent performance for miRNA-21 detection with the linear range from 0.33 fM to 3.3 pM, and a lower detection limit of 42 aM. Moreover, this strategy exhibited effectively base discrimination capability and was successfully applied for monitoring the expression levels of miRNA-21 in different cancer cell lines and human serum.
