Abstract:The overwhelming potential of porous coordination polymers (PCP), also known as Metal-Organic Frameworks (MOFs), especially their nanostructures for various biomedical applications, have made these materials worth investigating for more applications and uses. MOFs unique structure has enabled them for most applications, particularly in biomedical and healthcare. A number of very informative review papers are available on the biomedical applications of MOFs for the reader's convenience. However, many of those reviews focus mainly on drug delivery applications, and no significant work has been reported on other MOFs for biomedical applications. This review aims to present a compact and highly informative global assessment of the recent developments in biomedical applications (excluding drug-delivery) of MOFs along with critical analysis. Researchers have recently adopted both synthetic and post-synthetic routes for the fabrication and modification of MOFs that have been discussed and analyzed. A critical review of the latest reports on the significant and exotic area of bio-sensing capabilities and applications of MOFs has been given in this study. In addition, other essential applications of MOFs, including photothermal therapy, photodynamic therapy, and antimicrobial activities, are also included. These recently grown emergent techniques and cancer treatment options have gained attention and require further investigations to achieve fruitful outcomes. MOF's role in these applications has been thoroughly discussed, along with future challenges and valuable suggestions for the research community that will help meet future demands.
Metal-organic frameworks for biomedical applications: A review

Abstract:Metal-organic frameworks (MOFs) are emergent materials in diverse prospective biomedical uses, owing to their inherent features such as adjustable pore dimension and volume, well-defined active sites, high surface area, and hybrid structures. The multifunctionality and unique chemical and biological characteristics of MOFs allow them as ideal platforms for sensing numerous emergent biomolecules with real-time monitoring towards the point-of-care applications. This review objects to deliver key insights on the topical developments of MOFs for biomedical applications. The rational design, preparation of stable MOF architectures, chemical and biological properties, biocompatibility, enzyme-mimicking materials, fabrication of biosensor platforms, and the exploration in diagnostic and therapeutic systems are compiled. The state-of-the-art, major challenges, and the imminent perspectives to improve the progressions convoluted outside the proof-of-concept, especially for biosensor platforms, imaging, and photodynamic therapy in biomedical research are also described. The present review may excite the interdisciplinary studies at the juncture of MOFs and biomedicine.

Abstract:In the past decade,nanoscale metal-organic frameworks（nMOFs） have drawn a great attention due to their high porosity,wide range of pore shapes,tunable frameworks and relatively low toxic.With the development of nanotechnology,many researchers studied the synthesis,characterization,functionalization and biotoxicity of nMOFs,and a more thorough understanding was developed about numerous nMOFs as promising platforms for biomedical applications.This review highlights the up-to-date progress of nMOFs related to their bio-applications such as drug delivery,bioimaging,biosensing and biocatalysis,and the common surface modification methods we re classified into four categories:covalent post-synthetically modification,coordinative post-synthetically modification,noncovalent postsynthetically modification and modification on the external surface.At the same time,the challenges and perspectives of nMOFs were discussed as well.
Bone marrow transplantation for β‐thalassaemia major: the UK experience in two paediatric centres

Abstract:Summary. Stem cell transplantation (SCT) remains the only cure for thalassaemia major. Recent advances in medical treatment make it even more important that accurate information is available regarding outcome of SCT in relevant patient populations in order to guide informed decisions regarding the most appropriate treatment for individual thalassaemia patients. We report the results of 55 consecutive first related allogeneic bone marrow transplants (BMT) for children with β‐thalassaemia major performed in two UK paediatric centres over 10 years. Between February 1991 and February 2001, 55 children underwent 57 allogeneic BMT. The median age at BMT was 6·4 years and the majority of patients (73%) originated from the Indian subcontinent. Using the Pesaro risk classification, 17 patients were class 1, 27 were class 2 and 11 were class 3. Actuarial overall survival and thalassaemia‐free survival at 8 years were 94·5% (95% CI 85·1–98·1) and 81·8% (95% CI 69·7–89·8) respectively. Despite the majority of patients being in class 2 or 3, transplant‐related mortality was low (5·4%). The principal complication was graft rejection accompanied by autologous reconstitution that occurred in 13·2% of transplants. Following modification of the conditioning regimen in 1993, the rejection rate fell to 4·6% and remained low. Acute graft‐versus‐host disease (GVHD) of grade II–IV occurred in 31% and chronic GVHD in 14·5%. These data compare favourably with survival with medical treatment for thalassaemia major and suggest that allogeneic BMT remains an important treatment option for children with β‐thalassaemia major, particularly when compliance with iron chelation is poor.

Abstract:Abundant connectivity among organic ligands and inorganic metal ions makes the physical and chemical characters of metal-organic frameworks (MOFs) could be precisely devised and modulated for specific applications. Especially nanoscale MOFs (NMOFs), a unique family of hybrid nanomaterials, with merits of holding the nature as the mainstay MOFs and demonstrating particle size in nanoscale range which enable them prospect platform in clinic. Adjustability of composition and structure allows NMOFs with different constituents, shapes, and characteristics. Oriented frameworks and highly porous provide enough space for packing therapeutic cargoes and various imaging agents efficiently. Moreover, the relatively labile metal-ligand bonds make NMOFs biodegradable in nature. So far, as a significant class of biomedically relevant nanomaterials, NMOFs have been explored as drug carriers, therapeutic preparation, and biosensing and imaging preparation owing to their high porosity, multifunctionality, and biocompatibility. This review provides up-to-date developments of NMOFs in biomedical applications with emphasis on size control, synthetic approaches, and surfaces functionalization as well as stability, degradation, and toxicity. The outlooks and several crucial issues of this area are also discussed, with the expectation that it may help arouse widespread attention on exploring NMOFs in potential clinical applications.

Abstract:Novel materials have been developed because of technological advancements combined with material research. Metal-organic frameworks (MOF) technology has been investigated for biomedical applications in this line. Nonetheless, as our team has learned from current literature, selecting metal ions/organic linkers, synthesis techniques, water stability/solubility, toxicity, and the possibility of biomolecules/drugs (enzyme, protein, DNA/RNA, and antibodies, among others) tagging/conjugation are the major challenges/factors. These issues/factors have an impact on MOFs' performance in biomedical applications, and they also raise a lot of doubts about its real-time biological utility in the near future. We targeted a comprehensive review on the MOFs for biomedical applications to keep these considerations in mind. The evolution of MOF technology is based on their interesting features such as biological or pharmacological activity, biocompatibility, limited toxicity, and particular host–guest interactions, as well as environmental friendliness. In this paper, we have summarized the state-of-the-art progress pertaining to MOFs' biomedical applications such as biosensing, biomedical, and drug delivery applications in this field that is still very new.

Abstract:Owing to their unique features, including high cargo loading, biodegradability, and tailorability, metal–organic frameworks (MOFs) and their composites have attracted increasing attention in various fields. In this review, application strategies of MOFs and their composites in nanomedicine with emphasis on their functions are presented, from drug delivery, therapeutic agents for different diseases, and imaging contrast agents to sensor nanoreactors. Applications of MOF derivatives in nanomedicine are also introduced. Besides, we summarize different functionalities related to MOFs, which include targeting strategy, biomimetic modification, responsive moieties, and other functional decorations. Finally, challenges and prospects are highlighted about MOFs in future applications.

Abstract:Metal-organic frameworks (MOFs) demonstrate excellent intrinsic characteristics for a variety of applications, including energy storage, catalysis, optics, sensory responses, and biomedical sciences. MOFs are being synthesized using various methods, and now efforts are mainly focused on designing a series of novel MOF composites with multifunctional attributes and physicochemical structures. Furthermore, current investigations are directed toward deploying the morphological and size properties to open new application windows. Cancer has turned into a hazard to human life, killing millions of people every year. Early-stage cancer detection is a challenging task, and most cancer deaths occur due to late-stage diagnosis because people do not get regular cancer screenings. Extensive research has been conducted on theranostic nanoplatforms that integrate several imaging technologies and therapy techniques for the detection and treatment of cancer. However, MOFs have been demonstrated to be effective as nanoplatforms for a wide range of biomedical applications due to their unique features, low cytotoxicity, high porosity, biodegradability due to labile metal–ligand bond, and tunable framework. MOF is an advanced material in the fight against cancer with significant importance. This review unlocks new routes to intricate MOFs in cancer diagnosis, drug delivery, bioimaging and highlights the development of these mechanisms for theranostic nanoplatforms. Furthermore, the future potential and significant obstacles of MOFs for cancer theranostics, bioimaging, and drug delivery are also demonstrated in this review.

Abstract:Metal-organic frameworks (MOFs) are a very promising platform for applications in various industries. In recent years, a variety of methods have been developed for the preparation and modification of MOFs, providing a wide range of materials for different applications in life science. Despite the wide range of different MOFs in terms of properties/sizes/chemical nature, they have not found wide application in biomedical practices at present. In this review, we look at the main methods for the preparation of MOFs that can ensure biomedical applications. In addition, we also review the available options for tuning the key parameters, such as size, morphology, and porosity, which are crucial for the use of MOFs in biomedical systems. This review also analyses possible applications for MOFs of different natures. Their high porosity allows the use of MOFs as universal carriers for different therapeutic molecules in the human body. The wide range of chemical species involved in the synthesis of MOFs makes it possible to enhance targeting and prolongation, as well as to create delivery systems that are sensitive to various factors. In addition, we also highlight how injectable, oral, and even ocular delivery systems based on MOFs can be used. The possibility of using MOFs as therapeutic agents and sensitizers in photodynamic, photothermal, and sonodynamic therapy was also reviewed. MOFs have demonstrated high selectivity in various diagnostic systems, making them promising for future applications. The present review aims to systematize the main ways of modifying MOFs, as well as the biomedical applications of various systems based on MOFs.

Abstract:Metal-Organic Frameworks (MOFs) have emerged as a promising biomedical material due to its unique features such as high surface area, pore volume, variable pore size, flexible functional groups, and excellent efficiency for drug loading. In this review, we explored the use of novel and smart metal organic frameworks as drug delivery vehicles to discover a safer and more controlled mode of drug release aiming to minimize their side effects. Here, we systematically discussed the background of MOFs following a thorough review on structural and physical properties of MOFs, their synthesis techniques, and the important characteristics to establish a strong foundation for future research. Furthermore, the current status on the potential applications of MOF-based stimuli-responsive drug delivery systems, including pH-, ion-, temperature-, light-, and multiple responsive systems for the delivery of anticancer drugs has also been presented. Lastly, we discuss the prospects and challenges in implementation of MOF-based materials in the drug delivery. Therefore, this review will help researchers working in the relevant fields to enhance their understanding of MOFs for encapsulation of various drugs as well as their stimuli responsive mechanism.

Abstract:Metal-organic frameworks (MOFs), also known as coordination polymers, have attracted extensive research interest in the past few decades due to their unique physical structures and potentially vast applications. In this review, we outline the recent progress in the synthesis, functionalization and applications of MOFs in biomedicine, mainly focusing on two promising, yet challenging areas, i.e., drug delivery and biosensing applications. A major challenge is the proper functionalization of MOFs with demanding properties suitable for biomedical applications. Extensive studies on MOFs in biomedicine have led to substantial progress in the control of key properties of MOFs such as toxicity, size and shape, and biological stability. Due to their flexible composition, pore size and easy functionalization properties, MOFs can be utilized as key components for the development of various functional systems, and their applications in drug delivery and biosensing are reviewed. Future trends and perspectives in these research areas are also outlined.

Abstract:A nanozyme is a nanoscale material having enzyme-like properties. It exhibits several superior properties, including low preparation cost, robust catalytic activity, and long-term storage at ambient temperatures. Moreover, high stability enables repetitive use in multiple catalytic reactions. Hence, it is considered a potential replacement for natural enzymes. Enormous research interest in nanozymes in the past two decades has made it imperative to look for better enzyme-mimicking materials for biomedical applications. Given this, research on metal–organic frameworks (MOFs) as a potential nanozyme material has gained momentum. MOFs are advanced hybrid materials made of inorganic metal ions and organic ligands. Their distinct composition, adaptable pore size, structural diversity, and ease in the tunability of physicochemical properties enable MOFs to mimic enzyme-like activities and act as promising nanozyme candidates. This review aims to discuss recent advances in the development of MOF-based nanozymes (MOF-NZs) and highlight their applications in the field of biomedicine. Firstly, different enzyme-mimetic activities exhibited by MOFs are discussed, and insights are given into various strategies to achieve them. Modification and functionalization strategies are deliberated to obtain MOF-NZs with enhanced catalytic activity. Subsequently, applications of MOF-NZs in the biosensing and therapeutics domain are discussed. Finally, the review is concluded by giving insights into the challenges encountered with MOF-NZs and possible directions to overcome them in the future. With this review, we aim to encourage consolidated efforts across enzyme engineering, nanotechnology, materials science, and biomedicine disciplines to inspire exciting innovations in this emerging yet promising field.

Abstract:Abstract Metal–organic frameworks (MOFs) have a high specific surface area, adjustable pores and can be used to obtain functional porous materials with diverse and well-ordered structures through coordination and self-assembly, which has intrigued wide interest in a broad range of disciplines. In the arena of biomedical engineering, the functionalized modification of MOFs has produced drug carriers with excellent dispersion and functionalities such as target delivery and response release, with promising applications in bio-detection, disease therapy, tissue healing, and other areas. This review summarizes the present state of research on the functionalization of MOFs by physical binding or chemical cross-linking of small molecules, polymers, biomacromolecules, and hydrogels and evaluates the role and approach of MOFs functionalization in boosting the reactivity of materials. On this basis, research on the application of functionalized MOFs composites in biomedical engineering fields such as drug delivery, tissue repair, disease treatment, bio-detection and imaging is surveyed, and the development trend and application prospects of functionalized MOFs as an important new class of biomedical materials in the biomedical field are anticipated, which may provide some inspiration and reference for further development of MOF for bio-medical applications.

Abstract:Metal−organic frameworks (MOFs) are a novel class of porous hybrid organic−inorganic materials that have attracted increasing attention over the past decade. MOFs can be used in chemical engineering, materials science, and chemistry applications. Recently, these structures have been thoroughly studied as promising platforms for biomedical applications. Due to their unique physical and chemical properties, they are regarded as promising candidates for disease diagnosis and drug delivery. Their well-defined structure, high porosity, tunable frameworks, wide range of pore shapes, ultrahigh surface area, relatively low toxicity, and easy chemical functionalization have made them the focus of extensive research. This review highlights the up-to-date progress of MOFs as potential platforms for disease diagnosis and drug delivery for a wide range of diseases such as cancer, diabetes, neurological disorders, and ocular diseases. A brief description of the synthesis methods of MOFs is first presented. Various examples of MOF-based sensors and DDSs are introduced for the different diseases. Finally, the challenges and perspectives are discussed to provide context for the future development of MOFs as efficient platforms for disease diagnosis and drug delivery systems.

Abstract:For decades, nanoscale metal-organic frameworks (nMOFs) have attracted extensive interest in biomedicine due to their distinct characteristics, including facile synthesis, porous interior, and tunable biocompatibility. With high porosity, versatile nMOFs allow for the facile encapsulation of various therapeutic agents with exceptionally high payloads. Constructed from metal ions and organic linkers through coordination bonds, nMOFs with plentiful functional groups enable the surface modification for active targeting and enhanced biocompatibility. This review outlines the up-to-date progresses on the exploration of nMOFs in the field of biomedicine. First, the classification and synthesis of nMOFs are discussed, followed by the concrete introduction of drug loading strategies of nMOFs and mechanisms of stimulation-responsive drug release. Second, the smart designs of the nMOFs-based platforms for anticancer and antibacterial treatment are summarized. Finally, the basic challenges faced by nMOFs research and the great potential of biomimetic nMOFs are presented. This review article affords an inspiring insight into the interdisciplinary research of nMOFs and their biomedical applications, which holds great expectation for their further clinical translation.

Abstract:Metal–organic frameworks (MOFs), a burgeoning class of coordination polymers, have garnered significant attention due to their outstanding structure, porosity, and stability. They have been extensively studied in catalysis, energy storage, water harvesting, selective gas separation, and electrochemical applications. Recent advancements in post-synthetic strategies, surface functionality, and biocompatibility have expanded the application scope of MOFs, particularly in various biomedical fields. Herein, we review MOF-based nanomaterials bioimaging nanoplatforms in magnetic resonance imaging, computed tomography, and fluorescence imaging. MOFs serve as the foundation for biosensors, demonstrating efficiency in sensing H 2 O 2 , tumor biomarkers, microRNA, and living cancer cells. MOF-based carriers are well designed in drug delivery systems and anticancer treatment therapies. Additionally, we examine the challenges and prospects of MOFs in surface modification, release of metal ions, and interaction with intracellular components, as well as their toxicity and long-term effects.

Abstract:Since the early reports of MOFs and their interesting properties, research involving these materials has grown wide in scope and applications. Various synthetic approaches have ensued in view of obtaining materials with optimised properties, the extensive scope of application spanning from energy, gas sorption, catalysis biological applications has meant exponentially evolved over the years. The far‐reaching synthetic and PSM approaches and porosity control possibilities have continued to serve as a motivation for research on these materials. With respect to the biological applications, MOFs have shown promise as good candidates in applications involving drug delivery, BioMOFs, sensing, imaging amongst others. Despite being a while away from successful entry into the market, observed results in sensing, drug delivery, and imaging put these materials on the spot light as candidates poised to usher in a revolution in biology. In this regard, this review article focuses current approaches in synthesis, post functionalization and biological applications of these materials with particular attention on drug delivery, imaging, sensing and BioMOFs.

Abstract:Recently, metal-organic frameworks (MOFs) have garnered enormous attention from researchers owing to their superior physicochemical properties, which are of particular interest in various fields such as catalysis and the diverse areas of biomedicine. Despite their position in the utilization for various applications compared to other innovative nanocarriers such as dendrimers and mesoporous silica nanoparticles (MSNs), in terms of advantageous physicochemical attributes, as well as attractive textural properties, ease of characterization, and abundant surface chemistry for functionalization and other benefits, MOFs yet suffer from several issues such as poor degradability, which might lead to accumulation-induced biocompatibility risk. In addition, some of the MOFs suffer from a shortcoming of poor colloidal stability in the aqueous solution, hindering their applicability in diverse biomedical fields. To address these limitations, several advancements have been made to fabricate polymeric nanocomposites of MOFs for their utility in various biomedical fields. In this review, we aim to provide a brief emphasis on various organic polymers used for coating over MOFs to improve their physicochemical attributes considering a series of recently reported intriguing studies. Finally, we summarize with perspectives.

Abstract:Abstract Investigation of metal–organic frameworks (MOFs) for biomedical applications has attracted much attention in recent years. MOFs are regarded as a promising class of nanocarriers for drug delivery owing to well-defined structure, ultrahigh surface area and porosity, tunable pore size, and easy chemical functionalization. In this review, the unique properties of MOFs and their advantages as nanocarriers for drug delivery in biomedical applications were discussed in the first section. Then, state-of-the-art strategies to functionalize MOFs with therapeutic agents were summarized, including surface adsorption, pore encapsulation, covalent binding, and functional molecules as building blocks. In the third section, the most recent biological applications of MOFs for intracellular delivery of drugs, proteins, and nucleic acids, especially aptamers, were presented. Finally, challenges and prospects were comprehensively discussed to provide context for future development of MOFs as efficient drug delivery systems.

Abstract:This review based on Zn-based MOF is summarized on new insights for targeted drug delivery of medicinal compounds and developed for medicinal applications. The stability of the MOF may be controlled by functionalization and the proper structure and composition choices. Thus, one of the most crucial factors to consider when it comes to bio-applications of MOF is the management of their particle size distribution and the alteration of their surface. Recent research was shown that Zn-based MOF can be used as a viable delivery system for non-toxic/biocompatible medicines and therapeutic agents in biological and medical applications. The toxicity, biocompatibility, stability, and recently reported production techniques for MOF's zinc-based building block as a possible platform with biological applications were explored and presented in this review study to offer ideal venues for further research. The factors including The surface area and pore volumes, functional groups, and synthetic methods for adsorption and desorption of drugs were studied. Antibacterial and anticancer activities of Zn-based MOF and their mechanisms were investigated. At last, the adsorption and release mechanism of drugs was investigated.
