Abstract:This study aimed to reveal the interaction and inhibitory mechanisms of tangeretin (TAN), nobiletin (NBT), and their acidic hydroxylated forms, 5-demethyltangeretin (5-DT) and 5-demethylnobiletin (5-DN) on porcine pancreatic lipase (PPL) using spectroscopic techniques and molecular dynamics (MD) simulation. PPL inhibition assay showed that the inhibitory activity of NBT (IC<sub>50</sub> value of 3.60 ± 0.19 μM) was superior to those of three polymethoxylated flavones (PMFs), indicating it may be related to the methoxy groups at the 3'-position in its molecular structure. Inhibition kinetic analyses demonstrated that the inhibition types of the 4 PMFs were consistent with the mixed inhibition model, which agreed well with the results from the ultraviolet-visible (UV-Vis) spectroscopy, Circular dichroism (CD), fluorescence spectroscopy, molecular docking, and MD simulation that PMFs could bind to the PPL catalytic site and non-catalytic site, affecting the normal spatial conformation of PPL and weakening its ability to decompose the substrate. All these findings suggest that PMFs are a kind of natural lipase inhibitors, and NBT has the potential as a lipase inhibition precursor because of its unique flavone skeleton structure.

Abstract:To expand the application of nobiletin (NOB) in semi-solid functional foods, bovine serum albumin (BSA)/carboxymethyl inulin (CMI) complexes-stabilized Pickering emulsion (BCPE) (φ oil = 60%, v/v) was fabricated, and the swallowing index and bioavailability of the NOB-loaded Pickering emulsion was evaluated. Confocal laser scanning microscope (CLSM) and cryo-scanning electron microscopy (cryo-SEM) images revealed that BSA/CMI complexes attached to the oil–water interface. NOB-loaded BCPE exhibited a viscoelastic and shear-thinning behavior. Fork drip test results suggested that the textural value of unloaded and NOB-loaded emulsions was International Dysphagia Diet Standardisation Initiative Level 4, which could be swallowed directly without chewing. The in vitro lipolysis model suggested that NOB had a faster digestive profile and a higher bioaccessibility in the BCPE than in the oil suspension. The in vivo rat model revealed that the oral bioavailability of NOB was increased by 2.07 folds in BCPE compared to its bioavailability in unformulated oil. Moreover, BCPE led to a higher plasma concentration of the major demethylated metabolite of NOB (4′-demethylnobiletin) than the unformulated oil. Accordingly, BCPE enhanced the oral bioavailability of NOB by improving bioaccessibility, absorption, and biotransformation.

Abstract:Nobiletin (NOB) is a naturally occurring compound, commonly found in citrus peel, that shows hepatoprotective and lipid-reducing effects. However, the lipid biomarkers and the potential improvement mechanisms have not been adequately explored. Therefore, we investigated the ameliorative effect and the molecular mechanism of NOB on NAFLD induced by a high-fat diet in mice. The results showed that supplementation with NOB over 12 weeks markedly improved glucose tolerance, serum lipid profiles, inflammatory factors, hepatic steatosis, and oxidative stress. These beneficial effects were mainly related to reduced levels of potential lipid biomarkers including free fatty acids, diacylglycerols, triacylglycerols, and cholesteryl esters according to hepatic lipidomic analysis. Twenty lipids, including DGs and phosphatidylcholines, were identified as potential lipid biomarkers. Furthermore, RT-qPCR and Western blot analysis indicated that NOB inhibited the expression of lipogenesis-related factors such as SREBP-1c, SCD-1, and FAS, and upregulated the expression of lipid oxidation (PPARα) and cholesterol conversion (LXRα, CYP7A1, and CYP27A1) genes as well as antioxidation-related factors (Nucl-Nrf2, NQO1, HO-1, and GCLC), indicating that NOB intake may reduce lipid biosynthesis and increase lipid consumption to improve hepatic steatosis and oxidative stress. This study is beneficial for understanding the ameliorative effects of NOB on NAFLD.

Abstract:Disturbance of the gut microbiota plays a critical role in the development of nonalcoholic fatty liver disease (NAFLD). Increasing evidence supports that natural products may serve as prebiotics to regulate the gut microbiota in the treatment of NAFLD. In the present study, the effect of nobiletin, a naturally occurring polymethoxyflavone, on NAFLD was evaluated, and metabolomics, 16S rRNA gene sequencing, and transcriptomics analysis were performed to determine the underlying mechanism of nobiletin, and the key bacteria and metabolites screened were confirmed by in vivo experiment. Nobiletin treatment could significantly reduce lipid accumulation in high-fat/high-sucrose diet-fed mice. 16S rRNA analysis demonstrated that nobiletin could reverse the dysbiosis of gut microbiota in NAFLD mice and nobiletin could regulate myristoleic acid metabolism, as revealed by untargeted metabolomics analysis. Treatment with the bacteria Allobaculum stercoricanis, Lactobacillus casei, or the metabolite myristoleic acid displayed a protective effect on liver lipid accumulation under metabolic stress. These results indicated that nobiletin might target gut microbiota and myristoleic acid metabolism to ameliorate NAFLD.

Abstract:Exploring the effect of bile salts on the properties of emulsion carriers containing hydrophobic bioactive compounds is particularly critical to understanding the stability and bioavailability of these hydrophobic bioactive compounds in the digestive process. In this study, the effects of bile salts on the stability and digestive characteristics of the ursolic acid (UA) self-stabilized water-in-oil (W/O) emulsion were investigated via static and dynamic (with or without enzyme) in vitro simulated digestive systems. The results showed that under the static system, the basic conditions had less interference, while the bile salts had a significant effect on the appearance and microstructure of the emulsion. The primary mechanism of emulsion instability is hydrophobic binding and depletion flocculation. Under the dynamic condition, it was found that the low concentrations of bile salts can promote the release amount and the rate of free fatty acids via displacement, while high concentrations of bile salts inhibit the decomposition of lipid, which may be related to the secondary coverage formed at the interface by the bile salts. These findings provide a theoretical basis for understanding the digestive behavior of the UA emulsion and its interaction with bile salts, which are conducive to developing and designing new emulsions to improve the bioaccessibility of UA.

Abstract:BACKGROUND The Pickering emulsion delivery technique is widely acknowledged for its efficacy in serving as a carrier that can encapsulate functional components effectively. Previous studies have shown significant differences in the stability of Pickering emulsions composed of different oil phases and in the bioaccessibility of the encapsulated functional ingredients. This study therefore investigated the effects of different carrier oils in the betulin self-stabilized water-in-oil (W/O) Pickering emulsion on the stability of the emulsion and bioaccessibility of betulin. RESULTS The results showed that the oil type was one of the main factors affecting the stability of the emulsion. Palm oil and coconut oil provided better storage stability and centrifugal stability due to the high saturated fatty acid content. The bioavailability of betulin correlated significantly with the composition and characteristics of fatty acids in carrier oils. Carrier oils rich in low-saturation long-chain fatty acids tended to release more free fatty acids (FFAs), thus forming larger and more mixed micelles with stronger swelling and dissolution ability, resulting in a relatively high bioaccessibility of betulin. In contrast, the bioaccessibility of betulin in the emulsion prepared by coconut oil (with high saturated fatty acid content) was relatively low (1.17%). CONCLUSION The results of this study indicate that selecting an appropriate carrier oil is important for the design of self-stabilized W/O Pickering emulsions to improve the bioaccessibility of betulin and other lipophilic bioactivities effectively. © 2024 Society of Chemical Industry.

Impact of molecular interactions between hydrophilic phytosterol glycosyl derivatives and bile salts on the micellar solubility of cholesterol

Abstract:Hydrophilic phytosterol glycosyl derivatives are synthetic phytosterol analogues by coupling with the glycosyl moiety to improve the water solubility and bioaccessibility of free phytosterols. The aim of this study is to clarify the molecular interaction of phytosterol glycosyl derivatives with bile salts and the consequent impact on cholesterol solubilization. Sharp nonlinear decrease in the micellar solubility of cholesterol and accompanying changes in particle size, zeta potential and microtopography of mixed micelles were observed when phytosterol glycosyl derivatives were introduced in cholesterol-loaded bile salt micelles. These results suggested that β-sitosterol glycosyl derivatives molecules indeed participated in the formation of mixed micelles. Further, nuclear magnetic resonance showed that the structural change of mixed micelles was caused by the insertion of β-sitosterol glycosyl derivatives via hydrogen bonds with sodium taurocholate, which resulted in the low cholesterol solubilization. Moreover, the hydrogen-bond interactions were apparently influenced by the glycosyl moiety of β-sitosterol glycosyl derivatives. These molecular mechanisms may contribute to the development of cholesterol-absorption inhibitors.
Nobiletin Prevents High-Fat Diet-Induced Dysregulation of Intestinal Lipid Metabolism and Attenuates Postprandial Lipemia

Abstract:Objective: Nobiletin is a dietary flavonoid that improves insulin resistance and atherosclerosis in mice with metabolic dysfunction. Dysregulation of intestinal lipoprotein metabolism contributes to atherogenesis. The objective of the study was to determine if nobiletin targets the intestine to improve metabolic dysregulation in both male and female mice. Approach and Results: Triglyceride-rich lipoprotein (TRL) secretion, intracellular triglyceride kinetics, and intestinal morphology were determined in male and female LDL (low-density lipoprotein) receptor knockout ( Ldlr –/– ), and male wild-type mice fed a standard laboratory diet or high-fat, high-cholesterol (HFHC) diet ± nobiletin using an olive oil gavage, radiotracers, and electron microscopy. Nobiletin attenuated postprandial TRL levels in plasma and enhanced TRL clearance. Nobiletin reduced fasting jejunal triglyceride accumulation through accelerated TRL secretion and lower jejunal fatty acid synthesis with no impact on fatty acid oxidation. Fasting-refeeding experiments revealed that nobiletin led to higher levels of phosphorylated AKT (protein kinase B) and FoxO1 (forkhead box O1) and normal Srebf1c expression indicating increased insulin sensitivity. Intestinal length and weight were diminished by HFHC feeding and restored by nobiletin. Both fasting and postprandial plasma GLP-1 (glucagon-like peptide-1; and likely GLP-2) were elevated in response to nobiletin. Treatment with a GLP-2 receptor antagonist, GLP-2(3-33), reduced villus length in HFHC-fed mice but did not impact TRL secretion in any diet group. In contrast to males, nobiletin did not improve postprandial lipid parameters in female mice. Conclusions: Nobiletin opposed the effects of the HFHC diet by normalizing intestinal de novo lipogenesis through improved insulin sensitivity. Nobiletin prevents postprandial lipemia because the enhanced TRL clearance more than compensates for increased TRL secretion.
Inhibitory Effect of Condensed Tannins from Banana Pulp on Cholesterol Esterase and Mechanisms of Interaction

Abstract:In the present study, the inhibitory effect of condensed tannins (CTs) on cholesterol esterase (CEase) was studied. The underlying mechanisms were evaluated by reaction kinetics, turbidity and particle size analyses, multispectroscopy methods, thermodynamics, and computer molecular simulations. CTs showed potent CEase inhibitory activity with an IC50 value of 64.19 μg/mL, and the CEase activity decreased with increasing CT content in a mixed-competitive manner, which was verified by molecular docking simulations. Fluorescence and UV-vis measurements revealed that complexes were formed from CEase and CTs by noncovalent interaction. Isothermal titration calorimetry indicated that the interaction between CEase and CTs occurred through hydrogen bonding and hydrophobic interactions. Circular dichroism analysis suggested that CTs inhibited the activity of CEase by altering the secondary structure of CEase. The inhibition of CTs on CEase in the gastrointestinal tract might be one mechanism for its cholesterol-lowering effect.

Abstract:An efficient and environmentally friendly method for the online screening of lipase inhibitors by capillary electrophoresis was established to investigate the inhibitory activity of citrus flavonoids. To maintain high lipase activity during the experiment, the type, concentration and pH value of the running buffer were optimized. At the same time, the enzymatic reaction time was optimized to ensure sufficient product formation. Under the optimized conditions, the Michaelis-Menten constant (Km) of lipase was determined to be 1.345 mM, and the half-maximal inhibitory concentration (IC50) and inhibition constant (Ki) of orlistat were measured to be 0.1059 and 0.0607 µM, respectively. Then, the developed method was applied to evaluate the inhibitory activity of citrus fruit extracts and showed that the extract of Tangerine Pith had the highest inhibitory activity, with an inhibition percentage of 40.67%. Additionally, molecular docking technology was used to validate these inhibition experiments, indicating that tangeretin, nobiletin, hesperetin and naringenin could affect the activity of lipase by forming hydrogen bonds with lipase. This method can be used as an effective method for the preliminary screening of lipase inhibitors from citrus fruits.

Abstract:Bile salts as two-dimension facial amphiphile effectively emulsify lipids at low concentrations to promote gut digestion. Hence, we select various bile salts sodium cholate (NaC), sodium glycocholate (NaGC) and sodium taurocholate (NaTC) to study the interfacial tension and rheological dilational modulus to illustrate the interfacial adsorption mechanism and interface film strength, which is closely related to emulsion stability. Unlike linear surfactant molecules, the interfacial behavior of bile salts contains into two stages. At the low concentration for the first stage, the rigid facial molecules adsorb at the lipid-water interface with the hydrophilic concave and hydrophobic convex surface. And the interfacial relaxation is mainly dominated by intermolecular interaction and diffusion exchange, resulting in that the adsorption layer is especially an elastic film. During the second stage, aggregates are gradually generated at the interface as the concentration increases, and the interfacial relaxation is mainly dominated by the rapid diffusion exchange between the adsorption film and the sublayer, leading to the faster formation of the adsorption film. The mechanism of facial amphiphile adsorbed at lipid-water interface indicates soft molecular membrane established at interfaces on account of intermolecular interaction, revealing that higher interfacial strength benefits emulsion stability. We also found that NaTC with excellent adsorption capacity can stabilize edible oil for 8 h to facilitate lipid digestion. Compared with linear surfactant, bile salts with great biocompatibility can form two-dimension adsorption layer to emulsify and stabilize lipids, which can be widely used in lipid delivery system.
Modulation of Lipid Digestion by Nanoarchitectonics of Complex Interfacial Structures with Tween 80 and Lecithin

Abstract:In recent years, there has been a growing interest in regulating lipid digestion through the construction of various interfacial structures. In the present work, a series of complex interfacial structures were designed by combining Tween 80 in the aqueous phase and lecithin in the oil phase at different concentration ratios. The emulsification properties, the roles in regulating lipid digestion, and the interfacial dilatational rheological properties of the composite emulsifying systems were characterized. The results showed that the combination of Tween 80 and lecithin at different ratios could effectively modulate the rate of lipid digestion. The polyoxyethylene chains of Tween 80 formed a network, that provided a spatial obstacle for the adsorption of bile salts and lipases. Thus, Tween 80 significantly delayed the lipid digestion. The introduction of lecithin gradually replaced Tween 80 molecules at the interface, thus providing space for the adsorption of bile salts and lipases. In addition, as the ratio of lecithin concentration to Tween 80 increased, lecithin gradually became the dominant factor in the interfacial properties. As a result, the rate of lipid digestion was accelerated. Therefore, by compounding different ratios of lecithin and Tween 80, a series of emulsions with different lipid digestion rates were obtained. This research provides a basis for rationally designing food emulsions according to specific needs.
Bile conjugation and its effect on in vitro lipolysis of emulsions

Abstract:Bile Salts (BS) are responsible for stimulating lipid digestion in our organism. Gut microbiota are responsible for the deconjugation process of primary conjugated to secondary unconjugated BS. We use two structurally distinct BS and characterize the rate of lipolysis as a compound parameter. A static in-vitro digestion model as well as meta-analysis of literature data has been performed to determine the most influential factors affecting the lipid digestion process. The results demonstrate that lipolysis of emulsions using conjugated BS (NaTC, FFA = 60.0 %, CMC in SIF = 5.58 mM, MSR of linoleic acid = 0.21, rate of adsorption = -0.057 mN/m.s) enhances the release of FFA compared to deconjugated BS (NaDC, FFA = 49.5 %, CMC in SIF = 2.49 mM, MSR of linoleic acid = 0.16 rate of adsorption = -0.064 mN/m.s). These results indicate that conjugation plays an important role in controlling the rate of lipolysis in our organism which can be in turn, tuned by the microflora composition of our gut, ultimately controlling the rate of deconjugation of the BS.

Abstract:The evolving vision of the Internet-of-Things (IoT) will revolutionize various applications such as remote health monitoring, home automation and remote surveillance. It has been projected that by 2025, there will be 1 trillion IoT devices influencing our daily lives. This will result in the generation of an enormous amount of data, which will have to be stored, processed and transmitted efficiently and reliably. Although advancements in Integrated Circuit (IC) design and the availability of various Ultralow Power (ULP) circuit components have helped us to visualize an ecosystem of numerous internetconnected devices, the overall system integration will become a major challenge. A System-on-Chip (SoC), catering to IoT applications is expected to contain many different circuit components such as sensors and Analog-Front-Ends (AFEs) for real-time signal acquisition, analog-to-digital converters, digital signal processors, memories, wireless transceivers etc. All these components have different supply voltage requirements and power profiles. Hence, power delivery to such components in an SoC will play an important role in the overall system architecture. Although, battery-powered systems have traditionally worked well in portable electronics but in an IoT ecosystem, the cost of battery replacement in a trillion-sensor node network will be enormous. In many applications, such as remote surveillance, systems require a long operational lifetime. Moreover, system deployment should be unobtrusive and hence such systems should have small form factors. The above requirements are hard to meet using conventional battery-powered systems. Hence, in most IoT SoCs, there is a strong motivation for an integrated Power Management Unit (PMU), with energy harvesting capability for near-perpetual battery-less operation, which can provide a range of supply voltage rails to satisfy the electrical specifications of different functional units. This dissertation will address the design challenges related to energy autonomy and power-delivery in a wireless sensor node. We propose a fully integrated energy harvesting platform with a capability to harvest from multiple sources of energy such as indoor solar and thermoelectric generators (TEGs). Additionally, we also propose a power-efficient supply regulation scheme to meet the electrical specifications of the various components of a self-powered, battery-less SoC. Finally, we demonstrate several ULP digital and mixed-signal circuit components, which can be leveraged in an energyautonomous system. The proposed solutions to power delivery will enhance the operational lifetime, reduce the overall form factor and contribute towards attaining energy-autonomy to facilitate a wide range of applications related to the IoT.

Abstract:β-Carotene (BC) nanoemulsions were successfully prepared by microfluidization. BC micellarization was significantly affected by bile salts and pancreatin concentration. Positive and linear correlation was observed between BC release and bile salts concentration. Pancreatin facilitated BC's release in simulated digestion. Compared to the control (bulk oil) (4.6%), nanoemulsion delivery systems significantly improved the micellarization of BC (70.9%). The amount of BC partitioned into micelles was positively proportional to the length of carrier oils. Unsaturated fatty acid (UFA)-rich oils were better than saturated fatty acid (SFA)-rich oils in transferring BC (p < 0.05). No significant difference was observed between monounsaturated fatty acid (MUFA)-rich oils and polyunsaturated fatty acid (PUFA)-rich oils (p > 0.05). A positive and linear relationship between the degree of lipolysis and the release of BC in vitro digestion was observed. Bile salts showed cytotoxicity to Caco-2 cells below 20 times dilution. BC uptake by Caco-2 cells was not affected by fatty acid (FA) compositions in micelles, but BC uptake was proportional to its concentration in the diluted micelle fraction. The results obtained are beneficial to encapsulate and deliver BC or other bioactive lipophilic carotenoids in a wide range of commercial products.
Extraction of naringin dihydrochalcone and its effect on reducing blood lipid levels in vitro

Abstract:Response surface methodology (RSM) was used to optimize the extraction process of naringin. The central component design included three parameters of extraction, namely, temperature (X ), solid-liquid ratio (X ), and extraction time (X ). The optimum extraction temperature was 67℃; the ratio of material to liquid was 54:1mL/g, and the extraction time was 2.8h. According to the best extraction conditions, naringin was processed to verify the accuracy of the model. Five parallel experiments were set up, and a yield of 32.48 mg/g naringin was obtained, that was equivalent to the predicted yield of 32.56 mg/g. Naringin was purified to obtain naringin-refined products using DM101 macroporous adsorption resin. Naringin dihydrochalcone was synthesized following catalytic hydrogenation of purified naringin. The structures of naringin and naringin dihydrochalcone were determined via Fourier infrared spectrometer and nuclear magnetic resonance spectrometry. Evaluate the ability of naringin dihydrochalcone to bind with glycine bile sodium and bovine bile sodium by simulating the gastrointestinal environment in vitro. Further focusing on HepG2 cells, a high cholesterol induced high-fat HepG2 cell model was established. We measured the effects of different concentrations of naringin dihydrochalcone on intracellular lipids in denatured HepG2 cells and further validated the lipid-lowering effect of naringin at the cellular level. The results showed that naringin dihydrochalcone has potential application in functional foods for lowering blood lipids.

Unravelling the behaviour of curcumin nanoemulsions during in vitro digestion: effect of the surface charge

Abstract:Oil-in-water nanoemulsions containing curcumin were prepared through high-pressure homogenization using corn oil and three different emulsifiers: Tween 20 (non-ionic), Sodium Dodecyl Sulphate (SDS, anionic) and DodecylTrimethylAmmonium Bromide (DTAB, cationic). A human gastric simulator was used as the in vitro digestion model (in which the stomach, duodenum, jejunum and ileum steps were performed) to evaluate the impact of surface charge on the digestion of the curcumin nanoemulsions. This model allowed the simulation of continuous peristaltic movements and consequently enabled a more mechanically realistic simulation of the dynamic digestion process than simple stirred vessel models. The emulsifier charge had a significant effect on the droplet size, particle electric charge and microstructure of curcumin nanoemulsions during the simulated digestion, which consequently influenced the free fatty acid release and curcumin bioavailability. The results showed the positively charged DTAB-stabilized emulsions to be the least stable during the digestion process, exhibiting the largest increase in droplet size and eventual phase separation. This also contributed to the low bioavailability of curcumin. Conversely, emulsions stabilized with Tween 20 showed retention of emulsion structure (high surface area) and greater free fatty acid production, which could explain the increased curcumin bioavailability. The emulsifier charge influenced the lipid digestion process and the bioavailability of the bioactive compound incorporated, probably by altering the ability of bile salts and digestive enzymes to adsorb onto the emulsion surfaces, thus altering the droplet size (and consequently the surface area) due to droplet breakup or coalescence within the digestive tract. The results of this work also highlighted the importance of subjecting the emulsions to a simulated gastric environment, since changes in pH, ionic strength, gastric enzyme activity and shear will impact the emulsion properties in the small-intestine. This manuscript has provided important insights into the effect of emulsifier charge on the behaviour of nanoemulsions during in vitro digestion, which is important to determine their functional performance, aiming at the optimization of nanoemulsion-based delivery systems to protect and release bioactive lipophilic compounds.

Abstract:Naringenin (NGN) can be used to inhibit the progression of nonalcoholic fatty liver disease (NAFLD) in mice, but its poor water solubility limits its applications. Nanostructured lipid carriers (NLCs) have recently attracted much attention in the field of nanodrug delivery systems because they increase the drug loading capacity and impressively enhance the solubility of indissolvable drugs. Herein, a thin-film dispersion method was used to prepare naringenin-loaded nanostructured lipid carriers (NGN-NLCs). These NGN-NLCs have a narrow size distribution of 171.9 ±2.0 nm, a high drug loading capacity of 23.7 ± 0.3%, a high encapsulation efficiency of 99.9 ± 0.0% and a drug release rate of 86.2 ± 0.4%. NGN- NLCs elevated the pharmacokinetic parameters (Cmax and AUC0→t) of NGN, accelerated NGN transepithelial transport in MDCK cells and intestinal absorption in the jejunum and ileum, and reduced hepatic lipid accumulation in an oleic acid (OA) plus lipopolysaccharide (LPS)-induced lipid deposition cell model in primary hepatocytes and in a methionine/choline deficient (MCD) diet-induced NAFLD mouse model. A detailed study of the mechanism showed that this NLC formulation elevated the drug release rate in simulated intestinal solutions in vitro, the transepithelial transport in MDCK cells, the oral absorption in mice and the ex vivo intestinal absorption of NGN. Thus, NGN-NLCs significantly enhanced the inhibitory effects of NGN on MCD diet induced mouse NAFLD.
