Abstract:Abstract In order to explore the influence of temperature and salinity on stress response of P. monodon , four experimental groups were set, respectively were normal temperature and normal salt (Control group), normal temperature and low salt(Group A), high temperature and low salt(Group B), high temperature and normal salt(Group C). The stress experiment was conducted for 96 hours, and the activities of acid phosphatase(ACP), alkaline phosphatase(AKP), total superoxide dismutase (T-SOD) and total antioxidant capacity (T-AOC) in hepatopancreas, the changes of tissue structure and the expression of related heat shock protein genes were measured．The results showed that the branchial structures of the three stress groups were found to be damaged to varying degrees. In Group A, AKP and ACP levels increased initially and then decreased, while T-SOD levels decreased initially and then steadily increased. In Group B and Group C, the four enzymes showed similar trends.There was no significant difference in the expression level of Hsps in the low salt group. The expression levels of Hsp-10, Hsp-60 and Hsp-90 genes showed a similar trend, and the maximum expression levels of the four genes were all at 96h. The expression levels of Hsps in muscle tissue were similar to those in hepatopancreas tissue. However, the highest expression levels of Hsp-70 and Hsp-90 in high temperature and low salt group were at 24h. Through this work on P. monodon stress responses to high temperature and low salinity, we have gained a deeper understanding of the physiological responses to environmental stressors, which will be useful in the development of protective measures for the shrimps under aquaculture conditions that replicate these stressors.

Abstract:Determining optimum stocking density of the white shrimp Litopenaeus vannamei (Boone) is a big concern for shrimp farmers. However, few studies have assessed the influence of stocking density on the antioxidant status, immunology, digestive enzyme activities, and growth performance of white shrimp in biofloc systems. In this study, these parameters of white shrimp in a biofloc system were compared at three stocking densities: 300 orgs m−3 as low stocking density (LD), 400 orgs m−3 as medium stocking density (MD), and 500 orgs m−3 as high stocking density (HD). The feed conversion ratio in the LD group was significantly lower than that in the MD and HD groups (P < 0.05), and the ultimate individual weight in the LD group was significantly higher than that in the other two groups (P < 0.05). The antioxidant status and immunology parameters, including complement 3, complement 4, lysozyme, superoxide dismutase, glutathione peroxidase and malondialdehyde were all depressed in the HD groups. Furthermore, activities of the digestive enzymes, amylase, trypsin, and lipase were lower in the MD and HD groups than that in the LD group. The highest relative percentage survival was observed in the LD group 10 days after challenge with the pathogen Vibrio harveyi. Results of this study indicated that the immune status and welfare of white shrimp can be seriously impaired in the HD condition (i.e., ≥500 m−3) in biofloc systems. These findings can be used to determine suitable stocking densities in the white shrimp farming industry using the biofloc system.
Optimizing white shrimp aquaculture: Feeding frequency and automatic feeders impact on growth and antioxidant capacity

Abstract:Abstract The feeding regime is a critical aspect of white shrimp ( Litopenaeus vannamei ) aquaculture, but research on intensive shrimp culture using automatic feeders is scarce. Our study investigated different feeding strategies for adult white shrimp in intensive aquaculture tanks, evaluating growth, survivability, feed utilization, shrimp digestive and antioxidant capacity, and conducting an economic analysis. Shrimp (8.0 ± 0.5 g) were cultured for 35 days with feeding frequencies of 6, 8, 10, and 12 times/day (A6, A8, A10, and A12 groups, respectively) using automatic feeders, and one group was fed manually 6 times/day (M6). The results show that the automatic feeding groups had a higher final body weight and specific growth rate, and a lower feed conversion ratio compared to the manual group. Among the automatic feeding groups, A6 and A8 exhibited the best performance, with a quadratic regression suggesting 7.83 times/day as an optimal feeding frequency. Digestive enzyme activity (TP and α-AMS) was significantly higher in the A6 and A8 groups ( p < 0.05). However, the A6 and A8 groups showed significantly higher levels of antioxidant enzymes (SOD and GPx) and lower levels of lipid oxidant enzyme (MDA), indicating better digestive and antioxidant abilities in shrimp with higher growth rates. Moreover, the antioxidant genes expression of MnSOD and GPx testified the enzyme changing. Pearson's correlation confirmed the positive correlation between SOD and GPx. Economically, the A8 group showed the highest profitability among all groups. Overall, the study recommends a feeding frequency of 6–8 times/day using automatic feeders as a suitable and feasible approach for white shrimp culture in intensive settings.

Abstract:The objective of this study is to explore the immune response of the shrimp Penaeus vannamei to low temperature and air exposure during the mimic waterless transportation. Shrimp were cold shocked at 13 °C for 3 min, then exposed to air at 15 °C for 12 h, and finally revived in water at 25 °C. Hemocyte structure remained intact with only slight distortions of some organelles and nuclear membrane under the stress. Phenoloxidase (PO), lysozyme (Lys) and gamma-glutamyl transferase (GGT) activities, glutamine (Gln) content and relative mRNA expressions of prophenoloxidase (proPO), β-1,3-glucan binding protein (LGBP), ferrin (Fer) and glucose regulated protein 78 (GRP 78) increased and reached peak levels after 3 h-9 h of air exposure, and then decreased to relatively stable levels in the prolonged period of air exposure. The total hemocyte count (THC) and gene expressions of proPO, Fer and LGBP at the end of waterless storage were significantly lower (p < 0.05) than those in fresh shrimp while no significant difference (p ≥ 0.05) was found between revived and fresh shrimp in PO, Lys, GGT activities, Gln content and gene expression level of GRP 78. Of all the hemocytes, the percentage of semi granular cells (SGC) and granular cells (GC) significantly decreased after 6-9 h of storage, while hyaline cells (HC) dramatically increased after 9 h of storage to compensate the loss of SGC and GC. Results suggested that the low temperature and air exposure caused significant immunological response to the shrimp, but the damages to the immune system were partly reparable.

Abstract:Free fatty acids have long been used as dietary supplements in aquaculture, but the application of monoglycerides has increased interest in more recent times. The study aimed to investigate the effects of dietary short- and medium-chain fatty acid monoglyceride and cinnamaldehyde (SMMG) on the growth performance, survival, immune responses, and tolerance to hypoxic stress of Pacific white shrimp (Litopenaeus vannamei). In Experiment 1, shrimp post-larvae were divided into 4 groups with 6 replicates and fed with diets supplemented with 0 (control), 0.3, 0.4, and 0.5% diet for 30 days. The final body weight and survival rate were determined. In Experiment 2, the juvenile shrimp from Experiment 1 were subjected to hypoxic stress conditions (dissolved oxygen level 2-2.5 mg/L) for 14 days, then the specific growth rate (SGR), survival rate, intestinal Vibrio spp. count, immune responses, and histopathological change of the hepatopancreas were analyzed. Following the 30-day feeding trial, the results revealed that the final body weight and survival of the 0.3-0.5% SMMG groups (2.81-3.06 g and 74.00-84.33%, respectively) were significantly higher than the control shrimp (1.96 g and 68.33%, respectively). In the hypoxic stress experiment, the survival rates of shrimp fed 0.4-0.5% SMMG (71.67-80.00%) were significantly higher than the control (51.67%). Although the SGR were not affected by SMMG supplementation, all immune parameters evaluated were significantly enhanced, and the intestinal Vibrio spp. counts were significantly decreased in the 0.4-0.5% SMMG-fed shrimp; the histopathological structure of the hepatopancreas was also improved in these shrimp compared to the control. Our findings indicated that SMMG as a feed additive has beneficial effects in improving shrimp health and increasing tolerance to hypoxic conditions.
Oxygen Supply in White-leg Shrimp Litopenaeus Vannamei Juvenile Culture by Coated Sodium Sulphate-hydrogen Peroxide-sodium Chloride Adduct

Abstract:The coated sodium sulphate-hydrogen peroxide-sodium chloride adduct was prepared mainly from hydrogen peroxide, sodium sulphate and sodium chloride. Surface structure, stability and release profiles of the coated adduct were analyzed. It was found that layers on the surface of the particles were loose macro-voids membrane. The decomposing rate of hydrogen peroxide was only 1.23% in the air for 10d and the release rate in 5min was over 50% in water with pH 6.0, 7.0, 8.5 and 10. The coated adduct was used into water in three white-leg shrimp Litopenaeus vannamei juvenile culture ponds at a rate of 1.0, 2.0 and 3.0g/m^3 (adduct to water), respectively. The results showed that the dissolved oxygen (DO) levels in the water increased by 11.6%, 18.2% and 26.2% 10min after administration of the coated adduct compared with the initial stage, and, the DO level was the maximum, 27.5%, 32.5% and 40% higher than that in the control tanks about 20min after administration of the coated adduct, respectively. The white-leg shrimp juvenile hypoxia was found in the control ponds about 20min after administration of the coated adduct. A few juveniles appeared hypoxia in the experimental pond supplemented with at a rate of 1.0g/m^3 coated adduct only about 60min after the administration. In other two experimental ponds, however, the phenomenon was not found.

Abstract:This study investigated the synergistic effects of oxolinic acid (OA) combined with oxytetracycline (OTC) on white shrimp (Penaeus vannamei). Disk diffusion susceptibility testing was performed to analyze the sensitivity of Vibrio alginolyticus and Vibrio parahaemolyticus to different concentrations of OA and OTC. The results revealed that 50 mg OA/L combined with 50 mg OTC/L exhibited stronger antibacterial effects on V. alginolyticus and V. parahaemolyticus. The results of in vitro tests indicated that cotreatment with OA and OTC significantly reduced superoxide anion production and phenoloxidase activity, but not phagocytic activity. Subsequently, feeding trials were performed to investigate the immunomodulatory effects and bioaccumulation of dietary OA combined with OTC on shrimp. The healthy shrimp (15.13 ± 1.02 g) were divided into four groups: control, 100 mg OA/kg combined with 50 mg OTC/kg, 50 mg OA/kg combined with 100 mg OTC/kg, and 50 mg OA/kg combined with 50 mg OTC/kg. The shrimp were sampled to determine innate immunity parameters and residual OA and OTC levels in the muscle during a 28-day feeding regimen; the shrimp were fed the experimental diet from day 1 to day 5 and a commercial diet from day 6 to day 28. Residual OA levels were considerably higher in the group fed 100 mg OA/kg combined with 50 mg OTC/kg compared with the other groups and peaked on day 4. The residual OA levels of all the groups were below the detection limit after without providing OA. The residual OTC levels of the group fed 50 mg OA/kg combined with 100 mg OTC/kg were considerably higher from day 1 to day 4. The residual OTC levels in all the groups decreased rapidly and could not be detected on day 28. The administration of 50 mg OA/kg combined with 100 mg OTC/kg exerted the least effect on the white shrimp. Moreover, the survival rates of the treatment groups after the V. parahaemolyticus challenge were higher than those of the control group, especially the group fed 50 mg OA/kg combined with 100 mg OTC/kg. This result indicated that the synergistic effects of dietary OA and OTC are safe and effective. Combination therapy is a new method of antibiotic use in aquaculture.
Inorganic Nitrogen Control, Growth, and Immunophysiological Response of Litopenaeus Vannamei (boone, 1931) in a Biofloc System and in Clear Water with or Without Commercial Probiotic

Abstract:A 5-week feeding trial was conducted to evaluate the effects of biofloc in situ and commercial probiotic supplementation on white shrimp (1.87 ± 0.03 g) inorganic nitrogen control, growth, and immunophysiological response. For this purpose, four treatments were conducted: clear water with no probiotic application (CW), clear water with probiotic application (CW+P), biofloc with no probiotic application (FLOC), and biofloc with probiotic application (BFT+P); each group had three replicates. Growth parameters (final body weight, daily weight gain, specific growth rate) were significantly higher in the two biofloc systems (P < 0.05), and the FLOC and FLOC+P group did not have a significant difference (P > 0.05). The immune responses (total hemocyte count, complement component protein, and lysozyme) and antioxidant status (glutathione, catalase) in the CW+P, FLOC, and FLOC+P groups were increased significantly at the end of the experiment compared with the CW group (P < 0.05), and the FLOC and FLOC+P groups did not have a significant difference (P > 0.05). Results of a 10-day Vibrio harveyi challenge test show that the survival rates in CW+P, FLOC, and FLOC+P groups were significantly higher (P < 0.05), and the FLOC and FLOC+P groups did not have a significant difference (P > 0.05). These results suggest that probiotic addition in the biofloc system had little advantage, but probiotics can improve the immune status of the shrimp in the clear water system. Further, cost-effectiveness analysis showed that the biofloc system was an efficient and economical option for the production of white shrimp.
Effects of the glutathione administration via dietary on intestinal microbiota of white shrimp, Penaeus vannamei, under cyclic hypoxia conditions

Abstract:Environmental stress can impair the survival, growth performance, and intestinal environment of shrimp. The objective of this study was to investigate the effect of glutathione (GSH) on the survival, growth performance, intestinal oxidation, microbiota, and histology of shrimp under hypoxia. Four treatments were used: (1) normoxia, (2) cyclic serious/medium hypoxia (CSMH), (3) CSMH and 75 mg kg−1 GSH, and (4) CSMH and 150 mg kg−1 GSH. White shrimp (Penaeus vannamei) in groups 3 and 4 were fed a commercial diet supplemented with 75 and 150 mg kg−1 GSH for a 28 day period, respectively, and they were cultured under CSMH (0.8 – 3.5 mg L−1 dissolved oxygen) for the last 14 days of the experiment. P. vannamei supplemented with 75 mg kg−1 GSH showed significantly improved survival and growth performance under CSMH compared with the CSMH condition alone. The dose of 75 mg kg−1 GSH completely eliminated overproduction of reactive oxygen species and malondialdehyde to suppress serious histopathological lesions and improve bacterial diversity and the relative abundance of beneficial bacteria, such as Rhodobacteraceae, thereby preventing pathogen (e.g., Vibrio) invasion in the intestine of shrimp under CSMH. However, the dose of 150 mg kg−1 GSH was excessive, as it led to serious impairment of survival and growth. These results indicate that 75 mg kg−1 GSH has the potential to control shrimp mortality and growth inhibition under CSMH in the shrimp farm setting.

Effects of Low-Fish-Meal Diet Supplemented with Coenzyme Q10 on Growth Performance, Antioxidant Capacity, Intestinal Morphology, Immunity and Hypoxic Resistance of Litopenaeus vannamei

Abstract:The aim of this study was to evaluate the effects of a low-fish-meal diet supplemented with coenzyme Q10 on the growth, antioxidant capacity, immunity, intestinal health and hypoxic resistance of Litopenaeus vannamei. L.vannamei with an initial weight of 0.66 g were fed with the experimental diets for 56 days. Diets D1 (20% FM level) and D2-D7 (15% FM level), supplemented with 0%, 0.002%, 0.004%, 0.006%, 0.008% and 0.01% coenzyme Q10 were formulated. In terms of growth performance, the weight gain and specific growth rate in the D2 diet were significantly lower than those in the D1 diet (p < 0.05). The final body weight, weight gain and specific growth rate in the D2-D7 diets had an upward trend, and the condition factor in the D2-D7 diets was lower than those in the D1 diet (p < 0.05). There were no significant differences in the crude protein and crude lipid levels in the whole body among all diet treatments (p > 0.05). In terms of hepatopancreas antioxidant parameters, the D5 and D6 diets significantly promoted the total antioxidant capacity and total superoxide dismutase activity, and significantly decreased the malondialdehyde content (p < 0.05). The expression levels of cat, mnsod and gpx in shrimp fed with the D5 and D6 diets were significantly higher than those of shrimp fed with the D2 diet (p < 0.05). In addition, the mRNA level of ProPO was increased in the D4 and D5 diets, and LZM expression was increased in the D6 diet compared with the D1 diet (p < 0.05). The villus height of shrimp fed with diets supplemented with coenzyme Q10 was significantly increased (p < 0.05), and the intestinal thickness and submucosal thickness of shrimp fed with the D6 diet were the highest (p < 0.05). After acute hypoxia stress, lethal dose 50 time in the D3-D7 diets was significantly increased compared with the D1 and D2 diets (p < 0.05), and the highest value was found in the D4 diet (p < 0.05). After stress, the expression levels of TLR pathway-related genes (Toll, Myd88, Pelle, TRAF6 and Dorsal) in the D4 and D6 diets were significantly increased compared with the D2 diet. In general, Litopenaeus vannamei fed with the D6 diet achieved the best growth, antioxidant capacity, immunity, and intestinal morphology among all low FM diets and D4-D6 diets improved hypoxic resistance.
Effects of organic acids and essential oils blend on growth, gut microbiota, immune response and disease resistance of Pacific white shrimp (Litopenaeus vannamei) against Vibrio parahaemolyticus

Abstract:An 8-week feeding trial was undertaken to evaluate supplemental effects of AviPlus® (AP), a blend of organic acids [citric acid, 25%; sorbic acid, 16.7%] and essential oils [thymol, 1.7%; vanillin, 1.0%], on growth, gut microbiota, innate immunity and disease resistance of Pacific white shrimp (Litopenaeus vannamei) against Vibrio parahaemolyticus. A basal experimental diet was formulated and supplemented with 0, 0.3, 0.6, 0.9 and 1.2 g kg-1 AP to produce five test diets (Con, AP0.3, AP0.6, AP0.9 and AP1.2). Each diet was fed to triplicate groups of shrimp (0.2 ± 0.01 g, mean ± SE) to apparent satiation three times daily. Growth performance and survival rate were not significantly influenced by AP supplementation (P > 0.05). Significantly (P < 0.05) higher serum total protein was found in groups fed ≥ 0.6 g kg-1 AP compared to control. Serum alkaline phosphatase and phenoloxidase activities were significantly increased in AP0.9 and AP1.2 groups. Also, the group received AP0.6 diet showed significantly higher glutathione peroxidase activity than control. Expression of gut pro-inflammatory genes including TNF-α, LITAF and RAB6A were down-regulated by AP administration. Gut microbiota analysis showed the significant enhancement of the operational taxonomic unit (OTU) diversity and richness indices by AP application. AP supplementation led to increased abundance of Firmicutes and a reduction in abundance of Proteobacteria. Also, dietary inclusion of 1.2 g kg-1 AP led to a significant increase in the abundance of Lactobacillus in shrimp gut. The group offered AP0.3 diet showed significantly higher disease resistance than control group. Furthermore, AP application significantly enhanced relative expression of immune related genes including lysozyme, penaeidin and catalase at 48 h post challenge. In conclusion, these findings show that the tested organic acids and essential oils mixture beneficially affects intestinal microflora and improves immune response and disease resistance of L. vannamei.

Abstract:Pacific white shrimp ( Penaeus vannamei ) is one of the most productive and economically important species globally. However, the development and continuous expansion of the farming scale led to an increase in the risk of disease occurrence in shrimp farming. The application of probiotics as an effective method for controlling diseases in aquaculture has been widely considered. In shrimp farming, several probiotics have been used and shown benefits to the health of the host. To diverse the sources of bacterial species as probiotics in shrimp farming, in this study, we aimed to elucidate the effects of dietary probiotics ( Clostridium butyricum I9 (I9), Clostridium butyricum G15 (G15), or Paraclostridium bifermentans X13) on the growth, immune response and intestinal microbiome of white shrimp. Shrimps were fed with diets containing either phosphate-buffered saline (PBS), I9 (10 7 CFU/g feed), G15 (10 7 CFU/g feed), or X13 (10 7 CFU/g feed) for 30 days and followed by the challenge with Vibrio parahaemolyticus ( Vp ). The results showed that the survival rate, body weight gain, and special growth rate of shrimps in the I9, X13, and G15 groups significantly increased, compared to the PBS. The supplementation of probiotics increased the content of short-chain fatty acids and effectively maintained the normal morphology and structure of the intestinal tract and hepatopancreas. The I9, X13, or G15 groups showed a positive change in the diversity and abundance of gut bacteria. There was a significant up-regulation of CTL, SOD, proPO, Crustin, PEN2-4, and ALF1-3 genes in shrimps in the I9, X13, and G15. Additionally, dietary probiotics significantly increased the survival rate, maintained the intestinal structure, promoted the activities of SOD, AKP, ACP, and T-AOC enzymes, and reduced the level of MDA in shrimps after Vp infection. In conclusion, dietary supplementation of I9, G15, or X13 improved the growth, immunity, and disease resistance of Pacific white shrimp, providing a scientific basis for shrimp farming.

Abstract:Vibrio genus is a common pathogen in aquaculture and causes acute hepatopancreatic necrosis disease (AHPND) and massive mortality of shrimp. Many studies have suggested that a single functional ingredient such as plant extract or organic acid can reduce the dependence on antibiotics and promote the growth and immunity of aquatic animals. In this study, we evaluated the effects of a phytobiotic-based compound additive (Sanacore® GM, SNGM), which had a successful trajectory of commercial application in fish farming. However, its effects on the hepatopancreas health and intestinal microbiota of shrimp after Vibrio challenge have not been well evaluated. In the present study, Pacific white shrimp were fed diets with or without supplementation of SNGM, and the SNGM grades were 0-g/kg (CON), 3-g/kg (SNGM3), and 5-g/kg (SNGM5) diets. The feed trial lasted 60 days, after which a Vibrio parahaemolyticus challenge was performed. The results showed that compared to the CON group, both the SNGM3 and SNGM5 groups had a significantly higher weight gain and a lower feed conversion ratio as well as higher survival after Vibrio parahaemolyticus challenge. In the growth trial, the SNGM3 group had a significantly increased total protein, albumin concentration, and acid phosphatase activity in hemolymph compared to the CON group. In the challenge experiment, the SNGM3 and SNGM5 groups had increased albumin and glucose contents as well as the activities of phenoloxidase, lysozyme, alkaline phosphatase, and superoxide dismutase in hemolymph. Both the SNGM3 and SNGM5 groups had improved morphology of the hepatopancreas and intestine. The SNGM5 group had alleviated gut microbiota dysbiosis induced by Vibrio infection by increasing the potential probiotic bacterium abundance (Shewanella) and decreasing the potential pathogenic bacteria abundance (Vibrio, Photobacteriuma, Pseudoalteromonas, and Candidatus_Bacilloplasma). In conclusion, the dietary phytobiotic-based additive at 3-g/kg level increased the growth and Vibrio parahaemolyticus resistance of Pacific white shrimp by promoting immune-related enzyme activities and improving the morphological structure of the hepatopancreas and intestine and the intestinal microbiota composition.

Abstract:The intestine is a host-pathogen interaction site and improved intestinal barrier function help to prevent disease in shrimp. Alginate oligosaccharides (AOS) are derived from resourceful brown algae. The intestine protection properties of AOS were widely recognized, and their benefits in fish have been reported. Nevertheless, there are no reports on AOS in shrimp and other crustaceans. In the present work, we measured the effects of AOS on growth performance and disease resistance in the white shrimp Litopenaeus vannamei and investigated their effects on intestinal health. Shrimps with an initial weight of about 2 g were fed with diets supplemented with 0 (control), 0.07%, 0.2%, 0.6%, or 1.2% of AOS for 56 days and were sampled and challenged with Vibrio parahaemolyticus. Dietary AOS did not significantly influence weight gain or feed utilization (P > 0.05). However, AOS considerably decreased the seven-day cumulative mortality after the challenge at any dose (P < 0.05). Dietary AOS improved the intestinal structure, significantly boosted the intestinal villus height at 0.6% and 1.2% levels, and increased intestinal wall thickness by 0.2%, 0.6%, and 1.2%. The alkaline phosphatase and maltase activities were also increased, suggesting that AOS improved the intestinal condition. Redox homeostasis in intestinal was improved by AOS, as expressed by the enhanced total antioxidant capacity and decreased malonaldehyde content, partly due to the increased superoxide dismutase and catalase activities. Compared with the antioxidant system, AOS's stimulating effects on immunity were more significant. At any level, AOS significantly activated lysozyme activity, the expression of propo and two antimicrobial peptide genes (pen-3 and crusin). However, the lowest concentration of AOS did not stimulate the gene expression of all three assayed pattern recognition receptors (LGBP, Toll, and IMD), and only the highest concentration of AOS increased the expression of imd. These findings suggest that AOS are highly efficient immunostimulants, and various immune pathways in shrimp are differentially sensitive to AOS. Finally, our findings suggest that AOS significantly alter the gut microbiota and their relative abundance at the phylum, family, and genus levels. In conclusion, AOS significantly enhances disease resistance in L. vannamei, possibly attributed to improved intestinal development, increased intestinal immunity and altered microbiota. These findings could provide a basis for future studies on the practical use of AOS and its mechanisms of action.

Abstract:Use of antibiotics and other chemicals to combat disease outbreaks has been a bottleneck for the sustainable growth of shrimp industry. Among various replacements proposed, organic acid (OA) and their salts (OS) are commonly used by farmers and feed millers. However, in free forms, their requirement is very high (2–3 kg/MT) as they tend to disassociate before reaching the hindgut. The dosage can be reduced by microencapsulation of the ingredients. In this study, a 63-day trial was conducted to assess the effects of OA and OS (COMP) microencapsulated (ENCAP) with fat (HF), fat + alginate (HA), wax esters (WE) and HA + WE (HAWE) on performance, digestive enzymes, immunity and resistance to Vibrio parahaemolyticus. A positive control (PC, 200 g/kg fishmeal-FM) and a negative control (NC, 130 g/kg FM) diet were formulated. Eight other diets were formulated, supplementing an NC diet with microencapsulated OA (OAHF, OAHA, OAWE, OAHAWE) and OS (OSHF, OSHA, OSWE, OSHAWE). Among the ENCAPs, significant difference was observed in serum malondialdehyde (p = 0.026), where HF showed the lowest level (6.4 ± 0.3 mmol/L). Significant interactions between COMP and ENCAP were observed in lipid deposition (p = 0.047), serum alkaline phosphatase, acid phosphatase, hepatopancreatic and serum phenol oxidase (p < 0.0001). Despite no differences, 96-h mortality during pathogenic Vibrio parahaemolyticus challenge in all treatment diets (45–56%) was lower compared to the NC diets (63%). In conclusion, use of HF microencapsulated OA diets could provide improved performance and disease resistance that could contribute to the reduction of antibiotic use by the shrimp industry.

Abstract:A 58-day feeding trial was conducted to evaluate the effects of dietary myo-inositol (MI) supplementation on growth performance, haematological parameters, hepatopancreas histopathology and antioxidant status of Litopenaeus vannamei fed with oxidized fish oil (OFO). Control diet contained fresh fish oil (FFO) without MI supplementation. The other four diets contained two oxidation levels of OFO (peroxide value: 133.2 and 268.7 meq kg-1) with or without 200 mg MI kg-1 diets (MI0+L, MI0+H, MI200 + L and MI200 + H). Results showed that OFO-supplemented groups (without MI supplementation) showed better growth performance and lower whole-body inositol content when opposed to control group. MI supplementation significantly improved whole-body inositol content in high-oxidized fish oil (HOFO) groups, and also reduced whole-body lipid in low-oxidized fish oil (LOFO) groups. Moreover, Supplementation of OFO and MI markedly hit the fatty acid profile of muscle. HOFO caused severe histopathological changes in hepatopancreas of shrimp, which slightly alleviated by MI supplementation. MI supplementation also grew the total protein (TP) content and alkaline phosphatase (AKP) activity and decreased the activities of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) of serum in OFO-supplemented groups. Ingestion of OFO increased levels of lipid peroxidation and protein oxidation in serum or hepatopancreas, which partly ameliorated by MI supplementation. Activities of antioxidant enzymes exhibited different expression patterns because of OFO and MI. In addition, HOFO markedly increased mRNA expression levels of antioxidant genes including ferritin (FT), thioredoxin (Trx), GPX, glutathione S-transferase (GST) and catalase (CAT) and decreased peroxiredoxin (Prx) expression, in which expression of GPX and Prx were increased owing to MI supplementation. Therefore, it suggested that dietary OFO stimulated growth performance, but also induced oxidative stress and caused impairment to hepatopancreas in L. vannamei. The negative impact brought about by OFO was partially mitigated by dietary MI supplementation.

Abstract:The present study investigated the effects of short-chain fructooligosaccharides (FOS) as dietary supplements on stress, immune response, gut microbiota, growth, and survivability of three different treatment groups of Pacific white shrimp, Litopaneous vennamei, cultured in a recirculating system. The experiment was conducted over a 35-day trial period. Shrimps were fed diets, 15 times a day using automated feeder, supplemented with GOS at 0%, 0.15%, and 0.30% by weight. Denaturing gradient gel electrophoresis analysis revealed that the enteric microbial community of shrimp fed the basal diet differed markedly (<80.0% similarity coefficient) from those fed FOS-supplemented diets. However, shrimp survival, weight gain, and immune responses among the treatment groups were good but not significantly different (P > 0.05), probably due to the limited length of the feeding trial.

Abstract:Aims: This study aimed to assess the effects of chlorine dioxide (ClO2) in water on whiteleg shrimp Penaeus vannamei, evaluating its impact on the stomach microbiota, gill transcriptome, and pathogens. Methods and results: ClO2 was added to the aquarium tanks containing the shrimp. The application of ClO2 to rearing water was lethal to shrimp at concentrations above 1.2 ppm. On the other hand, most of the shrimp survived at 1.0 ppm of ClO2. Microbiome analysis showed that ClO2 administration at 1.0 ppm significantly reduced the α-diversity of bacterial community composition in the shrimp stomach, and this condition persisted for at least 7 days. Transcriptome analysis of shrimp gill revealed that ClO2 treatment caused massive change of the gene expression profile, including stress response genes. However, after 7 days of the treatment, the gene expression profile was similar to that of shrimp in the untreated control group, suggesting a recovery to the normal state. This 1.0-ppm ClO2 significantly reduced shrimp mortality in artificial challenges with an acute hepatopancreatic necrosis disease-causing Vibrio parahaemolyticus and white spot syndrome virus, which were added to rearing water. Conclusions: The use of ClO2 at appropriate concentrations effectively eliminates a significant portion of the bacteria in the shrimp stomach and pathogens in the water. The results of this study provide fundamental knowledge on the disinfection of pathogens in water using ClO2 and the creation of semi germ-free shrimp, which has significantly decreased microbiome in the stomach.

Abstract:The objective of this study was to evaluate the effect of dietary supplementation of a probiotic bacterium, Bacillus subtilis, on the growth, immune response and antioxidant activities of shrimp (Litopenaeus vannamei). Shrimps with an average initial weight of 2.11 +/- 0.17 g were randomly assigned to four groups with three replicates. The control group was fed a basal diet, and three treated groups were fed diets supplemented with B. subtilis at doses of 1 x 104, 5 x 104 and 10 x 104 colony-forming unit (CFU) g-1 feed respectively. After 40 days of culture, 10 shrimps from each replicate were taken randomly for the determination of immune response and oxidization resistance indices. The results showed that the shrimps fed with B. subtilis at a dose of 1 x 104, 5 x 104 CFU g-1 feed showed significantly better growth than that of the control diet. The phenoloxidase activities showed a tendency to increase with an increased dose of B. subtilis in diets but there was no significant difference among the three treated groups. In addition, phenoloxidase activities were found to be significantly higher (P < 0.05) in the groups treated with B. subtilis than that of the control group. Shrimps treated with 5 x 104 CFU g-1 feed probiotic bacterium showed the highest lysozyme activity and it was significantly higher (P < 0.05) than the other groups. However, there was no significant difference in acid phosphatase and alkaline phosphatase activity across all the groups. The total antioxidant capacity, superoxide dismutase and glutathione peroxidase activities in the probiotic-treated groups were significantly increased (P < 0.05) as compared with the control groups. Both maleic dialdehyde concentration and superoxide anion activities in the probiotic-treated groups were significantly lower (P < 0.05) than those of the control. The probiotic did not affect the nitric oxide synthase and the catalase activity in any of the control and treated groups. These results indicated that the probiotic B. subtilis could significantly promote the growth rate of shrimp by increasing the immune function and antioxidant capacity. The most effective dose of B. subtilis in the diet was 5 x 104 CFU g-1 feed.

Abstract:An ammonia stress experiment was conducted by adjusting the concentration of total ammonia nitrogen (TAN) to 20 mg/L using ammonium chloride. The shrimp without ammonia stress was set as the control group. A total of 300 shrimp were randomly divided into six circular fiberglass tanks, each tank containing 50 shrimp. Antioxidant and immunological enzyme activities were investigated at five time points: 0 h, 12 h, 24 h, 48 h, and 96 h. The shrimp sampled at the initial time point were set as the control group. The samples used for intestinal microbiota determination were taken at two time points: 0 h and 96 h. All of the enzyme activities showed an initial increase followed by a decreasing trend. The enzyme activities (SOD, CAT, LZM, ACP, AKP) generally were higher at 12 h and 24 h compared to the control group, and then decreased some at 48 h and 96 h. The richness and diversity of intestinal microbiota decreased under ammonia stress. Proteobacteria, Firmicutes, and Bacteroidetes dominated in both the control and the 96-h group. Principal component analysis indicated a relatively small alteration of the microbiota by ammonia. Chitinophagaceae and Chloroflexi were the main species in the 96-h group, while Clostridium sensu stricto 1 and Paracoccus were the main species in the control group. Our findings suggest that ammonia exposure suppressed the antioxidant and immunological defenses of Litopenaeus vannamei. Ammonia exposure induced variation in the expression of intestinal microbiota abundance and composition, but the changes were unremarkable.
