Abstract:The need for dynamic, elastomeric polymeric biomaterials remains high, with few options with tunable control of mechanical properties, and environmental responses. Yet the diversity of these types of protein polymers pursued for biomaterials-related needs remains limited. Robust high-throughput synthesis and characterization methods will address the need to expand options for protein-polymers for a range of applications. Here, a combinatorial library approach with high throughput screening is used to select specific examples of dynamic protein silk-elastin-like polypeptides (SELPs) with unique stimuli responsive features, including tensile strength and adhesion. Using this approach, 64 different SELPs with different sequences and molecular weights are selected out of over 2000 recombinant E. coli colonies. New understanding of sequence-function relationships with this family of proteins is gained through this combinatorial-screening approach and can provide a guide to future library designs. Further, this approach yields new families of SELPs to match specific material functions.

Abstract:The combinational density of immobilized functional molecules on biomaterial surfaces directs cell behaviors. However, limited by the low efficiency of traditional low-throughput experimental methods, investigation and optimization of the combinational density remain daunting challenges. Herein, we report a high-throughput screening set-up to study biomaterial surface functionalization by integrating photo-controlled thiol-ene surface chemistry and machine learning-based label-free cell identification and statistics. Through such a strategy, a specific surface combinational density of polyethylene glycol (PEG) and arginine-glutamic acid-aspartic acid-valine peptide (REDV) leads to high endothelial cell (EC) selectivity against smooth muscle cell (SMC) was identified. The composition was translated as a coating formula to modify medical nickel-titanium alloy surfaces, which was then proved to improve EC competitiveness and induce endothelialization. This work provided a high-throughput method to investigate behaviors of co-cultured cells on biomaterial surfaces modified with combinatorial functional molecules.

Abstract:We introduce a novel hybrid microfluidic platform that utilizes hydrodynamic trapping and dielectrophoretic (DEP) forces to transfer cells and particles into semi-isolate microchambers. The electromicrofluidic chip is designed to form a high-throughput single-cell assay in a massively parallel manner. In the first operational phase, hydrodynamic forces transfer single particles into trap sites. Then, in the second phase, DEP forces transport them into adjacent low-shear microchambers. After assembling thousands of individual cells into an array of 10 000 microchambers, the chip adds single fluorescently labeled beads sensitive to the target proteins to study the cell protein secretion profiles at the single-cell level. To show this capability, we used this multiplexed on-chip immunoassay to analyze galectin-3 (Gal-3) and Gal-3 binding protein (Gal-3bp) secretion profiles in cancer (MCF-7) and normal (HEK-293T) cells. The ability to controllably add tens of required single beads sensitive to the target proteins in this high-throughput system is an answer to the challenges in previously introduced similar devices. The proposed biomedical microdevice has great potential for running fundamental long-term single-cell phenotypic and genotypic studies, with crucial applications in biology and medicine.

Abstract:The complete nucleotide sequence of the RSV cpts-248/404 live attenuated vaccine candidate was determined from cloned cDNA and was compared to that of the RSV A2/HEK7 wild-type, cold-passaged cp-RSV, and cpts-248 virus, which constitute the series of progenitor viruses. RSV cpts-248/404 is more attenuated and more temperature sensitive (ts) (shut-off temperature 36 degrees) than its cpts-248 parent virus (shut-off temperature 38 degrees) and is currently being evaluated in phase I clinical trials in humans. Our ultimate goal is to identify the genetic basis for the host range attenuation phenotype exhibited by cp-RSV (i.e., efficient replication in tissue culture but decreased replication in chimpanzees and humans) and for the ts and attenuation phenotypes of its chemically mutagenized derivatives, cpts-248 and cpts-248/404. Compared with its cpts-248 parent, the cpts-248/404 virus possesses an amino acid change in the polymerase (L) protein and a single nucleotide substitution in the M2 gene start sequence. In total, the cpts-248/404 mutant differs from its wild-type RSV A2/HEK7 progenitor in seven amino acids [four in the polymerase (L) protein, two in the fusion (F) glycoprotein, and one in the (N) nucleoprotein] and one nucleotide difference in the M2 gene start sequence. Heterogeneity at nucleotide position 4 (G or C, negative sense, compared to G in the RSV A2/HEK7 progenitor) in the leader region of vRNA developed during passage of the cpts-248/404 in tissue culture. Biologically cloned derivatives of RSV cpts-248/404 virus that differed at position 4 possessed the same level of temperature sensitivity and exhibited the same level of replication in the upper and lower respiratory tract of mice, suggesting that heterogeneity at this position is not clinically relevant. The determination of the nucleotide sequence of the cpts-248/404 virus will allow evaluation of the stability of the eight mutations that are associated with the attenuation phenotype during vaccine production and following replication in humans.

Abstract:Materiobiology is an emerging field focused on the physiochemical properties of biomaterials concerning biological outcomes which includes but is not limited to the biological responses and bioactivity of surface-modified biomaterials. Herein, we report a novel in vitro characterization platform for characterizing nanoparticle surface-modified 3D printed PLA scaffolds. We have introduced innovative design parameters that were practical for ubiquitous in vitro assays like those utilizing 96 and 24-well plates. Subsequently, gold and silica nanoparticles were deposited using two low-temperature plasma-assisted processes namely plasma electroless reduction (PER) and dusty plasma on 3D scaffolds. Materiobiological testing began with nanoparticle surface modification optimization on 96 well plate design 3D scaffolds. We have employed 3D laser confocal imaging and scanning electron microscopy to study the deposition of nanoparticles. It was found that the formation and distribution of the nanoparticles were time-dependent. In vitro assays were performed utilizing an osteosarcoma (MG-63) cell as a model. These cells were grown on both 96 and 24 well plate design 3D scaffolds. Subsequently, we performed different in vitro assays such as cell viability, and fluorescence staining of cytoskeletal actin and DNA incorporation. The actin cytoskeleton staining showed more homogeneity in the cell monolayer growing on the gold nanoparticle-modified 3D scaffolds than the control 3D PLA scaffold. Furthermore, the mineralization and protein adsorption experiments conducted on 96 well plate design scaffolds have shown enhanced mineralization and bovine serum albumin adsorption for the gold nanoparticle-modified scaffolds compared to the control scaffolds. Taken together, this study reports the efficacy of this new in vitro platform in conducting more reliable and efficient materiobiology studies. It is also worth mentioning that this platform has significant futuristic potential for developing as a high throughput screening platform. Such platforms could have a significant impact on the systematic study of biocompatibility and bioactive mechanisms of nanoparticle-modified 3D-printed scaffolds for tissue engineering. It would also provide unique ways to investigate mechanisms of biological responses and subsequent bioactive mechanisms for implantable biomaterials. Moreover, this platform can derive more consistent and reliable in vitro results which can improve the success rate of further in vivo experiments.

Abstract:In the field of tissue engineering, regenerative medicine, and life sciences, the topological biochemical cues regulate cell attachment and alignment within the construct. In a native biological system, these cues are inherent. However, most of the biological materials utilized in the fabrication of tissue construct do not possess the appropriate cues required to develop an architecture to support the cell attachment and growth of a functional tissue Therefore the ability to manipulate structural and biochemical cues plays an important role in biofabrication process, and it is a key element to evaluate a engineered cellular model. Plasma surface functionalization and biologics printing have been investigated and validated as two effective techniques to guide cell functions by creating microenvironments. The objective of this work is to develop a novel dual functional platform for freeform microplasma surface patterning and biologics printing process as well as to study the underlying process science and the process induced cellular functions. The microplasma jet system was assembled by two parts. The upper part is a plastic NPT connector surrounding an extending high voltage copper electrode. The lower part is a dielectric Pasteur pipette connected with a capillary micro-scale nozzle tip. The lower part is interchangeable and the diameter of the tip ranges from 50 pm to 1 mm. With up to 20 kV output capability, a high-voltage power supply was connected to the copper electrode through the NPT connector which also served as gas inlet. A high-voltage probe linked to an oscilloscope is used to monitor the real time voltage. The whole microplasma jet system was set up on automation platform, which allows X-Y-Z motion control and switch control. This integrated system operates at atmospheric pressured environment. All tissue constructs could be fabricated at room temperature without the use of a mask. Clear polystyrene microplates were used as plasma treatment substrates. After O-2-He mixed microplasma treatment, 7F2 mouse osteoblastic cells were cultured in the microplates for cell biology studies. We demonstrated the capability of our dual functional platform by applying microplasma in the polystyrene wells and control group (without any treatment) in other wells of the same microplate substrate. The results show that the microplasma treatment changed the surface properties and improved cell attachment. This dual functional freeform system allows for surface patterning and printing of cells, proteins, growth factors, etc. to fabricate three-dimensional tissue constructs.

Abstract:The lack of prediction accuracy during drug development and screening risks complications during human trials, such as drug-induced liver injury (DILI), and has led to a demand for robust, human cell-based, in vitro assays for drug discovery. Microporous polymer-based scaffolds offer an alternative to the gold standard flat tissue culture plastic (2D TCPS) and other 3D cell culture platforms as the porous material entraps cells, making it advantageous for automated liquid handlers and high-throughput screening (HTS). In this study, we optimized the surface treatment, pore size, and choice of scaffold material with respect to cellular adhesion, tissue organization, and expression of complex physiologically relevant (CPR) outcomes such as the presence of bile canaliculi-like structures. Poly-l-lysine and fibronectin (FN) coatings have been shown to encourage cell attachment to the underlying substrate. Treatment of the scaffold surface with NaOH followed with a coating of FN improved cell attachment and penetration into pores. Of the two pore sizes we investigated (A: 104 ± 4 μm; B: 175 ± 6 μm), the larger pore size better promoted cell penetration while limiting tissue growth from reaching the hypoxia threshold. Finally, polystyrene (PS) proved to be conducive to cell growth, penetration into the scaffold, and yielded CPR outcomes while being a cost-effective choice for HTS applications. These observations provide a foundation for optimizing microporous polymer-based scaffolds suitable for drug discovery. © 2018 American Institute of Chemical Engineers Biotechnol. Screening of perfused combinatorial 3D microenvironments for cell culture

Abstract:Biomaterials combining biochemical and biophysical cues to establish close-to-extracellular matrix (ECM) models have been explored for cell expansion and differentiation purposes. Multivariate arrays are used as material-saving and rapid-to-analyze platforms, which enable selecting hit-spotted formulations targeting specific cellular responses. However, these systems often lack the ability to emulate dynamic mechanical aspects that occur in specific biological milieus and affect physiological phenomena including stem cells differentiation, tumor progression, or matrix modulation. We report a tailor-made strategy to address the combined effect of flow and biochemical composition of three-dimensional (3D) biomaterials on cellular response. We suggest a simple-to-implement device comprising (i) a perforated platform accommodating miniaturized 3D biomaterials and (ii) a bioreactor that enables the incorporation of the biomaterial-containing array into a disposable perfusion chamber. The system was upscaled to parallelizable setups, increasing the number of analyzed platforms per independent experiment. As a proof-of-concept, porous chitosan scaffolds with 1 mm diameter were functionalized with combinations of 5 ECM and cell-cell contact-mediating proteins, relevant for bone and dental regeneration, corresponding to 32 protein combinatorial formulations. Mesenchymal stem cells adhesion and production of an early osteogenic marker were assessed on-chip on static and under-flow dynamic perfusion conditions. Different hit-spotted biomaterial formulations were detected for the different flow regimes using direct image analysis. Cell-binding proteins still poorly explored as biomaterials components - amelogenin and E-cadherin - were here shown as relevant cell response modulators. Their combination with ECM cell-binding proteins - fibronectin, vitronectin, and type 1 collagen - rendered specific biomaterial combinations with high cell adhesion and ALP production under flow. The developed versatile system may be targeted at widespread tissue regeneration applications, and as a disease model/drug screening platform. STATEMENT OF SIGNIFICANCE: A perfusion system that enables cell culture in arrays of three-dimensional biomaterials under dynamic flow is reported. The effect of 31 cell-binding protein combinations in the adhesion and alkaline phosphatase (ALP) production of mesenchymal stem cells was assessed using a single bioreactor chamber. Flow perfusion was not only assessed as a classical enhancer/accelerator of cell growth and early osteogenic differentiation. We hypothesized that flow may affect cell-protein interactions, and that key components driving cell response may differ under static or dynamic regimes. Indeed, hit-spotted formulations that elicited highest cell attachment and ALP production on static cell culture differed from the ones detected for dynamic flow assays. The impacting role of poorly studied proteins as E-cadherin and amelogenin as biomaterial components was highlighted.

Abstract:A polymer microarray based on the supramolecular ureido‐pyrimidinone (UPy) moiety was fabricated to screen antimicrobial materials for their ability to support cell adhesion. UPy‐functionalized additives, either cell‐adhesive, antimicrobial or control peptides, were used, and investigated in different combinations at different concentrations, resulting in a library of 194 spots. These were characterized on composition and morphology to evaluate the microarray fabrication. Normal human dermal fibroblasts were cultured on the microarrays and cell adhesion to the spots was systematically analyzed. Results demonstrated enhanced cell adhesion on spots with combinations including the antimicrobial peptides. This study clearly proves the power of the high throughput approach in combination with supramolecular molecules, to screen additive libraries for desired biological response. Harnessing Colloidal Self-Assembled Patterns (cSAPs) to Regulate Bacterial and Human Stem Cell Response at Biointerfaces In Vitro and In Vivo

Abstract:The generation of complex physicochemical signals on the surface of biomedical materials is still challenging despite the fact that a broad range of surface modification methods have been developed over the last few decades. Colloidal self-assembled patterns (cSAPs) are combinations of unique colloids differing in size and surface chemistry acting as building blocks that can be programmed to generate surface patterns with exquisite control of complexity. This study reports on producing a variety of pre-modified colloids for the fabrication of cSAPs as well as post-assembly modifications to yield complex surfaces. The surface of cSAPs presents hierarchical micro- and nanostructures, localized hydrophilic/hydrophobic characteristics, and tunable surface functionality imparted by the individual colloids. The selected cSAPs can control bacterial adhesion (S. aureus, P. aeruginosa, and E. coli) and affect the cell cycle of human bone marrow stem cells (hBMSCs). Moreover, in a mouse subcutaneous model, cSAPs with selective [2-(methacryloyloxy)ethyl]dimethyl-(3-sulfopropyl)ammonium (SBMA) modification can reduce the inflammatory response after being challenged with bacteria. Developing Novel Interface and Signal Amplification Strategies for Study of Biological Interactions by Surface Plasmon Resonance(SPR) and SPRimaing

Abstract:Surface plasmon resonance (SPR) has been widely used as a powerful analytical technique for the study of a broad range of biomolecular interactions. With the capability of real-time detection, SPR allows convenient and nondestructive measurement of analyte concentration and binding kinetics. To improve the performance of SPR biosensing, we have developed a series of novel methods that lead to ultrasensitive detection via signal amplification by coupling inline atom transfer radical polymerization (ATRP) to a biospecific interaction. To adapt this enhancement method to the fragile lipid membrane-based system, initiator functionalized gold nanoparticles (AuNPs) has been used with in situ ATRP reaction, which allows sensitive measurement of lipid membrane binding proteins. In addition, conjugated nanoparticles AuNPs and Fe3O4 NPs have been employed to enhance protein detection sensitivity with SPRimaging. A novel Au-well microarray, fabricated based on spatial variation of metal thickness, is utilized to eliminate background resonance and provide better performance. To mimic cell membrane recognition processes, a membrane bilayer has been assembled onto a nanoglassified surface, and a synthesized water-soluble deep cavitand was incorporated into the supported lipid membrane. The incorporated cavitand retains its host properties and real-time analysis of the host:guest properties is carried out by SPR and fluorescence microscopy. To further explore the biomolecule interactions on cell membrane, we have studied the polymer growth at the lipid membrane-water interface using functional guest initiator that triggers an ATRP process. A variety of functional polymers have been grown at the lipid membrane surface, demonstrating different hydrophilicity and protein binding properties. Mass spectrometry in combination with SPR for comprehensive protein analysis has been explored to build a new technical platform. We have designed a thermoresponsive poly(N-isopropylacrylamide) (PNIPAAM) grafted surface, which demonstrates temperature-mediated hydrophobicity for enrichment of proteins based on hydrophobic interaction. SPR quantitatively measures the binding of proteins and peptides, and the biochip is directly interfaced with matrix assisted laser desorption/ionization (MALDI)-MS to characterize the bound analytes. It provides outstanding results in simple steps of on-plate desalting, offering news tools for effective proteomic analysis.

Mycobacteria in Crohn’s disease: how innate immune deficiency may result in chronic inflammation

Abstract:Crohn’s disease (CD) is often considered to be an autoimmune condition or, alternatively, an autoinflammatory condition, based on the observation of host-directed inflammatory processes. However, the underlying basis of this deleterious inflammatory response remains elusive. Recent findings from genetic and genomic studies have altered the perspective on the pathogenesis of CD, hinting at defects in innate immune sensing of intracellular bacteria and the handling of these organisms through autophagy. These findings are consistent with emerging data from immunological studies that point to a systemic immune deficiency in CD patients. Both sets of data (genetic predisposition and immunodeficiency) are consistent with the longstanding hypothesis that mycobacteria might be involved in the etiology of CD. In this article, we discuss the convergence of these three lines of investigation and highlight important knowledge gaps required in order to address the mycobacterial hypothesis with greater clarity. In the coming years, clinical immunological investigations should focus on defining the specificity of functional immune defects with regards to microbes and their associated ligands. Should CD result from a dysfunctional host–pathogen interaction, elucidation of the microbes that can exploit such defects to induce a chronic inflammatory disease is critical for the development of subsequent diagnostic assays and clinical interventions.

Automated Microcontact Bioprinting for High-Throughput Manufacturing of Lubricant-Infused Microarrays

Abstract:Microarrays are widely used for detecting target analytes and biomarkers, with fabrication methods ranging from non-contact to contact bioprinting techniques. Microcontact bioprinting (μCP), which utilizes elastomeric stamps to transfer biorecognition molecules (bioinks) onto substrates, offers advantages such as customizability, cost-effectiveness, and versatility in handling bioinks with high viscosities. Despite its prevalent use in laboratory settings, μCP faces challenges in achieving the repeatability and reproducibility required for industrial manufacturing. In this study, we address these limitations by developing and optimizing a μCP protocol using industrial techniques. A key innovation in our approach is the combination of microcontact printing with fluorosilanization, enabling the use of lubricant-infused surfaces to prevent non-specific attachment. Additionally, we enhance biomolecule immobilization through covalent attachment using a modified bioink formulation. We identify and mitigate high-risk failure modes including bioink formulation, application and removal, environmental conditions, and force application during stamping. Furthermore, we integrate an automated syringe pump and a standardized force application system, taking critical steps toward industrial scalability. Using lubricant-infused substrates, our optimized μCP protocol demonstrates significant improvements in repeatability and reproducibility, achieving intra-assay and inter-assay coefficients of variance below 10% and signal-to-noise ratios exceeding 15. These advancements validate our μCP method for high-throughput, scalable microarray fabrication, paving the way for its implementation in industrial manufacturing.

Abstract:A series of magnesium sulphate phosphors MgSO4:RE3+,X ( RE = Dy, Tm, Eu and X = P, Mn) have been prepared and studied. Based on the experimental results of thermoluminescence (TL) emission spectra and dose responses studies, it is proposed that in these phosphors large defect complexes are formed, which include intrinsic imperfections and dopants. These defect complexes were formed in the course of preparation of phosphors and could be regarded as basic elements in the TL multi-stage processes. This defect model might be applicable to the most of TL materials.
Fabrication of Miniature Drug Screening Platform Using Low Cost Bioprinting Technology

Abstract:In the pharmaceutical industry, new chemicals and substances are being tested to find appropriate compounds or mix for treating a specific disease. The demand for screening large compound collections against and increasing number of therapeutic targets has stimulated technology development in the areas of assay automation and miniaturization. Unfortunately, some of these new compounds are rather hard to obtain, which causes an expensive research and limited material availability; therefore, increasing the time development for future cures.We have developed a new and low-cost deposition method to fabricate miniature drug screening platform that can realistically and inexpensively evaluate biochemical reactions up to 4 substances per trial in a picoliter-scale volume. This paper describes the development of the controls for a deposition method (inkjet printing technology) which will simultaneously place therapeutic drugs and cells onto target sites to fabricate cell/drug chips for drug screening application. Using a modified HP 5360 CD printer, droplets of GFP expressing Escherichia coli have been deposited in an agar coated coverslip chip as small reliable volume of 180 picoliters per each colony dot, along with this bacteria it has been patterned different antibiotics in such a way that we evaluated the growth of the bacteria under antibiotics presence. The viability and function of the printed cells were evaluated by the live/dead and plasmid gene transfection experiments resulting in more than 98% viability and maintaining DNA function. Moreover, it has been recorded as high throughput process printing 213 assays per second. Due to the reduction of volume, this method will increase the effectiveness of the resources utilized for emerging drug screening processes. The results show promising usage of resources for future drug screening through new biochemicals.
