Abstract:Generative AI models have recently achieved mainstream attention with the advent of powerful approaches such as stable diffusion, DALL-E and MidJourney. The underlying breakthrough generative mechanism of denoising diffusion modeling can generate high quality synthetic images and can learn the underlying distribution of complex, high-dimensional data. Recent research has begun to extend these models to medical and specifically neuroimaging data. Typical neuroimaging tasks such as diagnostic classification and predictive modeling often rely on deep learning approaches based on convolutional neural networks (CNNs) and vision transformers (ViTs), with additional steps to help in interpreting the results. In our paper, we train conditional latent diffusion models (LDM) and denoising diffusion probabilistic models (DDPM) to provide insight into Alzheimer’s disease (AD) effects on the brain’s anatomy at the individual level. We first created diffusion models that could generate synthetic MRIs, by training them on real 3D T1-weighted MRI scans, and conditioning the generative process on the clinical diagnosis as a context variable. We conducted experiments to overcome limitations in training dataset size, compute time and memory resources, testing different model sizes, effects of pretraining, training duration, and latent diffusion models. We tested the sampling quality of the disease-conditioned diffusion using metrics to assess realism and diversity of the generated synthetic MRIs. We also evaluated the ability of diffusion models to conditionally sample MRI brains using a 3D CNN-based disease classifier relative to real MRIs. In our experiments, the diffusion models generated synthetic data that helped to train an AD classifier (using only 500 real training scans) -and boosted its performance by over 3% when tested on real MRI scans. Further, we used implicit classifier-free guidance to alter the conditioning of an encoded individual scan to its counterfactual (representing a healthy subject of the same age and sex) while preserving subject-specific image details. From this counterfactual image (where the same person appears healthy), a personalized disease map was generated to identify possible disease effects on the brain. Our approach efficiently generates realistic and diverse synthetic data, and may create interpretable AI-based maps for neuroscience research and clinical diagnostic applications.

In our study, we trained latent diffusion models (LDM) and denoising diffusion probabilistic models (DDPM) specifically for generating synthetic Diffusion Tensor Imaging (DTI) maps. We developed models that generate synthetic DTI maps of mean diffusivity by training on real 3D DTI scans, and evaluating the synthetic data's realism and diversity using maximum mean discrepancy (MMD) and multi-scale structural similarity index (MS-SSIM). Additionally, we aim to assess the transfer learning capability of a 3D CNN-based Alzheimer′s disease classifier, by pretraining it on sex classification on real and synthetic DTIs. Our approach efficiently produces realistic and diverse synthetic data, potentially helping to create interpretable AI-driven maps for neuroscience research and clinical diagnostics.

An Empirical Analysis of Diffusion, Autoencoders, and Adversarial Deep Learning Models for Predicting Dementia Using High-Fidelity MRI

Abstract:This study explores cutting-edge computational technologies and intelligent methods to create realistic synthetic data, focusing on dementia-centric Magnetic Resonance Imaging (MRI) scans related to Alzheimer’s and Parkinson’s diseases. The research delves into Generative Adversarial Networks (GANs), Variational Autoencoders, and Diffusion Models, comparing their efficacy in generating synthetic MRI scans. Using datasets from Alzheimer’s and Parkinson’s patients, the study reveals intriguing findings. In the Alzheimer dataset, diffusion models produced non-dementia images with the lowest Frechet Inception Distance (FID) score at 92.46, while data-efficient GANs excelled in generating dementia images with an FID score of 178.53. In the Parkinson dataset, data-efficient GANs achieved remarkable FID scores of 102.71 for dementia images and 129.77 for non-dementia images. The study also introduces a novel aspect by incorporating a classification study, validating the generative metrics. DenseNets, a deep learning architecture, exhibited superior performance in disease detection compared to ResNets. Training both models on images generated by diffusion models further improved results, with DenseNet achieving accuracies of 80.84% and 92.42% in Alzheimer’s and Parkinson’s disease detection, respectively. The research not only presents innovative generative architectures but also emphasizes the importance of classification metrics, providing valuable insights into the synthesis and detection of neurodegenerative diseases through advanced computational techniques.

Counterfactual MRI Data Augmentation using Conditional Denoising Diffusion Generative Models

Abstract:Deep learning (DL) models in medical imaging face challenges in generalizability and robustness due to variations in image acquisition parameters (IAP). In this work, we introduce a novel method using conditional denoising diffusion generative models (cDDGMs) to generate counterfactual magnetic resonance (MR) images that simulate different IAP without altering patient anatomy. We demonstrate that using these counterfactual images for data augmentation can improve segmentation accuracy, particularly in out-of-distribution settings, enhancing the overall generalizability and robustness of DL models across diverse imaging conditions. Diffusion Models for Counterfactual Generation and Anomaly Detection in Brain Images

Abstract:Segmentation masks of pathological areas are useful in many medical applications, such as brain tumour and stroke management. Moreover, healthy counterfactuals of diseased images can be used to enhance radiologists' training files and to improve the interpretability of segmentation models. In this work, we present a weakly supervised method to generate a healthy version of a diseased image and then use it to obtain a pixel-wise anomaly map. To do so, we start by considering a saliency map that approximately covers the pathological areas, obtained with ACAT. Then, we propose a technique that allows to perform targeted modifications to these regions, while preserving the rest of the image. In particular, we employ a diffusion model trained on healthy samples and combine Denoising Diffusion Probabilistic Model (DDPM) and Denoising Diffusion Implicit Model (DDIM) at each step of the sampling process. DDPM is used to modify the areas affected by a lesion within the saliency map, while DDIM guarantees reconstruction of the normal anatomy outside of it. The two parts are also fused at each timestep, to guarantee the generation of a sample with a coherent appearance and a seamless transition between edited and unedited parts. We verify that when our method is applied to healthy samples, the input images are reconstructed without significant modifications. We compare our approach with alternative weakly supervised methods on the task of brain lesion segmentation, achieving the highest mean Dice and IoU scores among the models considered.

Generating Realistic Brain MRIs via a Conditional Diffusion Probabilistic Model

Abstract:As acquiring MRIs is expensive, neuroscience studies struggle to attain a sufficient number of them for properly training deep learning models. This challenge could be reduced by MRI synthesis, for which Generative Adversarial Networks (GANs) are popular. GANs, however, are commonly unstable and struggle with creating diverse and high-quality data. A more stable alternative is Diffusion Probabilistic Models (DPMs) with a fine-grained training strategy. To overcome their need for extensive computational resources, we propose a conditional DPM (cDPM) with a memory-efficient process that generates realistic-looking brain MRIs. To this end, we train a 2D cDPM to generate an MRI subvolume conditioned on another subset of slices from the same MRI. By generating slices using arbitrary combinations between condition and target slices, the model only requires limited computational resources to learn interdependencies between slices even if they are spatially far apart. After having learned these dependencies via an attention network, a new anatomy-consistent 3D brain MRI is generated by repeatedly applying the cDPM. Our experiments demonstrate that our method can generate high-quality 3D MRIs that share a similar distribution to real MRIs while still diversifying the training set. Adversarial Learning for MRI Reconstruction and Classification of Cognitively Impaired Individuals

Abstract:Game theory-inspired deep learning using a generative adversarial network provides an environment to competitively interact and accomplish a goal. In the context of medical imaging, most work has focused on achieving single tasks such as improving image resolution, segmenting images, and correcting motion artifacts. We developed a dual-objective adversarial learning framework that simultaneously 1) reconstructs higher quality brain magnetic resonance images (MRIs) that 2) retain disease-specific imaging features critical for predicting progression from mild cognitive impairment (MCI) to Alzheimer's disease (AD). We obtained 3-Tesla, T1-weighted brain MRIs of participants from the Alzheimer's Disease Neuroimaging Initiative (ADNI, N=342) and the National Alzheimer's Coordinating Center (NACC, N = 190) datasets. We simulated MRIs with missing data by removing 50% of sagittal slices from the original scans (i.e., diced scans). The generator was trained to reconstruct brain MRIs using the diced scans as input. We introduced a classifier into the GAN architecture to discriminate between stable (i.e., sMCI) and progressive MCI (i.e., pMCI) based on the generated images to facilitate encoding of disease-related information during reconstruction. The framework was trained using ADNI data and externally validated on NACC data. In the NACC cohort, generated images had better image quality than the diced scans (Structural similarity (SSIM) index: 0.553 ± 0.116 versus 0.348 ± 0.108). Furthermore, a classifier utilizing the generated images distinguished pMCI from sMCI more accurately than with the diced scans (F1-score: 0.634 ± 0.019 versus 0.573 ± 0.028). Competitive deep learning has potential to facilitate disease-oriented image reconstruction in those at risk of developing Alzheimer's disease.

Abstract:In recent years, brain health has received increasing attention, but conventional acquisition of brain MRI (magnetic resonance imaging) images still suffer from issues such as missing data, artifacts, and high costs, which hinders research and diagnosis. With the application of deep learning in medical image synthesis, low-cost, efficient, and high-quality medical MRI synthesis techniques have become a prominent research focus and have gradually matured. However, traditional methods for synthesizing magnetic resonance imaging (MRI) mostly rely on generative adversarial networks, which require fine-tuning of parameters and learning rates to achieve stringent Nash equilibrium conditions, leading to problems such as gradient explosions and mode collapse. Building upon the latest research in synthetic models DDPM (denoising diffusion probabilistic model), we propose a novel approach for 2D brain MRI image synthesis based on a lightweight denoising diffusion probabilistic model. This method improves the attention module in the denoising diffusion probabilistic model to make it more lightweight. Additionally, we adopt the smooth L1 loss function as a replacement for the traditional mean absolute error (L1 loss) by comparing the error between the 2D brain MRI images with added noise and the real noise for training the model. Finally, we validate the proposed model on the MRI Brain Tumor Classification dataset, demonstrating that it achieves high-quality synthesis results while significantly reducing the parameter count of the DDPM model.
A Quantitatively Interpretable Model for Alzheimer's Disease Prediction Using Deep Counterfactuals

Abstract:Deep learning (DL) for predicting Alzheimer's disease (AD) has provided timely intervention in disease progression yet still demands attentive interpretability to explain how their DL models make definitive decisions. Recently, counterfactual reasoning has gained increasing attention in medical research because of its ability to provide a refined visual explanatory map. However, such visual explanatory maps based on visual inspection alone are insufficient unless we intuitively demonstrate their medical or neuroscientific validity via quantitative features. In this study, we synthesize the counterfactual-labeled structural MRIs using our proposed framework and transform it into a gray matter density map to measure its volumetric changes over the parcellated region of interest (ROI). We also devised a lightweight linear classifier to boost the effectiveness of constructed ROIs, promoted quantitative interpretation, and achieved comparable predictive performance to DL methods. Throughout this, our framework produces an ``AD-relatedness index'' for each ROI and offers an intuitive understanding of brain status for an individual patient and across patient groups with respect to AD progression.

Abstract:By utilizing model interpretability, our deep learning framework enhances assessment of cognitive impairment from MRI scans, paving the way for improved generalizability in machine learning models. Development of deep learning models to evaluate structural brain changes caused by cognitive impairment in MRI scans holds significant translational value. The efficacy of these models often encounters challenges due to variabilities arising from different data generation protocols, imaging equipment, radiological artifacts, and shifts in demographic distributions. Domain generalization (DG) techniques show promise in addressing these challenges by enabling the model to learn from one or more source domains and apply this knowledge to new, unseen target domains. Here we present a framework that utilizes model interpretability to enhance the generalizability of classification models across various cohorts. We used MRI scans and clinical diagnoses from four independent cohorts: Alzheimer's Disease Neuroimaging Initiative (ADNI, n = 1821), the Framingham Heart Study (FHS, n = 304), the Australian Imaging Biomarkers & Lifestyle Study of Ageing (AIBL, n = 661), and the National Alzheimer's Coordinating Center (NACC, n = 4647). With this data, we trained a deep neural network to focus on areas of the brain identified as relevant to the disease for model training. Our approach involved training a classifier to differentiate between structural neurodegeneration in individuals with normal cognition (NC), mild cognitive impairment (MCI), and dementia due to Alzheimer's disease (AD). This was achieved by aligning class‐wise attention with a unified visual saliency prior, which was computed offline for each class using all the training data. Our method not only competes with state‐of‐the‐art approaches but also shows improved correlation with postmortem histology. This alignment with the gold standard evidence is a significant step towards validating the effectiveness of DG frameworks, paving the way for their broader application in the field.

Conditional Diffusion Models for Semantic 3D Brain MRI Synthesis

Abstract:Artificial intelligence (AI) in healthcare, especially in medical imaging, faces challenges due to data scarcity and privacy concerns. Addressing these, we introduce Med-DDPM, a diffusion model designed for 3D semantic brain MRI synthesis. This model effectively tackles data scarcity and privacy issues by integrating semantic conditioning. This involves the channel-wise concatenation of a conditioning image to the model input, enabling control in image generation. Med-DDPM demonstrates superior stability and performance compared to existing 3D brain imaging synthesis methods. It generates diverse, anatomically coherent images with high visual fidelity. In terms of dice score accuracy in the tumor segmentation task, Med-DDPM achieves 0.6207, close to the 0.6531 accuracy of real images, and outperforms baseline models. Combined with real images, it further increases segmentation accuracy to 0.6675, showing the potential of our proposed method for data augmentation. This model represents the first use of a diffusion model in 3D semantic brain MRI synthesis, producing high-quality images. Its semantic conditioning feature also shows potential for image anonymization in biomedical imaging, addressing data and privacy issues. TADM: Temporally-Aware Diffusion Model for Neurodegenerative Progression on Brain MRI

Abstract:Generating realistic images to accurately predict changes in the structure of brain MRI is a crucial tool for clinicians. Such applications help assess patients' outcomes and analyze how diseases progress at the individual level. However, existing methods for this task present some limitations. Some approaches attempt to model the distribution of MRI scans directly by conditioning the model on patients' ages, but they fail to explicitly capture the relationship between structural changes in the brain and time intervals, especially on age-unbalanced datasets. Other approaches simply rely on interpolation between scans, which limits their clinical application as they do not predict future MRIs. To address these challenges, we propose a Temporally-Aware Diffusion Model (TADM), which introduces a novel approach to accurately infer progression in brain MRIs. TADM learns the distribution of structural changes in terms of intensity differences between scans and combines the prediction of these changes with the initial baseline scans to generate future MRIs. Furthermore, during training, we propose to leverage a pre-trained Brain-Age Estimator (BAE) to refine the model's training process, enhancing its ability to produce accurate MRIs that match the expected age gap between baseline and generated scans. Our assessment, conducted on the OASIS-3 dataset, uses similarity metrics and region sizes computed by comparing predicted and real follow-up scans on 3 relevant brain regions. TADM achieves large improvements over existing approaches, with an average decrease of 24% in region size error and an improvement of 4% in similarity metrics. These evaluations demonstrate the improvement of our model in mimicking temporal brain neurodegenerative progression compared to existing methods. Our approach will benefit applications, such as predicting patient outcomes or improving treatments for patients.

Abstract:Deep learning has been widely investigated in brain image computational analysis for diagnosing brain diseases such as Alzheimer's disease (AD). Most of the existing methods built end-to-end models to learn discriminative features by group-wise analysis. However, these methods cannot detect pathological changes in each subject, which is essential for the individualized interpretation of disease variances and precision medicine. In this article, we propose a brain status transferring generative adversarial network (BrainStatTrans-GAN) to generate corresponding healthy images of patients, which are further used to decode individualized brain atrophy. The BrainStatTrans-GAN consists of generator, discriminator, and status discriminator. First, a normative GAN is built to generate healthy brain images from normal controls. However, it cannot generate healthy images from diseased ones due to the lack of paired healthy and diseased images. To address this problem, a status discriminator with adversarial learning is designed in the training process to produce healthy brain images for patients. Then, the residual between the generated and input images can be computed to quantify pathological brain changes. Finally, a residual-based multi-level fusion network (RMFN) is built for more accurate disease diagnosis. Compared to the existing methods, our method can model individualized brain atrophy for facilitating disease diagnosis and interpretation. Experimental results on T1-weighted magnetic resonance imaging (MRI) data of 1,739 subjects from three datasets demonstrate the effectiveness of our method.

Abstract:Abstract Recent advances in computer vision have shown promising results in image generation. Diffusion probabilistic models have generated realistic images from textual input, as demonstrated by DALL-E 2, Imagen, and Stable Diffusion. However, their use in medicine, where imaging data typically comprises three-dimensional volumes, has not been systematically evaluated. Synthetic images may play a crucial role in privacy-preserving artificial intelligence and can also be used to augment small datasets. We show that diffusion probabilistic models can synthesize high-quality medical data for magnetic resonance imaging (MRI) and computed tomography (CT). For quantitative evaluation, two radiologists rated the quality of the synthesized images regarding "realistic image appearance", "anatomical correctness", and "consistency between slices". Furthermore, we demonstrate that synthetic images can be used in self-supervised pre-training and improve the performance of breast segmentation models when data is scarce (Dice scores, 0.91 [without synthetic data], 0.95 [with synthetic data]).

Abstract:Deep learning models based on convolutional neural networks (CNNs) have been used to classify Alzheimer’s disease or infer dementia severity from T1-weighted brain MRI scans. Here, we examine the value of adding diffusion-weighted MRI (dMRI) as an input to these models. Much research in this area focuses on specific datasets such as the Alzheimer’s Disease Neuroimaging Initiative (ADNI), which assesses people of North American, largely European ancestry, so we examine how models trained on ADNI, generalize to a new population dataset from India (the NIMHANS cohort). We first benchmark our models by predicting “brain age” - the task of predicting a person’s chronological age from their MRI scan and proceed to AD classification. We also evaluate the benefit of using a 3D CycleGAN approach to harmonize the imaging datasets before training the CNN models. Our experiments show that classification performance improves after harmonization in most cases, as well as better performance for dMRI as input.

Abstract:High-resolution magnetic resonance imaging (MRI) can enhance lesion diagnosis, prognosis, and delineation. However, gradient power and hardware limitations prohibit recording thin slices or sub-1 mm resolution. Furthermore, long scan time is not clinically acceptable. Conventional high-resolution images generated using statistical or analytical methods include the limitation of capturing complex, high-dimensional image data with intricate patterns and structures. This study aims to harness cutting-edge diffusion probabilistic deep learning techniques to create a framework for generating high-resolution MRI from low-resolution counterparts, improving the denoising diffusion probabilistic model (DDPM) by minimizing its unpredictability and uncertainty. DDPM includes two processes. The forward process employs a Markov chain to systematically introduce Gaussian noise to low-resolution MRI images. In the reverse process, a U-Net model is trained to denoise the forward process images and produce high-resolution images conditioned on the features of their low-resolution counterparts. The proposed framework was demonstrated using T2-weighted MRI images from institutional prostate patients and brain patients collected in the Brain Tumor Segmentation Challenge 2020 (BraTS2020). For the prostate dataset, the bicubic interpolation model (Bicubic), conditional generative-adversarial network (CGAN), and our proposed DDPM framework improved the noise quality measure from low-resolution images by 4.4%, 5.7%, and 12.8%, respectively. Our method enhanced the signal-to-noise ratios by 11.7%, surpassing Bicubic (9.8%) and CGAN (8.1%). In the BraTS2020 dataset, the proposed framework and Bicubic enhanced PSNR from resolution-degraded images by 9.1% and 5.8%. The multi-scale structural similarity indexes were 0.970±0.019, 0.968±0.022, and 0.967±0.023 for the proposed method, CGAN, and Bicubic, respectively. This study explores a deep learning-based diffusion probabilistic framework for improving MR image resolution. Such a framework can be used to improve clinical workflow by obtaining high-resolution images without penalty of the long scan time. Future investigation will likely focus on prospectively testing the efficacy of this framework with different clinical indications.

Brain Imaging-to-Graph Generation using Adversarial Hierarchical Diffusion Models for MCI Causality Analysis

Abstract:Effective connectivity can describe the causal patterns among brain regions. These patterns have the potential to reveal the pathological mechanism and promote early diagnosis and effective drug development for cognitive disease. However, the current methods utilize software toolkits to extract empirical features from brain imaging to estimate effective connectivity. These methods heavily rely on manual parameter settings and may result in large errors during effective connectivity estimation. In this paper, a novel brain imaging-to-graph generation (BIGG) framework is proposed to map functional magnetic resonance imaging (fMRI) into effective connectivity for mild cognitive impairment (MCI) analysis. To be specific, the proposed BIGG framework is based on the diffusion denoising probabilistic models (DDPM), where each denoising step is modeled as a generative adversarial network (GAN) to progressively translate the noise and conditional fMRI to effective connectivity. The hierarchical transformers in the generator are designed to estimate the noise at multiple scales. Each scale concentrates on both spatial and temporal information between brain regions, enabling good quality in noise removal and better inference of causal relations. Meanwhile, the transformer-based discriminator constrains the generator to further capture global and local patterns for improving high-quality and diversity generation. By introducing the diffusive factor, the denoising inference with a large sampling step size is more efficient and can maintain high-quality results for effective connectivity generation. Evaluations of the ADNI dataset demonstrate the feasibility and efficacy of the proposed model. The proposed model not only achieves superior prediction performance compared with other competing methods but also predicts MCI-related causal connections that are consistent with clinical studies.

An interpretable generative multimodal neuroimaging-genomics framework for decoding Alzheimer's disease

Abstract:Alzheimer's disease (AD) is the most prevalent form of dementia with a progressive decline in cognitive abilities. The AD continuum encompasses a prodormal stage known as Mild Cognitive Impairment (MCI), where patients may either progress to AD or remain stable. In this study, we leveraged structural and functional MRI to investigate the disease-induced grey matter and functional network connectivity changes. Moreover, considering AD's strong genetic component, we introduce SNPs as a third channel. Given such diverse inputs, missing one or more modalities is a typical concern of multimodal methods. We hence propose a novel deep learning-based classification framework where generative module employing Cycle GANs was adopted to impute missing data within the latent space. Additionally, we adopted an Explainable AI method, Integrated Gradients, to extract input features relevance, enhancing our understanding of the learned representations. Two critical tasks were addressed: AD detection and MCI conversion prediction. Experimental results showed that our model was able to reach the SOA in the classification of CN/AD reaching an average test accuracy of $0.926\pm0.02$. For the MCI task, we achieved an average prediction accuracy of $0.711\pm0.01$ using the pre-trained model for CN/AD. The interpretability analysis revealed significant grey matter modulations in cortical and subcortical brain areas well known for their association with AD. Moreover, impairments in sensory-motor and visual resting state network connectivity along the disease continuum, as well as mutations in SNPs defining biological processes linked to amyloid-beta and cholesterol formation clearance and regulation, were identified as contributors to the achieved performance. Overall, our integrative deep learning approach shows promise for AD detection and MCI prediction, while shading light on important biological insights.

Predictive Modeling Of Alzheimer's Disease Prognosis Using Anatomical & Diffusion MRI

Abstract:Mild cognitive impairment (MCI) is an intermediate stage between healthy aging and Alzheimer's disease (AD), and AD is a progressive neurodegenerative disorder that affects around 50 million people worldwide. As new AD treatments begin to be developed, one key goal of AD research is to predict which individuals with MCI are most likely to progress to AD over a given interval (such as 2 years); if successful, these individuals could be preferentially enrolled in drug trials that aim to slow AD progression. Here we benchmarked a range of MCI-to-AD predictive models including linear regressions, support vector machines, and random forests, using predictors from anatomical and diffusion-weighted brain MRI, age, sex, APOE genotype and standardized clinical scores. In evaluations on 2,448 subjects (1,132 MCI, 883 healthy controls, 433 with dementia) from the ADNI study, models including PCA-compacted features achieved a balanced accuracy of 75.3% (using cortical features) and 89.7% using diffusion MRI measures on test set, suggesting the added prognostic value of microstructural metrics obtainable with diffusion MRI.
MCDDPM: Multichannel Conditional Denoising Diffusion Model for Unsupervised Anomaly Detection in Brain MRI

Abstract:Detecting anomalies in brain MRI scans using supervised deep learning methods presents challenges due to anatomical diversity and labor-intensive requirement of pixel-level annotations. Generative models like Denoising Diffusion Probabilistic Model (DDPM) and their variants like pDDPM, mDDPM, cDDPM have recently emerged to be powerful alternatives to perform unsupervised anomaly detection in brain MRI scans. These methods leverage frame-level labels of healthy brains to generate healthy tissues in brain MRI scans. During inference, when an anomalous (or unhealthy) scan image is presented as an input, these models generate a healthy scan image corresponding to the input anomalous scan, and the difference map between the generated healthy scan image and the original anomalous scan image provide the necessary pixel level identification of abnormal tissues. The generated healthy images from the DDPM, pDDPM and mDDPM models however suffer from fidelity issues and contain artifacts that do not have medical significance. While cDDPM achieves slightly better fidelity and artifact suppression, it requires huge memory footprint and is computationally expensive than the other DDPM based models. In this work, we propose an improved version of DDPM called Multichannel Conditional Denoising Diffusion Probabilistic Model (MCDDPM) for unsupervised anomaly detection in brain MRI scans. Our proposed model achieves high fidelity by making use of additional information from the healthy images during the training process, enriching the representation power of DDPM models, with a computational cost and memory requirements on par with DDPM, pDDPM and mDDPM models. Experimental results on multiple datasets (e.g. BraTS20, BraTS21) demonstrate promising performance of the proposed method.
